Abstract Objective It was to verify the association between the definition of sex of rearing and, clinical and cytogenetic features among patients with genital ambiguity referred without a sex assignment. Methods The sample consisted of 133 patients with genital ambiguity seen at a single reference service. These patients did not have a defined social sex at the first consultation and their etiological diagnosis was obtained during follow-up. Results A total of 133 cases were included, 74 of which were reared as males and 59 as females. No correlation was found between the year of birth and the year of the first consultation with the definition of sex of rearing. However, the definition of sex of rearing was associated with age at the first consultation, severity of genital ambiguity, presence of palpable gonad(s), presence of uterus on ultrasound, karyotype, and diagnosis. Palpable gonad(s), more virilized genitalia, absence of a uterus on ultrasound, 46, XY karyotype, or a karyotype with sex chromosome abnormalities emerged as strong predictors for defining male sex. All 77 (58 %) patients over 18 years old had a gender identity in accordance with the sex of rearing; though 9 of 77 (12 %) had homo or bisexual orientation, especially girls with Congenital Adrenal Hyperplasia. Conclusions Clinical and cytogenetic data were strongly associated with the definition of the sex of rearing of children with genital ambiguity referred to a DSD center without sex assignment. The compound 2-hydrazinyl-4-(4-methoxyphenyl)thiazole (PD76) is a novel thiazolyl hydrazine derivative with proven antifungal activity against different fungal species, mainly Candida and Cryptococcus. Considering the advantages of oral route for clinical therapy, the aim of this work was to evaluate the potential intestinal permeability of this new antifungal drug. For the quantitation of PD76, a high-performance liquid chromatography method was developed and fully validated. The cytotoxicity of the compound in Caco-2 cells was analyzed and intestinal permeability of PD76 was assessed by means of the comparison of in vitro assay in Caco-2 cells and in silico platforms ADMETlab and admetSAR. Cell viability above 70% was obtained at all PD76 studied concentrations. Using Caco-2 cell model, the compound showed apparent permeability coefficients (Papp) of 5.25 × 10-6 and 23.28 × 10-6 cm s-1 in apical-basolateral and basolateral-apical directions, respectively. Experiments performed using verapamil as P-gp inhibitor demonstrated that PD76 is slightly susceptible to active efflux. Both in silico platforms inferred that PD76 presents permeability in Caco-2 cells, with Log P values of 2.82 (ADMETlab) and 2.10 (admetSAR). Resumo Fundamento: As ferramentas de telecardiologia são estratégias valiosas para melhorar a estratificação de risco. Objetivo: Objetivamos avaliar a acurácia da tele-eletrocardiografia (ECG) para predizer anormalidades no ecocardiograma de rastreamento na atenção primária. Métodos: Em 17 meses, 6 profissionais de saúde em 16 unidades de atenção primária foram treinados em protocolos simplificados de ecocardiografia portátil. Tele-ECGs foram registrados para diagnóstico final por um cardiologista. Pacientes consentidos com anormalidades maiores no ECG pelo código de Minnesota e uma amostra 1:5 de indivíduos normais foram submetidos a um questionário clínico e ecocardiograma de rastreamento interpretado remotamente. A doença cardíaca grave foi definida como doença valvular moderada/grave, disfunção/hipertrofia ventricular, derrame pericárdico ou anormalidade da motilidade. A associação entre alterações maiores do ECG e anormalidades ecocardiográficas foi avaliada por regressão logística da seguinte forma: 1) modelo não ajustado; 2) modelo 1 ajustado por idade/sexo; 3) modelo 2 mais fatores de risco (hipertensão/diabetes); 4) modelo 3 mais história de doença cardiovascular (Chagas/cardiopatia reumática/cardiopatia isquêmica/AVC/insuficiência cardíaca). Foram considerados significativos valores de p < 0,05. Resultados: No total, 1.411 pacientes realizaram ecocardiograma, sendo 1.149 (81%) com anormalidades maiores no ECG. A idade mediana foi de 67 anos (intervalo interquartil de 60 a 74) e 51,4% eram do sexo masculino. As anormalidades maiores no ECG se associaram a uma chance 2,4 vezes maior de doença cardíaca grave no ecocardiograma de rastreamento na análise bivariada (OR = 2,42 [IC 95% 1,76 a 3,39]) e permaneceram significativas (p < 0,001) após ajustes no modelo 2 (OR = 2,57 [IC 95% 1,84 a 3,65]), modelo 3 (OR = 2,52 [IC 95% 1,80 a 3,58]) e modelo 4 (OR = 2,23 [IC 95% 1,59 a 3,19]). Idade, sexo masculino, insuficiência cardíaca e doença cardíaca isquêmica também foram