Medicinal products based on the Cannabis sativa plant are available in Brazil, among them, drug, cannabis products, individual import products, and those provided by patient associations. Despite this, there is no monograph for the evaluation of cannabinoids in Brazilian Pharmacopoeia (BP) or other compendia accepted. Therefore, this work aimed to develop a quick and easy method to quantify cannabinoids in authorized products for medicinal purposes. The method was developed by HPLC-UV, gradient elution composed of formic acid in acetonitrile and water, the flow rate of 0.6 mL min-1 and 220 nm detection. It was possible to resolve the peaks of cannabidiol (CBD), tetrahydrocannabinol (THC), cannabidiolic acid and cannabinol and to quantify CBD (105.35%; 104.07%) and THC (106.62%). The developed method was applied to two products and met the established requirements of precision, accuracy, linearity, and selectivity. This procedure has been tested on four cannabis-based products to attest to its effectiveness. Considering the importance of establishing minimum quality requirements for medicinal products and considering the available products in Brazil, it was found that it is important to evaluate the implementation of other products in the method developed for future validation and later inclusion in the BP.

RESUMO Embora a região semiárida do Nordeste brasileiro seja uma grande produtora de maracujá amarelo, os problemas de sais no solo e nas fontes hídricas, em muitas áreas, têm limitado o crescimento e a produção dessa cultura, o que realça a importância de estudos visando a reduzir tais efeitos. Nesse contexto, objetivou-se com este estudo, avaliar o efeito da aplicação foliar de piruvato, como mitigador do estresse salino, sobre a morfofisiologia do maracujazeiro cv. Redondo Amarelo, em fase de mudas. Os tratamentos constaram de três condutividades elétricas da água de irrigação (CEa: 0,8, 2,4 e 4,0 dS m-1) e três concentrações de piruvato (0, 25 e 50 mM), distribuídos em delineamento experimental de blocos casualizados, em esquema fatorial 3 × 3, com quatro repetições e duas plantas por parcela. A irrigação com águas de CEa de 4,0 dS m-1 reduz as variáveis de crescimento, fitomassa, indice de qualidade de Dickson e aumenta o extravasamento de eletrólitos em mudas de maracujazeiro cv. Redondo Amarelo. A aplicação exógena de piruvato de cálcio (25 e 50 mM) favorece as variáveis de crescimento, fitomassa, indice de qualidade de Dickson, teor relativo de água, suculência foliar e reduz o extravasamento de eletrólitos no limbo foliar do maracujá. A aplicação exógena de piruvato atenua o estresse salino, com efeitos benéficos na taxa de assimilação de CO2, transpiração, eficiência da carboxilação instantânea, eficiência intrínseca do uso da água e massa seca das raízes.
ABSTRACT Although the semi-arid region of Northeast Brazil is a major producer of yellow passion fruit, the problems of salts in the soil and water sources, in many areas have limited the growth and production of this crop, which highlights the importance of studies aimed at reducing such effects. In this context, the objective of this study was to evaluate the effect of foliar application of pyruvate, as a mitigator of salt stress, on the morphophysiology of yellow passion fruit, in seedling phase. The treatments consisted of three electrical conductivities of irrigation water (ECw: 0.8, 2.4, and 4.0 dS m-1) and three concentrations of pyruvate (0, 25, and 50 mM), distributed in a randomized block experimental design in a 3 × 3 factorial scheme, with four replications and two plants per plot. Irrigation with ECw of 4.0 dS m-1 reduces growth variables, phytomass, Dickson quality index, and increases electrolyte leakage in passion fruit seedlings cv. Redondo Amarelo. Exogenous application of calcium pyruvate (25 and 50 mM) increased growth, phytomass, Dickson quality index, relative water content and leaf succulence and reduced electrolyte leakage in the leaf blade in passion fruit. Exogenous application of pyruvate attenuates salt stress, with beneficial effects on CO2 assimilation rate, transpiration, instantaneous carboxylation efficiency, intrinsic water use efficiency, and root dry mass.

