The essential oil extracted from Anemia tomentosa (EOAT) has shown larvicidal potential against Aedes aegypti, based on methods in vitro and in silico. Chromatographic and spectrometric techniques (gas chromatography-mass spectrometry (CG-MS), gas chromatography-flame ionization detector (GC-FID)), as well as mono and bidimensional nuclear magnetic resonance spectroscopy (NMR) were used to assess 10 essential oil components. Chemical composition of essential oil comprised 87.51% sesquiterpenes, with emphasis on presilphiperfolan-1-ol (42.13%) and silphiperfol-5-ene (19.47%). Larvicidal assay conducted in vitro with EOAT has evidenced potential cytotoxic activity up to 48 h exposure to it; mortality rate observed for A. aegypti larvae exposed to essential oil reached 100%. Study conducted in silico with chemical compounds deriving from the herein investigated plant species has evidenced its potential to inhibit acetylcholinesterase in A. aegypti. Activity of triquinane sesquiterpenes ranging from -6.8 to -8.2 kcal mol-1 stood out in comparison to that of temephos (-7.5 kcal mol-1). Chemical compounds identified in the investigated essential oil presented low human and environmental toxicity, as observed in absorption, distribution, metabolism and excretion, and toxicity (ADMETox) predictions. Resumo O aumento da modificação dos habitats e da perda de espécies demanda esforços consistentes para descrever e compreender os padrões de biodiversidade. O programa BIOTA/FAPESP foi criado nesse contexto e é uma iniciativa de sucesso para promover estudos em biodiversidade e conservação no Brasil. O BIOTA/Araçá é um projeto interdisciplinar que promoveu uma avaliação detalhada da biodiversidade da Baía do Araçá, um ecossistema costeiro localizado ao Norte do estado de São Paulo, Sudeste do Brasil. A baía engloba múltiplos habitats, tais como praias, manguezais, costões rochosos, e uma planície de maré, e também fornece importantes serviços ecossistêmicos. Infelizmente, a baía está sujeita à conflitos sócio-ambientais complexos que contrastam demandas econômicas, sociais e ambientais (i.e. a expansão das atividades do porto vizinho vs. a pesca artesanal de pequena escala e a proteção da biodiversidade). O presente estudo apresenta um levantamento das espécies bentônicas que ocorrem nos diferentes habitats da Baía do Araçá, incluindo dados obtidos durante o projeto BIOTA/Araçá e de investigações realizadas anteriormente na área. As espécies bentônicas desempenham um papel importante no ambiente marinho, e estudar a diversidade desses organismos que vivem associados ao fundo é indispensável para compreender o funcionamento do meio ambiente. A macrofauna, meiofauna, e microorganismos associados aos fundos consolidado e inconsolidado foram listados, e informações adicionais foram fornecidas para cada espécie, tais como a distribuição geográfica e nos habitats. O checklist inclui 826 espécies, quase 70% registradas durante o projeto BIOTA/Araçá. Os taxa mais especiosos foram os anelídeos (225 spp.), moluscos (194 spp.), e crustáceos (177 spp.). Entre as espécies bentônicas listadas, sete são endêmicas da Baía do Araçá, 14 são consideradas ameaçadas de extinção, e sete são exploradas economicamente. Abstract Increasing habitat modification and species loss demand consistent efforts to describe and understand biodiversity patterns. The BIOTA/FAPESP Program was created in this context and it has been a successful initiative to promote studies on biodiversity and conservation in Brazil. The BIOTA/Araçá is an interdisciplinary project that provided a detailed evaluation of the biodiversity of Araçá Bay, a coastal seascape located on the North coast of the state of São Paulo, Southeast Brazil. The bay encompasses multiple habitats, such as beaches, mangroves, rocky shores, and a tidal flat, and provides important ecosystem services. Unfortunately, the bay is the subject of complex social-environmental conflicts that oppose economic, social, and environmental demands (i.e., the expansion of neighboring harbor activities vs. small-scale artisanal fisheries and protection of biodiversity). The present study presents a survey of the benthic species occurring in the different habitats of Araçá Bay, including data obtained during the BIOTA/Araçá project and previous assessments of the area. The benthic species play an important role in marine environments and studying the diversity of these organisms that live associated with the bottom is indispensable for comprehending the environment’s functioning. The macrofauna, meiofauna, and microorganisms associated with soft and hard bottom were listed, and additional information, such as the habitat and geographical distribution, were provided for each species. The checklist includes 826 species, almost 