Lipid Composition and Cytokines IL-10 and TGF-β1 Evaluation of Prebiotic Ice Cream Containing Human Milk

Human milk (HM) is recognized as an ideal nutritional source to meet the developmental needs of babies. However, a significant portion does not meet quality standards for immediate consumption, rendering it an interesting raw material for the production of by-products. This study aimed to evaluate the lipid composition and cytokines interleukin-10 (IL-10) and transforming growth factor-beta 1 (TGF-β1) in ice cream containing HM. Three formulations were developed: F1 with 93.46% HM, F2 with 46.73% cows’ milk and 46.73% HM, and F3 with 46.73% infant formula and 46.73% HM. Fatty acids (FA) and their nutritional quality, the triacylglycerol (TAG) profile, and the concentrations of cytokines IL-10 and TGF-β1 were analyzed. F1 and F3 exhibited the highest concentrations of essential FAs (18:2n-6 and 18:3n-3) compared to F2. This trend extended to eicosapentaenoic acid + docosahexaenoic acid, with higher values in F1 (10.62 ± 0.26) and F3 (11.16 ± 0.28) versus F2 (3.67 ± 0.18). Both cytokines IL-10 and TGF-β1, present in the ice creams, experienced a decrease in concentration due to the processing of the raw materials. Therefore, ice creams produced with HM have promising potential, making them recommended for use, providing a new destination for waste, and creating a novel product that can be used as a complementary food.

Reuse made by the separation of whey can reduce the waste of human milk. However, the authors are not aware of the literature on treatments in human whey, made available by pasteurization holder, freeze-drying, spray drying, and high hydrostatic pressure. In this context, the present study applied treatments to human whey and evaluated their lipid and immunological quality. Among the results, a different formation in the triacylglycerol profile was evidenced after the application of spray drying and high hydrostatic pressure, while pasteurization and freeze-drying presented ion intensity close to the control human whey. In addition, pasteurization proved to be adequate for lipid nutritional quality and transformation factor-β1 (TGF-β1) concentration, while an increase in interleukin-10 (IL-10) levels was promoted, between 73 and 80%, after freeze-drying, spray drying, and high hydrostatic pressure. Through the principal component analysis, it is noteworthy that the processes presented divergences in terms of the effects caused, with similarity only between pasteurization and freeze-drying in the composition of fatty acids. However, it was observed that all processes were able to maintain the nutrients. Nevertheless, it is relevant to consider individual characteristics presented and the interest in the desired quality, which can be promising as a complementary product to infant feeding.