Abstract Natural resources are depleted in fragmented landscapes that have their vegetation also altered. As a result, the microhabitat diversity and the composition and distribution of local species are affected. In this study, we evaluated the small mammals’ community diversity, composition and microhabitat selection in two Atlantic Forest fragments, in an ecotone area with the Pampa biome, southern Brazil. We recorded five rodents (Akodon montensis, Oligoryzomys nigripes, Sooretamys angouya, Juliomys pictipes and the exotic Rattus rattus) and one marsupial (Didelphis albiventris). Both fragments were dominated by the generalist rodent A. montensis. Akodon montensis and O. nigripes showed similar habitat preferences: ground covered by rocks and higher values of vegetation obstruction. Sooretamys angouya preferred places with higher abundance of trees. Fruit availability was important for A. montensis and D. albiventris, highlighting the importance of this food resource for local wildlife, and the potential role of these species as seed predators and dispersers. Small species richness, the presence of an exotic species and high dominance level suggest that the study area is highly degraded.

RESUMO Objetivo: Descrever as práticas de ressuscitação volêmica em unidades de terapia intensiva brasileiras e compará-las com as de outros países participantes do estudo Fluid-TRIPS. Métodos: Este foi um estudo observacional transversal, prospectivo e internacional, de uma amostra de conveniência de unidades de terapia intensiva de 27 países (inclusive o Brasil), com utilização da base de dados Fluid-TRIPS compilada em 2014. Descrevemos os padrões de ressuscitação volêmica utilizados no Brasil em comparação com os de outros países e identificamos os fatores associados com a escolha dos fluidos. Resultados: No dia do estudo, foram incluídos 3.214 pacientes do Brasil e 3.493 pacientes de outros países, dos quais, respectivamente, 16,1% e 26,8% (p < 0,001) receberam fluidos. A principal indicação para ressuscitação volêmica foi comprometimento da perfusão e/ou baixo débito cardíaco (Brasil 71,7% versus outros países 56,4%; p < 0,001). No Brasil, a percentagem de pacientes que receberam soluções cristaloides foi mais elevada (97,7% versus 76,8%; p < 0,001), e solução de cloreto de sódio a 0,9% foi o cristaloide mais comumente utilizado (62,5% versus 27,1%; p < 0,001). A análise multivariada sugeriu que os níveis de albumina se associaram com o uso tanto de cristaloides quanto de coloides, enquanto o tipo de prescritor dos fluidos se associou apenas com o uso de cristaloides. Conclusão: Nossos resultados sugerem que cristaloides são usados mais frequentemente do que coloides para ressuscitação no Brasil, e essa discrepância, em termos de frequências, é mais elevada do que em outros países. A solução de cloreto de sódio 0,9% foi o cristaloide mais frequentemente prescrito. Os níveis de albumina sérica e o tipo de prescritor de fluidos foram os fatores associados com a escolha de cristaloides ou coloides para a prescrição de fluidos.
Abstract Objective: To describe fluid resuscitation practices in Brazilian intensive care units and to compare them with those of other countries participating in the Fluid-TRIPS. Methods: This was a prospective, international, cross-sectional, observational study in a convenience sample of intensive care units in 27 countries (including Brazil) using the Fluid-TRIPS database compiled in 2014. We described the patterns of fluid resuscitation use in Brazil compared with those in other countries and identified the factors associated with fluid choice. Results: On the study day, 3,214 patients in Brazil and 3,493 patients in other countries were included, of whom 16.1% and 26.8% (p < 0.001) received fluids, respectively. The main indication for fluid resuscitation was impaired perfusion and/or low cardiac output (Brazil: 71.7% versus other countries: 56.4%, p < 0.001). In Brazil, the percentage of patients receiving crystalloid solutions was higher (97.7% versus 76.8%, p < 0.001), and 0.9% sodium chloride was the most commonly used crystalloid (62.5% versus 27.1%, p < 0.001). The multivariable analysis suggested that the albumin levels were associated with the use of both crystalloids and colloids, whereas the type of fluid prescriber was associated with crystalloid use only. Conclusion: Our results suggest that crystalloids are more frequently used than colloids for fluid resuscitation in Brazil, and this discrepancy in frequencies is higher than that in other countries. Sodium chloride (0.9%) was the crystalloid most commonly prescribed. Serum albumin levels and the type of fluid prescriber were the factors associated with the choice of crystalloids or colloids for fluid resuscitation.

