Cancer treatment represents one of the main scientific study targets today, mainly due to the pronounced side effects arising from chemotherapy. This study reports the synthesis, characterization, and anticancer activity of ten compounds from the Morita-Baylis-Hillman reaction. Ethylene glycol diacrylate was used as a double Michael acceptor in reactions with isatin derivatives to give homodimers of 3-hydroxyindolin-2-one core, recognized in the literature for its extensive pharmacological profile. The use of 1,4-diazabicyclo[2,2,2]octane (DABCO) as a catalyst and room temperature were the optimal conditions for the study reaction. The isolated yields were up to 63%, with most reaction times inferior to 24 h, some as fast as 15 min. The anticancer potential of the synthesized dimers was evaluated in vitro against three cancer strains, resulting in average inhibitory concentrations up to 0.72 µM. It was also found that the best performing homodimers are more active than their monomeric counterparts. Considering the promising selectivity indices observed, the preliminary results obtained here act as a basis for broader tests regarding the effectiveness of homodimeric adducts against cancer cells.

This investigation describes the synthesis of eugenol analogs presenting 1,2,3-triazole fragments and evaluation of their antileishmanial activity. The alkylation of guaiacol (1) with allyl bromide afforded 1-(allyloxy)-2-methoxybenzene (2) (93% yield). The Claisen rearrangement conducted with 1 gave ortho eugenol (3) (82% yield). Alkylation procedures performed with 3 produced 1-allyl-3-methoxy-2-(prop-2-yn-1-yloxy)benzene (4) (73% yield) and 1-allyl-3-methoxy-2-(pent-4-yn-1-yloxy)benzene (6) (53% yield). The copper(I)-catalyzed alkyne-azide cycloaddition (CuAAC) reactions involving alkynes 4 and 6 with different benzylic azides afforded twenty-two eugenol analogs with 1,2,3-triazole functionalities (48-93% yield). We screened the compounds at 10 μmol L 1 against Leishmania braziliensis intracellular amastigotes during macrophage infection. The action of these compounds was compared with the known leishmanicidal drug amphotericin B. None of the analogs were toxic to macrophages at 10 μmol L-1. The cytotoxic concentration at 50% (CC50), effective concentration at 50% (EC50), and selectivity index (SI) were determined to the best compounds 4-((2-allyl-6-methoxy)phenoxymethyl)-1-(4-chlorobenzyl)-1 H-1,2,3-triazole (8c) and 4-((2-allyl-6-methoxy)phenoxymethyl)-1-(4-trifluoromethoxybenzyl)-1 H-1,2,3-triazole (8h). They showed a significant leishmanicidal effect, with EC50 of 28.09 µmol L-1 (8c) and 52.03 µmol L-1 (8h). The SIs were 9.7 for 8c and > 5.7 for 8h. These compounds have the potential as new leishmanicidal agents against L. braziliensis and may represent a starting point for the development of alternative treatments for cutaneous leishmaniasis.

A exposição excessiva à radiação solar (RS) é o principal causador do câncer de pele. O objetivo desse trabalho foi investigar a percepção de estudantes do ensino médio sobre RS e suas implicações à saúde humana. Os dados foram coletados através de questionários e as respostas foram avaliadas por tabelas de frequências relativa, absoluta e percentual. Devido ao tamanho da amostra, utilizamos o teste de Fisher a 5% de significância (e hipótese alternativa bilateral) para correlacionar as respostas. Os alunos, independente de terem estudado ou não sobre RS não sabem definir RS e não têm percepção sobre os efeitos da RS à saúde humana. Este diagnóstico mostra, por um lado, a carência conceitual dos estudantes. Por outro, ele sugere também a ineficácia dos recursos didáticos convencionais e meios de informação acessíveis. Portanto, a inserção deste conceito na educação básica é primordial dada sua relevância social e por se tratar de um problema de saúde pública.
The excessive exposure to solar radiation (SR) is the main cause of skin cancer. The main aim of this paper was to investigate the perception of high school students about the SR and its implications for human health. The data have been collected through questions and the answers were evaluated by tables of relative, absolute and percentual frequency. Due to the size of our sample, we have used the Fischer test at 5% of significance (and bilateral alternative hypothesis) to correlate the answers. The students, whether they studied or not SR, don’t know to define SR and don’t have perception about its effects on the human health. This diagnosis shows, on the one hand, the conceptual lack of students. On the other, it also suggests the ineffectiveness of conventional didactic resources and accessible information media. Thus, the insertion of this concept in basic education is essential due to its social relevance and because it’s a public health problem.

