ABSTRACT: The objective of the present study was to detect the genetic diversity of Anaplasma marginale strains in naturally infected calves from a rural property located in the northeastern region of the state of Pará, Eastern Amazon, which has a history of mortality due to anaplasmosis. Fourteen calves positive for A. marginale were selected using a semi-nested polymerase chain reaction for the target msp1α gene, with asymptomatic (n=3) and symptomatic (n=11) infections. After sequencing the samples, two genotypes were verified in the E and C regions and the structures in tandem repeats were determined. Nine different strains were found: eight related to the E genotype (α-β-β-Γ = one animal, asymptomatic; 16-F-17-F-F = two animals, symptomatic; α-β-F-F-F-F = one animal, asymptomatic; 31-62-62-61 = one animal, symptomatic; τ-10-3 = three animals, two symptomatic and one asymptomatic; α-β-β-β = one animal, symptomatic; τ-22 -13-18 = two animals, both symptomatic; β-β-β-BRA1-31 = two animals, both symptomatic), and one related to genotype C (23-24-25-31-27-27 = one animal, asymptomatic). Genotype E was predominant in 92.86% of the samples (13/14), followed by genotype C (7.14%). This study made it possible to detect the genetic diversity of A. marginale in calves from the selected dairy farm, in addition to identifying the BRA1 sequence in the animals of the present study, which was recently diagnosed in Minas Gerais, demonstrating the dispersion of A. marginale strains in herds from different Brazilian states. Genetic diversity of A. marginale was observed in both symptomatic and asymptomatic calves. There were no significant differences when clinical signs were compared to the genotype verified in the infected animals. The prevalence of pathogenicity was not observed.

RESUMO: O objetivo do presente trabalho foi detectar a diversidade genética de cepas de Anaplasma marginale em bezerros naturalmente infectados oriundos de uma propriedade rural localizada na região nordeste do estado do Pará, Amazônia Oriental, a qual apresentava histórico de mortalidade devido à anaplasmose. Foram selecionados 14 bezerros positivos para A. marginale pela técnica de semi-nested PCR (nPCR) para o alvo no gene msp1α, com infecção assintomática (n=3) e sintomáticos (n=11). Após o sequenciamento das amostras foram verificados dois genótipos nas regiões E e C, e determinadas as estruturas em tandem repeats. Nove diferentes estirpes foram encontradas, sendo oito relacionadas ao genótipo E (α-β-β-Γ = um animal, assintomático; 16-F-17-F-F = dois animais, sintomáticos; α-β-F-F-F-F = um animal, assintomático; 31-62-62-61 = um animal, sintomático; τ-10-3 = três animais, dois sintomáticos e um assintomático; α-β-β-β = um animal, sintomático; τ-22-13-18 = dois animais, sintomáticos; β-β-β-BRA1-31 = dois animais, sintomáticos) e uma relacionada ao genótipo C (23-24-25-31-27-27 = um animal, assintomático). O genótipo E foi predominante em 92,86% das amostras (13/14), seguido pelo genótipo C (7,14%). O estudo possibilitou a detecção da diversidade genética de A. marginale em bezerros dessa propriedade leiteira, além de identificar a sequência BRA1 nos animais do presente estudo, a qual foi diagnosticada recentemente em Minas Gerais, o que demonstra a dispersão das estirpes de A. marginale nos rebanhos de diferentes estados brasileiros. A diversidade genética de A. marginale foi observada tanto em bezerros sintomáticos quanto em assintomáticos e não houve diferença significativa quando se comparou os sinais clínicos ao genótipo verificado no animal infectado, não observando a prevalência de patogenicidade de estirpes.

ABSTRACT: The study aimed to describe the clinicopathological aspects of 37 cases of pythiosis, 34 in horses and three in mules, from properties located in the Amazon biome of Pará, Brazil. The clinical signs observed in the animals were weakness, poor-to-regular nutritional status, pale mucous membranes, itching at the lesion site, and lameness when the limbs were affected. The lesions were located on the lips, nostrils, rib region, thoracic and abdominal walls, scapular, distal limbs, foreskin, perineum and udder. Macroscopically, ulcerative and granulation-tissue-like masses were observed, with fistulous tracts filled with yellowish and foul-smelling serosanguinous discharges. In the biopsy, it was possible to visualize white and firm areas with foci of yellowish necrotic material and hardened masses, called kunkers. The histopathological examination showed a pyogranulomatous inflammatory reaction with the presence of Pythium insidiosum hyphae, which were impregnated with black, confirming the diagnosis of pythiosis in equids in the Amazon biome, being the first report of the disease in mules in the region.

RESUMO: O trabalho objetivou descrever os aspectos clínico-patológicos de 37 casos suspeitos de pitiose, 34 em equinos e três em muares, provenientes de propriedades localizadas no Pará, bioma amazônico brasileiro. Os sinais clínicos observados nos animais eram caracterizados por debilidade, estado nutricional de ruim a regular, mucosas pálidas, prurido no local da lesão, além de claudicação quando os membros foram acometidos. As lesões eram localizadas nos lábios, narinas, região das costelas, parede torácica e abdominal, escapular, distais dos membros, prepúcio, períneo e úbere. Macroscopicamente observavam-se extensas lesões ulceradas com intensa proliferação de tecido de granulação, de bordos irregulares, com tratos fistulosos, de consistência firme, denominados de “kunkers”, preenchidos com material amarelado e friável, possuindo exsudação serossanguinolenta de odor fétido. Ao exame histopatológico observou-se reação inflamatória piogranulomatosa com presença de hifas de Pythium insidiosum, as quais se impregnaram de negro, confirmando o diagnóstico de pitiose em equídeos no Bioma Amazônico, sendo o primeiro relato da doença em muares na região.