preditores independentes de doença cardíaca grave no ecocardiograma. Conclusões: As anormalidades do tele-ECG aumentaram a probabilidade de doença cardíaca grave no ecocardiograma de rastreamento, mesmo após ajustes para variáveis demográficas e clínicas. Abstract Background: Tele-cardiology tools are valuable strategies to improve risk stratification. Objective: We aimed to evaluate the accuracy of tele-electrocardiography (ECG) to predict abnormalities in screening echocardiography (echo) in primary care (PC). Methods: In 17 months, 6 health providers at 16 PC units were trained on simplified handheld echo protocols. Tele-ECGs were recorded for final diagnosis by a cardiologist. Consented patients with major ECG abnormalities by the Minnesota code, and a 1:5 sample of normal individuals underwent clinical questionnaire and screening echo interpreted remotely. Major heart disease was defined as moderate/severe valve disease, ventricular dysfunction/hypertrophy, pericardial effusion, or wall-motion abnormalities. Association between major ECG and echo abnormalities was assessed by logistic regression as follows: 1) unadjusted model; 2) model 1 adjusted for age/sex; 3) model 2 plus risk factors (hypertension/diabetes); 4) model 3 plus history of cardiovascular disease (Chagas/rheumatic heart disease/ischemic heart disease/stroke/heart failure). P-values < 0.05 were considered significant. Results: A total 1,411 patients underwent echo; 1,149 (81%) had major ECG abnormalities. Median age was 67 (IQR 60 to 74) years, and 51.4% were male. Major ECG abnormalities were associated with a 2.4-fold chance of major heart disease on echo in bivariate analysis (OR = 2.42 [95% CI 1.76 to 3.39]), and remained significant after adjustments in models (p < 0.001) 2 (OR = 2.57 [95% CI 1.84 to 3.65]), model 3 (OR = 2.52 [95% CI 1.80 to3.58]), and model 4 (OR = 2.23 [95%CI 1.59 to 3.19]). ABSTRACT Casselia, a small South American genus of Verbenaceae found in Brazil, Bolivia, and Paraguay, has often been misidentified within other angiosperm families. Based on a careful review of herbarium specimens plus fieldwork, the present study provides taxonomic, nomenclatural, and geographical distribution alignments in Casselia, while also shedding light on the reasons why it is a poorly known and overlooked genus. The novelties encompass the redefinition of the boundaries of C. confertiflora, recognition of C. confertiflora var. laciniata at specific level, reestablishment of C. zelota as an accepted species with an epitype designation, as well as geographic distribution readjustment of the C. integrifolia and C. serrata. Consequently, Casselia comprises eight species, occurring along the Atlantic Forest, Cerrado, and Caatinga domains. The only species with a narrow distribution is C. serrata. However, the genus is still poorly collected, and the species populations are not abundant. Furthermore, to aid in the recognition of Casselia and prevent misidentifications, a detailed description of the genus accompanied by plant photographs and an updated identification key are also provided. Niobium pentoxide shows an interesting reactivity that allows the control of different aspects of its morphology and chemistry. In this study, Nb2O5 nanoparticles were modified with protoporphyrin IX (PPIX) and tris(ethynylphenyl) pyrene derivative (PyPh3) by using 3-aminopropyltriethoxysilane as linkage group and used as photosensitizers against lung cancer. Abstract This study evaluated the nutritional and productive performance of Nellore purebred heifers and crossbred Brangus x Nellore (BGNE) and Braford x Nellore (BFNE) in a feedlot system. Thirty heifers (10 of each genetic group) with an average age of 18 months and an initial body weight of 261 kg were used. The experiment was structured and conducted according to a completely randomized design, with three treatments. Heifers received two diets (60 days each) during the experimental period. The experiment lasted 120 days with four experimental periods. Nellore heifers had a lower intake than crossbred heifers (P <0.05). There were no differences between BGNE and BFNE heifers, which had higher final body weight, average daily gain, feed efficiency, hot carcass weight and carcass length than NE heifers. Crossed heifers presented better fat cover than NE heifers. This study presents the synthesis and characterization of five Schiff bases derived from the reaction of 4-(2-aminoethyl)-benzenesulfonamide (compound 1) with corresponding aldehydes, (benzaldehyde, 2-pyridinecarboxaldehyde, 2-quinolinecarboxaldehyde, 8-hydroxy2-quinolinecarboxaldehyde and 4-imidazolecarboxaldehyde, for compounds 2-6, respectively). Characterization was performed by