ABSTRACT. In soybeans, the integument or seed coat is an important modulator between the external and internal environment. It plays a fundamental role in seed vitality, and its lignin content may influence the seed quality. The objective of this experiment was to evaluate the quality of soybean seeds from a partial diallel and their reciprocals and its relationship with lignin content, seed coat thickness, and deposition location. The seeds were also evaluated for physiological quality through germination and accelerated aging tests. The lignin content was quantified by absorbance, and the integument thickness was analyzed and measured using scanning electron microscopy. The most contrasting cultivars for lignin content were analyzed using fluorescence microscopy and histochemical techniques. Cultivars and their reciprocals differed in seed physiological quality. We found differences in the genotypes for integument thickness. Using histochemical techniques, autofluorescence was observed in the same regions identified as lignified. Positive staining for lignin was observed in the hilum region. Among the genotypes, we found variations in the physiological quality of seeds (germination and accelerated aging test), lignin content, and integument thickness. Resumo: A seleção e utilização de genótipos de soja com sementes de alta qualidade fisiológica é importante para possibilitar maior probabilidade de estabelecimento e sucesso da cultura em campo, e, consequentemente, alcançar altas produtividades. Objetivou-se neste trabalho avaliar o controle genético da qualidade fisiológica de sementes de soja a partir de um dialelo parcial. Seis cultivares de soja, previamente selecionadas, grupo 1 (CD 201, CA 115, MS 8400) e grupo 2 (CD 202, Syn 1263, Syn 1279), foram intercruzadas utilizando-se um esquema de dialelo parcial, gerando 24 tratamentos. Sementes dessas cultivares e destes cruzamentos foram avaliadas quanto à qualidade fisiológica das sementes a partir de testes de germinação, primeira contagem de germinação, envelhecimento acelerado, emergência e índice de velocidade de emergência de plântulas. Foram avaliados os teores de lignina nos tegumentos das sementes de soja. Foram analisados os efeitos da capacidade geral (CGC) e específica de combinação (CEC) e os efeitos recíprocos. Há efeito significativo dos genes aditivos e não aditivos para os caracteres de qualidade de sementes, e efeito pronunciado de recíprocos, o que sugere a presença do efeito materno. Não há correlação entre o teor de lignina e testes fisiológicos para os genótipos estudados.

Abstract: The selection of soybean genotypes with seeds of high physiological quality is key to increasing the likelihood of establishment and success in the field and thus reaping higher yields. The aim of this study was to evaluate the genetic control of the physiological quality of soybean seeds from a partial diallel cross. Six previously selected soybean cultivars, group 1 (CD 201, CA 115, MS 8400) and group 2 (CD 202, Syn 1263, Syn 1279), were intercrossed by a partial diallel cross, totalizing 24 treatments. Seeds from these cultivars and crosses were evaluated for seed physiological quality based on germination tests, first germination count, accelerated aging, seedlings emergence, and emergence speed index. The lignin content in the soybean seed coat was evaluated. The effects on general and specific combining ability and reciprocal effects were analyzed. There were significant additive and non-additive effects of the genes on the seed quality traits and pronounced effects on the reciprocal traits, which suggest the presence of a maternal effect. Lignin content was not correlated with the physiological test results in the studied genotypes.

Resumo: Objetivou-se verificar a interferência do tratamento com inseticidas e fungicidas sobre a qualidade fisiológica de sementes de milho, classificadas em dois tamanhos, em diferentes períodos de armazenamento. Sementes do híbrido BM915 PRO, classificadas em dois tamanhos (CH20/64 e CH24/64), foram tratadas com calda caracterizada pela mistura de Carbendazim + Thiram (Derosal Plus®), Deltametrina (Actellic®), Pirimifós metílico (K-obiol®) e água (tratamento padrão). Além do tratamento padrão adotado pela empresa, foram acrescentados à mistura da calda tratamentos com clotianidina (Poncho®), tiametoxam (Cruizer®) e fipronil (Shelter®). As sementes foram armazenadas em ambiente não climatizado em embalagens de papel multifoliado. A qualidade fisiológica das sementes foi avaliada, a cada noventa dias, por um período de 270 dias por meio do teste de germinação, teste de frio e envelhecimento acelerado. Foi avaliada, ainda, a qualidade sanitária por meio do Blotter test. Concluiu-se que a qualidade fisiológica das sementes do híbrido BM915 PRO é mantida até os noventa dias de armazenamento, independente do tratamento químico utilizado e do tamanho das sementes. A adição do inseticida ao tratamento padrão utilizado no tratamento químico das sementes não afeta a ação dos fungicidas sobre os fungos presentes nestas.

