Determination of Lisdexamfetamine in Human Plasma by LC-MS/MS Method LCMS/MS LCMSMS LC MS/MS MS LCMS MSMS

Lisdexamfetamine is a prodrug that is converted to d-amphetamine in the body, mainly used to treat attention deficit hyperactivity disorder in children and adults. Its mechanism involves the enhancement of neurotransmitters such as norepinephrine and dopamine, which are critical for attention and behavioral regulation. With the upcoming patent expiration of Vyvanse®, bioanalytical methods for the quantification of lisdexamfetamine in plasma are needed for use in pharmacokinetic studies. In this work, we developed and validated a sensitive bioanalytical method using liquid chromatography-tandem mass spectrometry (LC-MS/MS) for lisdexamfetamine quantification in human plasma. The method developed was performed for 4 min per sample and using the concentration range of 0.3-100 ng mL-1. Intra-batch, inter-batch and instrument reproducibility, the precision was lower than 10%. Resumen La aplicación de herbicidas preemergentes es una estrategia de control de malezas, sin embargo, estos productos deben presentar residualidad para el control de diferentes flujos germinativos. Este estudio tuvo como objetivo evaluar la eficacia de herbicidas preemergentes en diferentes flujos de germinación de Merremia aegyptia, Mucuna aterrima y Ricinus communis, cuando se aplican sobre diferentes cantidades de paja y diferentes períodos secos simulados. El experimento se realizó en un diseño factorial 4 × 2 × 2 con cuatro repeticiones, el tratamiento incluye cuatro períodos secos (0, 30, 60 y 90), dos cantidades de paja (0 y 10 t ha-1) y dos flujos de germinación. Los herbicidas amicarbazona (1225 g ha-1); imazapic (147 g ha-1), sulfentrazona (800 g ha-1) y tebuthiuron (900 g ha-1), se aplicaron para el control de malezas antes de la emergencia, y los flujos de flujo de germinación se evaluaron a los 7, 14, 21, 28 y 35 días después de la emergencia de especies (DAE), mientras se verifica la masa seca. La amicarbazona presentó una reducción del control para todas las especies en los periodos secos más prolongados y presencia de paja. La amicarbazona mostró menos del 80% de control para todas las especies a los 90 días del período seco en presencia de paja. Imazapic no presentó residuo control para ninguna de las especies analizadas. Para sulfentrazona, la cantidad de paja no afectó el control de las malezas en diferentes flujos de germinación, mostrando el mismo patrón de control independientemente de la cantidad de paja, porcentajes de control superiores al 80% independientemente de la cantidad de paja. Los herbicidas asociados a cantidades de paja y periodos secos tienen impacto sobre las especies de malezas M.aegyptia, M.aterrima y R. communis.
Abstract Preemergent herbicides are a frequent weed control strategy. Considering different crop germinative fluxes, these products must present long-lasting weed control. This study evaluated preemergent herbicides in different germination fluxes of Merremia aegyptia, Mucuna aterrima and Ricinus communis when applied to different quantities of straw and different simulated dry periods. The experiment was conducted in a 4 × 2 × 2 factorial design with four replications. The treatments included four dry periods (0, 30, 60, and 90 days), two straw quantities (0 and 10 t ha-1), and two germination fluxes. The herbicides amicarbazone (1225 g ha-1), imazapic (147 g ha-1), sulfentrazone (800 g ha-1), and tebuthiuron (900 g ha-1) were applied for preemergence weed control, and germination flush fluxes were evaluated at 7, 14, 21, 28, and 35 days after emergence (DAE) while verifying plant dry mass. Amicarbazone controlled less than 80% of the studied species at the 90-day dry period in the presence of straw. Imazapic did not present control residue for any of the species analyzed. Sulfentrazone showed the same control pattern at all germination fluxes, regardless of the amount of straw. Tebuthiuron successfully controlled all species in the first germination flush, exceeding 80% regardless of the amount of straw. Herbicides associated with straw quantities and dry periods have a significant impact on M. aegyptiaca, M. aterrima and R. communis.

ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. This study describes the validation of a headspace gas chromatography with flame ionization (HS-GC-FID) method for the determination of propanal, pentanal, hexanal, octanal, and trans-2-nonenal originated by lipid oxidation from cat food samples, as well as the evaluation of the matrix effect in the results. This method was applied to the analysis of commercial cat food and proved to be suitable for the determination of these volatile compounds in different samples. Mean recoveries between 88 and 109% were obtained and repeatability expressed as relative standard deviation was always lower than 6.95%. The intra- and inter-assay precisions ranged from 0.44 to 20.88% and from 0.45 to 20.52%, respectively. In addition, the matrix effect of cat food samples was determined by comparing the slopes of the standard addition method, and the external calibration curve and its influences were verified. These results highlight the high potential of this method, which allows the determination of lipid oxidation products in cat food samples directly, without requiring prior sample preparation techniques.

Progenesis QI (PQI) is a multiplatform bioinformatics tool that facilitates the identification workflow for metabolomics experiments. PQI uses fragmentation data provided by MassBank of North America (MoNA) libraries, among others, for metabolite annotation. However, PQI does not officially support MoNA libraries and other libraries based on structure-data files (.sdf). This paper describes the development and application of a software named MoNA to Progenesis QI Library Converter, allowing PQI and MoNA by correcting the fragmentation data of the library for Progenesis readability. We evaluated several public experimental datasets, including human plasma, plant extracts, cultured cells, bacteria, rat serum, and rat hippocampus. The results showed that it is mandatory to proceed with file conversion of each library to allow PQI to access fragmentation information from .msp (main spectra profile) files. This work reports the use of the Octave, a free computational software, to optimize the experimental design in the synthesis of ethyl oleate from oleic acid and ethanol catalyzed by a native lipase from Aspergillus niger. The central composite factorial design (CCD) was performed with 2 levels and 3 variables (temperature, mass of native A. niger lipase and oleic acid:ethanol molar ratio) in conjunction with response surface methodology (RSM). The results indicate that variable 1 (oleic acid:ethanol molar ratio; X1) and 2 (mass of A. niger lipase, X2), as well as the interaction between them, are statistically significant. The optimal reaction condition was estimated using ANOVA data and the response surface. The experimental results are consistent with those estimated, and thus, ethyl oleate was obtained with conversion (> 90%) using oleic acid and ethanol in a 1:2 molar ratio, A. niger lipase (8,00 mg) and temperature of 45 °C in 6 hours of reaction.

Resumen El objetivo del presente estudio fue investigar la posible influencia de la alelopatía de los extractos acuosos de cultivos forrajeros, aislados o en consorcio, sobre rendimiento fisiológico de las semillas de soja [Glycine max (L.) Merr.] y de maíz (Zea mays L.), además de su fitotoxicidad en la hierba Bidens pilosa L. Se prepararon extractos acuosos a una concen tración del 5%. Los estudios con soja y maíz se llevaron a cabo humedeciendo los papeles de germinación correspondientes a los diferentes tratamientos y se realizaron dentro de los estándares de las Reglas de Análisis de Semillas de Brasil. Estudios con los extractos en las semillas de B. pilosa se llevaron a cabo en una cámara de tipo B.O.D. Se colocaron cincuenta semillas de malezas en caja gerbox conteniendo papel de filtro humedecido con los diferentes tratamientos en cinco repeticiones. El agua destilada se utilizó como control en todas las pruebas. Los extractos de Avena strigosa y Raphanus sativus L., cultivados tanto en forma aislada como en consorcio, aumentaron el porcentaje de germinación de semillas (%G) y la longitud de las plántulas de soja. Asimismo, los extractos de Pennisetum glaucum (L.) R.Br. y Crotalaria spectabilis Roth, cultivados aislados y en consorcio, proporcionaron ganancias en el %G de maíz. Además, todos los extractos evaluados suprimieron la germi nación y redujeron la velocidad del índice de germinación de la maleza en comparación con el control, siendo el extracto de R. sativus el que más redujo el % G de B. pilosa.
Abstract The aim of the present study was to investigate the potential allelopathic effects of isolated or intercropped aqueous forage extracts on the physiological performance of soybean [Glycine max (L.) Merr.] and corn (Zea mays L.) seeds, as well as their phytotoxicity to the weed species Bidens pilosa L. Aqueous extracts were prepared at a concentration of 5%, and the tests with soy and corn were conducted by wetting the germination papers with different treatments and performing procedures within the standards of the Brazilian Rules of Seed Analysis. Tests with the cover plant extracts on the seeds of B. pilosa used BOD-type repetitions. Fifty weed seeds were placed in a gerbox with filter paper moistened with the different treatments with five repetitions. Distilled water was used as the control in all tests. Extracts of Avena strigosa Schreb and Raphanus sativus L. (radish), cultivated individually and in combination, increased the seed germination percentage (%G) and the length of soybean seedlings. Similarly, extracts of Pennisetum glaucum (L.) R.Br and Crotalaria spectabilis Roth, in individual and combined cultivation, provided gains in corn %G. Furthermore, all tested extracts suppressed germination and decreased the speed of the germination index of the hairy beggarticks when compared to the control, with the radish extract showing the greater reduction effect on the %G of the weed plant B. pilosa.

