The reproductive biology of Marathrum rubrum Novelo & C. T. Philbrick: morphological and anatomical changes during flower development

Resumen Antecedentes: Las especies de la familia Podostemaceae poseen rasgos inusuales en angiospermas. Crecen sumergidas en ríos de corriente fuerte y su ciclo de vida está ligado a la estacionalidad. Debido a su estructura floral reducida, el análisis de las etapas de desarrollo y eventos como la antesis o la receptividad del estigma ha sido complicado. Pregunta: ¿Cuál es la relación entre la morfología, el desarrollo y la receptividad del estigma en diferentes etapas del desarrollo de la flor de Marathrum rubrum? Especie de estudio: Marathrum rubrum Novelo & C. T. Philbrick. Sitio y años de estudio: Río Horcones, Jalisco, México. Enero 2014 y 2015. Métodos: Se realizó un análisis in situ y en laboratorio de la morfología floral de M. rubrum para evaluar la relación entre la morfología y desarrollo floral, y la receptividad del estigma en distintas etapas. Resultados: Cuando las flores emergen de la espatela, los óvulos y el polen están completamente desarrollados, por ello, la esporogénesis y la gametogénesis ocurren dentro del tallo, estando las plantas aún sumergidas. Conclusiones: Los cambios morfológicos observados en las flores de M. rubrum durante las etapas analizadas se relacionan únicamente con eventos asociados con la fecundación y el desarrollo del fruto. Las observaciones detalladas de la biología reproductiva de M. rubrum en el ambiente aéreo tienen importantes implicaciones para el conocimiento básico de la biología de las plantas de esta familia, la estructura genética de las poblaciones, el éxito reproductivo y la biología de la conservación en ambientes amenazados.
Abstract Background: Podostemaceae is the largest family of strictly aquatic angiosperms. Species of this family possess unusual morphological and reproductive characters, grow attached to rocks in rivers with strong currents, and have a life cycle tightly linked to seasonality. Due to its reduced floral structures, which characterizes most species, the analysis of developmental stages and events such as anthesis or stigma receptivity has proven difficult. Question: How floral morphology, floral development and stigma receptivity of different stages of flower development of Marathrum rubrum Novelo & C. T. Philbrick are related among them? Studied species: Marathrum rubrum Novelo & C. T. Philbrick. Study site and dates: Horcones River, Jalisco, Mexico. January, 2014 and 2015. Methods: We made an in situ and lab analyses of flower morphology to assess the relationships between flower morphology, stages of development, reproductive structures, and stigma receptivity, during different stages of flower development. Results: When flowers emerge from the spathella, both the ovules and pollen grains are fully developed, implying that sporogenesis and gametogenesis take place within the stem while the plant is still submerged. Conclusions: The morphological changes observed in the flowers of M. rubrum during the analyzed stages are related only to events associated with fertilization and fruit development. The time-course of morphological changes in the flowers, from anthesis to fruit formation, lasts 20 to 30 days. The detailed observation of the aerial reproductive biology of M. rubrum bears important implications to the genetic structure of populations, plant fitness and conservation biology in threatened environments.