Abstract: The aim of this study was to investigate the effect of treatment with insecticides and fungicides on the physiological quality of corn seeds, classified in sizes, in different periods of storage. Seeds of the hybrid BM915 PRO, classified in two sizes (CH20/64 and CH24/64), were treated with a mixture of carbendazim + thiram (Derosal Plus®), pirimiphos methyl (Actellic®), deltamethrin (K-obiol®), and water (standard treatment). In addition to the standard treatment adopted by the company, treatments with clothianidin (Poncho®), thiamethoxam (Cruizer®), and fipronil (Shelter®) were added to the spray mixture. The seeds were stored in a non-climate-controlled environment in multi-ply paper packages. The physiological quality of the seeds was evaluated every ninety days over a period of 270 days by the germination test, cold test, and accelerated aging. Sanitary quality was also evaluated through the Blotter test. The physiological quality of seeds of the hybrid BM915 PRO is maintained up to ninety days of storage, regardless of the chemical treatment used and the size of the seeds. The addition of the insecticide to the standard treatment used in chemical seed treatment does not affect the action of the fungicides on the fungi in the seeds.

ABSTRACT. The objective of this study was to evaluate gene expression related to water deficit tolerance in maize lines. For this, lines previously classified as tolerant (91-T and 32-T) and non-tolerant (24-NT and 57-NT) to water deficit were used. The seeds of the four lines were evaluated for emergence and emergence speed index, and the seedlings were evaluated for root and shoot length under two conditions of water availability (70 and 10% substrate water retention capacity). In transcript analysis, the expression of several genes associated with water deficit tolerance, ZmDBP3, ZmALDH9, ZmAN13, and ZmDREB2A, was evaluated by qRT-PCR for the 91-T and 57-NT lines. It can be concluded that soil water deficiency did not reduce root development. However, the shoot length was significantly lower under dry conditions. Through transcript analysis, the genes ZmDBP3 and ZmAN13 were identified as potential markers for the early selection of maize lines tolerant to water deficit.

RESUMO Para investigar se diferentes condições de armazenamento interferem no potencial fisiológico, sanitário e no teor de óleo de sementes de mamona (Ricinus communis) foram utilizadas sementes de duas cultivares, Guarani e IAC-80, armazenadas em dois ambientes (câmara fria e armazém convencional) em duas embalagens (saco de papel Kraft multifoliado e saco plástico - com e sem acondicionamento a vácuo a 1 atm). Testou-se ainda outro tipo de acondicionamento, utilizando-se papel aluminizado para criopreservação das sementes (-196 °C). A qualidade das sementes foi avaliada antes do armazenamento e após 4, 8 e 12 meses por meio dos testes de geminação (contagem aos 7 e 14 dias), emergência, sanidade, teor de água e teor de óleo. O delineamento experimental utilizado foi o inteiramente casualizado, em esquema fatorial 7x4, sendo sete condições de armazenamento e quatro épocas. A criopreservação (-196 °C) é a condição ideal para manutenção do potencial fisiológico de sementes de Ricinus communis, cultivares IAC-80 e Guarani, ao longo do armazenamento. Independente das condições de armazenamento de sementes de R. communis, o teor de óleo decresce e a incidência dos fungos Aspergillus spp. e Fusarium spp. aumenta ao longo do armazenamento.

ABSTRACT The objective of this work was to evaluate the effects of different storage conditions on the physiological and sanitary quality and oil content of castor bean (Ricinus communis) seeds. Seeds of castor bean plants of the Guarani, and IAC-80 cultivars were stored in two environments (cold room, and room conditions), using three package types (multifoliate Kraft paper bag, and polyethylene bag, and polyethylene bag with vacuum at 1 atm). In addition, another storage condition was evaluated: cryopreservation (-196 °C) in foil paper bags. Seed quality was evaluated before storage and at 4, 8, and 12 months after storage by testing their 7-day and 14-day germination, emergence, health, water content, and oil content. The experiment was conducted in a completely randomized design, with 7×4 factorial arrangement consisting of seven storage conditions and four evaluation times. Cryopreservation is the ideal condition for maintaining the seed physiological quality of the Ricinus communis cultivars used throughout storage. The oil content of the R. communis seeds decreases, and the incidence of Aspergillus spp. and Fusarium spp. fungi increases throughout storage, regardless of the storage conditions.

