DIMENSIONING OF VINYLSULFONIC SUPPORTS FROM CASHEW APPLE BAGASSE BIOMASS IN THE IMMOBILIZATION OF LIPASES

In this work, the support, cashew apple bagasse (CAB), was chemically modified with divinyl sulfone (DVS), and it was evaluated to immobilize Candida antarctica lipase A (CAL-A). The best activation conditions of CAB support were defined by an advanced experimental design using the Taguchi method, assessing five factors at four levels (concentration of DVS, ionic strength, pH, temperature, and time). The support and biocatalyst (CAB-DVS-CAL-A) were characterized by Fourier transform infrared spectroscopy (FTIR), elemental analysis, thermogravimetry (TGA), scanning microscopy (SEM), fluorescence spectroscopy (XRF) and electrophoresis. The optimal conditions to activate the support were DVS concentration of 1.4 mol L-1 (3 mL of DVS in 20 mL of reaction volume), a concentration of sodium bicarbonate buffer at 5 mmol L-1, pH 3.0 at 30 °C for 12 h. The immobilization on CAB-DVS promoted increased thermal stability at 70 °C and different pHs of CAL-A. Therefore, the selected conditions allowed for a catalyst with a catalytic activity of 6.8 U g-1 and more stable than the free enzyme (CAL-A). ABSTRACT In 2000, Cleide Costa published a paper presenting the state of knowledge of the Neotropical Coleopte ra, with a focus on the Brazilian fauna. Twenty-four years later, thanks to the development of the Coleoptera section of the Taxonomic Catalog of the Brazilian Fauna (CTFB - Catálogo Taxonômico da Fauna do Brasil) through the collaboration of 100 coleopterists from all over the globe, we can build on Costa’s work and present an updated overview of the state of knowledge of the beetles from Brazil. There are currently 35,699 species in 4,958 genera and 116 families known to occur in the country, including representatives of all extant suborders and superfamilies. Our data show that the Brazilian beetle fauna is the richest on the planet, concentrating 9% of the world species diversity, with some estimates accounting to up to 15% of the global total. The most diverse family in numbers of genera is Cerambycidae (1,056 genera), while in number of species it is Chrysomelidae (6,079 species). Conotrachelus Dejean, 1835 (Curculionidae) is the most species-rich genus, with 570 species. The French entomologist Maurice Pic is the author who has contributed the most to the naming of species recorded from Brazil, with 1,794 valid names in 36 families, whereas the Brazilians Ubirajara R. Martins and Maria Helena M. Galileo are the only ones among the top-ten authors to have named species in the 21st century. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Objective: Bipolar disorder (BD) is a major cause of disability-adjusted life years in young adults. Pregnancy complications have previously been associated with BD. The current study aimed to examine the association between perinatal factors and BD. Methods: We included 3,794 subjects from the 1993 Pelotas population-based birth cohort study. We assessed 27 variables at birth and modeled BD onset at 18 and 22 years. Bivariate analysis was performed by means of binomial logistic regression models. The variables with p-values less than 0.05 were included in a multiple regression with confounders. Results: Maternal smoking was associated with a 1.42-fold increased risk of BD at 18 or 22 years old (95%CI 1.091-1.841), and maternal passive exposure to tobacco with a 1.43-fold increased risk (95%CI 1.086-1.875). No association was found between other perinatal factors and BD after controlling for confounders. Conclusion: The results of the present cohort study corroborate previous reports in the literature indicating a negative effects of maternal smoking during pregnancy. These findings can be further tested and support the development of strategies to prevent the onset development of BD. ABSTRACT Purpose: To evaluate the modulatory properties of Calendula officinalis L. (Asteraceae) (C. officinalis) extract on cafeteria diet-fed rats. Methods: A cafeteria diet was administered ad libitum for 45 days to induce dyslipidemia. Then, the rats were treated with the formulations containing C. officinalis in the doses of 50, 100, and 150 mg/kg or only with the vehicle formulation; the control group received a commercial ration. Results: The cafeteria diet decreased glutathione S-transferase activity and high-density lipoprotein plasmatic levels and damaged the hepatic architecture. The C. officinalis extract was able to reduce lipid infiltration in liver tissue and to modulate oxidative stress and lipid profile markers. Conclusions: The correlations between the variables suggest a pathological connection between oxidative stress markers and serum lipid profile. Abstract The main factors governing Hevea brasiliensis germination and seedling establishment remains unclear. We examined the effect of growth regulators Indole 3-Acetic Acid (IAA) and 6-Benzylaminopurine (BAP), and their interactions on germination and the development of mature zygotic embryos (MZE) and protein profile of Hevea brasiliensis seedlings from wild and cultivated (clone PB 250) genotypes. Embryonic axes excised from seeds (wild and clone PB 250) were inoculated in Murashige and Skoog medium (control) and supplemented with IAA (3 µM) and BAP (6 µM) individually and their combination (3 µM IAA + 6 µM BAP). For both genotypes, the mature embryos displayed a high percentage of germination and establishment, and the seedlings were characterized by protein bands ranging from 7 to 30 kDa. Notably, the wild genotype showed proteins in the 14 kDa range, which may be associated with one of the major rubber elongation factors (REF). The wild and clone genotypes presented different behavior and strategies in relation to the protein profile in the presence of different growth regulators. Although the latex biosynthetic pathway and its mechanisms of regulation still remain largely unknown, our results aid in our understanding of the dynamics of proteins in different rubber tree clones in vitro.

