Isolation of potential zoonotic Mycobacterium spp. from diseased freshwater angelfish (Pterophyllum scalare) from an aquarium

Resumo A infecção por micobactérias não tuberculosas é uma das doenças bacterianas crônicas mais comuns em peixes ornamentais de aquário e parece estar diretamente relacionada a práticas de manejo estressantes. Além disso, também representa potencial zoonótico. Aqui, apresentamos o isolamento e a caracterização de micobactérias não tuberculosas de peixes-anjo de água doce doentes (Pterophyllum scalare) em São Paulo, Brasil. Foram avaliadas nove fêmeas reprodutoras descartadas com sinais de doença. Os peixes exibiam letargia, perda de apetite, caquexia, úlceras cutâneas e exoftalmia. Na necropsia, quatro peixes apresentaram granulomas macroscópicos no baço. Mycobacterium chelonae, M. fortuitum, M. gordonae, M. intracellulare e M. peregrinum foram isolados e identificados por análise de restrição por PCR de hsp65. A análise histopatológica revelou lesões microscópicas compatíveis com micobacteriose, e o bacilo de Mycobacterium foi observado pela coloração de Ziehl-Neelsen. Notavelmente, todas as espécies de Mycobacterium identificadas neste estudo já foram relatadas em pacientes humanos; portanto, animais doentes podem ser uma fonte de infecção para pessoas que manipulam peixes e aquários.
Abstract Nontuberculous mycobacteria infection is one of the most common chronic bacterial diseases in ornamental aquarium fish and appears to be directly related to stressful husbandry practices. Furthermore, it also represents zoonotic potential. Here we present the isolation and characterization of non-tuberculous mycobacteria from diseased freshwater angelfish (Pterophyllum scalare) in São Paulo, Brazil. Nine discarded breeding females with signs of disease were evaluated. The fish exhibited lethargy, loss of appetite, cachexia, skin ulcers, and exophthalmia. At necropsy, four fishes presented macroscopic granulomas in the spleen. Mycobacterium chelonae, M. fortuitum, M. gordonae, M. intracellulare and M. peregrinum were isolated and identified by hsp65 PCR restriction analysis. Histopathological analysis revealed microscopic lesions compatible with mycobacteriosis, and Mycobacterium bacillus were observed by Ziehl-Neelsen stain. Notably, all Mycobacterium species identified in this study have already been reported in human patients; therefore, diseased animals may be a source of infection for people who handle fish and aquariums.

Abstract Low density sugarcane plantation (LDSP) has been implemented by some sugarcane producers in Brazil, aiming to save seeds and operational costs. The study was carried out in the municipality of Areia, Paraíba, Brazil. Five planting densities were used, varying from 5 to 25 m-2 of buds arranged in randomized blocks, with four replications. Data were measured annually over three cultivation cycles (2017 to 2020), during which the field was fertilized with NPK and the harvests were carried out manually without prior burning. The lower planting density presents higher productivity only in the cane plant (101.03 t ha-1) due to the higher plant height (2.37 m) and the higher number of stalks (11 stalks m-2), suggesting that these variables are due to the greater availability of light, water and photosynthate. However, there is a drastic reduction in sugarcane yield for this lower population in the 2nd ratoon by up to 65.62%, which is correlated with number of stalks per meter. We demonstrate the agronomic viability of LDSP in the population of 10 buds m-2 in relation to conventional planting of sugarcane until the 2nd ratoon. Data are important for future studies to present additional considerations for other production factors, such as the effects of mechanized harvesting and the management of nutrients and water, assessing the sustainability of this large-scale planting system.
Resumo O método de plantio de cana-de-açúcar de baixa densidade (LDSP) vem sendo implementado por alguns produtores de cana-de-açúcar no Brasil, visando economia de sementes e custos operacionais. O estudo foi realizado no município de Areia, Paraíba, Brasil. Utilizou-se cinco densidades de plantio, variando de 5 a 25 m-2 de gemas dispostas em blocos casualizados, com quatro repetições. Os dados foram medidos anualmente ao longo de três ciclos de cultivo (2017 a 2020). Menor densidade de plantio mostrou maior produtividade (101,03 t ha-1) devido à maior altura de planta (2,37 m) e ao maior número de hastes (NS) (11 hastes m-2), sugerindo que essas variáveis são devido à maior disponibilidade de luz, água e fotoassimilados. No entanto, houve redução drástica na produtividade da cana-de-açúcar para esta população menor na 2ª soca em 65,62%, estando correlacionado com o número de colmos por metro. Demonstramos a viabilidade agronômica do LDSP em uma população de 10 gemas m-2 em relação ao plantio convencional de cana-de-açúcar até a 2ª soca. Os dados são importantes para que estudos futuros apresentem considerações adicionais sobre outros fatores de produção, como os efeitos da colheita mecanizada e do manejo de nutrientes e água, avaliando a sustentabilidade desse sistema de plantio em larga escala.

