This study presents a solution to solid waste problems, focusing on spent lithium-ion batteries (LiBs) and iron ore tailings (IOT) from the Mariana environmental accident in Brazil. The approach involves the production CoFe2O4 from LiBs and IOT, which serves as a catalyst for solar photo-Fenton reactions for methylene blue (MB) decolorization and as an electrochemical sensor for ascorbic acid (AA) detection. Chemical analysis showed recycling potential, with 45.22 ± 0.22% m m−1 Co from LiBs and 14.9 ± 1.5% m m−1 Fe from IOT, determined by inductively coupled plasma atomic emission spectroscopy (ICP OES) and flame atomic absorption spectrometry (FAAS). The sol-gel synthesized CoFe2O4 exhibited a crystallite size of 51.9 ± 1.3 nm and agglomerated crystal clusters. Recycled-CoFe2O4 exhibited a 98.1% MB decolorization efficiency in 60 min under solar irradiation and remained above 92.3% in all 7 reuse cycles. The electrochemical sensor exhibited a coefficient of determination of 0.9987, a sensitivity of 3.352 ± 0.0428 μA mol L−1, and a limit of detection of 0.5511 µM in the concentration range of 1.96 to 23.08 mmol L−1 for AA detection. The antifouling booster biocides are frequently studied for toxic effects on the aquatic ecosystems. The present investigation proposes passive silicone rubber samplers as a collection method for biocides, once these methods can concentrate substances in aqueous matrices at very low levels. Through the passive sampler-water partition coefficient (Ksw) and the analyte chemical nature, we can optimize their extraction from the membrane to apply in the sample medium. We used the co-solvent method to determine the Ksw of three third-generation antifouling biocides, chlorothalonil, dichlofluanid, and dichlorooctylisothiazolinone (DCOIT), with log Ksw = 2.24, 4.01, and 2.38, respectively. Improving extraction also led to a recovery range higher than 70%, determinations were carried out by gas chromatography with an electron capture detector. In last decades, technical-scientific investigations into cultural heritage assets have increasingly become part of analysis methodologies accessible to professionals from different areas for a broad understanding of the complex material and historical nature of objects. In this interdisciplinary word, the painting “Costureiras” by Tarsila do Amaral (1886-1973), from 1950, was studied by ultraviolet-induced visible luminescence (UVL), infrared image (IR) and macro X-ray fluorescence scanning (MA-XRF) techniques. UVL and MA-XRF images enabled the exploration of the conservation status of the artwork, the creative process of the artist, and areas featuring materials distinct from the original painting. By combining the results of IR and MA-XRF images, it was confirmed that changes occurred in the creative process during the composition of the artwork. This study details the preparation, antileishmanial assay, and in silico analysis of twenty 2-hydroxy-3-(1,2,3-triazolyl)propyl vanillin derivatives. These compounds were synthesized in three steps and evaluated against Leishmania infantum, Leishmania amazonensis, and Leishmania braziliensis promastigotes. Compounds 3s and 3t were the most effective, showing good activity against all Leishmania species tested. Molecular docking indicated that all compounds bind favorably to the sterol 14α-demethylase (CYP51) enzyme from L. infantum. ADMET (absorption, distribution, metabolism, excretion and toxicity) analysis indicated good oral bioavailability, non blood-brain barrier penetration, and high gastrointestinal absorption. Posaconazole and compounds 3e, 3s, and 3t remained stable in the CYP51 binding region during 100 ns molecular dynamics (MD) simulations. Root mean square deviation (RMSD) and root mean square fluctuations (RMSF) analyses from the MD trajectory revealed significant conformational fluctuations of the CYP51 N-terminal, suggesting occasional expulsion of 3e, potentially explaining its higher IC50 (half-maximal inhibitory concentration) values. RESUMO Dentre os métodos de controle de plantas daninhas, o químico, por meio de herbicidas, é um dos mais utilizados devido à sua praticidade e eficiência. No entanto, ainda faltam informações