RESUMO O aporte de resíduos vegetais ao longo dos ciclos da cana-de-açúcar favorece o incremento de matéria orgânica e atividade de microrganismos no solo, especialmente nas camadas superficiais. A textura do solo também tem um efeito importante nos processos ecológicos e na qualidade do solo. Nesse sentido, o objetivo desta pesquisa foi avaliar atributos biológicos do solo em diferentes ciclos de cultivo de cana-de-açúcar sob colheita mecanizada, em um Latossolo da região de cerrado do Brasil Central. O estudo foi realizado em áreas comerciais de cultivo de cana-de-açúcar na safra 2018/2019, consideradas homogêneas em termos de condições edafoclimáticas, tendo como fonte de variação entre as áreas os ciclos de cultivo (C1: um ciclo de cultivo; C3: três ciclos de cultivo; C7: sete ciclos de cultivo) e uma área de vegetação de cerrado selecionada como referência. Foram determinadas variáveis microbiológicas nas camadas de 0-0,10 e 0,10-0,20 m. As variáveis relacionados à biomassa microbiana e textura foram submetidos à análise de componentes principais. Áreas com ciclos de cultivo de cana-de-açúcar mais longos apresentaram maior proporção de carbono da biomassa microbiana no carbono orgânico total nas camadas subterrâneas (quociente microbiano). ABSTRACT The contribution of plant residues throughout the sugarcane cycles favors the increase of organic matter and the activity of microorganisms in the soil, especially in the surface layers. Soil texture also has an important effect on ecological processes and soil quality. In this context, the objective of this study was to evaluate soil biological attributes in different sugarcane cultivation cycles under mechanized harvesting in an Oxisol in the Savanna region of Central Brazil. The study was conducted in commercial areas under sugarcane cultivation during the 2018/2019 season, which were considered homogeneous in terms of soil and climatic conditions, with the source of variation among the areas being the cultivation cycles (C1: one cultivation cycle; C3: three cultivation cycles; C7: seven cultivation cycles) and a savanna vegetation area selected as a reference. Microbiological variables were determined in two layers, 0-0.1 and 0.1-0.2 m. The variables related to microbial biomass and texture were subjected to principal component analysis. Areas with longer sugarcane cultivation cycles show higher proportion of microbial biomass carbon in the total organic carbon in subsurface layers (microbial quotient). Caparaó and the Fluminense northwest regions are nationally recognized by the important contribution on coffee production and exportation. Adulterations involving specialty coffees result in a decrease in the quality of the final product. However, obtaining many different samples from the same region is unfeasible in some cases, needing strategies to work with a limited number of samples for pattern recognition. Thus, this work is the first to use the construction of synthetic samples (SS) for analysis of coffees, and its objective is to identify adulterations in specialty coffees with bark, straw and low-quality beans, using UV-Vis spectroscopy, associated with chemometric methods. The synthetic samples partial least square discriminant analysis (SS-PLS-DA) showed better specificity, sensitivity and reliability rates than the Hard PLS-DA models. One-class methods (soft independent modeling of class analogy (SIMCA) and data driven soft independent modeling of class analogy (DD-SIMCA)) showed low specificity and reliability. RESUMO Os recursos hídricos, especialmente em regiões semiáridas, devem ser utilizados com o emprego de tecnologias que permitam aumentar a eficiência e uso da água para irrigação. Uma estratégia para lidar com esse problema é o uso de porta-enxertos tolerantes à seca. Com o objetivo de selecionar porta-enxertos de videira adequados para o cultivo em regiões semiáridas, com base na biomassa, variáveis bioquímicas e nas trocas gasosas, o experimento foi conduzido no Campo Experimental de Bebedouro, pertencente à Embrapa Semiárido, em Petrolina-PE. Foram utilizados os portaenxertos ‘Paulsen1103’, ‘SO4’, ‘IAC 313’, ‘IAC 572’, ‘IAC 766’, ‘Ramsey’ e ‘101-14 MgT’ submetidos a três lâminas de irrigação (100, 50 e 20% da ETo). O delineamento experimental foi em blocos casualizados com quatro repetições, em parcelas subdivididas, com a parcela principal sendo a lâmina de irrigação e a subparcela os portaenxertos. Características bioquímicas, de biomassa e relacionadas às trocas gasosas foram utilizadas para seleção dos melhores portaenxertos quanto à tolerância à seca, por meio de análise multivariada de componentes principais (PCA) e análise de variância. Os portaenxertos ‘IAC 313’ e ‘IAC 766’ destacaram-se pela maior síntese de prolina radicular, o ‘IAC 766’ apresentou melhor resultado para síntese de sacarose foliar, enquanto ‘Paulsen1103’ se destacou em prolina foliar, carotenoide e razão clorofila