Abstract Background: Tele-cardiology tools are valuable strategies to improve risk stratification. Objective: We aimed to evaluate the accuracy of tele-electrocardiography (ECG) to predict abnormalities in screening echocardiography (echo) in primary care (PC). Methods: In 17 months, 6 health providers at 16 PC units were trained on simplified handheld echo protocols. Tele-ECGs were recorded for final diagnosis by a cardiologist. Consented patients with major ECG abnormalities by the Minnesota code, and a 1:5 sample of normal individuals underwent clinical questionnaire and screening echo interpreted remotely. Major heart disease was defined as moderate/severe valve disease, ventricular dysfunction/hypertrophy, pericardial effusion, or wall-motion abnormalities. Association between major ECG and echo abnormalities was assessed by logistic regression as follows: 1) unadjusted model; 2) model 1 adjusted for age/sex; 3) model 2 plus risk factors (hypertension/diabetes); 4) model 3 plus history of cardiovascular disease (Chagas/rheumatic heart disease/ischemic heart disease/stroke/heart failure). P-values < 0.05 were considered significant. Results: A total 1,411 patients underwent echo; 1,149 (81%) had major ECG abnormalities. Median age was 67 (IQR 60 to 74) years, and 51.4% were male. Major ECG abnormalities were associated with a 2.4-fold chance of major heart disease on echo in bivariate analysis (OR = 2.42 [95% CI 1.76 to 3.39]), and remained significant after adjustments in models (p < 0.001) 2 (OR = 2.57 [95% CI 1.84 to 3.65]), model 3 (OR = 2.52 [95% CI 1.80 to3.58]), and model 4 (OR = 2.23 [95%CI 1.59 to 3.19]). Age, male sex, heart failure, and ischemic heart disease were also independent predictors of major heart disease on echo. Conclusions: Tele-ECG abnormalities increased the likelihood of major heart disease on screening echo, even after adjustments for demographic and clinical variables. Resumo Fundamento: As ferramentas de telecardiologia são estratégias valiosas para melhorar a estratificação de risco. Objetivo: Objetivamos avaliar a acurácia da tele-eletrocardiografia (ECG) para predizer anormalidades no ecocardiograma de rastreamento na atenção primária. Métodos: Em 17 meses, 6 profissionais de saúde em 16 unidades de atenção primária foram treinados em protocolos simplificados de ecocardiografia portátil. Tele-ECGs foram registrados para diagnóstico final por um cardiologista. Pacientes consentidos com anormalidades maiores no ECG pelo código de Minnesota e uma amostra 1:5 de indivíduos normais foram submetidos a um questionário clínico e ecocardiograma de rastreamento interpretado remotamente. A doença cardíaca grave foi definida como doença valvular moderada/grave, disfunção/hipertrofia ventricular, derrame pericárdico ou anormalidade da motilidade. A associação entre alterações maiores do ECG e anormalidades ecocardiográficas foi avaliada por regressão logística da seguinte forma: 1) modelo não ajustado; 2) modelo 1 ajustado por idade/sexo; 3) modelo 2 mais fatores de risco (hipertensão/diabetes); 4) modelo 3 mais história de doença cardiovascular (Chagas/cardiopatia reumática/cardiopatia isquêmica/AVC/insuficiência cardíaca). Foram considerados significativos valores de p < 0,05. Resultados: No total, 1.411 pacientes realizaram ecocardiograma, sendo 1.149 (81%) com anormalidades maiores no ECG. A idade mediana foi de 67 anos (intervalo interquartil de 60 a 74) e 51,4% eram do sexo masculino. As anormalidades maiores no ECG se associaram a uma chance 2,4 vezes maior de doença cardíaca grave no ecocardiograma de rastreamento na análise bivariada (OR = 2,42 [IC 95% 1,76 a 3,39]) e permaneceram significativas (p < 0,001) após ajustes no modelo 2 (OR = 2,57 [IC 95% 1,84 a 3,65]), modelo 3 (OR = 2,52 [IC 95% 1,80 a 3,58]) e modelo 4 (OR = 2,23 [IC 95% 1,59 a 3,19]). Idade, sexo masculino, insuficiência cardíaca e doença cardíaca isquêmica também foram preditores independentes de doença cardíaca grave no ecocardiograma. Conclusões: As anormalidades do tele-ECG aumentaram a probabilidade de doença cardíaca grave no ecocardiograma de rastreamento, mesmo após ajustes para variáveis demográficas e clínicas. The aim of this work was to extract and characterize pectin from lyophilized noni pulp (Morinda citrifolia Linn.) at three different pHs using different extractor agents and to evaluate anti-viral activity. The extraction of high yield (acid extraction) was subjected to sulfation procedure. Fourier transform infrared spectroscopy and proton nuclear magneticresonance analysis were used to calculate the degree of methylation of pectin from acid extraction and its sulfated derivative. The data obtained for the degree of methylation of noni pectin and sulfated pectin were 30.3 and 30.7, respectively. Pectin was subjected to acute oral toxicity testing in mice. The results showed that pectin had a pro-inflammatory effect, and sulfated pectin had an