The indiscriminate use of herbicides makes weeds resistant, increasing the demand for more efficient herbicides. Novel potential herbicides were prepared from sorbic acid, being 8 dienamides and 6 epoxides. Subsequently, phytotoxic activities of sorbic acid and 14 synthesized compounds were evaluated against sorghum, onion, cucumber, lettuce, and beggartick seeds. Furthermore, the protein target of the most active substance in plants was identified in silico. Among the tested compounds, (2E,4E)-N-(p-fluorophenyl)hexa-2,4-dienamide (2c), (2E,4E)-N -(o-naphthyl) hexa-2,4-dienamide (2d), (E)-N-methylphenyl-3-((2R,3R)-3-methyloxiran-2-yl)acrylamide (3b), (E)-N-bromophenyl-3-((2R,3R)-3-methyloxiran-2-yl)acrylamide (3e), and (E)-N-methoxyphenyl-3-((2R,3R)-3-methyloxiran-2-yl)acrylamide (3f) showed pronounced inhibitory activities, both of shoots and roots, making them candidates for promising herbicides. All compounds altered seed development, and compounds 2c, 2d, 3b, 3e, and 3f presented equal or superior results to the commercial control herbicide S-metolachlor. Food fraud, such as the adulteration of extra virgin olive oil (EVOO), is found to cause substantial negative impacts on both the economy and the health of consumers. This work was aimed at evaluating 19 EVOO samples commercialized in Foz do Iguaçu, a frontier city located around the triple border of Brazil, Argentina, and Paraguay. To detect the presence of adulteration in EVOO samples, the present study employed gas chromatography coupled to flame ionization detector (GC-FID) and headspace solid phase microextraction with gas chromatography coupled to mass spectrometry (HS-SPME-GC-MS) in order to evaluate fatty acids composition and volatile organic compounds (VOCs), respectively. The quantitative results obtained from the analysis of fatty acids composition showed that 32% of the EVOO samples were adulterated, presumably with soy oil, due to the high levels of linoleic, linolenic, and myristic acids present in them. Principal component analysis (PCA) conducted using the complete chromatographic aroma profiles obtained from the VOCs helped distinguish authentic EVOO samples from adulterated ones and the country of origin of the samples. The microwave-assisted interesterification of babassu oil with methyl, ethyl and butyl acetates was carried out via acid catalysis, using sulfuric, phosphoric, methanesulfonic and acetic acids for the production of biodiesel with triacetin as an additive. Dielectric measurements in the range of 0.3 to 13 GHz of pure reactants and reactions media lead to a clear understanding of the interaction between the medium and microwave irradiation, allowing achieving optimal conditions in terms of microwave power, kind and catalyst concentration, reactant molar ratios, and temperatures. Using ethyl acetate, reactions were carried out at various oil to acetate molar ratio (1:12 to 1:72) using sulfuric acid (0.5% m/mT) as a catalyst. For 1:60 molar ratio, the experiments were carried out with 0.5 to 2% m/mT sulfuric acid. The kind of catalyst and the applied microwave power to mass ratio were investigated using H3PO4, CH3COOH and CH3SO3H. The best catalytic activities were achieved with H2SO4 and CH3SO3H. It was found that under microwave or conventional heating the reactivity follows the ethyl→methyl→butyl trend. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Resumo Ensaios pré-clínicos demonstraram que uma biomembrana composta de álcool polivinílico a 1%, contendo proteínas de látex (10%) obtidas da planta medicinal Calotropis procera (Aiton) era biocompatível e estimulava a cicatrização de feridas incisionais e excisionais em modelos murinos, e aspectos mecanísticos foram estabelecidos. No estudo atual, foi investigada a eficácia da biomembrana (BioMemCpLP) em promover a cicatrização de úlceras crônicas em pacientes acometidos de hanseníase. O estudo foi iniciado com 28 voluntários. Cinco foram excluídos, posteriormente, por diferentes desconformidades. Úlceras de 15 pacientes foram tratadas continuamente com BioMemCpLP durante 56 dias. Outros cinco pacientes foram tratados apenas com sulfadiazina de prata e mais três pacientes receberam curativos simples de hidrocoloide com alta capacidade de absorção. Em todos os casos, os curativos foram renovados três vezes por semana e as úlceras foram avaliadas semanalmente quanto à contração e ao progresso da cicatrização, durante 56 dias. A extensão da área cicatrizada, nas úlceras tratadas com BioMemCpLP, foi maior do que nos dois grupos controles. Aproximadamente 88% das úlceras tratadas com BioMemCpLP foram totalmente curadas, antes do dia 56 de tratamento, contra 6% em ambos os grupos