Phytochemical study of Psychotria schlechtendaliana (Rubiaceae) aerial parts resulted in the isolation of alkaloids 4 N oxide harmane (1) and strictosidinic acid (2), and the terpenoids sitosterol, estigmasterol, α-amyrin, β-amyrin and betulinic acid. The crude extract (CE), its fractions (hexane FH, chloroform FC, ethyl acetate FEA, hydromethanolic FHM, alkaloidal chloroform FCOH, alkaloidal aqueous FAq) and majority alkaloid (1) were investigated for their antiproliferative potential against nine human tumor cells lines and one non-tumoral human cell line (HaCat). CE, FH and FEA fractions exhibited strong growth inhibition for ovary cells (OVCAR-3, GI50 = 5.89; 1.36 and 6.59 μg mL-1, respectively) and FC, FH and FAq fractions showed potent activity on the growth of leukemia cell lines (K562, GI50 = 1.92; 7.23 and 8.81 μg mL-1, respectively). Compound 1 exhibited selective antiproliferative activity to breast cancer (MCF-7, GI50 = 32.7 μg mL-1) and was non-toxic to HaCat cells. To evaluate the anti-inflammatory effect, models of ear edema induced by croton oil and the enzyme myeloperoxidase (MPO) were used. FH and FC fractions exhibited anti-inflammatory effect and reduce ear edema compared to the control group. These fractions showed results superior to those exhibited by indomethacin (75.1%), FH (97.5%) and FC (95.5%) fractions. Abstract This study aimed to investigate the antioxidant and anti-inflammatory properties of quercetin on the cellular components of the Enteric Nervous System in the ileum of rats with arthritis. Rats were distributed into five groups: control (C), arthritic (AIA), arthritic treated with ibuprofen (AI), arthritic treated with quercetin (AQ) and arthritic treated with both ibuprofen and quercetin (AIQ). The ileum was processed for immunohistochemical techniques for HuC/D, calcitonin gene-related peptide, and vasoactive intestinal polypeptide. Measurements in histological sections, chemiluminescence assays, and total antioxidant capacity were also performed. Rheumatoid arthritis resulted in a decrease in neuronal density, yet neuroplasticity mechanisms were evident through observed changes in varicosities size and neuronal area compared to the control group. Reduced paw edema and neuroprotective effects were predominantly noted in both plexuses, as evidenced by the increased density preservation of HuC/D-IR neurons in the AIQ group. The increase of lipoperoxidation levels and paw edema volume in the AQ group was observed compared to the arthritic, whereas the AIQ group mainly showed similar results to those observed in the control. Pisonia aculeata L. (Nyctaginaceae) is a shrub widely used in Brazilian folk medicine, where the roots are used as purging and barks and leaves have anti-inflammatory properties. The aim of this study is to evaluate the antiedematogenic activity of crude extract and derived fractions from stems of P. aculeata and the acute toxicity of crude methanolic extract. From methanolic extract from stems of P. aculeata were obtained steroids, triterpenoids and nitrogenous compounds. All fractions tested show inhibitory activity of mice edema ear induced by croton oil, mainly hexane and chloroform fractions. Crude methanolic extract was also active, inhibiting 27.9% of the swelling occasioned by croton oil. In addition, treatment of mice with the crude extract of the stems of P. aculeata in the acute toxicity test did not cause death of animals or change in behavioral parameters, indicating that the extract may be considered non-toxic.

Pisonia aculeata L. (Nyctaginaceae) es un arbusto ampliamente utilizado en la medicina popular de Brasil, donde las raíces se utilizan como purgante, mientras que la corteza y hojas por sus propiedades anti-inflamatorias. El objetivo de este estudio es evaluar la actividad antiedematogénica del extracto crudo y las fracciones derivadas de los tallos de P. aculeata y la toxicidad aguda del extracto metanólico crudo. Del extracto metanólico obtenido de los tallos de P. aculeata se obtuvieron esteroides, triterpenoides y compuestos nitrogenados. Todas las fracciones probadas mostraron antiinflamatoria actividad en el edema de oreja inducido por aceite de crotón en ratones, sobre todo las fracciones de hexano y cloroformo. El extracto metanólico crudo también fue activo, inhibiendo el 27,9% de la inflamación ocasionada por el aceite de crotón. Además, el extracto crudo de los tallos de P. aculeata en el ensayo de toxicidad aguda no causaron la muerte de los animales o el cambio en su comportamiento, lo que indica que el extracto puede ser considerado como no tóxico.

