Solid and liquid fractions of hydrothermally pretreated sugarcane bagasse (SCB) were simultaneously used as substrate of a novel continuous compartmentalized reactor. The effect of four (56, 42, 28, and 14 h) hydraulic retention time (HRT) and three (0.5, 3.0, and 9.0 g L-1) chemical oxygen demand (COD) levels were evaluated on hydrogen (H2) and organic acids production. Higher H2 production and yield (686 mL and 1.63 mol mol-1 carbohydrate, respectively) were obtained under an HRT of 28 h, probably due to the Clostridium and Thermoanaerobacterium metabolisms, which accounted for almost 60% of the microbial relative abundance. Under lower and higher HRT (14 and 56 h, respectively) lactic acid prevailed without hydrogen production. Other value-added chemicals such as citric, valeric and caproic acids were also obtained according to the HRT. From the functional point of view, enzymes from the glycoside hydrolases group (GHs) potentially performed important roles in the lignocellulosic biomass bioconversion. ABSTRACT The knowledge of the bat fauna from Minas Gerais, the largest state in southeastern Brazil, has greatly changed in recent years due to increased sampling and taxonomic revisions with reports of range extension and/or new species descriptions. The first annotated list of bats for the state dates to over 10 years ago. Our study aimed to update it by incorporating new localities sampled since then and by adding new species records to the list. We also conducted a comprehensive review of the literature and museum databases and analyzed 691 specimens collected in the state from all mesoregions and ecosystems deposited in collections. We documented the occurrence of 99 bat species from nine families, distributed in 181 municipalities, covering all mesoregions and biomes of the state. This is an increment of 30% in bat richness and of 241% in locality sites with bat records in Minas Gerais, when compared with the previous compilation. We report two previously unrecorded species for the state, the phylostomids Rhinophylla pumilio Peters, 1865 and Trinycteris nicefori (Sanborn, 1949). The mesoregions with the largest number of bat collection localities were “Metropolitana” (46), “Zona da Mata” (26), and “Norte de Minas” (21). The Atlantic Forest had the largest number of collection sites compared to the other biomes present in the state (102). There are a lot of sampling gaps in the state, and most of the conservation areas have not yet been sampled for bats. In face of that and based on the recent findings of several new species and records as a result of sampling and revisionary efforts, we suggest the implementation of rapid survey programs accompanied by taxonomic studies within the conservation units of Minas Gerais. RESUMO Análises hematológicas e bioquímicas possuem grande importância na avaliação da saúde das populações e de seus habitats e, por estarem sujeitas a influências de diversos fatores, tornam-se necessários conhecer os valores próprios de cada espécie. Neste estudo objetivou-se determinar os parâmetros hematológicos e bioquímicos, assim como a análise morfométrica das células sanguíneas de catetos criados em cativeiro no semiárido do nordeste brasileiro. Utilizou-se 30 animais adultos divididos em 2 grupos (15 fêmeas e 15 machos), com peso médio de 20kg e idade entre 2 e 2.5 anos. Foram coletadas amostras de sangue e soro, no período da manhã, para realizar as análises hematológicas, bioquímica sérica e morfometria celular. Os dados foram analisados estatisticamente quanto à normalidade, pelo teste de Shapiro-Wilk, e homocedasticidade, por Levene. Quando a diferença estatística entre sexos os dados foram paramétricos, utilizando o teste t para medidas independentes e Mann-Whitney quando não paramétricos. Os resultados mostraram que o sexo não possui influência na maioria das variáveis estudadas, excetuando as análises bioquímicas cálcio, proteínas totais e ureia e a contagem relativa dos eosinófilos. ABSTRACT Hematological and biochemical analyses are paramount in assessing the health of different populations and their habitats, and knowledge on the specific values of each species is necessary, due to influence of several factors. In this context, this study aimed to determine the hematological and biochemical parameters of collared peccaries raised in captivity in the semi-arid Brazilian northeast associated to morphometric blood cell analyses. A total of 30 adult animals were investigated, divided into two groups (15 females and 15 males), averaging 20 kg and between 2 and 2.5 years old. Blood and serum samples were collected in the morning for hematological, serum biochemistry and cellular morphometry analyses. Data normality was determined by the Shapiro-Wilk test, and homoscedasticity, by Levene’s test. Concerning statistical differences between sexes, parametric data were analyzed by the t test for independent measurements and non-parametric data, by the Mann-Whitney test. Sex did not influence most of the studied variables, except for calcium, total proteins and urea and relative eosinophil counts. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract Objective: To investigate the effect of a single oral dose of 200,000 IU of vitamin D3 on antiphospholipid antibodies in hospitalized patients with moderate to severe COVID-19. Methods: This is a post-hoc, exploratory analysis from a double-blind, placebo-controlled, randomized clinical trial performed in two centers in São Paulo, Brazil. Hospitalized patients with COVID-19 were randomly assigned to receive either vitamin D3 (n = 97) or placebo (n = 97). In this post-hoc analysis, the endpoints were titers and frequency of anti–β2-Glycoprotein-I (aβ2-GP) and Anticardiolipin (aCL) antibodies [Immunoglobulin G, M and A (IgG, IgM and IgA)]. Results: Overall mean (SD) age was 55.3 (13.9) years, Body Mass Index (BMI) was 32.2 (7.1 kg/m²), and 106 participants (54.6 %) were male. There was a significant group by time interaction (p = 0.046) for frequency of aCL IgG, with increased values from baseline to discharge in the placebo group [n (%), from 13 (13.4) to 25 (25.8)] compared to the vitamin D3 [from 25 (25.8) to 29 (29.9)]. However, the frequency of aCL IgG did not change between the groups on discharge. No significant differences between vitamin D3 and placebo groups were found for any other autoantibodies. Conclusion: These findings do not support the use of a single oral dose of 200,000 IU of vitamin D3 to modulate autoantibodies in hospitalized patients with moderate to severe COVID-19. Abstract Leprosy is a chronic infectious disease caused by the bacilli Mycobacterium leprae and Mycobacterium lepromatosis. In addition to humans, animals such as nine-banded armadillos and red squirrels are species naturally infected. The objective of this study was to investigate the presence of M. leprae and M. lepromatosis in non-volant small mammals of the order Didelphimorphia and Rodentia through Polymerase Chain Reaction (PCR) assay. During 2015 and 2018, field expeditions were carried out in three municipalities, covering biotic elements of the Amazon and Cerrado biomes, in the Mato Grosso State, Midwest of Brazil. A specific primer for repetitive sequences of the genomic DNA of M. leprae and M. lepromatosis targeting the RLEP and RLPM gene, respectively, was used to screen for these agents. The molecular detection of M. leprae DNA in the samples was 13.8%. M. lepromatosis was not detected. ABSTRACT Introduction: The Glanzmann Thrombasthenia (GT) and Bernard-Soulier Syndrome (BSS) are rare hereditary disorders of platelet function. Their treatment often requires platelet transfusion, which can lead to the development of alloantibodies. Objective: In this study, we aim to develop a strategy for alloantibody detection and to describe the frequency of alloimmunization in a patient population from a single center in southeastern Brazil. Methods: Samples from patients with GT or BSS were tested using the Platelet Immunofluorescence Test (PIFT). If a positive result was obtained, a confirmatory step using the Monoclonal Antibody Immobilization of Platelet Antigens (MAIPA) and Luminex bead-based platelet assay (PAKLx) was executed. Main results: Among 11 patients with GT, we detected the presence of alloantibodies in 5 using PIFT, with confirmation through MAIPA and PAKLx in 2 (1 anti-HLA and 1 anti-HPA), resulting in a frequency of 18.1%. Among 4 patients with BSS, PIFT was positive in 3, with confirmation by MAIPA and PAKLx in 1 (anti-HLA), showing a frequency of 25%. The two patients with anti-HLA antibodies exhibited a panel reactive antibody (PRA-HLA) testing greater than 97%. Conclusion: Our study highlights the importance of identifying platelet alloimmunization in this patient population. RESUMO: Mormo é uma enfermidade causada pela bactéria Burkholderia mallei que acomete primariamente cavalos, mulas e burros. A doença pode causar lesões na pele, pulmões e em diversos outros órgãos, entretanto frequentemente manifesta-se como uma enfermidade assintomática. No Brasil são utilizados testes sorológicos de elevada sensibilidade e especificidade para auxiliar na detecção de anticorpos contra B. mallei e contribuir