ABSTRACT Hepatic injuries in COVID-19 are not yet fully understood and indirect pathways (without viral replication in the liver) have been associated with the activation of vascular mechanisms of liver injury in humans infected with SARS-CoV-2. Golden Syrian hamsters are an effective model for experimental reproduction of moderate and self-limiting lung disease during SARS-CoV-2 infection. As observed in humans, this experimental model reproduces lesions of bronchointerstitial pneumonia and pulmonary vascular lesions, including endotheliitis (attachment of lymphoid cells to the luminal surface of endothelium). Extrapulmonary vascular lesions are well documented in COVID-19, but such extrapulmonary vascular lesions have not yet been described in the Golden Syrian hamster model of SARS-CoV-2 infection. The study aimed to evaluate microscopic liver lesions in Golden Syrian hamsters experimentally infected with SARS-CoV-2. In total, 38 conventional Golden Syrian hamsters, divided into infected group (n=24) and mock-infected group (n=14), were euthanized at 2-, 3-, 4-, 5-, 7-, 14-, and 15-days post infection with SARS-CoV-2. Liver fragments were evaluated by histopathology and immunohistochemical detection of SARS-CoV-2 Spike S2 antigens. The frequencies of portal vein endotheliitis, lobular activity, hepatocellular degeneration, and lobular vascular changes were higher among SARS-CoV-2-infected animals. Spike S2 antigen was not detected in liver. The main results indicate that SARS-CoV-2 infection exacerbated vascular and inflammatory lesions in the liver of hamsters with pre-existing hepatitis of unknown origin. A potential application of this animal model in studies of the pathogenesis and evolution of liver lesions associated with SARS-CoV-2 infection still needs further evaluation. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Resumo: O fungo Sclerotinia sclerotiorum, agente causal do mofo-branco, encontra-se disseminado em todo o mundo. A doença é considerada uma das principais da cultura da soja e do feijão no Brasil. O patógeno S. sclerotiorum é disseminado pelas sementes de soja e feijão tanto na forma de escleródios como na forma de micélio dormente no interior das sementes. O objetivo deste trabalho foi avaliar a relação entre diferentes potenciais de S. sclerotiorum nas sementes de soja e feijão e o desempenho dessas sementes, bem como verificar a localização e a quantificação do inóculo nas sementes inoculadas pela técnica de PCR em tempo real (qPCR), validando o termo potencial de inóculo. Sementes de soja e feijão foram inoculadas com o fungo pelo método do condicionamento osmótico baseado na exposição das sementes ao fungo pelos períodos de 24 h, 48 h, 72 h e 96 h. A análise molecular foi realizada por meio de qPCR em sementes inteiras e dissecadas em tegumento, cotilédone e eixo embrionário. Os resultados demonstraram que os efeitos de S. sclerotiorum na germinação e vigor das sementes foram progressivos e proporcionais aos aumentos dos potenciais de inóculo, visto que no maior potencial (P96) foi possível observar danos mais severos nas sementes e, consequentemente, nas plantas emergidas. O inóculo do patógeno foi encontrado em todas as partes das sementes avaliadas, mesmo no seu menor potencial de inóculo (P24), apresentando uma concentração de DNA crescente, onde o tegumento obteve maior quantidade de DNA que o embrião quando comparadas.
Abstract: The fungus Sclerotinia sclerotiorum, the causal agent of white mold, is widespread throughout the world. The disease is considered to be one of the major diseases of soybean and bean crops in Brazil. The pathogen S. sclerotiorum is spread by soybean and bean seeds both in the form of sclerotia and dormant mycelium inside the seeds. The objective of this work was to evaluate the relationship between different potentials of S. sclerotiorum in soybean and bean seeds and the performance of these seeds, as well as to verify the localization and quantification of the inoculum of the pathogen in the seeds inoculated by Real-time PCR (qPCR), validating the term inoculum potential. Soybean and bean seeds were inoculated with the fungus by the osmotic conditioning method based on the exposure of the seeds to the fungus for periods of 24 h, 48 h, 72 h, and 96 h. Molecular analysis was carried out by means of qPCR in whole seeds and dissected in the integument, cotyledon and embryonic axis. The results showed that the effects of S. sclerotiorum on seed germination and vigor were progressive and proportional to the increases in inoculum potentials, since there was more severe damage to the seeds and consequently to the emerged plants at the highest potential (P96). The inoculum of the pathogen was found in all parts of the evaluated seeds, even at its lowest inoculum potential (P24), with an increasing DNA concentration, and the integument obtained a greater amount of DNA than the embryo, in comparison.

