Abstract In Compost-Bedded Pack Barn (CBP) systems, air velocity is linked with the thermal comfort of housed dairy cattle and bedding quality and, therefore, assessing ventilation efficiency is essential. In this context, the objective of this study was to evaluate and characterize dependence and spatial distribution of air velocity at the 1.5 m height (vair,M) and at bedding level (vair,B) in an open CBP system with positive pressure ventilation. The study was conducted in 2021, in a facility located in the Zona da Mata region, Minas Gerais, Brazil. The facility area was divided into a mesh composed of 55 equidistant points, where vair,M and vair,B data were collected in the morning (09:00 a.m.) and afternoon (03:00 p.m.) periods, during three weeks in Brazilian winter. Geostatistics techniques were used to assess dependence and spatial distribution. In both periods evaluated, there were a strong occurrence of spatial dependence and non-uniform vair,M and vair,B distributions. The vair,M and vair,B values were lower than recommended (1.8 m∙s–1) in more than 65.0% of the area. Adequate ventilation levels were observed only in the first 20.0 m of the facility, from Southeast to Northwest, because of the fan lines present.

Abstract Acute myeloid leukemia (AML) comprises a heterogeneous group of hematopoietic cell neoplasms of myeloid lineage that arise from the clonal expansion of their precursors in the bone marrow, interfering with cell differentiation, leading to a syndrome of bone marrow failure. AML is a consequence of genetic and epigenetic changes (point mutations, gene rearrangements, deletions, amplifications, and arrangements in epigenetic changes that influence gene expression) in hematopoietic precursor cells, which create a clone of abnormal cells that are capable of proliferating but cannot differentiate into mature hematopoietic cells or undergo programmed cell death. The diagnosis requires more than 20% myeloid blasts in the bone marrow and certain cytogenic abnormalities. Treatment will depend on age, comorbidities, and cytogenetic risk among the most frequent.

Risk factors for postoperative pulmonary complications and prolonged hospital stay in pulmonary resection patients: a retrospective study

Abstract Background Postoperative pulmonary complications are the main cause of morbidity and mortality after pulmonary resection. This study was undertaken to determine the risk factors associated with postoperative pulmonary complications (PPCs) and length of hospital stay (LOS) in pulmonary resection patients in a tertiary teaching hospital in Brazil. Methods A retrospective data gathering from 196 patients who underwent pulmonary resection between 2012 and 2016 was conducted. Demographic and hospital admission data were collected from patients with complete medical records. Univariate analysis was performed, followed by Poisson's regression for predicting the prevalence of postoperative pulmonary complications and length of hospital stay. Results Thirty-nine patients (20%) displayed pulmonary complications in the postoperative period. The risk factors associated with an increased prevalence of postoperative pulmonary complications in a multivariate analysis were: American Society of Anesthesiologists physical status (ASA) ≥ 3 (PR 4.77, p = 0.03, 95% CI: 1.17 to 19.46), predicted diffusion capacity of the lungs for carbon monoxide- corrected single breath (PR 0.98, p < 0.001, 95% CI: 0.96 to 0.99) and age of the patient (PR 1.04; p = 0.01; 95% CI: 1.01 to 1.06). Those associated with an increased prevalence of prolonged hospital stay were: duration of surgical procedure longer than five hours (PR 6.94, p = 0.01, 95% CI: 1.66 to 12.23), male sex (PR 5.72, p < 0.001, 95% CI: 1.87 to 9.58), and presence of postoperative pulmonary complications (PR 11.92, p < 0.001, 95% CI: 7.42 to 16.42). Conclusions The rate of postoperative pulmonary complications in the study population is in line with the world average. Recognizing risk factors for the development of PPCs may help optimize allocation resources and preventive efforts.

