ABSTRACT Inactivated COVID-19 vaccines data in immunocompromised individuals are scarce. This trial assessed the immunogenicity of two CoronaVac doses and additional BNT162b2 mRNA vaccine doses in immunocompromised (IC) and immunocompetent (H) individuals. Adults with solid organ transplant (SOT), hematopoietic stem cell transplant, cancer, inborn immunity errors or rheumatic diseases were included in the IC group. Immunocompetent adults were used as control group for comparison. Participants received two CoronaVac doses within a 28-day interval. IC received two additional BNT162b2 doses and H received a third BNT162b2 dose (booster). Blood samples were collected at baseline, 28 days after each dose, pre-booster and at the trial end. We used three serological tests to detect antibodies to SARS-CoV-2 nucleocapsid (N), trimeric spike (S), and receptor binding domain (RBD). Outcomes included seroconversion rates (SCR), geometric mean titers (GMT) and GMT ratio (GMTR). A total of 241 IC and 100 H adults participated in the study. After two CoronaVac doses, IC had lower SCR than H: anti-N, 33.3% vs 79%; anti-S, 33.8% vs 86%, and anti-RBD, 48.5% vs 85%, respectively. IC also showed lower GMT than H: anti-N, 2.3 vs 15.1; anti-S, 58.8 vs 213.2 BAU/mL; and anti-RBD, 22.4 vs 168.0 U/mL, respectively. After the 3rd and 4th BNT162b2 doses, IC had significant anti-S and anti-RBD seroconversion, but still lower than H after the 3rd dose. After boosting, GMT increased in IC, but remained lower than in the H group. CoronaVac two-dose schedule immunogenicity was lower in IC than in H. Abstract Objective To review current literature to support the use of mesna as a preventive therapy for hemorrhagic cystitis and bladder cancer in patients with systemic autoimmune diseases and systemic vasculitis treated with cyclophosphamide. Materials and methods The search for articles was conducted systematically through MEDLINE, LILACS, Cochrane Library, and Embase databases. Only articles in English were selected. For available records, titles and abstracts were selected independently by two investigators. Results Eighteen studies were selected for analysis. The known adverse effects of cyclophosphamide were hematological toxicity, infections, gonadal toxicity, teratogenicity, increased risk for malignancy and hemorrhagic cystitis. Long-term toxicity was highly dependent on cyclophosphamide cumulative dose. The risk of bladder cancer is especially higher in long-term exposure and with cumulative doses above 36 g. The risk remains high for years after drug discontinuation. Hemorrhagic cystitis is highly correlated with cumulative dose and its incidence ranges between 12 and 41%, but it seems to be lower with new regimens with reduced cyclophosphamide dose. No randomized controlled trials were found to analyze the use of mesna in systemic autoimmune rheumatic diseases and systemic vasculitis. Retrospective studies yielded conflicting results. Uncontrolled prospective studies with positive results were considered at high risk of bias. No evidence was found to support the use of mesna during the treatment with cyclophosphamide for autoimmune diseases or systemic vasculitis to prevent hemorrhagic cystitis and bladder cancer. In the scenarios of high cumulative cyclophosphamide dose (i.e., > 30 g), patients with restricted fluid intake, neurogenic bladder, therapy with oral anticoagulants, and chronic kidney disease, mesna could be considered. Conclusion The current evidence was found to be insufficient to support the routine use of mesna for the prophylaxis of hemorrhagic cystitis and bladder cancer in patients being treated for systemic autoimmune diseases and systemic vasculitis with cyclophosphamide. The use may be considered for selected cases. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Resumo Objetivo Analisar o trofismo muscular e a de interleucina-6 no músculo bíceps braquial de ratas com lesão medular cervical incompleta tratados com estimulação elétrica neuromuscular (EENM). Métodos Ratas adultas foram submetidas à hemissecção da medula espinal em C5-C7 e a um protocolo de EENM de 5 semanas. O trofismo do bíceps braquial foi avaliado pela relação peso muscular/peso corporal e análise histológica. A expressão de interleucina-6 no bíceps braquial foi medida usando ensaio de imunoabsorção enzimática. Resultados O grupo tratado com EENM apresentou preservação do trofismo muscular, assim como prevenção da redução dos níveis de interleucina-6. Conclusão A lesão da medula espinal causa atrofia muscular e diminui a expressão de interleucina-6. Essas alterações são parcialmente prevenidas pela EENM. Abstract Objective To analyze the muscle trophism and expression of interleukin-6 in the biceps brachii muscle of rats with incomplete cervical spinal cord injury treated with neuromuscular electrical stimulation (NMES). Methods Adult rats underwent C5-C7 spinal cord hemisection and a 5-week NMES protocol. Trophism of the biceps brachii was assessed using muscle weight/body weight ratio and histological analysis. Interleukin-6 expression from biceps brachii was measured using the enzyme-linked immunosorbent assay technique. Results Preservation of the biceps brachii muscle trophism was found in the NMES treated group, along with prevention of interleukin-6 level reduction. Conclusion Spinal cord injury causes muscle atrophy and decreases interleukin-6 levels. These alterations are partially prevented by NMES. Abstract Sporothrix spp. are the major dimorphic fungus associated with a type of subcutaneous mycosis, sporotrichosis. The limitation of antifungal availability and the past reports of in vitro resistance of Sporothrix spp. clinical isolates makes it important to search for new compounds with antifungal activities. In this study, we therefore evaluate the in vitro activities of complexes coordinated with Co(II) and cobalt chloride hexahydrate against clinical isolates of Sporothrix spp. Broth microdilution test was performed as per M38-A2 from CLSI (2008) in duplicate for 31 clinical isolates of Sporothrix spp. (27 S. brasiliensis e 04 S. schenckii stricto sensu). The antifungal activities of the complexes coordinated with Co(II) and cobalt chloride hexahydrate were detected at a concentration range of 32-128 µg/mL for all isolates. None of the compounds demonstrated any cytotoxicity (to macrophage cells) at the concentration of 200 µg/mL. The activity against Sporothrix spp. recorded in this study instigate the continuity of experimental studies with Co(II) to search for the mechanisms of antifungal action as well as to evaluate its interaction with the commercial antifungal drugs.