total/carotenoide. ABSTRACT Water resources used for irrigation should be managed using technologies that improve water use efficiency, mainly in semiarid regions. Using drought-tolerant rootstocks is a strategy to handle this challenge. The objective of this study was to select suitable grapevine rootstocks for cultivation in semiarid regions based on their biomass, biochemical, and gas exchange attributes. The experiment was conducted at the Bebedouro Experimental Field of the Brazilian Agricultural Research Corporation (EMBRAPA Semiarid), in Petrolina, PE, Brazil. Rootstocks from grapevine plants of the cultivars Paulsen 1103, SO4, IAC 313, IAC 572, IAC 766, Ramsey, and 101-14 MgT were subjected to three irrigation water depths (100%, 50%, and 20% ET0). A randomized block experimental design with four replications was used, in a split-plot arrangement consisting of irrigation water depths in the plots and rootstocks in the subplots. Biochemical, biomass, and gas exchange attributes were assessed for selecting the best rootstocks regarding tolerance to drought using multivariate principal component analysis (PCA) and analysis of variance. The rootstocks IAC 313 and IAC 766 presented the highest root proline synthesis; IAC 766 presented better result for leaf sucrose synthesis; and Paulsen 1103 presented the highest leaf proline synthesis and carotenoid contents, as well as total chlorophyll-to-carotenoid ratio. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract Angiogenesis (budding of new blood vessels) is involved in several processes, including the development of embryos and growth of tumors. Schinus terebinthifolia leaves express an antitumor lectin (SteLL). This work hypothesized that SteLL can interfere with the formation of a vascular network from preexisting vessels. To test this hypothesis, the effect of SteLL on the angiogenesis process was assessed using an in vivo model of yolk sac membrane of Coturnix japonica embryos. SteLL was isolated with purification factor of 46.6. As expected, polyacrylamide gel electrophoresis (PAGE) for native basic proteins confirmed the homogeneity and PAGE in presence of dodecyl sodium sulphate revealed a single 14-kDa polypeptide band. The fractal analysis by box counting and information dimension measurements indicated that SteLL at 1.35 mg/mL significantly decreased by ca. 12% the angiogenesis within the C. japonica yolk sac membrane regarding the control. The inhibition of the vascular network formation in the yolk sac membrane resulted in decreased blood supply to the embryos. Consequently, the area of embryos was significantly reduced by 9.2% regarding the control, which corroborated with the antiangiogenic activity of SteLL. The findings implicate SteLL as an antiangiogenic agent and add to the panel of biological activities of this lectin.

ABSTRACT Background/objective: Dawson’s fingers have been traditionally associated with multiple sclerosis. However, this imaging biomarker has also been linked to white matter hyperintensities related to cerebral small vessel disease. In the latter, Dawson’s fingers could represent damage of small venules in subjects with severe small vessel disease and could theoretically be associated with cognitive decline. In this study, we aimed to assess the association between Dawson’s fingers and cognitive performance in a population of older adults. Methods: Population-based study conducted in individuals aged 60 years or older, residing in three rural villages of coastal Ecuador (Atahualpa, El Tambo and Prosperidad). Of 712 older adults identified by means of a door-to-door survey, 590 underwent a brain MRI. Of them, 575 also had the Montreal Cognitive Assessment (MoCA). We selected the 157 individuals with moderate-to-severe white matter hyperintensities to assess the presence of Dawson’s fingers. The independent association between Dawson’s fingers and cognitive performance (as the dependent variable) was assessed by means of a linear regression model, after adjusting for demographics, cardiovascular risk factors, and the other biomarkers of cerebral small vessel disease. Results: Of 157 individuals with moderate-to-severe White matter hyperintensities, 17 (11%) had Dawson’s fingers on MRI. The mean MoCA score in subjects with Dawson’s fingers was 14.5±6.4 points and that of those without this neuroimaging biomarker was 17.3±6.2 points. The association between Dawson’s fingers and the MoCA score was marginal in univariate models (p=0.082), but it completely vanished in a multivariate linear regression model adjusted for relevant covariates (β: -0.31; 95% C.I.: -3.23 - 2.60; p=0.833). A mediation model disclosed that 83.5% of the effect of Dawson’s finger on cognitive performance was mediated by age. Conclusion: Dawson’s fingers are not independently associated with cognitive performance in individuals with cerebral small vessel disease. Most of the effect of this association is mediated by age.
