ABSTRACT Objective: Adipose tissue-derived stromal/stem cells (ASCs) and vitamin D have immunomodulatory actions that could be useful for type 1 diabetes (T1D). We aimed in this study to investigate the safety and efficacy of ASCs + daily cholecalciferol (VIT D) for 6 months in patients with recent-onset T1D. Materials and methods: In this prospective, dual-center, open trial, patients with recent onset T1D received one dose of allogenic ASC (1 x 106 cells/kg) and cholecalciferol 2,000 UI/day for 6 months (group 1). They were compared to patients who received chol-ecalciferol (group 2) and standard treatment (group 3). Adverse events were recorded; C-peptide (CP), insulin dose and HbA1c were measured at baseline (T0), after 3 (T3) and 6 months (T6). Results: In group 1 (n = 7), adverse events included transient headache (all), mild local reactions (all), tachycardia (n = 4), abdominal cramps (n = 1), thrombophlebitis (n = 4), scotomas (n = 2), and central retinal vein occlusion at T3 (n = 1, resolution at T6). Group 1 had an increase in basal CP (p = 0.018; mean: 40.41+/-40.79 %), without changes in stimulated CP after mixed meal (p = 0.62), from T0 to T6. Basal CP remained stable in groups 2 and 3 (p = 0.58 and p = 0.116, respectively). Group 1 had small insulin requirements (0.31+/- 0.26 UI/kg) without changes at T6 (p = 0.44) and HbA1c decline (p = 0.01). At T6, all patients (100%; n = 7) in group 1 were in honeymoon vs 75% (n = 3/4) and 50% (n = 3/6) in groups 2 and 3, p = 0.01. Conclusions: Allogenic ASC + VIT D without immunosuppression was safe and might have a role in the preservation of β-cells in patients with recent-onset T1D. ClinicalTrials.gov: NCT03920397.

RESUMO: As abelhas mostram-se fundamentais em diversos aspectos, especialmente com relação à biodiversidade de plantas e polinização. Recentemente, estão sendo enfrentadas imensas perdas no número de colônias brasileiras, principalmente nos estados do sul do país, com forte atividade apícola. Há indicativos de que, dentre as razões para as perdas, possam estar envolvidos patógenos que afetam a saúde das abelhas. Dentre eles, o microsporídio Nosema e o vírus da realeira negra (BQCV) destacam-se pela prevalência. Neste estudo, foram avaliadas 92 colônias, de 17 apiários do sul do Brasil, a respeito da infecção por Nosema ceranae, Nosema apis e BQCV. Foram realizadas extrações de ácidos nucleicos e síntese de cDNA a partir de amostras de abelhas adultas, seguidos de Reação em Cadeia da Polimerase-Transcriptase Reversa (RT-PCR). Oito amostras positivas para BQCV foram submetidas a sequenciamento. Os resultados mostraram que N. ceranae e BQCV estão circulando na região sul do país, podendo ser a razão para as perdas de colônias. N. apis não foi encontrado. N. ceranae foi encontrado em 57.6% (53/92) das colônias e BQCV em 32.6% (30/92). Foi encontrada coinfecção por ambos em 25% (23/92) das colônias estudadas, fator que sugere a diminuição da longevidade do hospedeiro por ação sinérgica dos patógenos. As amostras submetidas ao sequenciamento indicaram similaridade de 96.8 a 100% entre elas, além de forte similaridade com sequências da Ásia, Estados Unidos, Alemanha e Peru. Este estudo relata a circulação de N. ceranae e BQCV nos apiários do sul do Brasil, além de ser a primeira análise filogenética da sequência do BQCV brasileiro.

ABSTRACT: Bees are fundamental in several aspects, especially in relation to plant biodiversity and pollination. Recently, immense losses are being faced in the number of Brazilian colonies, mainly in southern states of the country, which has a strong beekeeping activity. There are indications that, among the reasons for the losses, pathogens that affect the health of bees may be involved. Among them, the microsporidium Nosema and the black queen cell virus (BQCV) stand out for their prevalence. In this study, 92 colonies of 17 apiaries from southern Brazil were evaluated for infection by Nosema ceranae, Nosema apis and BQCV. Nucleic acid extractions and cDNA synthesis were performed from adult bee samples, followed by Reverse Transcription Polymerase Chain Reaction (RT-PCR) and multiplex PCR. Eight BQCV positive samples were subjected to sequencing. The results showed that N. ceranae and BQCV are circulating in the Southern region of the country, which may be the reason for the loss of colonies. N. apis was not found. N. ceranae was found in 57.6% (53/92) of the colonies and BQCV in 32.6% (30/92). Co-infection was found in 25% (23/92) of the colonies studied, a factor that is suggested to be reducing the hosts’ longevity due to the synergistic action of the pathogens. The samples submitted to sequencing indicated similarity of 96.8 to 100% between them, in addition to strong similarity with sequences from Asia, United States, Germany and Peru. This study reports the circulation of N. ceranae and BQCV in apiaries in southern Brazil, in addition to being the first phylogenetic analysis of the Brazilian BQCV sequence.