RESUMO O conhecimento de genes relacionados ao déficit hídrico é uma importante ferramenta visando auxiliar e acelerar o processo de seleção de genótipos de milho mais adaptados às condições de seca. Diante disso, objetivou-se neste trabalho avaliar a expressão de genes relacionados à tolerância ao déficit hídrico em linhagens de milho, por meio das análises proteômicas e transcriptômicas. Para isso, foram utilizadas duas linhagens previamente classificadas como tolerante (91-T) e não tolerante (57-NT) a baixa disponibilidade hídrica. Para as análises proteômicas, utilizaram-se sementes secas, ponta de espigas e plântulas coletadas com três, cinco e sete dias após a semeadura, sendo as plântulas submetidas a duas condições contrastantes quanto a disponibilidade hídrica (70% e 10% da capacidade de retenção de água). Avaliou-se a expressão das enzimas catalase, peroxidase, esterase e das proteínas resistentes ao calor por meio da técnica de eletroforese. Sementes secas, ponta de espigas e plântulas (coletadas sete dias após a semeadura, submetidas a duas condições de disponibilidade hídrica) de duas linhagens de milho (91-T e 57-NT) foram utilizadas para as análises transcriptômicas. A expressão dos genes associados a tolerância a baixa disponibilidade hídrica (ZmLEA3, AOX2 and ZmPP2C) foi realizada pela técnica de qRT-PCR. Pode-se concluir que houve maior expressão em ponta de espigas na linhagem 91-T para as enzimas catalase, peroxidase e esterase. Para as enzimas catalase e esterase, as plântulas de sete dias de desenvolvimento apresentaram uma redução da expressão, nas duas linhagens analisadas. Maior atividade das proteínas resistentes ao calor ocorreu nas sementes secas da linhagem 91-T. Identificou-se o gene AOX2 como um potencial marcador para seleção de genótipos de milho tolerantes ao déficit hídrico.

ABSTRACT Knowing genes related to water deficit is a valuable tool to assist and accelerate the selection process of maize genotypes more adapted to drought conditions. Therefore, this work aimed to evaluate genes expression related to tolerance to water deficit in maize lines, using proteomic and transcriptomic analyses. Two lines previously classified as tolerant (91-T) and non-tolerant (57-NT) to low water availability were used. Dried seeds, ears tips, and seedlings (collected at three, five, and seven days after sowing) of the two maize lines (91-T and 57-NT) were used in proteomic analyses. Seedlings were subject to two contrasting water-availability conditions (70% and 10% of the water retention capacity). The expression of catalase, peroxidase, and esterase enzymes and heat-resistant proteins were evaluated by electrophoresis. Dried seeds, ears tips, and seedlings (collected at seven days after sowing, subject to two contrasting water-availability conditions) of two maize lines (91-T and 57-NT) were used in the transcriptomic analyses. The expression of genes associated with tolerance to low water availability (ZmPP2C, ZmLEA3, and AOX2) was evaluated by the qRT-PCR technique. Results revealed that catalase, peroxidase, and esterase enzymes had a higher expression on the ears tips of the 91-T line. The expression of catalase and esterase enzymes reduced in seven-day-old seedlings in the two lines analyzed. Heat-resistant proteins had greater activity on the dried seeds of the 91-T line. The AOX2 gene was identified as a potential marker for the selection of maize genotypes tolerant to water deficit.