ABSTRACT A great challenge to overcome is how to maintain and increase the productivity of vegetables, such as jambu (Acmella oleracea), by using natural processes and living organisms that stimulate plant production and release fewer toxic residues into the environment. The objective of this study was to evaluate the growth of A. oleracea, based on biometric, and physiological responses, following the application of the entomopathogenic fungi Beauveria bassiana and Metarhizium anisopliae and the growth promoter Trichoderma asperellum, under protected and unprotected planting systems, in the rainy and dry seasons of the Amazon. Two trials were conducted simultaneously, in randomized blocks, in a commercial area of peri-urban agriculture in the municipality of Ananindeua, Pará state, Brazil, in protected and unprotected hanging beds. Metarhizium anisopliae, B. bassiana, and T. asperellum isolates promoted growth in jambu in protected and unprotected plantation systems, in both crop cycles. Of note, M. anisopliae matched the results obtained with the growth promoter T. asperellum and stood out for favoring greater performance in all of the evaluated growth variables, especially in the protected cultivation system and in rainy season. In addition, A. oleracea grew better in all treatments in a protected planting system and in both evaluated periods. RESUMO Um dos grandes desafios a serem superados é manter e aumentar a produtividade de hortaliças, como o jambu (Acmella oleracea), utilizando processos naturais e organismos vivos que estimulem a produção vegetal, com menos resíduos tóxicos no meio ambiente. O objetivo deste estudo foi avaliar o crescimento de plantas de A.oleracea, por meio de respostas biométricas e fisiológicas, após a aplicação dos fungos entomopatogênicos Beauveria bassiana e Metarhizium anisopliae e do promotor de crescimento Trichoderma asperellum, sob sistemas de plantio protegido e não protegido, nas estações chuvosa e seca da Amazônia. Dois ensaios foram conduzidos simultaneamente, em blocos casualizados, em uma área comercial de agricultura periurbana no município de Ananindeua, Pará, Brasil, em canteiros suspensos protegidos e não protegidos. Os isolados fúngicos de M. anisopliae, B. bassiana e T. asperellum promoveram o crescimento em plantações de jambu, em sistemas de plantio protegido e desprotegido, em ambos os ciclos de cultivo. No entanto, M. anisopliae se igualou aos resultados obtidos com o promotor de crescimento T. asperellum e se destacou por favorecer maior desempenho em todas as variáveis de crescimento avaliadas, principalmente no sistema de cultivo protegido e no período chuvoso amazônico. Além disso, as plantas de A. oleracea apresentaram melhor crescimento em todos os tratamentos, em sistema de plantio protegido, nos dois períodos avaliados. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. ABSTRACT In 2000, Cleide Costa published a paper presenting the state of knowledge of the Neotropical Coleopte ra, with a focus on the Brazilian fauna. Twenty-four years later, thanks to the development of the Coleoptera section of the Taxonomic Catalog of the Brazilian Fauna (CTFB - Catálogo Taxonômico da Fauna do Brasil) through the collaboration of 100 coleopterists from all over the globe, we can build on Costa’s work and present an updated overview of the state of knowledge of the beetles from Brazil. There are currently 35,699 species in 4,958 genera and 116 families known to occur in the country, including representatives of all extant suborders and superfamilies. Our data show that the Brazilian beetle fauna is the richest on the planet, concentrating 9% of the world species diversity, with some estimates accounting to up to 15% of the global total. The most diverse family in numbers of genera is Cerambycidae (1,056 genera), while in number of species it is Chrysomelidae (6,079 species). Conotrachelus Dejean, 1835 (Curculionidae) is the most species-rich genus, with 570 species. The French entomologist Maurice Pic is the author who has contributed the most to the naming of species recorded from Brazil, with 1,794 valid names in 36 families, whereas the Brazilians Ubirajara R. Martins and Maria Helena M. Galileo are the only ones among the top-ten authors to have named species in the 21st century. Abstract Introduction: Methylene Blue (MB) has been shown to attenuate oxidative, inflammatory, myocardial, and neurological lesions during ischemia-reperfusion and has great potential during cardiac arrest. This study aimed to determine the effects of MB combined with epinephrine during cardiac arrest on myocardial and cerebral lesions. Method: Thirty-eight male Wistar rats were randomly assigned to four groups: the sham group (SH, n = 5), and three groups subjected to cardiac arrest (n = 11 /group) and treated with EPI 20 μg.kg−1 (EPI), EPI 20 μg.kg−1 + MB 2 mg.kg−1 (EPI + MB), or saline 0.9% 0.2 ml (CTL). Ventricular fibrillation was induced by direct electrical stimulation in the right ventricle for 3 minutes, and anoxia was maintained for 5 minutes. Cardiopulmonary Resuscitation (CPR) consisted of medications, ventilation, chest compressions, and defibrillation. After returning to spontaneous circulation, animals were observed for four hours. Blood gas, troponin, oxidative stress, histology, and TUNEL staining measurements were analyzed. Groups were compared using generalized estimating equations. Results: No differences in the Returning of Spontaneous Circulation (ROSC) rate were observed among the groups (EPI: 63%, EPI + MB: 45%, CTL: 40%, p = 0.672). The mean arterial pressure immediately after ROSC was higher in the EPI+MB group than in the CTRL group (CTL: 30.5 [5.8], EPI: 63 [25.5], EPI+MB: 123 [31] mmHg, p = 0.007). Serum troponin levels were high in the CTL group (CTL: 130.1 [333.8], EPI: 3.70 [36.0], EPI +MB: 43.7 [116.31] ng/mL, p < 0.05). Conclusion: The coadministration of MB and epinephrine failed to yield enhancements in cardiac or brain lesions in a rodent model of cardiac arrest. Colostrum human milk samples obtained from human milk banks received different treatments and were evaluated for their proximal composition, fatty acids composition and lipid quality. The total lipids content was maintained in the treatments. There was no difference in moisture, ash, total proteins, carbohydrates and energetic value for raw colostrum human milk and pasteurized colostrum human milk, and there was difference in ash, total proteins and energetic value for freeze-dried colostrum human milk and pasteurized + freeze-dried colostrum human milk. The fatty acids composition demonstrated no difference in the processes for 14 of 31 fatty acids identified. Furthermore, lipid quality indices were obtained, which were unpublished for colostrum human milk submitted to pasteurization and freeze drying. Principal component analysis showed similar behaviors for freeze-dried colostrum human milk and pasteurized and freeze-dried colostrum human milk. So, the processes used may be promising for application in colostrum human milk, due to the maintenance of its proximal characteristics and fatty acid composition.