70% recorded during the BIOTA/Araçá project. The most speciose taxa were the annelids (225 spp.), mollusks (194 spp.), and crustaceans (177 spp.). Seven benthic species are endemic to Araçá Bay, 14 are considered threatened, and seven are economically exploited. Furthermore, the bay is the type locality of many taxa, and 11 new benthic species were described based on specimens sampled during the project. ABSTRAT: Feline injection-site sarcomas in felines account for more than 40% of cutaneous and subcutaneous neoplasms in felines. The present study aimed to describe the macroscopic and histomorphological findings of feline application/injection sarcomas. Samples from 31 feline tumors with a history of feline application/inflammation sarcoma were re-evaluated regarding histological subtype, mitotic index and score, depth of tissue invasion, and presence of inflammation considering the location, intensity and predominant cell types. Of the 31 samples from felines diagnosed with sarcoma at the application/inflammation site, 87.15% were cats with no defined breed (NDB), with a mean age of 8.5 years. The predominant anatomical sites were the back and flank/abdomen, both with 29% (9/31), and the prevalent histological subtype was fibrosarcoma at 77.4% (24/31), followed by anaplastic giant cell sarcoma at 12.9% (4/31) and myxosarcoma 9.6% (3/31). The histological grade with the highest number of cases was III (51.6%), followed by Grade II (35.4%) and I (12.9%). The mean of the longest axis measurements varied between the different tumor grades without being significant, with the average being 2.5±2.79cm in Grade I tumors and 3.2±2.28cm in Grade II tumors. and 4.68±2.07cm in Grade III tumors. Necrosis was observed in 74.2% of tumors. The tissue inflammation score was mild to moderate in 58% of cases and severe in 32.2%, with lymphocytic and lymphoplasmacytic infiltrates being prevalent, with 25.8% each, followed by lymphoplasmohistiocytic with 22.6%. The infiltration depth was 38.7% in muscle tissue, followed by 32.2% in the subcutaneous tissue. Pleomorphism was accentuated in 51.6%. Desmoplasia was moderate in 45.1%. Satellite nodules were present in 29% of cases, and 19.4% had macrophages with intracytoplasmic content suggestive of adjuvants. Surgical margins were infiltrated (M1) in 48.4% and narrowed in 25.8% (M2). The anatomical locations observed were different from those recommended by the Vaccine-Associated Feline Sarcoma Task Force (VAFST); in most cases, the adjuvanted macrophage was not present. From this data, we can suggest that sarcomas in felines are not only correlated to the vaccine application, corroborating the hypothesis that any material, whether liquid or solid, and any chronic inflammatory process in the subcutaneous tissue of cats can induce the entity if they are predisposed to do so. The detailed histomorphological data evaluated in this study were key points and provided important information about tumor behavior, being a tool for clinical-oncological decision-making. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Hematological problems are associated with Coronavirus disease 2019 (COVID-19). Respiratory impairment is the higher point studied, although without experimental studies related to the oxygen transport performed by erythrocytes. Therefore, we decided to investigate if erythrocytes from COVID-19 patients have their functionality changed. The case-control study included hospitalized patients with a positive real-time polymerase chain reaction (RT-PCR) result admitted to University Hospital. Volunteers (negative RT-PCR results) were recruited as a control group. Thus, we assessed different erythrocytes parameters, oxidative stress markers, and biophysical studies using whole blood and isolated hemoglobin. We found a decrease of 51% in oxygen uptake and reduced antioxidant enzyme activity in COVID-19 patients compared to controls. Raman spectrometry showed structural changes in the hemoglobin and lipids of the erythrocytes from COVID-19 patients; thus, these results were consolidated with an increase in Young’s modulus in erythrocytes followed by morphology changes. Besides, isolated hemoglobin from COVID-19 patients has a distinct interaction profile using a ligand model compared to the control. RESUMO O sensoriamento remoto, por meio das técnicas de razão de bandas para o monitoramento nutricional de plantios clonais de eucalipto, é fundamental para garantir a sanidade, produtividade e reduzir custos de empreendimentos florestais. Objetivou-se aplicar índices de vegetação a partir de imagens de satélite de alta resolução na diagnose nutricional de macronutrientes em plantios de híbridos clonais de Eucalyptus urophylla S. T. Blake. O estudo foi realizado em 62 áreas de seis municípios do estado da