Multi-flow calibration (MFC) is based on monitoring the analytical signal from a single calibration standard solution at several different nebulization gas flow rates (Q), which normalizes plasma conditions and minimizes matrix effects. In the present study, MFC was evaluated, for the first time, applied to inductively coupled plasma optical emission spectrometry (ICP OES) and inductively coupled plasma mass spectrometry (ICP-MS) to analyze different and complex-matrix samples. Al, As, Cd, Cr, Cu, Mn, Pb, and Zn were determined by ICP OES, while As, Cd, Co, Cu, Hg, Mo, Mn, Ni, Se, Sb, Pb, and V were determined by ICP-MS. MFC results were compared with those obtained using external standard calibration (EC) for both techniques and in all cases, MFC showed equal or superior accuracy (recoveries between 80-120%) compared to EC, and lower relative standard deviation (RSDs ≤ 10%). Several tests were also performed using only two nebulization gas flows to build linear models for calibration (called two-flow calibration, TFC) and the accurate results (recoveries ranged from 80 to 110% for ICP OES and from 81 to 102% for ICP-MS) suggests that this strategy can be also applied, resulting in a method with high sample throughput.

Resumen Las diferencias en la proporción líquido/polvo pueden afectar las propiedades de los materiales a base de silicato de calcio. Este estudio evaluó la influencia de la proporción polvo/gel en las propiedades fisicoquímicas del cemento NeoMTA Plus. El tiempo de fraguado (minutos), la fluidez (mm y mm²), el pH (en diferentes períodos), la radiopacidad (mmAl) y la solubilidad (% de pérdida de masa) fueron evaluados utilizando las consistencias para el material de reparación radicular (NMTAP-RP; 3 cucharadas de polvo/2 gotas de gel) y para cemento sellador del conducto radicular (NMTAP-SE; 3 cucharadas de polvo/3 gotas de gel), en comparación con el cemento Biodentine (BIO) y el cemento TotalFill BC (TFBC). El análisis estadístico se realizó utilizando las pruebas ANOVA y Tukey unidireccionales (α=0.05). BIO tuvo el tiempo de fraguado más corto, seguido de NMTAP-RP y NMTAP-SE. TFBC mostró el mayor tiempo de fraguado y radiopacidad. BIO, NMTAP-RP y NMTAP-SE tuvieron una radiopacidad similar. Todos los materiales promovieron un pH alcalino. NMTAP-RP/ SE tuvieron una solubilidad menor que BIO y TFBC. Con respecto a la fluidez, TFBC tuvo los valores más altos, seguido de NMTAP-SE y NMTAP-RP. BIO tuvo la fluidez más baja. En conclusión, NMTAP en la relación polvo/gel mostró un pH alto y una baja solubilidad. El aumento en la proporción de polvo disminuyó el tiempo de fraguado y la fluidez. Estos hallazgos son importantes con respecto a su consistencia y tiempo de trabajo durante la aplicación clínica.
Abstract Differences in liquid-to-powder ratio can affect the properties of calcium silicate-based materials. This study assessed the influence of powder-to-gel ratio on physicochemical properties of NeoMTA Plus. Setting time (minutes), flow (mm and mm²), pH (at different periods), radiopacity (mm Al) and solubility (% mass loss) were evaluated using the consistencies for root repair material (NMTAP-RP; 3 scoops of powder to 2 drops of gel) and root canal sealer (NMTAP-SE; 3 scoops of powder to 3 drops of gel), in comparison to Biodentine cement (BIO) and TotalFill BC sealer (TFBC). Statistical analysis was performed using one-way ANOVA and Tukey tests (α=0.05). BIO had the shortest setting time, followed by NMTAP-RP and NMTAP-SE. TFBC showed the highest setting time and radiopacity. BIO, NMTAP-RP, and NMTAP-SE had similar radiopacity. All materials promoted an alkaline pH. NMTAP-RP/SE presented lower solubility than BIO and TFBC. Regarding the flow, TFBC had the highest values, followed by NMTAP-SE, and NMTAP-RP. BIO had the lowest flow. In conclusion, NMTAP in both powder-to-gel ratios showed high pH and low solubility. The increase in the powder ratio decreased the setting time and flow. These findings are important regarding the proper consistency and work time to clinical application.

