ABSTRACT This study assessed the genetic variability in open-pollinated progenies of Eucalyptus urophylla for resistance to rust (Puccinia psidii). The progeny trial was conducted on a statistical randomized block design with 20 progenies, five plants per plot, and nine replications. Analysis of variance showed high genetic variability for the studied trait, with potential for selection gains. The genetic variability of this population provides support to conduct a breeding program with superior individuals for rust resistance, allowing low costs and minimizing the yield losses on eucalyptus plantations.

RESUMO O uso de clones adaptáveis a regiões com déficit hídrico e épocas bem definidas e prolongadas de seca, como acontece no Oeste da Bahia, é uma forma de superar tal estresse. O manejo do K também interfere neste aspecto, pois regula a abertura e o fechamento dos estômatos, impedindo que a planta perca água desnecessariamente, tornando-a mais eficiente no uso da água. Logo, o objetivo deste trabalho foi avaliar o desempenho produtivo de clones de eucalipto e doses de potássio para as condições edafoclimáticas do município de Luis Eduardo Magalhães, localizado no Oeste do estado da Bahia, visando à produção energética. O delineamento experimental foi em blocos ao acaso com quatro repetições, no esquema de parcelas subdivididas. Foram testados seis clones de eucalipto (AEC-056, AEC-144, AEC-220, AEC-224, AEC-103 e AEC-1528), e quatro doses de K2O (0, 30, 60 e 120 kg ha-1). Aos dois anos de idade o clone AEC 1528 foi o que apresentou maior crescimento em altura e diâmetro enquanto o AEC 056 o menor. O clone AEC 1528 apresentou maior produtividade. Notou-se exigência diferenciada de K entre os clones de eucalipto tanto para o crescimento quanto produtividade.

ABSTRACT The use of clones adapted to regions with water deficit caused by well-defined and prolonged dry periods, as happens in the western part Bahia, is a way to overcome water stress. The adjustment of potassium (K) also influences this aspect, because it regulates the opening and closing of stomata, impeding water loss by plants and making them more efficient in water use. Therefore, the aim of this study was to evaluate the performance of eucalyptus clones grown for energy production in response to potassium levels in soil and climate conditions, in the municipality of Luis Eduardo Magalhães, located in western Bahia state. A randomized block with four replications in a split plot was used as experimental design. Six eucalyptus clones (AEC-056, CEA-144, CEA-220, CEA-224, CEA-103 and CEA-1528) and four doses of K2O (0, 30, 60 and 120 kg ha-1) were tested. At two years old, clone 1528 showed greatest productivity, with the tallest height and trunk diameter, while 056 showed the lowest performance. Different K requirements were observed among eucalyptus clones for both growth and productivity.

RESUMO O reflorestamento comercial de espécies nativas brasileiras, visando à produção de madeira nobre para serraria, tem se intensificado nos últimos anos em diversas regiões do Brasil. Dentre as espécies nativas potenciais que veem sendo plantadas por empresas do setor florestal, o guanandi (Calophyllum brasiliense Cambess.) é destaque, uma espécie com ampla distribuição natural, alta adaptabilidade a diferentes condições de solo e clima, boa forma fuste e ótima qualidade madeira. O desenvolvimento de um bom programa de melhoramento genético requer prioridade para a obtenção de ganhos de produtividade e rendimento no médio e longo prazo. Nesta linha de pesquisa, o estudo das técnicas de propagação vegetativa, visando abreviar etapas no melhoramento florestal e viabilizar a produção maciça de material melhorado, tem importância fundamental no sucesso do programa de melhoramento. Com objetivo de avaliar a viabilidade da propagação vegetativa da espécie, foram realizados dois experimentos sucessivos, instalados e avaliados ao longo de dois anos, a fim de identificar os melhores tipos de estacas, doses de hormônio e manejo. Foram testados diferentes tipos de estacas, submetidas a doses crescentes de ácido indolbutírico (AIB), avaliados principalmente os dados de sobrevivência, emissão de brotos e formação de raiz. Os resultados indicaram que a espécie se mostrou viável a propagação vegetativa, obtendo índices de 85% a 90% de enraizamento de estacas provenientes de mudas, para as doses de ácido indolbutírico (AIB) nas concentrações entre 3.000 a 7.000 mg.L-1. Para as estacas de brotações da base de árvores adultas 3.000 mg.L-1 foi a melhor dose de AIB.

