Abstract Temperature affects the rate of biochemical and physiological processes in amphibians, influencing metamorphic traits. Temperature patterns, as those observed in latitudinal and altitudinal clines, may impose different challenges on amphibians depending on how species are geographically distributed. Moreover, species’ response to environmental temperatures may also be phylogenetically constrained. Here, we explore the effects of acclimation to higher temperatures on tadpole survival, development, and growth, using a meta-analytical approach. We also evaluate whether the latitude and climatic variables at each collection site can explain differences in species’ response to increasing temperature and whether these responses are phylogenetically conserved. Our results show that species that develop at relatively higher temperatures reach metamorphosis faster. Furthermore, absolute latitude at each collection site may partially explain heterogeneity in larval growth rate. Phylogenetic signal of traits in response to temperature indicates a non-random process in which related species resemble each other less than expected under Brownian motion evolution (BM) in all traits, except survival. Abstract The disease coronavirus COVID-19 has been the cause of millions of deaths worldwide. Among the proteins of SARS-CoV-2, non-structural protein 12 (NSP12) plays a key role during COVID infection and is part of the RNA-dependent RNA polymerase complex. The monitoring of NSP12 polymorphisms is extremely important for the design of new antiviral drugs and monitoring of viral evolution. This study analyzed the NSP12 mutations detected in circulating SARS-CoV-2 during the years 2020 to 2022 in the population of the city of Manaus, Amazonas, Brazil. The most frequent mutations found were P323L and G671S. Reports in the literature indicate that these mutations are related to transmissibility efficiency, which may have contributed to the extremely high numbers of cases in this location. In addition, two mutations described here (E796D and R914K) are close and have RMSD that is similar to the mutations M794V and N911K, which have been described in the literature as influential on the performance of the NSP12 enzyme. These data demonstrate the need to monitor the emergence of new mutations in NSP12 in order to better understand their consequences for the treatments currently used and in the design of new drugs. Drug repositioning involves the use of a determined drug in a different indication and has been widely used for some therapeutic classes. Artificial intelligence, in turn, has been a trend in the modern world of innovation, including in drug design. Those approaches are apparently paradoxical (the former is not considered as properly an innovative method to introduce new molecules/drugs into the market, although the latter is) can be used as complementary. In this review, we present some concepts of both methods, their advantages, possible disadvantages, and some applications in drug design in general to improve different aspects of drug development. Examples of the use of both methods together have been given for many therapeutic classes. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. RESUMO: As aves e os produtos de origem aviária são fontes de contaminação predominantes de Salmonella enterica e importantes reservatórios de bactérias com resistência antimicrobiana. Objetivou-se identificar Salmonella com fenótipos de multirresistência a drogas (MDR), com a capacidade de formação de biofilmes e a presença de genes que codificam resistência antimicrobiana em isolados da cadeia de frangos de corte, do estado Maranhão, Brasil. Avaliaram-se 121 cepas de S. enterica de sorovares diferentes quanto ao teste de suscetibilidade aos antimicrobianos e destas, 26 cepas para detecção da capacidade de formar biofilmes e genes de resistência pela técnica de PCR. Foram encontradas resistência antimicrobiana em 95 (78,5%) dos isolados de Salmonella e 57 (47,1%) apresentaram fenótipos MDR. Os isolados apresentaram maior resistência aos princípios sulfonamida (58,7%), trimetoprim (48,8%), tetraciclina (45,4%), ácido nalidíxico (44,6%), amoxicilina e ampicilina (26,4%) e cefazolin (22,3%). Os sorovares Salmonella Schwarzengrund (n=21/61.7%), Albany (n=15/62.5%) e Enteritidis (n=4/44.5%) apresentaram os maiores índices de fenótipos MDR. A capacidade de formar biofilmes foi encontrada em 13 cepas avaliadas, consideradas fracamente produtoras. Nos sorovares S. Schwarzengrund, Albany, Enteritidis, Heidelberg e Typhimurium foram detectados os genes de resistência blaCTX-M, blaCTX-M2, blaSHV, sul1, sul2, tetA, tetB, tetC, tetE, dfrA12 e dfrA1. Os resultados evidenciaram a elevada ocorrência de fenótipos de S. enterica com resistência múltipla a antimicrobianos convencionais, com capacidade de formar biofilmes, na cadeia produtiva de aves destinadas ao consumo humano. ABSTRACT: Poultry and poultry