ABSTRACT Immunocompromised individuals were considered high-risk for severe disease due to SARS COV-2 infection. This study aimed to describe the safety of two doses of COVID-19 adsorbed inactivated vaccine (CoronaVac; Sinovac/Butantan), followed by additional doses of mRNA BNT162b2 (Pfizer/BioNTech) in immunocompromised (IC) adults, compared to immunocompetent/healthy (H) individuals. This phase 4, multicenter, open label study included solid organ transplant and hematopoietic stem cell transplant recipients, cancer patients and people with inborn errors of immunity with defects in antibody production, rheumatic, end-stage chronic kidney or liver disease, who were enrolled in the IC group. Participants received two doses of CoronaVac and additional doses of mRNA BNT162b2. Adverse reactions (AR) data were collected within seven days after each vaccination. Serious adverse events and of special interest (AESI) were monitored throughout the study. We included 241 immunocompromised and 100 immunocompetent subjects. Arthralgia, fatigue, myalgia, and nausea were more frequent in the IC group after CoronaVac. Following the first additional dose of mRNA BNT162, pain, induration, and tenderness at injection site, fatigue and myalgia were more frequent in the H group. A heart transplant recipient had a graft rejection temporally associated with the second CoronaVac dose, but there was no literature evidence of causal association. Four cases of AESI were considered related to the vaccine: three erythema multiforme after CoronaVac, all in IC participants, and one paresthesia after mRNA, in a H participant. Our findings were comparable to other studies that evaluated the safety of COVID-19 vaccines in different immunocompromised populations. Both vaccines were safe for immunocompromised participants.

ABSTRACT Purpose: To evaluate the economic impact of the following initial treatment scenarios for glaucoma on the Brazilian Public Health System (SUS): (1) traditional continuous instillation of hypotensive eye drops and (2) single session of selective laser trabeculoplasty. Methods: Economic impact was analyzed in three scenarios, from the least to the most conservative, for a hypothetical cohort of 5,000 individuals with open-angle glaucoma. Thereafter, projections were made on the basis of a glaucoma prevalence of 3% in the 2021 Brazilian population size. Results: All three scenarios demonstrated that selective laser trabeculoplasty exhibited a significantly lower economic impact than the eye drops on SUS over one and five years. Furthermore, the difference was more than United States Dollar 8 billion at five years when considering 3% of the Brazilian population aged >40 years in 2021. Conclusion: As the initial treatment for primary open-angle glaucoma, selective laser trabeculoplasty exhibited a lower economic impact on SUS than latanoprost and timolol maleate eye drop instillation in all the studied scenarios over one and five-year periods.

Microparticulated whey protein can be used as a fat substitute in foods to enhance their nutritional value and improve their sensory and rheological characteristics. Microparticulation denatures whey proteins through heat treatment, followed by shear rate control to determine the aggregate sizes formed. The processing method used during microparticulation can modify the protein denaturation rate, which may enable the application of new proteins. This study sought to develop a methodology for quantifying whey protein fractions in commercial samples before and after microparticulation at different concentrations and physical states. The developed methodology proved to be applicable because of its selectivity, quantification, detection limits, and precision. Resumen Antecedentes: Los genomas de cloroplasto de Cactaceae exhiben modificaciones en los límites de las regiones repetidas inversas (IRs), inversiones de genes y deleciones. En nueve especies de Mammillaria, se han encontrado tres estructuras de cloroplasto distintas, aunque no todas corresponden a las clasificaciones basadas en morfología. Pregunta: ¿Existe una estructura de genoma de cloroplasto distinta en las especies de Mammillaria serie Stylothelae? Especies estudiadas: Mammillaria bocasana y M. erythrosperma. Sitio y años de estudio: México, de 2019 a 2023. Métodos: ADN de cloroplasto fue secuenciado y los plastomas fueron ensamblados de novo usando el programa Fast-Plast. Las secuencias completas fueron anotadas y verificadas. Las secuencias fueron alineadas en el programa MAUVE para detectar posibles cambios estructurales. Un análisis filogenético con máxima verosimilitud evaluó las relaciones de las especies estudiadas. Resultados: Los plastomas variaron desde 107,368 pb de longitud en Mammillaria bocasana hasta 108,069 pb en M. erythrosperma. Ambos presentaron una estructura cuadripartita y contuvieron 108 genes. Las IRs tuvieron ~1,600 pb y contuvieron los genes rpl2, rpl23 (pseudo) y trnI-CAU. MAUVE identificó una inversión de ~21 kb en la región larga de copia única que contiene genes relacionados con la fotosíntesis. El análisis filogenético recuperó a ambas especies como grupos hermanos dentro de Mammillaria subg. Mammillaria. Conclusiones: Las dos especies estudiadas de Mammillaria serie Stylothelae presentaron una estructura de plastoma sinapomórfica. Otras estructuras de plastomas de Mammillaria han evolucionado independientemente en linajes diferentes.