ABSTRACT Commercial reforestation of Brazilian native species to produce hardwood for sawmills has been recently intensified in the country. Among the potential species planted by the logging industry is guanandi (Calophyllum brasiliense Cambess) because it is widely distributed in the country, highly adapted to different soil and climate conditions, good bole form and high quality timber. The development of genetic improvement programs should prioritize gains in productivity and yields in the medium and long term. For such programs to be successful, the study of vegetative propagation techniques to abbreviate steps in forest improvement and allow its mass production is fundamental. To assess the viability of vegetative propagation of the species, two successive experiments were carried out during two years testing the best type of cutting, hormone concentration and management. Different cuttings types submitted to increasing doses of indole-butyric acid (IBA) were tested to evaluate survival, sprouting, rooting and callus formation. Results indicate that the species is viable for vegetative propagation with 85 to 90% rooting of cuttings from seedlings in the IBA concentrations of 3000 to 7000 mg.L-1. For the cuttings, sprouting from the base of adult trees 3000 mg.L-1 was the best concentration of IBA.

O objetivo deste estudo foi estimar os parâmetros genéticos quantitativos das progênies sob o efeito de geada, bem como determinar a interação genótipos x ambientes das progênies de Eucalyptus dunnii em dois locais de estudo. Os testes de progênies foram implantados em 2010, utilizando 20 progênies de polinização aberta de E. dunnii. Foram avaliados: altura de planta (ALT), diâmetro à altura do peito (DAP) e volume de madeira em metros cúbicos (VOL). Foram realizadas as análises estatísticas e descritivas, correlação fenotípica/genotípica por local e análise de variância conjunta, sendo estimados os parâmetros genéticos. Foram detectadas diferenças significativas (p≤0,05) entre progênies para as características silviculturais, o que sugere a possibilidade de melhoramento pela seleção. Pela análise conjunta, a interação progênies x ambientes (G x E) não apresentou diferença significativa, confirmando que as progênies superiores identificadas nos testes podem ser indicadas para ambos os locais. Os resultados de correlações genéticas e fenotípicas variaram de baixos a altos (0,23 a 1,0) para as características estudadas, sendo as correlações, em sua grande maioria, significativas (p≤0,05), em que o DAP foi altamente correlacionado com o VOL. A população, por apresentar alta variação genética e controle genético das características, permite a obtenção de ganhos com a seleção entre as progênies.

ABSTRACT The aim of this study was to estimate quantitative genetic parameters of progenies under frost situations and to determine the interaction of genotype x environments for Eucalyptus dunnii progenies in two different sites. The progeny trials were set up in 2010, using 20 open pollination progenies of E. dunnii. The following variables were evaluated: plant height (HGT), diameter at breast height (DBH), and wood volume (VOL) in cubic meters. Descriptive and statistical analysis were used as well as phenotypic/genotype correlation by location and joint variance analysis and the genetic parameters were estimated. Significant differences were detected (p≤ 0,05) among progenies for silvicultural characters, suggesting the possibilities for selection gain. By the joint analysis, the interaction of progenies x environments (P x E) did not show significant differences, confirming that the superior progenies identified in the tests can be used for both studied locations. The genetic and phenotypic correlations results ranged from low to high (0.23 to 1.0) values for the studied characters, and the great majority of correlations were significant (p≤ 0,05) in which the diameter at breast height was highly correlated with volume. Because of high genetic variability and genetic control of the characters, the population allows to obtain gains with selection between in the progenies.

Avaliou-se a variabilidade genética em progênies de Eucalyptus dunnii Maiden para resistência à ferrugem (Puccinia psidii). Experimentos de campo foram instalados em duas regiões edafoclimáticas.Progênies de polinização aberta foram estabelecidas em um delineamento emblocos casualizados. Em condições de campo foram avaliadas 60 e 48 progênies emdois locais, com seis repetições e oito plantas por parcela. Instalou-se tambémum experimento em ambiente controlado, com 53 progênies, em blocoscasualizados, com seis repetições e nove plantas por parcela. Foram avaliadas altura da planta e a severidade de ataque que determinam a fase mais suscetível à doença foliar. A população de E. dunnii estudada apresentou altavariabilidade genética para resistência à ferrugem, com coeficiente de variaçãogenética de 36, 07% e 70% das progênies avaliadas foram imunes a ferrugem. Istoindica alto potencial para seleção e melhoramento da espécie.