ABSTRACT It is essential to determine the productive potential and quality of forage sorghum (Sorghum bicolor (L.) Moench) silages using alternative and organic inputs, such as poultry litter (PL), intending to higher efficiency, and sustainable development of animal production systems in the world. Poultry litter fertilization has been widely used in crops for silage production, where it is believed that fertilization with PL can influence the fermentation profile and composition of forage sorghum silages. The objective of this study was to evaluate different levels of fertilization with PL on the agronomic and quality characteristics of forage sorghum silage ‘BRS Ponta Negra’. For determination of agronomic and productive characteristics, randomized blocks design was adopted, the treatments were different PL levels: 5, 10, and 15 Mg ha-1. While for the evaluation of silage characteristics, the design was completely randomized. With fertilizer doses, a linear increase was observed for plant height, stem diameter, leaf length, and consequently for fresh and DM productions. Linear reduction was observed for neutral detergent fiber, while crude protein and pH contents of silages increased linearly with PL levels. Quadratic effect occurred for gas and effluent losses. Poultry litter may be an alternative source to supply the N needs in the production of sorghum for silage under experimental similar conditions. Associating productivity with cultivation efficiency, the PL level 13.46 Mg ha-1 is recommended for presenting the highest N agronomic efficiency and good nutritional quality of the silage.

