The aerospace industry is constantly looking for innovative materials that exhibit good mechanical and corrosion properties. The 2198-T851 (Al-Cu-Li) alloy was developed to replace the conventional Al-Cu-Mg in aircraft structures. Despite the usefulness of the 2198-T851 alloy, its performance may be affected when subjected to an aggressive medium containing chloride ions. The deposition of Nb2O5 coatings by using the reactive sputtering technique on the 2198-T851 alloy surface appears as a powerful tool to improve the corrosion resistance of this material. Recently, groundbreaking research findings have demonstrated the positive effect of Nb2O5 coatings on corrosion protection of alloy 2198-T851. However, the corrosion products originated from the 2198-T851 aluminium alloy are poorly understood. The use of Raman spectroscopy and XPS techniques may help to shed some light on the corrosion products of 2198-T851 alloy. Results demonstrated the corrosion products are mainly composed by CuCl2 x H2O, CuCl, Cu2Cl(OH)3, Al(OH)3, and AlO(OH).

This investigation describes the synthesis of eugenol analogs presenting 1,2,3-triazole fragments and evaluation of their antileishmanial activity. The alkylation of guaiacol (1) with allyl bromide afforded 1-(allyloxy)-2-methoxybenzene (2) (93% yield). The Claisen rearrangement conducted with 1 gave ortho eugenol (3) (82% yield). Alkylation procedures performed with 3 produced 1-allyl-3-methoxy-2-(prop-2-yn-1-yloxy)benzene (4) (73% yield) and 1-allyl-3-methoxy-2-(pent-4-yn-1-yloxy)benzene (6) (53% yield). The copper(I)-catalyzed alkyne-azide cycloaddition (CuAAC) reactions involving alkynes 4 and 6 with different benzylic azides afforded twenty-two eugenol analogs with 1,2,3-triazole functionalities (48-93% yield). We screened the compounds at 10 μmol L 1 against Leishmania braziliensis intracellular amastigotes during macrophage infection. The action of these compounds was compared with the known leishmanicidal drug amphotericin B. None of the analogs were toxic to macrophages at 10 μmol L-1. The cytotoxic concentration at 50% (CC50), effective concentration at 50% (EC50), and selectivity index (SI) were determined to the best compounds 4-((2-allyl-6-methoxy)phenoxymethyl)-1-(4-chlorobenzyl)-1 H-1,2,3-triazole (8c) and 4-((2-allyl-6-methoxy)phenoxymethyl)-1-(4-trifluoromethoxybenzyl)-1 H-1,2,3-triazole (8h). They showed a significant leishmanicidal effect, with EC50 of 28.09 µmol L-1 (8c) and 52.03 µmol L-1 (8h). The SIs were 9.7 for 8c and > 5.7 for 8h. These compounds have the potential as new leishmanicidal agents against L. braziliensis and may represent a starting point for the development of alternative treatments for cutaneous leishmaniasis.

A exposição excessiva à radiação solar (RS) é o principal causador do câncer de pele. O objetivo desse trabalho foi investigar a percepção de estudantes do ensino médio sobre RS e suas implicações à saúde humana. Os dados foram coletados através de questionários e as respostas foram avaliadas por tabelas de frequências relativa, absoluta e percentual. Devido ao tamanho da amostra, utilizamos o teste de Fisher a 5% de significância (e hipótese alternativa bilateral) para correlacionar as respostas. Os alunos, independente de terem estudado ou não sobre RS não sabem definir RS e não têm percepção sobre os efeitos da RS à saúde humana. Este diagnóstico mostra, por um lado, a carência conceitual dos estudantes. Por outro, ele sugere também a ineficácia dos recursos didáticos convencionais e meios de informação acessíveis. Portanto, a inserção deste conceito na educação básica é primordial dada sua relevância social e por se tratar de um problema de saúde pública.
The excessive exposure to solar radiation (SR) is the main cause of skin cancer. The main aim of this paper was to investigate the perception of high school students about the SR and its implications for human health. The data have been collected through questions and the answers were evaluated by tables of relative, absolute and percentual frequency. Due to the size of our sample, we have used the Fischer test at 5% of significance (and bilateral alternative hypothesis) to correlate the answers. The students, whether they studied or not SR, don’t know to define SR and don’t have perception about its effects on the human health. This diagnosis shows, on the one hand, the conceptual lack of students. On the other, it also suggests the ineffectiveness of conventional didactic resources and accessible information media. Thus, the insertion of this concept in basic education is essential due to its social relevance and because it’s a public health problem.