Resumo Clyomys laticeps é um roedor equimídeo encontrado em áreas abertas dos biomas Cerrado e Pantanal, ao longo do Brasil central e Paraguai. Essa espécie apresenta hábitos semi-fossoriais e atividade noturna. Até o momento, não existem estudos sobre seus parasitas. O objetivo deste trabalho foi descrever a comunidade de helmintos encontrados em C. laticeps, capturados no Parque Estadual da Serra de Caldas Novas, uma área de Cerrado no Brasil Central. Os indivíduos foram capturados em área de “campo sujo” entre janeiro e outubro de 2016. Os intestinos delgado e grosso de 14 roedores foram examinados quanto à presença de helmintos. Três espécies de nematoides foram encontradas: Fuellebornema almeidai, Pterygodermatites (Paucipectines) sp. e Subulura forcipata. Considerando-se as três espécies, 85,7% dos roedores apresentaram pelo menos um helminto. F. almeidai foi a espécie mais prevalente, enquanto Subulura forcipata foi a mais abundante. Este estudo é o primeiro registro da estrutura da comunidade de helmintos para C. laticeps. Como resultado, foi registrado um novo hospedeiro e o aumento da área de distribuição geográfica para o nematódeo F. almeidai e o primeiro caso de Pterygodermatites (Paucipectines) sp. infectando um roedor equimídeo. Além disso, este estudo também traz o primeiro registro de S. forcipata infectando um mamífero.
Abstract The broad-headed spiny rat, Clyomys laticeps, is an echimyid rodent found in open areas of Cerrado and Pantanal biomes in central Brazil and Paraguay. Little is known about the parasites associated with this semi-fossorial species, as no previous studies have been conducted on their helminth fauna. The aim of this study was to report the helminth community structure of C. laticeps inhabiting Serra de Caldas Novas State Park, a Cerrado area in central Brazil. Trappings were carried out in dry grasslands from January to October 2016, and the large and small intestines of 14 C. laticeps individuals were examined for the presence of helminths. Three nematode species were found: Fuellebornema almeidai, Pterygodermatites (Paucipectines) sp., and Subulura forcipata, and 85.7% of the studied species were infected with at least one of these helminths. F. almeidai was the most prevalent species among hosts, and S. forcipata was the most abundant. This study is the first report on helminth community structure in C. laticeps. We report a new host species and increase the known geographical range of F. almeidai, and provide the first record of Pterygodermatites (Paucipectines) sp. infecting echimyids. This is also the first report of S. forcipata in a mammal host.

Identificação e seleção de espécies de Trichoderma spp. endofíticos de bromélias de restingas como agentes de biocontrole da fusariose em frutos de abacaxi