ABSTRACT The thermal and luminous conditions in aviaries affect the production and quality of eggs. Thus, the aim of the present study was to evaluate the thermal and luminous environment of commercial laying hen aviaries equipped with compact fluorescent (CF) and light-emitting diode (LED) lamps and their influence on egg production and quality. The study was carried out in two commercial laying hen facilities with similar construction features and management. The first facility was illuminated with CF lamps and the second with LED lamps. The thermal environment was assessed via air temperature, air relative humidity, and enthalpy. The evaluated lamps provided a similar thermal environment in both facilities. The aviary equipped with LED lamps showed higher illuminance than the one equipped with a CF lamp. Production performance was evaluated through feed intake (FI), feed conversion (FC), mortality (M), and laying percentage (LP). Specific gravity (SG) was used to evaluate eggshell quality. The variables related to production performance and eggshell quality of laying hens raised in aviaries equipped with LED lamps were statistically equal (F-test, p > 0.05) to those raised in aviaries equipped with CF lamps. These results indicate that the replacement of CF lamps with LED lamps does not adversely affect the yield and quality of eggs.

Association between SNP rs527616 in lncRNA AQP4-AS1 and susceptibility to breast cancer in a southern Brazilian population

Abstract Breast cancer (BC) is the leading cause of death by this disease in women worldwide. Among the factors involved in tumorigenesis, long non-coding RNAs (lncRNAs) and their differential expression have been associated. Differences in gene expression may be triggered by variations in DNA sequence, including single nucleotide polymorphisms (SNPs). In the present study, we analyzed the rs527616 (C>G), located in the lncRNA AQP4-AS1, using PCR-SSP in 306 BC patients and 312 controls, from a Brazilian population. In the BC group, the frequency found for CG heterozygotes was above the expected and the overdominant model is the best one to explain our results (OR: 1.70, IC 95%: 1.23-2.34, P<0.001). Furthermore, the SNP were associated with age at BC diagnosis and the risk genotype more frequent in the older age group. According to TCGA data, AQP4-AS1 is down-regulated in BC tissue, and the overexpression is associated with better prognoses, including Luminal A, HER2-, stage 1 of disease and smaller tumor. In conclusion, the CG genotype is associated with increased susceptibility in the southern Brazilian population. This SNP is mapped in the lncRNA AQP4-AS1, showing differential expression in BC samples. Based on these results, we emphasize the potential of the role of AQP4-AS1 in cancer.

Abstract Red cabbage (Brassica oleracea L. var. capitata f. rubra DC.) extract has been demonstrated hypolipidemic and antioxidant capacity. Herein, we investigated the effect of red cabbage aqueous extract (RC) or fenofibrate (FF) in oxidative stress induced by Triton WR-1339 in rats. The antioxidant capacity was evaluated through the superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities and, thiobarbituric reactive species (TBARS) and protein carbonyl (PC) levels in erythrocytes, liver, kidneys, cerebral cortex and hippocampus of male rats. The alterations promoted by Triton WR-1339 in enzymatic antioxidant defense in the liver, kidneys and hippocampus were reversed by RC or FF treatments. The TBARS and PC levels increased in the liver, cerebral cortex and hippocampus of hyperlipidemic rats were decreased by the treatments with RC or FF. These findings demonstrated that RC is a potential therapy to treat diseases not only involving dyslipidemic condition but also oxidative stress.

ABSTRACT Pentavalent antimonials and amphotericin B remain as the main drugs to treat human leishmaniasis. However, the high toxicity and variable efficacy of treatment have stimulated the search for novel drug candidates. Naturally occurring alkaloids have a long history of antileishmanial activity. Here, we investigate the effects of the β-carboline-1-propionic acid alkaloid isolated from Quassia amara L., Simaroubaceae, against Leishmania amazonensis and Leishmania infatum. The alkaloid was isolated after liquid-liquid fractionation followed by chromatographic purification of the Q. amara methanol extract. The antileishmanial activity was evaluated by the microdilution method, using resazurin as the viability indicator. In addition, annexin and propidium iodide were used to detect parasites undergoing apoptosis. The anti-amastigote activity of the β-carboline-1-propionic acid alkaloid was determined by the infection of RAW 264.7 macrophages. The alkaloid displayed leishmanicidal activity against Leishmania amazonensis and L. infantum promastigotes and intracellular amastigotes with 50% inhibitory concentration ranging from 2.7 ± 0.82 to 9.4 ± 0.5 µg/ml and selectivity indexes >10. Moreover, apoptotic Leishmania amazonensis (19.5%) and L. infantum (40.4%) promastigotes were detected after 5 h incubation with the alkaloid. Finally, the β-carboline-1-propionic acid alkaloid inhibited the production of NO of infected macrophages, suggesting that the intracellular amastigote elimination occurs in a nitrosative stress-independent way. The results shown here suggest that the β-carboline-1-propionic acid alkaloid has potential as an antileishmanial agent.