Resumo Pouco se sabe se a morte celular induzida pela terapia fotodinâmica (PDT) compromete os macrófagos (MΦ), envolvidos nas respostas imunes induzidas pela PDT. Neste estudo, foram avaliados parâmetros de citotoxicidade dos MΦ mediada pela PDT e a produção de citocinas, frente à protoporfirina IX (PpIX). MΦ peritoneais de camundongos BALB/c foram estimulados in vitro com PDT, luz, PpIX ou lipopolissacarídeo (LPS). Após isto, a viabilidade celular (VC), a lipoperoxidação, os níveis de óxido nítrico (NO), de DNA degradado, de TNF-α, IL-6 e IL-10 foram avaliados. A exposição curta à PDT reduziu a VC em 10-30%. Os níveis de NO e de DNA degradado duplicaram, sem aumento da lipoperoxidação. Houve aumento de TNF-α e IL-10, sendo maior na presença de LPS. Já a produção de IL-6 reduziu em dez vezes. A PDT induz estresse celular, gera radicais NO e causa dano ao DNA, tornando o microambiente citotóxico. Ainda, modula citocinas pró e anti-inflamatórias em MΦ.
Abstract Little is known regarding whether photodynamic therapy (PDT)-induced cell death can substantially compromise macrophages (MΦ), which are important cells in PDT-induced immune responses. Here, parameters of PDT-mediated MΦ cytotoxicity and cytokine production in response to protoporphyrin IX (PpIX) were evaluated. Peritoneal MΦ from BALB/c mice were stimulated in vitro with PDT, light, PpIX, or lipopolysaccharide (LPS). After that, cell viability, lipid peroxidation, Nitric Oxide (NO), DNA damage, TNF-α, IL-6 and IL-10 were evaluated. Short PDT exposure reduced cell viability by 10–30%. There was a two-fold increase in NO and DNA degradation, despite the non-increase in lipoperoxidation. PDT increased TNF-α and IL-10, particularly in the presence of LPS, and decreased the production of IL-6 to 10-fold. PDT causes cellular stress, induces NO radicals and leads to DNA degradation, generating a cytotoxic microenvironment. Furthermore, PDT modulates pro- and anti-inflammatory cytokines in MΦ.

Experimental analysis of eccentrically loaded reinforced concrete columns with an added jacket of self-compacting concrete

Resumo O presente trabalho apresenta o estudo experimental de pilares de concreto armado reforçados com parafusos chumbadores e acréscimo de camada com 35 mm de concreto autoadensável na face comprimida. Nove pilares, com alturas de 2000 mm, foram submetidos à flexão composta reta com excentricidade inicial de 60 mm, assim discriminados: um pilar original (PO) com seção transversal de 120 mm x 250 mm; um pilar de referência (PREF) com seção transversal de 155 mm x 250 mm e sete pilares, inicialmente moldados com seção transversal de 120 mm x 250 mm, e após, reforçados através da adição de camada de reforço de 35 mm e diversas configurações de posições de parafusos chumbadores na face comprimida. Exceto o pilar original (PO), os pilares foram submetidos à excentricidade inicial de carga de 42,5 mm, devido ao acréscimo da camada de reforço na face comprimida. Apesar de não ocorrer ruptura dos parafusos chumbadores, não foi possível evitar o desplacamento da camada de reforço. Os resultados obtidos indicam que é possível a reabilitação estrutural de pilares de concreto armado com o uso da metodologia de reforço empregada, obtendo ganho de capacidade de carga média de 271 % em relação ao pilar original (PO).
Abstract This paper presents the experimental study of eccentrically loaded reinforced concrete columns with an added 35 mm self-compacting concrete jacket attached to the column’s most compressed face using wedge bolts. Nine columns with a 2000 mm height were tested under compression and one-way bending until failure. Columns were denominated as original column (PO) with a cross section of 120 mm x 250 mm; reference column (PR) with a cross section of 155 mm x 250 mm, and seven columns with an initial cross section of 120 mm x 250 mm and later reinforced by the addition of 35 mm self-compacting concrete layer and various configurations of wedge bolts. Except for the original column PO, the columns were submitted to a 42.5 mm load eccentricity due to the added concrete layer at the compressed face. Although failure of the wedge bolts did not occur, it was not possible to prevent detachment of the added layer. The results indicate that it is possible to structurally rehabilitate reinforce concrete columns with the use of the strengthening methodology used in this research, resulting in average ultimate load capacity gains of 271% compared to original column’s ultimate load.