RESUMEN Antecedentes/objetivo: Los dedos de Dawson, tradicionalmente asociados con esclerosis múltiple, han sido - además - recientemente vinculados con hiperintensidades de sustancia blanca causadas por enfermedad de pequeño vaso cerebral. En este contexto, dichas lesiones corresponderían a daño venular, podrían representar estadíos avanzados de enfermedad de pequeño vaso y estarían teóricamente asociados con deterioro cognitivo. En el presente estudio se intentó determinar si los dedos de Dawson se asociaron con un peor desempeño cognitivo en adultos mayores. Métodos: Estudio poblacional realizado en personas de 60 años o más, residentes de tres poblados del litoral Ecuatoriano (Atahualpa, El Tambo y Prosperidad). De un total de 712 adultos mayores identificados mediante censos puerta-a-puerta, 590 fueron sometidos a IRM cerebral y de éstos, a 575 también se les realizó valoración cognitiva mediante la prueba de Montreal (MoCA). Se seleccionaron los 157 individuos con hiperintensidades de sustancia blanca de moderada a severa y se determinó la presencia de dedos de Dawson en dichos sujetos. La asociación independiente entre dedos de Dawson y desempeño cognitivo (como variable dependiente) fue evaluada mediante un modelo de regresión linear ajustado por factores demográficos y de riesgo cardiovascular, así como por los demás biomarcadores de enfermedad de pequeño vaso cerebral. Resultados: De los 157 individuos con hiperintensidades de sustancia blanca de moderada a severa, 17 (11%) presentaron dedos de Dawson en IRM. La puntuación promedio en el MoCA fue de 14.5±6.4 en sujetos con dedos de Dawson y de 17.3±6.2 en aquellos sin este biomarcador de neuroimagen. La asociación entre dedos de Dawson y puntaje en el MoCA fue marginal en el modelo univariado (p=0.082), mientras que dicha asociación se diluyó completamente luego de ajustar por las covariables de interés (β: -0.31; 95% I.C.: -3.23 - 2.60; p=0.833). El 83.5% del efecto de los dedos de Dawson en el desempeño cognitivo estuvo mediado por la edad. Conclusión: Los dedos de Dawson no se asocian independientemente con el desempeño cognitivo en individuos con enfermedad de pequeño vaso. La mayor parte de esta asociación se encuentra mediada por la edad.

In the present work five acetylene derivatives (1-5), including three unknowns (1, 3 and 4), were isolated from seeds of Porcelia macrocarpa (Annonaceae). The structures of isolated compounds were determined as docos-13-yn-21-enoic acid (1), 3-hydroxy-4-methylene-2-(eicos-11’-yn-19’-enyl)but-2-enolide (2), 3-hydroxy-4-methylene-2-(octadec-9’-yn-17’-enyl)but-2-enolide (3), 3-hydroxy-4-methylene-2-(hexadec-7’-yn-15’-enyl)but-2-enolide (4), and (2S,3R,4R)-3-hydroxy-4-methyl-2-(eicos-11’-yn-19’-enyl)butanolide (5) by analysis of nuclear magnetic resonance (NMR) and electrospray ionization high-resolution mass spectrometry (ESI-HRMS) data. Moreover, all isolated compounds demonstrated selectivity towards intracellular amastigotes of Leishmania (L.) infantum, especially 2-4 with 50% inhibitory concentration (IC50) values of 9.2, 10.4 and 11.0 µM, respectively, indicating superior activity of that determined to positive control miltefosine (IC50 of 17.8 µM). Furthermore, these compounds showed higher selectivity index (SI) in comparison with miltefosine. Since related acetylene fatty acid 1 displayed reduced antiparasitic potential (IC50 of 48.5 µM), the obtained results suggested that the γ-lactone plays an important role in the antileishmanial activity. However, 2-4 exhibited cytotoxicity to mammalian NCTC cells (CC50 ca. 80 µM), which could be a result of the presence of a conjugated carbonyl system in the lactone ring, since 5, the only acetogenin that presents the saturated ring, lacked mammalian cytotoxicity (CC50 > 200 µM).