This study investigated the genetic variability in progenies of Eucalyptus dunnii Maiden for resistance against rust (Puccinia psidii). Field experiments were installed in two regions with differentsoil-climatic conditions. Open-pollinated progenies were established in arandomized complete block design. Sixty and 48 progenies were evaluated underfield conditions at two sites, respectively, with six replications and eight trees per plot. In another experiment in a controlled environment, 53 progenies were evaluated in randomized blocks with six replications and nine plants perplot. The following traits were evaluated: plant height, severity of pestattack and the most susceptible stage to the leaf disease. The genetic variability for rust resistance in the E. dunnii population under studywas high, with a genetic coefficient of variation of 36.07%; 7% of thee valuated progenies were rust-resistant. It indicates a high potential for selection and breeding of the species.

The guapuruvu is pioneer specie by fast growing of legume family with great importance in restoration projects. The aim of the study was to assess the genetic variability of offspring of Schizolobium parahyba through quantitative genetic parameters. The progeny test was installed in a randomized block design in three replications four plants per plot with 60 progenies. Was evaluated plant height, diameter at breast height with the ages: 2, 7, 14, 20 and 66 months and the percentage of survival at 66 months. For the estimates of genetic parameters, the data were analyzed by software SELEGEN. The heritability coefficients at individual level (ĥa²) were higher by height (0.318, 0.012, 0.247, 0.390 and 0.012) compared to DBH (0.021, 0.024 and 0.015) in the 14, 20 and 66 months, respectively. The highest genetic correlation between characters and plant height DBH occurred in the period of 14 months (0.85). The progenies studied, despite not show significant genetic variability, have the potential to initiate programs of forest breeding and for conservation genetics.

O guapuruvu é uma espécie pioneira de rápido crescimento da família Leguminosae de grande importância em projetos de restauração. O objetivo do estudo foi verificar a variabilidade genética de progênies de Schizolobium parahyba da região de Botucatu/SP, por meio de parâmetros genéticos quantitativos. O teste de progênies foi instalado no delineamento experimental em blocos casualizados, três repetições, quatro plantas por parcelas e 60 progênies. Foram avaliados os caracteres: altura de plantas (ALT) e diâmetro á altura do peito (DAP) nas idades de 2, 7, 14, 20 e 66 meses e a porcentagem de sobrevivência, aos 66 meses, transformada em log (x+10). Para as estimativas dos parâmetros genéticos, os dados coletados foram analisados pelo software SELEGEN. Os coeficientes de variação genética individual (CVgi %), variação genotípica entre progênies (CVgp %) e variação relativa (CVr), apresentaram estimativas que demonstram uma variabilidade genética restrita e muito influenciada pelo ambiente. Os coeficientes de herdabilidade ao nível individual (ĥa²) foram maiores para altura (0,318, 0,012, 0,247, 0,390 e 0,012) em relação ao DAP (0,021, 0,024 e 0,015) aos 14, 20 e 66 meses, respectivamente. As correlações genotípicas no geral foram altas entre os caracteres ALT e DAP. A maior correlação genética entre os caracteres DAP e ALT de plantas ocorreu entre a idade 14 meses (0,85). As progênies estudadas, apesar de não mostrarem significativa variabilidade genética, são potenciais para se dar início a programas de melhoramento e conservação genética.

Handroanthus heptaphyllus (Vell.) Mattos, popularmente conhecida por ipê-roxo, é uma espécie pertencente à família Bignoneaceae, muito apreciada por sua beleza, madeira de excelente qualidade e utilizada em produtos medicinais e programas de reflorestamento de áreas degradadas, paisagismo e restauração. Neste trabalho, objetivou-se estudar a diversidade genética entre e dentro das populações de H. heptaphyllus por meio de marcadores microssatélites. Foram estudadas 192 plântulas, formadas a partir de sementes colhidas de duas populações, em um total de 30 árvores, de fragmentos florestais naturais na região de Botucatu - SP. Foram analisados oito locos microssatélites, com polimorfismo alélico, variando de seis alelos para o loco TAU22 a 14 alelos para os locos TAU12, TAU30 e TAU31, com número efetivo médio de alelos por loco (Âe) igual a 4,9. As médias para a heterozigosidade esperada (Ĥe), para as duas populações foi de 0,785, a heterozigosidade observada (Ĥo)foi de 0,609 e o índice de fixação (^F) variou pouco entre as populações, com média de 0,222. O valor médio da divergência genética entre as duas populações (Ĝst') foi de 0,100. Conclui-se que a maior diversidade genética ocorre dentro das populações; portanto, em programa de coleta de germoplasma, para a região de Botucatu, é recomendado realizar uma maior amostragem de indivíduos dentro das populações, o que possibilitaria uma boa representatividade genética. As populações estudadas possuem diversidade genética para subsidiar programas de melhoramento genético e conservação de germoplasma.