RESUMO O óleo de Caryocar villosum é usado na medicina popular amazônica para tratar dores e condições inflamatórias. Assim, avaliamos as propriedades antiinflamatórias e antinociceptivas do extrato etanólico obtido das cascas dos frutos desta espécie. Os testes de contorções induzidas por ácido acético, hiperalgesia mecânica induzida por carragenina, formalina, edema de pata induzido por carragenina e peritonite induzida por carragenina foram usados em camundongos. O extrato etanólico de C. villosum obtido das cascas dos frutos inibiu significativamente o número de contorções abdominais, a hiperalgesia mecânica e o tempo de lambida da pata na segunda fase do teste de formalina. Na dose de 300 mg kg-1, o extrato também reduziu significativamente o volume de edema formado na fase tardia e reduziu o recrutamento de leucócitos e neutrófilos na cavidade peritoneal, bem como os níveis de quimiocina CXCL1. Sugere-se que o extrato atenua o recrutamento de leucócitos por meio da inibição da ativação de CXCL1. A atividade antinociceptiva periférica ocorre por meio da modulação da via opioide pois o pré-tratamento com o extrato etanólico de C. villosum reverteu a antinocicepção induzida pela naltrexona.
ABSTRACT The oil of Caryocar villosum is used in Amazonian folk medicine to treat pain and inflammatory conditions. So, we assessed the anti-inflammatory and antinociceptive properties of the ethanolic extract obtained from the fruit peels of this species. The acetic acid-induced writhing, carrageenan-induced mechanical hyperalgesia, formalin, carrageenan-induced paw edema and carrageenan-induced peritonitis tests were used on mice. The C. villosum ethanolic extract significantly inhibited the number of abdominal writhes, mechanical hyperalgesia and paw licking time in the second phase of the formalin test. At a dose of 300 mg kg-1, the extract also significantly reduced the volume of edema formed in the late phase and reduced the recruitment of leukocytes and neutrophils in the peritoneal cavity, as well as CXCL1 chemokine levels. It is suggested that the extract attenuates the leukocyte recruitment by inhibiting the CXCL1 activation. The peripheral antinociceptive activity occured through opioid pathway modulation because pretreatment with C. villosum ethanolic extract reversed the naltrexone-induced antinociception.

Acerola is a tropical fruit with a large industrial and commercial appeal due to its organoleptic qualities and bioactive compound-rich nature. Thus, as bioactive compounds (BC) stability is of great interest in the food industry, assessment of preserving techniques to prolong shelf life has become imperative. Hence, the purpose of this study was to evaluate how storage affects BC conservation in microencapsulated and lyophilized acerola pulps over 180 days. For this matter, antioxidant tests, physical-chemical analyzes, and vitamin C analysis by mass spectrometry were performed. Results indicated that microencapsulation better preserved the antioxidants in acerola pulp compared to the other preservation technologies employed. This is due to the greater BC concentration in the microencapsulated, as well as the pH reduction. Furthermore, over 180 days, microencapsulation better preserved the target compounds in acerola pulp compared to lyophilization, as evidenced by the superior content of BC in the former treatment, making it an appealing option for the food sector.

