GATA2 variants in patients with non-tuberculous mycobacterial or fungal infections without known immunodeficiencies

ABSTRACT Introduction Haploinsufficiency of the hematopoietic transcription factor GATA2 is associated with a broad spectrum of diseases, including infection susceptibility and neoplasms. We aimed to investigate GATA2 variants in patients with non-tuberculous mycobacterial (NTM) and/or fungal infections (FI) without known immunodeficiencies. Method We performed GATA2 genotyping in patients with NTM and/or FI. Results Twenty-two patients were enrolled (seventeen FI, four NTM and one with both infections). The pathogenic variant NG_029334.1:g.16287C>T was found in one patient (4.5%) and two asymptomatic offsprings. We also found the likely-benign variant NG_029334.1:g.12080G>A (rs2335052), the benign variant NG_029334.1:g.16225C>T (rs11708606) and the variant of uncertain significance NG_029334.1:g.16201G>A (rs369850507) in 18.2%, 27.3%, and 4.5% of the cases, respectively. Malignant diseases were additionally diagnosed in six patients. Conclusion Although detected in 45.4% of the patients, most GATA2 variants were benign or likely benign. Identifying a pathogenic variant was essential for driving both the patient's treatment and familial counseling. Pathogenic variants carriers should receive genetic counseling, subsequent infection prevention measures and malignancies surveillance. Additionally, case-control genotyping should be carried out in Brazil to investigate whether the observed variants may be associated with susceptibility to opportunistic infections and/or concurrent neoplasms.

Considering the promising antitumor effects of compounds with dual anti-inflammatory and antiproliferative activities, thus benzophenones analogs (2-7) were evaluated on in vivo anti-inflammatory assay and molecular docking analysis. Those with the best molecular docking results were in vitro evaluated on cyclooxygenase (COX) enzymes and tested regarding antiproliferative activity. All derivatives displayed in vivo anti-inflammatory activity. Among them, the substances 2’-hydroxy-4’-benzoylphenyl-β-D-glucopyranoside (4), 4-hydroxy-4’-methoxybenzophenone (5) and 4’-(4’’-methoxybenzoyl)phenyl-β-D-glucopyranoside (7) showed the best values of Glide Score in COX-2 docking evaluation and 4 and 5 selectively inhibited COX-2 and COX-1 in vitro enzymatic assay, respectively. Thus, 4 and 5 were tested against breast cancer (MCF-7, MDA MB-231, Hs578T) and non-small-cell-lung cancer (A549) cell lines. The estrogen-positive MCF-7 cell line was more responsive compared to other tested cell lines. They induced cell cycle arrest at G1/S transition in MCF-7 cell line once there was an increase in G0/G1 population with concomitant reduction of S population. The antiproliferative activity of these substances on MCF-7 was associated with their ability to inhibit cyclin E expression, a critical regulator of G1/S transition. Taken together, the data indicate that 4 and 5 have dual anti-inflammatory and antiproliferative activities and support further studies to evaluate their antitumor potential.

Meliaceae representatives are economically important in several aspects including the production of highly prized woods (mahogany, cedar, etc.), constituents for cosmetics, and insecticides. The present study aimed to verify the chemical composition as well as leishmanicidal and cytotoxic potential of essential oils from leaves of two different populations of Guarea macrophylla collected at cities of São Paulo (population I) and Cubatão (population II), São Paulo State, Brazil. Chemically, the oils showed the predominance of sesquiterpenes: cis-β-guaiene, bicyclogermacrene, viridiflorol, and isolongifolan-7α-ol from population I and α-copaene, E-caryophyllene, cis-β-guaiene, and γ-amorphene from population II. In vitro antileishmanial activity against promastigote forms of Leishmania (L.) amazonensis of essential oils was evaluated and displayed 50% effective concentration (EC50) values ranging from 11.8 to 20.5 µg mL-1. Furthermore, toxicity against peritoneal macrophages of BALB/c mice was observed, with 50% cytotoxic concentration (CC50) ranging from 17.7 to > 100 µg mL-1. Multivariate statistical analysis revealed the influence of each constituent of the oils against L. amazonensis being 1,10-di-epi-cubenol, α-amorphene, E-caryophyllene, isopimara-7,15-diene, and β-elemene associated with the antileishmanial potential.