Handroanthus heptaphyllus (Vell.) Mattos, popularly known as ipê-roxo, is a species of the family Bignoneaceae much appreciated for its beauty, excellent quality wood which is used for making medicinal products and also in reforestation programs of degraded areas, as well as landscaping and restoration. The aim of this study was to investigate the genetic diversity between and within populations of H. heptaphyllus using microsatellite markers. The 192 seedlings were produced from seeds collected on 30 trees into the two populations of natural forest fragments in Botucatu region, São Paulo, Brazil. Eight microsatellite loci were analyzed, with allelic polymorphism varying from six alleles for locus TAU22 to 14 alleles for loci TAU12, TAU30, and TAU31, with an expected mean number of alleles per locus (Âe) of 4.9. The mean expected heterozygosity(Ĥe) for the two populations was 0.785, the mean observed heterozygosity(Ĥo) was 0.609, and the fixation index (^F) was low between populations, with a mean of 0.222. The gene differentiation between the two populations (Ĝst') was 0.100. We concluded that the higher genetic diversity is within populations; therefore, as far as germplasm collection programs in Botucatu region are concerned, it is recommended that a larger sampling of individuals should be considered within populations, thereby providing good genetic representativeness. The populations have enough genetic diversity to support genetic improvement and germplasm preservation programs.

A cultura do Eucalipto tem importância econômica muito elevada no Brasil e vem sofrendo inúmeros ataques de patógenos, além de essa cultura estar sujeita aos inúmeros estresses ambientais. A resistência e sobrevivência desta cultura frente às circunstâncias ambientais desfavoráveis, e ataques de pragas e doenças, revelaram que o Eucalipto apresenta um sistema de defesa evoluído e altamente eficiente. Aqui nós apresentamos os resultados obtidos no projeto do genoma de ESTs do Eucalyptus (FORESTs). Agrupando as seqüências de cDNA expressas (ESTs) obtidas pelo projeto, montou-se grupos (contigs) de seqüências similares provenientes de cada biblioteca de cDNA induzidas e não induzidas por agentes estressantes, identificou-se os grupos de seqüências de cDNA similares a outras moléculas já conhecidas, como moléculas de plantas-sinalizadoras, fitoalexinas e biossíntese de vias de lignina, proteínas-RP e supostos genes correspondendo às enzimas envolvidas na detoxificação de moléculas de oxigênio reativo. Apresentamos também, considerações gerais sobre os mecanismos de defesa do Eucalipto contra o estresse biótico e abiótico. Estas informações são de extraordinário valor para a produção de Eucalipto e ajudaram no futuro a direcionar programas de melhoramento do Eucalipto, visando assim obter plantas com melhor resistência contra os patógenos e estresses ambientais.

Eucalypt plantation has high economical importance in Brazil; however, it has been attacked by various pathogens under different environmental stress conditions. Disease resistance and survival under unfavorable environmental conditions have revealed that the eucalypt has developed highly efficient defense systems. Here we show the results of the Eucalyptus ESTs Genome Project (FORESTs). Using the expressed sequence tags (ESTs) obtained by the Project, contigs of similar sequences from each cDNA library induced and not induced by stress agents were formed, and cDNA sequences similar to other already known molecules, such as plant-signaling molecules, phytoalexins, lignin biosynthesis pathways, PR-proteins and putative genes corresponding to enzymes involved in the detoxification of reactive oxygen species, were identified. We also present general considerations about the mechanisms of Eucalyptus defense against biotic and abiotic stresses. These data are of extreme importance for future eucalypt breeding programs aimed at developing plants with enhanced resistance against pathogens and environmental stresses.