The present work intends to obtain and characterize a milk by-product: the human whey (HW), in order to avoid the excessive disposal of human milk (HM). The characterization was performed through analyzes of the chemical composition and fatty acids (FAs) and the triacylglycerol (TAG) lipid profile. The results indicated that the chemical composition altered significantly after the HW obtained, except for the percentage of ash and carbohydrates. About the FA composition, it was observed that strictly essential FA, essential FA and other FAs found in HM remained present in HW. For the nutritional lipids quality, the atherogenicity and the thrombogenicity indices presented desirable values, and the TAGs lipid profile revealed a variation in the analyzed samples. Therefore, it can be concluded that the characterized HW has potential for application in isolated form or to be used as an ingredient in other foods.

Considering the increasing number of low cost vegetable oil product adulterations, it is necessary to assess the authenticity of the lipid sources used in the dog food manufacture, consequently different brands and classifications of feed were analyzed to verify the product authenticity by gas chromatography (GC) with flame ionization detector (FID) and by electrospray ionization mass spectrometry (ESI-MS). Fatty acid (FA) composition analysis was performed to compare the FAs amount in samples with the respective guarantee levels stated by the manufacturers on its product label. Gas chromatography with flame ionization detector (GC-FID) results for omega-3 and 6 percentages are within the limits presented by each manufacturer, while the timnodonic + cervonic acids amount information presented on feed sample labels is not in accordance with GC-FID results. Consequently, the samples were analyzed by ESI-MS, the results exposed the chicken oil presence in the feed and eicosapentaenoic (EPA) + docosahexaenoic (DHA) acids absence, since no lipid profiles similar to fish oils or triacylglycerols (TAGs) containing these two FAs were found. Thus, the information presented on labels are in disagreement with the results obtained both by GC-FID FA composition analysis and by ESI-MS lipid profile analysis. Principal component analysis (PCA) confirmed that the main contribution was from FAs found by GC-FID.

Abstract Objective: To analyze the impact of vacuum-assisted venous drainage (VAVD) on arterial pump flow in a simulated pediatric cardiopulmonary bypass circuit utilizing a centrifugal pump (CP) with an external arterial filter. Methods: The simulation circuit consisted of a Quadrox-I Pediatric oxygenator, a Rotaflow CP (Maquet Cardiopulmonary AG, Rastatt, Germany), and a custom pediatric tubing set primed with Lactated Ringer's solution and packed red blood cells. Venous line pressure, reservoir pressure, and arterial flow were measured with VAVD turned off to record baseline values. Four other conditions were tested with progressively higher vacuum pressures (-20, -40, -60, and -80 mmHg) applied to the baseline cardiotomy pressure. An arterial filter was placed into the circuit and arterial flow was measured with the purge line in both open and closed positions. These trials were repeated at set arterial flow rates of 1500, 2000, and 2500 mL/min. Results: The use of progressively higher vacuum caused a reduction in effective arterial flow from 1490±0.00 to 590±0.00, from 2020±0.01 to 1220±0.00, and from 2490±0.0 to 1830±0.01 mL/min. Effective forward flow decreased with increased levels of VAVD. Conclusion: The use of VAVD reduces arterial flow when a CP is used as the main arterial pump. The reduction in the forward arterial flow increases as the vacuum level increases. The loss of forward flow is further reduced when the arterial filter purge line is kept in the recommended open position. An independent flow probe is essential to monitor pump flow during cardiopulmonary bypass.