RESUMO Uma das doenças fúngicas de grande importância que ataca a cultura do abacaxizeiro é a fusariose, causado pelo fungo Fusarium guttiforme. A utilização do Trichoderma spp, que são fungos habitantes do solo e de ocorrência natural na forma endofítica em bromélias, ainda não foi estudada visando o biocontrole da gomose do abacaxizeiro. O trabalho objetivou identificar espécies de Trichoderma endofíticos de bromélias em Restinga e avaliar o antagonismo in vitro e in vivo a F. guttiforme, agente etiológico da gomose do abacaxi. Foram avaliados 5 isolados de Trichoderma mantidos em armazenamento na Clínica Fitossanitária da Universidade Estadual do Norte Fluminense Darcy Ribeiro (CF/UENF440, CF/UENF441, CF/UENF442, CF/UENF443 CF/UENF444). Um isolado de F. guttiforme patogênico ao abacaxizeiro também foi utilizado. Para identificação específica dos isolados de Trichoderma, efetuou-se a extração do DNA e o sequenciamento dos genes ITS. Procedeu-se a análise filogenética e os isolados endofíticos de bromélias foram agrupados na seção Longibrachiatum e, com baixa homologia dentre as espécies conhecidas, podendo tratar-se, portanto, de nova espécie, o que ainda será confirmado com a obtenção de novas sequências de genes específicos. Foram realizados testes de antibiose in vitro para avaliação e seleção dos isolados antagonistas (antagonismo em cultivo pareado, efeito inibitório de compostos voláteis, não voláteis e não voláteis termoestáveis) e avaliou-se o tratamento biológico de frutos de abacaxi com ferimentos, visando-se avaliar o biocontrole da gomose em pós-colheita. O isolado CF/UENF440 se mostrou como um potencial agente de biocontrole in vitro de F. guttiforme, pois demonstrou forte antibiose em co-cultivo meio de cultura, tanto para compostos não-voláteis quanto para não-voláteis termoestáveis, diferindo significativamente dos demais isolados testados e do controle. Na avaliação do biocontrole in vivo em frutos de abacaxi com ferimentos, os isolados de Trichoderma testados não apresentaram significativo efeito protetor no biocontrole da gomose em frutos na pós-colheita.
ABSTRACT One of the major fungal diseases affecting the pineapple crop is fusariosis, caused by the fungus Fusarium guttiforme. The use of Trichoderma spp., which are soil fungi naturally occurring as endophytic forms in Bromeliaceae, has not been studied for the biocontrol of fusariosis in pineapple plants. The present study aimed to identify Trichoderma species endophytic to Bromeliaceae in “Restinga” and to evaluate the in vitro and in vivo antagonism against F. gutifforme, the etiological agent of pineapple fusariosis. Five Trichoderma isolates were evaluated, which were stored in the Phytosanitary Clinics of the State University of North Fluminense Darcy Ribeiro (CF/UENF440, CF/UENF441, CF/UENF442, CF/UENF443 CF/UENF444). A F. guttiforme isolate, pathogenic to pineapple, was also used. For specific identification of Trichoderma spp., DNA extraction and sequencing and of ITS genes were carried out. Phylogenetic analysis was conducted and the endophytic isolates from Bromeliaceae were grouped in the Longibrachiatum section, showing low homology among the known species, which may indicate a new species that will be confirmed with the obtaining of additional sequences of specific genes. In vitro antibiosis tests were then carried out to select antagonistic isolates (antagonism in matched culture, inhibitory effect of volatile, non-volatile and non-volatile thermostable compounds) and the biological treatment of wounded pineapple fruits was evaluated with the aim of assessing the biocontrol of fusariosis in the post-harvest. The isolate CF/UENF440 was shown to be a potential in vitro biocontrol agent of F. guttiforme since it demonstrated strong antibiosis in co-cultivation culture medium, for both non-volatile and non-volatile thermostable compounds, significantly differing from the remaining tested isolates and control. Considering the evaluation of in vivo biocontrol in wounded pineapple fruits, the tested Trichoderma isolates had no significant protectant effect in the biocontrol of fusariosis in post-harvest fruits.

Agroindustrial performance and phenotypic parameters evaluation of sugarcane varieties in different crop seasons