RESUMO: Clostridioides (Clostridium) difficile é o principal agente envolvido em diarreias associadas ao uso de antimicrobianos em seres humanos e um enteropatógeno de grande relevância em quadros de diarreia em potros e equinos adultos. Em adição, estudos tem sugerido que animais são possíveis reservatórios de estirpes toxigênicas de C. difficile para humanos. Apesar da importância na saúde animal e humana, a epidemiologia da infecção por C. difficile (ICD) é ainda pouco conhecida. Dessa forma, o presente estudo tem como objetivo caracterizar seis casos de diarreia por C. difficile ocorridos em Minas Gerais, Brasil. Com exceção de um animal, todos os equinos incluídos no presente estudo desenvolveram ICD após antibioticoterapia, três dos quais durante a hospitalização. Coinfecção por Salmonella Heidelberg e S. Infantis foi detectada em três casos, tornando o tratamento antimicrobiano desafiador. Todos os animais recuperaram após administração de metronidazol. Os isolados obtidos no presente estudo foram sensíveis a metronidazol e vancomicina, porém três estirpes foram resistentes a moxifloxacina e duas a clindamicina. Os isolados foram classificados como ribotipos 126 (n=4), 078 (n=1) e 014/020 (n=1), todos previamente relatados em seres humanos com ICD no Brasil e em outros países.
ABSTRACT: Clostridioides (Clostridium) difficile is the main causative agent of antimicrobial-related diarrhea in humans and a major pathogen-associated enteric disorder in foals and adult horses. Moreover, studies have suggested that animals are a possible reservoir of toxigenic C. difficile strains for humans. Despite this known importance, the epidemiology of C. difficile infection (CDI) in equine is still largely unknown. Therefore, this study described six cases of equine CDI occurring in Minas Gerais, Brazil, including the characterization of the isolates. All but one equine included in this research developed CDI after antimicrobial therapy, three of which occurred during hospitalization. Coinfection with Salmonella Heidelberg and S. Infantis was detected in three cases, making the antimicrobial treatment challenging. All animals recovered after metronidazole administration. All C. difficile isolates were susceptible to metronidazole and vancomycin, while three were resistant to moxifloxacin and two were resistant to clindamycin. The isolates were classified as RT126 (n = 4), RT078 (n = 1), and RT014/020 (n = 1), all previously reported infecting humans and animals worldwide.

RESUMO: Bacillus toyonensis é um micro-organismo probiótico que há décadas é utilizado na nutrição animal em todo o mundo. O objetivo deste trabalho foi avaliar o efeito imunomodulador da suplementação oral de B. toyonensis em cães vacinados contra o parvovírus canino. Os filhotes foram selecionados aleatoriamente e divididos em dois grupos, um recebeu B. toyonensis na concentração de 2 × 108 esporos viáveis por dia e outro grupo sem suplementação como controle. Os filhotes foram vacinados contra o parvovírus canino tipo 2. A suplementação com B. toyonensis foi eficiente em estimular IgG específica para parvovírus com títulos de 2, 3 e 2,5 vezes maior que os controles aos 7, 21 e 35 dias pós-vacinação, respectivamente. Células mononucleares do sangue periférico (PBMCs) de cães foram cultivadas e estimuladas com DNA de B. toyonensis, células vegetativas e esporos. A transcrição do mRNA das citocinas IL-4, IL-17 e IFN-γ foi modulada pelos estímulos. Assim, concluímos neste estudo que a suplementação com B. toyonensis pode amplificar a resposta imune da vacina contra o parvovírus canino.
ABSTRACT: Bacillus toyonensis is a probiotic microorganism that for decades has been used in animal nutrition around the world. The objective of this work was to evaluate the immunomodulatory effect of oral B. toyonensis supplementation in dogs vaccinated against canine parvovirus. Puppies were randomly selected and divided in two groups, one received B. toyonensis at a concentration of 2x108 viable spores per day and another group without supplementation was left as control. The puppies were vaccinated against canine parvovirus type 2. B. toyonensis supplementation was efficient in stimulating specific IgG for parvovirus with titers of 2, 3, and 2.5-fold higher than controls at 7, 21, and 35 pos-vaccination days respectively. Peripheral blood mononuclear cells (PBMCs) from dogs were cultured and stimulated with B. toyonensis DNA, vegetative cell and spores. The mRNA transcription of cytokines IL-4, IL-17, and IFN-γ up modulated by the stimuli. Thus, we conclude in this study that B. toyonensis supplementation may amplify the vaccine immune response against canine parvovirus.

