Protein profile as a quality indicator of cryopreserved semen from Tambaqui Colossoma macropomum (Cuvier, 1818)

Resumo O objetivo desse estudo foi de avaliar a associação entre a presença de proteínas no plasma seminal do tambaqui Colossoma macropomum (Cuvier, 1818) com indicadores de qualidade seminal pós-descongelamento. O semen foi criopreservado com diluidor a base de BTS com 8% DMSO. Uma amostra de 200 µL de semen de cada animal foi diluída em 800 µL de BTS, e centrifugada em 800 rpm, e somente o sobrenadante foi criopreservado para posterior análise do perfil proteico do plasma seminal, através da eletroforese unidimensional (SDS-PAGE). Decorridos 15 dias da criopreservação, uma palheta com semen criopreservado foi descongelado para análise dos parâmetros quali-quantitativos. Considerando todas as coletas, o SDS-PAGE identificou 15 bandas proteicas no plasma seminal do tambaqui. Quando se avaliou a interação (presença ou ausência) das proteínas encontradas no plasma seminal, com os parâmetros espermáticos pós-descongelamento, observou-se grande influência da presença das proteínas na qualidade espermática. Observou-se maior taxa de fertilização (P<0,05) com a presença das proteínas 12, 34, 44, 85 e 90 kDa. As proteínas do plasma seminal de tambaqui influenciaram na qualidade espermática após descongelamento, podendo ser utilizadas como indicadores para a qualidade espermática após descongelamento, principalmente as proteínas com peso molecular ≤50 kDa.
Abstract The aim of this study was to evaluate the association between proteins in the seminal plasma of tambaqui Colossoma macropomum (Cuvier, 1818) with seminal quality indicators after thawing. The semen was cryopreserved with a dilution based on BTS with 8% DMSO. A 200 µL sample of semen from each animal was diluted in 800 µL BTS, centrifuged at 800 rpm, and the supernatant was cryopreserved to further analyze of the protein profile of seminal plasma through one-dimensional electrophoresis (SDS-PAGE). After 15 days of cryopreservation, a cryopreserved semen straw was thawed to analyze both qualitative and quantitative parameters. When considering all collections, the SDS-PAGE identified 15 protein bands in the seminal plasma of tambaqui. When the interaction (presence or absence) between proteins observed in the seminal plasma and the post thawed spermatic parameters was evaluated, we observed a great influence of the presence of proteins on spermatic quality. A greater (P<0.05) fertilization rate was observed with the presence of proteins 12, 34, 44, 85, and 90 kDa. Proteins in seminal plasma of tambaqui influenced the spermatic quality after thawing, and thus, they can be utilized as an indicator of sperm quality, especially the proteins with a molecular weight ≤ 50 kDa.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Severity score system for progressive myelopathy: development and validation of a new clinical scale

Progressive myelopathies can be secondary to inborn errors of metabolism (IEM) such as mucopolysaccharidosis, mucolipidosis, and adrenomyeloneuropathy. The available scale, Japanese Orthopaedic Association (JOA) score, was validated only for degenerative vertebral diseases. Our objective is to propose and validate a new scale addressing progressive myelopathies and to present validating data for JOA in these diseases. A new scale, Severity Score System for Progressive Myelopathy (SSPROM), covering motor disability, sphincter dysfunction, spasticity, and sensory losses. Inter- and intra-rater reliabilities were measured. External validation was tested by applying JOA, the Expanded Disability Status Scale (EDSS), the Barthel index, and the Osame Motor Disability Score. Thirty-eight patients, 17 with adrenomyeloneuropathy, 3 with mucopolysaccharidosis I, 3 with mucopolysaccharidosis IV, 2 with mucopolysaccharidosis VI, 2 with mucolipidosis, and 11 with human T-cell lymphotropic virus type-1 (HTLV-1)-associated myelopathy participated in the study. The mean ± SD SSPROM and JOA scores were 74.6 ± 11.4 and 12.4 ± 2.3, respectively. Construct validity for SSPROM (JOA: r = 0.84, P < 0.0001; EDSS: r = -0.83, P < 0.0001; Barthel: r = 0.56, P < 0.002; Osame: r = -0.94, P < 0.0001) and reliability (intra-rater: r = 0.83, P < 0.0001; inter-rater: r = 0.94, P < 0.0001) were demonstrated. The metric properties of JOA were similar to those found in SSPROM. Several clinimetric requirements were met for both SSPROM and JOA scales. Since SSPROM has a wider range, it should be useful for follow-up studies on IEM myelopathies.