A cana-de-açúcar pode ser cultivada como matéria-prima para a produção de cachaça artesanal e forragem para alimentação animal, porém normalmente não se utiliza um bom aporte tecnológico, pois são plantadas variedades usadas para produção de açúcar e álcool. Este trabalho teve como objetivo avaliar o desempenho agroindustrial e a estimativa de parâmetros fenotípicos em variedades de cana-de-açúcar avaliadas em duas safras. O delineamento experimental utilizado foi o látice triplo 4 x 4, sendo avaliadas dezasseis variedades de três diferentes instituições. Foram avaliados caracteres agronómicos e tecnológicos com importância agroindustrial. Houve diferença significativa entre as variedades e as safras para os caracteres avaliados na análise conjunta. A análise dos parâmetros fenotípicos contribuiu para o entendimento do desempenho dos caracteres ao longo das duas safras. As variedades possuem comportamento dessemelhante, porém, apresentam regularidade no desempenho, ao longo do tempo, para a maioria dos caracteres agronómicos e industriais avaliados. As características fenotípicas POL e ATR, denotam boa confiabilidade em prever o valor genotípico para estas variáveis. O alto número de medições necessárias para os caracteres TCH, PUREZA e MMC, dificultam a seleção de variedades potenciais. A correlação fenotípica avaliada em duas safras não é suficiente para realizar a recomendação de variedades.
Sugarcane can be cultivated as raw material for craft cachaça production and forage for animal feed, but usually a good technological input is not used, because varieties for sugar and ethanol production are planted. The aim of this study was to evaluate the agroindustrial performance and the estimation of phenotypic parameters of sugarcane varieties of the first and second crop seasons. The experimental design was a triple lattice 4 x 4, being assessed sixteen varieties of three different institutions. Agronomic and technological characters with agroindustrial importance were evaluated. There was a significant difference between the varieties and the crop seasons for the traits evaluated in the joint analysis. The phenotypic parameters analysis contributed to the understanding of the traits performance throughout two crop seasons. Varieties do not present a similar behavior, however, they have regularity in the performance, over time, for most agronomic and industrial characters evaluated. Phenotypic characteristics POL and TRS, show good reliability in predicting the genotypic value for these variables. The high number of measurements required for the TSH, Purity and SM characters make it difficult to select potential varieties. Phenotypic correlation evaluated in two harvests is not sufficient to carry out the recommendation of varieties.

In this study, the elemental composition of nine medicinal plants and phytomedicines was evaluated by using axial view inductively coupled plasma optical emission spectrometry (ICP OES) after three digestion procedures: dry ashing (DA) and two wet digestions with a mineral acid (HNO3/H2O2) mixture on a conventional hot plate (CD), besides microwave digestion (MW). Accuracy was assessed from spinach leaves (CRM 1570a) and recovery values were in the range of 82 to 113.8%. Precisions, such as relative standard deviation (RSD), below 10% were reached. The MW procedure was preferred for its accuracy (recovery values of 94 to 102%), reduced contamination, increase in the efficiency of the decomposition process, lower residual acidity and reduced time required for digestion. The concentrations of analytes in the samples (minimum-maximum in μg g-1) were: Ca (100.71-462.20), Co (0.54-0.85), Cu (0.32-7.82), Fe (2.74-18.03), Mg (18.40-521.28), Mn (0.17-55.14), Ni (0.56-1.25) and Zn (2.96-20.92). The levels of Al, Ba, Cd, Cr, Mo, Pb, Se and V in all investigated samples were found to be below the limit of detection of ICP OES. The multielement analysis by ICP OES showed that the method is simple, fast and reliable for the multielement determination in medicinal plants and phytomedicines.

RESUMO Os efeitos do tempo da alimentação inicial sobre o ponto de não retorno (PNR) e o crescimento de larvas de jundiá Rhamdia voulezi foram estudados em condições laboratoriais. O experimento foi iniciado imediatamente após a completa absorção do saco vitelínico, 3 dias após a eclosão (DAE). O alimento foi fornecido pela primeira vez no 3o, 4o, 5o, 6o, 7o, 8o e 9o DAE. Zooplâncton e Artemia spp. recém-eclodida foram usados para a alimentação das larvas. Para avaliar os efeitos da privação alimentar sobre o crescimento, o comprimento padrão e peso de 594 larvas foram analisados. O crescimento larval foi significativamente afetado pelo tempo da primeira alimentação exógena. As larvas que receberam alimento do 3 o ao 5 o DAE apresentaram desenvolvimento significativamente maior que aquelas alimentadas mais tardiamente, a partir do 6 o DAE. As larvas alcançaram o PNR no 6 o DAE. A sobrevivência e o crescimento das larvas dependem do momento da alimentação inicial. Para evitar a mortalidade em massa e obter bom crescimento, as larvas de R. voulezi precisam estabelecer com sucesso a alimentação inicial até o 5 o dia após a eclosão.
ABSTRACT The effects of the timing of first feeding on the point-of-no-return (PNR) and growth of laboratory reared silver catfish Rhamdia voulezi larvae were studied. The experiment began immediately after the complete absorption of the yolk sac, 3 days after hatching (DAH). The food was provided for the first time on 3rd, 4th, 5th, 6th, 7th, 8th, and 9th DAH. Zooplankton and newly-hatched Artemia spp. were used to feed the larvae. To evaluate the effect of food deprivation on growth, the standard length and weight of 594 larvae were assessed. Larval growth was significantly affected by the time of first exogenous feeding. The larvae fed from 3rd to 5th DAH showed a significantly higher development than those fed from 6th DAH. The larvae reached the PNR on 6th DAH. Survival and larval growth strongly depended on the timing of initial feeding. In order to avoid potential mortality by starvation and obtain good growth, the silver catfish larvae R. voulezi must establish successful initial feeding within 5 days after hatching.