anti-inflammatory effect. Both pectins were also evaluated for herpes simplex virus activity in vitro, and although they successfully inhibited virus replication, the sulfated pectins were cytotoxic. Morinda Linn. Abstract Thiosemicarbazones are promising classes of compounds with antitumor activity. For this study, six 2,4-dihydroxy-benzylidene-thiosemicarbazones compounds were synthesized. These compounds were submitted to different assays in silico, in vitro and in vivo to evaluate the toxicological, antioxidant and antitumor effects. The in silico results were evaluated by the SwissADME and pkCSM platforms and showed that all compounds had good oral bioavailability profiles. The in vitro and in vivo toxicity assays showed that the compounds showed low cytotoxicity against different normal cells and did not promote hemolytic effects. The single dose acute toxicity test (2000 mg/kg) showed that none of the compounds were toxic to mice. In in vitro antioxidant activity assays, the compounds showed moderate to low activity, with PB17 standing out for the ABTS radical capture assay. The in vivo antioxidant activity highlighted the compounds 1, 6 and 8 that promoted a significant increase in the concentration of liver antioxidant enzymes. Finally, all compounds showed promising antitumor activity against different cell lines, especially MCF-7 and DU145 lines, in addition, they inhibited the growth of sarcoma 180 at concentrations lower than 50 mg/kg. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. SUMMARY Beckwith-Wiedemann syndrome (BWS) is a common genetic congenital disease characterized by somatic overgrowth and its broad clinical spectrum includes pre- and post-natal macrosomia, macroglossia, visceromegaly, increased risk of neonatal hypoglycemia, and development of embryonic tumors. BWS occurs due to genetic/epigenetic changes involving growth-regulating genes, located on region 11p15, with an important genotype-phenotype correlation. Congenital adrenal hyperplasia (CAH) comprises a spectrum of autosomal recessive diseases presenting a variety of clinical manifestations due to a deficiency in one of the enzymes involved in cortisol secretion. Early diagnosis based on newborn screening prevents the adrenal crisis and early infant death. However, high 17-hydroxyprogesterone (17-OHP) levels can occur in newborns or premature infants without CAH, in situations of stress due to maternal or neonatal factors. Here, we report new cases of false-positive diagnosis of 21-hydroxylase deficiency during newborn screening – two girls and one boy with BWS. Methylation-specific multiplex ligation-dependent probe amplification revealed a gain of methylation in the H19 differentially methylated region. Notably, all three cases showed a complete normalization of biochemical changes, highlighting the transient nature of these hormonal findings that imitate the classical form of CAH. This report sheds light on a new cause of false-positive 21-hydroxylase deficiency diagnosis during newborn screening: Beckwith-Wiedemann syndrome. Abstract Objectives: To determine the cut-off point of the cochlear radiation dose as a risk factor for hearing loss in patients with vestibular schwannoma treated with radiosurgery. Methods: A systematic review of the literature was performed without language or publication year restrictions in the MEDLINE/PubMed, EMBASE, Web of Science, LILACS/VHL and Cochrane Library databases. Studies that met the following criteria were included: 1) population: adults of both sexes who underwent radiosurgery for vestibular schwannoma treatment; 2) exposure: cochlear radiation; 3) outcome: hearing loss; 4) type of study: cohort. Two independent reviewers conducted the entire review process. The registration number in PROSPERO was CRD42020206128. Results: From the 333 articles identified in the searches, seven were included after applying the eligibility criteria. There was no standardization as to how to measure exposure or outcome in the included studies, and most studies did not present sufficient data to enable meta-analysis. Conclusion: It was not possible to determine a cut-off point for high cochlear dose that could be considered a risk factor for hearing loss. Hematological problems are associated with Coronavirus disease 2019 (COVID-19). Respiratory impairment is the higher point studied, although without experimental studies related to the oxygen transport performed by erythrocytes. Therefore, we decided to investigate if erythrocytes from COVID-19 patients have their functionality changed. The case-control study included hospitalized patients with a positive real-time polymerase chain reaction (RT-PCR) result admitted to University Hospital. Volunteers (negative RT-PCR results) were recruited as a control group. Thus, we assessed different erythrocytes parameters, oxidative stress markers, and biophysical studies using whole blood and isolated hemoglobin. We