Abstract New biodegradable packaging has been developed from renewable sources, mainly of vegetable origin. Arrowroot starch has been recently used to produce high-quality biodegradable films capable of behaving well when incorporating oils, extracts, metal, and metal nanocomposites. The study aimed to verify the impact of incorporating metals in the sulfate and chloride forms in a biopolymeric matrix from arrowroot starch in terms of biodegradability, physicochemical and microbiological parameters. Different arrowroot films were produced to incorporate solutions with a concentration of 1 Mol L−1 of sulfate and chloride metals. The action of biodegradability in soil, UV transmittance, and visible light were observed in UV-Vis spectrophotometry and antimicrobial action on Escherichia coli, Staphylococcus aureus, Salmonella serovar Typhimurium, and Salmonella serovar Enteritidis. Good results were obtained, such as biodegradability time between 81.70 to 100 % (30 days), a low transmission rate of UV radiation and visible light between 250 to 890 nm, high capacity for bacterial inhibition between 22.08 to 10.05 mm for E. coli, among 25.59 to 11.10 mm for S. aureus, between 22.14 to 11.66 mm for S. serovar Typhimurium and between 21.11 to 8.26 mm for S. serovar Enteritidis. It is concluded that the biodegradable films of arrowroot starch incorporated with metals showed potential in all the evaluated tests, thus characterizing possible new products for different uses, such as low time available in the environment, preservation of the characteristics of special products, and antimicrobial capacity.
Resumen Se han desarrollado nuevos embalajes biodegradables de fuentes renovables principalmente de origen vegetal. El almidón de arrurruz se ha usado en la producción de películas biodegradables de alta calidad, capaces de tener un buen comportamiento al incorporar aceites, extractos, metales y nanocompuestos metálicos. El estudio tiene como objetivo verificar el impacto de la incorporación de metales en las formas de sulfato y cloruro en una matriz biopolimérica de almidón de arrurruz en términos de biodegradabilidad, parámetros fisicoquímicos y microbiológicos. Se produjeron diferentes películas de arrurruz con soluciones a una concentración de 1 Mol L−1 de metales sulfato y cloruro. La acción de biodegradabilidad en el suelo, transmitancia UV y luz visible se observó en espectrofotometría UV-Vis, y acción antimicrobiana sobre Escherichia coli, Staphylococcus aureus, Salmonella serovar Typhimurium y Salmonella serovar Enteritidis. Se obtuvieron buenos resultados, como tiempo de biodegradabilidad entre 81,70 a 100 % (30 días), baja tasa de transmisión de radiación UV y luz visible entre 250 a 890 nm, alta capacidad de inhibición bacteriana entre 22,08 a 10,05 mm para E. coli, entre 25,59 a 11,10 mm para S. aureus, entre 22,14 a 11,66 mm para S. serovar Typhimurium y entre 21,11 a 8,26 mm para S. serovar Enteritidis. Se concluye que las películas biodegradables de almidón de arrurruz incorporadas con metales mostraron potencial en todas las pruebas evaluadas, caracterizando así posibles nuevos productos para diferentes usos, tales como bajo tiempo disponible en el ambiente, preservación de las características de productos especiales y capacidad antimicrobiana.