Abstract Background: The chloroplast genomes of Cactaceae exhibit boundary modifications in the inverted repeat regions (IRs), gene inversions, and deletions. Among nine Mammillaria species, three distinct chloroplast structures have been identified, although not all of these correspond to the morphology-based classification of the genus. Question: Is there a distinct chloroplast genome structure in the species of Mammillaria series Stylothelae? Studied species: Mammillaria bocasana and M. erythrosperma. Study site and dates: Mexico from 2019 to 2023. Methods: Chloroplast DNA was sequenced, and chloroplast genomes were de novo assembled using the Fast-Plast program. Complete plastome sequences were annotated and verified. The sequences were aligned in MAUVE program to detect possible structural changes. A maximum likelihood phylogeny was executed to evaluate the relationships of the studied species. Results: The plastomes ranged from 107,368 bp in Mammillaria bocasana to 108,069 bp in M. erythrosperma. Both presented a quadripartite structure and contained 108 genes. The IRs were ~ 1,600 bp long and included the genes rpl2, rpl23 (pseudo), and trnI-CAU. MAUVE identified a ~ 21 kb inversion in the large single copy containing a block of genes related to photosynthesis. The phylogenetic analysis placed both species in a single clade separated from the other species within Mammillaria subg. Mammillaria. Conclusions: The studied species of Mammillaria series Stylothelae exhibited a different and synapomorphic chloroplast genome structure. Other Mammillaria chloroplast genome structures have evolved independently in different lineages.

Abstract Products derived from medicinal plants with antimicrobial activity are considered a promising alternative in the treatment of fungal infections. In this perspective, this study proposed to evaluate the antifungal activity of the dichloromethane fraction of Annona crassiflora Mart. against C. albicans strains. Tests were carried out to determine Minimum Inhibitory Concentration (MIC), Minimum Fungicide Concentration (MFC), microbial growth kinetics, fungal cell wall and membrane mechanisms of action, antifungal biofilm activity, and cytotoxic effects on human erythrocytes. The extract presented MIC and MFC values that ranged from 256 µg/mL to 1,024 µg/mL, with fungicidal activity in the microbial growth kinetics assay. The mechanism of action did not occur through damage to the cell wall or via binding to ergosterol in the membrane, though the fraction presents activity against biofilm and is not cytotoxic in human erythrocytes. Resumo Produtos derivados de plantas medicinais com atividade antimicrobiana são considerados uma alternativa promissora no tratamento de infecções fúngicas. Nesta perspectiva, o estudo propôs avaliar a atividade antifúngica da fração diclorometano da Annona crassiflora Mart. frente às cepas de C. albicans. Realizou-se ensaios para determinação de Concentração Inibitória Mínima (CIM) e Concentração Fungicida Mínima (CFM), interferência sobre a cinética de crescimento microbiano, mecanismos de ação sobre a parede e a membrana celular fúngica, atividade antifúngica sobre o biofilme e os efeitos citotóxicos sobre eritrócitos humanos. Esse extrato apresentou valores da CIM e CFM que variaram de 256 µg/mL a 1,024µg/mL e no ensaio de cinética de crescimento microbiana, demonstrou atividade fungicida. O mecanismo de ação não ocorreu por danos na parece celular, tampouco por via ligação ao ergosterol da membrana. Demonstrou-se, ainda, que a fração tem ação frente ao biofilme e não é citotóxico em eritrócitos humanos. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. ABSTRACT We present an updated checklist of Brazilian bats, commenting on the endemic and threatened status of the species listed and providing information on recent taxonomic and nomenclatural changes. The bats of Brazil comprise 186 species, 68 genera, and nine families, with 13 species exclusive to the country. From the previous checklists, we add eight species to Brazil: Artibeus amplus, Choeroniscus godmani, Glossophaga bakeri, Lichonycteris obscura, Platyrrhinus guianensis, Trachops ehrhardti, Molossus melini and Myotis pampa. The latter is reported for the country for the first time in this study. The Brazilian list of threatened species includes three species as vulnerable (Furipterus horrens, Lonchophylla bokermanni, and Natalus macrourus) and Lonchophylla dekeyseri as endangered. The International Union for the Conservation of Nature (IUCN) lists Natalus