ABSTRACT: Birds, cartilaginous and teleost fish, reptiles, and some amphibians have intrascleral cartilage and/or bone; however, these are rarely reported in therian mammals. This study aimed to investigate and characterize a nonpathological formation of cartilage in the posterior sclera of sheep macroscopically, histologically, and by immunohistochemical exam (IHC). Ninety eyes from 45 domestic sheep were collected, underwent gross examination, fixed in formalin, and embedded in paraffin for the microscopical assessment. Sections with histological shreds of cartilage were selected to perform IHC to confirm the presence of cartilage. Intrascleral cartilage was detected in 60 eyeballs (66.66%) from 37 sheep (82.22%). A slight whitish thickening was grossly seen in the posterior sclera. The histologic exam revealed a few scattered, isolated chondrocytes to larger aggregates of cartilaginous islands in the posterior sclera. Eighteen (30%) of 60 eyeballs revealed marked anti-collagen type II immunolabeling. The development of cartilaginous structures in the eyes is considered rare in mammalian animals. The high occurrence of intrascleral cartilage in the examined sheep eyes suggests that this finding corresponds to an anatomical component of sheep sclera, despite the age, breed, or body condition.

RESUMO: Aves, peixes cartilaginosos e teleósteos e répteis, bem como alguns anfíbios possuem cartilagem e/ou osso intraescleral, contudo, isso é raramente descrito em marsupiais. O objetivo deste trabalho foi investigar e caracterizar a formação não patológica de cartilagem hialina na esclera posterior de ovinos, por exame macroscópico, histológico e imuno-histoquímico. Olhos de 45 ovinos foram coletados, submetidos a exame macroscópico, fixados em formalina a 10% tamponada e incluídos em parafina para o exame histológico. Amostras com evidência de formação cartilaginosa ao exame histológico foram selecionadas para a realização da técnica de imuno-histoquímica para confirmação. Cartilagem intraescleral foi detectada em 60 amostras oculares (66,66%) de 37 ovinos (82,22%), caracterizada na avaliação macroscópica como discretos espessamentos esbranquiçados na esclera posterior. O exame histológico revelou formações que variaram de condrócitos isolados e dispersos a grandes agregados, formando ilhas cartilaginosas na esclera. Dezoito (30%) de 60 bulbos oculares mostraram imunomarcação anti-colágeno tipo II. O desenvolvimento de estruturas cartilaginosas nos bulbos oculares é considerado raro em mamíferos. A identificação de cartilagem intraescleral em 66% dos olhos ovinos avaliados no estudo indica que a formação de cartilagem corresponde a um componente anatômico normal na esclera de ovinos, independente de idade, raça ou condição corporal.

ABSTRACT: This study collected samples from 50 Holstein cows, most intensively bred, and from the Campos Gerais region, Paraná, with an average milk production of 30.21L/day. Samples of the liver, spleen and lymph nodes were collected to determine the levels of copper, cobalt, iron, molybdenum and zinc. Spleen and lymph nodes were subjected to histological analysis and evaluation of the degree of hemosiderosis. The average concentrations of copper (495.05ppm), molybdenum (4.19ppm), and zinc (274.49ppm) were higher than those established for the bovines. For cobalt 26% of the animals presented levels below the established level, which characterized cobalt deficiency. Only iron (299.12ppm) exhibited an adequate average level. Histopathologically hemosiderosis was observed mainly in the spleen (78%) and less frequently in the lymph nodes (20%). The observation of hemosiderin in the spleen and lymph nodes is not related to copper deficiency. Still it may be related to high levels of molybdenum, zinc, iron, or other undetermined causes.

RESUMO: Para este estudo foram coletadas amostras de 50 vacas, da raça Holandesa, a maior parte criada intensivamente e oriunda da região dos Campos Gerais/PR, com média individual de produção de leite de 30,21L/dia. Foram coletadas amostras de fígado, baço e linfonodos para determinação dos níveis de cobre (Cu), cobalto (Co), ferro (Fe), molibdênio (Mo) e zinco (Zn). Baço e linfonodo foram submetidos à análise histológica e avaliação do grau de hemossiderose. A média das concentrações de Cu (495,05ppm), Mo (4,19ppm) e Zn (274,49ppm) encontrava-se acima dos níveis estabelecidos para bovinos. Para o Co, observou-se que 26% dos animais apresentaram níveis abaixo do estabelecido, o que caracterizou deficiência de Co. Apenas o Fe (299,12ppm) apresentou níveis médios adequados. Na histopatologia, hemossiderose foi observada, principalmente no baço (78%) e com menos frequência nos linfonodos (20%). A observação de hemossiderina no baço e linfonodos não está relacionada à deficiência de Cu, porém pode estar relacionada a elevados níveis de Mo, Zn e Fe ou a outras causas não determinadas.

ABSTRACT: The samples were taken from 106 cows with various-looking lesions on their teats and ranged in age from 2 to 8 years. Enzyme-linked immunosorbent assay (ELISA) antigen (Ag) positive for the bovine papillomavirus (BPV) was found in 59 (55.7%) blood serum samples. PCR using FAP59/64 primers was positive for 24 (22.6%) samples. BPV-2 (40, 37.7%), BPV-6 (28, 26.4%), BPV-8 (30, 28.3%), BPV-9 (36, 34%), BPV-10 (32, 30.3%), and BPV-12 (22, 20.8%) were found in a PCR type-specific analysis of single and mixed type teat warts. The highest positivity was observed in BPV-2, BPV-9 and BPV-10 in flat and round forms, BPV-6, BPV-10, BPV-12, and mixed types in rice grain-cauliflower forms, BPV-9 and mixed types in filiform in the distribution of types based on the macroscopic appearance of teat lesions. As for the distribution of BPV types according to age, the most BPV-2 types were found in the age group of two years, the most BPV-10 types in the age group of three years, the most BPV-9 types in the age group of four years, the most BPV-8+BPV-12 types in the age group of five years, and the most mixed types between the ages of six and eight years. The existence of the virus was then checked using electron microscopy on the chosen samples (at least one investigation was conducted), and it was positively identified using BPV type-specific primers. The authors concluded that BPV detection using an ELISA (Ag) test from blood serum samples was shown to be less sensitive than BPV type-specific PCR from wart samples.