ABSTRACT The objective of this work was to evaluate the thermal environment inside conventional cages for laying hens, with and without the use of protective screens. This research was conducted in two conventional cages (open cage), where each cage had half of its area surrounded by high density polyethylene mesh (HDPE) with a diameter of 2.0 cm, and the other half was kept fully open. Hyline W-36 and Dekalb Brown laying hens were housed in these cages. The wind speed, dry bulb temperature (tdb), black globe temperature, humidity index (BGHI), and enthalpy (H) were analyzed. The results indicate that the use of the screen reduced the wind speed inside the cage by 0.22 m s−1. The presence of the screen affected tdb, BGHI, and H in some of the evaluated periods evaluated, making these values higher than those of the treatment without screen. However, the obtained values are within or below the thermal comfort range for laying birds. Therefore, the use of HDPE screen in the cages altered their internal environment. However, it is not enough to hinder the thermal comfort of the housed animals.

O presente estudo tem o objetivo de investigar diretamente os efeitos comportamentais e antioxidantes da sinvastatina em um modelo de mania bipolar induzida por dimesilato de lisdexanfetamina. Ratos Wistar foram tratados durante 30 dias com simvastatina. No 24º dia após o início do tratamento, foi administrada dimesilato de lisdexanfetamina durante 7 dias em cada rato. Os resultados sugerem que sinvastatina combinada com dimesilato de lisdexanfetamina induziu um aumento significativo na locomoção, e administração de dimesilato de lisdexanfetamina provoca um desequilíbrio oxidativo determinado por um aumento na peroxidação lipídica, oxidação de proteínas e alterações nas atividades de enzimas antioxidantes em áreas do cérebro; adicionalmente, na presença de sinvastatina a maioria destes efeitos foram bloqueados. Estes resultados contribuem para um melhor entendimento dos papeis críticos do tratamento dos distúrbios neuropsiquiátricos com dimesilato de lisdexanfetamina, associado com o aumento do estresse oxidativo e com alterações das enzimas antioxidantes. Tendo em vista o papel central exercido pelo dimesilato de lisdexanfetamina, o efeito antioxidante estabelecido pela terapia com sinvastatina é de grande interesse.
The present study aims to directly investigate the behavioral and antioxidant effects of simvastatin in a model of bipolar mania induced by lisdexamfetamine dimesylate. Wistar rats were treated for 30 days with simvastatin. On the 24th day after the start of treatment, each rat was administered lisdexamfetamine dimesylate for 7 days. The results suggest that simvastatin combined with lisdexamfetamine dimesylate induced a significant increased locomotion and lisdexamfetamine dimesylate administration causes an oxidative imbalance determined by an increment in lipid peroxidation, protein oxidation and alterations in the activities of antioxidant enzymes in brain areas; moreover, in the presence of simvastatin, most of these effects were prevented. These findings contribute to a better understanding of the critical roles of lisdexamfetamine dimesylate in the treatment of neuropsychiatric disorders, associated with increased oxidative stress and changes in antioxidant enzymatic defense. In view of the central role played by lisdexamfetamine dimesylate, the established antioxidant effect of simvastatin therapy is of major interest.