ABSTRACT In response to the Zika epidemics in Brazil, the ZDC molecular assay (Bio-Manguinhos) was developed and registered at the Brazilian Regulatory Agency of Health Surveillance - ANVISA. The circulation of Zika (ZIKV) Dengue (DENV) and Chikungunya (CHIKV) viruses and their clinical similarities are challenges to correctly diagnose these viruses. The simultaneous detection of ZIKV, DENV and CHIKV is an important tool for diagnosis and surveillance. Here, we present the analytical and clinical performance evaluation of ZDC molecular assay (Bio-Manguinhos) at the public health laboratories three years after its registration at ANVISA. The clinical performance demonstrates the ZDC molecular assay (Bio-Manguinhos) has 100% sensitivity and 100% specificity to detect and discriminate ZIKV, CHIKV, and DENV from clinical plasma samples. The ZDC molecular assay (Bio-Manguinhos) results were highly reproducible and no cross-reactivity was seen during testing with a panel of other infectious agents. In conclusion, the ZDC molecular assay (Bio-Manguinhos) is an accurate and reliable tool to monitor Zika, dengue and chikungunya infections in countries like Brazil with simultaneous circulation of the three viruses.

A novel anti-inflammatory hybrid 3-ibuprofenyl-copalic acid (3-IbuCA) was synthesized from 3-hydroxy-copalic acid isolated from Amazonian copaiba oil (Copaifera multijuga Hayne), and the anti-inflammatory ibuprofen. After full characterization, several assays to verify its anti-inflammatory effects were performed in vitro, in vivo and in silico (molecular docking). Induced fit docking was performed to observe the interactions with the enzymes cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2). In vitro tests of cytotoxicity and tumor necrosis factor (TNF)-α inhibition, and in vivo tests of pleurisy, protein expression and gastrocytotoxicity were performed. Molecular docking studies with COX-1 and 2 showed binding free energies (ΔG) of -2.2 and -7.8 kcal mol-1, respectively, while for mofezolac and indomethacin, the binding free energies ΔG presented values of -8.5 and -10.1 kcal mol-1, which makes 3-IbuCA selective for COX-2 inhibition. This hybrid showed no toxicity against human macrophage at concentrations up to 2 µM, and inhibited TNF-α production in lipopolysaccharide (LPS)-stimulated macrophages. In the pleurisy assays, 3-IbuCA reduced the total leukocytes and mononuclear cells, which was followed by reduction of p-IKBα (phosphorylated nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor, alpha) protein expression. Compared with ibuprofen alone, the hybrid caused less gastric damage. Thus, the docking, together with in vitro and in vivo studies suggest that this novel hybrid has potential as a new anti-inflammatory agent.

Resumo O presente artigo traz uma revisão da principal literatura produzida pelo Grupo de Estudos Arqueométricos do Instituto de Pesquisas Energéticas e Nucleares, Comissão Nacional de Energia Nuclear, de São Paulo (IPEN-CNEN/SP), criado em 1997, como consequência de uma iniciativa internacional para motivar o uso de técnicas nucleares para caracterização de materiais arqueológicos. Desde aquele momento, e até hoje, o grupo tem desenvolvido parcerias com pesquisadores/as de diferentes regiões do Brasil e de outros países para oferecer subsídios à compreensão interdisciplinar das cadeias operatórias de produção da cerâmica. Além disso, também vem aportando dados para o estudo multielementar e mineralógico de depósitos arqueológicos. Ao descrever brevemente os princípios das técnicas empregadas e a síntese dos resultados obtidos para cada estudo de caso, o objetivo foi demonstrar a contribuição das análises físico-químicas para questões relacionadas a aspectos culturais, como escolhas tecnológicas compartilhadas, redes de trocas e interações sociais em diferentes períodos do passado.
Abstract The present paper brings together key literature produced by the Group of Archaeometric Studies of the Nuclear and Energy Research Institute, IPEN – National Nuclear Energy Commission (São Paulo, Brazil). This group was founded in 1997, as a result of an international initiative to encourage the use of nuclear techniques for the characterization of archaeological materials. Since then, the Group has developed interdisciplinary partnerships with researchers from Brazil and other countries to offer analytical tools for unraveling ceramic chaîne opératoires in different contexts. Such studies have contributed towards wider cultural interpretations regarding shared technological choices and artifact exchange networks in different social groups. Recently, the Group has also used multi-elemental and mineralogical techniques to tackle questions over the formation of archaeological deposits. This paper briefly explains the analytical principles of the employed techniques, and emphasizes how the results of each case study contribute to the interpretation of specific archaeological problems.