Resumo As características intrínsecas de muitas frutas tropicais causam grandes perdas pós-colheita e impedem sua comercialização como frutas frescas. As informações sobre sua composição são cruciais para definir as condições de processamento e identificar oportunidades de desenvolvimento de produtos. No entanto, os métodos analíticos geralmente usados para quantificar açúcares e ácidos orgânicos são caros e demorados. A análise simultânea por Cromatografia Líquida-Ionização por Electrospray-Espectrometria de Massa (LC-ESI-MS/MS) é uma técnica muito sensível e reproduzível, permitindo a quantificação simultânea precisa em sistemas complexos. Assim, desenvolvemos um método LC-ESI-MS/MS de monitoramento de reações múltiplas (MRM) usando coluna de fase reversa para detectar e quantificar açúcares e alguns ácidos orgânicos em apenas quatro minutos, em frutas selecionadas: araza (Psidium cattleianum), ceriguela (Spondias purpurea), manga (Mangifera indica), goiaba (Psidium guajava) e pitanga (Eugenia uniflora L.). Todas as frutas apresentaram concentrações semelhantes de glicose e frutose, exceto a pitanga, que apresentou valores superiores. O conteúdo de ácidos cítrico, málico e tartárico foi quantificado; algumas frutas se destacaram pelo alto teor de ácidos orgânicos. Os dados gerados sobre a composição das frutas tropicais permitem estabelecer uma correlação entre o teor de carboidratos e as propriedades físicas das polpas das frutas, auxiliando no desenvolvimento de novos produtos.
Abstract The intrinsic characteristics of many tropical fruits cause high post-harvest losses and prevent their commercialization as fresh fruits. Information about their composition is crucial for defining processing conditions and identifying opportunities for product development. However, the analytical methods generally used to quantify sugars and organic acids are costly and time-consuming. Simultaneous analysis by Liquid Chromatography-Electrospray Ionization-Mass Spectrometry (LC-ESI-MS/MS) is a very sensitive and reproducible technique, allowing for accurate simultaneous multi-analyte quantitation in complex systems. Thus, a LC-ESI-MS/MS Multiple Reaction Monitoring (MRM) method was developed using reverse phase column for detecting and quantifying sugars and some organic acids in only 4 min, in selected fruits as following: araza (Psidium cattleianum L.), ciriguela (Spondias purpurea L.), mango (Mangifera indicaL.), guava (Psidium guajava L.) and pitanga (Eugenia uniflora L.). All fruits had a similar concentration of glucose and fructose, except for pitanga that presented higher values of both. The content of citric, malic and tartaric acids was quantitated; some fruits stood out by their high content of organic acids, for instance, araza was rich in citric acid. The newly generated data on the composition of tropical fruits allowed establishing a correlation between the carbohydrate content and the physical properties of the fruit pulps, assist in product development.