Bahia sob cultivo de plantios homogêneos de eucalipto, com idades entre 1,3 e 1,8 anos. O delineamento foi inteiramente casualizado com seis tratamentos constituídos pelo regime pluviométrico (1000 a 1300 mm ano-1 e 1300 a 1600 mm ano-1) e tipos de solo (classe e textura). A diagnose nutricional de nitrogênio (N), fósforo (P), potássio (K), cálcio (Ca), magnésio (Mg) e enxofre (S) foi realizada a partir da análise foliar e da determinação dos índices de vegetação: índice de vegetação por diferença normalizada (NDVI), índice de diferença normalizada da água (NDWI) e índice de vegetação ajustado ao solo (SAVI). As áreas avaliadas possuem alto vigor (NDVI > 0,70) e os maiores valores foram observados nas regiões chuvosas (1300 a 1600 mm ano-1) (médio de 0,78). ABSTRACT Remote sensing through band ratio techniques for nutritional monitoring of clonal eucalyptus plantations is essential to guarantee health and productivity, reducing costs of forestry enterprises. The objective was to apply vegetation indices from high resolution satellite images in the nutritional diagnosis of macronutrients in plantations of clonal hybrids of Eucalyptus urophylla S. T. Blake. The study was carried out in 62 areas of six municipalities in the state of Bahia under cultivation of homogeneous eucalyptus plantations, aged between 1.3 and 1.8 years. The design was completely randomized with six treatments constituted by the rainfall regime (1000 to 1300 mm year-1 and 1300 to 1600 mm year-1) and soil types (class and texture). The nutritional diagnosis of nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg) and sulfur (S) was performed from leaf analysis and determination of vegetation indices: normalized difference vegetation (NDVI), normalized difference water index (NDWI) and soil adjusted vegetation index (SAVI). Cellulosic materials have several applications, from rheological modifiers to structural reinforcement and packaging components. Using cellulosic materials may also contribute to environmental sustainability because it can be sourced from agricultural byproducts. In this work, self-sustained composite films were produced by the casting of TEMPO-oxidized cellulose nanofibers/graphite composite dispersions. Oscillatory rheology and tensile strength tests showed that the presence of graphite did not significantly contribute to the enhancement of neither the rheological nor mechanical properties of the dispersions and films. Morphological analysis showed that particle segregation and setting occurred during film casting, resulting in particle concentration gradient along the thickness of the film. These results could indicate the low exfoliation efficiency of the microfluidization process and, therefore, justify why the graphite did not act as a reinforcement of the cellulose matrix. ABSTRACT The expression ‘you need to know to conserve’ is a well-known cliche among biologists. Documenting the richness of a group of organisms is the first step towards understanding biodiversity and preparing efficient conservation plans. In this context, many efforts have been made to quantify the number of species on Earth and estimate the number of species still unknown to science. A few countries have complete and integrated databases estimating the approximate number of species recorded for their territory, particularly in the Global South. In Brazil, a country of continental dimensions, revealing the richness of the second most diverse clade of invertebrates (=Mollusca) has been a goal of taxonomists. Recently, in an unprecedented, collective, and integrated effort among Brazilian malacologists, it was possible to estimate how many valid species of molluscs are there in Brazil. In this effort, more than 30 mollusc experts joined together to update the Taxonomic Catalogue of the Brazilian Fauna (TCBF), a governmental website that allows a quick and real-time updating of all Metazoan. So far, more than 5,000 updates have been made in TCBF, indicating the presence of 3,552 valid species of molluscs in Brazil, distributed among the main clades as follows: Caudofoveata (10 spp.), Solenogastres (6 spp.), Polyplacophora (35 spp.), Scaphopoda (43 spp.), Cephalopoda (92 spp.), Bivalvia (629 spp.) and Gastropoda (2,737 spp.). RESUMO: A esporotricose é uma infecção fúngica crônica causada por espécies do gênero Sporothrix. A ocorrência de casos refratários ao tratamento de longo prazo, especialmente na região nasal de gatos, alerta para a importância do estudo de sua patogênese. Este estudo teve como objetivo analisar o processo inflamatório das lesões cutâneas de esporotricose felina refratária ao tratamento, por meio de exames citopatológico e histopalógico, e comparar com aspectos clínicos. Treze gatos com lesões cutâneas refratárias ao tratamento com Itraconazol