various spectroscopic techniques and supported by density functional theory (DFT) calculations. The crystal structures revealed how the substituent groups influenced the present supramolecular interactions. Compounds 1-4 and 6 showed no cytotoxicity to BHK-21 and VERO E6 cells at the highest concentration of 50 µmol L-1, while compound 5 was cytotoxic at this concentration. Compound 5 was active against the Chikungunya virus at the concentration of 10 µmol L-1, highlighting the effect of the 8-hydroxyquinoline substituent for the antiviral activity. For Zika virus, compound 6 was the only one active at 50 µmol L-1. ABSTRACT Using organic residues and bioinputs is a promising and sustainable practice to produce seedlings with forest essences, such as Cedrela fissilis Vell. (cedar), a vulnerable species close to extinction due to intensive exploitation in native areas. Thus, we aimed to evaluate the effect of different substrates based on organic residues associated with or without the application of Parachlorella sp. microalgae in the emergence and morphophysiology of C. fissilis seedlings. Sowing was carried out on six substrates: Oxisol with a clayey texture; Oxisol + Parachlorella sp.; Oxisol with sheep manure (3:1, v v–1); Oxisol with sheep manure + Parachlorella sp.; Oxisol with cattle manure (3:1, v v–1); and Oxisol with cattle manure + Parachlorella sp. The addition of organic residues to the soil, especially cattle manure, contributes to increasing the percentage of emergence, plant height, chlorophyll index, CO2 assimilation rate, and instantaneous carboxylation efficiency of Rubisco due to the superior chemical attributes in the substrate, which promote greater physiological efficiency. Organic residues increased the water use efficiency of seedlings. The application of Parachlorella sp. microalgae contributes to increases in the CO2 assimilation rate and stomatal conductance when seedlings are grown only in Oxisol. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract The threatened orchid Cattleya milleri is a microendemic orchid of a Brazilian savanna hotspot. As endophytes and mycorrhizae may improve its propagation and conservation, we investigated its root fungal community. Cattleya milleri roots were sampled in five natural sites and at a greenhouse. Fungal root endophytes were isolated for characterization and molecular ITS (Internal Transcribed Spacer) identification. Total DNA was extracted from endorhiza and rhizosphere for ITS amplification and sequencing. Sixteen fungal isolates were clustered in 6 Operational Taxonomic Units (OTUs) and endorhizal and rhizospheric sequences were clustered in 327 OTUs. Endorhiza presented from 25 to 89 OTUs, and rhizosphere 56 OTUs. Cluster analysis showed high divergence between natural and greenhouse fungal communities, but similarity among natural samples. From the 94 genera, 24 were annotated as endophytes, two mycorrhizas, 33 pathotrophs, 40 saprotrophs, and 17 symbiotrophs based on the FunGuild database. Endophytes of the orders Capnodiales, Hypocreales, Pleosporales, and mycorrhizae of Sebacinales occurred in all sites. The mycorrhizae Tulasnella occurred in all natural samples. The interaction with only two mycorrhizal taxa may limit C. milleri distribution. However, the recruitment of many non-mycorrhizal endophytes is essential to natural development. Abstract Candida albicans is the primary species causing oral candidiasis. Its increasing drug resistance drives the search for more effective antifungal agents. Therefore, we assessed toxicological parameters and the antimicrobial activity and mechanisms of action of the monoterpene (-)-fenchone against oral C. albicans. We conducted an in silico study using PASS online and AdmetSAR, followed by evaluation of antifungal activity through Minimum Inhibitory Concentration (MIC), Minimum Fungicidal Concentration (MFC), association study with miconazole, and assays with sorbitol and ergosterol. Inhibition of biofilm formation and disruption of preformed biofilm were considered. Toxicity was also assessed through hemolysis assay. The in silico study revealed a higher likelihood of the compound being active for antifungal activity, as well as promising pharmacokinetic and toxicity characteristics. Subsequently, (-)-fenchone exhibited predominantly fungicidal activity (MIC90 = 8 μg/mL; MFC = 16 μg/mL), including against miconazole-resistant C. albicans isolates. The substance does not appear to act by damaging the fungal cell wall or plasma membrane, and exhibited synergy with miconazole. There was activity in inhibiting biofilm formation but not in disrupting preformed biofilm. Finally, the product exerted low hemolytic activity at more than