Resumo Pouco se sabe se a morte celular induzida pela terapia fotodinâmica (PDT) compromete os macrófagos (MΦ), envolvidos nas respostas imunes induzidas pela PDT. Neste estudo, foram avaliados parâmetros de citotoxicidade dos MΦ mediada pela PDT e a produção de citocinas, frente à protoporfirina IX (PpIX). MΦ peritoneais de camundongos BALB/c foram estimulados in vitro com PDT, luz, PpIX ou lipopolissacarídeo (LPS). Após isto, a viabilidade celular (VC), a lipoperoxidação, os níveis de óxido nítrico (NO), de DNA degradado, de TNF-α, IL-6 e IL-10 foram avaliados. A exposição curta à PDT reduziu a VC em 10-30%. Os níveis de NO e de DNA degradado duplicaram, sem aumento da lipoperoxidação. Houve aumento de TNF-α e IL-10, sendo maior na presença de LPS. Já a produção de IL-6 reduziu em dez vezes. A PDT induz estresse celular, gera radicais NO e causa dano ao DNA, tornando o microambiente citotóxico. Ainda, modula citocinas pró e anti-inflamatórias em MΦ.
Abstract Little is known regarding whether photodynamic therapy (PDT)-induced cell death can substantially compromise macrophages (MΦ), which are important cells in PDT-induced immune responses. Here, parameters of PDT-mediated MΦ cytotoxicity and cytokine production in response to protoporphyrin IX (PpIX) were evaluated. Peritoneal MΦ from BALB/c mice were stimulated in vitro with PDT, light, PpIX, or lipopolysaccharide (LPS). After that, cell viability, lipid peroxidation, Nitric Oxide (NO), DNA damage, TNF-α, IL-6 and IL-10 were evaluated. Short PDT exposure reduced cell viability by 10–30%. There was a two-fold increase in NO and DNA degradation, despite the non-increase in lipoperoxidation. PDT increased TNF-α and IL-10, particularly in the presence of LPS, and decreased the production of IL-6 to 10-fold. PDT causes cellular stress, induces NO radicals and leads to DNA degradation, generating a cytotoxic microenvironment. Furthermore, PDT modulates pro- and anti-inflammatory cytokines in MΦ.

Abstract Identification of stagnant regions of viscoplastic fluid flows in production lines and equipment is of paramount importance owing to potential material degradation and process contamination. The present work introduces an assessment strategy to identify, classify and quantify unyielded regions with the objective of optimizing the flow conditions with the purpose of minimizing stagnant regions. Flow of Carbopol® 980 in a T-bifurcation channel is adopted to illustrate the procedure. The rheological behavior of Carbopol® 980 was simulated using the Herschel-Bulkley viscoplastic model regularized by Papanastasiou’s exponential approach. The analysis shows that three distinct types of stagnant unyielded regions take place in the bifurcation channel depending upon the Reynolds condition. Furthermore, the rheological characteristics of the fluid indicate the existence of an ideal Reynolds condition which allows the smallest flow stagnant area at the bifurcation zone.

The structures of two methoxylated chalcones, namely (E)-1-(4-methoxyphenyl)-3-(3,4,5-trimethoxyphenyl)prop-2-en-1-one and (E)-3-(4-ethoxyphenyl)-1-(4-methoxyphenyl)prop-2-en-1-one, reveal the effect of the inclusion of the methoxyl and ethoxyl substituents of the conformation on methoxy-chalcone. Structural comparative study between two chalcones was done in this work and some effects on geometric parameters, such as planarity and dihedral angles, were described. In addition, intermolecular interactions responsible for crystalline packaging were investigated by Hirshfeld surfaces and the values of those interactions were analysed by comparing experimental and theoretical models. The molecular stability was expressed in terms of softness and hardness, both obtained from frontier molecular orbitals. Finally, there is a good agreement between calculated and experimental infrared spectrum, which allowed the assignment of the normal vibrational modes.

Integrated furfural and first generation bioethanol production: process simulation and techno-economic analysis

Abstract Furfural is a base chemical with a wide range of applications and with a great opportunity for market growth in the near term. Derived from biomass, its production may be incorporated to the Brazilian chemical industry using sugarcane bagasse as feedstock. In this context, the integration of a furfural plant to a first generation bioethanol facility, within the biorefinery concept, was simulated considering different scenarios compared to an autonomous bioethanol distillery. The economic analysis of the different scenarios showed that the revenues from furfural commercialization increase the internal rate of return of the project for maximum furfural production (22.0%) in comparison to a conventional ethanol distillery (13.5%), despite the decrease in electricity output. Moreover, the economic analysis of the results pointed out the possibility of lowering furfural prices to levels that could lead to its use as a precursor for biofuels.

