Pancreatic bioengineering is a potential therapeutic alternative for type 1 diabetes (T1D) in which the pancreas is decellularized, generating an acellular extracellular matrix (ECM) scaffold, which may be reconstituted by recellularization with several cell types to generate a bioartificial pancreas. No consensus for an ideal pancreatic decellularization protocol exists. Therefore, we aimed to determine the best-suited detergent by comparing sodium dodecyl sulfate (SDS), sodium deoxycholate (SDC), and Triton X-100 at different concentrations. Murine (n=12) and human pancreatic tissue from adult brain-dead donors (n=06) was harvested in accordance with Institutional Ethical Committee of the University of São Paulo Medical School (CEP-FMUSP) and decellularized under different detergent conditions. DNA content, histological analysis, and transmission and scanning electron microscopy were assessed. The most adequate condition for pancreatic decellularization was found to be 4% SDC, displaying: a) effective cell removal; b) maintenance of extracellular matrix architecture; c) proteoglycans, glycosaminoglycans (GAGs), and collagen fibers preservation. This protocol was extrapolated and successfully applied to human pancreas decellularization. The acellular ECM scaffold generated was recelullarized using human pancreatic islets primary clusters. 3D clusters were generated using 0.5×104 cells and then placed on top of acellular pancreatic slices (25 and 50 μm thickness). These clusters tended to connect to the acellular matrix, with visible cells located in the periphery of the clusters interacting with the ECM network of the bioscaffold slices and continued to produce insulin. This study provided evidence on how to improve and accelerate the pancreas decellularization process, while maintaining its architecture and extracellular structure, aiming at pancreatic bioengineering. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). RESUMO: Este estudo teve como objetivo relatar um leiomiossarcoma associado a abdome agudo e torção uterina em uma cateta (Tayassu tajacu) de oito anos, nulípara, mantida sob cuidados humanos. O animal apresentou uma história clínica de dois dias de aumento de volume abdominal, secreção vaginal e abdome agudo. Achados de ultrassonografia indicaram um tumor intrabdominal, e uma laparotomia exploratória seguida de ovariohisterectomia de emergência foi realizada para remover o tumor. Macroscopicamente, o corno direito do útero exibia um tumor firme, friável medindo 23cm de diâmetro e pesava 9,3kg. Na superfície de corte, o tumor drenava um fluido de odor fétido e conteúdo sanguinolento. A parede do tumor tinha 6cm de espessura, era de cor vermelha escura e firme, e apresentava um material fibrilar amarelo, friável e macio aderido firmemente à sua superfície. Microscopicamente, a lesão consistia em proliferação maligna dos leiomiócitos acompanhada de necrose e hemorragia difusas de endométrio e miométrio. O citoplasma das células neoplásicas foi fortemente imunopositivo para vimentina e actina de músculo liso e negativo para citoqueratina e desmina. Leiomiossarcomas são raros em Tayassuidae e sua apresentação clinicopatológica pode ser concomitante com abdome agudo, torção uterina e piometra. Essas condições devem ser incluídas no diagnóstico diferencial de doenças reprodutivas observadas em catetas. ABSTRACT: This study aimed to report a leiomyosarcoma associated with acute abdomen and uterine torsion in an 8-year-old, nulliparous, white-collared peccary (Tayassu tajacu) kept under human care. The animal presented a two-day clinical history of abdominal bulging, vaginal discharge, and acute abdomen. Ultrasound findings indicated an intrabdominal tumor, and an exploratory laparotomy followed by an emergency ovariohysterectomy was performed to remove the tumor. Grossly, the right horn exhibited a firm, friable tumor measuring 23cm in diameter and weighing 9.3kg. On the cut surface, the tumor drained a foul-smelling fluid and bloody content. The tumor wall was 6cm thick, dark red, firm, and had a brittle, soft, yellow fibrillar material firmly adhered to its surface. Microscopically, the lesion consisted of malignant proliferation of leiomyocytes with necrosis and hemorrhage of the endometrium and myometrium. The cytoplasm of the neoplastic cells was strongly immunopositive for vimentin and smooth muscle actin but negative for