Abstract Introduction The aberrant expression of the inhibitor of DNA binding (ID1) gene has been frequently associated with the leukemogenesis and prognostication acute myeloid leukemia (AML), although its clinical importance has never been investigated in patients treated outside well-controlled clinical trials. Methods Using quantitative real-time polymerase chain reaction, we investigated the role of the ID1 expression in the clinical outcomes of non-selected patients with acute myeloid leukemia treated in a real-life setting. Results Overall, 128 patients were enrolled. Patients with high ID1 expression had a lower 3-year overall survival (OS) rate of 9%, with the 95% confidence interval (95%CI) at 3 to 20%, compared to patients with a low ID1 expression (22%, 95%CI: 11 - 34%) (p = 0.037), although these findings did not retain significance after adjustment (hazard ratio (HR): 1.5, 95%CI: 0.98 - 2.28; p = 0.057). The ID1 expression had no impact on post-induction outcomes (disease-free survival, p = 0.648; cumulative incidence of relapse, p = 0.584). Conclusions Although we are aware thar our data are confronted with many variables that cannot be fully controlled, including drug unavailability, risk-adapted treatment, comorbidities and the time from diagnosis to treatment initiation, we are firm believers that such an initiative can provide more realistic data on understudied populations, in particular those from low- and middle-income countries. RESUMO Espécies vegetais de ocorrência no bioma Caatinga quando submetidas a diferentes flutuações hídricas podem ter seu desempenho morfofisiológico e metabólico alterado. A depender do nível de estresse, a produção de mudas de qualidade é afetada. Logo, objetivou-se avaliar as respostas às diferentes disponibilidades hídricas no desenvolvimento morfofisiológico e bioquímico de Croton blanchetianus Baill. O experimento foi desenvolvido em casa de vegetação e foram avaliados quatro regimes hídricos: 25%, 50%, 75% e 100% da capacidade de vaso e conduzido em delineamento experimental em blocos ao acaso, totalizando 5 blocos. As variáveis analisadas foram: altura e diâmetro dos ramos; área foliar, número de folhas, volume radicular; massa seca da raiz, parte aérea, total e relação raiz/parte aérea; teor relativo de água, prolina, proteínas solúveis totais, malondialdeído, peróxido de hidrogênio e pigmentos fotossintéticos (clorofila ɑ, clorofila b e carotenóides). A espécie Croton blanchetianus Baill. apresenta mecanismos de tolerância à seca, demonstrados pela redução de crescimento, área foliar, massa seca das mudas e da redução dos pigmentos fotossintéticos. Se ajusta osmoticamente com aumento na produção de prolina que atua na osmoproteção quando há déficit hídrico e, aumentando a produção de proteínas no menor e maior regime hídrico. ABSTRACT Plant species occurring in the Caatinga biome, when subjected to different water fluctuations, may have their morphophysiological and metabolic performance altered. Depending on the level of stress, the production of quality seedlings is affected. Therefore, the objective of this study was to evaluate the responses to different water availability levels in the morphophysiological and biochemical development of Croton blanchetianus Baill. The experiment was carried out in a greenhouse, evaluating four water regimes: 25%, 50%, 75% and 100% of pot capacity, and conducted in a randomized block design, with a total 5 blocks. The variables analyzed were: branch height and branch diameter; leaf area, number of leaves, root volume; root dry mass, shoot dry mass, total dry mass and root/shoot ratio; relative water content, proline, total soluble proteins, malondialdehyde, hydrogen peroxide, and photo synthetic pigments (chlorophyll ɑ, chlorophyll b, and carotenoids). The species Croton blanchetianus Baill. shows drought tolerance mechanisms, demonstrated by the reductions of growth, leaf area, dry mass of seedlings and photo synthetic pigments. Resumo As florestas urbanas fornecem cobertura vegetal de médio e grande porte em áreas urbanas. O plantio de árvores nativas nas calçadas é uma abordagem viável para reduzir os danos causados pela extensa urbanização. O uso de hidrogéis parece aumentar o sucesso do transplante de mudas em ambientes urbanos. Assim, este estudo tem como objetivo avaliar a eficácia de Schinus terebinthifolia na arborização urbana, focando na sua adaptação e sobrevivência pós-transplante utilizando hidrogel e sendo monitorado por sistema arduino. As concentrações do hidrogel utilizadas foram 0,75; 1,5; 3,0 e 6,0 g L-1, sendo também estabelecidos dois controles: um tratamento controle sem