para controle da doença. Porém, devido à obrigatoriedade da eutanásia de animais sororeagentes, os equídeos com sorologia positiva para B. mallei e assintomáticos geraram grandes embates entre criadores, médicos-veterinários e laboratórios de diagnóstico. Este trabalho esclarece as limitações dos testes diagnósticos complementares para detecção de B. mallei e descreve os achados clínicos, morfológicos e de exames laboratoriais em 24 equídeos, procedentes de diferentes municípios do estado de Mato Grosso, Brasil, que reagiram ao teste de fixação de complemento e foram positivos no teste de “western blotting” para mormo. Foram colhidos dados e amostras de tecidos de 24 equídeos para análise histológica, microbiológica e molecular. Em 23 equídeos não existiam sinais clínicos, alterações morfológicas, isolamento microbiológico ou detecção molecular que caracterizassem infecção por B. mallei. Por outro lado, amostras de um cavalo assintomático e sem alterações lesionais apresentaram amplificação de sequência compatível com B. mallei na PCR. Considerando que a infecção por B. mallei é passível da aplicação de medidas de defesa sanitária animal e que por exigência internacional e da legislação nacional, os resultados sorológicos são ferramentas que devem amparar os procedimentos de saneamento para erradicação da bactéria no estado de Mato Grosso, Brasil. ABSTRACT: Glanders is a disease caused by the bacterium Burkholderia mallei that primarily affects horses, mules and donkeys. The disease can cause lesions in the skin, lungs and several other organs. However, it often manifests as an asymptomatic disease. In Brazil, serological tests of high sensitivity and specificity are used to assist in the detection of antibodies against B. mallei and to contribute to the control of the disease. However, due to the mandatory euthanasia of seroreactive animals, equids with positive serology for B. mallei and asymptomatic generated great conflicts between breeders, veterinarians and diagnostic laboratories. This study clarifies the limitations of complementary diagnostic tests for detecting B. mallei. It describes the clinical, morphological and laboratory findings in 24 equines from different municipalities in the Mato Grosso State, Brazil, which reacted to the complement fixation test and were positive in the western blotting test for glanders. Data and tissue samples were collected from 24 horses for histological, microbiological and molecular analysis. In 23 horses, no clinical signs, morphological alterations, microbiological isolation, or molecular detection would characterize B. mallei infection. On the other hand, samples from an asymptomatic horse without lesional alterations showed sequence amplification compatible with B. mallei in the PCR. Considering that the infection by B. mallei is subject to the application of animal sanitary defense measures and that, by international requirement and national legislation, the serological results are tools that should support the sanitation procedures for the error of the bacteria in the Mato Grosso State, Brazil. RESUMO A batata-doce é uma hortaliça alimentar básica para a população. Este trabalho objetivou-se avaliar o rendimento da batata-doce em resposta à adubação potássica com diferentes fontes e parcelamento. O trabalho foi conduzido na Universidade Federal da Paraíba, em Areia-PB. O delineamento experimental foi de blocos casualizados em arranjo fatorial 7 × 2, com sete épocas de aplicação (100% no plantio; 100% aos 30 dias após o plantio (DAP); 100% aos 60 DAP; 50% no plantio e 50% aos 30 DAP; 50% no plantio e 50% aos 60 DAP; 50% aos 30 e 50% aos 60 DAP; 33% no plantio 33% aos 30 e 33% aos 60 DAP) e duas fontes, cloreto e sulfato de potássio, com três repetições. As variáveis analisadas foram à massa fresca por planta, massa média de raízes comerciais, produção de raízes comerciais por planta, teor de K foliar, produtividades total e comercial de raízes. O cloreto e sulfato de potássio incrementaram a massa fresca da planta e a massa de raízes comerciais, respectivamente. As produtividades comerciais de raízes (25,16 e 22,28 Mg ha-1) foram superiores à média nacional (14,07 Mg ha-1), quando se forneceu K2O nas fontes sulfato e cloreto de potássio, respectivamente. O teor de K foliar ficou dentro do padrão para a cultura, sob adubação de cloreto e sulfato de potássio. A aplicação de sulfato de potássio proporciona maior rendimento de batata-doce. O cloreto e o sulfato de potássio, quando fornecidos em uma única aplicação, aumentam a produtividade total e comercial somente quando as plantas estão aos 60 DAP.