cytokeratin and desmin. Leiomyosarcoma is uncommon in Tayassuidae, and its clinicopathological presentation may be concomitant with acute abdomen, uterine torsion, and pyometra. These conditions should be included in the differential diagnosis of reproductive diseases observed in female peccaries. High-intensity interval training (HIIT) has stood out as a treatment for obesity, leading to adaptations of the cardiovascular system and reducing body adiposity. In addition, the search for alternative therapies for weight loss has intensified. The administration of Hibiscus sabdariffa (Hs) has been described as an efficient supplement in weight loss and in the treatment of metabolic changes associated with obesity. In this context, the objective was to investigate the effects of the association of Hs and HIIT on metabolic adaptations and lipid metabolism in obese rats. Wistars rats were subjected to obesity and subsequently randomized into 4 groups: obese (Ob), obese + HS (ObHs), obese + HIIT (ObHIIT), and obese + HS + HIIT (ObHsHIIT). For 8 weeks, ObHs and ObHsHIIT rats received Hs extract daily (150 mg/kg of body weight) and trained groups (ObHIIT and ObHsHIIT) were subjected to a HIIT program on a treadmill. Nutritional profile, glycemic curve, biochemical profile, and liver glycogen were determined. HIIT decreased caloric intake, feed efficiency, body adiposity, total body fat, and body weight gain, associated with improvements in physical performance parameters and a smaller glycemic curve and area. Hs had a hepatoprotective effect, reducing alkaline phosphatase values, but its effects were more pronounced when associated with HIIT. Therefore, the combination of treatments promoted a reduction in food consumption and body adiposity, as well as an improvement in physical performance and glycemic profile, but without changes in lipid metabolism. RESUMO Os distúrbios neurológicos em aves incluem vários agentes infecciosos ou não infecciosos, alguns envolvidos em malformações embrionárias. A hidrocefalia é uma importante desordem neurológica a ser pesquisada em aves. O presente relato tem como objetivo investigar um caso de hidrocefalia em um papagaio-verdadeiro (Amazona aestiva) de um criadouro conservacionista, localizado no estado de Minas Gerais, Brasil, clinicamente acometido por sinais neurológicos. As lesões post mortem observadas foram megaloencefalia, distensão e adelgaçamento da calota craniana, cérebro subdesenvolvido, espessamento das meninges e presença de grande volume (aproximadamente 10mL) de líquido cefalorraquidiano (LCR) claro. As avaliações histopatológicas demonstraram discreta malácia multifocal, associada à gliose e satelitose neuronal no sistema nervoso central. A triagem molecular para vírus e parasitos revelou a ausência de várias etiologias comuns, incluindo Alphavirus (protocolo universal do gênero), Bornavirus aviário, Alphaherpesvirinae (primers universais para subfamília), Paramyxovirus (primers universais da família), vírus da doença de Newcastle (Orthoavulavirus aviário 1), vírus influenza A (Alphainfluenzavirus), vírus da doença de Marek (Mardivirus, Gallid herpesvirus 2), vírus do Nilo Ocidental, Flavivirus (Orthoflavivirus primers universais), Apicomplexa (primers universais para a ordem) e Toxoplasma gondii, todos testados negativos por PCR ou RT-PCR. A bacteriologia e a micologia aeróbias do líquido cefalorraquidiano não revelaram crescimento. Os resultados sugerem que a hidrocefalia resultou do acúmulo crônico craniano de LCR, com aumento da pressão craniana e distensão do crânio antes da ossificação. ABSTRACT Hydrocephalus is an important neurological disorder rarely described in birds. The present report aims to describe a natural case of hydrocephalus in a blue-fronted parrot (Amazona aestiva) from a conservationist breeding (Minas Gerais, Brazil), clinically affected with neurological signs. The post-mortem lesions noted were megalencephaly, distension and thinning of the skullcap, underdeveloped brain, thickening of the meninges and the presence of a large volume of approximately 10mL of clear cerebrospinal fluid (CSF). The histopathological evaluations demonstrated discrete multifocal malacia, associated with gliosis and neuronal satellitosis in the central nervous system. The screening for viruses revealed the absence of several common etiologies, including Alphavirus (genus universal primers), avian