RESUMO: Com grande diversidade genética e distribuição geográfica, Hylaeamys megacephalus (G. Fisher, 1814) ocorre tanto na Amazônia quanto no Cerrado. Visto seu aspecto generalista, esta espécie tolera diversos hábitos alimentares e habitats, ocorrendo em áreas inundadas ou não, sendo predominantemente terrestre, permitindo maior fluxo de genes entre as populações, mesmo em longas distâncias. Apresenta ampla distribuição, e carece de estudos que busquem um melhor entendimento sobre as variações morfológicas resultantes das diferenças impostas pelo meio ao longo de sua distribuição. O estudo teve como objetivo, analisar as diferenças entre as populações de H. megacephalus, com base na craniometria investigando se o ambiente interfere na morfologia. Analisamos um total de 142 espécimes oriundos de coleções científicas de mamíferos, do Museu Nacional, Universidade Federal do Rio de Janeiro (MN-UFRJ), Laboratório de Biologia e Parasitologia de Mamíferos Reservatórios Silvestres, Instituto Oswaldo Cruz, Fundação Oswaldo Cruz (LBCE-Fiocruz) e Laboratório de Biodiversidade, Universidade Federal de Goiás, Regional Jataí, nos quais foram tomadas 20 medidas craniométricas. A craniometria foi explorada nas análises estatísticas de agrupamento de pares não ponderados com médias aritméticas (UPGMA), variação canônica e análise dos Componentes Principais (PCA). Os resultados encontrados nos levaram a concluir a existência de três grupos craniométricos da espécie de H. megacephalus com tendência a se diferenciarem, por influências geográficas.

ABSTRACT: Hylaeamys megacephalus (G. Fisher, 1814) presents great genetic diversity and wide geographical distribution, and occurs in both the Amazon and Cerrado biomes. Because of its generalist aspect, this species tolerates different eating habits and habitats. It occurs in flooded and dry areas and is predominantly terrestrial, which allows greater gene flow between populations even over long distances. Studies that seek a better understanding of morphological variations resulting from differences imposed by the environment throughout this species’ distribution are still lacking. This study aimed to analyze the differences between H. megacephalus populations based on craniometry, investigating whether the environment has an influence on morphology. We analyzed a total of 142 specimens from three scientific mammal collections: National Museum, “Universidade Federal do Rio de Janeiro” (MN-UFRJ); “Laboratório de Biologia e Parasitologia de Mamíferos Reservatórios Silvestres”, “Instituto Oswaldo Cruz”, “Fundação Oswaldo Cruz”(LBCE-Fiocruz); and “Laboratório de Biodiversidade”, “Universidade Federal de Goiás”, “Regional Jataí” (LZE-UFG), and took 20 craniometric measurements. Craniometry was explored using unweighted pair group method with arithmetic mean (UPGMA), canonical variate analysis, and principal component analysis (PCA). The results led us to conclude that there are three craniometric groups of H. megacephalus with a tendency to differentiate as a result of geographical influences.

Resumen: El síndrome de Kabuki consiste en una serie de anomalías congénitas que involucran discapacidad intelectual, hipotonía y dismorfismo facial y corporal. Además de estos signos cardinales, con frecuencia se observan cambios craneofaciales y dentales, aunque no se entienden completamente. Por lo tanto, el objetivo de este artículo es presentar los hallazgos dentales en pacientes con síndrome de Kabuki. Esta es una revisión sistemática desarrollada de acuerdo con el protocolo PRISMA. Se aplicó una estrategia de búsqueda con descriptores ("síndrome de kabuki" AND "odontología") a seis bases de datos. Como criterios de inclusión, seleccionamos informes de casos clínicos que abordaban los hallazgos dentales en uno o más pacientes con síndrome de Kabuki entre 1999 y 2019, sin restricciones de género, edad o idioma. La recolección de datos en todas las bases de datos fue realizada por un evaluador calibrado y los títulos, resúmenes y estudios de texto completo fueron analizados sistemáticamente. Se encontraron once informes de casos que se ajustan a los criterios de inclusión propuestos. Después de la síntesis cualitativa, la ausencia de dientes o la agenesia fueron los hallazgos más reportados, siendo observados por nueve autores. Los hallazgos dentales menos frecuentes en pacientes con este síndrome son taurodoncia, microdoncia, fusión, geminación y dientes cónicos o con forma de destornillador. De hecho, cuando cualquier síndrome o condición genética se relaciona con manifestaciones de interés dental, el cirujano dental debe poder investigarlas, especialmente la agenesia o ausencia de los dientes. Por esta razón, el dentista debe ser consciente de las manifestaciones del síndrome de Kabuki, lo que contribuye a la comprensión y el manejo de los pacientes.

