This work evaluated the use of corncob as a support for the immobilization of an extracellular fructosyltransferase (E.C. 2.4.1.9) from Aspergillus oryzae IPT-301, aiming at the synthesis of a stable biocatalyst for the production of fructooligosaccharides. The transfructosylation activity of the immobilized enzyme was maximized via experimental design. Additionally, the biochemical properties and stability of the immobilized enzyme, as well as the reaction kinetics, were determined. The enzyme immobilized on alkali-treated corncob showed the highest transfructosylation activity at 50 °C and pH 5.5. It also presented a wide pH stability and a half-life around 1.4 times greater than the soluble enzyme. Furthermore, the immobilized enzyme showed 53% of retention of catalytic activity in a second consecutive reaction cycle, showing the potential for reuse. These results suggest that alkali-treated corncob is a promising support material for the synthesis of heterogeneous biocatalysts aiming at fructooligosaccharide production, which can contribute to the valorization of this abundant agro-waste. Microparticulated whey protein can be used as a fat substitute in foods to enhance their nutritional value and improve their sensory and rheological characteristics. Microparticulation denatures whey proteins through heat treatment, followed by shear rate control to determine the aggregate sizes formed. The processing method used during microparticulation can modify the protein denaturation rate, which may enable the application of new proteins. This study sought to develop a methodology for quantifying whey protein fractions in commercial samples before and after microparticulation at different concentrations and physical states. The developed methodology proved to be applicable because of its selectivity, quantification, detection limits, and precision. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. RESUMO Análises hematológicas e bioquímicas possuem grande importância na avaliação da saúde das populações e de seus habitats e, por estarem sujeitas a influências de diversos fatores, tornam-se necessários conhecer os valores próprios de cada espécie. Neste estudo objetivou-se determinar os parâmetros hematológicos e bioquímicos, assim como a análise morfométrica das células sanguíneas de catetos criados em cativeiro no semiárido do nordeste brasileiro. Utilizou-se 30 animais adultos divididos em 2 grupos (15 fêmeas e 15 machos), com peso médio de 20kg e idade entre 2 e 2.5 anos. Foram coletadas amostras de sangue e soro, no período da manhã, para realizar as análises hematológicas, bioquímica sérica e morfometria celular. Os dados foram analisados estatisticamente quanto à normalidade, pelo teste de Shapiro-Wilk, e homocedasticidade, por Levene. Quando a diferença estatística entre sexos os dados foram paramétricos, utilizando o teste t para medidas independentes e Mann-Whitney quando não paramétricos. Os resultados mostraram que o sexo não possui influência na maioria das variáveis estudadas, excetuando as análises bioquímicas cálcio, proteínas totais e ureia e a contagem relativa dos eosinófilos. ABSTRACT Hematological and biochemical analyses are paramount in assessing the health of different populations and their habitats, and knowledge on the specific values of each species is necessary, due to influence of several factors. In this context, this study aimed to determine the hematological and biochemical parameters of collared peccaries raised in captivity in the semi-arid Brazilian northeast associated to morphometric blood cell analyses. A total of 30 adult animals were investigated, divided into two groups (15 females and 15 males), averaging 20 kg and between 2 and 2.5 years old. Blood and serum samples were collected in the morning for hematological, serum biochemistry and cellular morphometry analyses. Data normality was determined by the Shapiro-Wilk test, and homoscedasticity, by Levene’s test. Concerning statistical differences between sexes, parametric data were analyzed by the t test for independent measurements and non-parametric data, by the Mann-Whitney test. Sex did not influence most of the studied variables, except for calcium, total proteins and urea and relative eosinophil counts. RESUMO Composta por parasitoides e predadores, a família Dryinidae é a terceira maior família de Chrysidoidea, com 1.924 espécies descritas no mundo. A diversidade da família pode ser ainda maior, entretanto, estudos sobre a diversidade e distribuição geográfica são escassos, especialmente em regiões semiáridas. O objetivo deste trabalho foi determinar a ocorrência e a diversidade de gêneros de Dryinidae no Semiárido da Bahia. Foram utilizadas cinco armadilhas do tipo Malaise distribuídas em duas áreas de vegetação nativa e uma constituída por eucalipto (Eucalyptus urophylla), com monitoramento mensal por dois anos consecutivos. Os indivíduos foram identificados a nível de gênero e morfoespécies. Foram coletados 77 indivíduos de Dryinidae, distribuídos em quatro subfamílias (Anteoninae, Aphelopinae, Dryininae, Gonatopodinae), contidas em sete gêneros (Anteon Jurine, 1807; Aphelopus Dalman, 1823; Crovettia Olmi, 1984; Deinodryinus, Perkins, 1907; Dryinus Latreille, 1804; Gonatopus Ljungh, 1810 e Thaumatodryinus Perkins, 1905), e 29 morfoespécies. O gênero Dryinus foi o mais frequente nas três áreas de estudo. As áreas nativas (principalmente na área florestal) apresentaram maior diversidade do que a área de eucalipto. ABSTRACT Dryinidae is a family of parasitoids and predators; it is the third largest family in the superfamily Chrysidoidea, with 1,924 species worldwide. The diversity of this family may be even greater, but studies on its diversity and geographical distribution are scarce, especially in semiarid regions. The objective of this study was to describe the occurrence and diversity of Dryinidae genera in the Semiarid region of Bahia, Brazil. Five Malaise traps were distributed in two areas with native vegetation and in one with eucalyptus (Eucalyptus urophylla) plantation and monitored monthly for two years. Specimens were identified to genus and morphospecies levels. The collection consisted of 77 Dryinidae specimens from four subfamilies (Anteoninae, Aphelopinae, Dryininae, Gonatopodinae), seven genera (Anteon Jurine, 1807; Aphelopus Dalman, 1823; Crovettia Olmi, 1984; Deinodryinus, Perkins, 1907; Dryinus Latreille, 1804; Gonatopus Ljungh, 1810 and Thaumatodryinus Perkins, 1905), and 29 morphospecies. Abstract The present study aimed to evaluate the influence of increasing levels of corn straw replacement of sorghum silage on average daily gain, loin eye area, subcutaneous fat thickness, weight of commercial cuts, leg tissue composition, and physico-chemical characteristics of the meat from F1 Santa Inês × Dorper lambs maintained in a feedlot system. Treatments consisted of 0, 33, 66 and 100% corn straw replacement of sorghum silage. There were thirty-six 150-day-old male lambs. Corn straw replacement of sorghum silage in lamb feed reduced the average daily gain and weight of the ribs, besides promoting a quadratic influence on leg fat content. However, there was no influence on commercial carcass cuts, leg muscle index, leg tissue composition, and physico-chemical characteristics of the meat. Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. This project contributes to the sustainable development of the SB, in line with the guidelines of the United Nations Decade of Ocean Science for Sustainable Development. ABSTRACT Several drugs are available for the treatment of osteoporosis in postmenopausal women. Over the last decades, most patients requiring pharmacological intervention were offered antiresorptive drugs as first-line therapy, while anabolic agents were considered a last resource for those with therapeutic failure. However, recent randomized trials in patients with severe osteoporosis have shown that anabolic agents reduce fractures to a greater extent than antiresorptive medications. Additionally, evidence indicates that increases in bone mineral density (BMD) are maximized when patients are treated with anabolic agents first, followed by antiresorptive therapy. This evidence is key, considering that greater increases in BMD during osteoporosis treatment are associated with a more pronounced reduction in fracture risk. Thus, international guidelines have recently proposed an individualized approach to osteoporosis treatment based on fracture risk stratification, in which the stratification risk has been refined to include a category of patients at very high risk of fracture who should be managed with anabolic agents as first-line therapy. In this document, the Brazilian Society of Endocrinology and Metabolism and the Brazilian Association of Bone Assessment and Metabolism propose the definition of very high risk of osteoporotic fracture in postmenopausal women, for whom anabolic agents should be considered as first-line therapy. This document also reviews the factors associated with increased fracture risk, trials comparing anabolic versus antiresorptive agents, efficacy of anabolic agents in patients who are treatment naïve versus those previously treated with antiresorptive agents, and safety of anabolic agents.