ABSTRACT The Atlantic Forest is made up of different vegetation types over extensive latitude. Most of its remnants are in mountain ranges, which are responsible for the maintenance of most endemic and threatened species in this domain. Here we present the floristic composition of the Área de Proteção Ambiental Mestre Álvaro (APAMA), a coastal massif at Espírito Santo State with expressive altitudinal amplitude (100-800 a.s.l.) containing remnants never floristically studied. Knowing the composition of this protected area is important because it includes an altitudinal range not yet evaluated in floristic comparisons involving highland and lowland forests between the latitudes 18-21° S. We found 493 species (in 292 genera and 91 families): ten are regional endemic species, nine are new records of occurrence for the state, and 52 are under some degree of threat of extinction. Our results indicate the presence of flora elements' typical of both highland and lowland forests, which makes the APAMA essential in a scenario extremely fragmented. Finally, our data provide an unprecedented floristic list for regional forest restoration projects, as well as an invaluable source of data for biogeographic studies that seek to understand the effect caused by neighboring vegetation types on the composition of lowland forests.

This report describes a rapid and inexpensive colorimetric method based on biuret assay for total serum protein analysis using wax printed microzone plates and a desktop scanner. The colorimetric assay was performed in 96-microzone plates fabricated on polyester films through a wax printing protocol. The image scanning mode and the concentration of the biuret reagents were optimized to achieve the highest detectability levels. The best analytical response was obtained using the slide scanning mode as well as 4% CuSO4 and 150 mmol L-1 potassium sodium tartrate solutions. The proposed method offered linear relationship for the bovine serum albumin concentration range between 25 and 200 µmol L-1 with a limit of detection of 9.7 µmol L-1. The colorimetric approach revealed satisfactory precision (relative standard deviation values between 0.6 and 4.4%). The bioanalytical feasibility was demonstrated through the total protein analysis in serum samples collected from dogs previously diagnosed with kidney disease. The total serum protein concentrations varied from 960 ± 19 to 1661 ± 38 µmol L-1 and the achieved values revealed good accordance with those obtained through a reference method. Based on the reported results, wax printed microzone plates have demonstrated simplicity and potential to be adopted as alternative platforms for bioassays.

Abstract This study was carried out to determine the toxicity and biochemical and morphophysiological changes caused by Serjania erecta leaf aqueous extract in pacu (Piaractus mesopotamicus). For acute toxicity testing (CL50-4h), pacu juveniles were exposed during 4 h to Serjania erecta aqueous extract concentrations of 2.5, 12.5, 25, 50, 100, and 150 µg mL-1, which were added directly to the water in the tanks. In the control group, the animals were kept in water free from aqueous extract. CL50-4 h was estimated at 57.43 µg mL-1. After exposure to the aqueous extract, the highest (P<0.05) glucose concentration and the lowest (P<0.05) plasma sodium level were when the fish were exposed to the S. erecta concentration of 50 µg mL-1. Mortality occurred at S. erecta extract levels higher than 50 μg mL-1, and all fish died at concentrations greater than 100 μg mL-1. In addition, exposure to this extract caused severe histological changes in the gills and liver with higher prevalence of necrosis (30.2%), and fatty degeneration (77.4%) respectively. At the concentrations tested here, S. erecta aqueous extract causes morphofunctional alterations in this fish species.