Abstract This study assessed the effect of non-thermal plasma (NTP) on Nannochloropsis gaditana cell rupture and subsequent lipid extraction. Micrograph images showed that the surface of original N. gaditana cells appeared smooth and had no apparent holes, whereas after NTP the cells showed a non-uniform apparently fractured surface with many pores and large splits. Maximum lipid recovery from N. gaditana cells (~19%) for NTP-assisted rupture was observed at 10 min, similar to extraction after 10 min sonication (19.0%). The lipid recovery after both pretreatments (average 19.0%) was 2-fold higher than the control treatment (no pretreatment 9.5%). In addition, lipids from unruptured algal cells were mainly composed of ω-3polyunsaturated fatty acids (~31% of total fatty acids), while lipids after NTP were predominantly composed of saturated fatty acids (36.0% of total fatty acids), which could be advantageous for biodiesel production. This study demonstrates an alternative approach based on NTP-technology for algal cell rupture prior to lipid extraction with potential uses for algal biodiesel production.

A simple and efficient microwave-assisted extraction process has been developed in order to extract plumieride from Allamanda cathartica flowers. The extracts have been obtained through microwave-assisted extraction (MAE) process with ethanol and ethyl acetate at different time, power and temperatures. Static maceration at room temperature with ethanol and ethyl acetate was obtained in order to compare the efficiency of extraction. HPLC method was validated to quantify plumieride in the extracts. The analyzed parameters: linearity (R2 = 0.999), precision (CV 3.3%), recovery (98%), specificity (97%), limit of detection (1 µg mL-1), and limit of quantification (2 µg mL-1) were satisfactory. The yield of plumieride reached 43% in the extracts under the optimal MAE conditions (10 min, 300W). Plumieride recoveries by conventional extraction method were 12% with ethanol and 22% with ethyl acetate. Time and power parameters on plumieride extraction yield was determined by applying a factorial design and surface plots. Ethanol was the best extractor solvent and it was possible to obtain high contents (52 mg g-1 of dry flowers) at optimized conditions. It can be concluded that MAE process has actual advantages over the conventional extraction method in terms of shorter time and higher efficiency to recover plumieride from A. cathartica flowers.

ABSTRACT To characterize methicillin-resistant Staphylococcus aureus isolates from an intensive care unit of a tertiary-care teaching hospital, between 2005 and 2010. A total of 45 isolates were recovered from patients admitted to the intensive care unit in the study period. Resistance rates higher than 80% were found for clindamycin (100%), erythromycin (100%), levofloxacin (100%), azithromycin (97.7%), rifampin (88.8%), and gentamycin (86.6%). The SCCmec typing revealed that the isolates harbored the types III (66.7%), II (17.8%), IV (4.4%), and I (2.2%). Four (8.9%) isolates carried non-typeable cassettes. Most (66.7%) of the isolates were related to the Brazilian endemic clone from CC8/SCCmec III, which was prevalent (89.3%) between 2005 and 2007, while the USA100/CC5/SCCmec II lineage emerged in 2007 and was more frequent in the last few years. The study showed high rates of antimicrobial resistance among methicillin-resistant S. aureus isolates and the replacement of Brazilian clone, a well-established hospital lineage, by the USA100 in the late 2000s, at the intensive care unit under study.