Abstract Germline TP53 mutations are associated with Li-Fraumeni syndrome (LFS), a disease that predisposes carriers to a wide variety of early onset tumors. In southern and southeastern Brazil, a high frequency of a germline TP53 mutation, p.R337H, was diagnosed in 0,3% of the population due to a founder effect. Carriers are at risk for developing cancer but the penetrance is lower than in typical DNA binding domain mutations. To date, only a few families were detected and diagnosis of carriers remains a challenge. Therefore, the inclusion of additional criteria to detect p.R337H carriers is necessary for the Brazilian population. We assessed the A.C. Camargo Cancer Center Oncogenetics Department database in search of common characteristics associated with p.R337H families that did not fulfill LFS/LFL clinical criteria. Among 42 p.R337H families, three did not meet any LFS/LFL criteria. All cases were young female patients with breast cancer diagnosed before age 45 and with no family history of LFS linked-cancers. Our results suggest that screening for the germline TP53 p.R337H mutation should be indicated, along with BRCA1 and BRCA2 genetic testing, for this group of patients, especially in the South and Southeast of Brazil.

RESUMO Este estudo descreveu o desenvolvimento inicial de Pseudoplatystoma reticulatum, bem como as mudanças nos padrões de crescimento, testando a hipótese da ocorrência de crescimento diferencial durante a ontogenia inicial da espécie. Variáveis morfométricas e merísticas foram analisadas durante os períodos embrionário, larval e juvenil, assim como os coeficientes de crescimento alométrico para larvas e juvenis. Os ovos apresentaram diâmetro médio de 1,79 mm e espaço perivitelino amplo (23,41%). O comprimento total (CT) das larvas variou de 3,05 a 125,72 mm e o número total de miômeros de 45 a 50 (pré-anal=13-16 e pós-anal=30-35). A pigmentação inicial é escassa, concentrada nas extremidades do saco vitelino. A partir do estágio de flexão a pigmentação se intensifica, formando uma faixa longitudinal que se estende do focinho ao opérculo e duas no corpo, uma dorsal e outra ventral, que se unem no pedúnculo caudal. Inicialmente há um crescimento diferenciado da cabeça e da cauda, com posterior crescimento do tronco, que demonstra mudanças nas prioridades ontogenéticas para as capacidades de alimentação, natação e respiração. Nossos resultados são de grande importância para a ecologia, sistemática e larvicultura, especialmente em termos de variação ontogenética na morfologia, crescimento, alimentação, comportamento e mortalidade P. reticulatum.
ABSTRACT This study described the initial development of Pseudoplatystoma reticulatum, as well as changes in growth patterns. Morphometric and meristic variables were analyzed during embryonic, larval and juvenile periods, even as allometric growth coefficients for larvae and juveniles. Eggs showed an average diameter of 1.79 mm and broad perivitelline space (23.41%). The total length (TL) of the larvae ranged from 3.05 to 25.72 mm, and the total number of myomeres ranged from 45 to 50 (preanal = 13-16 and postanal = 30-35). Initial pigmentation is poor, concentrated at the extremities of the yolk sac. In the flexion stage, the pigmentation intensifies and forms a longitudinal stripe extending from the snout to the operculum and two longitudinal stripes on the body, one dorsal and the other ventral, that unite at the caudal peduncle. At first, there is a differential growth of the head and tail, and then there is the growth of the body, which shows changes in the ontogenetic priorities of feeding, swimming, and breathing capacities. Our results are very important for ecology, systematics, and hatchering, especially in terms of ontogenetic variation in morphology, growth, feeding, behavior, and mortality of P. reticulatum.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Desenvolvimento embrionário, larval e juvenil de Hemisorubim platyrhynchos (Siluriformes, Pimelodidae) da bacia do rio Paraná