confiáveis sobre a eficácia de aplicações em baixo volume e o espectro de gotas a ser utilizado para herbicidas de contato, como o fomesafen. O objetivo deste trabalho foi determinar o volume de calda e espectro de gotas para aplicação do herbicida fomesafen, bem como a eficácia no controle de picão-preto (Bidens pilosa) e corda-de-viola (Ipomoea triloba). O herbicida foi aplicado utilizando pulverizador pressurizado por CO2, com pontas TT11002 espaçadas de 0,50 m, operando nas pressões de 100 kPa (gotas muito grossas) e 400 kPa (gotas médias) nos volumes de calda de 35, 70, 140 e 280 L ha-1, obtidos por meio da variação na velocidade de aplicação. No momento da aplicação foram avaliados a porcentagem de área coberta e a densidade de gotas (gotas cm-²) em etiquetas de papel hidrossensível, utilizando o programa DropScope®. Melhores índices de controle do picão-preto são obtidos quando a aplicação foi realizada com gotas muito grossas, na pressão de 100 kPa, com volume de calda entre 65 e 280 L ha-1, densidade de gotas superior a 60 gotas cm-2 e cobertura maior que 10%. ABSTRACT Among weed control methods, chemical control using herbicides is one of the most widely employed due to its practicality and efficiency. However, there is still a lack of reliable information regarding the effectiveness of low-volume spraying and the droplet spectrum for contact herbicides, such as fomesafen. The objective was to determine the spray volume and droplet spectrum for applying the fomesafen herbicide and its efficacy in controlling hairy beggarticks (Bidens pilosa) and littlebell (Ipomoea triloba). The herbicide was applied using a CO2-pressurized knapsack sprayer with TT11002 spray tip spaced at 0.50 m, operating at pressures of 100 kPa (very coarse droplets) and 400 kPa (medium-sized droplets) with spray volumes of 35, 70, 140, and 280 L ha-1, obtained by varying the application speed. At the time of application, the percentage of covered area and droplet density (droplets cm-²) were evaluated on water-sensitive paper labels using the DropScope® program. RESUMO A soja é uma das mais importantes comodities agrícolas mundiais, no entanto, devido ao intenso processo de melhoramento e sua forma de reprodução, vem perdendo diversidade genética ao longo das últimas décadas. Assim, o objetivo deste estudo foi avaliar o desempenho agronômico, estimar a diversidade genética entre as linhagens e identificar cruzamentos promissores. Para isto, foram avaliados 66 genótipos de soja, sendo 61 linhagens pré-comerciais e cinco cultivares comerciais (AS3797, DESAFIO, M7110, M7739, M8210) como controles. O delineamento experimental foi o de blocos ao acaso com três repetições. A matriz de distância entre as linhagens foi calculada pela distância de Mahalanobis e o agrupamento pelo método da ligação média entre grupos (UPGMA). A distância máxima foi de 1.776,26 entre as linhagens M7110 e G23, e a mínima de 1,50 entre as linhagens G13 e G36, sendo a distância média de 364,40, demonstrando a existência de diversidade genética. Os caracteres que mais contribuíram para a divergência genética foram número de dias para o florescimento (66,7%) e maturação fisiológica (28,55%). As linhagens G60 (maior produtividade de grãos), G51 e G33 (menor maturação fisiológica) e a cultivar M7110 (maior divergência) se destacaram. Formaram-se quatro grupos, sendo o grupo G1 com maior produtividade de grãos e menor número de dias para o florescimento e maturação fisiológica. ABSTRACT Although soybean is one of the most important agricultural commodities in the world, it has been losing genetic diversity in recent decades, due to the intense breeding process and reproduction method. Thus, the objective of this study was to evaluate the agronomic performance and estimate the genetic diversity among inbred lines, and identify promising crossings. A total of 66 soybean genotypes were evaluated, 61 of which were pre-commercial lines, and five commercial cultivars (AS3797, DESAFIO, M7110, M7739, M8210) as controls. The experimental design consisted of randomized blocks with three replicates. The distance matrix between the inbred lines was calculated by Mahalanobis distance and grouping by the unweighted pair