RESUMO A falta de aproveitamento dos resíduos gerados pelo processamento mecânico da madeira e seu descarte inadequado são desafios para a indústria madeireira. Esses poderiam ser utilizados em produtos de maior valor agregado. O objetivo deste trabalho foi avaliar a utilização de resíduos gerados pelo processamento mecânico das madeiras amazônicas Caryocar villosum, Hymenolobium excelsum, Mezilaurus itauba, Erisma uncinatum, Tachigali myrmecophyla e Qualea paraensis na produção de painéis aglomerados de alta densidade. Os painéis foram produzidos com massa específica nominal de 850 kg.m-3, espessura nominal de 15,7 mm e adesivo fenol-formaldeído na proporção de 8%. As propriedades físico-mecânicas dos painéis produzidos a partir de cada espécie e de uma mistura entre todas elas em partes iguais foram avaliadas em sete tratamentos. Corpos- de-prova, retirados dos painéis produzidos, foram confeccionados para ensaios físicos (massa específica, teor de umidade, absorção de água e inchamento em espessura) e mecânicos (flexão estática - MoR e MoE, e ligação interna) de acordo com a norma ABNT NBR 14810-3. As propriedades mecânicas dos painéis aglomerados, manufaturados com os resíduos de C. villosum, H. excelsum e T. myrmecophyla foram, de maneira geral, superiores às das demais espécies e atenderam um ou mais dos requisitos mínimos indicados pela norma ANSI A208.1 para painéis de alta densidade (classificação H-1) e destinados a produção de pisos (classificação PBU).
ABSTRACT The lack of use of the residues generated by the mechanical processing of wood and their inadequate disposal are challenges for the timber industry. They have potential for products with higher value. The purpose of this research was to evaluate the use of residues generated by the mechanical processing of Amazonian woods Caryocar villosum, Hymenolobium excelsum, Mezilaurus lindaviana, Erisma uncinatum, Tachigali myrmecophyla and Qualea paraensis in the high-density particleboard production. The panels produced had nominal density of 850 kg.m-3, nominal thickness of 15.7 mm and 8% of phenol formaldehyde adhesive. The physical-mechanical properties of the panels produced from each species and with a mixture of all of them in equal parts were evaluated. The specimens from the panels were prepared for physical tests (apparent density, moisture content, water absorption and thickness swelling) and mechanical tests (static bending - modulus of elasticity and modulus of rupture, and internal bond) according to the standard ABNT NBR 14810-3. The mechanical properties of the particleboards manufactured with the C. villosum, H. excelsum and T. myrmecophyla residues were generally superior than those of the other species and met one or more of the minimum requirements indicated by the ANSI A208.1 standard for (H-1 classification) and for floors production (PBU classification).