<p>Falhas podem ocorrer em conexões de implantes dentários, em especial em hexágonos externos (EH). Devido à ocorrência de deformação nesta porção dos implantes, este estudo objetivou avaliar os níveis de deformação de conexões EH submetidas ao torque interno. Dois tipos de implantes foram utilizados: grupo N e grupo S. Foram aplicados torques de 0, 32, 45, 60 e 80 Ncm nos implantes do grupo N e torques de 0, 30, 40, 60 Ncm e infinito nos implantes do grupo S. Medidas referentes à distância interna (ID), área interna (AI) e área externa (AE) foram obtidas por meio de fotos digitais analisadas em software. A análise estatística foi feita pelo teste de Scott-Knott. Os resultados demonstraram que quanto maior o torque aplicado, maior a alteração de todas as dimensões avaliadas em ambos os grupos. No grupo S, torques iguais ou superiores a 40 Ncm e 30 Ncm causaram maior deformação na AE e AI respectivamente, enquanto no grupo N, torques iguais ou superiores a 60 Ncm e 32 Ncm causaram maior deformação na AE e AI respectivamente. Os níveis de deformação foram maiores no grupo S em comparação ao grupo N. Nossos resultados indicam que a AI, a AE e a DI do EH podem ser influenciadas pelos diferentes torques internos.</p>
<p>Failures may occur in the connections of dental implants, especially in external hexagon (EH). Due to the deformations in this portion of implants, this study aimed to evaluate the levels of deformation of EH connections subjected to internal toque. Two types of implants were used: N group and S group. Torques of 0, 32, 45, 60 and 80 Ncm were applied to the N group, and torques of 0, 30, 40, 60 and infinite Ncm were applied to the S group implants. The internal distance (ID), internal area (IA) and external area (EA) of the EH were obtained from digital pictures, which were analyzed by a specific software. Statistical analysis was performed by the Scott-Knott test. The results showed that the higher the torque applied, the greater were the changes in the evaluated dimensions in both groups. In the S group, torque levels equal or greater than 40 Ncm and 30 Ncm caused greater deformation of EA and IA respectively, while in the N group, torque levels equal or greater than 60 Ncm and 32 Ncm caused greater deformation of EA and IA respectively. Levels of deformation were greater in the S group as compared with the N group. These findings suggest that the IA, EA and ID of the EH may be affected by different internal torque levels.</p>

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The aim of this research was to evaluate the fractions obtained from the leaf, stem and roots of Allamanda schottii Pohl, Apocynaceae, responsible for the cytotoxicity, using several cell lines. Cytotoxicity was correlated with the season the part of the plant, and the major compounds were assessed. The ethanol extracts of leaves, stems and roots obtained at different seasons were evaluated in the human erythromyeloblastoid leukemia cell line (K562). Subsequently the ethanol extracts and dichloromethane fractions collected in winter were evaluated in mouse fibroblast cell line (Mus musculus) (L929), cervix adenocarcinoma (HeLa), human pre-B leukemia (Nalm6), as well as K562 cell line. The compounds plumericin, plumieride and ursolic acid isolated from ethanol extracts of the stems were evaluated in the same cell lines, as well as on breast adenocarcinoma cell line (MCF-7), and Mus musculus skin melanoma cell line (B16F10). The chromatographic profiles of the dichloromethane fractions were obtained by high performance liquid chromatography. The results revealed that the season during which A. schottii was collected, and the part of the plant analyzed, influence the cytotoxicity on the K562 cells tested. On the other hand the dichloromethane fractions, mainly from the stems and roots, are responsible for the cytoxicity on the cells tested. These results may be associated with the seasonal variation of plumericin in these parts of the plant. This information is in accordance with the HPLC analysis. The results clearly show the potential for the phytotherapeutic use of this species, and suggest that the cytotoxic activity observed may be due to the presence of plumericin, or to minor compounds not yet identified. The seasonal influence on the production of secondary metabolites was verified.