de controle. Esse resultado não foi correlacionado com idade/sexo, tempo de lesão ou localização das úlceras, deformidade ou se o paciente alcançou ou não a cura da hanseníase. Abstract Pre-clinical assays demonstrated that a 1% polyvinyl alcohol biomembrane containing latex proteins (10%) from the medicinal plant Calotropis procera was biocompatible and stimulated healing of incisional and excisional wounds in murine models, and the mechanistic aspects were established. The efficacy of the biomembrane (BioMemCpLP) to promote healing of chronic ulcers in leprosy patients was investigated. The study started with 28 volunteers. Five were excluded later due to different disconformities. Ulcers from 15 patients were continuously treated with BioMemCpLP for 56 days. Five patients were treated only with silver sulfadiazine and three patients received plain hydrocolloid wound dressings with high absorption capacity. In all cases, wound dressings were renewed three times a week for 56 days and ulcers were evaluated weekly for contraction and healing progress. The extent of the healed area in the ulcers treated with BioMemCpLP was greater than in the control groups. Approximately 88% of ulcers treated with BioMemCpLP were fully healed before day 56, against 6% in both control groups. This result was not correlated with age/gender, duration or location of ulcers, deformity or whether or not the patient was cured of leprosy. The results showed that BioMemCpLP was beneficial for treatment of ulcers suffered by leprosy patients without noticeable side effects. ABSTRACT Introduction: Immunosenescence is associated with an increased risk of cancer development. Among the malignant hemopathies that affect this age group, it is chronic lymphoid leukemia (CLL), characterized by disorders in adaptive immunity, which include subpopulations of T lymphocytes. Objective: To determine frequency of T lymphocyte subpopulations in older adult patients with chronic lymphoid leukemia evaluated at the Institute of Hematology and Immunology of Cuba. Methods: A cross-sectional study was conducted in 30 older adults with chronic lymphoid leukemia. TCD3+CD4+ and TCD3+CD8+ lymphocytes were quantified in peripheral blood by flow cytometry. A Beckman Coulter Gallios flow cytometer was used to read and analyze the data. The percentage values, the mean and the standard deviation were used. It was considered statistically significant if p≤0.05. Results: There was a predominance of men who represented 56.7% and the age group of 70-79 years. No older adults with CLL with high or normal values of TCD3+CD4+ lymphocytes were reported. Men predominated with low percentage values of TCD3+CD4+, TCD3+CD8+ lymphocytes and inversion of the CD4/CD8 ratio in relation to women. Conclusions: Older adult with CLL present alterations in the number of T lymphocyte subpopulations. The role of these cells in relation to the growth of malignant B cells it is unknown and it turns out important to determine if this may reflect an attempt to evade tumor cells from immune control.
RESUMEN Introducción: la inmunosenescencia está asociada con un mayor riesgo de desarrollo de cáncer. Dentro de las hemopatías malignas que afectan a este grupo de edad, está la leucemia linfoide crónica (LLC), caracterizada por trastornos en la inmunidad adaptativa que incluye las subpoblaciones de linfocitos T. Objetivo: Determinar la frecuencia de las subpoblaciones de linfocitos T en los pacientes adultos mayores con leucemia linfoide crónica evaluados en el Instituto de Hematología e Inmunología de Cuba. Métodos: Se realizó un estudio transversal en 30 adultos mayores con leucemia linfoide crónica. Se cuantificaron los linfocitos TCD3+CD4+ y TCD3+CD8+ en sangre periférica por citometría de flujo. Para la lectura y el análisis de los datos se empleó un citómetro de flujo Beckman Coulter Gallios. Se utilizaron los valores porcentuales, la media y la desviación estándar. Se consideró estadísticamente significativo si p≤0.05. Resultados: Hubo un predominio de hombres que representaron el 56,7 % y del grupo de 70-79 años de edad. No se reportó ningún adulto mayor con LLC con valores altos ni normales de linfocitos TCD3+CD4+. Predominaron los hombres con valores bajos porcentuales de linfocitos TCD3+CD4+, TCD3+CD8+ e inversión del índice CD4/CD8 en relación con las mujeres. Conclusiones: Los adultos mayores con LLC presentan alteraciones en el número de las subpoblaciones de linfocitos T. La acción de estas células en relación al crecimiento de células B malignas aún es desconocido y resulta importante determinar si esto puede reflejar un intento de evasión de las células tumorales al control inmunológico.