Decolorization of textile industry wastewater in solid state fermentation with Peach-Palm (Bactris gasipaes) residue

Resumo Neste trabalho, avaliamos a descoloração de efluentes têxteis durante seu tratamento em um sistema de fermentação em estado sólido (SSF). Os ensaios foram conduzidos com resíduo de pupunha (Bactris gasipaes) utilizando o fungo de podridão branca Ganoderma lucidum EF 31. A influência da concentração de corante, as quantidades de resíduo e da fase líquida foram estudadas tanto na eficiência de descoloração como na produção de enzima. De acordo com os resultados, independentemente das condições experimentais utilizadas, a lacase foi a principal enzima ligninolítica produzida por G. lucidum. A atividade de lacase mais elevada foi obtida em baixas concentrações de efluentes, sugerindo um efeito inibitório no metabolismo fúngico. A maior remoção de cor foi obtida com 10 gramas de resíduo da pupunha e 60 mL do efluente final. Nos ensaios de controle realizados com o corante sintético RBBR, foram atingidos cerca de 20% mais descoloração do que os obtidos com o efluente industrial. A adsorção de RBBR no resíduo de pupunha também foi investigada. Os testes de equilíbrio mostraram que a adsorção deste corante seguiu as isotermas de Langmuir e Freundlich. Assim, os resultados experimentais indicam que o resíduo de pupunha é um substrato adequado tanto para a produção de lacase quanto para a remoção de cor em efluentes industriais.
Abstract In this work we have assessed the decolorization of textile effluents throughout their treatment in a solid-state fermentation (SSF) system. SSF assays were conducted with peach-palm (Bactris gasipaes) residue using the white rot fungus Ganoderma lucidum EF 31. The influence of the dye concentration and of the amounts of peach-palm residue and liquid phase on both the discoloration efficiency and enzyme production was studied. According to our results, independently of experimental conditions employed, laccase was the main ligninolytic enzyme produced by G. lucidum. The highest laccase activity was obtained at very low effluent concentrations, suggesting the existence of an inhibitory effect of higher concentrations on fungal metabolism. The highest percentage of color removal was reached when 10 grams of peach palm residue was moistened with 60 mL of the final effluent. In control tests carried out with the synthetic dye Remazol Brilliant Blue R (RBBR) decolorization efficiencies about 20% higher than that achieved with the industrial effluent were achieved. The adsorption of RBBR on peach-palm residue was also investigated. Equilibrium tests showed that the adsorption of this dye followed both Langmuir and Freundlich isotherms. Hence, our experimental results indicate that peach-palm residue is suitable substrate for both laccase production and color removal in industrial effluents.

Integrated furfural and first generation bioethanol production: process simulation and techno-economic analysis

Abstract Furfural is a base chemical with a wide range of applications and with a great opportunity for market growth in the near term. Derived from biomass, its production may be incorporated to the Brazilian chemical industry using sugarcane bagasse as feedstock. In this context, the integration of a furfural plant to a first generation bioethanol facility, within the biorefinery concept, was simulated considering different scenarios compared to an autonomous bioethanol distillery. The economic analysis of the different scenarios showed that the revenues from furfural commercialization increase the internal rate of return of the project for maximum furfural production (22.0%) in comparison to a conventional ethanol distillery (13.5%), despite the decrease in electricity output. Moreover, the economic analysis of the results pointed out the possibility of lowering furfural prices to levels that could lead to its use as a precursor for biofuels.