ABSTRACT Sweet potatoes are an important staple food for human consumption. This study evaluated sweet potato yield in response to potassium fertilization using different sources and splitting. It was conducted at the Universidade Federal da Paraíba, Areia-PB. A complete randomized block experimental design was used in a 7 × 2 factorial arrangement. The fertilization was split into seven treatments (100% after planting; 100% at 30 days after planting (DAP); 100% at 60 DAP; 50% after planting and 50% at 30 DAP; 50% after planting and 50% at 60 DAP; 50% at 30 DAP and 50% 60 at DAP; 33% after planting, 33% at 30 DAP, and 33% at 60 DAP). Two fertilization sources were used, namely potassium chloride and potassium sulfate, with three replicates. Plant fresh mass, mass of marketable roots, production of marketable roots per plant, leaf K concentration, total and marketable root yields were evaluated. Chloride and sulfate potassium efficiently increased the fresh mass of the plant and the mass of marketable roots, respectively. The marketable yield of the roots (25.16 and 22.28 Mg ha-1) was higher than the national average (14.07 Mg ha-1) when K2O was supplied in the sulfate and potassium chloride sources, respectively. The leaf K concentration remained within the standard levels for the crops under chloride and potassium sulfate fertilization. Potassium sulfate application results in higher sweet potato yields. When supplied in a single application, chloride and sulfate potassium increase the total and commercial yield only when the plants are at 60 DAP.

Resumo Apresentamos um levantamento comentado de projetos financiados pelo programa BIOTA-Micro-organismos. Estes projetos geraram uma variada gama de resultados, incluindo a identificação de novos micro-organismos produtores de compostos antibacterianos, a caracterização de novas enzimas microbianas para usos industriais, classificação taxonômica de novos micro-organismos presentes em diversos ambientes, investigação de ecossistemas microbianos em solos e mangues e sua influência sobre plantas ameaçadas, e o sequenciamento de vários novos genomas microbianos derivados de metagenomas. Os resultados descritos demonstram o papel-chave de micro-organismos em ecossistemas importantes para atividades humanas, assim como o potencial que vários desses micro-organismos tem de contribuir para aplicações biotecnológicas cruciais para a sobrevivência humana no século 21.
Abstract We present a survey of projects that have been funded by FAPESP under the BIOTA-Microorganisms program. These projects generated a wide variety of results, including the identification of novel antibacterial-producing microorganisms, the characterization of novel microbial enzymes for industrial applications, taxonomic classification of novel microorganisms in several environments, investigation of the soil and mangrove microbial ecosystems and its influence on endangered plant species, and the sequencing of novel metagenome-assembled genomes. The results surveyed demonstrate the importance of microorganisms in environments that play important roles in human activities as well as the potential that many of these microorganisms have in contributing to biotechnological applications crucial for human survival in the 21st century.