found a decrease of 51% in oxygen uptake and reduced antioxidant enzyme activity in COVID-19 patients compared to controls. Raman spectrometry showed structural changes in the hemoglobin and lipids of the erythrocytes from COVID-19 patients; thus, these results were consolidated with an increase in Young’s modulus in erythrocytes followed by morphology changes. Besides, isolated hemoglobin from COVID-19 patients has a distinct interaction profile using a ligand model compared to the control. Abstract: Studies on seed maturation directly contribute to obtaining seeds with a higher standard of physical, physiological, and sanitary quality. Thus, the aim of this study was to verify the morphological and physiological changes during the maturation of Cenostigma tocantinum Ducke seeds. The maturation stages were analyzed: I (293 days after anthesis - DAA), II (322 DAA), III (350 DAA), and IV (356 DAA) through visual, biometric analyses, and quantification of moisture content, dry mass, viability, and seed vigor. During the development of C. tocantinum seeds, changes in fruit coloration and an increase in fruit and seed length, width, and thickness were observed. In the initial stages, the seeds had a high moisture content, which decreased in the later stages. On the other hand, the dry mass of the seeds showed an inversely proportional behavior to the moisture content. Physiological variables performed better in stages III and IV, except for electrical conductivity. It can be concluded that C. tocantinum seeds showed superior physiological quality in stages III or IV, recommending the collection of seeds during these maturation periods. Resumo: Estudos sobre maturação de sementes contribuem diretamente para obtenção de sementes com maior padrão de qualidade física, fisiológica e sanitária. Dessa forma, objetivou-se verificar as alterações morfológicas e fisiológicas durante a maturação de sementes de Cenostigma tocantinum Ducke. Analisou-se os estádios de maturação: I (293 dias após antese - DAA), II (322 DAA), III (350 DAA) e IV (356 DAA) através de análises visuais, biométricas e a quantificação do grau de umidade, massa seca, viabilidade e vigor das sementes. Durante o desenvolvimento das sementes de C. tocantinum observou-se mudanças na coloração dos frutos e um aumento no comprimento, largura e espessura dos frutos e sementes. Nos estádios iniciais, as sementes apresentaram elevado grau de umidade e redução nos estádios finais. Por outro lado, a massa seca das sementes apresentou comportamento inversamente proporcional ao grau de umidade. As variáveis fisiológicas apresentaram desempenho superior nos estádios III e IV, exceto para condutividade elétrica. Lycium humile Phil., es una planta halófita extrema, endémica de Argentina, Bolivia y Chile, que crece entre los 2000 y 4000 m s.m. en humedales salinos. El objetivo del presente trabajo es identificar caracteres adaptativos, morfo-anatómicos e histoquímicos de órganos vegetativos de L. humile relacionados a su tolerancia a ambientes extremos de humedales salinos de altura, con el fin de evaluar su potencial como fuente de metabolitos de interés comercial o de genes relacionados a la tolerancia a factores abióticos. Para llevar a cabo el análisis, durante el periodo de febrero 2018, se obtuvieron muestras de plantas pertenecientes a 4 poblaciones (5 individuos por población) ubicadas en los departamentos de Antofagasta de la Sierra, Santa María y Ambato en la provincia de Catamarca, Argentina. Hojas y tallos se estudiaron mediante técnicas histológicas convencionales y análisis histoquímicos para microscopia óptica. Se realizaron también estudios de composición elemental de diferentes tejidos foliares mediante microscopía electrónica de barrido acoplada a espectroscopia elemental de difracción de rayos X (EDX). Las hojas y tallos de las poblaciones estudiadas mostraron características anatómicas e histoquímicas uniformes. Se describe por primera vez cómo se alcanza la suculencia de las hojas por el plegamiento de los tejidos evidenciado en un patrón de venación tridimensional. Se destaca la presencia de mesófilo isolateral-radial y parénquima acuífero como caracteres únicos para la especie. Se evidencian tricomas glandulares y parénquima en empalizada con contenidos complejos de fenoles, flavonoides, proteínas, lípidos, terpenos y alcaloides; así como depósitos de fenoles, lípidos y terpenos en la capa externa de la cutícula. El análisis EDX mostró idioblastos cristalíferos ricos en contenidos de Ca2+ y Na+ . Los tallos se caracterizaron por presentar hipodermis y cámaras de aire en los estratos corticales y medulares. Estos caracteres anatómicos e histoquímicos de L. humile representan rasgos adaptativos a ambientes salinos de humedales de altura. Se discute su función en relación al ambiente y su potencial como fuente de genes de resistencia y de metabolitos interesantes para la industria cosmética y farmacéutica.