Abstract In the way to reduce costs in the poultry production, high densities of birds are adopted in the production systems. Such high densities can lead to an inappropriate environment, compromising the welfare and decreasing the animal performance. In this work we aimed to evaluate the effects of different densities on the productive yield, egg quality and immunological aspects of Japanese quails. It was used 816 Japanese quails, during egg production phase, in a random design, with four treatments (121.43 cm2/bird; 106.25 cm2/bird; 94.44 cm2/bird and 85.00 cm2/bird) with six repetitions each. Results indicate density did not interfere on egg quality and all treatments can be considered excellent, once the Haugh Unit was 88.03. Therefore, higher densities decline the yield, decreasing the eggs weight (10.35 g), the laying percentage (88%), egg mass (9.16 g) and the week average feed consumption (157.35 g). IgY values were higher at smaller cage density, suggesting birds in this condition presented better immunological condition, related to welfare and stress factors, all linked to immunoglobulins IgY.

Assessment of phosphorus in fruit juices is of great interest due to its essentiality and nutritional properties. This study applied sample treatment strategies to determine phosphorus in fractions of juices. Firstly, total phosphorus (Ptotal) was determined from digestion and sample suspension exploring direct analysis by inductively coupled plasma optical emission spectrometry, which presented no significant difference, for 95% of confidence level. Then, free phosphorus (Pfree) was determined by spectrophotometry and represented 30-90% of Ptotal with an inverse relationship respecting Ptotal concentration. Fractioning according to particle size evidenced highest fraction of Pfree in samples after filtration. Fractioning of phosphorus based on its charge was also performed and the high percentages of P in the anionic fraction (from 91.2 to 95.9%) are related to free inorganic phosphate in equilibrium with its protonated forms. Thus, it is more assailable after consumption, giving this food great functionality on human diet.

A produção de ovos de codorna tem experimentado um aumento acentuado na última década. A nutrição é o principal fator que afeta o potencial produtivo na indústria de aves, assim como o manejo nutricional adequado é necessário para garantir a manutenção das condições físicas ideais, crescimento e produção de produtos de alta qualidade. A farinha de carne e ossos (FCO) tem sido frequentemente utilizada na indústria avícola como uma fonte alternativa e de baixo custo de proteína em substituição parcial do farelo de milho e soja. No entanto, até esta data, não houve estudos que tinham investigado o efeito da dieta FCO sobre o desempenho de codornas ou sobre os custos de produção na fase inicial de criação. Isto é particularmente importante tendo em vista que esta fase é caracterizada por grandes investimentos por parte dos produtores, sem retorno econômico imediato. Neste estudo, nós investigamos se a substituição parcial do farelo de soja (FS) por farinha de carne e ossos (FCO) na dieta de codornas japonesas durante a fase inicial de criação é uma alternativa viável que venha a manter ou melhorar o seu desempenho produtivo e econômico. Nossos resultados mostram que a inclusão da FCO na dieta de codornas reduz os custos de alimentação por até 6% sem prejudicar o desempenho produtivo.
Quail egg production has experienced a steep rise in the last decade. Nutrition is the main factor affecting productive potential in the poultry industry, as appropriate nutritional management is necessary to ensure the maintenance of optimal physical conditions, growth and the production of high quality products. Meat and bone meal (MBM) has often been used in the poultry industry as an alternative and cost-effective source of protein in partial replacement of corn and soybean meal. However, there have been no studies to date that have investigated the effect of dietary MBM on the performance of quail or on the costs of production in the starter phase. This is particularly important considering that this phase is characterized by large investments by producers, without immediate economic return. In this study, we investigated whether partial replacement of soybean meal (SBM) by meat and bone meal (MBM) in the diet of Japanese quail during the starter phase is a viable alternative that would maintain or improve their productive and economic performance. Our results show that the inclusion of MBM in the diet of quail reduces feeding costs by up to 6% without impairing productive performance.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.
