Abstract Background: Elderly patients are more susceptible to Coronavirus Disease-2019 (COVID-19) and are more likely to develop it in severe forms, (e.g., Acute Respiratory Distress Syndrome [ARDS]). Prone positioning is a treatment strategy for severe ARDS; however, its response in the elderly population remains poorly understood. The main objective was to evaluate the predictive response and mortality of elderly patients exposed to prone positioning due to ARDS-COVID-19. Methods: This retrospective multicenter cohort study involved 223 patients aged ≥ 65 years, who received prone position sessions for severe ARDS due to COVID-19, using invasive mechanical ventilation. The PaO2/FiO2 ratio was used to assess the oxygenation response. The 20-point improvement in PaO2/FiO2 after the first prone session was considered for good response. Data were collected from electronic medical records, including demographic data, laboratory/image exams, complications, comorbidities, SAPS III and SOFA scores, use of anticoagulants and vasopressors, ventilator settings, and respiratory system mechanics. Mortality was defined as deaths that occurred until hospital discharge. Results: Most patients were male, with arterial hypertension and diabetes mellitus as the most prevalent comorbidities. The non-responders group had higher SAPS III and SOFA scores, and a higher incidence of complications. There was no difference in mortality rate. A lower SAPS III score was a predictor of oxygenation response, and the male sex was a risk predictor of mortality. Conclusion: The present study suggests the oxygenation response to prone positioning in elderly patients with severe COVID-19-ARDS correlates with the SAPS III score. Furthermore, the male sex is a risk predictor of mortality.

Abstract Centrosema coriaceum Benth belongs to Fabaceae family and have few studies of biological activity and chemical composition. Thus, the aims of this work were to determine chemical profile of the ethanolic extract of C. coriaceum leaves (CCE) by UFLC-QTOF-MS and to evaluate its in vitro biological potential. CCE showed MIC value of 1000 µg/mL against Candida glabrata (fungistatic effect) and high affinity in cell envelope by increasing cell permeability in nucleotide leakage, sorbitol and ergosterol assays. CCE showed antioxidant activity in all assays performed. For the anti-inflammatory and cytotoxicity activities, CCE, at all tested concentrations, significantly inhibited the production of nitric oxide and did not decrease J774A.1 cell viability below 70%. Finally, rutin, kaempferol-3O-rutinoside, caffeic acid, and sucrose were identified in CCE by UFLC-QTOF-MS. These results suggest, for the first time, that C. coriaceum has interesting antifungal, antioxidant, and anti-inflammatory activities.

Abstract The water retention curve is widely used in studies involving soil. The management systems directly influence soil structure by altering water retention dynamics. Several equations are used to adjust the retention of water in the soil, but most of the time, the choice of these models occurs in an arbitrary way. From this problem, it was proposed with the present study to relate the best mathematical model to water retention, taking into account the different management systems adopted, based on previously established adjustment criteria. For the accomplishment the study, a soil of caulinitic mineralogy and average texture was utilized. The treatments were area under native Cerrado (Savanna), eucalyptus plantation with six and twelve years of cultivation, pasture with two and six years of cultivation, conventional plantation with two and eight years of cultivation, no-till with three and six years of cultivation. From the adjustment criteria for non-linear models, it was found that the management influences the choice of the water retention model. It is possible to observe that the greatest divergences between the models occurred close to the soil saturation zone, and that the Fredlung-Xing model is more efficient in adjusting the water retention curve under conservationist management systems.

Abstract In canine visceral leishmaniasis, coinfections can aggravate the disease. Our aim was to investigate Brucella canis in dogs infected with Leishmania infantum. One hundred and six L. infantum-seropositive dogs were submitted to serology for B. canis, PCR for B. canis and L. infantum, and histopathological analysis of the genital tract. Anti-B. canis antibodies were detected in seven dogs whose clinical signs, L. infantum load and histological alterations were similar to those of seronegative animals. The circulation of anti-B. canis antibodies was low but demonstrates the exposure of dogs to this bacterium in a visceral leishmaniasis-endemic area.

Abstract In this work, a compact ultra-wide-band microstrip antenna loaded with C-shaped parasitic elements, for Digital Video Broadcasting - Terrestrial (DVB-T) and LTE 450 / 700 signal reception, is designed and presented. The proposed antenna is named “HI Monopole”, and its geometry has been designed to be simple, lightweight, and compact. Fabrication process is inexpensive and suitable for electronic gadgets. The HI Monopole operates with ultra-wide bandwidth characteristics, encompassing Digital Video Broadcasting - Terrestrial (DVB-T) standard (470-890 MHz), LTE-450 and LTE-700 bands. This feature has been accomplished by inserting an indent in its ground plane, adjusting dimensions of the feeding line, and adding two C-shaped parasite elements onto the region posterior to the antenna's main radiating element. It was verified via electromagnetic measuring that this device is capable of covering the 429-1022 MHz range. Hence, its total bandwidth is 593 MHz. The superior and inferior cutoff frequencies have been defined using the -10 dB return loss reference level. Therefore, the proposed antenna can be employed for receiving DTV and LTE signals broadcasted in any country, independently of its adopted standard.