macrourus as “near threatened”, and Lonchophylla bokermanni and L. dekeyseri as “endangered”. Twenty-two additional species are probable for the country. Compared to previous lists, five species are now considered doubtful records and 19 are considered erroneous records. Abstract The objective of the current study was to carry out a survey of the main anatomopathological alterations in raising quails and evaluate possible interference of these in the bone tissue. To obtain the data, 23 quails were collected from farm in the central Serrana region of Espírito Santo. Necropsies with macroscopic descriptions, microbiological, coproparasitological, radiographic and histomorphometric tests were carried out. It was done data descriptive analysis and average comparision using Student T test. It was found that they presented lesions predominantly in the digestive system, followed by urinary and reproductive, and muscular system, were the altered color of the liver (47%) was the most frequent lesion. In the parasitological exams, it was found oocysts of Eimeira sp. (39.13%). In the microbiological exams, it was detected predominantly Escherichia coli (83%). Moderate osteopenia in quails, but the percentage of trabecular bone on bones was similar between healthy and diseased quails, without bone changes in histology. Microscopically, it was observed lung congestion as predominant lesion. Abstract Increasing habitat modification and species loss demand consistent efforts to describe and understand biodiversity patterns. The BIOTA/FAPESP Program was created in this context and it has been a successful initiative to promote studies on biodiversity and conservation in Brazil. The BIOTA/Araçá is an interdisciplinary project that provided a detailed evaluation of the biodiversity of Araçá Bay, a coastal seascape located on the North coast of the state of São Paulo, Southeast Brazil. The bay encompasses multiple habitats, such as beaches, mangroves, rocky shores, and a tidal flat, and provides important ecosystem services. Unfortunately, the bay is the subject of complex social-environmental conflicts that oppose economic, social, and environmental demands (i.e., the expansion of neighboring harbor activities vs. small-scale artisanal fisheries and protection of biodiversity). The present study presents a survey of the benthic species occurring in the different habitats of Araçá Bay, including data obtained during the BIOTA/Araçá project and previous assessments of the area. The benthic species play an important role in marine environments and studying the diversity of these organisms that live associated with the bottom is indispensable for comprehending the environment’s functioning. The macrofauna, meiofauna, and microorganisms associated with soft and hard bottom were listed, and additional information, such as the habitat and geographical distribution, were provided for each species. The checklist includes 826 species, almost 70% recorded during the BIOTA/Araçá project. The most speciose taxa were the annelids (225 spp.), mollusks (194 spp.), and crustaceans (177 spp.). Seven benthic species are endemic to Araçá Bay, 14 are considered threatened, and seven are economically exploited. Furthermore, the bay is the type locality of many taxa, and 11 new benthic species were described based on specimens sampled during the project. Resumo O aumento da modificação dos habitats e da perda de espécies demanda esforços consistentes para descrever e compreender os padrões de biodiversidade. O programa BIOTA/FAPESP foi criado nesse contexto e é uma iniciativa de sucesso para promover estudos em biodiversidade e conservação no Brasil. O BIOTA/Araçá é um projeto interdisciplinar que promoveu uma avaliação detalhada da biodiversidade da Baía do Araçá, um ecossistema costeiro localizado ao Norte do estado de São Paulo, Sudeste do Brasil. A baía engloba múltiplos habitats, tais como praias, manguezais, costões rochosos, e uma planície de maré, e também fornece importantes serviços ecossistêmicos. ABSTRACT Purpose: This study aimed to compare an teriorchamber parameters acquired by a swept-source anteriorsegment optical coherence tomography before and after laser peripheral iridotomy. Methods: This study prospectively evaluated 14 patients with primary-angle closure and six patients with primary-angle closure glaucoma. Gonioscopy and anterior-segment optical coherence tomography using the DRI OCT Triton® were performed before and after laser peripheral iridotomy. Anterior-segment optical coherence tomography parameters