por mais de um ano foram incluídos no estudo. Amostras de lesões cutâneas foram coletadas para análises citopatológicas, histopatológicas e cultura fúngica. Fragmentos de tecidos foram processados e corados pela hematoxilina-eosina (HE) e Prata Metenamina de Grocott (GMS). Duas apresentações clínicas tiveram maior ocorrência: a forma localizada cutânea em animais com bom estado geral e doença estável (n=9, 69,2%); e a forma disseminada cutânea em gatos com estado geral ruim (n=4, 30,8%). A região nasal (84,6%) foi a localização mais frequente das lesões nos gatos com esporotricose refratária ao tratamento. No estudo citopatológico, felinos com menos de duas lesões, e em bom estado geral, (n=9, 69,2%) revelaram ausência ou leve intensidade de leveduras (até 5 leveduras por campo), menor intensidade de macrófagos e neutrófilos, e maior intensidade de células epitelióides, linfócitos, plasmócitos e eosinófilos. Enquanto, (n=4, 30,8%) dos felinos que apresentavam a forma disseminada da esporotricose associada a um estado geral ruim, revelaram acentuada intensidade de leveduras em sua maioria fagocitadas por acentuado número de macrófagos e neutrófilos. Dos animais com bom estado geral, a maioria (n=6, 66,7%) apresentava maior intensidade de eosinófilos. Na histopatologia, o granuloma supurativo mal formado foi o tipo predominante (n=9, 69,2%) nas lesões de esporotricose felina, e (n=4, 30,8%) foram de granulomas bem formados. Granulomas mal formados apresentaram leve a moderada intensidade fúngica (55,6%) nos animais com bom estado geral e lesões localizadas, e acentuada intensidade fúngica (44,4%) nos gatos com a forma disseminada da doença e estado geral ruim. Granulomas bem formados (n=4, 30,7%) apresentaram leve a moderada intensidade de carga fúngica, sendo 75% dos animais com esse tipo de granuloma com apenas uma lesão e bom estadogeral. O tratamento de longo prazo com itraconazol, nesses gatos com esporotricose refratária, pode manter a infecção controlada e lesões localizadas estáveis de forma temporária, contudo a reativação do fungo pode ocorrer levando a uma resposta imunológica exuberante e ineficiente.
ABSTRACT: Sporotrichosis is a chronic fungal infection caused by Sporothrix species. The occurrence of cases that are resistant to long-term treatment, especially in the nasal planum of cats, emphasizes the importance of studying its pathogenesis. The purpose of this study was to analyze and compare the inflammatory process of cutaneous lesions of feline refractory sporotrichosis to clinical aspects through cytopathological and histopathological examination. Moreover, the study included 13 cats with cutaneous lesions that had been resistant to itraconazole treatment for more than a year. Cutaneous lesions samples were collected for cytopathological, histopathological, and fungal culture analyses. Tissue fragments were processed and stained with hematoxylin-eosin (HE) and Grocott methenamine silver (GMS). Further, two clinical presentations had the highest occurrence: the localized cutaneous form in animals with good general condition and stable disease (n=9, 69.2%) and the disseminated cutaneous form in cats with poor general condition (n=4, 30.8%). In cats with refractory sporotrichosis, the nasal planum (84.6%) was the most common location of lesions. In the cytopathological study, cats with fewer than two lesions and in good general condition (n=9, 69.2%) showed absence or mild yeast intensity (up to 5 yeasts per field), lower intensity of macrophages and neutrophils, and higher intensity of epithelioid cells, lymphocytes, plasma cells, and eosinophils. On the other hand, (n=4, 30.8%) of the cats with disseminated sporotrichosis and a poor general condition had a marked intensity of yeasts, which were mostly phagocytosed by an increased number of macrophages and neutrophils. Of those animals with good general condition, the majority (n=6, 66.7%) had higher eosinophil intensity. In histopathology, malformed suppurative granuloma was the predominant type (n=9, 69.2%) in feline sporotrichosis lesions, followed by well-formed granulomas (n=4, 30.8%). Malformed granulomas showed mild to moderate fungal intensity (55.6%) in animals with good general condition and localized lesions while marked fungal intensity (44.4%) in cats with the disseminated form of the disease and poor general condition. Well-formed granulomas (n=4, 30.7%) had mild to moderate intensity of fungal load, and 75% of the animals with this type of granuloma had only one lesion and were in good general condition. Long-term itraconazole treatment in these cats with refractory sporotrichosis can keep the infection under control and localized lesions stable; however, fungus reactivation can occur, resulting in an exuberant and inefficient immune response.