group method with arithmetic mean (UPGMA). The maximum distance was 1,776.26 between cultivar M7110 and the G23 inbred line, and the minimum 1.50 between inbred lines G13 and G36, with an average distance of 364.40, demonstrating genetic diversity. The traits that contributed most to genetic divergence were number of days to flowering (66.7%) and physiological maturation (28.55%). Inbred lines G60 (higher grain yield), G51 and G33 (lower physiological maturation) and cultivar M7110 (greater divergence) stood out. Evaluation of miRNAs regulation of BDNF and IGF1 genes in T2DM insulin resistance in experimental models: bioinformatics based approach

Resumo Os microRNAs (miRNAs) são reconhecidos como biomarcadores do diabetes mellitus tipo 2 (DM2), úteis para a compreensão do metabolismo da doença, e possuem grande potencial como alvos terapêuticos. O aumento da expressão de BDNF e IGF1 está altamente envolvido nos benefícios as vias de insulina e glicose, porém, são regulados negativamente em condições de resistência à insulina, enquanto seu aumento de expressão está correlacionado com a melhora do metabolismo da glicose e da insulina. Estudos sugerem a regulação desses genes por microRNA em vários contextos diferentes, proporcionando uma nova abordagem de investigação para compreender o metabolismo do DM2 e revelar potenciais alvos terapêuticos. No presente estudo, investigamos em diferentes modelos animais (humanos, ratos e camundongos) miRNAs que têm como alvo BDNF e IGF1 em tecido muscular esquelético com condições fisiológicas de DM2. As análises foram realizadas utilizando ferramentas de bioinformática e bancos de dados para predição de miRNA, homologia molecular, validação experimental de interações, expressão na condição fisiológica estudada e interação em rede. Os resultados mostraram três candidatos a miRNAs para IGF1 (miR-29a, miR-29b e miR-29c) e um para BDNF (miR-206). As avaliações experimentais e a busca pela expressão no músculo esquelético de indivíduos com DM2 confirmaram a interação prevista entre miRNA-mRNA para miR-29b e miR-206 através de modelos humanos, ratos e camundongos. Essa interação foi reafirmada em múltiplas análises de rede. Em conclusão, nossos resultados mostram a relação de regulação entre miR-29b e miR-206 com os genes investigados, em diversos tecidos, sugerindo um padrão de inibição. Contudo, esses dados mostram um grande número de possíveis processos fisiológicos de interação para perspectivas biotecnológicas.
Abstract microRNAs (miRNAs) are recognized as diabetes mellitus type 2 (T2DM) biomarkers useful for disease metabolism comprehension and have great potential as therapeutics targets. BDNF and IGF1 increased expression are highly involved in the benefits of insulin and glucose paths, however, they are down-regulated in insulin resistance conditions, while their expression increase is correlated to the improvement of glucose and insulin metabolism. Studies suggest the microRNA regulation of these genes in several different contexts, providing a novel investigation approach for comprehending T2DM metabolism and revealing potential therapeutic targets. In the present study, we investigate in different animal models (human, rat, and mouse) miRNAs that target BDNF and IGF1 in skeletal muscle tissue with T2DM physiological conditions. Bioinformatics tools and databases were used to miRNA prediction, molecular homology, experimental validation of interactions, expression in the studied physiological condition, and network interaction. The findings showed three miRNAs candidates for IGF1(miR-29a, miR-29b, and miR-29c) and one for BDNF (miR-206). The experimental evaluations and the search for the expression in skeletal muscle from T2DM subjects confirmed the predicted interaction between miRNA-mRNA for miR-29b and miR-206 through human, rat, and mouse models. This interaction was reaffirmed in multiple network analyses. In conclusion, our results show the regulation relationship between miR-29b and miR-206 with the investigated genes, in several tissues, suggesting an inhibition pattern. Nevertheless, these data show a large number of possible interaction physiological processes, for future biotechnological prospects.