Abstract Harvest time is correlated to the seed physiological quality and can be determined through germination and vigor tests. X-ray is a good technique for assessing seed quality, since it can reduce analysis subjectivity, making the process faster and more efficient. Our aim was to evaluate jiló (Solanum gilo) seed internal free area, through X-ray image analysis, and to relate results with seed germination in different stages of maturation. The variables evaluated were: object area, perimeter, standard deviation, and fraction area, which represents the internal free areas of seeds. The experimental design was completely randomized and treatments for each variety consisted of fruits harvested at: 35 d after anthesis (DAA); 40 DAA; 45 DAA and 45 DAA plus 7 d at rest in dark room. Four replicates of 50 seeds were used for each treatment. Data from germination test and seeds image analysis were used for ANOVA analysis, and treatments means were compared with the Scott-Knott test (p≤0.05). Jiló seeds came from Morro redondo and Tinguá varieties. Jiló seeds from each treatment were analyzed by X-ray test and subsequently led to germination test. X-ray images were analyzed using ImageJ software, which was efficient. There were no statistically significant differences in each maturation stage between varieties. However, there was a statistically significant difference in seeds free areas of both varieties. Seeds with free area (embryo - endosperm) ≤ 10 % produced normal seedlings, and harvest at 45DAA + 7 d was recommended and germination seeds was 100%. Thus, adiographic image analysis of eggplant seeds allows measurement of internal free areas, as well as determination of the relationship between these and germination, allowing identification of seeds with greater potential to germinate, helping to reduce jiló seeds economic losses.

Resumen El momento de cosecha se correlaciona con la calidad fisiológica de la semilla y puede definirse a través de la germinación y las pruebas de vigor. Los rayos X son una buena técnica para evaluar la calidad de la semilla, ya que pueden reducir la subjetividad del análisis, volviendo el proceso más rápido y más eficiente. Nuestro objetivo fue evaluar el área libre interna de la semilla de jiló (Solanum gilo), a través del análisis de la imagen de rayos X, y relacionar los resultados con la germinación de la semilla en distintas etapas de maduración. Las variables evaluadas fueron: área del objeto, perímetro, desviación estándar, y área de fracción, que representa las áreas libres internas de las semillas. El diseño experimental fue completamente aleatorio y los tratamientos para cada variedad fueron frutos cosechados: 35 d después de la antesis (DDA); 40 DDA; 45 DDA; y 45 DDA más 7 d en descanso en un cuarto oscuro. Cuatro réplicas de 50 semillas se utilizaron para cada tratamiento. Los datos de las pruebas de germinación y el análisis de imágenes de semillas se usaron para realizar un análisis de ANOVA, y las medias de los tratamientos se compararon con la prueba Scott-Knott (p ≤ 0.05). Las semillas de jiló (berenjena) provinieron de las variedades Morro Redondo y Tinguá. Las semillas de jiló de cada tratamiento se analizaron con la prueba de rayos X y despues se realizó la prueba de germinación. Las imágenes de rayos X se analizaron usando el programa ImageJ, que fue eficiente. No hubo diferencias estadísticamente significativas en cada etapa de maduración entre las variedades. Sin embargo, se encontró una diferencia estadísticamente significativa en áreas libres de las semillas de ambas variedades. Las semillas con área libre (embrión-endospermo) ≤ 10 % produjeron plántulas normales, y se recomienda la cosecha a 45 DDA + 7 d cuando la germinación de semillas fue de 100 %. Por lo tanto, el análisis de imagen adiográfico de semillas de jiló permite la medición de áreas libres internas, así como la definición de la relación entre las mismas y la germinación, permitiendo identificar las semillas con mayor potencial para germinar, y ayudando a reducir las pérdidas económicas de semillas de jiló.

RESUMO. As sementes da cultura do gergelim contém cerca de 50% de teor de óleo, sendo uma das principais razões que estimulam o seu cultivo. No entanto, informações sobre metodologias para avaliação da qualidade de sementes dessa cultura são escassas. Dessa forma, objetivou-se adequar as metodologias dos testes de envelhecimento acelerado para sementes de gergelim e investigar alterações no comportamento de isoenzimas das sementes submetidas ao teste. Foram utilizados quatro cultivares de sementes de gergelim da safra 2014/2015. Foi feita a caracterização fisiológica das sementes e a análise eletroforética das isoenzimas superóxido dismutase, esterase, catalase, álcool desidrogenase, malato desidrogenase e isocitrato liase. Para o teste de envelhecimento acelerado, as sementes foram submetidas ao método tradicional e com solução saturada de NaCl, pelos períodos de envelhecimento de 0, 24, 48, 72 e 96 horas. Concluiu-se que é possível avaliar o vigor de sementes de gergelim pelo teste de envelhecimento acelerado, pelo método tradicional por 72 horas e com solução saturada de NaCl a 45ºC por 48 horas. Quando associado à atividade das isoenzimas EST, CAT, ADH e ICL foi possível visualizar que houve variação significativa na intensidade da expressão das bandas conforme avança o processo deterioração das sementes.