irrigação e sem hidrogel, e um controle com irrigação diária com água. A avaliação foi contínua durante 14 dias, e os parâmetros analisados foram temperatura e umidade do substrato, teor relativo de água (CR), extravasamento de eletrólitos (EEE), teor de clorofila e compostos bioquímicos. Adotou-se delineamento inteiramente casualizado, composto por 6 tratamentos com 9 repetições cada. Os resultados indicam que S. terebinthifolia é uma espécie altamente resiliente e adequada para arborização urbana, apresentando notável tolerância ao transplante e à restrição hídrica. Observou-se que o uso de hidrogéis contribui significativamente para a manutenção da umidade do substrato, resultando em maior estabilidade das mudas transplantadas. O sistema Arduino permitiu avaliação contínua e precisa das condições do substrato, otimizando o manejo da arborização urbana e validando a eficiência dos tratamentos aplicados. Respostas positivas foram observadas ao utilizar hidrogéis em termos de conteúdo relativo de água, estabilidade da membrana e atividade antioxidante, mesmo sob restrição hídrica. Abstract Urban forest provide medium and large vegetation cover in urban areas. Planting native trees on sidewalks is a viable approach to reduce damages caused by extensive urbanization. The use of hydrogels seems to increase the success of seedling transplantation in urban environments. Thus, this study aims to evaluate the efficacy of Schinus terebinthifolia in urban afforestation, focusing on its adaptation and post-transplant survival using hydrogel and being monitored by an arduino system. The concentrations of the commercial hydrogel used were 0.75, 1.5, 3.0, and 6.0 g L-1, and two controls were also established: a control treatment without irrigation and without hydrogel, and a control with daily water irrigation. The evaluation was continuous for 14 days, and the parameters analyzed were substrate temperature and moisture, relative water content (RWC), electrolyte extravasation (EEE), chlorophyll content, and biochemical compound. A completely randomized design was adopted, consisting of 6 treatments with 9 replicates each. The results indicate that S. terebinthifolia is a highly resilient species suitable for urban afforestation, showing remarkable tolerance to transplantation and water restriction. It was observed that the use of hydrogels significantly contributes to maintaining substrate moisture, resulting in greater stability of the transplanted seedlings. The Arduino system allowed for continuous and precise evaluation of substrate conditions, optimizing the management of urban afforestation and validating the efficiency of the applied treatments. Positive responses were observed when using hydrogels in terms of relative water content, membrane stability, and antioxidant activity, even under water restriction. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. RESUMO O sistema de plantio direto é um sistema conservacionista que auxilia na sustentabilidade e a produção agrícola. A eficácia de controle do glyphosate, aplicado isoladamente ou em associação a outros herbicidas, pode ser alterada em função da formulação do produto. O objetivo do presente trabalho foi avaliar a eficácia de glyphosate em formulações contendo diferentes sais e concentrações, aplicado isolado e em associações a outros herbicidas, no controle de plantas daninhas em estádios avançados na operação de dessecação pré-semeadura. O experimento foi realizado a campo em área com histórico de elevada infestação de plantas daninhas. O experimento foi conduzido no delineamento de blocos casualizados, avaliando-se onze tratamentos e quatro repetições. Os tratamentos foram compostos pela aplicação de três formulações à base de glyphosate isolado e em associações com os herbicidas clethodim e 2,4-D amina, além de uma testemunha sem aplicação. As variáveis avaliadas foram porcentagem de controle das plantas daninhas e porcentagem de dessecação. Foi realizado uma comparação de médias por contrastes para analisar a porcentagem de controle de plantas daninhas. ABSTRACT The no-tillage system is a conservation system that helps sustainability and agricultural production. The effectiveness of glyphosate control, applied alone or in combination with other herbicides, can be altered depending on the product’s formulation. The objective of the present study was to evaluate the effectiveness of glyphosate in formulations containing different salts and concentrations, applied alone and in combination with other herbicides, in controlling weeds in advanced stages in the pre-sowing burndown operation. The experiment was carried out in the field