O foco de análise do presente estudo é a assimetria que a líquida lateral apresenta nas posições de onset e de coda de sílaba, uma vez que mostra estabilidade fonética no onset e variação na posição de coda. Como o funcionamento da lateral pré e pós-vocálica apresenta diferenças no português e no espanhol, este estudo elegeu, como objeto de análise, o português dos Campos Neutrais, na fronteira Brasil-Uruguai, por seu contato com o espanhol. A base empírica é o português dos Campos Neutrais, emprestada de Espiga (2001), já que, segundo o autor, nessa região a lateral em posição de onset se manifesta como alveolar, enquanto pode assumir cinco formas fonéticas na posição de coda: alveolar, velarizada, labializada, vocalizada e zero fonético. A explicitação e a formalização da assimetria da líquida lateral são propostas com base na Teoria da Otimidade Estocástica, por meio de restrições. Com base no mecanismo de alinhamento relacional, o presente estudo propõe restrições de distância de sonoridade inspiradas em Gouskova (2004), dela diferenciando-se por ter a particularidade de considerar a relação entre o onset e o núcleo e entre o núcleo e a coda de sílaba.
The focus of this study is the asymmetry of the lateral liquid in syllable onset and coda positions, since it shows phonetic stability in onset position and variation in coda position. Taking into account that the pre- and postvocalic laterals perform differently in Portuguese and in Spanish, this study aims at analyzing the Portuguese spoken in the Campos Neutrais, a region on the Brazil-Uruguay border, because of its contact with Spanish. Its empirical basis, which was taken from Espiga (2001), is the Portuguese spoken in the Campos Neutrais, since this author states that, in this region, the lateral in onset position performs as an alveolar whereas it may take five phonetic forms in coda position, i.e., alveolar, velar, labial, glide and phonetic null. The explanation and the formalization of the asymmetry of the lateral liquid have been based on the Stochastic Optimality Theory (StOT), through constraints. Having the mechanism of relational alignment as its background, this study proposes sonority distance constraints adapted from Gouskova (2004). However, differences lie in the fact that this study takes into account the relation between the onset and the nucleus, besides the one between the nucleus and the syllable coda.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The diversity, extracellular enzymatic activities and photoprotective compounds of yeasts isolated in Antarctica

The diversity of yeasts collected from different sites in Antarctica (Admiralty Bay, King George Island and Port Foster Bay and Deception Island) and their ability to produce extracellular enzymes and mycosporines were studied. Samples were collected during the austral summer season, between November 2006 and January 2007, from the rhizosphere of Deschampsia antarctica, ornithogenic (penguin guano) soil, soil, marine and lake sediments, marine water and freshwater from lakes. A total of 89 isolates belonging to the following genera were recovered: Bensingtonia, Candida, Cryptococcus, Debaryomyces, Dioszegia, Exophiala, Filobasidium, Issatchenkia (Pichia), Kodamaea, Leucosporidium, Leucosporidiella, Metschnikowia, Nadsonia, Pichia, Rhodotorula, and Sporidiobolus, and the yeast-like fungi Aureobasidium, Leuconeurospora and Microglossum. Cryptococcus victoriae was the most frequently identified species. Several species isolated in our study have been previously reported to be Antarctic psychophilic yeasts, including Cr. antarcticus, Cr. victoriae, Dioszegia hungarica and Leucosporidium scottii. The cosmopolitan yeast species A. pullulans, C. zeylanoides, D. hansenii, I. orientalis, K. ohmeri, P. guilliermondii, Rh. mucilaginosa, and S. salmonicolor were also isolated. Five possible new species were identified. Sixty percent of the yeasts had at least one detectable extracellular enzymatic activity. Cryptococcus antarcticus, D. aurantiaca, D. crocea, D. hungarica, Dioszegia sp., E. xenobiotica, Rh. glaciales, Rh. laryngis, Microglossum sp. 1 and Microglossum sp. 2 produced mycosporines. Of the yeast isolates, 41.7% produced pigments and/or mycosporines and could be considered adapted to survive in Antarctica. Most of the yeasts had extracellular enzymatic activities at 4ºC and 20ºC, indicating that they could be metabolically active in the sampled substrates.