O objetivo deste estudo foi descrever o desenvolvimento embrionário, larval e juvenil da jurupoca, Hemisorubim platyrhynchos (Valenciennes, 1840), bem como as mudanças nos padrões de crescimento alométrico durante a ontogenia inicial da espécie. Um total de 90 ovos, 210 larvas e 24 juvenis provenientes de reprodução induzida foram analisados quanto a variáveis morfométricas e merísticas, além do coeficiente de crescimento alométrico em relação à cabeça, tronco e cauda durante o período larval e juvenil inicial. Os ovos apresentaram diâmetro médio de 1,74 mm, espaço perivitelino amplo (21,29%), com média de 0,37 mm, e diâmetro médio do vitelo de 1,08 mm. O comprimento padrão (CP) das larvas variou de 3,47 a 11,85 mm, com a maioria das medidas apresentando aumento proporcional ao longo do desenvolvimento. O número total de miômeros variou de 40 a 46 (pré-anal=15-17 e pós-anal=24-30). As larvas iniciais de H. platyrhynchos apresentam pigmentação na cabeça e na região ântero-ventral do corpo (anterior e posterior do saco vitelino). No estágio de pós-flexão, a pigmentação se intensifica, distribuindo-se na região dorsal da cabeça, formando uma faixa longitudinal que se estende do focinho ao opérculo, assim como uma faixa transversal, de um flanco a outro, passando pela região anterior da nadadeira dorsal, com máculas distribuídas ao longo do corpo nos juvenis (CP=19,5-49,09 mm). Nos primeiros estágios de desenvolvimento larval, a cabeça e a cauda crescem muito mais rapidamente do que o tronco, o que indica prioridades relacionadas à alimentação e natação, as quais posteriormente tendem à isometria, com um crescimento rápido do tronco nos juvenis iniciais.
This study aimed to describe the initial stages of development of the Pothole Shovelnose Catfish, Hemisorubim platyrhynchos (Valenciennes, 1840), as well as changes in the allometric growth pattern during early ontogeny. A total of 90 eggs 210 larvae, and 24 juveniles from hatching were analyzed for morphometric and meristic variables, and for besides the allometric growth coefficient in relation to head, trunk, and tail during larval and initial juvenile period. The eggs presented 1.74 mm of mean diameter; ample vitelline space (21.29%) with mean of 0.37 mm; and the average diameter of the yolk was 1.08 mm. The standard length (SL) of the larvae ranged from 3.47 to 11.85 mm, with most measurements a presenting proportional increase throughout development. The total number of myomeres ranged from 40 to 46 (pre-anal=15-17 and post-anal=24-30). The early larvae of H. platyrhynchos present pigmentation in the head and in the anteroventral region of the body (anterior and posterior regions of the yolk sac). The pigmentation intensifies in the post-flexion stage along the dorsal region of the head, forming a longitudinal strip extending from the snout to the operculum, as a transversal strip that goes from one side to the other, passing through the anterior region of the dorsal fin, with maculae distributed throughout the body in juveniles (SL=19.5-49.09 mm). In the early stages of larval development, the head and tail grow much faster than the trunk, indicating priorities related to food and swimming, which tend to isometry, with fast growth of the trunk in early juveniles.