products are considered the predominant sources of Salmonella enterica contamination and are important reservoirs of bacteria with antimicrobial resistance. The objective of this study was to identify Salmonella with multidrug resistance (MDR) phenotype, with the ability to form biofilms and elucidate the presence of genes that encode antimicrobial resistance in isolates from the broiler production chain in the state of Maranhão, Brazil. A total of 121 strains of S. enterica of different serovars were evaluated for antimicrobial susceptibility, and of these, 26 strains were used to detect the ability to form biofilms and identify resistance genes using PCR. Antimicrobial resistance was observed in 95 (78.5%) Salmonella isolates, and 57 (47.1%) showed MDR phenotype. The isolates showed greater resistance to the sulfonamide principles (58.7%), trimethoprim (48.8%), tetracycline (45.4%), nalidixic acid (44.6%), amoxicillin and ampicillin (26.4%), and cefazolin (22.3%). Salmonella Schwarzengrund (n=21/61.7%), Albany (n=15/62.5%), and Enteritidis (n=4/44.5%) showed the highest indices of MDR phenotype. The ability to form biofilms at 37°C was found in 13 of the 26 strains evaluated, which were considered poor producers. The resistance genes blaCTX-M, blaCTX-M2, blaSHV, sul1, sul2, tetA, tetB, tetC, tetE, dfrA12, and dfrA1 were observed in the serovars Schwarzengrund, Albany, Enteritidis, Heidelberg, and Typhimurium. The results showed a high occurrence of S. enterica, with multiple resistance to conventional antimicrobials and the ability to form biofilms in the poultry production chain. Resumo Barragens são umas das principais ameaças para peixes potamódromos Neotropicais, causando bloqueios de rotas migratórias e perdas de áreas de reprodução em um número crescente de sistemas de drenagem. Neste cenário, dados atuais defendem que a manutenção de tributários de fluxo livre em bacias hidrográficas fragmentadas seria uma importante estratégia na conservação de peixes migradores, tanto em situações de panmixia quanto em cenários com diferentes estoques coexistindo, tal como já relatado para Prochilodus lineatus. Este estudo analisou locos microssatélites e haplótipos mitocondriais em amostras de P. lineatus obtidas em diferentes anos ao longo da bacia do rio das Cinzas, no reservatório de Capivara (maior reservatório na calha principal do rio Paranapanema, alto rio Paraná) e no rio Tibagi, todos conectados por trechos livres, visando investigar as contribuições da bacia do rio das Cinzas na dinâmica populacional de peixes migradores na região do reservatório de Capivara. Ambos os marcadores detectaram altos níveis de diversidade genética e indicaram diferentes estoques de P. lineatus ao longo da bacia do rio das Cinzas, sugerindo alguma estruturação genética temporal, bem como a mistura destes estoques na amostra do reservatório de Capivara e de outros pontos, corroborando as relações de estoques neste reservatório e seus tributários. Abstract Dams are one of the main threats to Neotropical potamodromous fish, causing blockages of migratory routes and loss of breeding areas in an increasing number of drainage systems. In this scenario, current data argue that the maintenance of free-flowing tributaries along fragmented watersheds would be an important strategy to the migratory fish conservation, both in panmixia situations and in scenarios with different coexisting stocks, as already reported for Prochilodus lineatus. This study analyzed microsatellite loci and mitochondrial haplotypes in samples of P. lineatus obtained in different years along the Cinzas River basin, as well as in the Capivara Reservoir (largest Reservoir in the Paranapanema main channel, upper Paraná River) and in the Tibagi River, all connected by free stretches, aiming to investigate the contributions of the Cinzas River basin to the population dynamics of migratory fish in the Capivara reservoir region. The cariogenic processes are mainly caused by the bacterium Streptococcus mutans (S. mutans) and consist of the demineralization of the tooth that occurs when the acid production overcomes the natural repair or if a problem occurs in the last one. In this work, we performed the synthesis of twenty-one 1,4-naphthoquinones tethered to 1,2,3-1H-triazoles (8a-8k and 9a-9j), antibacterial evaluation against the S. mutans in vitro and the acute toxicity of the better ones in vivo. We observed strong inhibition results in the disc diffusion test ranging, the halos of inhibitions, from 18.66 (± 0.57) to 29 (± 2.64) mm, and good values in the minimum inhibitory concentration (5 to 50 μg), for the compounds 9e, 9h, 9i and 9j: Furthermore, they do not have a cytotoxic effect at the concentrations tested. Besides that, in the in vivo test, they show some slight alteration in the histopathological analyses and the biochemistry. Thus, we found four potential candidates to become instruments for the treatment of cavities.