RESUMODevido às ocorrências dos vírus de influenza aviária (H5N1 e H7N2), estudos relacionados à biosseguridade têm sido intensificados em que uma das soluções discutidas é o uso de tela para cercar a área dos galpões evitando o contato das galinhas poedeiras alojadas com outras aves. Neste contexto objetivou-se, com o presente trabalho, avaliar e comparar a utilização de tela para cercar o perímetro de um galpão visando à criação de galinhas poedeiras, sobre o ambiente acústico e aéreo no interior da instalação. Esta pesquisa foi realizada em um galpão comercial do tipo californiano para criação de galinhas poedeiras da linhagem Dekalb Brown. Metade da área do galpão foi cercada por tela de polietileno de alta densidade (PEAD) e a outra metade foi mantida totalmente aberta. Os níveis de ruído, concentração de amônia (NH3) e concentração de dióxido de carbono (CO2) foram mensurados durante 22 semanas. O uso de tela PEAD aumentou o nível de ruído e as concentrações de NH3 e de CO2 no interior da instalação; entretanto, esses valores foram inferiores àqueles que oferecem riscos à saúde das aves e dos trabalhadores.
ABSTRACTDue to the occurrence of the avian influenza viruses (H5N1 and H7N2), studies related to biosafety have been intensified, and one of the discussed solutions is the use of screen to enclose the poultry house area, avoiding the contact of the housed laying hens with other birds. In this context, the aim of the present study was to evaluate and to compare the use of screen, to enclose the perimeter of a laying hen house, on the acoustic and aerial environments inside the facility. This research was carried out in a Californian Dekalb Brown laying hen house. Half of the barn area was enclosed by high density polyethylene (HDPE) screen and the other half was maintained completely open. The levels of noise, ammonia (NH3) concentration and carbon dioxide (CO2) concentration were measured during 22 weeks. The use of HDPE screen increased the noise level and the concentrations of NH3 and CO2 inside of the facility, however, these values were smaller than those which offer health risks to the birds and workers.

Devido às ocorrências dos vírus de influenza aviária (H5N1 e H7N2), estudos relacionados à biosseguridade têm-se intensificado, e uma das soluções discutidas é o uso de tela para cercar completamente o perímetro dos galpões, visando a evitar o contato das galinhas poedeiras alojadas com outras aves. Desta forma, o presente trabalho teve como objetivo avaliar e comparar a utilização de tela cercando o perímetro de galpões convencionais de poedeiras, sobre o ambiente acústico e aéreo no interior da instalação. A pesquisa foi realizada em uma granja comercial para produção de ovos, localizada no sul do Estado de Minas Gerais, e foi conduzida em um galpão aberto, que teve a metade de sua área cercada por tela e a outra metade mantida totalmente aberta. Foram alojadas galinhas poedeiras da linhagem Hyline W-36, sendo analisadas as variáveis de intensidade sonora, concentração de amônia (NH3) e concentração de dióxido de carbono (CO2). As variáveis foram coletadas em seis pontos de medição, em cada região (com e sem tela), dois em cada corredor (norte, central e sul). Os resultados indicaram que o uso de tela não alterou o nível de ruído e as concentrações de NH3 e de CO2 no interior do galpão analisado. Contudo, os níveis mensurados foram inferiores àqueles que ofereceriam riscos à saúde das aves e dos trabalhadores, podendo-se concluir que o uso de tela não influenciou negativamente no ambiente aéreo do galpão.
Avian influenza viruses (H5N1 and H7N2) triggered a great number of studies on biosafety, and one of the discussed solutions is the use of screen to enclose completely the poultry house perimeter, since it can avoid the contact of laying hens with other birds. Thus, the present study aimed to evaluate and compare the use of screen enclosing a conventional laying house perimeter by assessing acoustic and aerial environment inside the facility. The research was performed in a commercial egg production farm located in Southern Minas Gerais State, Brazil. It was carried out in an open facility, in which half area was enclosed by screen and the other half was maintained completely open. Hyline W-36 laying hens were housed. Sound intensity, ammonia concentration (NH3) and carbon dioxide concentration (CO2) were analyzed as variables. These variables were collected from six measuring points in each region (with and without screen), two in each alley (north, central and south). The results indicated that the use of screen did not alter the noise level, NH3 and CO2 concentrations inside the analyzed poultry house. However, the levels measured were smaller than to the thresholds that can offer health risks to birds and workers. Therefore, it may be concluded that the use of screen did not influence negatively the facility aerial environment.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.