ABSTRACT Chemical investigation of the aqueous fraction of the ethanol extract from the Brazilian endemic marine sponge Clathria (Clathria) nicoleae Vieira de Barros, Santos & Pinheiro, 2013, Microcionidae, sampled from a 55 m deep rhodolith bed at the Amazon River mouth, led to the isolation of a new hexapeptide, clathriamide (1). HP-20 resin was used to capture compound 1 from the aqueous fraction, which was purified by additional chromatographic steps. The absolute configuration of the amino acids of 1 was determined by advanced Marfey's analysis using 5-fluoro-2,4-dinitrophenyl-Nα-L-tryptophanamide. The amino acid derivatives analyzed by ultra-performance liquid chromatography coupled to a mass spectrometry using a C8 column enabled a good chromatographic resolution of L-Ile and L-allo-Ile, previously unfeasible using C18 column.

Abstract Silicone breast implant is associated with complications inherent to the surgical procedure. Prosthesis coating with polyurethane, however, commonly reduces the incidence of such complications. In this paper, the authors evaluated the inflammatory histomorphometric profile and oxidative damage associated to the implant of polyester urethane sheets. Forty-eight Wistar rats were divided into Sham or polyester urethane groups (n = 8/group) and underwent a polyester urethane implant in the dorsal skinfold. Tissue samples were collected on days seven, 30, and 90 after surgery and subjected to histomorphometric analysis and biochemical tests. Results were analyzed by one-way ANOVA (p ≤ 0.05). Peri-implant tissue samples exhibited characteristic inflammatory response associated with the biomaterial, with increased vascularization on day seven and augmented levels of IL1-b and TNF-a after 30 days. Peri-implant fibrocystic population was small on day seven, but increased considerably after 90 days. A rise in the carbonyl group levels of skin samples in the polyester urethane group was observed on day seven. Findings suggest that polyester urethane sheets undergo biodegradation at an early stage after implantation, followed by increased vascularity and microencapsulation of biomaterial fragments, without persistent oxidative damage. Fiber arrangement inside the collagen matrix results in a fibrotic scar because of polyester urethane degradation.

RESUMO: O estudo objetivou verificar a ocorrência de Leishmania spp. e Leishmania (Leishmania) infantum em cavalos de uma região endêmica para leishmaniose visceral do Brasil. Amostras de DNA de sangue e suabe conjuntival (SC) foram testadas pela PCR e pela Reação de Imunofluorescência Indireta (RIFI). Embora nenhum cavalo estivesse clinicamente doente, animais infectados por Leishmania spp. e L. (L.) infantum foram encontrados em Ilha Solteira/SP. Dos 40 cavalos, 100% (40/40) foram positivos pela PCR de sangue, 90% (36/40) pela PCR de SC, e 2,5% (01/40) no sorodiagnóstico, pela RIFI. Seis desses 40 cavalos foram positivos para L. (L.) infantum pela PCR de sangue. O sequenciamento direto e a análise dos amplicons resultaram em uma sequência para análise evolutiva. Os resultados indicam a presença de Leishmania spp. e L. (L.) infantum infectando cavalos saudáveis no Brasil. A presença de DNA de Leishmania spp. and L. (L.) infantum em cavalos saudáveis sugere que eles podem ser importantes reservatórios desses parasitas, um achado altamente relevante para a vigilância epidemiológica das doenças que causam.
ABSTRACT: This study aimed to verify the occurrence of Leishmania spp. and Leishmania (Leishmania) infantum in horses from a visceral leishmaniasis endemic area in Brazil. DNA samples from blood and conjunctival swab (CS) were tested by PCR and Indirect Immunofluorescence Antibody Test (IFAT). Although none of the horses was clinically sick, animals infected by Leishmania spp. were found and some could be characterized as infected by L. (L.) infantum. From 40 horses, 100% of the animals were positive by blood PCR, 90% (36/40) by CS PCR, and 2.5% (01/40) in serodiagnosis, by IFAT. Six from these 40 horses were L. (L.) infantum positive by blood PCR. Direct sequencing and analysis of amplicons resulted in a sequence to evolutionary analysis. Results indicate the presence of Leishmania spp. and L. (L.) infantum infecting healthy horses in Brazil. The presence of Leishmania spp. and L. (L.) infantum DNA in asymptomatic horses suggests that they can be important reservoirs of these parasites, a highly relevant finding for the epidemiological surveillance of the diseases they cause.