ABSTRACT Background: Neotropical species lack autecological information. Among these, are phenological studies, which help to understand forest dynamics and provide important contributions to the collection, restoration, and conservation programs. This research aimed to characterize the vegetative and reproductive phenological behavior of neotropical species Geoffroea spinosa Jacq. in a seasonally dry tropical forest, as well as to verify if there is a correlation between phenophases and environmental parameters. Twenty individuals were accompanied monthly for two years, to detect and tally the presence of floral buds, flowers, immature fruits, ripe fruits, leaf emergence, presence of mature leaves, and defoliation. A semi-quantitative evaluation was carried out with the Fournier method. Results: The results showed that the population had synchrony of reproductive events, with more than 50% of the individuals blooming and fruiting during the same time interval. Furthermore, the circular statistical test indicated a high seasonality for reproductive events (r ≥ 0.50 and p < 0.001). Among the vegetative events, all were classified as non-seasonal (r < 0.5). G. spinosa, showed a significant correlation (p ≤ 0.05) for all meteorological variables evaluated. Conclusion: The reproductive phenophases of G. spinosa showed strongly seasonal behavior, unlike vegetative events. the period between mid-February and the end of March was the most propitious for fruit collection. No seasonality was observed for the vegetative events. The species proved to be sensitive to El niño conditions. Follow-up phenological studies associated with meteorological variables are important for a better understanding of the dynamics of vegetative behavior in response to climate change.

RESUMO Este estudo transversal foi realizado por meio de um questionário online. O questionário foi aplicado a endodontistas e coletou informagoes sobre modificagoes práticas durante o tratamento endodontico para combater o surto de COVID-19 e proteger profissionais e pacientes. Um total de 1105 participantes do Brasil participaram da pesquisa. Mais de 90% dos entrevistados identificam o alto risco de infecgao por COVID-19 para os dentistas e a necessidade de mudar algumas práticas clínicas. A maioria dos entrevistados (60,1%) segue parcialmente o isolamento social. A necessidade de mudangas dos Equipamentos de Protegao Individual (EPIs) durante as consultas odontológicas foi referida por 97,1% dos entrevistados. O uso de EPIs mínimos adequados durante o período pandémico foi associado á área de residencia e ao estado civil dos participantes. Apenas 30% dos participantes afirmou usar os EPIs mínimos durante a pandemia. A maioria dos entrevistados mudará a preparagao do acesso á cavidade para reduzir a disseminagao do virus. Outras mudangas nas consultas endodonticas foram descritas na pesquisa: maior atengao ás medidas de biosseguranga, duragao das consultas odontológicas e intervalos entre as consultas. Os endodontistas ainda precisam distinguir a melhor maneira para realizar seus procedimentos com seguranga, durante a pandemia de COVID-19. Diretrizes específicas requerem informagoes detalhadas para cada especialidade e seus procedimentos.
ABSTRACT This study evaluated the impact of COVID-19 on the endodontic treatment routine. It was a cross-sectional study using an online questionnaire applied to endodontists to collect information about practical modifications during endodontic treatment to protect professionals and patients against the COVID-19 outbreak. A total 1105 participants from Brazil participated in the survey. More than 90% of respondents identify the high risk of COVID-19 infection to dentists and the need to change some clinical practices. Most respondents (60.1%) are partially following social isolation. The need for a change in Personal Protective Equipment (PPE) during dental appointments was mentioned by 97.1% of respondents. The use of minimal adequate PPE during the pandemic period was associated with the area of residence and marital status of participants. Only 30% of respondents say they use the minimal adequate PPE. Most respondents will change cavity access preparation to reduce virus dissemination. Other changes in endodontic appointments were described in the survey: greater attention to biosafety measures, duration of dental appointments, and duration of intervals between appointments. Endodontists still need to identify the best arrangement for performing their procedures safely during the COVID-19 pandemic. Specific guidelines require detailed information for each specialty and its procedures.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

Abstract Candida spp. is considered an important cause of healthcare-associated infections worldwide. Currently, the emergence and spread of resistant Candida isolates are being increasingly reported, making the development of new agents urgently needed. In this study, we showed the in vitro anti-Candida activity of seven synthetic 3-alkylpyridine alkaloid analogs. Among them, alkaloid 1 presented a potent antifungal effect, which was independent of its capacity of binding with the fungal membrane ergosterol or cell wall. Analog 1 showed fungistatic and fungicidal effects against C. albicans (MIC 7.8 μg/mL and MFC 62.5 μg/mL), C. glabrata, C. krusei (MIC and MFC 31.2 μg/mL), and C. tropicalis (MIC 31.2 μg/mL and MFC 125 μg/mL). The time kill-curve study showed that compound 1 has a potent fungicidal effect in vitro, eliminating C. albicans cells. Furthermore, an in vitro synergistic effect with ketoconazole was observed for compound 1. This compound also eliminated the yeast-to-hypha transition. However, it showed high cytotoxicity against mammalian cells. Taken together, these findings support the use of compound 1 as a prototype to develop new anti-Candida agents, but molecular modifications should be done to minimize the high cytotoxicity obtained.