in an area with a history of high weed infestation. The experiment was conducted in a randomized block design, evaluating eleven treatments and four replicates. The treatments consisted of the application of three glyphosate-based formulations alone and in combination with clethodim and 2,4-D amine herbicides, in addition to a control without herbicide application. The evaluated variables were percentage of weed control and percentage of desiccation. A comparison of means by contrasts was performed to analyze the percentage of weed control. Ni-based superalloys are widely used in critical components of aircraft engines and turbines and also in the petrochemical industry, for applications in highly corrosive environments. These alloys have as main characteristics their superior mechanical, corrosion and oxidation resistance at high temperatures, as well as creep resistance. The chemical composition associated with carrying out heat treatments directly influences the phases formed (such as the ordered cubic phase γ’-Ni3(Al,Ti) in the fcc γ-Ni matrix), and depending on the alloying elements and fraction, there is the possibility of an increase in mechanical strength. There is a certain gap in the literature regarding the study of ternary superalloys based on Ni-Nb, and the influence of the third alloying element on the microstructure and microhardness. In this context, the objective of the study is to characterize pseudo-eutectic alloys of the Ni-15Nb-xM and Ni-20Nb-xM systems (xM = 2Al, 4Ti, 15Fe and 15Cr, wt.%) and investigate the influence of alloy elements and solution heat treating on their microstructure and properties through X-Ray Diffraction, Optical Microscopy, Scanning Electron Microscopy and Vickers Microhardness. Microhardness and microstructures were significantly influenced by the addition of alloying elements. The addition of Cr had a significant effect on the hardness of the cast samples. All alloys showed microhardness and microstructural changes after solution heat treatment. Abstract This study aimed to verify the action of bioactive compounds from Brazilian plants on the leader genes involved in the SARS-CoV-2 pathway. The main human genes involved were identified in GeneCards and UNIPROT platforms, and an interaction network between leader genes was established in the STRING database. To design chemo-biology interactome networks and elucidate the interplay between genes related to the disease and bioactive plant compounds, the metasearch engine STITCH 3.1 was used. The analysis revealed that SMAD3 and CASP3 genes are leader genes, suggesting that the mechanism of action of the virus on host cells is associated with the molecular effects of these genes. Furthermore, the bioactive plant compounds, such as ascorbate, benzoquinone, ellagic acid, and resveratrol was identified as a promising adjuvant for the treatment inhibiting CASP3-mediated apoptosis. Bioactive plant compounds were verified as the main pathways enriched with KEGG and related to viral infection, assessments/immune/infections, and cell proliferation, which are potentially used for respiratory viral infections. The best-ranked molecule docked in the CASP3 binding site was rutin, while the SMAD3 binding site was resveratrol. In conclusion, this work identified several bioactive compounds from Brazilian plants showing potential antiviral functions that can directly or indirectly inhibit the new coronavirus.

Abstract This study evaluated of the effects of açai oil during the close-up dry period of Holstein cows on colostrum quality, as well as on the immune and antioxidant responses of their calves. Sixteen multiparous cows were assigned randomly to two treatments: 1) CONTROL (n = 8) - 4.48% of soybean oil/concentrate; 2) AÇAI (n =8) - 4.48% of açai oil/concentrate. Cows fed with açai oil had greater (P≤0.04) colostrum concentrations of immunoglobulins (Ig) G (1st and 2nd milking), IgG heavy chains, IgA (only at 1st milking), alpha-lactalbumin (1st milking), total protein, and antioxidant capacity against peroxyl radicals (only at 1st milking). Cows fed with açai oil had greater serum concentrations of globulin (only on the day of calving) and total protein (only on the day of calving) (P = 0.03). Calves born of cows fed with açai oil had greater serum concentrations of total protein (only 24 and 48 h after calving) and serum concentration of IgG heavy chain (only 24 h after calving) and globulin (only 24 and 48 h after calving) (P = 0.01). These data suggest that the addition of açai oil in the cow feed during the close-up dry period boosted immunity in their calves by altering the composition of colostrum.