This work aims to optimize the process of extracting pigments from the peel of Amazonian fruits (bacupari, peach palm, and tucumã) using the enzyme pectinase. First, pigment extraction was carried out using different solvents (ethanol and acetone 80%, ethanol and acetone 50%, and water), temperatures (44, 40, 35, and 26 °C), and wavelengths (400, 450, and 490 nm). In the second stage, the best parameters found in the first stage were applied in a statistic with a response surface varying time (60, 120, and 180 min) and enzyme concentration (40, 80, and 120 U). The best results for extraction for bacupari and peach palm were acetone at 50% and 45 °C and ethanol at 50% and 35 °C for tucumã. In the second stage, the optimum point found was 1.5 h and 100 U concentration, and 2.5 h and 90 U concentration resulted in better extractions for bacupari, peach palm, and tucumã. Resumo A utilização de biomassa para a produção de energia representa uma estratégia promissora, justificando a busca por novas fontes de biomassa. O capim-elefante tem ganhado destaque devido ao seu elevado rendimento de matéria seca e rápido crescimento. Este estudo teve como objetivo quantificar a divergência genética entre novas famílias de meio-irmãos de capim-elefante, com a finalidade de identificar genótipos com maior divergência genética e potencial produtivo para hibridização. Utilizou-se uma metodologia de agrupamento hierárquico baseada em componentes principais. As famílias de meio-irmãos foram obtidas a partir de genótipos do Banco Ativo de Germoplasma de Capim-Elefante. O experimento foi conduzido em delineamento de blocos casualizados, com nove famílias de meio-irmãos, três repetições e oito plantas por parcela, totalizando a avaliação de 216 genótipos de capim-elefante. Foram realizadas análises de componentes principais (PC), biplot e agrupamento hierárquico para estimar a diversidade, utilizando o software R. Os dois primeiros PCs da análise biplot explicaram 64% da variação total. A produtividade de matéria seca foi a característica mais relevante para a discriminação genotípica (0,89), seguida pela altura da planta (0,67) e pelo diâmetro do caule (0,61) no PC1. Abstract The use of biomass for energy production constitutes a promising strategy that warrants the search for new sources of biomass. Elephant grass has been gaining notoriety due to its high dry matter yield and rapid growth. The present study was carried out to quantify the genetic divergence of nine elephant grass half-sib families in order to identify genotypes with greater genetic divergence and productive potential for hybridization, using the hierarchical clustering methodology based on principal components. Half-sib families were generated using genotypes from the Active Germplasm Bank of Elephant Grass. The experiment was laid out in a randomized-block design with nine half-sib families, three replicates, and eight plants per plot. A total of 216 genotypes of elephant grass were evaluated. Principal component (PC), biplot, and hierarchical clustering analyses for diversity estimation were conducted using R software. The first two PCs of biplot analysis accounted for 64% of the cumulative variation. Dry matter yield was the most important trait for genotype discrimination (0.89), followed by plant height (0.67) and stem diameter (0.61) in PC1. In this analysis, the distances between accessions were considered and there were no family links, which indicates the existence of wide variability within the evaluated families, since genotypes belonging to the same family were not grouped together, but rather distributed into different groups. In this study, a mixed oxide (SiO2/ZnO/Fe2O3/ZnFe2O4) was prepared using the anode recycled from alkaline batteries and the silt fraction of mining tailings and applied as a photocatalyst under solar irradiation and as a pseudocapacitor. The structure and morphology of the mixed oxide revealed clusters of nanometric particles with rhombic and octahedral shapes. A 23 full factorial design was performed to determine the effect of pH, hydrogen peroxide (H2O2), and photocatalyst on methylene blue (MB) decolorization. The maximum efficiency was 96%, obtained using 2.35 × 10-5 mol H2O2 and 20 mg of photocatalyst at pH 3 for 30 min under solar irradiation. This efficiency remained even after four successive decolorization cycles. Pseudocapacitive properties showed that the mixed oxide exhibited favorable characteristics for application as electrochemical pseudocapacitors due to its high capacitance (87.16 F g-1), energy density (27.24 Wh kg-1) at 524.41 W kg-1 of power density, and reversibility (93%). ABSTRACT The objective of this study was to evaluate the effects of prebiotics, alone and in combination with antibiotics, on broiler performance, carcass yield, intestinal permeability, and intestinal morphometry. A total of 1440 day-old male Cobb 500 chicks were distributed in a completely randomized design, with four treatments and 10 replications per treatment. The treatments used were a basal diet without prebiotics and antibiotics (BD), a basal diet with antibiotics (AB), a basal diet with prebiotics (PRE), and a basal diet with antibiotics and prebiotics (AB+PRE). To perform the treatment, the antibiotic zinc bacitracin and a prebiotic of S. cerevisiae-derived mannan-oligosaccharides and beta-glucans were used. The treatments did not affect the performance of birds from 1 to 7 and 1 to 21 days of age. From 1 to 