RESUMO A estabilidade lateral de vigas pré-moldadas esbeltas durante as fases transitórias e em serviço deve ser avaliada, observando que a presença de imperfeições geométricas torna o problema da instabilidade mais susceptível. O objetivo principal deste trabalho é avaliar o comportamento de vigas de concreto sobre apoios elastoméricos, considerando a influência de variáveis como: a resistência do concreto, a ação do vento e a rigidez a compressão das almofadas de apoio. Para a análise numérica não-linear utiliza-se neste trabalho o programa computacional ANSYS, baseado no método dos elementos finitos. As análises mostram que a influência da resistência do concreto é significante na estabilidade lateral da viga. A ação do vento pode ser responsável pela redução da área de contato entre a viga e a almofada de apoio (efeito de lift-off). Finalmente, a combinação destes fatores pode resultar em uma situação crítica de tensões na viga, para a qual não há condição de equilíbrio, resultando no tombamento da viga.
Abstract The behavior of slender precast beams related to lateral stability in the transitional and in service phases is worrying. The presence of geometric imperfections aggravates and makes the problems of instability more susceptible. The main objective of this work is to evaluate the behavior of concrete beams on elastomeric bearings and to analyze the influence of variables such as: concrete strength, wind load and bearing compression stiffness. For the numerical nonlinear analysis the software ANSYS based on the Finite Element Method was used. The analyses show that the influence of the strength of the concrete is significant in the lateral stability of the beam. The wind load represents a considerable decrease in the contact (lift off) between the beam and the bearing. Finally, the combination of these factors can result in a critical stress situation in the beam, and it is not possible to have equilibrium, causing its toppling.

Resumo A estabilidade lateral de vigas pré-moldadas longas e esbeltas tem sido associada a colapsos durante as fases transitórias de montagem. Os inevitáveis desvios de execução entre a seção transversal do apoio e a do meio do vão agravam e tornam mais suscetíveis os problemas de instabilidade. O objetivo deste trabalho é avaliar numericamente o comportamento de vigas de concreto que apresentam imperfeições geométricas sobre almofadas de apoio de elastômero e analisar a influência de variáveis, como: resistência característica do concreto, não linearidade física do concreto e rigidez à compressão da almofada. As análises numéricas foram desenvolvidas em um programa computacional baseado no Método dos Elementos Finitos. Dentre as principais conclusões, percebeu-se que o aumento da resistência característica do concreto confere à viga maior estabilidade e que a consideração do comportamento não linear físico do material não permite encontrar uma posição de equilíbrio no modelo numérico em alguns casos avaliados. A rigidez ao giro da almofada diminui na medida em que o ângulo de esconsidade aumenta. Deve-se ter em mente que, em situações muito críticas, pode não ser possível encontrar equilíbrio para a viga, levando ao seu colapso.
Abstract The stability of long and slender precast beams has been associated with structural collapse during the transitory phase of construction. The inevitable deviations regarding the execution between the support cross-section and midspan aggravate the instability problem, thus causing the beams to become even more susceptible to its effects. The focal point of this study is the numerical and behavioral evaluation of concrete beams that present geometric imperfections on elastomeric supports and analyze the influence exerted by the variables of: strength characteristics of concrete, physical non-linearity of the concrete and the compression stiffness of the bearing pad. The numerical analyses were developed on a computer program based on the finite element method (FEM). Among the principle conclusions drawn from the study, one finds that the increase in the characteristic strength of the concrete provides the beam with a higher degree of stability, and the consideration of the physical non-linear behavior of the material did not allow the equilibrium point on the numerical model to be found in some of the cases that were evaluated. The rotational stiffness of the pad decreases as the skew angle increases. Therefore, one needs to remember that in very critical situations, it may not be possible to find the point of equilibrium, thus leading to collapse.