RESUMO O objetivo desta revisão foi comparar dois modelos de agricultura sustentáveis, demostrando as vantagens e desvantagens de ambos, correlacionando-os entre si. As diversas práticas agrícolas contemporâneas compreendem modelos alternativos de agricultura, como permacultura, agricultura biológica, orgânica, biodinâmica, que são consideradas sustentáveis, e modelos intensivos de produção, que apresentam vários pontos negativos, contrastando com os modelos alternativos. A busca por uma agricultura menos agressiva ao meio ambiente, que permaneça produtiva em longo prazo, tem sido uma preocupação constante de pesquisadores e produtores. Os modelos de agricultura assemelham-se entre si, o que os difere são apenas algumas práticas que não são permitidas em todos, e suas bases de fundamentação. Nesta revisão dois modelos foram mais abordados de forma simplificada, para a percepção dos seus conceitos e de suas bases fundamentais, mostrando o que os diferencia, e que mesmo assim os leva para um mesmo objetivo, criar um ecossistema independente de recursos externos.
ABSTRACT The objective of this review was to compare two models of sustainable agriculture, demonstrating their advantages and disadvantages and correlating them with each other. The diverse contemporary agricultural practices comprise from alternative models of agriculture, such as permaculture, organic farming, organic, biodynamic, that are considered sustainable, and intensive production models, that present several negative points, contrasting with alternative models. The search for a less aggressive agriculture to the environment, what stay productive in the long run, it has been a constant concern of researchers and producers. Agricultural models are similar to each other, what differs are some practices that do not are allowed in all, and its foundation bases. In this review, two models were more approached in a simplified way, to the perception of their concepts and fundamental bases, showing what differentiates them, and that still leads to the same goal, creating an ecosystem independent of external resources.

RESUMO O nitrogênio exerce influência marcante no rendimento e qualidade da cebola. O presente trabalho foi desenvolvido no período de junho a outubro de 2016, tendo como objetivo avaliar a produtividade e qualidade de duas cultivares de cebola sob sistema adensado em função de doses de nitrogênio em Mossoró, RN. Empregou-se o delineamento em blocos casualizados, no esquema fatorial 2 x 7, correspondendo às cultivares IPA 11 e Rio das Antas e sete doses de nitrogênio (0, 45, 90, 135, 180, 225 e 270 kg ha-1 de N), fornecidas via fertigação, com quatro repetições. A dose de 92 kg ha-1 de N proporcionou as maiores produtividades total e comercial de bulbos. A adubação nitrogenada aumentou o teor de N, clorofilas a, b e total nas folhas da cebola, bem como os percentuais de bulbos das classes 3 e 4 e a pungência, reduzindo os percentuais de bulbos da classe 2 e não comerciais. As doses de 108 e 82 kg ha-1 de N proporcionaram as máximas taxas de retorno para ‘IPA 11’ (R$ 2,07) e ‘Rio das Antas’ (R$ 2,41), respectivamente.
ABSTRACT Nitrogen exerts a marked influence on the yield and quality of the onion. The present work was developed from June to October 2016, aiming to evaluate the yield and quality of two cultivars of onion under a system densified as a function of nitrogen doses in Mossoró, RN, Brazil. A randomized block design was used in a 2 x 7 factorial scheme, with four repetitions, corresponding to two cultivars (IPA 11 and Rio das Antas) and seven nitrogen doses (0, 45, 90, 135, 180, 225 and 270 kg ha-1 of N), provided by fertigation, with four repetitions. The dose of 92 kg ha-1 of N provides the highest total and marketable yields of bulbs. Nitrogen fertilization increases the concentrations of N, chlorophyll a, b and total in the leaves of the onion, as well as the percentage of bulbs of classes 3 and 4 and the pungency, reducing the percentage of bulbs of class 2 and non-commercial. The doses of 108 and 82 kg ha-1 of N provide the highest rates of return for ‘IPA 11’ (R$ 2.07) and ‘Rio das Antas’ (R$ 2.41), respectively.