Bornavirus, Alphaherpesvirinae (subfamily universal primers), Paramyxovirus (family universal primers), Newcastle’s disease virus (avian Orthoavulavirus 1), influenza A virus (Alphainfluenzavirus), Marek's disease virus (Mardivirus, Gallid herpesvirus 2), West Nile virus, Flavivirus (Orthoflavivirus universal primers), Apicomplexa (universal primers) and Toxoplasma gondii, all tested negative by PCR or RT-PCR. RESUMO Este trabalho objetivou o isolamento e a identificação das bactérias de diferentes microbiotas de lhamas. Para tanto, testes de disco difusão e diagnósticos moleculares para pesquisa de genes de resistência foram realizados. Foram isolados cinco Staphylococcus spp. coagulase positiva e 19 Staphylococcus spp. coagulase negativa, 19 Escherichia coli¸ duas Pantoea agglomerans, uma Koserella trabulsii, uma Enterobacter aerogenes e uma Klebsiella Pneumoniae. Entre os isolados de Staphylococcus spp., 79,17% foram resistentes ao sulfazotrim, 45,83% resistentes à penicilina e 20,83% à ampicilina. Foi confirmada a presença do gene mecA em apenas um isolado oxacilina resistente. No teste de disco difusão, 58,3% das enterobactérias foram resistentes à amoxicilina + ácido clavulânico e à cefotaxima, 50% à ceftazidima e ceftriaxona, e 33,3% à amoxicilina. Ainda entre os isolados Gram-negativos, não houve a expressão fenotípica de isolados ESBL. O presente trabalho expôs a presença de microrganismos resistentes a antibióticos em lhamas que não tiveram contato prévio com essas drogas, além da presença do gene mecA em um dos animais. Abstract Objective: To investigate the effects of different physical exercise programs and polymorphisms of the FTO (fat mass and obesity-associated gene) on body composition and cardiovascular risk factors in adolescents with overweight and obesity. Methods: A randomized, parallel, double-blind clinical trial consisting of the adolescent overweight from the state public network, in a simple representative random sample, who participated in an aerobic exercise or weight training intervention for 10 weeks. Anthropometry, body composition, biochemical markers, sexual maturation, and rs9939609 polymorphism in the FTO gene were assessed. 347 adolescents had their characterization of nutritional status. 72 individuals with overweight and obesity were invited to participate. 39 remained for the start of the program and were randomly allocated to both types of intervention. In the end, 26 subjects participated in the intervention programs, with 12 and 14 in the aerobic and weight training programs, respectively. Results: Heterozygous and homozygous bearers of risk allele A participating in the aerobic program showed improvements in glycemia (p = 0.002) and total cholesterol (p = 0.023) and a reduction in body fat mass (p = 0.041). The weight training program reduced glycemia in patients with the risk allele A (p = 0.027). Cameron’s stage four sexual maturation participants were 2.1 times more likely to improve their body fat (CI = 1.31–3.39). Conclusion: Aerobic exercises produced exclusively a significant decrease in fat mass and total cholesterol in patients with risk allele A. Distinct physical exercise programs may cause diverse changes in risk variables related to the health of adolescents. RESUMO Os países em desenvolvimento enfrentam grandes desafios para combinar a intensificação da agricultura com o desenvolvimento sustentável das cadeias de abastecimento. Este documento explora as oportunidades de inovação na conservação do gado Curraleiro Pé-Duro (CPD). O CPD é uma raça brasileira histórica, relacionada à cultura regional, que apresenta notável adaptabilidade em biomas tropicais. O método de pesquisa consistiu em entrevistar e analisar o discurso dos criadores de CPD no Tocantins, Brasil. A sistematização dos dados resultou em quatro categorias e onze subcategorias, que foram definidas a posteriori. Nossas descobertas revelam aspectos motivacionais, potencialidades e perspectivas de valorização, além das necessidades e desafios percebidos pelos produtores de CPD. Uma melhor compreensão da situação atual pode orientar as políticas públicas, incentivar os interessados, beneficiar os agricultores e levar a iniciativas agroalimentares sustentáveis. As contribuições empíricas de nosso diagnóstico são discutidas e mostramos como elas trazem e fornecem lições valiosas para prospectar inovações na conservação do gado de