Abstract Cholesterol is a lipid molecule of great biological importance to animal cells. Dysregulation of cholesterol metabolism leads to raised blood total cholesterol levels, a clinical condition called hypercholesterolemia. Evidence has shown that hypercholesterolemia is associated with the development of liver and heart disease. One of the mechanisms underlying heart and liver alterations induced by hypercholesterolemia is oxidative stress. In this regard, in several experimental studies, gold nanoparticles (AuNP) displayed antioxidant properties. We hypothesized that hypercholesterolemia causes redox system imbalance in the liver and cardiac tissues, and AuNP treatment could ameliorate it. Young adult male Swiss mice fed a regular rodent diet or a high cholesterol diet for eight weeks and concomitantly treated with AuNP (2.5 μg/kg) or vehicle by oral gavage. Hypercholesterolemia increased the nitrite concentration and glutathione (GSH) levels and decreased the liver’s superoxide dismutase (SOD) activity. Also, hypercholesterolemia significantly enhanced the reactive oxygen species (ROS) and GSH levels in cardiac tissue. Notably, AuNP promoted the redox system homeostasis, increasing the SOD activity in hepatic tissue and reducing ROS levels in cardiac tissue. Overall, our data showed that hypercholesterolemia triggered oxidative stress in mice’s liver and heart, which was partially prevented by AuNP treatment.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