Environmental indicators in effluent assessment of rainbow trout (Oncorhynchus mykiss) reared in raceway system through phosphorus and nitrogen

Resumo A descarga de fósforo e nitrogênio via efluente do sistema intensivo de truticultura foi quantificada através da utilização de indicadores ambientais. As cargas de nutrientes, o balanço de massa, a quantidade estimada de nutrientes na ração e a quantidade de nutrientes convertidos em biomassa de peixes foram calculados com base nas concentrações de fósforo (P) e nitrogênio (N) na ração e na água. Da ração oferecida, 24,75 kg estavam disponíveis como P e 99,00 kg como N, destes, 9,32 kg de P (38%) e 29,12 kg de N (25%) foram convertidos em biomassa de peixe e 15,43 kg P (62%) e 69,88 kg N (75%) foram exportados via efluente. As cargas e o balanço de massa mostram a descarga excessiva de nutrientes via efluente, corroborado pela taxa de conversão alimentar (2,12:1), devido à baixa eficiência na utilização da ração, portanto, propõe-se a utilização deste parâmetro zootécnico como indicador ambiental. Além disso, as práticas de manejo alimentar não são adequadas, destacando a baixa frequência de alimentação durante o dia, quantidade excessiva e baixa qualidade da alimentação ofertada. Esses resultados demonstram a necessidade de manejo alimentar adequado e de monitoramento do efluente.
Abstract The phosphorus and nitrogen discharge via effluent of intensive trout farming system was quantified through the use of environmental indicators. The nutrient loads, the mass balance, the estimated amount of nutrients in feed and the amount of nutrients converted in fish biomass were calculated based on the concentrations of phosphorus (P) and nitrogen (N) in the feed and in the water. Of the offered feed, 24.75 kg were available as P and 99.00 kg as N, of these, 9.32 kg P (38%) and 29.12 kg N (25%) were converted into fish biomass and 15.43 kg P (62%) and 69.88 kg N (75%) were exported via effluent. The loads and the mass balance show the excessive discharge of nutrients via effluent, corroborated by the feed conversion ratio (2.12:1) due to the low efficiency of feed utilization, therefore, it is proposed the use of this zootechnical parameter as environmental indicator. In addition, feed management practices are not adequate, highlighting the low frequency of feeding during the day, excessive amount and low quality of feed offered. These results demonstrate the need for adequate feed management and the need for careful monitoring of effluent.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Produtividade de duas cultivares de mamona submetidas a diferentes lâminas de irrigação suplementar

Objetivou-se com este trabalho avaliar a produtividade de grãos, os componentes da produção e a quantidade de óleo produzida por duas cultivares de mamona, irrigadas por gotejamento e submetidas a diferentes lâminas de água. O experimento foi conduzido no ano agrícola de 2009, em um Latossolo Vermelho distroférrico, textura argilosa, em área experimental no município de Dourados, MS. O delineamento experimental foi em blocos casualizados, no esquema fatorial, com cinco lâminas de água (0, 25, 50, 100 e 150% da evapotranspiração para irrigação localizada) em duas cultivares de mamona (IAC 2028 e IAC 80), com quatro repetições. O turno de rega foi predeterminado em duas irrigações semanais, excetuando-se os dias em que ocorreu precipitação pluviométrica. O aumento das lâminas de irrigação proporcionou incremento significativo da maioria dos componentes da produção e da produtividade da cultura sem, no entanto, alterar o teor de óleo contido nas sementes. A aplicação da maior lâmina de água aumentou em 80% a produtividade em relação ao tratamento que não recebeu irrigação suplementar.
This study evaluated the yield, components of production and oil content of two castor bean cultivars through drip irrigation with different water depths. The research was conducted in 2009 in an Oxisol clay in the experimental field in Dourados, Mato Grosso do Sul State. The experimental design was randomized blocks in factorial scheme with five water depths (0, 25, 50, 100 and 150% of evapotranspiration for drip irrigation) in two castor bean cultivars (IAC 2028 and IAC 80) with four replications. The irrigation schedule was predetermined up to two irrigations per week except on rainy days. The increase of irrigation provided significant increase in most components of production and crop yield without changing the oil content of seeds. The application of higher water depth increased yield by 80% in relation to the treatment that received no supplemental irrigation.