Resumo A produção de compostos via esterificação enzimática possui grande interesse científico e tecnológico devido às inúmeras inconveniências relacionadas com a catálise ácida, principalmente por estes sitemas não se adequarem ao atual termo “tecnologias limpas”. Além disso, produtos naturais como o óleo de cravo, apresentam compostos com excelentes potenciais biológicos. Compostos bioativos são quase sempre tóxicos em altas doses. A avaliação da letalidade em um organismo animal menos complexo pode ser usada para um monitoramento simples e rápido, servindo também para a identificação de compostos com potencial atividade inseticida contra larvas de insetos vetores de doenças. Neste sentido, foi determinada a toxicidade frente a Artemia salina do óleo essencial de cravo e do seu derivado acetato de eugenila obtido por esterificação enzimática com lipase Novozym 435. A conversão da reação de síntese de acetato de eugenila foi de 95,6%. Os resultados referentes à avaliação da toxicidade frente ao microcrustáceo Artemia salina demonstraram que tanto o óleo (LC50= 0,5993 µg.mL–1) quanto o éster (LC50= 0,1178 µg.mL–1) apresentam elevado potencial toxicológico, sendo que o éster apresenta aproximadamente 5 vezes mais toxicidade em relação ao óleo. Estes resultados demonstram o potencial emprego do óleo de cravo e de acetato de eugenila em formulações de inseticidas.
Abstract The production of compounds via enzymatic esterification has great scientific and technological interest due to the several inconveniences related to acid catalysis, mainly by these systems do not fit to the concept of “green chemistry”. Besides, natural products as clove oil present compounds with excellent biological potential. Bioactives compounds are often toxic at high doses. The evaluation of lethality in a less complex animal organism can be used to a monitoring simple and rapid, helping the identification of compounds with potential insecticide activity against larvae of insect vector of diseases. In this sense, the toxicity against Artemia salina of clove essential oil and its derivative eugenyl acetate obtained by enzymatic esterification using Novozym 435 as biocatalyst was evaluated. The conversion of eugenyl acetate synthesis was 95.6%. The results about the evaluation of toxicity against the microcrustacean Artemia salina demonstrated that both oil (LC50= 0.5993 µg.mL–1) and ester (LC50= 0.1178 µg.mL–1) presented high toxic potential, being the eugenyl acetate almost 5 times more toxic than clove essential oil. The results reported here shows the potential of employing clove oil and eugenyl acetate in insecticide formulations.

ABSTRACT Objective To investigate the acute effects of EPAP on the activity of sternocleidomastoid (SCM), parasternal muscles and ventilatory parameters in COPD patients. Method Twenty-four patients with COPD were studied using surface electromyography (sEMG) and a ventilometer. Patients were randomly assigned to EPAP 10 cmH2O-EPAP10 or 15 cmH2O-EPAP15 for 20 minutes. Results The parasternal muscle sEMG activity increased during EPAP10 and EPAP15; however, a greater and significant increase was observed with EPAP10 (mean between-group difference: 12.5% RMS, 95% CI: 9.5 to 15.4, p<0.001). In relation to the baseline, at 10 and 20 minutes and upon recovery, respectively parasternal activity increased by 23.9%, 28.9% and 19.1% during EPAP10 and by 10.7% at 10 and 20 minutes and upon recovery, respectively, 11.4% and 6.9% during EPAP15 at 10 and 20 minutes and upon recovery, respectively. The sEMG activity of SCM muscle showed an opposite pattern, increasing with EPAP15 and decreasing with EPAP10 (mean between-group difference: 15.5% RMS, 95% CI: 12.6 to 18.4, p<0.001). SCM muscle activity during EPAP15, increased by 4.8% and 6.1% at 10 and 20 minutes and decreased by -4.0% upon recovery compared to decreases of –5.6%, –20.6% and –21.3% during EPAP10 at 10, 20 minutes, and recovery. Ventilation at both EPAP intensities promoted significant reductions in respiratory rate (RR) and dyspnea, more pronounced in EPAP15: RR (mean between-group difference: –3,8bpm, 95%CI: –7,5 to –0,2, p=0,015) and dyspnea (mean between-group difference: –1.01, 95%CI: –1.4 to –0.53, p=0.028) . Conclusion In COPD patients, the use of EPAP10 was more effective in reducing accessory inspiratory activity and increasing parasternal activity, which was accompanied by an improvement in ventilation and a reduction in dyspnea.