In the present work, leaves of Pentacalia desiderabilis (Asteraceae) were subjected to extraction using aqueous 1-butyl-3-methylimidazolium bromide (BMImBr) in the microwave assisted extraction (MAE). The obtained extract was partitioned using CH2Cl2 and antifungal activity against Cladosporium cladosporioides and C. sphaerospermum of the organic phase was evaluated. Sequentially, this material was subjected to several chromatographic procedures to afford four quinoid derivatives identified as jacaranone (1), methyl-jacaranone (2), quinolacetic acid (3), and methyl 1-hydroxy-4-oxo-cyclohexaneacetate (4), being the first occurrence of compounds 2 - 4 in P. desiderabilis. Isolated quinoid derivatives were submitted to bioautography assay and the obtained results indicated that compounds 1 - 3 are active against Cladosporium cladosporioides (0.78, 12.5 and 12.5 µg, respectively) and C. sphaerospermum (3.13, 12.5 and 12.5 µg, respectively).

RESUMO Brycon nattereri é um peixe Neotropical ameaçado de extinção reportado para trechos conservados dos rios Paraná, Tocantins e São Francisco. Populações desta espécie têm sido muito raras em algumas sub-bacias do rio Paraná. Este estudo analisou a diversidade genética e a estrutura populacional de B. nattereri em uma área de ocorrência restrita recentemente identificada na bacia do alto rio Paraná. Sete locos microssatélites e 497 pb da região mitocondrial D-Loop foram examinados para 92 indivíduos de quatro pontos ao longo da área de ocorrência. Ambos os marcadores moleculares indicaram uma única população distribuída em um trecho de aproximadamente 80 km do rio. Embora alguns dados tenham sugerido um antigo gargalo genético, os atuais níveis de diversidade genética (H E = 0,574, h = 0,616) foram similares aos de outras espécies do gênero Brycon. Estes resultados sugerem que a população de B. nattereri tem mantido níveis satisfatórios de diversidade genética, apesar da pequena área de ocorrência. Estes dados destacaram uma importante área de conservação e ações podem melhorar a qualidade do ambiente, especialmente para a vida aquática e mata ciliar, se a área for eficientemente manejada e conservada.
ABSTRACT Brycon nattereri is an endangered Neotropical fish reported along conserved stretches of the upper Paraná, Tocantins and São Francisco rivers. Populations of this species have been very rare in some Paraná River sub basins. This study analyzes the genetic diversity and population structure of B. nattereri in a restricted area of occurrence recently identified in upper Paraná River basin. Seven microsatellite loci and 497 bp of D-Loop mitochondrial region were examined in 92 individuals from four points along the area of occurrence. Both molecular markers indicated a single population distributed along a stretch of the river approximately 80 km long. Although some of the data suggest an ancient bottleneck, current levels of genetic diversity (H E = 0.574 and h = 0.616) were similar to those of other species of the genus Brycon. The results suggest that the population of B. nattereri has been able to maintain satisfactory levels of genetic diversity, in spite of the small area of occurrence. These data have highlighted an important conservation area and action may prove essential to improve the quality of the environment, and especially the water and riparian plant life, if the area is to be managed and conserved efficiently.