were studied using scleral spur as reference: angle opening distance at 250, 500, and 750 µm, trabecular-iris space at 500 µm, trabecular-iris angle, trabecular-iris contact length, and iris curvature. Results: Anterior-segment optical coherence tomography identified 61% of the patients with two or more quadrants closed. Gonioscopy identified more closed angles than anterior-segment optical coherence tomography before laser peripheral iridotomy. In angle parameters, only the angle opening distance of 250 µm at the nasal quadrant was not significantly increased after laser peripheral iridotomy. The iris curvature and trabecular-iris contact length showed a significant reduction induced by the laser procedure. Even in eyes in which gonioscopy did not identify angular widening after laser peripheral iridotomy (n=7), the angle opening distance of 750 µm increased (nasal, 0.15 ± 0.10 mm to 0.27 ± 0.16 mm, p=0.01; temporal, 0.14 ± 0.11 mm to 0.25 ± 0.12 mm, p=0.001) and the iris curvature decreased (nasal, 0.25 ± 0.04 mm vs. 0.11 ± 0.07 mm, p=0.02; temporal, 0.25 ± 0.07 mm vs. 0.14 ± 0.08 mm, p=0.007). RESUMO Objetivo: Comparar os parâmetros de câmara anterior obtidos através da tomografia de coerência óptica de segmento anterior antes e após a iridectomia periférica a laser. Métodos: Quatorze pacientes com fechamento angular primário e seis com glaucoma primário de ângulo fechado foram prospectivamente avaliados neste estudo. Gonioscopia e tomografia de coerência óptica de segmento anterior com DRI OCT Triton® foram realizadas antes e após a iridectomia periférica a laser. Os seguintes parâmetros de tomografia de coerência óptica de segmento anterior, baseados na localização do esporão escleral, foram avaliados: ângulo de abertura angular a 250 µm, 500 µm e 750 µm, área do espaço entre a íris e o trabeculado a 500 µm, ângulo entre a íris e o trabeculado, extensão do contato entre a íris e o trabeculado e curvatura da íris. Resultados: A tomografia de coerência óptica de segmento anterior identificou 61% dos indivíduos com dois ou mais quadrantes fechados. A gonioscopia identificou mais quadrantes com ângulo fechado do que tomografia de coerência óptica de segmento anterior antes da iridectomia periférica a laser. Quanto aos parâmetros angulares, apenas ângulo de abertura angular a 250 µm no quadrante nasal não aumentou significativamente após a iridectomia periférica a laser. A curvatura da íris e a extensão do contato entre a íris e o trabeculado apresentaram redução significativa induzida pelo procedimento a laser. Mesmo nos olhos em que a gonioscopia não identificou aumento da amplitude angular após iridectomia periférica a laser (n=7), ângulo de abertura angular a 750 µm aumentou (nasal: 0,15 ± 0,10 mm para 0,27 ± 0,16 mm, p=0,01; temporal: 0,14 ± 0,11 mm para 0,25 ± 0,12 mm, p=0,001), e ICURVE diminuiu (nasal: 0,25 ± 0,04 mm vs. 0,11 ± 0,07 mm, p=0,02; temporal: 0,25 ± 0,07 mm vs. 0,14 ± 0,08 mm, p=0,007). ABSTRACT This study investigated the in vitro efficacy of emamectin benzoate and glyphosate against monogeneans from Colossoma macropomum and tolerance of this species to these therapeutic drugs. In vitro assays demonstrated that concentrations of emamectin benzoate (12.5, 16.6, 20.8 and 25.0g/L) and glyphosate (2,000; 3,000; 4,000 and 5,000mg/L) were 100% effective against Anacanthorus spathulatus, Notozothecium janauachensis and Mymarothecium boegeri. The highest concentrations of emamectin benzoate (20.8 and 25.0g/L) were 100% effective after 30 minutes of exposure. For glyphosate, the highest concentrations were the most effective, and 4,000 and 5,000 mg/L immobilized 100% of parasites in 45 minutes and 30 minutes, respectively. Scanning electron microscopy revealed that the parasites exposed to 20.8 and 25.0g/L of emamectin benzoate and 5,000 mg/L of glyphosate presented integument covered with deeper wrinkles. Behavioral changes occurred in C. macropomum exposed to all concentrations of both drugs, were as mortality occurred following exposure to emamectin benzoate and glyphosate at 2.0,12,5 and 25.0g/L and 500, 1,000, 2,000, 3,000, 4,000 and 5,000mg/L, respectively. RESUMO Este estudo investigou a eficácia in vitro do benzoato de emamectina e do glifosato contra monogenéticos de Colossoma macropomum e a tolerância dessa espécie a essas drogas terapêuticas. Ensaios in vitro demonstraram que as concentrações de benzoato de emamectina (12,5; 16,6; 20,8 e 25,0g/L) e de glifosato (2.000; 3.000; 4.000 e 5.000mg/L) foram 100% eficazes contra Anacanthorus spathulatus, Notozothecium