O trabalho foi desenvolvido em Botucatu, SP, e teve por objetivo determinar o momento de ocorrência do máximo potencial de germinação durante a maturação das sementes de canafístula, relacionando-a com a secagem dos frutos. Cinco árvores, em final de floração, tiveram 15 inflorescências etiquetadas em 06/02/ 2002. As colheitas, realizadas semanalmente, foram iniciadas na quinta semana após a etiquetagem (35 DAE) e finalizaram quando ocorreu o início da dispersão dos frutos (98 DAE), totalizando 10 colheitas. Os frutos das cinco plantas foram colhidos e avaliados separadamente. Em cada colheita, os frutos foram divididos em duas porções: uma teve as sementes extraídas (sementes frescas), e a outra foi posta para secar em ambiente natural de laboratório para, então, se extraírem as sementes (sementes secas). Determinaram-se o teor de água e a massa seca de 100 sementes frescas e 100 secas. Os testes de germinação das sementes, frescas e secas, foram realizados com e sem escarificação. O delineamento experimental utilizado foi o de blocos casualizados, considerando-se a árvore e o bloco. A maior capacidade germinativa das sementes foi atingida após a ocorrência do máximo acúmulo de massa. O máximo potencial de germinação, detectado nas sementes escarificadas, foi observado no início da dispersão, quando predominavam sementes duras. A maturação, a germinação e a instalação da dormência em semente imatura foram antecipadas com a sua secagem no interior do fruto separado da planta-mãe.

This study was carried out in Botucatu City, São Paulo State, Brazil with the objective of determining the time of maximum germination during maturation of horsebush seeds, in relation to fruit drying. Five trees, at the end of flowering had 15 inflorescences labeled on 06/02/2002. Weekly harvests were carried out from the fifth week after the inflorescences were labeled (35 DAL - days after labeling) and ended at the beginning of fruit dispersal (98 DAL), totalizing ten harvests. Fruits of five trees were harvested and evaluated separately. For each harvest, fruits were split in two lots: one lot had the seeds extracted for germination (fresh seeds) and the other was dried on room temperature before seed extraction (dry seeds). Water content and dry matter weight of 100 seeds were evaluated on both fresh and dry seeds. Germination tests of fresh and dry seeds were performed with and without scarification. The experiment was arranged in a complete randomized block design, considering a tree as a block. The highest seed germination capacity was reached after the maximum mass accumulation. Maximum germination, was found in scarified seeds at the beginning of seed dispersal, when hard seeds predominated. Maturation, germination, and induction of seed dormancy in immature seeds were anticipated with they dryed inside the fruits separated from the mother tree.

Este estudo visou avaliar a variabilidade e distância genética dentro de uma população-base de melhoramento genético de Eucalyptus grandis. A avaliação da variabilidade genética tem como objetivos principais analisar a base genética da população-base e montar um banco de dados marcadores moleculares da população em análise. Essa população é formada por 327 indivíduos, principalmente das procedências de Coff's Harbour, Atherton e Rio Claro. Devido à heterozigosidade natural dessa população, ela pode ser dividida em diversas subpopulações, de acordo com a latitude e longitude de origem; e dentro de subpopulações, em função do grau de melhoramento genético já realizado do material analisado no Brasil. Isso permitiu avaliar quanto da variabilidade detectada dentro da população-base foi devido a esses fatores: procedência e grau de melhoramento. A aplicação da técnica RAPD permitiu avaliar 70 locos polimórficos, que foram analisados utilizando-se o coeficiente de Jaccard, o que resultou em matrizes de similaridade genética entre os indivíduos. Os dados de similaridade genética posteriormente foram submetidos à análise estatística. Osdados indicaram que a população-base apresenta ampla base genética, com média de similaridade genética de 0,328. O subgrupo denominado Região 3, composto por material selvagem da macrorregião de Atherton, juntamente com material de APS da macrorregião de Coff's Harbour, foi um dos que mais contribuíram para a ampla base genética da população-base. Foi possível detectar diferença estatística entre as populações selvagens das procedências de Atherton e Coff's Harbour, assim como entre essas procedências e a de Rio Claro.