OBJETIVO: Estudar a imunogenicidade e a estabilidade do surfactante de origem porcina produzido pelo Instituto Butantan. MÉTODO: Experimento imunogenicidade: 16 coelhos da raça New-Zealand-White (Peso de 1000g) foram divididos em grupos de 4 animais. Cada grupo foi designado para receber: a) Surfactante do Butantan, b) Survanta® (Abbott Laboratories), c) Curosurf (Farmalab Chiesi) e d) nenhum tratamento com surfactante. Os surfactantes foram administrados via intratraqueal e o sangue dos animais foi coletado antes, 60 e 180 dias após a administração do surfactante. O soro obtido foi analisado quanto a presença de anticorpos anti-surfactante pelo método ELISA (enzyme-linked immunosorbent assay). Experimento estabilidade: O surfactante do Butantan usado neste experimento tinha sido armazenado por um ano em refrigerador (4 a 8°C) e sua estabilidade foi analisada em condições distintas de experimentação, usando o modelo de coelho prematuro. RESULTADOS: Experimento imunogenicidade: Nenhum dos surfactantes analisados determinou a produção de anticorpos contra seus constituintes. Experimento estabilidade: Os resultados deste estudo demonstraram que o surfactante do Instituto Butantan mostrou eficácia semelhante a do Curosurf após ter sido submetido à condições adversas ao longo do tempo. A eficácia foi demonstrada através da complacência pulmonar dinâmica, pressão ventilatória e da curva pressão-volume. CONCLUSÃO: Os resultados deste estudo demonstraram que o surfactante do Instituto Butantan pode representar um tratamento alternativo de reposição de surfactante.
PURPOSE: To study the immunogenicity and the stability of the porcine pulmonary surfactant preparation produced by the Instituto Butantan. METHOD: Immunogenicity assay: Sixteen New-Zealand-White rabbits (1000 g body weight) were divided into 4 study groups. Each group was assigned to receive either a) Butantan surfactant, b) Survanta® (Abbott Laboratories), c) Curosurf® (Farmalab Chiesi), or d) no surfactant. The surfactants were administered intratracheally, and the animals were collected immediately before and 60 and 180 days after surfactant administration. Sera were assayed for the presence of antisurfactant antibodies by enzyme-linked immunosorbent assay (ELISA). Stability assay: The Butantan surfactant used in this assay had been stored for one year in the refrigerator (4 to 8ºC) and its stability was evaluated in distinct assay conditions using a premature rabbit model. RESULTS: Immunogenicity assay: None of the surfactants analyzed triggered antibody immune responses against their components in any of the animals. Stability assay: The results of this study demonstrate that Butantan surfactant was as effective as Curosurf when both were submitted to the adverse circumstance of short- and long-term storage at room temperature. A similar level of efficacy for the Butantan surfactant, as compared to Curosurf was demonstrated by the pulmonary dynamic compliance, ventilatory pressure, and pressure-volume curve results. CONCLUSION: The results of our study demonstrate that Butantan surfactant may be a suitable alternative for surfactant replacement therapy.

A small-mass system has been developed for monitoring the flux of neutrons with energy up to 1 MeV at the new time-of-flight facility at CERN, n_TOF. The monitor is based on a thin Mylar foil with a 6Li deposit, placed in the neutron beam, and an array of Silicon detectors, placed outside the beam, for detecting the products of the 6Li(n, alpha)³H reaction. The small amount of material on the beam ensures a minimal perturbation of the flux and minimizes the background related to scattered neutrons. Moreover, a further reduction of the gamma-ray background has been obtained by constructing the scattering chamber hosting the device in carbon fibre. A detailed description of the flux monitor is here presented, together with the characteristics of the device, in terms of efficiency, resolution and induced background. The use of the monitor in the measurement of neutron capture cross-sections at n_TOF is discussed