Seed germination and dormancy break in Eragrostis polytricha, a native Brazilian grass species with potential for recovery of degraded lands

ABSTRACT Ferruginous Rocky Outcrops have high levels of species richness and endemism, but have been threatened by several anthropic actions, especially mining. Eragrostis polytricha, a common grass species in the vegetation of these outcrops, has shown promising features for use in the recovery of mining areas. However, in order to fully understand the species’ potential for such use, its requirements for germination, seed dormancy break and seedling development must be determined. Thus, we aimed to: (1) assess the temperature conditions needed for seeds of E. polytricha to germinate; (2) evaluate the effects of KNO₃ in breaking seed dormancy; and (3) analyze the germination efficiency of seeds that are still in spikelets. The experiment included seven treatments: 15-35 ºC with KNO₃, 20-30 ºC with KNO₃, 15 ºC with KNO₃, 25 ºC with KNO₃, 35 ºC with KNO₃, 20-30 ºC with water, and 20-30 ºC with KNO₃ using spikelets. The treatments with alternating temperatures associated with KNO3 yielded the highest germination rates, suggesting that these two factors combined can break seed dormancy. Seeds inside spikelets exhibited a high germination percentage, and thus represent an interesting alternative for seedling production.

The benzophenones are synthetic and natural compounds presenting a variety of activities, including photoprotective, cytotoxic and antiviral. It is herein described the preparation of a series of twenty-seven benzophenone derivatives bearing 1,2,3-triazole functionalities and the evaluation of their photoprotective, cytotoxic and antiviral on Zika Virus (ZIKV) activities. The compounds were prepared in three steps, namely reduction of benzophenone, alkylation of diphenylmethanol and CuAAC reactions. The in vitro evaluation of the photoprotective activity revealed that the most active derivative 4-((benzhydryloxy)methyl)-1-(4-nitrobenzyl)-1H-1,2,3-triazole (4k) displayed UVB sun protection factor equal to 6,9±0,53, which make this compound a possible candidate to be used in formulations for photoprotective applications. In terms of cytotoxicity, the compounds were evaluated against MDA-MB-231 and B16F10 cell lines. It was observed that the compounds were more active against MDA-MB-231 cells and three of them were capable of reducing cell viability by approximately 55% at 100 µmol L-1. In the antiviral screening against ZIKV, compound 4-(3-benzhydryloxy)propyl)-1-(3-methylbenzyl)-1H-1,2,3-triazole (5j) was the most effective in mantaining Vero cell viability.

Abstract Rheumatic heart disease (RHD) remains a major cause of preventable death and disability in children and young adults. Despite significant advances in medical technology and increased understanding of disease mechanisms, RHD continues to be a serious public health problem throughout the world, especially in low- and middle-income countries. Echocardiographic screening has played a key role in improving the accuracy of diagnosing RHD and has highlighted the disease burden. Most affected patients present with severe valve disease and limited access to life-saving cardiac surgery or percutaneous valve intervention, contributing to increased mortality and other complications. Although understanding of disease pathogenesis has advanced in recent years, key questions remain to be addressed. Preventing or providing early treatment for streptococcal infections is the most important step in reducing the burden of this disease.

Straightforward, microscale, Ellman's cholinesterase test procedures are presented, useful for screening of novel compounds aiming at neurodegenerative diseases and antidotes towards nerve agents and pesticides. Herein, we detail the different procedure features and set forth guidelines on experimental choices researchers may make. The detailed procedure lowers the costs for Ellman´s test, what is highly desirable for low-budget groups in science-lagging and developing countries.

Lupeol, a triterpene frequently found in Asteraceae plant species, showed moderate to low activity in different strains of Plasmodium falciparum, the most virulent malaria etiological agents. In this work, lupeol was isolated from Parahancornia fasciculata, a plant that is used to treat malaria in the Amazonia region. In the search of more activity lupeol derivatives, five new 1,2,3-triazole hybrid molecules were synthetized by copper-catalyzed azide-alkyne cycloaddition. The antiplasmodial activity of the semi-synthetic compounds were evaluated by the lactate dehydrogenase assay; the lupeol propargyl ether was the only one to disclosing increased activity (half maximal inhibitory concentration-IC50-62.0 ± 1.92 µmol L-1) in relation to lupeol (IC50 117.00 µmol L-1). Therefore, this work revealed a new class of interesting lupeol derivatives that can be obtained by linking electron donors to the hydroxy group at C-3.