ABSTRACT. The seeds of a sesame crop contain approximately 50% oil content, one of the main reasons for their cultivation. However, information regarding methodologies for the assessment of quality seeds of this culture are scarce. Therefore, the objective of this study was to adapt the methods of accelerated ageing tests for sesame seeds and investigate changes in the behaviour of isoenzymes. The physiological characterization of the seeds and electrophoretic analysis of the isoenzymes superoxide dismutase, esterase, catalase, alcohol dehydrogenase, malate dehydrogenase and isocitrate lyase were also performed. For the accelerated ageing test, the seeds were subjected to the traditional method, with a saturated NaCl solution, at ageing times of 0, 24, 48, 72, and 96 hours. It was found that it is possible to evaluate the effect of accelerated ageing tests on sesame seeds by using the traditional method for 72 hours and saturated NaCl solution at 45°C for 48 hours. When associated with the activity of isozymes EST, CAT, ADH, and ICL, it was possible to observe that there was a significant variation in the intensity of expression of the bands as the seed deterioration process progressed.

Resumo: Objetivou-se avaliar os efeitos de diferentes condições de armazenamento sobre a qualidade de sementes de mamona cultivar IAC-226, armazenadas por um período de 12 meses. Para isso, as sementes foram armazenadas em diferentes ambientes e tipos de embalagens: em câmara fria e armazém convencional, utilizando embalagens papel Kraft multifoliado e plástico com e sem acondicionamento a vácuo a 1 atm; e também armazenamento sob criopreservação (-196 °C). A qualidade das sementes foi avaliada antes e após 4, 8 e 12 meses de armazenamento pelos testes de geminação, primeira contagem de germinação, porcentagem de emergência, índice de velocidade de emergência e pela determinação das alterações nos sistemas enzimáticos catalase (CAT) e superóxido dismutase (SOD). A criopreservação (-196 °C) é eficiente na manutenção da qualidade fisiológica de sementes de mamona cultivar IAC-226 por 12 meses. A enzima catalase se destaca como um marcador da deterioração de sementes de mamona durante o armazenamento.

Abstract: The present study aimed to evaluate effects of different storage conditions on the castor bean seed cultivar IAC-226 quality, stored for 12 months. For this purpose, seeds were stored in different environment and packaging conditions: in a cold chamber and conventional storage, using multiwall Kraft paper and in no vacuum and vacuum plastic packages at 1 atm; as well as under cryopreservation storage (-196 °C). Seed quality was evaluated before and after 4, 8 and 12 months of storage by germination tests, first count of germination, emergence percentage, emergence speed index and determination of changes in catalase (CAT) and superoxide dismutase (SOD) enzyme systems. Cryopreservation (-196 °C) is efficient in maintaining the physiological quality of castor bean cultivar IAC-226 for 12 months. The enzyme catalase stands out as a marker of castor seed deterioration during storage.

Resumo O objetivo nesse trabalho foi avaliar a melhor época para a realização da colheita e verificar o efeito do armazenamento dos frutos na atividade de proteínas e na qualidade fisiológica de sementes do híbrido de abóbora 'Jabras'. Os frutos foram colhidos aos 15, 30, 45, 60 e 75 dias após a antese (DAA). Em cada época, foram colhidos trinta frutos, sendo que quinze frutos tiveram suas sementes extraídas imediatamente após a colheita e os outros quinze frutos foram armazenados por vinte dias em caixas plásticas em condições de galpão, e somente após este período, tiveram suas sementes extraídas. Avaliou-se a qualidade das sementes por meio do grau de umidade, germinação, primeira contagem, índice de velocidade de geminação, emergência, índice de velocidade de emergência, massa de 1000 sementes e massa seca de plântulas, além da análise do perfil eletroforético de proteínas resistentes ao calor (Late Embryogenesis Abundant - LEA). A maturidade fisiológica das sementes ocorreu em sementes oriundas de frutos colhidos aos 60 dias após a antese e armazenados por 20 dias. Nesse período, as sementes encontravam-se com o máximo de matéria seca, máxima germinação e vigor e alta concentração de proteínas LEA.