Morpho-anatomy and histochemistry of vegetative organs of Lycium humile (Solanaceae): adaptative traits to high altitude saline wetlands. Darwiniana, nueva serie 11(2): 466-484. Lycium humile Phil., is an extreme halophyte plant, endemic to Argentina, Bolivia, and Chile, which grows between 2000 and 4000 m a.s.l. in saline wetlands environments. The aim of this work is to identify morpho-anatomical and histochemical adaptive characteristics of the vegetative organs of L. humile linked to its tolerance to extreme environments of high altitude saline wetlands, in order to evaluate its potential as source of metabolites with commercial value or as source of resistance genes related to abiotic stress tolerance. To carry out the analysis, during the period of February 2018, samples were obtained from plants belonging to 4 populations of the species (5 individuals per population) located at the departments of Antofagasta de la Sierra, Santa María and Ambato, province of Catamarca, Argentina. Leaf and stems were studied using histochemical tests, conventional histological preparations and optical microscopy. Different foliar tissues were analyzed by scanning electron microscopy coupled to elemental X-ray diffraction spectroscopy (EDS). The leaves and stems of the analyzed populations showed uniform anatomical and histochemical features. It was stated for the first time that the succulence of the leaf is reached by the folding of the tissues evidenced in a tridimensional venation pattern. The presence of isolateral-radial mesophyll and aquifer parenchyma is highlighted as a unique feature characteristic for the species. The glandular trichomes and palisade parenchyma revealed complex contents of phenols, flavonoids, proteins, lipids, terpenes, and alkaloids; while the outer layer of the cuticles presented phenolic, terpenic and lipidic deposits. The EDS analysis showed crystalliferous idioblasts with high contents of Ca2+ and Na+. The stems exhibited hypodermis and air chambers formed in the cortical and medullary layers. These anatomical and histochemical characteristics of L. humile represent adaptive traits to saline high-altitude wetland environments. Its functionality in relation to the environment and its potential as source of resistance genes and metabolites interesting for the cosmetic and pharmaceutical industries are discussed.

Abstract Zoonotic spillover is a phenomenon characterized by the transfer of pathogens between different animal species. Most human emerging infectious diseases originate from non-human animals, and human-related environmental disturbances are the driving forces of the emergence of new human pathogens. Synthesizing the sequence of basic events involved in the emergence of new human pathogens is important for guiding the understanding, identification, and description of key aspects of human activities that can be changed to prevent new outbreaks, epidemics, and pandemics. This review synthesizes the connections between environmental disturbances and increased risk of spillover events based on the One Health perspective. Anthropogenic disturbances in the environment (e.g., deforestation, habitat fragmentation, biodiversity loss, wildlife exploitation) lead to changes in ecological niches, reduction of the dilution effect, increased contact between humans and other animals, changes in the incidence and load of pathogens in animal populations, and alterations in the abiotic factors of landscapes. These phenomena can increase the risk of spillover events and, potentially, facilitate new infectious disease outbreaks. Using Brazil as a study model, this review brings a discussion concerning anthropogenic activities in the Amazon region and their potential impacts on spillover risk and spread of emerging diseases in this region.