RESUMO Objetivou-se neste trabalho avaliar o desempenho de cultivares de cebola em função do espaçamento entre plantas. O delineamento experimental utilizado foi em blocos casualizados completos em esquema fatorial 3 x 4 com quatro repetições. Os tratamentos foram constituídos da combinação de três cultivares de cebola (Vale Ouro IPA 11, Serena e Rio das Antas) e quatro espaçamentos entre plantas (3, 6, 8 e 10 cm). As características avaliadas foram: produtividades total, comercial e não comercial de bulbos, porcentagens de bulbos das classes 1, 2, 3 e 4, massa média de bulbos, massa seca de bulbos. Nos cultivos de cebola de forma adensados, as cultivares Rio das Antas e Serena são as mais indicadas, com as maiores produtividades comerciais, respectivamente nos espaçamentos de 6 e 3 cm entre plantas. A cultivar Vale Ouro IPA 11 apresentou maior produtividade comercial no espaçamento de 5 cm entre plantas.
ABSTRACT The objective of this study was to evaluate the performance of onion cultivars as a function of the spacing between plants. The experimental design was a complete randomized block in a 3 x 4 factorial scheme, with four replicates. The treatments were composed of the combination between three onion cultivars (Vale Ouro IPA 11, Serena and Rio das Antas) and four plant spacings (3, 6, 8 and 10 cm). The evaluated characteristics were total, marketable and unmarketable yield of bulbs, percentages of bulbs in classes 1, 2, 3 and 4, bulb mean weight, bulb dry weight. In onion cultivation, the cultivars Rio das Antas and Serena are the most indicated, with the highest marketable yields at 6 and 3 cm spacings between plants, respectively. The cultivar Vale Ouro IPA 11 presented higher marketable yield for the spacing of 5 cm between plants.

RESUMO: Apesar da reconhecida importância de Clostridium perfringens como enteropatógeno de pequenos ruminantes, pouco se sabe sobre a frequência dos genótipos ou do papel de fatores de virulência adicionais de C. perfringens em cabras saudáveis, dificultando o diagnóstico laboratorial da infecção causada por esse micro-organismo. Dessa forma, o presente estudo teve como objetivo caracterizar C. perfringens de amostras de fezes de cabras adultas saudáveis. Amostras de fezes de 250 cabras saudáveis de 17 rebanhos diferentes em Minas Gerais, Brasil, foram submetidas ao isolamento e genotipagem de C. perfringens. C. perfringens tipo A foi isolado de 189 (75,6%) cabras, enquanto C. perfringens tipos C e D foram detectados em um animal (0.4%) cada. Todos os isolados foram negativos para os genes codificadores das toxinas NetB, NetE, NetF e enterotoxina. Os resultados apresentados confirmam C. perfringens tipo A como parte da microbiota de cabras saudáveis e sugere que C. perfringens tipos C e D são raramente encontrados em caprinos saudáveis.
ABSTRACT: Despite the known importance of Clostridium perfringens as an enteropathogen in small ruminants, little is known about the role of its additional virulence factors or the frequency of the various C. perfringens genotypes in healthy goats; this complicates the laboratory diagnosis of the infections caused by this microorganism. In light of this, the aim of the present study was to isolate and genotype C. perfringens from stool samples from healthy goats in Brazil. Stool samples from 250 apparently healthy adult goats from 17 different herds in Minas Gerais, Brazil were collected, and isolation and genotyping of C. perfringens was performed. C. perfringens type A was isolated from 189 (75.6%) goats, whereas C. perfringens types C and D were each detected in one goat (0.4%). All isolates were negative for enterotoxin-, NetB-, NetE-, and NetF-encoding genes. These results confirmed C. perfringens type A as part of the microbiota in these animals, and they suggested that C. perfringens type C and D are rarely isolated from healthy goats.

ABSTRACT Phytochemical investigation of Bauhinia acuruana Moric., Fabaceae, resulted in the isolation of sixteen constituents, including two new compounds 2'-hydroxy-2,3,5-trimethoxybibenzyl (1), (2R,3S)-2-(3,4'-dihydroxyphenyl)-5-methoxy-6-methylchroman-3,7-diol (2), together with fourteen known ones (3–16). The structures of the compounds were established by spectroscopic analysis including HR-ESI-MS, 1D and 2D NMR data, followed by comparison with previously reported data from the literature. Compounds 1, 2, 6, 7, 8 and 9 were evaluated for their cytotoxicity, which turned out to be marginal in a panel of six human cancer cell lines.