Resumo Apresenta-se uma síntese atualizada de informações sobre algas no Brasil objetivando refinar os dados reunidos até o presente, bem como avaliar os avanços sobre o conhecimento da diversidade de algas no país desde a publicação do Catálogo de Plantas e Fungos do Brasil. Os resultados de 2015 mostraram um acréscimo de 1.250 espécies (35.7%) a um total de 4.747 em relação a 2010. As classes mais diversas em número de espécies foram Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae e Euglenophyceae. Bacillariophyceae e Cyanophyceae tiveram o maior acréscimo de espécies no intervalo de cinco anos. A região Sudeste e Sul foram as mais diversas, porém, as regiões Nordeste com os estados do Piauí e Sergipe e Centro-Oeste com os estados de Mato Grosso, Goiás e Distrito Federal destacaram-se no cenário da biodiversidade nacional. Apesar da escassez de taxonomistas e da infraestrutura limitada, os resultados obtidos evidenciaram um avanço significativo no conhecimento da diversidade de algas no país nesse período de cinco anos, iniciando uma mudança quanto as diferenças geográficas regionais.
Abstract An updated synthesis of cyanobacteria and algae information is presented for Brazil aiming to refine the data gathered to date and evaluate the progress of the biodiversity knowledge about these organisms in the country since the publication of the Catálogo de Plantas e Fungos do Brasil. The results of 2015 showed an increase of 1,250 species (35.7%) when compared to 2010, reaching a total of 4,747 species. The most diverse classes in species number were the Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae and Euglenophyceae. Bacillariophyceae and Cyanophyceae had the highest increase in species number in the five-year interval. The Southeast and South regions were the most diverse, however, the Northeast, with the states of Piauí and Sergipe, and the Central-west region, with Mato Grosso, Goiás and Distrito Federal, also stood out in the national algal biodiversity scenario. Despite the shortage of taxonomists and limited infrastructure, the results showed a significant improvement in the knowledge regarding the diversity of cyanobacteria and algae in the country during the study period, starting to even out regional geographical differences caused by subsampling.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

MORPHOMETRIC ANALYSIS OF THE INFRAORBITAL FORAMEN AND ACESSORIES FORAMINAS IN BRAZILIAN SKULLS

The aim of the study was to establish morphometrically the infraorbital foramen (IOF) location in the sagittal and transversal plan of the skull and to analyze the variations, as well as verifying the symmetry with the contralateral IOF. 210 skulls were analyzed and as exclusion criterion, were not analyzed skulls of children. Digital paquimeter was used and had been carried through two bilateral measures: a)sagittal measure; from infraorbital margin until the superior margin of the IOF; b) transversal measure, of the lateral margin of piriformis opening until the medial margin of the IOF, using the transversal plan. These measures had formed an angle of 90 between itself. The data were analyzed by the Kolmogorov-Smirnov to verify the Gaussian distribution and and Person's test to evaluate the correlation, considering significantly the differences that had presented a p 0,05. The mean±SD in the sagital measures were respectively; right side 6,71±1.70 mm and 6,83±1.83 mm for left side (p0.0001). Transversal measures were: right side 13,28±2.17mm and 13,31±2.19 mm for left side (p0.0001). Were observed supranumerary foramens of the IOF, with presence of the double IOF in 50 cases, being bilateral in 08 specimens, left and right side 21 specimens for each side.The IOF are not absolutely symmetrical and may present pair bilateral and or unilateral opening. The presence of the supranumerary foraminas, as well as, the morphometrics measures in this study may intervene with the anesthesia, thus, suggest that they must be considered in the anaesthesical evaluation and be remembered during the procedures used for the localization of the forame
El objetivo del estudio fue establecer morfométricamente la localización del agujero infraorbital (FIO) en los planos sagital y transversal del cráneo y analizar las variaciones, así como verificar la simetría con el FIO contralateral. Fueron analizados 210 cráneos de individuos adultos. Utilizamos un paquímetro digital para efectuar las dos mediciones: a) sagital: del borde infraorbital hasta el borde superior del FIO; b) transversal: del borde lateral de la abertura piriforme hasta el bordemedial del FIO, utilizando el plano transversal. Estas mediciones forman un ángulo de 90 entre sí. Los datos fueron analizados por Kolmogorov-Smirnov y prueba de Person para evaluar la correlación, considerando un p 0.05. La media y ± SD en la distancia sagital fueron en el lado derecho 6,71± 1.70 mm y en el lado izquierdo 6,83±1.83 mm (p0.0001). Las medidas transversales fueron: lado derecho 13.28±2.17 mm y lado izquierdo 13.31±2.19 mm (p0.0001). Fueron observadas foraminas supranumerarias del FIO, con la presencia del IOF doble en 50 casos, siendo bilaterales en 8 especímenes izquierdos y derechos de la cara, 21 para cada lado. El FIO no era absolutamente simétrico y podía presentar el par bilateral y la abertura unilateral. La presencia de foraminas supranumerarias, como diversas medidas de este estudio, pueden interferir con la anestesia. Así sugerimos considerar estos parámetros en la evaluación anestésica y recordar las variantes durante los procedimientos usados para la localización del agujero infraorbital