Resumo Toxoplasma gondii é um coccídio que infecta quase todos os animais de sangue quente, incluindo aves. A Reserva Biológica do Atol das Rocas, localizada no Nordeste do Brasil, é o único atol do Atlântico Sul que abriga a maior população de aves marinhas tropicais do oeste do Oceano Atlântico. O objetivo deste estudo foi verificar a ocorrência de anticorpos anti-T. gondii em aves marinhas do Atol das Rocas que foram capturadas manualmente. Amostras de sangue foram obtidas e anticorpos avaliados pelo teste de aglutinação modificado (>5). Foram amostradas 267 aves de sete espécies, pertencentes a três famílias (Sternidae, Fregatidae e Sulidae) e duas ordens (Charadriiformes e Suliformes). Das 267 aves, 20 (7,3%) foram soropositivas sendo: nove das 48 viuvinhas-marrons (Anous stolidus), uma das 26 viuvinhas-negras (Anous minutus), três das 20 fragatas (Fregata magnificens), cinco dos 95 trinta-réis-das-rocas (Onychoprion fuscatus) e dois dos 20 atobás-de-pés-vermelhos (Sula sula). Nenhum dos 33 atobás grandes (Sula dactylatra) e dos 25 atobás-pardos (Sula leucogaster) estavam soropositivos. Os títulos de anticorpos foram 5 (n=6), 10 (n=4), 20 (n=3), 40 (n=6) e 160 (n=1). Abstract Toxoplasma gondii is a coccidian that infects almost all warm-blooded animals, including birds. Rocas Atoll Biological Reserve, located in the northeast of Brazil, is the only atoll in the South Atlantic, and home to the largest population of seabirds in the western Atlantic. In this study the occurrence of T. gondii antibodies in seabirds from Rocas Atoll were determined. Birds were manually captured, blood samples were taken, and antibodies detected by the modified agglutination test (>5). In total, 267 birds of seven species belonging to three families (Sternidae, Fregatidae and Sulidae) and two orders (Charadriiformes and Suliformes) were sampled. Out of the 267 samples, 20 (7.3%) were seropositive: nine out of 48 brown noddies (Anous stolidus), one out of 26 black noddies (Anous minutus), three out of 20 magnificent frigatebirds (Fregata magnificens), five out of 95 sooty terns (Onychoprion fuscatus) and two out of 20 red-footed boobies (Sula sula). None of the 33 masked boobies (Sula dactylatra) and the 25 brown boobies (Sula leucogaster) were seropositive. The antibody titers were 5 (n=6), 10 (n=4), 20 (n=3), 40 (n=6) and 160 (n=1). Essential oils (EO) have diverse applications, such as antibacterial and antifungal activity. However, they are susceptible to oxidation in the presence of air, light, and moisture. In addition, they are thermally unstable. In this sense, it is necessary to develop techniques to increase the lifespan of EO. In this work, EO was obtained from cloves and characterized by different techniques. The major compounds found were eugenol (83%), eugenol acetate (9%), and β-caryophyllene (8%). The EO was encapsulated by extrusion using a microfluidic device. The sodium alginate was used as wall material, and the CaCl2 solution was used as a crosslinking agent. The microcapsule presented sizes of 164.7 ± 0.3 μm, with an encapsulation yield of 64 ± 14%. Functional characteristic groups of EO were observed in the microcapsule by infrared and Raman spectroscopies. The microcapsule increased the thermal decomposition of the EO from 162 to 230 °C. Release kinetics of the capsule was performed, with an equilibrium time of 72 h and release of 54% of