OBJETIVO: Investigar mudanças longitudinais no uso da extremidade superior em três crianças com hemiplegia submetidas a um protocolo adaptado de terapia de movimento induzido por restrição (CIMT). MÉTODOS: Um desenho experimental de caso único (ABA) foi replicado em três crianças entre 8 e 11 anos de idade. Fases de baseline (A1) e (A2) e fase de intervenção (B) duraram duas semanas cada. Durante a fase de intervenção, as crianças usaram um splint na extremidade não afetada por dez horas por dia e foram submetidas a três horas de terapia diária por dez dias. O treinamento consistiu em atividades para a extremidade superior acometida, com aumento gradual da complexidade da tarefa e reforço verbal. A função manual foi classificada de acordo com o Manual Ability Classification System (MACS). As crianças foram avaliadas quatro vezes por semana com o Toddler Arm Use Test (TAUT) e três provas adaptadas do Teste Jebsen-Taylor de Função Manual (JTHF) e uma vez por semana com o Pediatric Motor Activity Log (PMAL) e escalas de autocuidado do Inventário de Avaliação Pediátrica de Incapacidade (PEDI). Os métodos Celeration Line, Banda de Dois Desvios-Padrão e análise visual foram usados para análise de dados. RESULTADOS: Resultados significativos demonstraram melhora na qualidade e frequência de uso da extremidade superior (PMAL), qualidade de uso do TAUT nas crianças 2 e 3, participação na criança 1, bem como diminuição do tempo gasto para completar as tarefas do JTHF para as crianças 2 e 3. Nenhuma mudança foi observada nas escalas de autocuidado do teste PEDI. CONCLUSÃO: Efeitos da CIMT associaram-se à melhora na destreza manual, frequência e qualidade de uso da extremidade acometida em crianças com hemiplegia.
OBJECTIVE: To investigate the profile of changes in the use of the upper extremity in three children with hemiplegia submitted to an adapted protocol of constraint-induced movement therapy (CIMT). METHODS: A single-subject design (ABA) was replicated in three children aged 8 to 11 years old. Baseline phases (A1) and (A2) and the intervention phase (B) lasted 2 weeks each. During the intervention period, children wore a splint on the non-affected extremity for 10 hours a day and were submitted to 3 hours of therapy a day during 10 days. Training consisted of activities with the affected upper extremity, with gradually increasing complexity and verbal feedback. Hand function was classified according to the Manual Ability Classification System (MACS). Children were assessed four times every week with the Toddler Arm Use Test (TAUT) and three adapted tasks from the Jebsen-Taylor Hand Function test (JTHF), and once a week with the Pediatric Motor Activity Log (PMAL) and self-care scales of the Pediatric Evaluation of Disability Inventory (PEDI). Celeration Line, Two-Standard Deviation Band and visual analysis methods were used for data analyses. RESULTS: Significant improvements in the amount and quality of upper extremity use (PMAL), TAUT quality of use for children 2 and 3, and participation for child 1, as well as decreased time to complete JTHF tasks for children 2 and 3 were observed. No changes were observed in the PEDI self-care scales. CONCLUSION: CIMT effects were associated with improvements in manual dexterity, amount and quality of use of the affected upper extremity in children with hemiplegia.

Contextualização: A imobilização tem sido amplamente utilizada para recuperação de lesões musculoesqueléticas, entretanto essa técnica causa déficits funcionais no sistema musculoesquelético. Existe uma série de evidências demonstrando redução da força devido à hipotrofia muscular e um incremento do percentual de fibras rápidas, embora as contribuições relativas à lesão ou à imobilização ainda não estejam totalmente esclarecidas. Objetivo: Verificar a influência da imobilização do cotovelo na relação torque-ângulo e na ativação elétrica dos flexores e extensores em sujeitos saudáveis. Métodos: Dezoito sujeitos do sexo masculino (22-42 anos) foram divididos em um grupo controle (n=11) e em um grupo imobilizado (n=7). Todos os sujeitos realizaram os mesmos testes duas vezes, com intervalo de 14 dias. O grupo imobilizado teve seu cotovelo não-dominante imobilizado com uma tala gessada no ângulo de 90º durante 14 dias. O torque máximo isométrico foi obtido nos ângulos de 150°, 120°, 90° e 60°. Resultados: Houve redução de 16% no torque máximo após a imobilização, o que não pode ser explicado por alterações na medida de perimetria ou na ativação elétrica muscular. Conclusões: A imobilização de 14 dias produziu uma redução no torque máximo isométrico do cotovelo, porém não alterou a ativação elétrica em sujeitos saudáveis. Essa redução parece estar relacionada à hipotrofia muscular e, provavelmente, menos intensa quando comparada à imobilização após lesões musculoesqueléticas.
BACKGROUND: Limb immobilization has been extensively used during the recovery process of musculoskeletal injuries despite the fact that this technique causes functional deficits in the skeletal muscle. There is evidence showing strength reduction due to muscle hypotrophy and an increase in the percentage of fast-twitch fibers, however it is still unclear how the injuries and the immobilization contribute to these changes. OBJECTIVE: To verify the influence of elbow joint immobilization on the torque-angle relationship and on electrical activation of the elbow flexor and extensor muscles in healthy subjects. METHODS: Eighteen male subjects (22 to 42 years) were assigned to either a control group (n=11) or to an immobilization group (n=7). All subjects performed the same tests twice with a 14-day interval period. The immobilization group had the non-dominant elbow joint immobilized with a cast at a joint angle of 90º during 14 days. Maximal isometric torques were obtained with the elbow joint positioned at 150º, 120º, 90º and 60º. RESULTS: There was a 16% reduction in maximal torque after immobilization that cannot be explained by changes in arm girth or by reduction in the electrical activation of the muscles. CONCLUSIONS: Fourteen days of immobilization produced a reduction in maximal elbow isometric torque but did not change muscle activation in healthy subjects. This reduction in maximal torque production seems to be related to muscle hypotrophy and is probably less pronounced when compared to that from immobilization after musculoskeletal injuries.