O trabalho teve o objetivo de identificar mudanças de conhecimentos e práticas de prevenção do dengue, e desenvolveu-se em duas áreas de Catanduva: Área de Estudo e Área Controle. Realizaram-se, entre 1999 e 2001, levantamentos quantitativo inicial e qualitativo; diagnóstico preliminar, apresentado à comunidade da Área de Estudo, desencadeando discussão para a definição e o direcionamento das ações; o desenvolvimento das ações na Área de Estudo, com a participação da comunidade; continuidade das atividades de rotina na Área Controle; e avaliação final. Na Área de Estudo, as ações realizadas foram: mudança do trabalho dos agentes de controle de vetores que passaram a demonstrar as medidas preventivas, sem a retirada de recipientes e sem utilização de larvicida; utilização de material educativo específico à realidade local; atividades relacionadas às prioridades dos moradores; atividades como música, teatro, gincanas e brincadeiras; demonstração do ciclo do vetor. Como resultado, houve a diminuição significante do número de recipientes por casa; o aumento significante da proporção de casas sem recipientes; e o aumento significante da proporção de pessoas que reconheceram as larvas do vetor na Área de Estudo em relação à de Controle.
This study aimed to identify changes in knowledge and practices learned to prevent dengue fever in two areas of Catanduva, São Paulo State, from 1999 to 2001: a study area and a control area. The study included an initial quantitative survey, qualitative research, a preliminary diagnosis presented to the community to launch a discussion aimed at defining future actions, implementation of the actions in the study area with community participation (but without changes in the control area), and a final comparison of the two areas. Changes in the study area included: vector control workers began demonstrating preventive measures without removing potential breeding places or using larvicide; use of educational aids specific to the local reality; activities related to the residents’ priorities; and activities such as music, theater skits, scavenger hunts, and games to demonstrate the vector cycle. Potential domiciliary breeding sites were significantly reduced; the proportion of houses without breeding sites was significantly increased; and there was an increase in the percentage of individuals who recognized the larval form of the vector in the study area as compared to the control area.