Abstract: Kabuki syndrome consists in a series of congenital anomalies involving intellectual disability, hypotonia, and facial and body dysmorphism. In addition to these cardinal signs, craniofacial and dental changes are frequently observed, although they are not fully understood. Thus, the aim of this article is to present the dental findings in patients with Kabuki syndrome. This is a systematic review developed according to the PRISMA protocol. A search strategy with descriptors ("kabuki syndrome" AND "dentistry") was applied to six databases. As inclusion criteria, we selected clinical case reports that addressed dental findings in one or more patients with Kabuki syndrome between 1999 and 2019, without restrictions regarding gender, age or language. Data collection in all databases was performed by a calibrated evaluator and titles, abstracts and full-text studies were systematically analyzed. Eleven case reports that fit the proposed inclusion criteria were found. After the qualitative synthesis, teeth absence or agenesis were the most reported findings, being observed by nine authors. Less frequent dental findings in patients with this syndrome are taurodontism, microdontia, fusion, gemination and conical or screwdriver-shaped teeth. In fact, when any syndrome or genetic condition is related to manifestations of dental interest, the dental surgeon should be able to investigate them, especially teeth agenesis or absence. For this reason, the dentist should be aware of the manifestations of Kabuki syndrome, contributing to patients' understanding and management.

BACKGROUND Biomphalaria glabrata snails are widely distributed in schistosomiasis endemic areas like America and Caribe, displaying high susceptibility to infection by Schistosoma mansoni. After the availability of B. glabrata genome and transcriptome data, studies focusing on genetic markers and small non-coding RNAs have become more relevant. The small RNAs have been considered important through their ability to finely regulate the gene expression in several organisms, thus controlling the functions like cell growth, metabolism, and susceptibility/resistance to infection. OBJECTIVE The present study aims on identification and characterisation of the repertoire of small non-coding RNAs in B. glabrata (Bgl-small RNAs). METHODS By using small RNA sequencing, bioinformatics tools and quantitative reverse transcription polymerase chain reaction (RT-qPCR), we identified, characterised, and validated the presence of small RNAs in B. glabrata. FINDINGS 89 mature miRNAs were identified and five of them were classified as Mollusk-specific. When compared to model organisms, sequences of B. glabrata miRNAs showed a high degree of conservation. In addition, several target genes were predicted for all the mature miRNAs identified. Furthermore, piRNAs were identified in the genome of B. glabrata for the first time. The B. glabrata piRNAs showed strong conservation of uridine as first nucleotide at 5’ end, besides adenine at 10th position. Our results showed that B. glabrata has diverse repertoire of circulating ncRNAs, several which might be involved in mollusk susceptibility to infection, due to their potential roles in the regulation of S. mansoni development. MAIN CONCLUSIONS Further studies are necessary in order to confirm the role of the Bgl-small RNAs in the parasite/host relationship thus opening new perspectives on interference of small RNAs in the organism development and susceptibility to infection.