OBJETIVO: Estudar a imunogenicidade e a estabilidade do surfactante de origem porcina produzido pelo Instituto Butantan. MÉTODO: Experimento imunogenicidade: 16 coelhos da raça New-Zealand-White (Peso de 1000g) foram divididos em grupos de 4 animais. Cada grupo foi designado para receber: a) Surfactante do Butantan, b) Survanta® (Abbott Laboratories), c) Curosurf (Farmalab Chiesi) e d) nenhum tratamento com surfactante. Os surfactantes foram administrados via intratraqueal e o sangue dos animais foi coletado antes, 60 e 180 dias após a administração do surfactante. O soro obtido foi analisado quanto a presença de anticorpos anti-surfactante pelo método ELISA (enzyme-linked immunosorbent assay). Experimento estabilidade: O surfactante do Butantan usado neste experimento tinha sido armazenado por um ano em refrigerador (4 a 8°C) e sua estabilidade foi analisada em condições distintas de experimentação, usando o modelo de coelho prematuro. RESULTADOS: Experimento imunogenicidade: Nenhum dos surfactantes analisados determinou a produção de anticorpos contra seus constituintes. Experimento estabilidade: Os resultados deste estudo demonstraram que o surfactante do Instituto Butantan mostrou eficácia semelhante a do Curosurf após ter sido submetido à condições adversas ao longo do tempo. A eficácia foi demonstrada através da complacência pulmonar dinâmica, pressão ventilatória e da curva pressão-volume. CONCLUSÃO: Os resultados deste estudo demonstraram que o surfactante do Instituto Butantan pode representar um tratamento alternativo de reposição de surfactante.
PURPOSE: To study the immunogenicity and the stability of the porcine pulmonary surfactant preparation produced by the Instituto Butantan. METHOD: Immunogenicity assay: Sixteen New-Zealand-White rabbits (1000 g body weight) were divided into 4 study groups. Each group was assigned to receive either a) Butantan surfactant, b) Survanta® (Abbott Laboratories), c) Curosurf® (Farmalab Chiesi), or d) no surfactant. The surfactants were administered intratracheally, and the animals were collected immediately before and 60 and 180 days after surfactant administration. Sera were assayed for the presence of antisurfactant antibodies by enzyme-linked immunosorbent assay (ELISA). Stability assay: The Butantan surfactant used in this assay had been stored for one year in the refrigerator (4 to 8ºC) and its stability was evaluated in distinct assay conditions using a premature rabbit model. RESULTS: Immunogenicity assay: None of the surfactants analyzed triggered antibody immune responses against their components in any of the animals. Stability assay: The results of this study demonstrate that Butantan surfactant was as effective as Curosurf when both were submitted to the adverse circumstance of short- and long-term storage at room temperature. A similar level of efficacy for the Butantan surfactant, as compared to Curosurf was demonstrated by the pulmonary dynamic compliance, ventilatory pressure, and pressure-volume curve results. CONCLUSION: The results of our study demonstrate that Butantan surfactant may be a suitable alternative for surfactant replacement therapy.