Abstract Microbial therapeutic enzymes are the protagonists in the pharmacological treatment of different human diseases. The intrinsic enzymatic characteristics, such as high affinity and specificity to the corresponding substrate, enable effective therapies, with minimal adverse effects and complete remission. However, immunogenicity, short half-life, low enzymatic yield, and low selectivity regarding available enzyme drugs are currently the main obstacles to their development and the broad adherence to therapeutic protocols. By harboring adapted and still unexplored microbial life, environments of extreme conditions, such as Antarctica, become especially important in the prospecting and development of new enzymatic compounds that present higher yields and the possibility of genetic improvement. Antarctic microorganisms have adaptation mechanisms, such as more fluid cell membranes, production of antifreeze proteins and enzymes with more malleable structures, more robust, stable, selective catalytic sites for their respective substrates, and high antioxidant capacity. In this context, this review aims to explore enzymes synthesized by bacteria and fungi from Antarctica as potential drug producers, capable of providing therapeutic efficacy, less adverse effects, and lower production costs with highlight to L-Asparaginase, collagenase, superoxide dismutase and ribonucleases. In addition, this review highlights the unique biotechnological profile of these Antarctic extremophile microorganisms.

Abstract In the Antarctic environment, yeasts are versatile eukaryotes that have shown wide dispersion in different substrates, producing active enzymes in extreme conditions, but their relevance in biotechnological applications is largely unknown. The aim of this study was to evaluate the production of extracellular hydrolases by yeasts isolated from Antarctic lichens and molecularly identify these isolates. From a total of 144 isolates on the screening, 109 (76%) produced at least one of the hydrolases tested, with most activities for proteases 59 (41%), cellulases 58 (40%), esterases 57 (39%), lipases 29 (20%), amylases 23 (16%) and pectinases 20 (14%). Among these isolates, 76 were identified, most belonged to the phylum Basidiomycota (n=73) with the dominance of Vishniacozyma victoriae (n=27), Cystobasidium alpinum (n=3), Mrakia niccombsii (n=3), Cystobasidium laryngis (n=2), Bannozyma yamatoana (n=2), Holtermanniella nyarrowii (n=2), and Glaciozyma martinii (n=2). This study is the first one reporting extracellular enzyme production by yeasts isolated from thallus of the species of Antarctic lichens Lecania brialmontii, Polycauliona candelaria, Usnea capillacea, Cladonia metacorallifera, and Polycauliona regalis. With these data, it’s possible to confirm lichens as a source of hydrolase-producing yeasts, reinforcing the potential of these microorganisms in bioprospecting studies of catalytic molecules from polar regions that may be useful in promising biotechnological applications.

Abstract Industrial sectors are searching for new compounds to improve the preservation of food and blood, human tissues, and fuels used at low temperatures. Antarctic microorganisms have mechanisms to overcome injuries caused by low temperatures, making them sources of compounds with antifreeze activity. However, it is mandatory that such compounds do not pose a risk to human health. The present study evaluated the potential of Antarctic bacteria to resist freezing, produce virulence factors, their tolerance to physiological pHs/temperature and resistance to antibiotics. Sixty-five isolates were tested for antifreeze compound production, among which, 31 grew after the test. Of these, 3 strains of Arthrobacter sp. (356, 358 and 443), one Psychromonas arctica (ESH238) and one unidentified strain (363) showed positive results for hemolytic activity. Psychrobacter sp. 456 showed proteinase activity. None of the isolates showed resistance to the antibiotics. All isolates were able to grow in one of the three pHs (4, 7 and 8) and/or temperature (36, 38 and 40 ºC). Antarctic bacterial present potential for the production of antifreeze compounds and may be considered safe in industrial processes. The characterization of the genes responsible for virulence factors should be carried out to reinforce the potential applicability of such bacteria.

Abstract Currently, antimicrobial resistance has become a global public health problem, which has made the need for new antimicrobial compounds to deal with resistant infections an emergency. However, environments that once offered so many innovative molecules, now already exhaustively exploited, do not meet this need. In this context, a geographically isolated, under-explored and extreme environment, such as Antarctica, which holds organisms with unique physiological and biochemical characteristics, assumes great importance as a potential source of new compounds with antimicrobial activity. In this patent review, we investigate the state of technological development in the field of antimicrobial compounds obtained from Antarctic organisms, highlighting the main countries and researchers active in the field, the species utilized, the compounds obtained, and their possible therapeutic applications. As results, few patent documents were found, however they encompass a wide diversity of compounds and species, indicating a great antimicrobial potential present in Antarctic biota, including compounds active against the most important human pathogenic microorganisms, such as including methicillin-resistant Staphylococcus aureus, vancomycin-resistant Enterococcus spp. and multi-resistant Mycobacterium tuberculosis. Furthermore, due to the increasing trend in patent applications, a significant rise in the number of patents in this area is expected in the coming years.