This study aimed to evaluate the genetic variability among individuals of a base population of Eucalyptus grandis and to build a molecular marker database for the analyzed populations. The Eucalyptus grandis base population comprised 327 individuals from Coff's Harbour, Atherton and Rio Claro. A few plants came from other sites (Belthorpe MT. Pandanus, Kenilworth, Yabbra, etc.). Since this base population had a heterogeneous composition, the groups were divided according to geographic localization (latitude and longitude), and genetic breeding level. Thus, the influence of those two factors (geographic localization and genetic breeding level) on the genetic variability detected was discussed. The RAPD technique allowed the evaluation of 70 loci. The binary matrix was used to estimate the genetic similarity among individuals using Jaccard's Coefficient. Parametric statistical tests were used to compare within-group similarity of the means. The obtained results showed that the base population had wide genetic variability and a mean genetic similarity of 0.328. Sub-group 3 (wild materials from the Atherton region) showed mean genetic similarity of 0.318. S.P.A. (from Coff's Harbour region) had a mean genetic similarity of 0.322 and was found to be very important for maintenance of variation in the base population. This can be explained since the individuals from those groups accounted for most of the base population (48.3% for it). The base population plants with genetic similarity higher than 0.60 should be phenotypically analyzed again in order to clarify the tendency of genetic variability during breeding programs.

Este trabalho foi realizado com o objetivo de avaliar os melhores métodos e épocas de realização da enxertia, sobre o pegamento de enxertos de aroeira-pimenteira (Schinus terebinthifolius Raddi). Foram conduzidos dois experimentos, instalados na Faculdade de Ciências Agronômicas, da Universidade Estadual Paulista - Câmpus de Botucatu-SP, nos períodos de 16 de dezembro de 2005 a 30 de janeiro 2006 e de 03 de junho a 17 de julho de 2006. O delineamento utilizado foi de blocos casualizados, com cinco tratamentos, três repetições cada e dez plantas por parcela. Os tratamentos foram: garfagem no topo em fenda cheia, inglês simples, inglês complicado; borbulhia em "T" normal e "T" invertido. As diferentes épocas do ano alteraram o índice de pegamento, o que restringe o período de coleta de garfos. A maior porcentagem de pegamento ocorreu no mês de dezembro, com a enxertia em fenda cheia (40%). No mês de junho, não houve pegamento significativo, mostrando então que o mês de dezembro é o mês ideal, entre os estudados, para a realização da enxertia em aroeira. As garfagens em geral foram superiores à borbulhia. Houve baixo índice de pegamento, quando não foi nulo, e, além disso, suas borbulhas não apresentaram brotos dentro de 45 dias. A maior porcentagem de plantas com brotos, aos 45 dias, foi pelo método inglês simples, com 92%.

The research was carried out aiming to evaluate the best grafting procedure and period for grafting healing of Brazilian Peppertree (Schinus terebinthifolius Raddi). Two experiments were set up at School of Agronomical Science (FCA) of São Paulo State University - UNESP - Botucatu, São Paulo State, Brazil, through the periods from December 16th, 2005 to January 30th, 2006 and from June 3rd to July 17th, 2006. The experimental design used was the randomized blocks with five treatments, three replications, and ten plants per plot. The treatments were: cleft grafting, splice grafting, whip grafting, normal T-budding, and inverted T-budding. The index of healing is different depending upon the time of the year which limited the period of scion harvest. The highest percentage of healing occurred in December, by cleft grafting (40%). In July there was no significant healing showing that December is the ideal epoch for grafting in Brazilian pepper tree. In general, the grafting was better than budding. There was a low index of healing on budding, when it was not null, and there were not presence of sprouts during the first 45 days. The highest percentage of plants with sprouts, in 45 days, was by the splice grafting procedure (92%).

O Pinus caribaea var. hondurensis Barret & Golfari tem elevada importância como espécie comercialmente plantada; aproximadamente 1,8 milhões de hectares estão ocupados por plantios desta espécie no Brasil. O trabalho teve como objetivo verificar por meio de marcadores microssatélites a variabilidade genética em Pinus caribaea var. hondurensis, bem como sua manutenção durante o processo de melhoramento genético, dentro de uma população- base de melhoramento, uma população de matrizes selecionadas e uma população melhorada F1. Para a realização das análises foi necessária a transferência de primers desenvolvidos para locos microssatélites de outras espécies do gênero. Dos 20 pares de primers testados, 8 foram transferidos para a espécie (RPS 25b, RPS 150, PSM 2, PR 4.6, PtTX 2037, PtTX 3029, RPTest 01 e RPTest 09). Verificou-se a existência de endogamia entre e dentro das populações estudadas, e o maior valor observado entre as populações foi F ST = 0,0213 (população base e F1). A heterozigosidade média observada e a heterozigosidade esperada na população-base foram, respectivamente, H0 = 0,2469 e He = 0,2489. A maior distância genética (D = 0,0119) foi observada entre as populações-base e a população melhorada F1. Através da distância genética entre as matrizes, foram indicados 10 cruzamentos potenciais entre as matrizes mais contrastantes, almejando a obtenção de vigor de híbrido nas progênies obtidas a partir destes cruzamentos.