Chronic migraine patients show cognitive impairment in an extended neuropsychological assessment

RESUMO O objetivo do presente estudo foi avaliar a presença de déficits cognitivos em pacientes com migrânea crônica e avaliar os principais fatores que desencadeiam transtornos cognitivos, como comorbidades e uso de medicações. Métodos: Pacientes com migrânea crônica (n = 30) e controles foram entrevistados, em um estudo caso-controle. A frequência e intensidade da cefaleia, medicações utilizadas e comorbidades associadas foram determinadas. Todos os pacientes foram submetidos a uma avaliação neuropsicológica estendida. Resultados: Os pacientes com migrânea crônica apresentaram uma pior performance no Montreal Cognitive Assessment (p = 0.00), Fluência Verbal (p = 0.00), Teste de Stroop (p = 0.00), Teste do Relógio (p = 0.00), Subteste Dígitos (p = 0.00) e Raciocínio matricial da WAIS-III (p = 0.01). Após ajuste estatístico através de Regressão Linear, a migrânea se manteve como único fator relevante para pior desempenho em MoCA, Fluência verbal, Teste do relógio e Teste de Stroop. Conclusão: Pacientes com migrânea crônica apresentam déficits cognitivos incluindo múltiplas tarefas, independentes da presença de comorbidades e uso de medicações.
ABSTRACT The objective of the present study was to assess the presence of cognitive deficits in patients with chronic migraine, and to assess the main factors that trigger cognitive disorders, such as comorbidities or the use of medications. Methods: Chronic migraine and control groups were interviewed in a case-control study. The frequency and intensity of the headache, medication used and associated comorbidities were determined. All patients were submitted to an extended neuropsychological assessment. Results: The chronic migraine group (n = 30) had a worse performance in the Montreal Cognitive Assessment Test (p = 0.00), Verbal Fluency (p = 0.00), Stroop (p = 0.00), Clock Drawing Test (p = 0.00), Digit Span (p = 0.00) and Matrix Reasoning (p = 0.01). After statistical adjustment by linear regression, migraine continued to be the only relevant factor in the poorer performance in the Montreal Cognitive Assessment, Verbal Fluency, Clock Drawing and Stroop tests. Conclusion: Patients with chronic migraine have cognitive deficits in multiple tasks, regardless of the presence of comorbidities or the use of medications.

Resumo O trabalho teve como objetivo determinar os fatores associados à contaminação de vegetais por protozoários zoonóticos. Amostras de água, solo e vegetais foram coletadas de julho/2014 a maio/2016, totalizando 83 amostras de 21 propriedades da região de Londrina, Paraná, Brasil. A amplificação de fragmentos de DNA de T. gondii, Cryptosporidium spp. e Giardia intestinalis foi realizada por meio da reação em cadeia da polimerase (PCR). Os resultados da PCR foram positivos para T. gondii em 12,9% (8/62), Cryptosporidium spp. em 11,3% (7/62) e G. intestinalis. em 25,8% (16/62) das amostras. O sequenciamento de DNA identificou C. parvum em cinco amostras e G. intestinalis, Assemblage E em três amostras. As associações estatísticas evidenciaram maior probabilidade de amostras serem positivas para T. gondii ou para pelo menos um dos três protozoários quando a fonte de água de irrigação era o rio; uma maior chance de amostras positivas para Cryptosporidium spp. quando havia cervos na propriedade; e uma menor chance das amostras serem positivas para pelo menos um dos três agentes etiológicos quando o solo era suplementado com calcário. Os resultados expõem alguns pontos críticos de contaminação, fornecendo suporte para capacitar os agricultores em boas práticas de gestão durante o processo de produção.
Abstract The objective of this study was to determine factors associated with vegetable contamination with zoonotic protozoan. Samples of water, soil and vegetables were collected from July/2014 to May/2016, totaling 83 samples, 21 properties of Londrina region, Paraná, Brazil. DNA amplification of Toxoplasma gondii, Cryptosporidium spp. and Giardia intestinalis in the samples was conducted using polymerase chain reaction (PCR). The PCR results were positive for T. gondii in 12.9% (8/62), Cryptosporidium spp. in 11.3% (7/62) and G. intestinalis in 25.8% (16/62) of the samples. DNA sequencing identified C. parvum in five samples and G. intestinalis Assemblage E in three. The statistical associations demonstrated greater probability of positive samples for T. gondii and for at least one of the three protozoa when the source of irrigation water was the river; a greater chance of positive samples for Cryptosporidium spp. when deer were present on the property; and a smaller chance of positive samples for at least one of the three etiologic agents when soil was supplemented with limestone. The results expose some critical contamination points, providing support for training farmers on good management practices during the production process.