RESUMO: O tamanho e a função atrial esquerda estão relacionados em humanos com eventos cardiovasculares adversos e prognóstico nas principais cardiopatias. Na doença valvar crônica de mitral (DVCM) em cães, o átrio esquerdo é a câmara cardíaca mais comprometida. Portanto, os objetivos deste estudo foram comparar os principais parâmetros de aumento atrial esquerdo (relação átrio esquerdo/aorta, índice do diâmetro atrial esquerdo e índice do volume atrial esquerdo) e função contrátil de átrio esquerdo (velocidade máxima e integral de velocidade da onda A do fluxo transvalvar mitral, fração atrial e força de ejeção atrial) nos diferentes estágios da doença valvar; correlacionar diâmetro e volume atrial esquerdo e os índices de função contrátil atrial esquerda com as variáveis ecocardiográficas preditivas de insuficiência cardíaca em cães (velocidade máxima da onda E do fluxo transvalvar mitral, relação E/TRIV, relação E/E’par e relação E/A). Os resultados mostraram que com a evolução progressiva da doença valvar crônica de mitral, ocorre dilatação progressiva do átrio esquerdo, bem caracterizada pela relação átrio esquerdo/aorta e pelo índice de volume atrial esquerdo. O índice do diâmetro atrial esquerdo, o índice de volume atrial esquerdo e a relação átrio esquerdo/aorta correlacionam-se, positivamente, com a velocidade da onda E e com a relação E/TRIV, que são importantes índices de função diastólica. Os índices de função contrátil do átrio esquerdo aumentam à medida que a DVCM evolui. Os índices de função contrátil do átrio esquerdo apresentam correlação positiva com os índices de pressão de enchimento ventricular esquerdo, exceção feita à fração atrial.
ABSTRACT: In humans, left atrial enlargement and reduced contractile functions are associated with adverse cardiovascular events and a poor prognosis in many dilatation of the left atrium occurs with the gradual evolution of chronic mitral valve disease and is well diseases. The left atrium is the most compromised cardiac chamber in dogs with chronic mitral valve disease (CMVD). Therefore, this study aimed to compare the main parameters of left atrial enlargement (left atrium/aorta ratio, left atrial diameter and volume indices) and contractile function (transmitral flow peak velocity A wave and time velocity integral, atrial fraction, and atrial ejection force) at different stages of valve disease, and correlate the left atrial diameter, volume, and contractile function indices with echocardiographic variables predictive of heart failure in dogs (transmitral flow peak velocity E wave, E wave/IVRT ratio, E wave/E´wave ratio, and E wave/A wave ratio). The results showed that progressive characterized by the left atrium/aorta ratio and left atrium volume index. The left atrial diameter and volume indices and left atrium/aorta ratio correlated positively with the transmitral flow peak velocity E wave and E wave/IVRT ratio, which are important indices of diastolic function. The left atrial contractile function indices increased as CMVD evolved. Except for the atrial fraction, the left atrial contractile function indices correlated with the left ventricular filling pressure indices.

A colheita mecanizada da cana-de-açúcar, quando realizada em épocas da safra em que o solo se encontra com conteúdos inadequados de água, tem sido responsável pela compactação do solo e a redução da produtividade nos ciclos posteriores da cultura. Este estudo teve como objetivo avaliar o efeito das operações de colheita da cana-de-açúcar em diferentes épocas da safra sobre a pressão de preconsolidação em um Latossolo Vermelho-Amarelo (LVA) e em um Cambissolo Háplico (CX). A colheita mecanizada consistiu no tráfego de duas passadas de uma colhedora de cana e do conjunto trator + transbordo em cada entrelinha da cultura. As amostras indeformadas foram utilizadas na determinação dos modelos de capacidade de suporte de carga do solo e na quantificação da compactação em decorrência da colheita mecanizada nos meses de novembro de 2005, março e agosto de 2006, e colheita manual sem tráfego, em março de 2006, por ser a época de maior intensidade pluvial. A porcentagem de amostras de solo, na região onde ocorre compactação adicional, indicou que o CX foi mais resistente à compactação que o LVA. A colheita mecanizada da cana-de-açúcar, mesmo sendo realizada na zona de friabilidade, causou compactação do solo, enquanto a colheita realizada manualmente não promoveu o mesmo efeito.
The mechanized harvesting of sugarcane when accomplished in times of crop where the soil meets the inappropriate water contents has been responsible for soil compaction and reduction of yield in subsequent cycles of the crop. This study aimed to evaluate the effect of the harvesting operations of sugarcane at different times of the crop on preconsolidation pressure in a Red-Yellow Latosol (LVA) and in a Haplic Cambisol (CX). Mechanized harvesting traffic consisted of two passes of a sugarcane harvester and tractor + sugarcane transfer in each interrow of the culture. Undisturbed soil samples were used to determine the soil bearing capacity models and quantification of soil compaction due to mechanical harvesting in the months of November 2005, March and August 2006, and manual harvesting without traffic, in March 2006, because it is a time of greater rainfall intensity. The percentage of soil samples, in the region where additional soil compaction occurs, indicated that the CX was more resistant to compaction than the LVA. A mechanized harvesting of sugarcane being held in the friability zone caused soil compaction while the harvest accomplished manually did not cause the same effect.