CPD. Estas descobertas podem apoiar acadêmicos e pessoas que também estão interessadas em acessar e apoiar a conservação e produção animal com uso de raças crioulas. ABSTRACT Developing countries face great challenges to couple agricultural intensification and sustainable supply chain development. This paper explores the opportunities for innovation in the conservation of the locally adapted Curraleiro Pé-Duro (CPD) cattle. The CPD is a historical Brazilian breed, related to regional culture, which exhibits remarkable adaptability in tropical biomes. The research method consisted of interviewing and analyzing the discourse of CPD breeders in Tocantins, Brazil. The systematization of the data resulted in four categories and eleven subcategories, which were defined a posteriori. Our findings reveal motivational aspects, potentialities, and prospects for valuation, besides the needs and perceived challenges faced by CPD farmers. A better understanding of the current situation may guide public policies, encourage stakeholders, benefit farmers, and lead to sustainable agri-food initiatives. The empirical contributions of our diagnosis are discussed, and we show how they bring forward and provide valuable lessons to prospect innovations in the conservation of CPD cattle. These findings may support scholars and people who are also interested in accessing and supporting the conservation of indigenous livestock. In December 2022, the first iAtlantic expedition in the South Atlantic set off to map and explore seafloor ecosystems in the Santos Basin slope (200-1,000 m depths) in collaboration with the Petrobras ‘Santos Basin - Regional Characterization Project.’ The 17-day ‘iAtlantic_BR10-Petrobras’ cruise was conducted on board the research vessel NPqHOc Vital de Oliveira (Brazilian Navy) and performed (a) water column structure characterization, (b) seafloor morphology mapping, (c) description of benthic habitats and communities by seafloor imagery and biological/ geological sampling, and (d) ex-situ experiments to assess the functioning of sedimentary ecosystems and their responses to climate-related environmental changes. RESUMO: Mycoplasma hyopneumoniae é um dos patógenos respiratórios mais desafiadores envolvidos com pneumonia suína em todo o mundo. É responsável por uma infecção crônica de alta morbidade, que predispõe a infecções bacterianas secundárias nas fases de crescimento e terminação. Avanços nas técnicas diagnósticas permitiram a identificação de características genéticas do agente, associadas à alta variabilidade antigênica e proteômica entre cepas. Este estudo teve como objetivo examinar a ocorrência e diversidade genética de cepas de M. hyopneumoniae em pulmões com lesões pneumônicas em 52 granjas de suínos no estado de Minas Gerais, um dos maiores estados produtores de suínos do Brasil. A genotipagem foi realizada utilizando a técnica de “multilocus variable number of tandem repeat analysis” (VTNR/MLVA), usando dois loci que codificam VNTR das adesinas P97 e P146. Os resultados mostraram que esse agente está amplamente disseminado em granjas de suínos e que existe alto polimorfismo das variantes de M. hyopneumoniae circulando no estado de Minas Gerais. Diferentes genótipos de M. hyopneumoniae estão distribuídos aleatoriamente em várias regiões do estado, sem um padrão de estrutura populacional e geográfica específico. ABSTRACT: Mycoplasma hyopneumoniae is one of the most challenging respiratory pathogens involved with swine pneumonia worldwide, responsible for a chronic infection with high morbidity, which predisposes secondary bacterial infections in growing and finishing pigs. Advances in diagnostic techniques allowed identification of genetic characteristics associated with high antigenic and proteomic variability among bacterial strains. This study aimed to evaluate the genetic diversity of M. hyopneumoniae strains in lungs with pneumonic lesions obtained from 52 pig farms located in Minas Gerais, one of the largest swine production states in Brazil. Genotyping was performed using multilocus variable number of tandem repeat (VNTR) analysis (MLVA), targeting two loci encoding P97 and P146 adhesins VNTR. The results showed that this agent is widely disseminated in pig farms and there is a high polymorphism of M. hyopneumoniae variants circulating in the state of Minas Gerais. Different M. hyopneumoniae genotypes are randomly