ABSTRACT Introduction: Vaccine strategies to enhance CTLs response are still a challenge since it is necessary to favor the exogenous antigens (Ag) cross-presentation by antigen presenting cells (APCs). For this purpose, liposomes encapsulating bacteria pore-forming proteins (PFPs) have been used. Objective. To explore the ability of liposomes co-encapsulating the Ag ovalbumin (OVA) and sticholysin II (StII), a PFP produced by Stichodactyla helianthus, to induce a CTL response, functional in a tumor model. Methods: Liposomes co-encapsulating OVA and StII were prepared by dehydration-rehydration (Lp/OVA/StII). The response of CTLs was evaluated, as well as the antitumor activity in a preventive setting. The ability of StII to stimulate maturation of dendritic cells (DCs) in vitro was also explored. Results: Lp/OVA/StII enhanced the OVA-specific CTL response and antitumor activity in mice challenged with the E.G7-OVA tumor. StII induced the maturation of DCs in vitro, which was dependent on the Toll-like receptor 4 (TLR4), with a contribution to the response of CTLs induced by the liposomal formulation in vivo. Lp/OVA/StII favored in vitro the cross-presentation of OVA and the activation of CD8+ T cells by macrophages, but not by DCs. These results are the first evidence that StII encapsulated into liposomes functions as a vaccine platform to induce a robust CTL response, with a contribution that goes beyond its ability to form pores in membranes.
RESUMEN Introducción: Las estrategias vacunales para potenciar respuestas de linfocitos T CD8+ citotóxicos (CTL) constituyen un reto debido a la necesidad de favorecer la presentación cruzada de antígenos (Ag) exógenos por las células presentadoras de antígenos (APC). Con este propósito se han empleado liposomas que encapsulan proteínas formadoras de poros (PFP) de origen bacteriano. Objetivo: explorar la habilidad de liposomas que co-encapsulan el Ag ovoalbúmina (OVA) y sticholysina II (StII), una PFP producida por Stichodactyla helianthus, de inducir una respuesta de CTL, funcional en un modelo tumoral. Métodos: Liposomas que co-encapsulaban OVA y StII se prepararon mediante deshidratación-rehidratación (Lp/OVA/StII). Se evaluó la respuesta de CTL, así como la actividad antitumoral en un escenario preventivo. La capacidad de StII de estimular la maduración de las células dendríticas (DC) in vitro fue también explorada. Resultados: Lp/OVA/StII potenció la respuesta de CTL específica a OVA y una actividad antitumoral en ratones retados con el tumor E. G7-OVA. StII indujo la maduración de las DC in vitro, lo que resultó dependiente del receptor tipo Toll 4 (TLR4), con una contribución en la respuesta de CTL inducida por la formulación liposomal in vivo. Lp/OVA/StII favoreció in vitro la presentación cruzada de OVA y la activación de células T CD8+ por macrófagos, pero no por DC. Estos resultados son la primera evidencia de que StII encapsulada en liposomas funciona como una plataforma vacunal para inducir una respuesta de CTL robusta, con una contribución que va más allá de su habilidad de formar poros en membranas.

Abstract Mitochondrial Myopathy is a rare pathology caused by a defect in the mitochondrial DNA metabolism, leading to defects in the formation of adenosine triphosphate, in the Krebs citric acid cycle, fatty acid oxidation and oxidative phosphorylation. It is manifested by exercise intolerance, muscle fatigue on small efforts, muscle weakness, tachycardia, and difficulty breathing. There are few case reports on the operative management of adult patients suffering from mitochondrial myopathy. With this report, we intend to describe the anesthetic management of a patient with mitochondrial myopathy who underwent laparoscopic gastroplasty and outline some anesthetic considerations about this pathology.

Abstract Obtaining low cost lignocellulolytic enzymes and efficient biomass pretreatment are key to increase the competitiveness of second-generation ethanol in comparison with fossil fuels. The enzymatic cocktail produced by the Chrysoporthe cubensis fungus as well as the mixture prepared with the cocktails of the Chrysoporthe cubensis and Penicillium pinophilum fungi have already proven to be efficient for hydrolyzing biomass pretreated with alkali. In this study, they were evaluated in saccharification of sugarcane bagasse pretreated with dilute acid or hot water at 121°C using an enzyme loading equal to 8 filter paper units per gram of biomass. The most promising results were obtained from the hydrolysis of biomass pretreated with hot water by the C. cubensis-P. pinophilum enzymes blend. In this condition, the glucose and xylose production were 25.2 g.L-1 and 4.6 g.L-1, respectively, that resulted in the conversion of 68% of glucan and 23% of xylan in only 48 hours. This study shows that the hydrothermal pretreatment is a promising alternative to improve the enzymes performance, produced by the fungi C. cubensis and P. pinophilum, in the sugarcane bagasse hydrolysis without the need of chemical compounds, generally used in the acid and alkali pretreatments. Furthermore, the hydrothermal pretreatment for 60 min allowed all cocktails applied to convert the cellulose efficiently with only 24 h of saccharification, which contributes to the energy savings employed in the process.