Abstract Cardiac hypertrophy and dysfunction are a significant complication of chronic Chagas disease, with heart failure, stroke, and sudden death related to disease progression. Thus, understanding the signaling pathways involved in the chagasic cardiac hypertrophy may provide potential targets for pharmacological therapy. Herein, we investigated the implication of focal adhesion kinase (FAK) signaling pathway in triggering hypertrophic phenotype during acute and chronic T. cruzi infection. C57BL/6 mice infected with T. cruzi (Brazil strain) were evaluated for electrocardiographic (ECG) changes, plasma levels of endothelin-1 (ET-1) and activation of signaling pathways involved in cardiac hypertrophy, including FAK and ERK1/2, as well as expression of hypertrophy marker and components of the extracellular matrix in the different stages of T. cruzi infection (60–210 dpi). Heart dysfunction, evidenced by prolonged PR interval and decrease in heart rates in ECG tracing, was associated with high plasma ET-1 level, extracellular matrix remodeling and FAK signaling activation. Upregulation of both FAK tyrosine 397 (FAK-Y397) and serine 910 (FAK-S910) residues phosphorylation as well as ERK1/2 activation, lead to an enhancement of atrial natriuretic peptide gene expression in chronic infection. Our findings highlight FAK-ERK1/2 signaling as a regulator of cardiac hypertrophy in Trypanosoma cruzi infection. Both mechanical stress, induced by cardiac extracellular matrix (ECM) augment and cardiac overload, and ET-1 stimuli orchestrated FAK signaling activation with subsequent activation of the fetal cardiac gene program in the chronic phase of infection, highlighting FAK as an attractive target for Chagas disease therapy.

RESUMO O cisto linfoepitelial oral (CLEO) é uma lesão incomum, cuja patogênese ainda é pouco elucidada. O objetivo deste estudo é relatar um caso de CLEO. Paciente do sexo feminino, leucoderma, 62 anos, apresentou aumento de volume nodular assintomático de consistência amolecida, em borda lateral da língua. Sob a hipótese clínica de hiperplasia de tecido linfoide, biópsia excisional foi realizada. O exame histopatológico revelou cavidade patológica revestida por epitélio e uma cápsula cística de tecido conjuntivo, contendo tecido linfoide. O diagnóstico de CLEO foi estabelecido. A paciente não apresentou sinais de recidiva após a remoção cirúrgica da lesão.
ABSTRACT Oral lymphoepithelial cyst (OLEC) is an uncommon lesion whose pathogenesis remains poorly understood. The purpose of this paper is to report a case of OLEC. Female patient, white, 62 years old, presented asymptomatic nodular swelling of soft consistency in the lateral border of the tongue. Under the clinical hypothesis of lymphoid tissue hyperplasia, an excisional biopsy was performed. Histopathological examination revealed a pathological epithelial-lined cavity and a cystic connective tissue capsule containing lymphoid tissue. The diagnosis of OLEC was established and the patient showed no signs of recurrence after surgical removal of the lesion.
RESUMEN El quiste linfoepitelial (QLE) oral es una lesión infrecuente, cuya patogénesis es aún poco conocida. El objetivo del presente estudio es reportar un caso de QLE oral. Mujer blanca de 62 años presentó un crecimiento nodular asintomático de consistencia blanda, en borde lateral de la lengua. Bajo la hipótesis clínica de hiperplasia del tejido linfoide, se realizó una biopsia excisional. El examen histopatológico reveló cavidad patológica revestida de epitelio y una cápsula quística de tejido conectivo, conteniendo tejido linfoide. Se estableció el diagnóstico de QLE oral. La paciente no ha presentado recidiva tras extirpación quirúrgica de la lesión.

Abstract RNA interference (RNAi) is a powerful gene silencing technology, widely used in analyses of reverse genetics, development of therapeutic strategies and generation of biotechnological products. Here we present a free software tool for the rational design of RNAi effectors, named siRNA and shRNA designer (SSD). SSD incorporates our previously developed software Strand Analysis to construct template DNAs amenable for the large scale production of mono-, bi- and trivalent multimeric shRNAs, via in vitro rolling circle transcription. We tested SSD by creating a trivalent multimeric shRNA against the vitellogenin gene of Apis mellifera. RT-qPCR analysis revealed that our molecule promoted a decrease in more than 50% of the target mRNA, in a dose-dependent manner, when compared to the control group. Thus, SSD software allows the easy design of multimeric shRNAs, for single or multiple simultaneous knockdowns, which is especially interesting for studies involving large amounts of double-stranded molecules.

Abstract Cadmium (Cd) is one of the most toxic elements to plants, so Cd-tolerant trees should be used in reforestation or remediation systems. In this study the effects of Cd on seed germination and early seedling growth were evaluated in Schinus molle plants. Seeds of S. molle were treated with Cd at concentrations of 0 (control), 0.01, 0.02, 0.05, and 0.125 mM using Cd(NO3)2. Seed germination parameters and seedling growth traits were evaluated 33 days after the experiment started. Seeds and roots were sampled for anatomical evaluation and processed with plant microtechniques. No effects of Cd were found on the germination percentage of S. molle seeds. However, seedlings’ shoot and root fresh masses and lengths increased with Cd treatment. Cadmium increased root cortex and endodermis thickness and reduced the percentage of vascular tissues in seedlings. Schinus molle seeds and seedlings thus showed tolerance to Cd related to anatomical adaptations.