OBJECTIVES: To prospectively evaluate demographic, anthropometric and health-related quality of life (HRQoL) in pediatric patients with laboratory-confirmed coronavirus disease 2019 (COVID-19) METHODS: This was a longitudinal observational study of surviving pediatric post-COVID-19 patients (n=53) and pediatric subjects without laboratory-confirmed COVID-19 included as controls (n=52) was performed. RESULTS: The median duration between COVID-19 diagnosis (n=53) and follow-up was 4.4 months (0.8-10.7). Twenty-three of 53 (43%) patients reported at least one persistent symptom at the longitudinal follow-up visit and 12/53 (23%) had long COVID-19, with at least one symptom lasting for >12 weeks. The most frequently reported symptoms at the longitudinal follow-up visit were headache (19%), severe recurrent headache (9%), tiredness (9%), dyspnea (8%), and concentration difficulty (4%). At the longitudinal follow-up visit, the frequencies of anemia (11% versus 0%, p=0.030), lymphopenia (42% versus 18%, p=0.020), C-reactive protein level of >30 mg/L (35% versus 0%, p=0.0001), and D-dimer level of >1000 ng/mL (43% versus 6%, p=0.0004) significantly reduced compared with baseline values. Chest X-ray abnormalities (11% versus 2%, p=0.178) and cardiac alterations on echocardiogram (33% versus 22%, p=0.462) were similar at both visits. Comparison of characteristic data between patients with COVID-19 at the longitudinal follow-up visit and controls showed similar age (p=0.962), proportion of male sex (p=0.907), ethnicity (p=0.566), family minimum monthly wage (p=0.664), body mass index (p=0.601), and pediatric pre-existing chronic conditions (p=1.000). The Pediatric Quality of Live Inventory 4.0 scores, median physical score (69 [0-100] versus 81 [34-100], p=0.012), and school score (60 [15-100] versus 70 [15-95], p=0.028) were significantly lower in pediatric patients with COVID-19 at the longitudinal follow-up visit than in controls. CONCLUSIONS: Pediatric patients with COVID-19 showed a longitudinal impact on HRQoL parameters, particularly in physical/school domains, reinforcing the need for a prospective multidisciplinary approach for these patients. These data highlight the importance of closer monitoring of children and adolescents by the clinical team after COVID-19.

Tilapia (Oreochromis niloticus) is the most cultivated fish species in Brazil. Studies have shown that fish consumption is related to the prevention of cardiovascular diseases due to its high contents of polyunsaturated fatty acids, as omega-3 (n-3) and omega-6 (n-6). In this sense, a new method to determine omega-3 and omega-6 by near-infrared (NIR) spectroscopy and multivariate calibration based on partial least squares (PLS) was proposed. Spectra of fillets of Nile tilapias packed under vacuum were obtained and compared to the reference method (gas chromatography) used for quantification of fatty acids. The outliers were evaluated in the models that were validated by calculation of the parameters for model performance estimation, showing promising results. The appropriate values achieved for accuracy, limits of detection and quantification, indicate that the NIR/PLS models can be an alternative to chromatography in the determination of omega-3 and omega-6 in fillets of Nile tilapia. Furthermore, the spectra acquisition on the vacuum-packed fillet contributes to the NIR/PLS method in terms of cost and quickness.

Resumo O presente artigo traz uma revisão da principal literatura produzida pelo Grupo de Estudos Arqueométricos do Instituto de Pesquisas Energéticas e Nucleares, Comissão Nacional de Energia Nuclear, de São Paulo (IPEN-CNEN/SP), criado em 1997, como consequência de uma iniciativa internacional para motivar o uso de técnicas nucleares para caracterização de materiais arqueológicos. Desde aquele momento, e até hoje, o grupo tem desenvolvido parcerias com pesquisadores/as de diferentes regiões do Brasil e de outros países para oferecer subsídios à compreensão interdisciplinar das cadeias operatórias de produção da cerâmica. Além disso, também vem aportando dados para o estudo multielementar e mineralógico de depósitos arqueológicos. Ao descrever brevemente os princípios das técnicas empregadas e a síntese dos resultados obtidos para cada estudo de caso, o objetivo foi demonstrar a contribuição das análises físico-químicas para questões relacionadas a aspectos culturais, como escolhas tecnológicas compartilhadas, redes de trocas e interações sociais em diferentes períodos do passado.
Abstract The present paper brings together key literature produced by the Group of Archaeometric Studies of the Nuclear and Energy Research Institute, IPEN – National Nuclear Energy Commission (São Paulo, Brazil). This group was founded in 1997, as a result of an international initiative to encourage the use of nuclear techniques for the characterization of archaeological materials. Since then, the Group has developed interdisciplinary partnerships with researchers from Brazil and other countries to offer analytical tools for unraveling ceramic chaîne opératoires in different contexts. Such studies have contributed towards wider cultural interpretations regarding shared technological choices and artifact exchange networks in different social groups. Recently, the Group has also used multi-elemental and mineralogical techniques to tackle questions over the formation of archaeological deposits. This paper briefly explains the analytical principles of the employed techniques, and emphasizes how the results of each case study contribute to the interpretation of specific archaeological problems.