Community-Acquired Pneumonia (CAP) is a major public health problem. In Brazil it has been estimated that 2,000,000 people are affected by CAP every year. Of those, 780,000 are admitted to hospital, and 30,000 have death as the outcome. This is an open-label, non-comparative study with the purpose of evaluating efficacy, safety, and tolerability levels of IV azithromycin (IVA) and IV ceftriaxone (IVC), followed by oral azithromycin (OA) for the treatment of inpatients with mild to severe CAP. Eighty-six patients (mean age 56.6 ± 19.8) were administered IVA (500mg/day) and IVC (1g/day) for 2 to 5 days, followed by AO (500mg/day) to complete a total of 10 days. At the end of treatment (EOT) and after 30 days (End of Study - EOS) the medication was evaluated clinically, microbiologically and for tolerability levels. Out of the total 86-patient population, 62 (72.1%) completed the study. At the end of treatment, 95.2% (CI95: 88.9% - 100%) reported cure or clinical improvement; at the end of the study, that figure was 88.9% (CI95: 74.1% - 91.7%). Out of the 86 patients enrolled in the study, 15 were microbiologically evaluable for bacteriological response. Of those, 6 reported pathogen eradication at the end of therapy (40%), and 8 reported presumed eradication (53.3%). At end of study evaluation, 9 patients showed pathogen eradication (50%), and 7 showed presumed eradication (38.89%). Therefore, negative cultures were obtained from 93.3% of the patients at EOT, and from 88.9% at the end of the study. One patient (6.67% of patient population) reported presumed microbiological resistance. At study end, 2 patients (11.11%) still reported undetermined culture. Uncontrollable vomiting and worsening pneumonia condition were reported by 2.3% of patients. Discussion and Conclusion Treatment based on the administration of IV azithromycin associated to ceftriaxone and followed by oral azithromycin proved to be efficacious and well-tolerated in the treatment of Brazilian inpatients with CAP.

Trombose de stent farmacológico no "mundo-real": análise crítica do Registro DESIRE (Drug-Eluting Stent in the Real World)