distributed in several regions of the state, with no specific geographic population structure pattern. Abstract In the current study, two euglossine species, Exaerete smaragdina and Eulaema nigrita, a cleptoparasite bee and its host, respectively, were used as models to: (i) access the genetic diversity and population structure of both species, sampled along a wide latitudinal range of Atlantic Forest, where the distribution of El. nigrita and Ex. smaragdina co-occurs; (ii) investigate the evolutionary history of these species through the Atlantic Forest, and in a wider scenario, to examine the evolutionary history of these species across others forest domains. Analyses involved males of El. nigrita and Ex. smaragdina sampled through Brazilian territory, including 19 sites in the Atlantic Forest. Bayesian Skyline Plot (BSP) was used to infer possible climate oscillations on population of both species over time. The BSP revealed stability in effective population size for both species in most of the Plio-Pleistocene period. However, BSP results aligned to the starlike configuration in the haplotype network, neutrality test, and population diversity patterns indicated population expansion of the two species during the late Pleistocene. Islet transplantation represents a therapeutic option for type 1 diabetes (T1D). Long-term viability of transplanted islets requires improvement. Mesenchymal stromal cells (MSCs) have been proposed as adjuvants for islet transplantation facilitating grafting and functionality. Stem cell aggregation provides physiological interactions between cells and enhances the in situ concentration of modulators of inflammation and immunity. We established a hanging-drop culture of adult human skin fibroblast-like cells as spheroids, and skin spheroid-derived cells (SphCs) were characterized. We assessed the potential of SphCs in improving islet functionality by cotransplantation with a marginal mass of allogeneic islets in an experimental diabetic mouse model and characterized the secretome of SphCs by mass spectrometry-based proteomics. SphCs were characterized as multipotent progenitors and their coculture with anti-CD3 stimulated mouse splenocytes decreased CD4+ T cell proliferation with skewed cytokine secretion through an increase in the Th2/Th1 ratio profile. SphCs-conditioned media attenuated apoptosis of islets induced by cytokine challenge in vitro and importantly, intratesticular SphCs administration did not show tumorigenicity in immune-deficient mice. Moreover, SphCs improved glycemic control when cotransplanted with a marginal mass of allogeneic islets in a diabetic mouse model without pharmacological immunosuppression. SphCs' protein secretome differed from its paired skin fibroblast-like counterpart in containing 70% of up- and downregulated proteins and biological processes that overall positively influenced islets such as cytoprotection, cellular stress, metabolism, and survival. In summary, SphCs improved the performance of transplanted allogeneic islets in an experimental T1D model, without pharmacological immunosuppression. Future research is warranted to identify SphCs-secreted factors responsible for islets' endurance. T1D. TD . The objective of this study was to evaluate the chemical composition and in situ degradability of elephant grass silage dry matter with increasing levels of sugarcane bagasse. A completely randomized design was adopted with five treatments: 0, 5, 10, 15, and 20% inclusion of sugarcane bagasse on an as-fed matter basis, with five replications, totalling 25 experimental units. The increasing proportion of sugarcane bagasse analysed was favourable only in increasing the concentration of dry matter and fibre of silages and in maintaining a considerable crude protein content. In the in situ degradation assay, the addition of sugarcane bagasse in silage decreased the ruminal degradability of dry matter, and as the passage rates increased, the effective degradability decreased. In the in vitro gas production assay, the total production of gases was decreased with the inclusion of sugarcane bagasse, as did the degradability of organic matter and neutral detergent fibre. The recommendation for the inclusion of sugarcane bagasse should be analysed according to the desired objective, emphasizing that values higher than 5% can greatly compromise the final nutritive value of elephant grass silage.