This work describes the importance of using renewable sources in experimental classes for undergraduate students, as well as to spread the ideas of using biomass wastes as a source of products of interest. Therefore, in the proposed experiment, citrus fruit peel waste was employed to obtain pure flavonoids by extraction and recristalization. These procedures are important to reveal to students how bioactive products can be easily obtained from natural sources. In this way, naringin and hesperidin could be efficiently extracted from grapefruit (Citrus x. paradisi L.) and common orange (Citrus sinensis L. osbeck) peel wastes, respectively, using simple alcohols as solvents.

Resumo: O objetivo deste trabalho foi caracterizar bacteriófagos autóctones e determinar sua atividade lítica em Staphylococcus aureus. Seis fagos foram isolados de água de lavagem de pisos de estábulos por meio do enriquecimento de cultura com três estirpes de S. aureus. Todos os fagos foram caracterizados pela digestão do DNA com enzimas de restrição e pelo sequenciamento do fragmento de DNA que codifica a endolisina. Cada fago foi testado contra 100 estirpes de S. aureus isoladas de casos de mastite bovina e de produtos lácteos pelo método de lise em placa. Sequências do gene de endolisina apresentaram alta conservação, com mais de 99% de similaridade a nível do nucleotídeo entre os fagos isolados. Foram identificados três domínios envolvidos no reconhecimento e na lise da parede celular bacteriana. Dois bacteriófagos isolados de estábulos apresentam alta atividade lítica em S. aureus, em ampla gama de estirpes, o que indica seu potencial para estudos de fagoterapia em gado leiteiro ou como agente de controle biológico para produtos lácteos.
Abstract: The objective of this work was to characterize autochthonous bacteriophages and to determine their lytic activity on Staphylococcus aureus. Six phages were isolated from dairy barn flush water through enrichment cultures with three S. aureus strains. All phages were characterized by DNA digestion by restriction enzymes and sequencing of the DNA fragment encoding endolysin. Each phage was tested against 100 S. aureus strains isolated from bovine mastitis and from dairy products using the lysis-plate method. The sequences of the endolysin gene were highly conserved, with nucleotide similarity higher than 99% among the isolated phages. Three domains involved in the recognition and lysis of the bacterial cell wall were identified. Two bacteriophages isolated from the dairy barns present high lytic activity on S. aureus, on a wide range of host strains, indicating their potential for studies on phage therapy in dairy cattle or as a biological control agent for dairy products.