This study has aimed to evaluate the best time to harvest squash seeds and verify the effect of fruit storage on protein activity and in the physiological quality of 'Jabras' squash hybrid seeds. The fruits were harvested at 15, 30, 45, 60 and 75 days after anthesis (DAA). In every period, thirty fruits were harvested and fifteen had their fruit extracted immediately after harvesting, and the other fifteen fruits were stored for twenty days in plastic boxes under shade conditions, and only after this period the seeds were extracted. Seed quality was evaluated for the following parameters: moisture content, germination, first count, germination rate, seedling emergence, emergence rate, seed mass and seedling dry matter. Also, the electrophoretic profile analysis of heat-resistant proteins (Late Embryogenesis Abundant - LEA) was performed. Seed physiological maturity occurred in fruits harvested at 60 days after anthesis and stored for 20 days. During this period, seeds reached the maximum dry matter, maximum germination and vigor, and a high concentration of LEA proteins.

RESUMOA duplicação cromossômica artificial está dentre as etapas mais importantes na obtenção de duplo-haploides em milho. Este estudo objetivou avaliar a capacidade de indução da linhagem indutora KEMS em clima tropical e testar a eficiência do marcador R1-navajo por meio de citometria de fluxo; avaliar dois protocolos de duplicação cromossômica e, analisar o desenvolvimento dos haploides duplicados no campo. Para isso, quatro genótipos (gerações F1 e F2) foram cruzados com a linhagem KEMS. As sementes obtidas foram selecionadas pelo marcador R1-navajo e submetidas a dois protocolos de duplicação cromossômica. A duplicação foi confirmada por meio de citometria de fluxo. As porcentagens de plantas autofecundadas após duplicação foram analisadas, bem como as quantidades de sementes duplo-haploides obtidas após as autofecundações. Foi observado que o germoplasma influencia a indução de haploides, mas não na taxa de duplicação dos protocolos testados. O protocolo 2 foi mais eficiente na duplicação de haploides, na porcentagem de plantas autofecundadas após duplicação, e na obtenção de linhagens duplo-haploides. Além disso, a linhagem indutora KEMS pode induzir haploides em clima tropical. Outros marcadores além do sistema do R1-navajo devem ser utilizados na seleção de sementes haploides.

ABSTRACTArtificial chromosome duplication is one of the most important process in the attainment of doubled haploids in maize. This study aimed to evaluate the induction ability of the inducer line KEMS in a tropical climate and test the efficiency of the R1-Navajo marker by flow cytometry to evaluate two chromosome duplication protocols and analyze the development of the doubled haploids in the field. To accomplish this goal, four genotypes (F1 and F2 generations) were crossed with the haploid inducer line KEMS. The seeds obtained were selected using the R1-Navajo marker and subject to two chromosome duplication protocols. Duplication was confirmed using flow cytometry. The percentages of self-fertilized plants after duplication as well as the quantities of doubled haploid seeds obtained after the self-fertilization processes were analyzed. It was observed that the germplasm influences haploid induction but not the duplication rates of the tested protocols. Protocol 2 was more efficient for the duplication of haploids, in the percentage of self-fertilized plants after duplication, and in the attainment of doubled haploid lines. Moreover, the haploid inducer line KEMS can produce haploids in a tropical climate. Other markers, in addition to the R1-Navajo system, should be used in the selection of haploid seeds.

O trabalho teve como objetivo investigar a tolerância a dessecação em sementes de Rhamnidium elaeocarpum Reissek por meio de alterações fisiológicas e bioquímicas. Os frutos foram coletados no município de Jaupaci, Estado de Goiás, quando as sementes apresentavam teor de água de 37% base úmida (b.u.), sendo em seguida mantidas em estufa de secagem com circulação de ar à temperatura de 35ºC até alcançarem os teores de água de 20 e 12% (b.u.). Foram avaliados os seguintes parâmetros: germinação; índice de velocidade de germinação; emergência; índice de velocidade de emergência; condutividade elétrica; taxa respiratória; perfil eletroforético das isoenzimas: catalase, peroxidase e esterase; atividade enzimática de endo-β-mananase e α-amilase e análise eletroforética de proteínas resistentes ao calor. Maior velocidade de germinação e taxa respiratória foi obtida para sementes com teor de água de 12% b.u. A atividade das isoformas de catalase foi mais pronunciada em sementes com maior grau de desidratação em relação à peroxidase que diminuiu sua atividade. As sementes apresentaram proteínas de baixo peso molecular resistentes ao calor, variando de 48,7 a 13,2 kDa. A aquisição de tolerância à secagem em sementes de R. elaeocarpum Reissek é mais associada à enzima catalase do que às enzimas esterase e peroxidase.