The Pinus caribaea var. hondurensis Barret & Golfari is highly important as a cultivated species for wooden production and approximately 1.8 million hectare are planted with the species, in Brazil. This research has aimed to verify through SSR markers the genetic variability in Pinus caribaea var. hondurensis, as well as its maintenance during genetic improvement, into the base population for breeding, a population of elite trees and an F1 hybrid population. For this study we have transferred primers of SSR loci developed for other Pinus species. Eight out of 20 tested primers have been successfully transferred to the P. caribaea (RPS 25b, RPS 150, PSM 2, PR 4.6, PtTX 2037, PtTX 3029, RPTest 01, and RPTest 09). Inbreeding was verified within and between populations, and the highest observed value between populations was F ST = 0.0213. The observed heterozigosity (Ho) and the expected heterozigosity (He) means were, respectively, 0.2469 and 0.2489. The highest genetic distance (D = 0.0119) was observed between the base populations and the F1 improved population. Based on genetic distance values obtained for elite trees, 10 potential crossings between contrasting elite trees have been indicated, aiming to reach the hybrid vigor between progenies from those crossings.

Boron (B) is a low mobility plant micronutrient whose molecular mechanisms of absorption and translocation are still controversial. Many factors are involved in tolerance to Boron excess or deficiency. Recently, the first protein linked to boron transport in biological systems, BOR1, was characterized in Arabidopsis thaliana. This protein is involved in boron xylem loading and is similar to bicarbonate transporters found in animals. There are indications that BOR1 is a member of a conserved protein family in plants. In this work, FORESTS database was used to identify sequences similar to this protein family, looking for a probable BOR1 homolog in eucalypt. We found five consensus sequences similar to BOR1; three of them were then used in multiple alignment analysis. Based on amino acid similarity and in silico expression patterns, a consensus sequence was identified as a candidate BOR1 homolog, helping deeper experimental assays that could identify the function of this protein family in Eucalyptus.

This work was aimed at locating Eucalyptus ESTs corresponding to the GS enzyme (Glutamine Synthetase, EC = 6.3.1.2) and to the D1 protein, which are directly related to resistance to herbicides that promote oxidative stress. Glutamine Synthetase corresponds to the site of action of the herbicide glufosinate. Herbicides that belong to groups such as ureas, uracils, triazines and triazinones act on the D1-Qb complex (receptor of electrons from the Photosystem II) by inactivating it. The clusters EGEQRT3302E01.g, EGEQRT3001F12.b; EGEZLV1203B04.g; EGBGFB1211H06.g and EGEZLV1205F09.g enclosed complete sequences (with 356 amino acids) of the Glutamine Synthetase enzyme. The cluster EGEQSL1054G06.g is a consensus of four reads and enclosed a complete sequence of D1 Protein (with 353 amino acids). The comparison of the sequences of Protein D1 from different species showed that the substitutions of serine (S) by glycine (G) or serine (S) by threonine (T) at the position 264 could produce plants resistant to herbicides that act on electron flow on Photosystem II. The sequence of amino acids corresponding to the cluster EGEQSL1054G06.g had a serine in position 264 indicating sensitivity of the Eucalyptus plants to herbicides that act on this site.

This work was aimed at locating Eucalyptus ESTs corresponding to the PROTOX or PPO enzyme (Protoporphyrinogen IX oxidase, E.C. 1.3.3.4) directly related to resistance to herbicides that promote oxidative stress, changing the functionality of this enzyme. PROTOX, which is the site of action of diphenyl-ether (oxyfluorfen, lactofen, fomesafen), oxadiazole (oxadiazon and oxadiargyl), and aryl triazolinone (sulfentrazone and carfentrazone) herbicides, acts on the synthesis route of porphyrins which is associated with the production of chlorophyll a, catalases, and peroxidases. One cluster and one single read were located, with e-values better than e-70, associated to PROTOX. The alignment results between amino acid sequences indicated that this enzyme is adequately represented in the ESTs database of the FORESTs project.