35 and 1 to 42 days, birds fed the AB+PRE treatment had higher feed intake than those fed the AB treatment. From 1 to 35 days, birds fed the AB+PRE treatment showed a worse feed conversion ratio. However, there was no difference in carcass yield between treatments. A higher number of goblet cells was observed in the duodenum and ileum of birds fed AB+PRE, but lower counts were obtained in birds fed AB. Intestinal histomorphometry displayed similar responses between both ages regarding antibiotic and prebiotic treatments, except in the ileum at 42 days, in which the antibiotic elicited a better response. Intestinal permeability analysis showed no significant difference between diets. Abstract Pulmonary arterial hypertension (PAH) is characterized by right ventricular failure and diminished cardiac output, potentially leading to renal and bone impairments. In contrast, resistance exercise training (RT) offers cardiovascular and bone health benefits. This study aimed to assess the impacts of stable PAH induced by monocrotaline (MCT) and RT on renal morphometry, as well as bone morphometry and biomechanical properties in male Wistar rats. Four experimental groups, untrained control (UC, n=7), trained control (TC, n=7), untrained hypertensive (UH, n=7), trained hypertensive (TH, n=7), were defined. After the first MCT or saline injection (20 mg/kg), trained rats were submitted to a RT program (i.e., Ladder climbing), 5 times/week. Seven days later the rats received the second MCT or saline dose. After euthanasia, renal and femoral histomorphometry and femoral biomechanical properties were assessed. PAH reduced renal glomerular area and volume, which was prevented by the RT. While PAH did not harm the femoral morphometry, structural and mechanical properties, RT improved the femoral parameters (e.g., length, percentage of trabeculae and bone marrow, ultimte and yield loads). Experimental stable PAH promotes renal but not bone damages, whereas RT prevents renal deteriorations and improves the femoral morphological and biomechanical properties. Abstract Birds kept in captivity are subject to various diseases that affect their well-being, with parasites being among the most common. The aim of this study was to report the occurrence of endoparasites in the feces of birds kept in captivity at Fortaleza Zoo, using the Willis Mollay and Hoffman techniques. Of the samples analyzed, 50.98% were positive for endoparasites, including the suborder Coccidia, the genera Capillaria spp., Strongyloides spp., Ascaridia spp., Libyostrongylus spp., cysts of a specific parasite of Ostriches, Balantidium spp., the Strongyloidea superfamily and cestodes. Resumo O feijão-caupi é uma planta leguminosa pertencente à família das Fabaceae cultivada nas regiões Norte e Nordeste do Brasil, com potencial produtivo. Entre os fatores abióticos, a deficiência hídrica é uma das principais limitações ambientais que influenciam a produção agrícola no mundo. O objetivo do trabalho foi estudar o conteúdo relativo de água e os osmorreguladores de plantas de feijão-caupi submetidas ao estresse hídrico. O experimento foi conduzido em casa de vegetação da Universidade Federal Rural da Amazônia (UFRA, Belém, PA), foram utilizadas plantas de feijão-caupi BR-17 Gurguéia Vigna unguiculata (L.) Walp. O delineamento experimental foi inteiramente casualizado (DIC) em esquema fatorial 2 × 2, duas condições hídricas (controle e déficit hídrico) e dois tempos de estresse (quatro e seis dias de suspensão hídrica), com 7 repetições, totalizando 28 unidades experimentais. O estresse hídrico afetou as plantas, provocando redução no conteúdo relativos de água (69,98%), amido (12,84% nas folhas e 23,48% nas raízes) e carboidratos (84,34%), e aumento na glicina-betaína, sacarose (114,11% nas folhas e 18,71% nas raízes) e prolina (358,86%) no tempo 2. O conteúdo relativo de água foi afetado negativamente pelas condições hídricas, com diminuição em relação à interação da parte aérea e do sistema radicular. Abstract Cowpea is a leguminous plant belonging to the fabaceae family cultivated in the North and Northeast regions of Brazil, with productive potential. Among the abiotic factors, water deficiency is one of the main environmental limitations that influence agricultural production in the world. The objective of this work was to study the relative water content and osmoregulators of cowpea plants subjected to water stress. The experiment was carried out in a greenhouse at the Universidade Federal Rural da Amazônia (UFRA, Belém, PA), cowpea plants BR-17 Gurguéia Vigna unguiculata (L.) Walp were used. The experimental design was completely randomized (DIC) in a 2 × 2 factorial scheme, two water conditions (control and water deficit) and two times of stress (four and six days of water suspension), with 7 replications, totaling 28 experimental units. The water deficit affected plants, causing a reduction in relative water content (69.98%), starch (12.84% in leaves and 23.48% in roots) and carbohydrates (84.34%), and an increase in glycine-betaine, sucrose (114.11% in leaves and 18.71% in roots) and proline (358.86%) at time 2. The relative water content was negatively affected by water conditions, with a decrease in relation to the interaction of the aerial part and the root system. Therefore, greater metabolic responses were noted in plants that were subjected to stress treatment at time 2 (6 days). ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals.