A densidade do solo de amostras indeformadas foi determinada por meio das técnicas de tomografia computadorizada e do torrão impermeabilizado, visando à identificação de processos de compactação do solo e a comparação de métodos. A pesquisa também visou auxiliar um levantamento pedológico elaborado com fins de classificação de terras para irrigação. As amostras de solo foram coletadas da região semi-árida de Petrolina (PE), ao longo de perfis do solo, da superfície até 1 m de profundidade, em camadas de 0,1 m e em duas áreas distintas quanto à sua ocupação vegetal e manejo: (1) vegetação natural (caatinga); (2) área cultivada e irrigada por pivô central. As tomografias foram realizadas com um tomógrafo de primeira geração, constituído por uma fonte de radiação gama de 137Cs e um detetor cintilador de NaI(Tl), com eletrônica padrão de espectrometria gama e sistema de movimentação de amostra controlados por uma interface específica acoplada a um microcomputador. Na área de caatinga, os resultados evidenciam a ocorrência de um adensamento do solo em camadas mais profundas, abaixo de 0,4 m, bem como de uma camada adensada e, ou, compactada entre aproximadamente 0 e 0,4 m, na área cultivada com pivô central, possivelmente em virtude da ação de máquinas e implementos utilizados no preparo e cultivo do solo.
The soil bulk density of undisturbed samples was evaluated using computed tomography and paraffin-sealed soil clod techniques, with the objective of identifying soil compaction processes and to compare the applied methodologies. Data were also used to complement a soil classification survey for irrigation purposes. Samples were collected from soil profiles of the semi-arid region of Petrolina (PE), Brazil, from the soil surface down to a depth of 1 m, in 10 cm layers. Two distinct areas were chosen: (1) native vegetation ("caatinga"), and (2) cultivated area under central pivot irrigation. Tomographies were obtained by a first generation tomograph, using a 137Cs gamma source, a NaI (Tl) scintillation crystal, and a standard electronic counting system linked to a sample moving system, controlled by a specific computational interface. Results suggest a density increase at greater depths (below 0.4 m) for the "caatinga" area. In the central pivot area, a compacted soil layer was detected between approximately 0 and 0.4 m, consisting of natural and/or dense layers, possibly due to machinery action during soil tillage.

OBJETIVO: apresentar a evolução clínica de recém-nascido portador de insuficiência respiratória grave neonatal secundária à Sindrome de Aspiração Meconial tratado por Circulação Extracorpórea por Membrana, ou, conforme o termo consagrado em língua inglesa, ECMO (Extracorporeal Membrane Oxygenation), o efeito do uso de surfactante exógeno neste caso e os custos do procedimento. MÉTODOS: Descrição de um caso de Síndrome de Aspiração Meconial, tratado na UCINE (Unidade de Cuidados Intensivos Neonatais) do Instituto da Criança Prof. Pedro de Alcantara, Hospital das Clínicas da Universidade de São Paulo. RESULTADOS: O suporte extracorpóreo teve a duração de 5 dias, sem complicações clínicas ou mecânicas. Surfactante exógeno de origem porcina foi administrado no 4o dia, após o quê observamos uma melhora significativa na complacência pulmonar. O recém-nascido pôde então ser rapidamente decanulado. Os custos do tratamento foram compatíveis com a realidade nacional em relação a um recém-nascido criticamente enfermo. CONCLUSÕES: a ECMO é indicada em casos de insuficiência respiratória neonatal que não respondam a outros tratamentos existentes. Deve ser disponível em Unidades de Tratamento Intensivo (UTIs) neonatais de hospitais terciários e ser empregada conforme critérios bem estabelecidos. A utilização de surfactante exógeno aparentemente antecipou a retirada da ECMO e, portanto, deve ser considerada em casos semelhantes. Os custos do tratamento justificam a organização de Equipes de ECMO nessas UTIs.
OBJECTIVES: to present the clinical outcome of a newborn with severe respiratory distress secondary to meconium aspiration syndrome and treated by extracorporeal membrane oxygenation (ECMO); and to present the effect of the use of exogenous surfactant in this case and the cost of the procedure. METHODS: Case report of a newborn with meconium aspiration syndrome and treated at the neonatal ICU of the Instituto da Criança Prof. Pedro de Alcantara, Hospital das Clínicas of the Universidade de São Paulo. RESULTS: ECMO was carried out for 5 days with no clinical or mechanical complications. On the 4th day of ECMO, we administered porcine exogenous surfactant; a significant improvement in lung compliance was observed and the newborn was decannulated shortly after that. Treatment costs were compatible with the situation of healthcare in Brazil for treatment of critically ill newborn patients. CONCLUSIONS: ECMO is indicated in cases of neonatal respiratory distress not responding to other treatments. The technique should be made available in neonatal Intensive Care Units (ICUs) of tertiary hospitals according to well-established protocols. The use of exogenous surfactant apparently allowed for earlier decannulation of the patient and should be considered in similar cases. The treatment costs do justify the organizing of ECMO teams in this type of ICUs.