the EO. Finally, the pure EO and encapsulated EO-microcapsules were applied in the Aedes aegypti larvae control, showing mean lethal concentration necessary to eliminate 50% (LC50) values of 74.4 and 96.9 μg mL-1 and lethal concentration necessary to eliminate 90% (LC90) of 106.2 and 133.3 μg mL-1 for pure EO and encapsulated EO, respectively. ABSTRACT The almond (Prunus dulcis) is an economically important crop in Portugal and is in the process of expansion. The assessment of the diversity of arthropods existing in the almond tree canopy becomes essential to know the trophic relationships that are established within it. The aim of this work was to identify the main arthropods present in the almond tree canopy in different soil management systems and water conditions. The trial was carried out in two almond groves, in Corujas, Macedo de Cavaleiros: rainfed and irrigated. They were subdivided into two treatments: 1) mobilized soil and 2) spontaneous vegetation, each with 3 blocks and 5 replications. Modalities 1 and 2 were installed in May 2022. Samples were collected fortnightly, between July and September 2022, performing 6 taps/tree/modality, using the tapping technique, to assess the auxiliary fauna and 20 leaves/ tree to monitor Monosteira unicostata (Hemiptera: Tingidae), Panonychus ulmi (Acari: Tetranychidae) and Tetranychus urticae (Acari: Tetranychidae). Posteriorly, the individuals were counted and their identification was made at order and family level, for the auxiliary fauna and at species level for the pests. The most frequent orders were Hymenoptera (30%) and Araneae (18%). There were no significant differences in the number of arthropods between treatments in the two almond groves. With regard to pests, T. urticae and M. unicostata predominated in both orchards, with greater abundance in the irrigated grove.
RESUMO A amendoeira (Prunus dulcis) é uma cultura economicamente importante em Portugal e atualmente encontra-se em fase de expansão. A avaliação da diversidade de artrópodes existente na copa da amendoeira torna-se essencial para conhecer as relações tróficas que se estabelecem no seu interior. O objetivo deste trabalho foi identificar os principais artrópodes presentes na copa das amendoeiras em diferentes sistemas de gestão do solo e condições hídricas. O ensaio foi conduzido em dois amendoais, em Corujas, Macedo de Cavaleiros: sequeiro e regadio. Eles foram subdivididos em dois tratamentos: 1) solo mobilizado e 2) vegetação espontânea, cada um com 3 blocos e 5 repetições. As modalidades 1 e 2 foram instaladas em maio de 2022. As amostras foram recolhidas quinzenalmente, entre julho e setembro de 2022, realizando-se 6 batidas/árvore/modalidade, na técnica das pancadas, para avaliar a fauna auxiliar e colhidas 20 folhas/árvore para monitorizar Monosteira unicostata (Hemiptera: Tingidae), Panonychus ulmi (Acari: Tetranychidae) e Tetranychus urticae (Acari: Tetranychidae). Posteriormente, contaram-se os indivíduos e a sua identificação foi feita ao nível de ordem e família para a fauna auxiliar e de espécie para as pragas. As ordens mais frequentes foram Hymenoptera (30%) e Araneae (18%). Não se verificaram diferenças significativas no número de artrópodes entre os tratamentos, nos dois amendoais. Quanto às pragas, predominaram T. urticae e M. unicostata nos dois amendoais, verificando-se maior abundância no amendoal de regadio.