janauachensis e Mymarothecium boegeri. As maiores concentrações de benzoato de emamectina (20,8 e 25,0g/L) foram 100% efetivas após 30 minutos de exposição. Para o glifosato, as maiores concentrações foram as mais efetivas, sendo que 4.000 e 5.000 mg/L imobilizaram 100% dos parasitos em 45 minutos e 30 minutos, respectivamente. A new phenylpropanoid derivative, named euploic acid (1), was isolated from Euploca procumbens (Mill.) Diane & Hilger, Boraginaceae, along with lithospermic acid B (2), lithospermic acid (3), 9’’-methyl lithospermate (4), and luteolin-7-O-glucoside (5). Compound structures were determined by mass spectrometry (MS), nuclear magnetic resonance (NMR) analysis and comparison with published data. Absolute configuration of 1 was established via electronic circular dichroism (ECD). Anti-inflammatory potential of euploic acid (1) was assessed by measuring its inhibition of cytokine and nitric oxide production in stimulated J774 macrophages. Compound 1 significantly reduced pro-inflammatory mediator production and release by stimulated macrophages at non-cytotoxic concentrations, comparable to the efficacy of dexamethasone. Our findings demonstrate the potential of euploic acid (1) as an effective anti-inflammatory agent. This study contributes to the structural elucidation of a new phenylpropanoid derivative and highlights its promising anti-inflammatory activity in the treatment of inflammatory disorders. 1, , Mill. ABSTRACT The knowledge of the bat fauna from Minas Gerais, the largest state in southeastern Brazil, has greatly changed in recent years due to increased sampling and taxonomic revisions with reports of range extension and/or new species descriptions. The first annotated list of bats for the state dates to over 10 years ago. Our study aimed to update it by incorporating new localities sampled since then and by adding new species records to the list. We also conducted a comprehensive review of the literature and museum databases and analyzed 691 specimens collected in the state from all mesoregions and ecosystems deposited in collections. We documented the occurrence of 99 bat species from nine families, distributed in 181 municipalities, covering all mesoregions and biomes of the state. This is an increment of 30% in bat richness and of 241% in locality sites with bat records in Minas Gerais, when compared with the previous compilation. We report two previously unrecorded species for the state, the phylostomids Rhinophylla pumilio Peters, 1865 and Trinycteris nicefori (Sanborn, 1949). The mesoregions with the largest number of bat collection localities were “Metropolitana” (46), “Zona da Mata” (26), and “Norte de Minas” (21). The Atlantic Forest had the largest number of collection sites compared to the other biomes present in the state (102). There are a lot of sampling gaps in the state, and most of the conservation areas have not yet been sampled for bats. In face of that and based on the recent findings of several new species and records as a result of sampling and revisionary efforts, we suggest the implementation of rapid survey programs accompanied by taxonomic studies within the conservation units of Minas Gerais. Isoquinoline alkaloids, especially from the Annonaceae family, have shown biological potential against parasites. Thus, this study aimed to evaluate the potential of the alkaloid fractions of the plant Fusaea longifolia against Plasmodium falciparum and annotate the compounds present in these samples. The tentative characterization of the alkaloids from the leaves and branches of F. longifolia was performed using liquid chromatography coupled to mass spectrometry (LC-MS/MS) and molecular networks. Through manual interpretation of the MS/MS spectra, 18 alkaloids were dereplicated from F. longifolia, 17 of which were reported for the first time in this species. An unpublished putative glycosylated alkaloid was annotated by interpreting the fragmentation data profile. Regarding biological activity, the fractions studied showed high activity against P. falciparum with half-maximal inhibitory concentration (IC50) of 2.42 and 1.60 μg mL-1 for branches and from the leaves, respectively, both similar to the reference standard quinine (IC50 of 1.24 μg mL-1). The structures of the 17 alkaloids were subjected to in silico analysis using molecular docking against four enzymes related to anti-Plasmodium activity (wild type (dm-PfDHFR) and mutant type (qm-PfDHFR), dihydroorotate dehydrogenase (PfDHODH) and purine nucleoside phosphorylase (PfPNP)).