ABSTRACT: Laminitis is a disease that affects the dermis and epidermis of the bovine hoof, generating changes in the hoof capsule. This study evaluated the effects of clinical laminitis diagnosed after the adaptation phase to confinement on the morphology, density, and mineral composition of the hoof of Nellore cattle after finishing. The animals were separated in the first weeks of confinement into a sick group (SG), with clinical laminitis, and a healthy group (HG). SG animals had higher heel length, dorsal wall length, toe height, and diagonal hoof length (p<0.05) than healthy animals. The dermal laminae had similar measurements for thickness, length, and spacing between them between SG and HG. Animals with laminitis showed congestion, hemorrhage, and basement membrane irregularities on histology. Computed microtomography (μCT) revealed that the hoof density of sick animals is lower than healthy ones. According to the mineral composition by energy-dispersive X-ray fluorescence (ED-XRF) spectrometry, the hooves of animals with laminitis (SG) and healthy ones (HG) were not biochemically different. Therefore, the occurrence of clinical laminitis in Nellore cattle in the first weeks of confinement causes an increase in the morphometric parameters of the hoof capsule and a reduction in the density of the abaxial hoof wall evaluated after the finishing period. This disease does not promote changes in the histomorphometric parameters of the dermal laminae and the percentage of minerals in the abaxial hoof wall.

RESUMO: A laminite é uma doença que afeta a derme e epiderme do casco de bovinos gerando alterações no estojo córneo. O estudo avaliou os efeitos da laminite clínica diagnosticada após a fase de adaptação ao confinamento na morfologia, densidade e composição mineral do casco de bovinos da raça Nelore após terminação. Nas primeiras semanas de confinamento, os animais foram separados em um grupo doente (GD) com laminite clínica e em um grupo saudável (GS). Os animais do GD apresentaram maior comprimento de talão, comprimento da parede dorsal, altura da pinça e comprimento diagonal do casco (p<0,05) do que os saudáveis. As lâminas dérmicas tiveram medidas semelhantes para espessura, comprimento e espaçamento entre elas entre GD e GS. Animais doentes apresentaram congestão, hemorragia e irregularidades da membrana basal na histologia. A microtomografia computadorizada (μCT) revelou que a densidade do casco de animais doentes é menor do que o saudável. Para a composição mineral por meio da espectrometria de fluorescência de raio-X por dispersão de energia (ED-XRF), o casco dos animais doentes (GD) e dos saudáveis (GS), não se mostraram diferentes bioquimicamente. Conclui-se que a ocorrência de laminite clínica em bovinos da raça Nelore nas primeiras semanas de confinamento ocasiona aumento de parâmetros morfométricos do estojo córneo e redução da densidade da parede abaxial do casco, avaliados após o período de terminação. Essa enfermidade não promove modificações nos parâmetros histomorfométricos das lâminas dérmicas e na porcentagem de minerais da parede abaxial do casco.

ABSTRACT: Mast cell tumors are one of the most common neoplasia in dogs and cytopathology and/or histopathology examinations are used for diagnosis. Histologic grading is considered the gold standard test to predict the prognosis of this neoplasia. However, studies have been conducted using the cytological grading system to provide similar information in a faster, less invasive, and more accessible way. This study aimed to investigate cytological graduation and correlate it with histological grading and the survival time of dogs diagnosed with cutaneous mast cell tumors at the Veterinary Hospital of “Universidade Federal de Uberlândia” over five years. For that, cytological and histological slides from 72 animals were reviewed. The statistical methods used were the kappa test for agreement between grading systems, the Kaplan-Meier for survival time, Cox regression for comparison of cytological and histological grades and survival time. The cytological grading when compared to the two-tier histologic grading, high and low grades, had a moderate agreement (kappa 0.566). When the correlation between survival time and the cytological grade was evaluated, there was a higher death rate in the group with high-grade mast cell tumors compared to low grade, pointing to a correlation between survival time and cytological grade (p=0.009). In conclusion, the cytological grade is useful to treatment planning and providing prognostic information that precedes tumor removal, showing a good correlation with the two-tier histologic grading and with the survival time of the animals.

RESUMO: O mastocitoma é uma das neoplasias cutâneas mais comum nos cães e os exames citopatológicos e/ou histopatológicos são utilizados para diagnóstico. A graduação histológica é considerada padrão ouro para prever o prognóstico dessa neoplasia. Contudo, estudos têm sido realizados visando utilizar graduação citológica para fornecer informações semelhantes de maneira rápida, menos invasiva e mais acessível. Esse trabalho objetivou realizar graduação citológica e correlacionar com as graduações histológicas e com a sobrevida de cães diagnosticados com mastocitoma cutâneo no Hospital Veterinário da Universidade Federal de Uberlândia durante 5 anos. Para isso, lâminas de citologia e histologia de mastocitomas de 72 animais foram revisadas. Os métodos estatísticos utilizados foram teste kappa para concordância entre os sistemas de graduação, método Kaplan-Meier para tempo de sobrevida dos animais, e análise pela regressão de Cox para comparação do grau citológico e grau histológico e o tempo de sobrevida global. A graduação citológica quando comparada com a histológica de dois níveis, alto grau e baixo grau, obteve uma concordância moderada (kappa 0,566). Na avaliação da correlação entre sobrevida e grau citológico, houve maior taxa de óbito no grupo de cães com mastocitoma de alto grau, comparado aos de baixo grau, sendo observada correlação entre a sobrevida e o grau citológico (p=0,009). Esse estudo concluiu que o grau citológico é útil para o planejamento do tratamento e para fornecer informações prognósticas que antecedem a exérese do tumor, tendo boa correlação com a graduação histológica de dois níveis e com a sobrevida dos animais.