RESUMO O estudo do sistema nervoso e suas funções é fascinante e complexo. Um dos mais intrigantes assuntos deste campo é como a neurobiologia produz estados conscientes, o notório problema da consciência (cérebro e mente). Para esse estudo, são necessários conhecimentos tanto de neuroanatomia e fisiologia como de questões linguísticas e filosóficas, o que torna o assunto temido ou ignorado pelos estudantes de Medicina. Assim, aquele que vai iniciar o estudo desse capítulo da neurociência se depara com um questionamento: por onde começar? A quantidade de informação é muito grande, e o ponto de partida, em geral, pode ser a biologia, a psicologia ou a filosofia. Portanto, este ensaio tem como objetivo sugerir um caminho, entre vários possíveis, para nortear aqueles que desejam informações preliminares sobre o tema. Isto porque alguns estudantes têm dificuldade com o assunto por não dominarem alguns conceitos fundamentais ou desconhecerem as teorias defendidas, fixando-se apenas na vertente biológica do assunto. Desta maneira, na introdução deste manuscrito são apresentadas três possibilidades conceituais para a palavra consciência e alguns problemas de limitação do idioma. Na sequência, a neurobiologia é abordada de forma sucinta, com ênfase na atividade eletroquímica neuronal. Em seguida, são relatadas as teorias sobre a consciência e os pontos de vista de dois autores: David Chalmers e Daniel Dennett. A escolha desses autores para este trabalho visa demonstrar que tais teorias possuem movimento pendular: iniciando com Descartes e sua teoria dualista em um extremo, passando por outras teorias intermediárias e chegando até os materialistas reducionistas no extremo oposto. A metodologia se resume à pesquisa de artigos científicos sobre o tema. Por fim, embora a corrente majoritária defenda que a consciência é um fenômeno bioquímico, pois, para haver consciência é necessário cérebro, também há aqueles que defendem que a consciência não seja um atributo da matéria. Discussões como esta devem ser encorajadas e embasadas em fundamentação teórica.
ABSTRACT The study of the nervous system and its functions is fascinating and complex. One of the most intriguing in this field is how neurobiology produces conscious states; the notorious problem of consciousness (the brain and mind). For this study, a knowledge of both neuroanatomy and physiology is required, as well as of linguistic and philosophical issues. For this reason, this is a topic that is feared or avoided by medical students. The student wishing to begin this chapter of neuroscience faces a question: where to start? There is a vast amount of information on the subject, and the starting point, in general, can be biology, psychology or philosophy. This essay suggests one of several possible means of guiding those seeking preliminary information on the subject. Some students have difficulty with the subject because they lack the fundamental concepts needed, or they do not know the theories defended, but are focused on subject from a purely biological perspective. This work begins by presenting three conceptual possibilities for the word ‘conscience’, and some language limitations related to it. Next, it gives a succinct overview of neurobiology, with emphasis on neuronal electrochemical activity. The theories of consciousness are outlined, giving the points of view of two authors: David Chalmers and Daniel Dennett. This choice of authors demonstrates that such theories have pendular movement: starting with Descartes and his dualist theory at one extreme, passing through other intermediate theories, and reaching the reductionist materialists at the opposite extreme. The methodology consists of a literature search of scientific articles on the subject. Finally, although the majority argue that consciousness is a biochemical phenomenon, because in order to have consciousness there must be a brain, there are also those who argue that consciousness is not an attribute of matter. Discussions like these should be encouraged, and based on theoretical studies.