Sazonalidade dos elementos do clima e sua influência na serapilheira em uma floresta sucessional na Amazônia oriental

Resumo A produção e decomposição da serapilheira é importante no funcionamento e na dinâmica de um ecossistema florestal, associando variáveis climatológicas à biomassa. Com isso, investigou-se o estoque de serapilheira em função da sozanalidade dos elementos do clima em uma floresta sucessional na Amazônia oriental. O estudo foi conduzido em uma floresta sucessional, na Região Metropolitana de Belém (RMB), Pará. No ecossistema foi realizada uma amostragem aleatória em três transectos alocados ao longo da perpendicular da linha de borda de 50 m. A análise do efeito da sazonalidade dos elementos do clima nas propriedades físicas (volume, espessura, densidade e massa seca) e no carbono da serapilheira foram realizadas utilizando o teste t student e ANOVA do teste F a 5 % de significância. A precipitação (495,2±125 mm e 109,8±60 mm) e a umidade relativa do ar (89,0±1,7 % e 79,6±2,8 %) apresentaram diferença estatística na sazonalidade (p<0,05), diferente da temperatura (27,1±0,2 ºC e 27,6±0,3 ºC) que não houve diferença. As variáveis da serapilheira não foram influenciadas pela sazonalidade dos elementos do clima e não diferiram estatisticamente entre os períodos chuvoso e menos chuvoso, respectivamente: quantidade de serapilheira acumulada (16,46±4,17 Mg ha-1 e 13,91±7,61 Mg ha-1), estoque de carbono da serapilheira (7,9±2,0 Mg ha-1 e 6,6 ± 3,61 Mg ha-1), volume (26,21±0,92 cm3 e 24,88±0,92 cm3), espessura (4,19±0,92 cm e 3,99±0,95 cm) e densidade (0,041±0,92 g/cm3 e 0,034±0,92 g/cm3). A sazonalidade dos elementos do clima não apresentou variação considerável a ponto de alterar as propriedades físicas e o carbono da serapilheira.
Abstract The production and decomposition of litter is important in the functioning and dynamic of a forest ecosystem, associating climatological variables to biomass. With that, the litter stock was investigated due to the seasonality of the climate in a successional forest in eastern Amazon. The study was conducted in a successional forest, in the metropolitan region of Belém (RMB), Pará. In the ecosystem, a random sampling was performed in three transects located along the perpendicular of the 50m border line. The analysis of the effect of the seasonality of the elements of the climate on the litter physical properties (volume, thickness, density and dry mass) and carbon content were carried out using the t Student test and ANOVA of the F test at 5 % significance level. Precipitation (495.2 ± 125 mm and 109.8 ± 60 mm) and relative humidity (89.0 ± 1.7 % and 79.6 ± 2.8 %) showed a statistical difference in seasonality (p<0.05), different from the temperature (27.1 ± 0.2 ºC and 27.6 ± 0.3 ºC), there was no difference. The litter variables were not influenced by the seasonality of the elements of the climate and did not differ statistically between the rainy and less rainy periods, respectively: amount of accumulated litter (16.46 ± 4.17 Mg.ha-1 and 13.91 ± 7.61 Mg.ha-1), litter carbon stock (7.9 ± 2, 0 Mg.ha-1 and 6.6 ± 3.61 Mg.ha-1), volume (26.21 ± 0.92cm3 and 24.88 ± 0.92 cm3), thickness (4.19 ± 0.92 cm and 3.99 ± 0.95 cm) and density (0.041 ± 0.92 g/cm3 and 0.034 ± 0.92 g/cm3). The seasonality of the elements of the climate did not show considerable variation to the point of altering the litter physical properties and carbon content.