Support vector machine-based classification of neuroimages in Alzheimer’s disease: direct comparison of FDG-PET, rCBF-SPECT and MRI data acquired from the same individuals

Objective: To conduct the first support vector machine (SVM)-based study comparing the diagnostic accuracy of T1-weighted magnetic resonance imaging (T1-MRI), F-fluorodeoxyglucose-positron emission tomography (FDG-PET) and regional cerebral blood flow single-photon emission computed tomography (rCBF-SPECT) in Alzheimer’s disease (AD). Method: Brain T1-MRI, FDG-PET and rCBF-SPECT scans were acquired from a sample of mild AD patients (n=20) and healthy elderly controls (n=18). SVM-based diagnostic accuracy indices were calculated using whole-brain information and leave-one-out cross-validation. Results: The accuracy obtained using PET and SPECT data were similar. PET accuracy was 68∼71% and area under curve (AUC) 0.77∼0.81; SPECT accuracy was 68∼74% and AUC 0.75∼0.79, and both had better performance than analysis with T1-MRI data (accuracy of 58%, AUC 0.67). The addition of PET or SPECT to MRI produced higher accuracy indices (68∼74%; AUC: 0.74∼0.82) than T1-MRI alone, but these were not clearly superior to the isolated neurofunctional modalities. Conclusion: In line with previous evidence, FDG-PET and rCBF-SPECT more accurately identified patients with AD than T1-MRI, and the addition of either PET or SPECT to T1-MRI data yielded increased accuracy. The comparable SPECT and PET performances, directly demonstrated for the first time in the present study, support the view that rCBF-SPECT still has a role to play in AD diagnosis.

Global Postural Reeducation for patients with musculoskeletal conditions: a systematic review of randomized controlled trials

ABSTRACT Objectives To systematically review randomized controlled trials that assessed the effects of Global Postural Reeducation (GPR) on patient-reported outcomes in conditions of the musculoskeletal system. Method An electronic search of MEDLINE (via PubMed), EMBASE, Cochrane CENTRAL, and SciELO was performed from their inception to June 2015. Randomized controlled trials that analyzed pain and patient-reported outcomes were included in this review. The Cochrane Collaboration’s Risk of Bias Tool was used to evaluate risk of bias, and the quality of evidence was rated following the GRADE approach. There were no language restrictions. Results Eleven trials were included totaling 383 patients. Overall, the trials had high risk of bias. GPR was superior to no treatment but not to other forms of treatment for pain and disability. No placebo-controlled trials were found. Conclusion GPR is not superior to other treatments; however, it is superior to no treatment. Due to the lack of studies, it is unknown if GPR is better than placebo. The quality of the available evidence ranges from low to very low, therefore future studies may change the effect estimates of GPR in musculoskeletal conditions.

AbstractEvening primrose (Oenothera biennis L., Onagraceae) seeds oil has great economic importance due to its wide industrial application, mainly for medicines and nutraceutics. However, to our knowledge, it remains almost unexplored regarding development of innovative formulations, such as nanoemulsions. On the present study, required Hydroprophile–Lipophile Balance of evening primrose seeds oil was determined (HLB 12) and a stable nanoemulsion (Day 1: mean droplet size: 214.3 ± 0.69 nm, polydispersity index: 0.253 ± 0.012. Day 7: mean droplet size: 202.8 ± 0.23 nm, polydispersity index: 0.231 ± 0.008) was achieved. Moreover, pseudo-ternary diagram allowed delimitation of nanoemulsion region, contributing to nanobiotechnology of natural products.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