RESUMO Uma correta caracterização de bancos de germoplasma é fundamental para melhoristas obterem sucesso em programas de melhoramento. Várias pesquisas têm buscado obtenção de genótipos com resistência a pragas. No entanto, não há um consenso em relação a qual metodologia é a mais adequada para caracterizar um banco de germoplasma de tomateiro com diferentes níveis de resistência a pragas. O trabalho foi realizado com o objetivo de comparar métodos de análise multivariada para avaliação da diversidade genética em genótipos de tomateiro com diferentes níveis de resistência a pragas. Os experimentos foram conduzidos na Estação Experimental de Hortaliças da Universidade Federal de Uberlândia - campus Monte Carmelo (18º 42’ 43,19” latitude Sul e 47º 29’ 55,8” longitude Oeste, 873 m de altitude), no período de abril de 2013 a novembro de 2016. Foram avaliados dezesseis genótipos provenientes do cruzamento interespecífico entre LA-716 (S. pennellii) versus linhagem pré-comercial (UFU-057) seguido de um retrocruzamento e uma autofecundação, juntamente com a linhagem pré-comercial UFU-057 (genitor recorrente), Santa Clara e o acesso selvagem S. pennellii (genitor doador). Foram analisados os teores de acilaçúcares, tricomas foliares, teste de repelência a traça-do-tomateiro e a larva-minadora. O delineamento experimental foi em blocos ao acaso totalizando 76 parcelas (19 genótipos x 4 blocos). Conclui-se que houve variabilidade genética entre os genótipos avaliados. O método de dispersão gráfica por componentes principais revelou maior poder de discriminação. Os genótipos UFU-057F2RC27#4.3, UFU-057F2RC28#2.2 e UFU-057F2RC27#4.7 contêm os maiores teores de acilaçúcares, resistência a Liriomyza spp. e a T. absoluta.
ABSTRACT The correct characterization of germplasm banks is fundamental for breeders to succeed in breeding programs. Several studies have sought to obtain genotypes with resistance to pests. However, there is no consensus about which methodology is the most appropriate to characterize a germplasm bank of tomato with different levels of resistance to pests. The objective of this study was to compare methods of multivariate analysis for the evaluation of genetic diversity in tomato genotypes with different levels of resistance to pests. The experiments were conducted at the Vegetable Experimental Station of the Federal University of Uberlândia - Monte Carmelo campus (18º 42’ 43.19” South latitude and 47º 29’ 55.8” West longitude, 873 m altitude), in the period from April 2013 to November 2016. Sixteen genotypes were evaluated from the interspecific cross between LA-716 (S. pennellii) versus pre-commercial line (UFU-057) followed by backcrossing and self-fertilization, along with the pre-commercial line UFU-057 (recurrent parent) Santa Clara and the wild accession S. pennellii (donor genitor). The contents of acylsugar, foliar trichomes, South American tomato pinworm and leaf miner repellency tests were analyzed. The experimental design was the randomized block design totaling 76 plots (19 genotypes x 4 blocks). It was concluded that there was genetic variability among the evaluated genotypes. The method of graphic dispersion by principal components revealed a greater power of discrimination. Genotypes UFU-057F2RC27#4.3, UFU-057F2RC28#2.2 and UFU-057F2RC27#4.7 contain the highest levels of acylsugar, resistance to Liriomyza spp. and T. absoluta.

Fast Ephedrine Quantification by Gas Chromatography Mass Spectrometry

Ephedrines are widely used in therapy. Because of their stimulant properties, these substances are relevant in different forensic fields. At present, the state of the art for ephedrines quantification relay based on a liquid chromatography mass spectrometry, mainly because of the dilute-and-shoot approach. Notwithstanding, several gas chromatography based methods have already been described, all of them include cleanup steps, with the potential disadvantage of incurring errors and increasing the workload. In this paper, a straightforward method for ephedrine quantification based on gas chromatographic mass spectrometry, without cleanup and based on Doehlert matrix optimization is presented. Only 10 µL of a urine sample is necessary and for N-methyl-N-(trimethylsilyl)trifluoroacetamide/N-methyl-bis-trifluoracetamide derivatives, the intermediate precision was 2.77% for ephedrine, 9.20% for cathine, 8.29% for norephedrine and 4.27% for pseudoephedrine. The limit of detection was 20 ng mL-1 for ephedrine, 30 ng mL-1 for cathine and 40 ng mL-1 for norephedrine and pseudoephedrine.