Resumo Os serviços de ecossistema, como a limitação natural de pragas, incluem-se nas estratégias de gestão de agro-ecossistemas e o seu uso pode ser otimizado com base no conhecimento provindo da modelação computacional das pragas, dos seus predadores, e da paisagem. Neste artigo focamo-nos no processamento de dados de temperatura do solo, necessários para o funcionamento dos modelos de praga-predador Bactrocera oleae (mosca da azeitona) e Haplodrassus rufipes (aranha do solo) no olival. A metodologia de processamento permitiu-nos (1) completar as lacunas existentes na série de dados de temperatura do solo, recolhidos com periodicidade horária, (2) convertê-la numa série com periodicidade diária, e ainda (3) criar uma variável adicional, a temperatura média diária do solo durante o crepúsculo vespertino. Este último passo possibilita que não haja uma perda de informação no que diz respeito àquele período específico do dia, reduzindo-se o erro associado à desaquação da resolução temporal da variável climática média diária para exprimir um fenómeno que ocorre apenas durante um determinado número de horas do dia.
Abstract Ecosystem services, such as natural pest control, are included in agro-ecosystem management strategies and their use can be optimized based on knowledge from computational modelling of pests, predators, and landscape. In this article we focus on the processing of soil temperature data, necessary for the operation of the predator-pest models Bactrocera oleae (olive fly) and Haplodrassus rufipes (soil spider) in the olive grove. The processing methodology allowed us to (1) complete the gaps in the series of soil temperature data, collected on an hourly basis, (2) convert it into a series with daily periodicity, and (3) create an additional variable, the average daily temperature of the soil during the twilight. This last step prevents the loss of information regarding that specific period of the day, reducing the error associated with the deaquation of the temporal resolution of the average daily climatic variable to express a phenomenon that occurs only during a certain number of hours of the day.

Resumo Neste estudo, foi feito um ensaio num campo agrícola inserido no perímetro de rega do Roxo, no Alentejo, usando o sensor de indução eletromagnética DUALEM21, de forma a analisar o seu potencial no mapeamento 3D da textura do solo. Os dados da condutividade elétrica aparente do solo (ECa), obtidos com o sensor, foram transformados utilizando um algoritmo de modelação inversa, Quasi-3D, para obter imagens da condutividade elétrica (EMCI) 3D da condutividade elétrica do solo (EC). Esta transformação é necessária, uma vez que ECa representa uma medição média, ponderada em profundidade, da EC e não a sua verdadeira variação com a profundidade. Analisou-se a relação linear entre a EC e a textura do solo, usando dados recolhidos em 13 pontos de amostragem, selecionados tendo em conta a variabilidade da ECa. Em cada ponto de amostragem foram colhidas amostras em três camadas até à profundidade de 0.6 m. Os resultados mostram que existe correlação significativa entre EC e a fração argila e entre EC e a fração areia (R2>0.6), sendo possível transformar as EMCI em mapas 3D do teor de argila e do teor de areia, até uma profundidade de 0.6 m.
Abstract Using a DUALEM21 sensor, we conducted electromagnetic induction (EMI) surveys in an agricultural field in the Roxo irrigation perimeter in Alentejo, Portugal, to evaluate the potential of using this technique for 3D mapping of soil texture. We first processed apparent electrical conductivity data (ECa) and then used a Quasi-3D inversion algorithm in order to obtain 3D electromagnetic conductivity images (EMCI) of the actual bulk soil electrical conductivity (EC). This is because the ECa is a depth-weighted, average conductivity measurement and does not represent the actual EC variation with depth. Afterward, we evaluated the possibility of establishing a linear regression (LR) relationship between EC and soil texture collected from 13 soil sample locations at 3 layers to a depth of 0.60 m. We selected the locations for soil sampling based on the ECa variations. Our analysis showed that it is possible to establish a relatively good LR between EC and clay, and between EC and sand (R2>0.60), allowing us to convert EMCI into sand and clay content and generate sand and clay 3D maps down to 0.60 m depth.