MIC×10. RESUMO O surgimento do HIV impactou os serviços de saúde abruptamente em 1981, inicialmente afetando grupos sociais restritos. Atualmente, com o desenvolvimento da terapia antirretroviral, nos deparamos com uma doença diferente da encarada na década de 80, agora com uma evolução crônica. Todavia, algumas barreiras permanecem as mesmas, como o impacto social, mental e físico do diagnóstico na vida da pessoa que vive com HIV. O objetivo do presente estudo é verificar carga viral, contagem de linfócitos CD4+, e qualidade de vida de pessoas que vivem com o vírus da imunodeficiência humana. A presente pesquisa trata-se de um estudo observacional transversal com amostra de 24 indivíduos de ambos os sexos, com sorologia positiva para HIV, em acompanhamento médico sob tratamento antirretroviral. Foi utilizado um instrumento próprio para a coleta de informações sociodemográficas. Os resultados de contagem de CD4+ e a Carga Viral do HIV foram coletados dos prontuários médicos dos pacientes. A Qualidade de vida (QV) foi avaliada através do WHOQOL-HIV-BREF. Foi utilizada a análise descritiva para a caracterização do perfil da amostra. Os participantes (46,8± 9,2 anos) possuem ensino médio completo (41,8%), com renda de um a três salários mínimos (37,5%). Apresentam de um a três anos de diagnóstico e contagem de linfócitos CD4 acima de 500 mm3 de sangue (75%). Em relação à QV, os participantes apresentaram média mais alta para o domínio físico (26,7± 5,3) e mais baixa para os domínios de meio ambiente (10,9± 2,7) e relações sociais (12,2± 2,8). O estudo conclui que as pessoas que vivem com HIV participantes do estudo são adultos jovens, com renda e nível de escolaridade baixas, apresentam baixa qualidade de vida em relação aos domínios meio ambiente e relações sociais.
ABSTRACT The emergence of HIV abruptly impacted health services in 1981, initially affecting restricted social groups. Currently, with the development of antiretroviral therapy, we are faced with a different disease from the one seen in the 80s, now with a chronic evolution. However, some barriers remain the same, such as the social, mental and physical impact of the diagnosis on the life of the person living with HIV. The objective of the present study was to verify the viral load, CD4+ lymphocyte count, and quality of life of people living with the human immunodeficiency virus. This was a cross-sectional observational research with a sample of 24 individuals of both sexes, with positive serology for HIV, in medical follow-up under antiretroviral treatment. A specific instrument was used to collect sociodemographic information. CD4+ count and HIV viral load results were collected from patients’ medical records. Quality of life (QoL) was assessed using the WHOQOL-HIV-BREF. Descriptive analysis was used to characterise the sample profile. Participants (46.8± 9.2 years) have completed high school (41.8%), with an income of one to three minimum wages (37.5%). They present a three-year diagnosis and CD4 lymphocyte count above 500 mm3 of blood (75%). Regarding QoL, the participants had a higher average for the physical domain (26.7± 5.3) and lower averages for the environment domains (10.9± 2.7) and social relationships (12.2± 2.8). The study concludes that the people living with HIV who participated in the study are young adults, with low income and level of education, with a low quality of life in relation to the environment and social relations domains.

Abstract Objective: To analyze the morphofunctional regeneration process of facial nerve injury in the presence of insulin-like growth factor-1 and mesenchymal stem cells. Methods: Fourteen Wistar rats suffered unilateral facial nerve crushing and were randomly divided into two groups. All received insulin-like growth factor-1 inoculation, but only half of the animals received an additional inoculation of mesenchymal stem cells. The animals were followed for 90 days and facial nerve regeneration was analyzed via spontaneous facial motor function tests and immunohistochemistry in the nerve motor nucleus. Results: The group that received the growth factor and stem cells showed a statistically superior mean in vibrissae movements (p<0.01), touch reflex (p = 0.05) and eye closure (p<0.01), in addition to better immunohistochemistry reactivity. There was a statistically significant difference in the mean number of cells in the facial nerve nucleus between the experimental groups (p = 0.025), with the group that received the growth factor and stem cells showing the highest mean. Conclusion: The association between growth factor and stem cells potentiates the morphofunctional regeneration of the facial nerve, occurring faster and more effectively.