RESUMO: O objetivo desta pesquisa foi avaliar a influência das células do colostro no desenvolvimento imune em bezerras leiteiras durante os primeiros 28 dias de vida. Os animais foram distribuídas em 2 grupos: COL+ (n=9) recebeu colostro fresco de suas próprias mães; e COL- (n=10) recebeu pool de colostro congelado sem células viáveis oriundo de vacas doadoras. Estas bezerras foram avaliadas antes da ingestão do colostro (D0); às 48 horas (D2) e semanalmente entre o D7 e D28. O desenvolvimento da imunidade foi avaliada pela fenotipagem das subpopulações celulares do sangue e produção de citocinas pelas células mononucleares sanguíneas após 72 horas de estimulação com concanavalina A (ConA), Staphylococcus aureus (S. aureus) e Escherichia coli (E. coli) inativadas. O histórico clínico das bezerras foi marcado por elevada frequência de diarreias em ambos os grupos. Entretanto, COL- apresentou maior intensidade de diarreia (escore de fezes ~ 3 de 4) e maior temperatura retal no D7 do que as bezerras COL+. Além disso, broncopneumonia (n=1) e inflamações umbilicais foram diagnosticadas apenas nas bezerras COL-. O grupo COL- apresentou menor concentração de ferro sérico e maior número de linfócitos no D7 do que o grupo COL+. COL- também apresentou maior frequência de anemias que o grupo COL+ no D21 e D28. Em geral, a fração das subpopulações celulares foram semelhantes entre os dois grupos ao longo do tempo estudado, exceto na semana 1, onde as bezerras COL- apresentaram maior proporção de CD45RO+ (P=0.07). Observou-se um constante aumento de CD45RO+ com declínio concomitante de CD45RO- ao longo do estudo, indicando o desenvolvimento da resposta imune. A proporção de células CD14MHCII+ aumentou de acordo com a idade (P≤0.05). As medianas das citocinas produzidas a partir do PBMC não estimuladas apresentaram valores abaixo do nível de detecção em ambos os grupos. O PBMC do COL+ estimulado com ConA secretou elevada concentração de IL17 na semana 2 (COL+=2060.0pg/mL e COL-=0.0pg/ml, P=0.00). PBMC do COL+ estimuladas com S. aureus inativado (P=0.05) e E. coli inativada (P=0.05) secretaram níveis mais elevados de IL17 que as bezerras COL- na semana 4. Outros picos de IL17 foram observados no COL+ na semana 2, porém não foi possível detectar diferenças estatísticas entre os grupos. Conclui-se que as bezerras alimentadas com colostro fresco e congelado apresentaram perfil similar entre as proporções das subpopulações celulares. O aumento de células expressando os marcadores de memória CD45RO+ e CD14MHCII+ demonstram, ao longo do experimento, o amadurecimento do sistema imune específico das bezerras após estimulação natural por patógenos após o nascimento. A maior secreção de IL17 pelas células das bezerras COL+ indica que as células maternas podem modular resposta imune Th17 direcionada aos antígenos bacterianos. Este mecanismo poderia ser responsável pela rápida e eficiente quimiotaxia de neutrófilos e eliminação dos microrganismos bacterianos. Os diferentes perfis de citocinas podem ser responsáveis pelos diferentes históricos clínicos relatados para as bezerras COL+ e COL-.
ABSTRACT: The objective of this study was to evaluate the influence of viable cells from colostrum on immune development in dairy heifer calves during the first 28 days of life. The animals were distributed between 2 groups: COL+ (n=9) receiving fresh whole colostrum from their own damns; and COL- (n=10) receiving pooled frozen colostrum, containing no viable cells, from a pool of donor cows. These calves were assessed before colostrum intake (D0), 48 hours of age (D2), and weekly from D7 to D28. The development of immunity was evaluated by assessment of the phenotype of blood leukocyte subsets, and induced cytokine production after 72 hours of stimulation in culture with concanavalin A (ConA), killed Staphylococcus aureus (S.aureus) and killed Escherichia coli (E. coli) by peripheral blood mononuclear cell (PBMC). The clinical history of these calves was marked by a high frequency of diarrhea in both groups. However, COL- had greater diarrhea intensity scores (fecal score~3 of 4), and rectal temperature on D7 than COL+ calves. Moreover, bronchopneumonia (n=1) and navel inflammation were observed only in COL- calves. COL- had a lower concentration of serum iron, and a higher absolute number of lymphocytes on D7 than COL+. COL- also had a higher percentage of anemic calves than the COL+ calves on D21 and D28. In general, the percent of cells within each subset of leukocytes was similar between the groups over the experiment, except on week 1 when COL- calves had a higher percentage of lymphocytes expressing CD45RO+ (P=0.07). A steady increase in CD45RO+ and concomitant decline in CD45RO- leukocytes was observed over the course of the study, indicating the development of immune memory. The proportion of CD14MHCII+ leukocytes increased with age (P≤0.05). The median background cytokine production by PBMC that were not stimulated was below the level of detection of the assays used for both groups. The PBMC from COL+ calves stimulated with ConA secreted a larger quantity of IL-17 week 2 (COL+=2060.0pg/mL and COL-=0.0pg/mL, P=0.00). PBMC from COL+ calves stimulated with killed S. aureus whole cell antigen (P=0.05) and killed E. coli whole cell antigen (P=0.05) also secreted higher levels of IL17 than COL- calves at week 4. Clear production of IL17 was observed in PBML from COL+ calves at week 2, but the difference was not statistical different between groups. In conclusion, calves fed fresh and frozen colostrum showed no difference in cells subset profile overall. The increased percentage of leukocytes expressing the memory CD45RO+ or CD14MHCII+ over the course of the experiment indicated a maturation of the adaptive immune response after natural exposure to pathogens in the environment of the calf. The enhanced IL17 secretion by COL+ calves indicated that viable maternal cells modulated T-cell Th17 production that was primed by bacterial antigens. This mechanism could be responsible for quick and efficient activation of neutrophils for bacterial clearance. The differences in cytokine production observed between groups may help to explain the different clinical pictures observed for calves COL+ and COL- calves.