Abstract Some experimental results concerning the performance of fiber-reinforced soil composites made of soil and shredded fibers from waste tires are presented. The composite mixtures were prepared with fiber addition of 0, 15, 30, and 45 wt%. After molding, the specimens were oven-dried and tested for unconfined compressive strength and water immersion durability. The results showed an improvement in durability with the addition of fiber. The results also indicated good compressive strength performance for the composites with a fiber content of 15 and 30 wt%. This study aimed to evaluate the effect of gamma irradiation (GI) on the physicochemical, technological, antioxidant and microbiological characteristics of different whole sorghum flours (WSF), as well as to characterize the profile of chemical constituents by paper spray mass spectrometry (PS-MS). The doses applied interfered in the staining characteristics of the evaluated flours, providing the darkening of the same. For the other physicochemical, technological, and antioxidant parameters, no significant influence of the process was observed. The fingerprint obtained in both ionization modes had not been influenced by irradiation, being flavonoids, phenylpropanoids, amino acids, benzoic acid derivatives, carboxylic acids, and sugars tentatively identified. In microbiological terms, there was a reduction of molds, yeasts, and Bacillus cereus in irradiated WSF compared to control (non-irradiated). Therefore, the application of low doses of gamma irradiation represents an advantageous alternative for the conservation of WSF and maintenance of bioactive compounds identified by the PS-MS technique. Resumen El estado de Nuevo León es uno de los principales productores de naranja en México, ya que cuenta con una superficie sembrada de 25 820 ha y una producción anual de 339 138 t. El cultivar que predomina es la naranja Valencia, que se caracteriza por presentar un ciclo de maduración tardío, lo cual favorece que la producción en la región se concentre en un periodo relativamente corto (febrero-abril), aunque puede permanecer en el árbol hasta junio si hay disponibilidad de agua. Por esta razón, es necesario ofrecer a los productores otros cultivares, que sean alternativas tanto en rendimiento como en su comercialización. El objetivo de este estudio fue evaluar y caracterizar a un grupo de cultivares de naranja de maduración temprana considerando algunos parámetros que inciden en la calidad del fruto, tales como el peso fresco, diámetro polar y ecuatorial, grosor de cáscara, número de semillas, oBrix, contenido de jugo y su dureza para el manejo postcosecha. Se utilizaron siete cultivares de naranja temprana conservados en el Banco de Germoplasma de Cítricos del Instituto Nacional de Investigaciones Forestales, Agrícolas y Pecuarias establecido en el Campo Experimental General Terán. De acuerdo con las características de calidad del fruto, los cultivares de naranja temprana se clasificaron en tres grupos diferentes: el primer grupo está representado por Marrs que tiene características sobresalientes, el segundo grupo está integrado por Parson Brown, Queen, San Miguel, Pineapple y Hamlin con características sobresalientes e intermedias y el tercer grupo está integrado por Temprana que tiene características sobresalientes y bajas. Los resultados obtenidos sugieren que existe una amplia variación entre los materiales de naranja temprana en el estado de Nuevo León, constituyéndose como una alternativa viable para diversificar la producción y no depender exclusivamente del cultivar Valencia.
Abstract The state of Nuevo León is one of the main producers of oranges in Mexico, since it has a planted area of 25 820 ha and an annual production of 339 138 t. The predominant cultivar is the Valencia orange, which is characterized by a late maturation cycle, which favors that production in the region concentrates in a relatively short period (February-April), although it can remain on the tree until June if water is available. For this reason, it is necessary to offer producers other cultivars, which are alternatives both in yield and in their commercialization. The objective of this study was to evaluate and characterize a group of early-maturing orange cultivars considering some parameters that affect fruit quality, such as fresh weight, polar and equatorial diameter, peel thickness, number of seeds, oBrix, juice content and its hardness for postharvest handling. Seven early orange cultivars preserved in the Citrus Germplasm Bank of the National Institute of Forestry, Agricultural and Livestock Research, established in the General Terán Experimental Field, were used. According to the quality characteristics of the fruit, the early orange cultivars were classified into three different groups: the first group is represented by Marrs which has outstanding characteristics, the second group is composed of Parson Brown, Queen, San Miguel, Pineapple and Hamlin with outstanding and intermediate characteristics and the third group is composed of Temprana which has outstanding and low characteristics. The results obtained suggest that there is a wide variation between early orange materials in the state of Nuevo León, constituting themselves as a viable alternative to diversify production and not depend exclusively on the cultivar Valencia.