Schistosomiasis is a major neglected tropical disease caused by trematodes from the genus Schistosoma. Because schistosomes exhibit a complex life cycle and numerous mechanisms for regulating gene expression, it is believed that spliced leader (SL) trans-splicing could play an important role in the biology of these parasites. The purpose of this study was to investigate the function of trans-splicing in Schistosoma mansoni through analysis of genes that may be regulated by this mechanism and via silencing SL-containing transcripts through RNA interference. Here, we report our analysis of SL transcript-enriched cDNA libraries from different S. mansoni life stages. Our results show that the trans-splicing mechanism is apparently not associated with specific genes, subcellular localisations or life stages. In cross-species comparisons, even though the sets of genes that are subject to SL trans-splicing regulation appear to differ between organisms, several commonly shared orthologues were observed. Knockdown of trans-spliced transcripts in sporocysts resulted in a systemic reduction of the expression levels of all tested trans-spliced transcripts; however, the only phenotypic effect observed was diminished larval size. Further studies involving the findings from this work will provide new insights into the role of trans-splicing in the biology of S. mansoni and other organisms. All Expressed Sequence Tags generated in this study were submitted to dbEST as five different libraries. The accessions for each library and for the individual sequences are as follows: (i) adult worms of mixed sexes (LIBEST_027999: JZ139310 - JZ139779), (ii) female adult worms (LIBEST_028000: JZ139780 - JZ140379), (iii) male adult worms (LIBEST_028001: JZ140380 - JZ141002), (iv) eggs (LIBEST_028002: JZ141003 - JZ141497) and (v) schistosomula (LIBEST_028003: JZ141498 - JZ141974).

Os fermentadores tipo air lift oferecem vantagens tais como: eficiente homogeneização dos componentes, baixo cisalhamento e economia de energia, pois o meio é agitado pelo processo de aeração, sem necessidade de agitação mecânica. O objetivo deste trabalho foi analisar a cinética de crescimento de Saccharomyces boulardii neste fermentador, com aeração de 1 e 1,5 vvm (volume de ar por volume de meio, por minuto), comparada com o crescimento em frascos agitados em shaker, visando a futura aplicação deste fermentador, em escala industrial. Os resultados indicaram que houve uma diminuição do pH com o consumo da glicose do meio, a qual foi totalmente consumida até o final da fase exponencial, de 5 e 6 horas para o shaker e o air lift, respectivamente. Após este período houve uma alteração na velocidade de crescimento de S. boulardii, em ambos os equipamentos, indicando uma possível mudança na fonte de carbono utilizada, uma vez que toda a glicose foi consumida após estes períodos. Os valores de velocidades específicas de crescimento foram semelhantes para o shaker e air-lift com 1,0 vvm, porém inferiores ao air-lift com 1,5 vvm, indicando que neste último reator há possibilidades de se conseguir uma velocidade de produção celular maior, dependendo apenas da eficiência de oxigenação oferecida.
This work aimed to analyze the kinetics of S. boulardii growth in an air lift fermentor under two aeration conditions: 1 and 1.5 air vvm (air volume per volume of culture medium per minute), in comparison with growth in shaken flasks, with a view to the future application of this fermenter on an industrial scale. Air lift fermentors offer advantages such as efficient homogenization of the components, low shear stress, and energy savings because the medium is stirred by aeration and does not require mechanical stirring. The pH of the medium decreased with the glucose uptake during the exponential phase, probably due to the formation of secondary metabolites. The entire process showed a similar specific growth rate during the exponential phase (up to 5 hours in the case of the shaker and 6 hours in that of the air-lift fermentor), indicating a possible change in the carbon source, since all the glucose was depleted after the aforementioned periods. After 6 hours, the growth rates in the shaker and the fermentor were similar under 1.0 vvm, but lower than in the fermentor under 1.5 vvm, suggesting that the fermentor may reach a higher cell production rate depending only on the oxygenation efficiency.

Radiative transition probabilities and lifetimes for the band systems A²pi - X²sigma+ of the isovalent molecules BeF, MgF and CaF

Accurate potential energy curves, transition moments, spectroscopic constants, radiative transition probabilities, and lifetimes for the states X²sigma+ and A²pi of BeF, MgF and CaF are reported using high level calculations. Diagonal transitions dominate for the A²<FONT FACE=Symbol>P ®</FONT> X²sigma+ system. Radiative lifetimes for nu' = 0 are predicted to be 6.81 ns, 7.16 ns and 19.48 ns, respectively for BeF, MgF and CaF. The result calculated for the CaF molecule are in excellent agreement with the experimental result equal to 21.9±4.0 ns.