RESUMO: O objetivo deste estudo foi avaliar a presença de organismos semelhantes a Helicobacter e as alterações endoscópicas e histopatológicas em estômago de gatos domésticos de sangue tipo A. Amostras de antro, corpo e fundo gástricos foram coletadas de 32 gatos, sem raça definida, não domiciliados através de gastroscopia. Teste de urease e análise citológica foram realizados em amostras frescas. Secções teciduais foram processadas e coradas com hematoxilina e eosina e pela prata pelo método de Warthin-Starry para avaliação histológica. Helicobacter spp. foi detectado em 100% das amostras submetidas às análises citológicas e coloração pela prata e em 96,9% das amostras submetidas ao teste de urease. Em 87,5% dos gatos foi identificado infiltrado inflamatório mononuclear. A graduação e distribuição do infiltrado inflamatório nestes gatos revelaram alterações leves (78,1%) a moderada (9,4%) em pelo menos uma região gástrica. Estas alterações eram independentes do escore de colonização. Hiperplasia de folículos linfoides foram detectadas em 3 gatos. Gatos do grupo sanguíneo A são frequentemente colonizados por Helicobacter spp. e os achados macro e microscópicos são consistentes com estudos em gatos domésticos realizados até a presente data. Conclui-se que o grupo sanguíneo mais comum em gatos não está associado com uma alta susceptibilidade a gastrite sintomática causada por Helicobacter spp.
ABSTRACT: The aim of this study was to evaluate the presence of gastric Helicobacter-like organisms and the endoscopic and histopathological changes in domestic cats with blood type A. Samples from the stomach antrum, body and fundus were collected from 32 mixed-breed stray domestic cats using gastroscopy. Urease testing and cytological analysis were performed in fresh samples. Tissue sections were processed and stained with hematoxylin and eosin (H&E) and the Warthin-Starry (WS) silver staining methods for histopathological examination. Helicobacter spp. were detected in 100% of samples subjected to silver staining and cytological analysis, and in 96.9% of samples subjected to urease testing. In 87.5% of the cats, mononuclear inflammatory-cell infiltrates were identified. The graduation and distribution of inflammatory infiltrates in these cats revealed mild (78.1%) to moderate (9.4%) inflammatory changes in at least one gastric region. These changes were independent of the colonization score. Hyperplasia of the lymphoid follicles was detected in three cats. Cats of blood group A are often colonized by Helicobacter spp. and the macroscopic and microscopic findings are consistent with studies in domestic cats reported to date, concluding that the most common blood group in cats is not associated with high susceptibility to symptomatic gastritis.

RESUMO: O presente estudo teve como objetivo avaliar macro e microscopicamente a mucosa gastrintestinal de cães domiciliados, que apresentaram sinais gastrintestinais crônicos, por meio de exame endoscópico e da avaliação histopatológica. Os sinais clínicos, a macro e a microscopia das mucosas gástrica e duodenal foram correlacionados. Foram avaliados 20 cães de diferentes raças, sendo 7 machos e 13 fêmeas, com idade variando entre 1,7 a 15,8 anos, que apresentavam cronicidade dos sinais característicos da Doença Inflamatória Intestinal (DII). O estudo foi prospectivo e transversal, realizado em cães atendidos no Hospital Universitário de Medicina Veterinária da Universidade Federal Fluminense (HUVET - UFF) e em clínicas privadas do município do Rio de Janeiro. Amostras de mucosa gastroduodenal foram obtidas por meio de biópsia endoscópica e foram processadas por técnica histológica de rotina e coradas por Hematoxilina-Eosina e Giemsa. As amostras foram submetidas à análise histopatológica para pesquisa de bactérias espiraladas sugestivas de Helicobacter spp. O teste rápido da urease também foi realizado. Os sinais clínicos, e os achados macro e microscópicos foram confrontados e correlacionados. Neste estudo, foi possível estabelecer correlação positiva entre a perda de peso e a presença de enantema no antro, no corpo e no duodeno além de correlação entre alteração do apetite, a presença de infiltrado inflamatório gástrico e a positividade no teste da urease. Este estudo demonstrou a importância da observação de alguns sinais clínicos como a perda de peso e alterações do apetite no curso da Doença Inflamatória Intestinal. Desta forma, enfatiza-se que cães que apresentem tais sinais clínicos devem ter a Doença Inflamatória Intestinal incluída no grupo de diagnósticos diferenciais evitando, com isso, negligenciar uma enfermidade que, cada vez mais, acomete animais de pequeno porte.