OBJECTIVES: The present coronavirus disease (COVID-19) pandemic has ushered in an unprecedented era of quality control that has necessitated advanced safety precautions and the need to ensure the adequate protection of healthcare professionals (HCPs). Endoscopy units, endoscopists, and other HCP may be at a significant risk for transmission of the virus. Given the immense burden on the healthcare system and surge in the number of patients with COVID-19, well-designed protocols and recommendations are needed. We aimed to systematically characterize our approach to endoscopic procedures in a quaternary university hospital setting and provide summary protocol recommendations. METHOD: This descriptive study details a COVID-19-specific protocol designed to minimize infection risks to patients and healthcare workers in the endoscopy unit. RESULTS: Our institution, located in São Paulo, Brazil, includes a 900-bed hospital, with a 200-bed-specific intensive care unit exclusively designed for patients with moderate and severe COVID-19. We highlighted recommendations for infection prevention and control during endoscopic procedures, including appropriate triage and screening, outpatient management and procedural recommendations, role and usage of personal protective equipment (PPE), and role and procedural logistics involving COVID-19-positive patients. We also detailed hospital protocols for reprocessing endoscopes and cleaning rooms and also provided recommendations to minimize severe acute respiratory syndrome coronavirus 2 transmission. CONCLUSION: This COVID-19-specific administrative and clinical protocol can be replicated or adapted in multiple institutions and endoscopy units worldwide. Furthermore, the recommendations and summary protocol may improve patient and HCP safety in these trying times.

Leishmaniasis is a neglected infectious disease caused by different species of the Leishmania parasite and is one of the major public health problems in developing countries. Despite the progress in fundamental knowledge about the Leishmania parasite, current therapy against leishmaniasis is still unsatisfactory due to limited efficacy, prolonged treatment, high cost and undesirable adverse effects. Thus, the research for new prototypes compounds of antileishmaniasis drugs remains a matter of current importance. The objective of this study was to evaluate the leishmanicidal activity of cytochalasin B, a natural compound isolated by our group in a previous study, against L. amazonensis, using experimental tests and computational simulation methodologies. The results of the biological evaluation showed that cytochalasin B has antileishmanial activity against the promastigote form of Leishmania amazonensis. These results are corroborated by the docking and molecular dynamics that showed that the activity occurs due to the inactivation of the trypanothione reductase enzyme (TryR).

Jatropha curcas L. is an oil crop that has been studied as a potential source of biodiesel. A high protein pie is produced as a byproduct of the biodiesel production, which could be used as animal feed. However, the pie has toxic compounds, as phorbol esters and other toxins, which prevents the use as animal feed. For this reason, Embrapa (Brazilian Agricultural Research Corporation) has been working in genetic breeding to develop non-toxic J. curcas genotypes. To evaluate this process, a simple and fast analytical technique was employed to obtain responses in a short time. Leaf spray (LS) is a recent ambient ionization mass spectrometry technique in which the sample itself serves as support and ion source. Here, toxic and non-toxic J. curcas leaves were differentiated by LS using a linear ion trap mass spectrometer and partial least squares discriminant analysis (PLS-DA) model chemometrics. It was possible to differentiate toxic and non-toxic leaves and to identify the m/z values that contribute to discrimination between the groups.

Abstract Choisya ternata Kunth, C. ternata var. sundance Kunth and the hybrid Choisya ‘Aztec-Pearl’ are three related species belonging to the Rutaceae family. Ethanol extracts were prepared from the leaves of these three species and evaluated in relation to their antioxidant activity using in vitro and ex vivo models. The ethanol extracts belonging to the three species produced a very high antioxidant profile as evidenced by the DPPH radical scavenging activity, the determination of total phenolics and flavonoid equivalent. The generation of reactive species of oxygen in leukocytes stimulated with LPS was dramatically reduced when the three ethanol extracts were used. The alkaloids anhydroevoxine and choisyine were isolated from the ethanol extract of C. ternata using HEMWat (4:6:5:5) as the solvent system by means of high-speed countercurrent chromatography. This was the first time quinoline alkaloids were isolated from this species using HSCCC. These compounds were also assayed for their capacity to inhibit the generation of ROS in leukocytes stimulated by LPS and the results also suggested that they are reactive oxygenase inhibitors.