FUNDAMENTOS: Reportamos a incidência e os preditores de trombose de stent numa grande coorte de pacientes complexos tratados com stents farmacológicos (SF) no "mundo-real". MÉTODO: Entre maio de 2002 e janeiro de 2008, 2.365 pacientes não-selecionados com > 1 lesão coronária com estenose > 50% foram tratados com SF. O seguimento clínico até cinco anos (média 2,2 ± 1,5 anos) foi completado em 98,3%. RESULTADOS: Entre os pacientes avaliados, 29% tinham diabetes, 60% eram portadores de doença multiarterial e 40% apresentavam síndrome coronária aguda (15%, infarto agudo do miocárdio [IAM]). A artéria descendente anterior foi o vaso-alvo mais freqüentemente tratado (35,8%), e 67% das lesões eram de alta complexidade (tipo B2/C). No total, 3.634 SF foram implantados e 40% dos pacientes receberam múltiplos stents. A trombose de stent (classificação Academic Research Consortium) incidiu em 1,6% (n = 38), 60,5% com confirmação angiográfica, 42% ocorreram entre 1 e 12 meses e 47% dos eventos tiveram desfecho fatal. Os preditores independentes de trombose de stent foram: tabagismo atual (razão de risco [RR] 2,59; intervalo de confiança [IC] 95% 1,18-5,67; p = 0,018), intervenção coronária percutânea (ICP) no IAM (RR 3,50; IC 95% 1,31-9,40; p = 0,013), calcificação moderada/grave (RR 2,38; IC 95% 1,34-4,23; p = 0,003), lesão excêntrica (RR 1,86; IC 95% 1,03-3,34; p = 0,039), > 1 SF implantado por território miocárdico (RR 1,81; IC 95% 1,09-3,02; p = 0,023), pósdilatação (RR 0,50; IC 95% 0,29-0,90; p = 0,020) e estenose residual intra-stent (RR 1,04 [por aumento de unidade %]; IC 95% 1,01-1,06; p = 0,003). CONCLUSÕES: Nesse registro prospectivo do "mundo-real", a incidência cumulativa de trombose de stent até cinco anos de seguimento foi rara (1,6%), e foi associada a tabagismo, ICP no IAM, morfologia complexa da lesão, múltiplos stents implantados por território miocárdico e subexpansão do stent.
BACKGROUND: We report the incidence of stent thrombosis (ST) predictors in a large cohort of complex patients treated with drug eluting stents (DES) in the real world. METHODS: From May 2002 until January 2008, 2,365 non-selected patients with > 1 coronary lesion with stenosis of > 50% were treated with DES. The clinical follow-up up to five years (mean time 2.2 ± 1.5 years) was completed in 98.3%. RESULTS: Twenty nine percent of patients had diabetes and 60% multi-vessel disease, 40% presented acute coronary syndrome (15%, acute myocardial infarction [AMI]). The anterior descending artery was the target vessel most frequently treated (35.8%), and 67% were complex lesions (Type B2/C). In all, 3,634 DES were implanted and 40% received multiple stents. Stent thrombosis (classification of the Academic Research Consortium) occurred in 1.6% (n = 38), 60.5% with angiographic confirmation, 42% occurred between 1 and 12 months and 47% of the events had a fatal outcome. The independent predictors of ST were: current smoking [relative risk (RR) 2.59; 95% confidence interval (CI) 1.18-5.67; p = 0.018], AMI intervention (RR 3.50; 95% CI 1.31-9.40; p = 0.013), moderate to severe calcification (RR 2.38; 95% CI 1.34-4.23; p = 0.003), excentric lesion (RR 1.86; 95% CI 1.03-3.34; p = 0.039), > 1 DES implanted per myocardial territory (RR 1.81; 95% CI 1.09-3.02; p = 0.023), post-dilatation (RR 0.50; 95% CI 0.29-0.90; p = 0.020) and intra-stent residual stenosis [RR 1.04 (per % unit increase); 95% CI 1.01-1.06; p = 0.003]. CONCLUSIONS: In this real world prospective registry, the cumulative incidence of stent thrombosis up to 5 year follow-up was rare (1.6%), and associated to smoking, PCI in the AMI, complex lesion morphology, multiple stents implanted per myocardial territory and subexpantion of the stent.

Análise da pele de três espécies de peixes: histologia, morfometria e testes de resistência