São propostas três causas principais para sincronia de populações: fatores exógenos, dispersão e interações interespecíficas. O presente trabalho teve por objetivo testar a influência dos fatores exógenos na sincronia de populações de Sitophilus zeamais (Mots.) (Coleoptera: Curculionidae), isoladas espacialmente (sem dispersão), em microcosmos com diferentes condições ambientais (umidade e temperatura). Doze populações com 20 indivíduos cada, foram divididas, aleatoriamente, em dois tratamentos: com lâmpada e sem lâmpada. O censo dos indivíduos adultos foi realizado semanalmente, durante sete meses. A tendência de crescimento da abundância ao longo do tempo foi eliminada através do ajuste de modelos autoregressivos. A sincronia entre as populações, detectada por meio dos coeficientes de correlação de Pearson e Spearman, foi maior dentro do que entre tratamentos, embora as populações mantidas sem lâmpada tenham sido mais sincrônicas do que as populações com lâmpada. Além de evidenciarem a influência do ambiente nas flutuações populacionais, esses resultados sugerem que o metabolismo e as interações intraespecíficas são fatores importantes na dinâmica populacional. Organismos em ambientes desfavoráveis podem apresentar taxas metabólicas anormais, contribuindo pouco para o crescimento populacional. Logo, populações pequenas sofrem maior influência da estocasticidade demográfica, reduzindo a probabilidade de sincronia entre elas. Nos ambientes mais favoráveis, espera-se que os indivíduos desenvolvam funções metabólicas normais, levando as populações a apresentar taxas de crescimento mais elevadas. Nesse caso, a estocasticidade demográfica tem menor influência, levando as populações sem lâmpada a flutuar de forma mais sincrônica.

Three main causes to population synchrony are proposed: exogenous factors, dispersal and inter-specific interactions. This paper had as main goal to test the influence of the exogenous factors in the synchrony in spatially isolated (i.e., no dispersal) populations of Sitophilus zeamais (Mots.) (Coleoptera: Curculionidae), in microcosms with different environmental conditions (humidity, temperature and light intensity). Twelve populations of 20 individuals each, were randomly assigned between two treatment conditions: with or without light. Population size and environmental factors (temperature and relative humidity) were weekly assessed for seven months. Temporal trend in populations increase was eliminated adjusting autoregressive models. Population synchrony, detected by means of Pearson’s and Spearman’s correlation coefficients, was higher within than between treatments, although the populations kept without lamp were more synchronized than populations with lamp. Besides demonstrating the influence of environment on population fluctuations, these results suggest that metabolism and intra-specific interactions are important factors in population dynamic. Organisms exposed to unsuitable environmental conditions may have abnormal metabolic rates, which negatively influences the population grow. Thus, small populations are more likely to suffer from demographic stochasticity, decreasing the probability of the synchrony among populations. On the other hand, in more suitable environments, individuals are expected to have normal metabolic functions, and so, to achieve higher rates of population grow. In this case, the demographic stochasticity has smaller influence, leading populations without lamp to fluctuate synchronously.

Os cupins normalmente constroem ninhos que apresentam formas e características diferenciadas de acordo com a espécie, oferecendo-lhes proteção, além de manter a coesão da sociedade. Algumas espécies constroem seus ninhos sobre a superfície do solo, enquanto outras preferem a subsuperfície, troncos e galhos de árvores como suporte ou madeira para construção das galerias. O objetivo deste trabalho foi analisar a relação entre a ocorrência de colônias do cupim arborícola Constrictotermes cyphergaster Silvestri e as espécies vegetais suporte em um Cerrado sentido restrito na Serra de Caldas Novas, GO. Os dados sugerem forte associação entre C. cyphergaster e algumas espécies vegetais de porte arbóreo, como Qualea grandiflora Mart., Annona crassiflora Mart., Caryocar brasiliense Camb. e Plathymenia reticulata Benth., evidenciado pelo alto valor do Qui-quadrado (chi2 = 214,986, gl. = 20, P < 0,001). Essa relação é verificada entre outras espécies de cupins e plantas, inclusive na região dos cerrados, e pode estar associada a vários fatores. Por um lado pode ser influenciada pela presença de inimigos naturais (competidores e predadores), e pela produção de substâncias tóxicas por essas plantas, ou por outro lado, pode haver benefício entre as espécies associadas, neste caso indicando relações ecológicas de mutualismo facultativo ou facilitação.

Termites usually build nests differently shaped and characterized according to each species, to protect and keep society cohesion. Some species build nests in the ground, some prefer tree thunks or branches as support, whereas other dig galleries in the wood. The purpose of this study is to analyze the relationship between the occurrence of arboreal termites Constrictotermes cyphergaster Silvestri and tree species that support the nest of this species, in a Cerrado sensu strictu of the Serra de Caldas Novas, GO. Data suggest a association relationship between C. Cyphergaster and the tree species Qualea grandiflora Mart., Annona crassiflora Mart., Caryocar brasiliense Camb. and Plathymenia reticulata Benth., shown by high Qui-squared values (c² = 214.986, gl. = 20, P < 0.001). This relationship may be found among other termites and tree species, including Cerrado biome, and may be due to several factors, such as natural competitors and predators, toxin production by other tree species or benefits between associated species (facultative mutualism or facilitation).