ABSTRACT: Cats are susceptible to feline panleukopenia virus (FPV) and canine parvovirus type 2 (CPV-2). Therefore, coinfection and superinfection with multiple parvovirus strains may occur, resulting in high heterogeneity and recombination. Considering the importance of cats as a potential source of genetic diversity for parvoviruses, we investigated the frequency of parvovirus infection in cats using their blood and fecal samples and performed molecular characterization of parvovirus strains circulating in cat populations. Accordingly, the fecal and blood samples of 60 cats with gastroenteritis symptoms were collected from Turkey’s Burdur, Isparta, and Izmit provinces. Of these 15 fecal samples tested as parvovirus-positive by PCR, 14 were confirmed to have been infected with true FPV strains by sequencing analysis. Through the phylogeny analysis, those were located in the FPV cluster, closely related to CPV-2, and one was discriminated in the CPV-2b cluster. Additionally, sequence analysis of the VP2 gene of CPV and FPV revealed that the FPV strains detected in Turkey and the vaccine strains were highly related to each other, with a nucleotide identity of 97.7- 100%. Furthermore, 13 variable positions were detected in VP2 of the field and reference FPV strains. Three synonymous mutations were determined in the VP2 gene. Some amino acid mutations in the VP2 protein-affected sites were considered responsible for the virus’s biological and antigenic properties. The partial sequence analysis of the VP2 gene revealed that four FPV strains detected in Turkey have a single nucleotide change from T to G at the amino acid position 384 between the nucleotides 3939-3941, which was reported for the first time. Therefore, these four isolates formed a different branch in the phylogenetic tree. The results suggest that both FPV and CPV-2b strains are circulating in domestic cats in Turkey and cats should be considered as potential sources of new parvovirus variants for cats, dogs and other animals.

RESUMO: Os gatos são suscetíveis ao vírus da panleucopenia felina (FPV) e ao parvovírus canino tipo 2 (CPV-2). Portanto, coinfecção e superinfecção com múltiplas cepas de parvovírus podem ocorrer, resultando em alta heterogeneidade e recombinação. Considerando a importância dos gatos como uma fonte potencial de diversidade genética para parvovírus, investigamos a frequência da infecção por parvovírus em gatos usando suas amostras de sangue e fezes e realizamos a caracterização molecular de cepas de parvovírus circulantes nas populações de gatos. Amostras fecais e de sangue de 60 gatos com sinais de gastroenterite foram coletadas nas províncias de Burdur, Isparta e Izmit, na Turquia. Destas, 15 amostras fecais testaram positivas para parvovírus por PCR e 14 foram confirmadas como infectadas com cepas verdadeiras de FPV por análise de sequenciamento. Através da análise filogenética, aqueles foram localizados no agrupamento FPV que está intimamente relacionado com o CPV-2, e um foi discriminado no agrupamento CPV-2b. Além disso, a análise da sequência do gene VP2 de CPV e FPV revelou que as cepas de FPV detectadas na Turquia e as cepas vacinais eram altamente relacionadas entre si, com uma identidade de nucleotídeos de 97,7-100%. Além disso, 13 posições variáveis foram detectadas em VP2 das cepas de campo e FPV de referência. Três mutações sinônimas foram determinadas no gene VP2. Algumas mutações de aminoácidos nos locais afetados pela proteína VP2 foram consideradas responsáveis pelas propriedades biológicas e antigênicas do vírus. A análise da sequência parcial do gene VP2 revelou que quatro cepas de FPV detectadas na Turquia têm uma única mudança de nucleotídeo de T para G na posição do aminoácido 384 entre os nucleotídeos 3939-3941, o que foi relatado pela primeira vez. Portanto, esses quatro isolados formaram um ramo diferente na árvore filogenética. Os resultados sugerem que ambas as cepas FPV e CPV-2b estão circulando em gatos domésticos na Turquia e os gatos devem ser considerados como fontes potenciais de novas variantes de parvovírus para gatos, cães e outros animais.

ABSTRACT: Canine transmissible venereal tumors (TVT) have a high incidence in Brazil. This is partly due to the large population of stray dogs and the ineffectiveness of epidemiological control programs. This study aimed to describe the epidemiological data, clinical manifestations, and treatments used in dogs affected by TVT. Data were retrospectively collected from the 2015-2020 records of the Veterinary Hospital of the Federal Rural University of Rio de Janeiro. A total of 252 dogs were diagnosed with TVT during the study period. Of these, 81.3% were mixed-breed, 50.4% were males, and 88.9% were young or adult animals. The genital region only was affected in 77.3% of cases. Exclusively extragenital lesions were observed in 22.6% of cases. Among the animals seen, 40.1% received no treatment. Of those treated, 99.3% underwent a vincristine sulfate protocol, and in 77.2%, the treatment resulted in total remission of the neoplasm after 4 to 6 chemotherapy sessions. It was concluded that TVT is a neoplasm most often seen in mixed-breed dogs and located in the genital region, with hemorrhagic secretion being the main clinical sign reported by owners. Vincristine sulfate is currently the most used therapy, with high efficacy. However, despite the good prognosis, there was a high rate of non-adherence or abandonment of treatment, and this is an important factor to be considered and addressed by veterinarians.