This study aimed to investigate the desiccation tolerance of Rhamnidium elaeocarpum Reissek seeds through physiological and biochemical alterations. Fruit were collected from the municipality of Jaupaci, Goias State, Brazil, when the water content of their seeds had a 37% wet basis (w.b.) and the seeds were subsequently kept in a drying oven with air circulation at a temperature of 35ºC until a water content was reached of 20 to 12% (w.b.). The following parameters were evaluated: germination, germination speed index, emergence, emergence speed index, electrical conductivity and respiration rate. Furthermore, the electrophoretic profile of the isoenzymes: catalase, peroxidase and esterase, the enzymatic activities of endo-β-mannanase and α-amylase and the electrophoretic analysis of heat-resistant proteins were performed. A higher germination speed and respiratory rate was obtained for seeds with a water content of 12% w.b., and the activity of catalase isoforms was more pronounced in seeds with a higher degree of dehydration, which was in contrast to peroxidase, showed decreased activity. The seeds contained heat-resistant proteins of low molecular weight that ranged from 48.7 to 13.2 kDa. We found that the acquisition of drying tolerance in R. Elaeocarpum Reissek seeds is associated more with catalase than with esterase and peroxidase.

Sementes de ipê-amarelo são consideradas ortodoxas e dispõem de mecanismos que auxiliam na tolerância à dessecação, como proteínas resistentes ao calor e sistemas de proteção contra radicais livres. Para investigar o efeito da secagem na qualidade das sementes de ipê-amarelo coletadas com 28% de teor de água utilizaram-se dois métodos: secagem lenta (30 ºC) e secagem rápida (45 ºC) até atingirem teores de água na massa de sementes iguais a 20%, 15%, 10% e 5%. A qualidade das sementes foi avaliada por meio de testes de germinação e vigor. Foi avaliada a expressão das proteínas resistentes ao calor e das enzimas catalase, esterase e isocitrato liase. A expressão dos genes sHSP17.5, CAT3 e ICL6 foi quantificada por meio da técnica de qRT-PCR. Maiores valores de germinação e vigor foram obtidos em sementes com teor de água de 20%, submetidas à secagem rápida. Houve a indução de expressão de proteínas resistentes ao calor nos dois métodos de secagem. Ocorreu variação no padrão eletroforético das enzimas catalase, esterase e isocitrato liase entre os diferentes tratamentos. A expressão de genes varia em função dos métodos de secagem e teores de água das sementes. A secagem até os teores de água entre 15 e 10% compromete a qualidade fisiológica das mesmas.

Handroanthus serratifolius seeds are considered orthodox and have some mechanisms that assist in desiccation tolerance, like heat-resistant proteins and the presence of protective systems against free radical production. In order to investigate the drying effect on the quality of Handroanthus serratifolius seeds collected with 28% water content two methods were used: slow drying (30 °C) and fast drying (45 °C) until water contents equaled 20%, 15%, 10% and 5%. Seed quality was assessed by germination test, first count of germination, speed germination, emergence and speed emergence. The expression of heat-resistant proteins and enzymes catalase, esterase and isocitrate lyase was assessed by electrophoresis. The gene expression of sHSP17.5, CAT3 and ICL6 was quantified by real time PCR. Higher values of germination and vigor were obtained in seeds with 20% water content, subjected to fast drying. There was induction of heat-resistant proteins expression for both drying methods. There was variation in the electrophoretic pattern of catalase, esterase and isocitrate lyase in seeds subjected to different treatments. The gene expression in Handroanthus serratifolius seeds varies depending on methods of drying and water content of seeds. The drying of Handroanthus serratifolius seeds until the water content between 15 and 10% compromises their physiological quality.