Effects of corticosteroids in very low birth weight newborns dependent on mechanical ventilation

Devido às suas ações anti-inflamatórias, os corticosteróides têm sido utilizados para prevenção de displasia broncopulmonar, sendo descrita, uma redução da incidência desta patologia. No entanto, efeitos adversos a curto e a longo prazo têm sido detectados, em recém-nascidos pré-termo. OBJETIVO: Analisar os efeitos sobre a incidência de displasia broncopulmonar, duração de ventilação mecânica e de internação, mortalidade, crescimento, além dos efeitos adversos dos corticosteróides, administrados entre 10-14 dias de vida, em recém-nascidos de muito baixo peso, dependentes de ventilação mecânica. MÉTODOS: Realizou-se estudo de coorte, incluindo-se todos os recém-nascidos com peso de nascimento < 1500 gramas dependentes de ventilação mecânica, entre 10-14 dias de vida. Foram divididos em: Grupo I - receberam dexametasona (16) e Grupo II - não receberam dexametasona (22). Administrou-se dexametasona, a partir do 10º dia de vida, dias 1 a 3 - 0,3 mg/kg/d, dias 4 a 6 - 0,2 mg/kg/d, dias 7 a 9 - 0,1 mg/kg/d. Analisou-se o desenvolvimento de displasia broncopulmonar (dependência de oxigênio aos 28 dias de vida), efeitos sobre a evolução respiratória e sobre o padrão de crescimento, além da ocorrência de efeitos adversos. RESULTADOS: A incidência de displasia broncopulmonar não diferiu entre os grupos (GI - 62,5%; GII - 22,7%;p = 0,07). Detectou-se desaceleração do crescimento no GI em relação ao GII(D P = 47g/semana, GI e 85,5g/semana, GII; p = 0,06; D PC - 0,75 cm/semana GI e 1cm/semana, no GII; p = 0,05). CONCLUSÃO: O uso de corticosteróides, em recém-nascidos pré-termo, entre 10 - 14 dias de vida não reduziu incidência de displasia broncopulmonar e causou uma desaceleração do crescimento.
Corticosteroids have been used in bronchopulmonary dysplasia prevention because of their antiinflammatory effects. Among their effects is a decrease in the incidence of bronchopulmonary dysplasia. However, short- and long-term side effects have been detected in preterm newborns. PURPOSE: To analyze the effects of corticosteroids on bronchopulmonary dysplasia, length of stay, mortality, growth, as well as the adverse effects in very low birth weight newborns between 10 and 14 days of life and dependent on mechanical ventilation. METHODS: Cohort study. All newborns with a birth weight under 1500 g, mechanical ventilation-dependent between 10 and 14 days of life, during the period January 2000 and June 2001 were included (n = 38). They were divided into 2 groups: Group I with corticosteroids (n = 16) and Group II without corticosteroids (n = 22). Dexamethasone administration: from the 10th day of life, d1 - d3, 0.3 mg/kg/d; d4 - d6, 0.2 mg/kg/d; d7 - d9, 0.1 mg/kg/d. Respiratory evolution, bronchopulmonary dysplasia (oxygen dependence at 28 days of life), growth pattern and the presence of adverse effects were analyzed. RESULTS: The incidence of bronchopulmonary dysplasia was 6.5% (Group I) and 30% (Group II), P = .07. A decrease in growth was detected in Group I compared with Group II (change in weight: Group I - 47 g/week, Group II - 85.5 g/week, P = .06; change in head circumference: Group I - 0.75 cm/week, Group II - 1 cm/week, P = .05). CONCLUSION: Use of corticosteroids in very low birth weight infants dependent on mechanical ventilation during the first 10 to 14 days of life did not affect the respiratory evolution and occurrence of bronchopulmonary dysplasia, but the velocity of growth was reduced.

A small-mass system has been developed for monitoring the flux of neutrons with energy up to 1 MeV at the new time-of-flight facility at CERN, n_TOF. The monitor is based on a thin Mylar foil with a 6Li deposit, placed in the neutron beam, and an array of Silicon detectors, placed outside the beam, for detecting the products of the 6Li(n, alpha)³H reaction. The small amount of material on the beam ensures a minimal perturbation of the flux and minimizes the background related to scattered neutrons. Moreover, a further reduction of the gamma-ray background has been obtained by constructing the scattering chamber hosting the device in carbon fibre. A detailed description of the flux monitor is here presented, together with the characteristics of the device, in terms of efficiency, resolution and induced background. The use of the monitor in the measurement of neutron capture cross-sections at n_TOF is discussed