RESUMO: O tumor venéreo transmissível (TVT) apresenta elevada incidência no Brasil, relacionada a elevada população de caninos errantes e a ineficácia dos programas de controle epidemiológicos. O objetivo do estudo foi descrever dados epidemiológicos, manifestação clínica e o tratamento empregado em cães acometidos pelo TVT no Hospital Veterinário da Universidade Federal Rural do Rio de Janeiro entre os anos de 2015 e 2020. Foram diagnosticados 252 cães com TVT durante o período do estudo, sendo 81,3% cães sem raça definida, 50,4% machos e 49,6% fêmeas, e com 88,9% animais jovens ou adultos. A região genital foi acometida em 77,3% dos casos. Lesões exclusivamente extragenitais foram observadas em 22,6% dos casos. Quanto ao tratamento, 40,1% dos cães não receberam tratamento. Entre os animais tratados, 99,3% utilizaram protocolo com sulfato de vincristina e em 77,2% o tratamento resultou em remissão total da neoplasia, com a realização de 4 a 6 sessões do quimioterápico. Conclui-se que o TVT é uma neoplasia frequentemente relacionada a cães sem raça definida, localizados na região genital, com secreção hemorrágica sendo o principal sinal clínico reportado pelos proprietários. O sulfato de vincristina é a terapia mais empregada, com alta eficácia. Entretanto, apesar de ser uma neoplasia com bom prognóstico, o alto índice de não adesão ou abandono ao tratamento representa um importante fator a ser considerado e trabalhado pelos médicos veterinários.

ABSTRACT: In this retrospective study, the clinical and anatomopathological findings of fungal diseases in dogs and cats in Northeastern Brazil are described. During the study period, 6,827 histopathological examinations (4,090 necropsies and 2,737 biopsies) of companion animals were reviewed, and fungal infections were diagnosed in 54 cases. Of these, 32 cases were diagnosed in cats and 22 in dogs. Sporotrichosis was the most prevalent mycosis (19/54), followed by dermatophytosis (9/54), cryptococcosis (8/54), candidiasis (7/54), and phaeohyphomycosis (4/54). Other diseases had two cases each, including aspergillosis, mucormycosis, and histoplasmosis, whereas coccidioidomycosis occurred once. Mycoses were diagnosed through microscopic lesions associated with the histomorphological characteristics of the agent, as evidenced by routine and special histochemical stains. They were occasionally confirmed by immunohistochemistry and microbiological cultivation. This retrospective study showed that despite being uncommon, fungal diseases are important causes of death in dogs and cats in the region, with cats being the most affected species and sporotrichosis being the most prevalent mycosis. Clinical signs were correlated with the portal of entry of the infection associated with localized or disseminated superficial and deep skin lesions or affecting parenchymal organs, especially in systemic infections. Microscopic lesions were mainly characterized by pyogranulomatous inflammation associated with fungal structures. In dogs, mycoses occurred mainly owing to an immunological impairment secondary to canine distemper virus co-infection.

RESUMO: Neste estudo retrospectivo, são descritos os achados clínicos e anatomopatológicos das doenças fúngicas em cães e gatos no Nordeste do Brasil. Durante o período de estudo, foram revisados 6.827 exames histopatológicos (4.090 necropsias e 2.737 biópsias) de animais de companhia e diagnosticados 54 casos de infecções fúngicas. Destes, 32 casos foram diagnosticados em gatos e 22 em cães. A esporotricose foi a micose mais prevalente (19/54), seguida pela dermatofitose (9/54), criptococose (8/54), candidíase (7/54) e feo-hifomicose (4/54). Outras doenças tiveram dois casos cada, incluindo aspergilose, mucormicose e histoplasmose, enquanto a coccidioidomicose ocorreu uma vez. As micoses foram diagnosticadas por meio das lesões microscópicas associadas às características histomorfológicas do agente, evidenciadas pelas colorações de rotina e histoquímicas especiais, e ocasionalmente confirmadas pela imuno-histoquímica e cultivo microbiológico. Esse estudo retrospectivo demonstrou que apesar de incomuns, as doenças fúngicas são importantes causas de morte em cães e gatos da região, sendo os gatos a espécie mais afetada e a esporotricose, a micose mais prevalente. Os sinais clínicos estavam correlacionados com a porta de entrada da infecção, associados a lesões cutâneas localizadas ou disseminadas superficiais e profundas ou afetando órgãos parenquimatosos, especialmente em infecções sistêmicas. As lesões microscópicas eram caracterizadas principalmente por inflamação piogranulomatosa associada a estruturas fúngicas. Nos cães, as micoses ocorreram principalmente devido ao comprometimento imunológico secundária a coinfecção pelo vírus da cinomose canina.

ABSTRACT: The molecular background of canine mast cell tumors (MCT) has been extensively investigated; however, the dynamic molecular changes that occur during carcinogenesis and metastasis are not fully understood. This study aimed to evaluate the incidence of mutations in the c-KIT proto-oncogene in canine MCTs and relative draining regional lymph nodes. Suspected or confirmed lymph node metastasis was classified accordingly to the HN Weishaar classification. The study included 34 dogs diagnosed with MCT; 19 patients were enrolled prospectively. These dogs had the primary MCT and regional lymph node resected and analyzed simultaneously. The second group was evaluated retrospectively and included fifteen patients resectioning the primary MCT without evaluation of regional lymph node. Analyzes of c-KIT mutation were performed for all primary MCTs and, in the first group, compared between primary MCT and HN-classified metastasis. Internal tandem duplications (ITD) in exon 11 of the c-KIT gene were detected in 20% of patients. Ten of the nineteen patients (52%) in the first group presented mast cell infiltration in the regional lymph node, and ITD in exon 11 of the c-KIT gene was detected in five and two dogs from Groups 1 and 2, respectively. ITD c-KIT mutations are common in canine MCT and may be found in the draining lymph node metastases/mast cell infiltrates in the absence of mutation of the primary tumor. Evaluation of c-KIT mutation in the primary tumor and metastases may be informative for defining both prognosis and therapeutic options in MCT cases.