Expression of FoxP3 in different forms of leprosy and reactions

RESUMO Embora as células T reguladoras (Treg) sejam importantes na regulação imunológica de vários processos patológicos, o papel delas na imunologia da hanseníase ainda precisa ser mais bem avaliado. O objetivo deste trabalho é quantificar o nuclear transcription factor P3 (FoxP3), principal marcador de células Treg, nas diferentes formas de hanseníase, com e sem reação hansênica. A expressão de células Treg foi investigada utilizando o método de ensaio de imunoabsorção enzimática (ELISA) para medir o marcador FoxP3 em plasma não estimulado, assim como no sobrenadante de células mononucleares do sangue periférico (PBMC) após incubação com antígeno sonicado de M. leprae (MLSA) durante 72 horas. Após estimulação com MLSA, um forte aumento da expressão de FoxP3 foi observado em pacientes com lepra multibacilar (LM) com reação do tipo 1 e 2. No entanto, o grupo que apresentou maior expressão de FoxP3 foi o de pacientes com reação do tipo 1, com cerca de seis vezes mais pacientes do que o de indivíduos saudáveis e duas vezes mais que os com reação do tipo 2. Portanto, este estudo sugere que o Treg desempenha um papel importante na etiopatogenia das reações hansênicas, regulando o processo inflamatório agudo exacerbado e evitando lesões graves e incapacitantes. Este estudo também demonstra que o ELISA, uma tecnologia simples e acessível, pode ser usado na medição de rotina do marcador FoxP3.
ABSTRACT Although regulatory T cells (Treg) are important in the immune regulation of various pathological processes, the role of these cells in the immunology of leprosy still needs to be better evaluated. The objective of this work is to quantify the FoxP3, the main marker of Treg cells, in the different forms of leprosy, with and without leprosy reaction. Expression of Treg cells was investigated using the enzyme-linked immunosorbent assay (ELISA) method to measure the FoxP3 marker in unstimulated plasma as well as in the peripheral blood mononuclear cells (PBMC) supernatant after incubation with M. leprae sonicated antigen (MLSA) for 72 hours. After stimulation with MLSA, a strong increase in FoxP3 expression was observed in multibacillary leprosy (MB), type 1 leprosy reaction (T1R) and type 2 leprosy reaction (T2R) patients. However, the group that presented the highest expression of FoxP3 was T1R, about six times more than that of healthy individuals and twice as many as T2R patients. Thus, this study suggests that Treg plays an important role in the etiopathogenesis of leprosy reactions, regulating the exacerbated acute inflammatory process, avoiding severe and incapacitating lesions. This study also demonstrates that ELISA, a simple and affordable technology, can be used in routine measurement of the FoxP3 marker.
RESUMEN Aunque las células T reguladoras sean importantes en la regulación inmunológica de varios procesos patológicos, su papel en la inmunología de la enfermedad de Hansen, o lepra, todavía necesita ser correctamente evaluado. El objetivo de este trabajo es cuantificar el factor de transcripción nuclear P3 (FoxP3), principal marcador de células Treg, en las diferentes formas de lepra, con y sin reacción leprosa. La expresión de células Treg fue investigada con el ensayo por inmunoabsorción ligado a enzimas (ELISA) para medición del marcador FoxP3 en plasma no estimulado así como en el sobrenadante de células mononucleares de sangre periférica (PBMC) luego de incubación con antígeno sonicado de M. leprae (MLSA) por 72 horas. Después de estimulación con MLSA, un fuerte aumento en la expresión de FoxP3 fue observado en pacientes con lepra multibacilar (MB) con reacción del tipo 1 y 2. Sin embargo, el grupo que presentó mayor expresión de FoxP3 fue el grupo de pacientes con reacción tipo 1, con alrededor de seis veces más pacientes que el de sujetos sanos y dos veces más que con los de reacción tipo 2. Por consiguiente, este estudio sugiere que Treg desempeña un papel importante en la etiopatogenia de las reacciones leprosas, regulando el proceso inflamatorio agudo agravado y evitando daños graves e incapacitantes. Este estudio también demuestra que ELISA, una técnica simple y accesible, puede ser usada en la medición de rutina del marcador FoxP3.