INTRODUÇÃO: A eficácia e a segurança do stent farmacológico (SF) de segunda geração liberador de everolimus XienceTM V (Abbott Vascular, Santa Clara, Estados Unidos) já foram estabelecidas no tratamento de pacientes selecionados com lesões coronárias. No entanto, o impacto do stent XienceTM V em populações da prática clínica com lesões complexas ainda não está totalmente definido. MÉTODOS: O Registro BRAVO foi um estudo prospectivo, não-randomizado, multicêntrico, que avaliou os resultados clínicos tardios de pacientes minimamente selecionados tratados com o SF XienceTM V na prática diária brasileira. No total, foram incluídos 535 pacientes em 25 centros clínicos entre setembro de 2008 e setembro de 2010. Eventos cardíacos adversos maiores (ECAM) foram definidos como morte cardíaca, infarto agudo do miocárdio (IAM) e revascularização do vaso-alvo (RVA). RESULTADOS: A média de idade dos pacientes era de 62,7 ± 11,1 anos, 40% dos quais tinham diabetes, 24,9% apresentavam IAM prévio e 41,9% apresentaram-se com síndrome coronária aguda. Cerca de dois terços dos pacientes tinham lesões tipo B2/C e 46,1% trataram a artéria descendente anterior. Implante de múltiplos stents ocorreu em 13,8% dos casos, e o sucesso angiográfico foi > 99%. Na fase intra-hospitalar, a taxa de IAM periprocedimento foi de 1,9%. Já no seguimento de 6 meses, as taxas cumulativas de óbito cardíaco, IAM e RVA foram de 1,1%, 2,2% e 1,3%, respectivamente (taxa de ECAM de 4,3%). Com relação à trombose de stent (definida de acordo com os critérios do Academic Research Consortium ARC), foram reportados 4 casos até 6 meses, representando taxa de evento de 0,75% (0,4% definitiva/provável). CONCLUSÕES: Neste Registro, que incluiu pacientes e lesões complexos tratados em múltiplos centros nacionais, o SF de segunda geração XienceTM V demonstrou excelentes resultados imediatos e efetividade clínica e segurança sustentadas até o seguimento a médio prazo (6 meses). Os resultados do seguimento a longo prazo são aguardados.
BACKGROUND: The XienceTM V everolimus-eluting stent (Abbott Vascular, Santa Clara, USA), a second-generation drug-eluting stent (DES) has demonstrated sustained efficacy and safety in the treatment of selected patients with coronary lesions. However, the impact of the XienceTM V stent in populations from daily clinical practice with complex lesions has not yet been fully determined. METHODS: The BRAVO Registry was a prospective, non-randomized, multicenter study that evaluated the late clinical outcomes of minimally selected patients treated with XienceTM V DES in the Brazilian daily clinical practice. Overall, 535 patients were included in 25 clinical sites between September/2008 and September/2010. Major adverse cardiac events (MACE) were defined as cardiac death, acute myocardial infarction (AMI) and target vessel revascularization (TVR). RESULTS: Mean age was 62.7 + 11.1 years, 40% had diabetes, 24.9% had a previous AMI and 41.9% presented with acute coronary syndrome. About two thirds of the patients had type B2/C lesions and 46.1% treated the left anterior descending artery. Multiple stenting procedures were performed in 13.8% of cases and angiographic success was > 99%. During hospitalization, periprocedural AMI rate was 1.9%. At the 6-month follow-up, cumulative rates of cardiac death, AMI and TVR were 1.1%, 2.2% and 1.3%, respectively (MACE rate: 4.3%). There were 4 cases of stent thrombosis (defined according to the Academic Research Consortium ARC) reported within 6 months, representing an event rate of 0.75% (0.4% definite/probable). CONCLUSIONS: In this Registry including complex patients and lesions treated at multiple sites in Brazil, the XienceTM V second generation DES demonstrated excellent immediate results and sustained clinical efficacy and safety at mid-term follow-up (6 months). Long-term results are expected.

Este trabalho descreve a construção e caracterização de uma célula eletroquímica microfluídica com a inserção dos três eletrodos (trabalho, pseudo-referência e auxiliar) em microcanais com espessuras menores que 20 µm. Os microcanais foram construídos entre duas bases de policarbonato com o uso de uma ou mais máscaras de toner sobrepostas como espaçador. Esta estratégia permite a construção de microcélulas com um volume interno sobre o eletrodo de trabalho entre 0,6 a 2,4 µL. Três diferentes materiais foram otimizados como eletrodos nas microcélulas: filmes de ouro ou compósito de grafite como eletrodo de trabalho, compósito de prata como pseudo-referência e compósito de grafite como eletrodo auxiliar. O desempenho das células microfluídicas foi avaliado usando as técnicas de voltametria cíclica, redissolução potenciométrica a corrente constante e voltametria de redissolução anódica por onda quadrada empregando ferrocianeto e alguns metais pesados (Cu2+, Hg2+, Pb2+, e Cd2+) como analitos modelo.
This work describes the construction and characterization of an electrochemical flow-through microcell with the three electrodes (working, pseudo-reference, and auxiliary) inserted in microchannels with thickness smaller than 20 µm. These microchannels were constructed between two stacked polycarbonate slides using one or more overlapped toner masks as spacer. This strategy allows the construction of microcells with a variable internal volume on the working electrode (0.6 to 2.4 µL). Three different materials were optimized as electrodes: gold film or graphite-epoxy composite as working electrode, silver-epoxy composite as pseudo-reference electrode and, graphite-epoxy composite as auxiliary electrode. The performance of the microfluidic cell was characterized by cyclic voltammetry, potentiometric stripping analysis at constant current, and square wave anodic stripping voltammetry using ferrocyanide and heavy metals (Cu2+, Hg2+, Pb2+, and Cd2+) as model analytes.