The Amazonian Rubiaceae species Uncaria guianensis (UG) is locally used as anti-inflammatory, antitumor, antidiabetic, anti-ulcers, and others. The phenolic content of its leaves is characterized by the great predominance of the flavonoid kaempferol-3,7-O-(α)-L-dirhamnoside (kaempferitrin). The present study quantitatively evaluates the kaempferitrin content in the leaves and branches of cultivated and wild UG specimens collected in different locations of the Brazilian Amazon rainforest by employing high-performance liquid chromatography coupled to a diode array detector (HPLC-DAD). Besides, the understanding of the polyphenol profile performed by electron spray ionization is deepened by tandem mass spectrometry analysis (ESI-MS/MS), using a previously approached leaf UG extract, and the flavonoid quercetin-3,7-O-(α)-L-dirhamnoside was first isolated from UG. All samples showed quite similar qualitative polyphenol profiles. Kaempferitrin in UG ranged from 1.1 to 1.9 mg 100 mg-1 for dry leaves of adult wild plants, 0.3 to 0.7 mg 100 mg-1 for dry leaves of cultivated young plants and 0.00 to 0.04 mg 100 mg-1 for dry branches of adult wild plants. Besides suggesting the distribution of kaempferitrin in the species, these results reinforce this flavonol as a suitable chemical marker for UG leaves and the products derived from them.

ABSTRACT Objective: Adipose tissue-derived stromal/stem cells (ASCs) and vitamin D have immunomodulatory actions that could be useful for type 1 diabetes (T1D). We aimed in this study to investigate the safety and efficacy of ASCs + daily cholecalciferol (VIT D) for 6 months in patients with recent-onset T1D. Materials and methods: In this prospective, dual-center, open trial, patients with recent onset T1D received one dose of allogenic ASC (1 x 106 cells/kg) and cholecalciferol 2,000 UI/day for 6 months (group 1). They were compared to patients who received chol-ecalciferol (group 2) and standard treatment (group 3). Adverse events were recorded; C-peptide (CP), insulin dose and HbA1c were measured at baseline (T0), after 3 (T3) and 6 months (T6). Results: In group 1 (n = 7), adverse events included transient headache (all), mild local reactions (all), tachycardia (n = 4), abdominal cramps (n = 1), thrombophlebitis (n = 4), scotomas (n = 2), and central retinal vein occlusion at T3 (n = 1, resolution at T6). Group 1 had an increase in basal CP (p = 0.018; mean: 40.41+/-40.79 %), without changes in stimulated CP after mixed meal (p = 0.62), from T0 to T6. Basal CP remained stable in groups 2 and 3 (p = 0.58 and p = 0.116, respectively). Group 1 had small insulin requirements (0.31+/- 0.26 UI/kg) without changes at T6 (p = 0.44) and HbA1c decline (p = 0.01). At T6, all patients (100%; n = 7) in group 1 were in honeymoon vs 75% (n = 3/4) and 50% (n = 3/6) in groups 2 and 3, p = 0.01. Conclusions: Allogenic ASC + VIT D without immunosuppression was safe and might have a role in the preservation of β-cells in patients with recent-onset T1D. ClinicalTrials.gov: NCT03920397.

Abstract The Equine Strangles, caused by Streptococcus equi subs. equi, is a contagious disease, causing high rates of morbidity been responsible for important economic losses. The M protein synthesized by S. equi plays an important role in the pathogenesis and is a promising candidate for a vaccine antigen. The innate immune system is responsible for the first immune response against microorganisms, this response is mediated by receptors that detect PAMPs and their activation trigger crucial modulation of the adaptative immune response. This work describes the immune response of S. equi subs. equi. recombinant SeM protein, using Escherichia coli BL21 (DE3) as an expression and delivery vaccine system. To characterize and to determine the vaccine efficacy, mice were vaccinated as followed: 1. Recombinant E. coli expressing rSeM protein; 2. The same recombinant E. coli, inactivated adsorbed in Alumen; 3. Purified rSeM protein adsorbed in Alumen; 4. Inactivated S. equi whole cells adsorbed in Alumen; 5. Control group. All vaccinated mice developed protective response against S. equi infection, however the groups that received the E. coli expressing rSeM presented significant higher IgG level than other vaccinated groups. The recombinant E. coli delivery vaccine system also induced a highest IgG response than inactivated S. equi or purified rSeM vaccines in horses. This study evidence that the recombinant E. coli, live or inactivated, enhanced the humoral response, reaching significant higher antibodies levels than those obtained in the vaccination with the bacterin or purified antigen, showing the feasibility of producing low-cost vaccines against strangles.