RESUMO: O perfil molecular do mastocitoma (MCT) tem sido bastante investigado, no entanto as dinâmicas moleculares que ocorrem durante a carcinogênese e metástase desta neoplasia não estão bem esclarecidas. O objetivo desse estudo foi avaliar a incidência de mutações no proto-oncogene c-KIT em MCTs caninos e respectivos linfonodos regionais. Os casos suspeitos ou confirmados de metástase para os linfonodos, foram classificados de acordo com a classificação HN de Weishaar. O estudo incluiu 34 cães diagnosticados com MCT e, desses, 19 pacientes foram avaliados de maneira prospectiva, em que o tumor primário e o linfonodo regional foram ressecados e analisados simultaneamente. O segundo grupo foi avaliado retrospectivamente e incluiu quinze pacientes que tiveram ressecção do MCT primário sem avaliação de linfonodo regional. A análise da mutação c-KIT foi realizada para todos os MCTs primários e, no primeiro grupo, comparados entre MCT primário e metástase classificada pelo sistema HN. Duplicações internas em tandem (DIT), no exon 11 do gene c-KIT, foram detectadas em um total de 20% dos pacientes. Dez dos dezenove pacientes (52%) do primeiro grupo apresentavam infiltração de mastócitos no linfonodo regional, e DIT no exon 11 do gene c-KIT foram identificadas em cinco e dois cães dos Grupos 1 e 2, respectivamente. Mutações do tipo DIT no gene c-KIT são comuns no MCT canino e podem estar presentes nas metástases/infiltrados de mastócitos na ausência de mutação do tumor primário. A avaliação da mutação no gene c-KIT no tumor primário e metástases pode ser informativa para definir tanto o prognóstico quanto as opções terapêuticas em casos de MCT.

ABSTRACT: Periodontitis affects the teeth supporting tissues, leading to tooth loss and damage to animal health. Evidence in humans suggests that oral microorganisms spread systemically, increasing the risk of pregnancy disorders such as miscarriage, prematurity, and low birth weight. This study aimed to verify whether periodontopathogenic microorganisms reach the transplacental unit, culminating in problems in pregnant ewes. After analyzing the oral cavity, 10 clinically healthy pregnant ewes (OGCH group) and 10 pregnant ewes with periodontitis (OGP group) were selected. The subgingival biofilm was collected for the polymerase chain reaction (PCR) test and amniotic fluid for both the PCR and interleukin (IL) analysis. Peripheral blood was collected for complete blood count, and analyses of IL-6, IL1-β, and tumor necrosis factor-α were performed. Placental fragments were collected to assess the inflammatory changes using optical microscopy. After giving birth, both the ewes and their lambs were weighed. On clinical examination, a positive correlation between bleeding and suppuration (correlation index - CI=0.54), suppuration and marginal gingivitis (CI=0.34), and marginal gingivitis and edema (CI=0.54) was observed. The weights of the ewes (p=0.013) and their respective lambs (p=0.04) in the OGP group were lower than those of their OGCH group counterparts. The hematological analysis revealed that the OGP group ewes showed a slight increase in the mean corpuscular volume (p=0.2447), segmented cells (p=0.3375), and eosinophils (p=0.3823) when compared with the OGCH group ewes, without a statistical difference. Regarding the microorganisms detected in the oral cavity, there was a significant difference between the occurrence of periodontal pockets and the presence of Fusobacterium necrophorum (p=0.0328), Porphyromonas asaccharolytica (p=0.0392), and the Mollicutes class (p=0.0352). Staphylococcus genus (p=0.9107) and Archaea domain (p=0.7245) were detected in the amniotic samples of both groups, without a significant difference, whereas P. asaccharolytica (p=0.2685) was only detected in one sample in the OGCH group. The expression of cytokine IL-6 in the OGP group differed significantly between the prepartum and postpartum periods (p=0.0039); moreover, it differed significantly in the postpartum period between the OGCH and OGP groups (p=0.0198). Histological examination showed a higher percentage of placental changes in the OGP group (70%) than in the OGCH group, such as the presence of macrophages, neutrophils, plasma cells, and multifocal areas of calcification. These results do not corroborate the hypothesis of dissemination of oral microorganisms to the placental unit, suggesting that it constitutes placental isolation in sheep.

RESUMO: A periodontite afeta os tecidos de suporte dos dentes levando à perda dentária e danos à saúde do animal. Evidências em humanos sugerem que os microrganismos orais se espalham sistemicamente, aumentando o risco de distúrbios da gravidez, como aborto espontâneo, prematuridade e baixo peso ao nascer. Este estudo teve como objetivo verificar se microrganismos periodontopatogênicos atingem a unidade transplacentária, culminando em problemas em ovelhas gestantes. Após análise da cavidade oral, foram selecionadas 10 ovelhas gestantes clinicamente saudáveis (grupo OGCH) e 10 ovelhas gestantes com periodontite (grupo OGP). O biofilme subgengival foi coletado para o teste de reação em cadeia da polimerase (PCR) e o líquido amniótico para teste de PCR e análise de interleucina (IL). Sangue periférico foi coletado para hemograma completo e análises de IL-6, IL1-β e fator de necrose tumoral-α foram realizadas. Fragmentos de placenta foram coletados para avaliação das alterações inflamatórias por meio de microscopia óptica. Após o parto, as ovelhas e seus cordeiros foram pesados. Ao exame clínico, observou-se correlação positiva entre sangramento e supuração (índice de correlação - IC=0,54), supuração e gengivite marginal (IC=0,34) e gengivite marginal e edema (IC=0,54). Os pesos das ovelhas (p=0,013) e de seus respectivos cordeiros (p=0,04) do grupo OGP foram inferiores aos do grupo OGCH. A análise hematológica revelou que as ovelhas do grupo OGP apresentaram discreto aumento no volume corpuscular médio (p=0,2447), células segmentadas (p=0,3375) e eosinófilos (p=0,3823) quando comparadas com as ovelhas do grupo OGCH, sem diferença estatística diferença. Em relação aos microrganismos detectados na cavidade oral, houve diferença significativa entre a ocorrência de bolsas periodontais e a presença de Fusobacterium necrophorum (p=0,0328), Porphyromonas asaccharolytica (p=0,0392) e da classe Mollicutes (p=0,0352). O gênero Staphylococcus (p=0,9107) e o domínio Archaea (p=0,7245) foram detectados nas amostras amnióticas de ambos os grupos, sem diferença significativa, enquanto P. asaccharolytica (p=0,2685) foi detectado apenas em uma amostra do grupo OGCH. A expressão da citocina IL-6 no grupo OGP diferiu significativamente entre os períodos pré e pós-parto (p=0,0039); além disso, diferiu significativamente no período pós-parto entre os grupos OGCH e OGP (p=0,0198). O exame histológico mostrou maior porcentagem de alterações placentárias no grupo OGP (70%) do que no grupo OGCH, como a presença de macrófagos, neutrófilos, plasmócitos e áreas multifocais de calcificação. Esses resultados não corroboram a hipótese de disseminação de microrganismos orais para a unidade placentária, sugerindo que se trata de um isolamento placentário em ovinos.