Leishmanicidal and cholinesterase inhibiting activities of phenolic compounds of Dimorphandra gardneriana and Platymiscium floribundum, native plants from Caatinga biome

Nos últimos anos, o Ministério da Saúde do Brasil e a Organização Mundial da Saúde tem apoiado a investigação de novas tecnologias que possam contribuir para a vigilância, novos tratamentos e controle da leishmaniose visceral no país. Assim, o objetivo deste trabalho foi isolar compostos de plantas do bioma Caatinga, e investigar a toxicidade destes compostos contra as formas promastigotas e amastigotas de Leishmania infantum chagasi, principal parasita responsável pela leishmaniose visceral na América do Sul, e avaliar a sua capacidade para inibir a enzima acetil-colinesterase (AChE). Após a exposição aos compostos em estudo, foram realizados testes utilizando a forma promastigota que expressa luciferase e ELISA in situ para medir a viabilidade das formas promastigotas e amastigota, respectivamente. O ensaio colorimétrico MTT foi realizado para determinar a toxicidade destas substâncias utilizando células monocíticas murina RAW 264.7. Todos os compostos foram testados in vitro para as sua propriedade anti-colinesterásica. Um cumarina, escoparona, foi isolada a partir de hastes de Platymiscium floribundum, e os flavonóides, rutina e quercetina, foram isolados a partir de grãos de Dimorphandra gardneriana. Estes compostos foram purificados, utilizando cromatografia em coluna gel eluída com solventes orgânicos em misturas de polaridade crescente, e identificados por análise espectral. Nos ensaios leishmanicidas, os compostos fenólicos mostraram eficácia contra as formas extracelulares promastigotas, com EC50 para escoporona de 21.4µg/mL e para quercetina e rutina 26 e 30.3µg/mL, respectivamente. Os flavonóides apresentaram resultados comparáveis à droga controle, a anfotericina B, contra as formas amastigotas com EC50 para quercetina e rutina de 10.6 e 43.3µg/mL, respectivamente. Os compostos inibiram a enzima AChE com halos de inibição variando de 0,8 a 0,6cm, indicando um possível mecanismo de ação para a atividade leishmanicida.
In recent years, the Brazilian Health Ministry and the World Health Organization have supported research into new technologies that may contribute to the surveillance, new treatments, and control of visceral leishmaniasis within the country. In light of this, the aim of this study was to isolate compounds from plants of the Caatinga biome, and to investigate their toxicity against promastigote and amastigote forms of Leishmania infantum chagasi, the main responsible parasite for South American visceral leishmaniasis, and evaluate their ability to inhibit acetylcholinesterase enzyme (AChE). A screen assay using luciferase-expressing promastigote form and an in situ ELISA assay were used to measure the viability of promastigote and amastigote forms, respectively, after exposure to these substances. The MTT colorimetric assay was performed to determine the toxicity of these compounds in murine monocytic RAW 264.7 cell line. All compounds were tested in vitro for their anti-cholinesterase properties. A coumarin, scoparone, was isolated from Platymiscium floribundum stems, and the flavonoids rutin and quercetin were isolated from Dimorphandra gardneriana beans. These compounds were purified using silica gel column chromatography, eluted with organic solvents in mixtures of increasing polarity, and identified by spectral analysis. In the leishmanicidal assays, the compounds showed dose-dependent efficacy against the extracellular promastigote forms, with an EC50 for scoporone of 21.4µg/mL, quercetin and rutin 26 and 30.3µg/mL, respectively. The flavonoids presented comparable results to the positive control drug, amphotericin B, against the amastigote forms with EC50 for quercetin and rutin of 10.6 and 43.3µg/mL, respectively. All compounds inhibited AChE with inhibition zones varying from 0.8 to 0.6, indicating a possible mechanism of action for leishmacicidal activity.