ABSTRACT: The aim of this study was to evaluate macro and microscopic gastrointestinal mucosa of domiciled dogs, with chronic gastrointestinal symptoms by endoscopy and histopathological evaluation, correlating clinical signs, macro and microscopy characteristics/aspects/changes of duodenal and gastric mucosa. Twenty dogs of different breeds were evaluated with with chronic characteristic signs of Inflammatory Bowel Disease (IBD), being 7 males and 13 females, with ages ranging from 1.7 to 15.8 years old. The study was prospective and cross-sectional, conducted in dogs treated at the Teaching Veterinary Medicine Hospital of the Universidade Federal Fluminense (HUVET-UFF) and private clinics in the city of Rio de Janeiro. Gastroduodenal mucosal samples were obtained by endoscopic biopsy and were processed by routine histological technique and stained with hematoxylin- eosin and Giemsa. The samples were submitted to histopathological analysis for the detection of spiral bacteria suggestive of Helicobacter spp. The rapid urease test was also performed. The clinical signs, the macro and microscopic findings were compared and correlated. In this study, we established a positive correlation between weight loss and the presence of erythema in the antrum, body and duodenum as well as correlation between change in appetite and the presence of gastric inflammatory infiltrate and positivity in the urease test. This study demonstrated the importance of the observation of some clinical signs such as weight loss and appetite changes in the course of IBD. Thus, it is emphasized that dogs showing these clinical signs should have the IBD included in the differential diagnoses group avoiding thereby neglecting a disease that increasingly affects small animals.

Background The skin secretions of toads of the family Bufonidae contain biogenic amines, alkaloids, steroids (bufotoxins), bufodienolides (bufogenin), peptides and proteins. The poison of Rhinella schneideri, formerly classified as Bufo paracnemis, presents components that act on different biological systems, including the complement system. The aim of this study was to isolate and examine the activity ofRhinella schneideri poison (RsP) components on the complement system.Methods The components active on the complement system were purified in three chromatographic steps, using a combination of cation-exchange, anion-exchange and gel filtration chromatography. The resulting fractions were analyzed by SDS-PAGE and screened for their activity in the hemolytic assay of the classical/lectin complement pathways. Fractions active on the complement system were also assessed for their ability to generate C3 fragments evaluated by two dimensional immunoelectrophoresis assay, C3a and C5a by neutrophil chemotaxis assay and SC5b-9 complex by ELISA assay.Results The fractionation protocol was able to isolate the component S5 from theRsP, as demonstrated by SDS-PAGE and the RP-FPLC profile. S5 is a protein of about 6000 Da, while S2 presents components of higher molecular mass (40,000 to 50,000 Da). Fractions S2 and S5 attenuated the hemolytic activity of the classical/lectin pathways after preincubation with normal human serum. Both components stimulated complement-dependent neutrophil chemotaxis and the production of C3 fragments, as shown by two-dimensional immunoelectrophoresis. S2 showed a higher capacity to generate the SC5b- 9 complex than the other fractions. This action was observed after the exposure of normal human serum to the fractions.Conclusions This is the first study to examine the activity of RsP components on the complement system. Fractions S2 and S5 reduced the complement hemolytic activity, stimulated complement-dependent neutrophil chemotaxis and stimulated the production of C3 fragments, indicating that they were able to activate the complement cascade. Furthermore, fraction S2 was also able to generate the SC5b-9 complex. These components may be useful tools for studying dysfunction of the complement cascade.