ABSTRACT: Spirochetes in the genus Borrelia (Spirochaetes) are the etiological agents of two zoonotic diseases that have rodents as reservoirs: the Lyme disease, transmitted by ixodid ticks (Ixodidae), and tick-borne relapsing fever, which is transmitted mostly by argasid ticks (Argasidae). The information on the epidemiology of pathogens that affect animals is scarce, especially in places that have been barely studied, such as the Caatinga biome. Thus, in the present study, we aimed to detect DNA of Borrelia in the blood of wild and domestic mammals, as well as in ticks collected in the Catimbau National Park, State of Pernambuco, Northeast Brazil. Blood was collected from 77 captured wild animals belonging to Didelphidae (45), Echimyidae (21), Cricetidae (7), Caviidae (2), and Dasypodidae (2) families. Additionally, blood samples from 120 sheep, 119 goats, 70 dogs, and 47 cattle were obtained. A total of 286 ticks were collected in the environment and identified as Ornithodoros cf. tabajara (188) and Ornithodoros rietcorreai (98); in addition, larvae of Ornithodoros spp. were collected on Thrichomys laurentius (4 specimens) and Monodelphis domestica (1); larvae of O. rietcorreai on Didelphis albiventris (2) and T. laurentius (1); Amblyomma parvum on T. laurentius (4); Rhipicephalus sanguineus (sensu lato) on dogs (121) and Rhipicephalus microplus on cattle (47). All blood samples, 120 free-living ticks, 46 O. rietcorreai and 74 O. cf. tabajara, and 12 R. sanguineus s.l. were submitted to DNA extraction to perform a polymerase chain reaction (nested PCR), aiming at amplifying a fragment of the flaB gene present in all species of Borrelia. Only one free-living specimen of O. cf. tabajara (1.3%) was positive. Negative detections in mammals may be related to the absence of infection or the low concentration of the spirochetes in the hosts. The low prevalence of Borrelia found in ticks can be explained by the low infection rates that these spirochetes depict in their vectors. New studies focusing on the detection of antibodies, as well as on the sequencing and isolating Borrelia spp. should be encouraged for a better understanding of the epidemiology of these bacterial agents in the region.

RESUMO: No gênero Borrelia (Spirochaetes) encontram-se dois grupos de agentes responsáveis por duas doenças zoonóticas, que têm roedores como reservatórios e acometem humanos: o grupo de Lyme, transmitido por carrapatos ixodídeos, e a febre recorrente, que pode ser transmitida por carrapatos argasídeos ou piolhos. Poucas são as informações sobre epidemiologia e agentes patogênicos que acometem animais, podendo representar uma riquíssima fonte de informações, principalmente em locais nunca estudados, pois podem ser bioindicadores de qualidade do ambiente e saúde. Desta forma, no presente estudo foi realizada a pesquisa de Borrelia spp. em amostras de sangue de mamíferos silvestres e domésticos, assim como de carrapatos, do Parque Nacional do Catimbau, Pernambuco. Foi realizada a captura e colheita sanguínea de 77 animais pertencentes às famílias Didelphidae (45), Echimyidae (21), Cricetidae (7), Caviidae (2) e Dasypodidae (2). Adicionalmente, amostras de sangue foram obtidas de 120 ovinos, 119 caprinos, 70 cães e 47 bovinos. Foram coletados 286 carrapatos no ambiente, identificados como Ornithodoros cf. tabajara (188) e Ornithodoros rietcorreai (98); além de carrapatos coletados nos animais: Ornithodoros spp. em Thrichomys laurentius (4 espécimes) e Monodelphis domestica (1); O. rietcorreai em Didelphis albiventris (2) e T. laurentius (1); Amblyomma parvum em T. laurentius (4); Rhipicephalus sanguineus (sensu lato) em cães (121) e R. microplus em bovinos (47). Todas as amostras sanguíneas, 120 carrapatos de vida livre, sendo 46 O. rietcorreai e 74 O. cf. tabajara, e 12 R. sanguineus s. l. foram submetidos à extração de DNA para realização da reação em cadeia da polimerase (nested-PCR), visando amplificação de um fragmento do gene flaB presente em todas as espécies de Borrelia. Não houve amplificação nas amostras sanguíneas dos animais, no entanto em um espécime de O. cf. tabajara de vida livre (1,3%) foi detectado o DNA de Borrelia. A não detecção nos mamíferos pode estar relacionada com a ausência de infecção ou à baixa concentração do agente nos hospedeiros. A baixa prevalência encontrada nos carrapatos pode ser justificada devido à baixa infecção nos vetores. Novos estudos focando na determinação de anticorpos, assim como no sequenciamento e isolamento de Borrelia devem ser estimulados para uma melhor compreensão da epidemiologia do agente na região.