O nabo-forrageiro tem ganhado importância no mercado brasileiro como adubo verde, planta de cobertura e pelo potencial para produção de biodiesel, no entanto estão registradas apenas duas cultivares que são morfologicamente muito semelhantes. A determinação da pureza genética é um dos requisitos para comercialização de sementes, pois garante a uniformidade de plantio, e é importante para os programas de melhoramento. Por tanto, objetivou-se com este trabalho avaliar a similaridade e padrões genotípicos que permitam diferenciar as cultivares comerciais de nabo-forrageiro 'IPR 116' e 'CATI AL-1000'. Para isso, foi analisado o padrão eletroforético de isoenzimas em sementes secas, sementes embebidas, folhas de plântulas e folhas jovens. As isoenzimas usadas foram: superóxido dismutase, catalase, esterase, glutamato - oxilacetato, malato desidrogenase e isocitrato liase. Foram analisados 37 primers RAPD e 10 primers ISSR em folhas das cultivares 'CATI AL-1000' e 'IPR-116'. Entre as isoenzimas estudadas, as mais polimórficas foram glutamato - oxilacetato, malato desidrogenase, esterase e superóxido dismutase, tendo o sistema superóxido dismutase realizado a melhor caracterização para todos os estádios de desenvolvimento. O sistema isoenzimático catalase não permitiu distinguir as cultivares em nenhum estádio de desenvolvimento e a isocitrato liase não revelou com o protocolo utilizado. Na análise dos marcadores 27 primers RAPD e oito primers ISSR apresentaram polimorfismo. Os resultados indicam que há possibilidade de determinar descritores confiáveis com base em isoenzimas em diferentes estádios de desenvolvimento de nabo-forrageiro e com uso de primers RAPD e ISSR.

The radish has been gaining importance in the Brazilian market due to its use as a green manure and cover crop, and also due to its potential for the production of biodiesel, however there are only two registered cultivars, which are morphologically very similar. Determination of genetic purity is a prerequisite in marketing seeds as it ensures uniformity of the crop and is important in breeding programs. The aim of this work therefore was to evaluate the similarity and genotype patterns which permit differentiation of the commercial radish cultivars IPR 116 and CATI AL-1000. In order to do this, isoenzyme electrophoretic patterns were analysed in dry seeds, soaked seeds, seedling leaves and young leaves; the isoenzymes used being: superoxide dismutase, catalase, esterase, glutamate-oxalocetate, malate dehydrogenase and isocitrate lyase. Thirty-seven RAPD primers and 10 ISSR primers in leaves of the CATI AL-1000 and IPR-116 cultivars were analysed. Among the isoenzymes under study, the most polymorphic were glutamate oxalocetate, malate dehydrogenase, esterase and superoxide dismutase, with the superoxide dismutase system giving the best characterisation for all stages of development. The catalase isoenzyme system did not make it possible to differentiate between cultivars at any stage of development, and isocitrate lyase was not revealed by the protocol used. In analysis of the markers, 27 RAPD primers and eight ISSR primers showed polymorphism. The results indicate that it is possible to determine reliable descriptors based on isoenzymes at different stages of development of the radish and with the use of RAPD and ISSR primers.

A semente do cafeeiro agrega toda a tecnologia desenvolvida pelos programas de melhoramento, porém, após o processamento e secagem, perdem a viabilidade em curto período de tempo, dificultando o seu armazenamento. Assim, objetivou-se com a pesquisa avaliar a qualidade de sementes de cafeeiro submetidas a diferentes métodos de secagem e ao armazenamento. Sementes de cafeeiro foram secadas até atingirem os teores de 40, 20, 12 e 5% de água por meio de dois métodos de secagem: lenta à sombra e rápida em secador mecânico. Após este processo, as sementes foram armazenadas em câmara fria por doze meses, sendo a qualidade, avaliada antes e ao final do armazenamento pelo teste de germinação, padrão eletroforético de proteínas resistentes ao calor e atividade das enzimas catalase, peroxidase, superóxido dismutase. A secagem lenta a sombra até 20% de teor de água propicia a conservação da qualidade em sementes de cafeeiro por até doze meses de armazenamento.

The technology developed by breeding programs is applied to coffee seeds; however, after processing and drying, they lose viability within a short period of time, thus making storage unsuitable. The objective of this research was to evaluate the quality of coffee seeds submitted to different drying methods and moisture contents during storage. The coffee seeds were submitted to conventional drying (slow shade drying) and fast drying in a static drier until they reached a moisture content of 40, 20, 12 and 5%. After this process, the seeds were stored in a cold chamber for 12 months, and seed quality was evaluated before and during storage by the germination test, electrophoretic patterns of heat resistant proteins, and the activity of isoenzyme systems. Conventional drying (slow shade drying) at 20% of moisture content maintains coffee seed quality until 12 months of storage.