Abstract The immobilization of the enzyme tyrosinase (Tyr) in lipid matrices can be explored to produce biosensors for detecting polyphenols, which is relevant for the food industry. Herein, we shall demonstrate the importance of the lipid composition to immobilize the enzyme tyrosinase in Langmuir-Blodgett (LB) films. Tyr could be incorporated into Langmuir monolayers of arachidic acid (AA), 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and 1,2-dipalmitoyl-sn-glycero-3-phospho-(1’-rac-glycerol) (sodium salt) (DPPG), having as the main effect an expansion in the monolayers. Results from polarization-modulated infrared reflection-absorption spectroscopy (PM-IRRAS) pointed to electrostatic interactions between the charged residues of Try and the lipid headgroups, in addition to changes in the order of lipid chains. The interaction between Tyr and DPPC in Langmuir monolayers can be correlated with the superior performance of DPPC/Tyr LB films used as biosensors to detect catechol by cyclic voltammetry. The molecular-level interactions assessed via PM-IRRAS are therefore believed to drive an immobilization process for Tyr in the lipid LB matrix and may serve as a general criterion to identify matrices that preserve enzyme activity.

This study presents a microwave-assisted procedure for sample preparation of cocoa beans using diluted HNO3 with H2O2 for the determination of Ba, Ca, Cu, K, Mg, Mn, P, S, Sr and Zn by inductively coupled plasma optical emission spectrometry (ICP OES). Doehlert modified design was used for optimization of experimental condition, and the volume of H2O2 (0, 0.5, 1.75 and 3 mL) and concentration of HNO3 (0.5, 3.75 and 7 mol L-1) were evaluated. The best experimental conditions were chosen considering lower residual acidity (RA), lower dissolved organic carbon (DOC) and the acceptable trueness range (80-120%), were 5 mL of 3.75 mol L-1 HNO3 and 1.75 mL of 30% v v-1 H2O2. The procedure was validate using certified reference materials (CRMs), digested using the optimized conditions. The proposed procedure was applied in twelve samples of cocoa beans, and the concentrations (mg kg-1) ranged from: 7.6-27 (Ba), 520-1955 (Ca), 11-26 (Cu), 6212-10855 (K), 1677-2516 (Mg), 13-29 (Mn), 4679-6104 (P), 1499-1898 (S), 7-32 (Sr) and 34-46 (Zn). A Pearson correlation plot was proposed to evaluate the correlation between elements, revealing high correlations between Mg and K; K and P; Cu and Mn; Cu and Zn; Cu and S.

The essential oil of Cymbopogon nardus, citronella, has been extensively studied. However, the chemical and biological properties of the ethanolic extract (EE) of C. nardus have not been evaluated. The aim of this study was to characterize the chemical composition of the EE of C. nardus and its active fraction (FrD). Moreover, the cytotoxic and antifungal properties of these extracts against Candida species with different resistance profiles to conventional drugs were evaluated. The compounds identified in EE were mono-C- and di-C-glycosyl flavones and phenylpropanoid glycosides. Phenylpropanoid glycosides were identified in FrD. EE showed antifungal activity, with minimum inhibitory concentration (MIC) values ranging from 62. 5 to 500 µg mL-1. FrD was more effective against C. glabrata, as evidenced by the lowest MIC value (15. 6 µg mL-1). EE inhibited yeast growth similar to amphotericin-B, as demonstrated by similar time-kill curves. EE inhibited C. albicans hyphae formation and mature biofilm of C. albicans, C. krusei and C. parapsilosis. The results of the chemical and biological analyses of EE and its fractions provided novel information and may contribute to control of infections caused by Candida species.