ABSTRACT: We describe an outbreak of leishmaniasis in seven guinea pigs (Cavia porcellus) in which nodular ulcerated skin lesions of varying sizes were observed in the nasal cavity, upper lip, pinnae, vulva, and periarticular region of the limbs. Cytologic exam of collected samples of the lesions in the auricle of one of the animals revealed macrophages containing parasitophorous vacuoles of approximately 4.0μm in diameter in their cytoplasm with morphology suggestive of Leishmania sp. Although skin lesions spontaneously regressed in two of the Guinea pigs, only one survived. All six animals that died were necropsied. Grossly, all animals showed bloody nodular cutaneous lesions with crusts. One of the guinea pigs had distended dark red and firm lungs. Histopathology of the skin lesions revealed histiocytic interstitial acanthotic dermatitis associated with a myriad of Leishmania organisms within macrophages cytoplasm. In the lung, the lesions were characteristic of broncho-interstitial pneumonia with focal infiltrates of neutrophils, epithelioid macrophages, and multinucleated giant cells containing 2µm basophilic amastigotes with morphology compatible with Leishmania spp. A focal granulomatous lesion ,associated with the causal agent in the lung is a novel description of leishmaniasis in guinea pigs caused by L. enriettii. The polymerase chain reaction (PCR) technique with mini-exon primer performed in samples of lesions from two affected guinea pigs was positive and equal to the reference strain, identifying Leishmania enriettii. The cytological, macroscopic, and histological lesions associated with the PCR technique allowed the diagnosis of leishmaniasis and the identification of the specie L. enriettii.

RESUMO: Descrevemos um surto de leishmaniose em sete cobaias (Cavia porcellus), com lesões cutâneas nodulares ulceradas de tamanhos variados observadas na cavidade nasal, lábio superior, pavilhões auriculares, vulva e região periarticular dos membros. No exame citológico foram encontrados macrófagos contendo vacúolos parasitóforos no citoplasma de aproximadamente 4.0μm em diâmetro com morfologia sugestiva de Leishmania sp. Apesar de regressão espontânea das lesões cutâneas terem ocorrido em duas das sete cobaias, apenas um sobreviveu. Seis dos sete animais afetados morreram e foram necropsiados. Macroscopicamente, todos os animais apresentaram lesões cutâneas nodulares, crostosas e sanguinolentas. Uma das cobaias tinha pulmões vermelho-escuros, distendidos e firmes. A histopatologia das lesões cutâneas revelou dermatite acantótica intersticial histiocítica associada a miríades de organismos de Leishmania no citoplasma de macrófagos. Nos pulmões as lesões eram características de pneumonia bronco-intersticial com infiltrado focal de neutrófilos, eosinófilos, macrófagos epitelioides e células gigantes multinucleadas contendo amastigotas basofílicos de 2µm com morfologia compatível com Leishmania spp. Lesões granulomatosas focais associadas ao agente no pulmão são um achado inédito na leishmaniose causada por L. enriettii em cobaias. A técnica de reação em cadeia da polimerase (PCR) com primer mini-exon realizada em amostras de lesões de duas cobaias afetadas foi positiva, identificando Leishmania enriettii. Os aspectos macroscópicos, citológicos, e histológicos associados à técnica da (PCR), permitiram o diagnóstico da leishmaniose e a identificação da espécie L. enriettii.

ABSTRACT: Many inconclusive diagnoses have been reported in studies carried out in veterinary diagnostic laboratories (VDLs). These inconclusive diagnoses are often associated with field veterinarians sending tissue samples and carcasses for histopathological examination. The use of social media in care has the potential as a tool for improvements in animal health. We evaluated the improvement in the rate of conclusive diagnoses in the VDL when using a messaging application for smartphones as a guidance tool for field veterinarians to refer animals and sample tissues for necropsy and histopathology. Veterinarians contacted the VDL team via a messaging app and forwarded videos, images, and clinical-epidemiological history. Based on this information, guidelines were created in real-time for contacting professionals about methods for collecting samples during necropsy and making diagnoses and scheduling visits to properties. The data obtained showed that smartphone messaging applications could be useful tools to expand the performance of VDLs and improve their diagnostic efficiency, especially when sending samples of animals that died is impossible due to certain sanitary conditions.

RESUMO: Muitos diagnósticos inconclusivos têm sido relatados em estudos realizados em laboratórios de diagnóstico veterinário (LDVs). Esses diagnósticos inconclusivos são frequentemente associados a veterinários de campo que enviam amostras de tecidos e carcaças para exame histopatológico. O uso das mídias sociais com cuidado tem potencial como ferramenta para melhorias na saúde animal. Avaliamos a melhora na taxa de diagnósticos conclusivos no LDV ao utilizar um aplicativo de mensagens para smartphones como ferramenta de orientação para veterinários de campo encaminharem animais e amostras de tecidos para necropsia e histopatologia. Os veterinários entraram em contato com a equipe do L por meio de um aplicativo de mensagens e encaminharam vídeos, imagens e histórico clínico-epidemiológico. Com base nessas informações, foram criadas orientações em tempo real para contato dos profissionais sobre métodos de coleta de amostras durante a necropsia e realização de diagnósticos, além do agendamento de visitas às propriedades. Os dados obtidos mostraram que os aplicativos de mensagens para smartphones podem ser ferramentas úteis para ampliar o desempenho dos VDLs e melhorar sua eficiência diagnóstica, principalmente quando o envio de amostras de animais que morreram é impossível devido a determinadas condições sanitárias.