Realizaram-se, entre 1999 e 2001, levantamentos quantitativo inicial e qualitativo; diagnóstico preliminar, apresentado à comunidade da Área de Estudo, desencadeando discussão para a definição e o direcionamento das ações; o desenvolvimento das ações na Área de Estudo, com a participação da comunidade; continuidade das atividades de rotina na Área Controle; e avaliação final. Na Área de Estudo, as ações realizadas foram: mudança do trabalho dos agentes de controle de vetores que passaram a demonstrar as medidas preventivas, sem a retirada de recipientes e sem utilização de larvicida; utilização de material educativo específico à realidade local; atividades relacionadas às prioridades dos moradores; atividades como música, teatro, gincanas e brincadeiras; demonstração do ciclo do vetor. Como resultado, houve a diminuição significante do número de recipientes por casa; o aumento significante da proporção de casas sem recipientes; e o aumento significante da proporção de pessoas que reconheceram as larvas do vetor na Área de Estudo em relação à de Controle.
This study aimed to identify changes in knowledge and practices learned to prevent dengue fever in two areas of Catanduva, São Paulo State, from 1999 to 2001: a study area and a control area. The study included an initial quantitative survey, qualitative research, a preliminary diagnosis presented to the community to launch a discussion aimed at defining future actions, implementation of the actions in the study area with community participation (but without changes in the control area), and a final comparison of the two areas. Changes in the study area included: vector control workers began demonstrating preventive measures without removing potential breeding places or using larvicide; use of educational aids specific to the local reality; activities related to the residents’ priorities; and activities such as music, theater skits, scavenger hunts, and games to demonstrate the vector cycle. Potential domiciliary breeding sites were significantly reduced; the proportion of houses without breeding sites was significantly increased; and there was an increase in the percentage of individuals who recognized the larval form of the vector in the study area as compared to the control area.

Liver morphology with emphasis on bile ducts changes and survival analysis in mice submitted to multiple Schistosoma mansoni infections and chemotherapy

Numa tentativa de estar o mais próximo possível a pacientes infectados e tratados nas áreas endêmicas de esquistosomose (S. mansoni) e também para obter um período mais longo de seguimento, camundongos foram repetidamente infectados com um número baixo de cercarias. Dados de sobrevivência e variáveis histológicas tais como granuloma esquistosomótico, alterações portais, necrose hepatocelular, regeneração hepática, pigmento esquistosomótico, fi-brose periductal e principalmente, alterações dos ductos biliares foram analisados nos animais infectados tratados e não tratados. A terapêutica por oxamniquina nos animais repetidamente infectados prolonga a sobrevivência de maneira significante (Chi-quadrado 9,24, p = 0,0024), portanto confirmando resultados anteriores com um modelo semelhante mas com um período mais curto de seguimento. Ainda, a mortalidade decresce rapidamente depois do tratamento, sugerindo uma abrupta redução na gravidade das lesões hepáticas. O fígado foi ainda estudado sob o ponto de vista morfológico e imunohistoquímico. Fibrose portal, com um quadro que lembra a fibrose humana do tipo Symmers está presente na fase tardia da infecção. As alterações de ductos biliares são muito próximas daquelas descritas na esquistosomose mansônica humana. Antígeno esquistosomótico foi observado em uma célula isolada do revestimento alterado de duetos biliares. A patogênese das alterações ductais e sua possível relação com a infecção parasitária e/ou seus antígenos foi discutida.
In an attempt to be as close as possible to the infected and treated patients of the endemic areas of schistosomiasis (S. mansoni) and in order to achieve a long period of follow-up, mice were repeatedly infected with a low number of cercariae. Survival data and histological variables such as schistosomal granuloma, portal changes, hepatocellular necrosis, hepatocellular regeneration, schistosomotic pigment, periductal fibrosis and chiefly bile ducts changes were analysed in the infected treated and non treated mice. Oxamniquine chemotherapy in repeatedly infected mice prolonged survival significantly when compared to non-treated animals (chi-square 9.24, p = 0.0024), thus confirming previous results with a similar experimental model but with a shorter term follow-up. Furthermore, mortality decreased rapidly after treatment suggesting an abrupt reduction in the severity of hepatic lesions. A morphological and immunohistochemical study of the liver was carried out. Portal fibrosis, with a pattern resembling human Symmers fibrosis was present at a late phase in the infected animals. Bile duct lesions were quite close to those described in human Mansonian schistosomiasis. Schistosomal antigen was observed in one isolated altered bile duct cell. The pathogenesis of the bile duct changes and its relation to the parasite infection and/or their antigens are discussed.