Low Expression of E-Cadherin and Cdh1 Variants Associated with Diffuse Gastric Cancer

ABSTRACT Background Reduced or null expression of E-cadherin protein is a frequent cause of diffuse gastric cancer (DGC). More than 50% of patients with DGC present somatic variants in CDH1 gene. Objectives The objectives of this study were to study E-cadherin expression and identify variants in the CDH1 gene in gastric tumors of patients with DGC. Methods We studied 18 Mexican DGC patients who attended a hospital of the Mexican Social Security Institute; E-cadherin expression was determined by immunohistochemistry, and variants were identified by Sanger sequencing in promoter and coding regions. Predictive analysis was performed using PolyPhen-2 and HOPE software. Results We found that 56% of DGC patients showed reduced expression of E-cadherin. All patients carried CDH1 variants; overall, 12 different CDH1 variants were identified. Predictive analysis revealed that the rs114265540 variant was probably damaging, with a value of 0.985, indicating a functional impact on the E-cadherin protein. Variants rs34939176 and rs33964119 were identified as risk factors for DGC (odds' ratios [OR] = 31.3, 95% CI 6.3-154.0, p < 0.001; OR = 6.1, 95% CI 2.0-19.0, p < 0.001, respectively) given their elevated frequency and by comparing it with those reported for MXL population in the 1000 Genomes Project database. Conclusions In this Mexican population, the percentage of diffuse gastric tumors with reduced expression of E-cadherin was similar to that reported in other populations. All gastric tumors of DGC patients studied had somatic CDH1 gene variants; however, the rs114265540, rs34939176, and rs33964119 variants were importantly related to DGC.

Abstract This study aimed to determine the occurrence of anti-Toxoplasma gondii, Neospora caninum, and Leptospira spp. antibodies in sheep and goats raised in villages of the Xukuru do Ororubá indigenous community, Pernambuco, Brazil. A total of 180 serum samples from sheep and 108 serum samples from goats of both sexes and different ages were analyzed. For antibody research, indirect immunofluorescence antibody test (IFAT) were used for the protozoa T. gondii and N. caninum, and microscopic agglutination test (MAT) for Leptospira spp., with a cutoff titer of 1:64, 1:50 and 1:100, respectively. The frequency of anti-T. gondii antibodies was 16.6% (30/180) for sheep and 11.1% (12/108) for goats. The frequency of anti-N. caninum antibodies was 10.55% (19/180) for sheep, and 20.37% (22/108) for goats, while for Leptospira spp., 2.2% (4/180) of sheep and 1.85% (2/108) of goats reacted positively. Resumo Objetivou-se determinar a ocorrência de anticorpos anti-Toxoplasma gondii, Neospora caninum e Leptospira spp., em ovinos e caprinos criados em aldeias da comunidade indígena Xukuru do Ororubá, Pernambuco, Brasil. Foram analisadas 180 amostras de soro de ovinos e 108 amostras de soro de caprinos de ambos os sexos e diferentes idades. Para a pesquisa de anticorpos foi utilizada a técnica de Reação de Imunofluorescência indireta (RIFI), para os protozoários T. gondii e N. caninum e Aglutinação Microscópica (MAT) para Leptospira spp., com ponto de corte de 1:64, 1:50 e 1:100, respectivamente. A frequência de anticorpos anti-T gondii foi de 16,6% (30/180) em ovinos e 11,1% (12/108) em caprinos. A frequência de anticorpos anti-N. caninum foi de 10,55% (19/180) para ovinos e 20,37% (22/108) para caprinos, enquanto para Leptospira spp., 2,2% (4/180) dos ovinos e 1,85% (2/108) dos caprinos reagiram positivamente. Os resultados obtidos neste estudo são inéditos em comunidades indígenas do país e alertam para o monitoramento de caprinos e ovinos da aldeia indígena Xukuru do Ororubá, quanto à ocorrência e impacto produtivo de infecções por T. gondii, N. caninum e Leptospira spp., além da ocorrência de zoonoses como a toxoplasmose e leptospirose na comunidade indígena.