O objetivo deste trabalho foi analisar a pele de três espécies de peixes (piavuçu Leporinus macrocephalus, pacu prata Mylossoma sp e piraputanga Brycon hilarii) através de análise histologica e morfometrica e testes de resistência após o curtimento. Amostras de peles da região dorsal foram coletadas, fixadas em Bouin por 24 horas, incluídas em parafina, cortadas em 5 µm de espessura e coradas por Hematoxilina-eosina. Os cortes foram analisados pela microscopia de luz. Foi utilizado o dinamômetro EMIC para a análise físico-mecânica do couro em 10 amostras no sentido longitudinal a partir da região dorsal do peixe de cada espécie. A derme desses peixes possui um padrão estrutural comum aos teleósteos, porém a disposição e distribuição das fibras colágenas apresentaram-se diferentes entre as espécies analisadas. Através da análise morfométrica constatou-se que a espessura da derme da pele diferiu entre as três espécies. Não houve diferença no teste de resistência à tração no couro da piraputanga (16,88 N/mm²) e piavuçu (18,50 N/mm²). Os resultados para os couros destas duas espécies de peixes foram superiores ao pacu (11,83 N/mm²). O valor de alongamento até a ruptura foi superior para piavuçu (52,83%) e piraputanga (60,45%), não diferindo entre eles, porém foi inferior para o pacu (33,83%). O rasgamento progressivo foi maior no couro de pacu (36,51 N/mm). As peles das três espécies analisadas podem ser transformadas em couro para sua aplicação em diversos artefatos.
This study was carried out to analyse the skin of three species of fish (piavuçu Leporinus macrocephalus, pacu prata Mylossoma sp e piraputanga Brycon hilarii) through histological and morphometrical analysis and resistance tests after hardening. Skin samples from the dorsal region were taken and fixed in Bouin solution for 24 hours. They were embedded in paraffin, cut 5 µm thick and stained by Hematoxylin-eosin technique. The cuts were analyzed by light microscopy. A EMIC dynamometer was used for the physical mechanical analysis of the skin in 10 samples, longitudinally oriented from the dorsal region of each specie of fish. The dermis of those fishes has a structural pattern common to teleosts, but the setting and distribution of collagen fibers was different among the analyzed species. The skin dermis thickness differed among the three species by the morphometrical analysis. No difference was observed for the resistance test to traction in piraputanga leather (16.88 N/mm²) as well as in piavuçu leather (18.50 N/mm²). The results for the skin of both fish species were higher than those for pacu (11.83 N/mm²). The value of stretching as to the point of rupture was higher for piavuçu (52.83%) and piraputanga (60.45%), showing no difference among them, but it was lower for pacu (33.83%). The progressive tearing was greater in pacu skin (36.51 N/mm). The skin of the three analyzed species can be turned into leather for the manufacture of several artifacts.

Manifestações respiratórias na malária por Plasmodium falciparum e Vivax: vivax

Foram selecionados 110 pacientes que procuraram a SUCAM/CUIABÁ para diagnóstico e tratamento de malária, com queixa de febre e procedente de área malarígena. Nestes pacientes foram realizados: gota espessa para a pesquisa de Plasmodium; aplicação de questionário padronizado para detectar sintomas respiratórios presentes no momento da entrevista; gasometria arterial e espirometria para avaliar a função pulmonar. Desta amostra, 62 pacientes foram positivos para malária sendo 41 P. vivax, 20 P. falciparum e 01 forma mista (grupo Malárico); 48 mostraram-se negativos à pesquisa de hematozoários através da gota espessa (grupo Não Malárico). As manifestações respiratórias encontradas nesta população quando comparadas os grupos Malárico e Não Malárico, foram respectivamente: 1º) Sintomáticos Respiratórios de 46,8% para 22 9% (p < 0,05); 2°) Alterações Espirométricas de 53,2% para 20,8% (p < 0,05); 3°) Alterações Gasométricas de 62,9% para 25,0% (p < 0,05). Com estes resultados concluímos que há nítida associação entre a Malária produzida pelo P. vivax e P. falciparum e as manifestações respiratórias apresentadas, nesta população de estudo.
A group of 110 patients (101 male, 9 female with mean age ± SD: 31,1 ± 11,0 years) with fever, coming from an endemic malaria region, was studied at SUCAM-CUIABÁ, for diagnosis and treatment of possible protozoal infection. For this group a simplified questionnaire on respiratory symptoms was applied and laboratory tests consisting of peripheral blood film, arterial blool gas analysis and pulmonary function test rwere done. 62 patients had a positive test of malaria (41 P. vivax, 20 P. falciparum 1 F + V) a¡rd 48 were negative, We conclude that there is an association between malarial infection and the presence of respiratory problems.