Abstract: Several microbiological indicators of soil quality present high sensitivity, but little is known about the influence of topographic factors on them. This work aimed to evaluate variability of biological indicators of soil quality across a hillslope under native forest and the influence of topographic factors on them. Four positions on a hillslope were evaluated. Activity of the enzymes β-glucosidase, acid phosphatase, urease and fluorescein diacetate (FDA) hydrolysis were determined, as well as basal and substrate-induced respiration, and density of microorganisms: total bacteria, total fungi, actinobacteria, phosphate solubilizers, ammonifiers, native rhizobia, free-living N2-fixing bacteria, spores of arbuscular mycorrhizal fungi and percentage of root colonization by arbuscular mycorrhizal fungi. Activity and density of microorganisms were correlated with topographic factors. The relation of these factors to the variations of the evaluated indicators was determined using the random forest algorithm. Microbiological indicators varied according to the hillslope positions. The indicators urease, basal respiration, spore density, mycorrhizal colonization, total bacteria and fungi, phosphate solubilizers, and free-living N2-fixing bacteria detected in JNFB and FAM culture medium did not vary with terrain attributes and were therefore more indicated in cases of topographic variations. This and future studies can help to select the best microbiological indicators for different conditions.

RESUMO A tangerineira ‘Dancy’ apresenta características desejáveis como cor, aroma, sabor e ampla aceitação pelo mercado consumidor. Trata-se de uma variedade bastante produzida no estado da Paraíba, porém, suscetível a mancha marrom de alternária (MMA), causada por Alternaria f. sp. citri, responsável por comprometer a qualidade dos frutos nas regiões produtores de tangerineiras. E para prevenir essa doença, tem-se ampliado pesquisas com o uso de produtos alternativos. Portanto, objetivo do trabalho foi determinar o potencial do extrato de Caesalpinia ferrea e da Quitosana® no manejo da MMA em frutos de tangerineira ‘Dancy’. Os frutos foram desinfestados e imersos em extrato de C. ferrea nas concentrações 0; 10; 100; 500 e 1000 µg/mL e a Quitosana® a 0,5; 1,0; 1,5 e 2,0 g/L. Após 24 h da aplicação dos produtos, foi inoculado o fungo A. alternata (105 esporos/mL). No segundo dia após a inoculação, foram realizadas diariamente a avaliação da severidade (AACPD). Para as análises das atividades da fenilalanina amônia-liase, peroxidase e polifenoloxidase foram retiradas três frutos/repetição no primeiro e no décimo segundo dia do experimento. O delineamento foi em DIC com 9 tratamentos (concentrações dos produtos) e 5 repetições com 5 frutos, constituindo uma unidade experimental com 225 frutos. Foi realizada análise de regressão e o teste de Dunnett (P ≤ 0,05) no programa SAS® System 9.3. As concentrações 500 µg/mL do extrato de C. ferrea e 1,5 g/L de Quitosana® reduziram a severidade da mancha marrom de alternária e promoveram altas atividades das enzimas em frutos de tangerineira ‘Dancy’.
ABSTRACT The tangerine ‘Dancy’ has desirable characteristics such as color, aroma, flavor and wide acceptance by the market. This variety is highly produced in the state of Paraíba but is susceptible to Alternaria brown spot (MMA), caused by Alternaria alternata, responsible for compromising the fruit quality in tangerine producing regions. To prevent this disease, research using alternative products has been expanded. Therefore, the objective of this study was to determine the potential of the extract of Caesalpinia ferrea and Chitosan® in the management of MMA in fruits of ‘Dancy’ tangerine. The fruits were disinfested and immersed in extract of C. ferrea at concentrations of 0, 10, 100, 500 and 1000 µg/mL, and Chitosan® at 0.5, 1.0, 1.5 and 2.0 g/L. After 24 h of the application of products, the fungus A. alternata was inoculated (105 spores/mL). From the second day after inoculation (DAI), severity (AUDPC) was daily evaluated. For analysis of the activities of phenylalanine ammonia-lyase, peroxidase and polyphenol oxidase, three fruits/replicate were taken on the first and twelfth days of the experiment. The design was completely randomized including 9 treatments (concentrations of products) and 5 replicates with 5 fruits, which constituted an experimental unit with 225 fruits. Regression analysis and Dunnett’s test (P ≤ 0.05) were performed by using SAS® System 9.3 software. The concentrations of 500 µg/mL of C. ferrea extract and 1.5 g/L Chitosan® reduced the severity of Alternaria brown spot and promoted high activities of the enzymes in ‘Dancy’ tangerine fruits.