O estudo químico de Aloysia gratissima (Gillies & Hook.) Tronc. e Aloysia virgata (Ruiz & Pav.) Pers., Verbenaceae, resultou no isolamento de quatorze substâncias. Suas estruturas foram determinadas com base nos dados de IV, RMN de ¹H e de 13C uni e bidimensionais e comparação com dados da literatura. A atividade antibacteriana do extrato bruto das folhas e caules e das frações de A. virgata, bem como as substâncias hoffmaniacetona e seu monoacetato, verbascosídeo e arenariosídeo, foram avaliadas através do método de bioautografia e a atividade antiedematogênica foi avaliada pelo modelo de edema de orelha induzido para os extratos brutos e frações metanólicas de A. gratissima e A. virgata..

Phytochemical study of Aloysia gratissima (Gillies & Hook.) Tronc. e Aloysia virgata (Ruiz & Pav.) Pers., Verbenaceae, afforded fourteen compounds. The structures were established using IR, 1D and 2D NMR and by comparison of its spectroscopic data to those of literature. The antibacterial activity of the crude extract from the leaves and branches and the fractions from the crude extract of A. virgata, besides the isolated compounds hoffmaniacetone, hoffmaniacetone monoacetate, verbascoside and arenarioside were evaluated by means of bioautography and the antiedematogenic activity was evaluated using induced ear oedema model to the crude extracts and methanol fractions for A. gratissima and A. virgata.

O extrato etanólico das folhas de Paullinia elegans Cambess., Sapindaceae, e as frações n-hexano, clorofórmio, acetato de etila e hidroetanólica, obtidas de seu fracionamento, foram avaliadas quanto às suas atividades anti-edematogênica e sequestradora de radicais livres. O extrato etanólico e a fração hexano produziram inibição significativa (74,4 e 76,0%, respectivamente) do edema da orelha induzido pelo óleo de cróton em ratos, em doses de 5 mg/orelha. As frações acetato de etila e hidroetanólica mostraram atividade sequestradora de radicais livres no ensaio de DPPH, com IC50 de 36,7 e de 30,1 µg/mL, respectivamente. O fracionamento dos extratos pelo uso de métodos cromatográficos resultou no isolamento do epifriedelanol, ácido 3-O-acetil oleanólico, mistura do stigmasterol 3-β-O-glucopiranosídeo e sitosterol 3-β-O-glucopiranosídeo, canferol, canferol 3,7-O-α-diramnopiranosídeo, canferol 3-O-α-ramnopiranosídeo e 2-O-metil-chiro-inositol. Os compostos foram identificados com base na comparação de seus dados espectroscópicos de RMN com os da literatura.

Ethanol extract of the leaves of Paullinia elegans Cambess., Sapindaceae, and its hexane, chloroform, ethyl acetate, and hydroethanol fractions were evaluated for their antiedematogenic and free radical scavenging activities. The ethanol extract and the hexane fraction produced statistically significant inhibition (74.4 and 76.0%, respectively) of the ear edema induced by croton oil in mice, observed at doses of 5 mg/ear. The ethyl acetate and hydroethanol fractions showed significant radical scavenging effect in the DPPH assay, with IC50 of 36.7 and 30.1 µg/mL, respectively. Fractionation of the extracts through chromatographic methods afforded epifriedelanol, oleanolic acid 3-O-acetyl, a mixture of stigmasterol 3-β-O-glucopyranoside and sitosterol 3-β-O-glucopyranoside, kaempferol 3,7-O-α-dirhamnopyranoside, kaempeferol-3-O-α-rhamnopyranoside and 2-O-methyl-chiro-inositol. The compounds were identified on the basis of their NMR spectral data and comparison with those of literature.

Eugenia caryophyllata, popularmente conhecida como "cravo-da-índia", cresce naturalmente na Indonésia e é cultivada em várias partes do mundo, incluindo o Brasil. O cravo-da-índia é utilizado em culinária, em farmácia, perfumaria e cosméticos. O óleo essencial extraído do cravo-da-índia cujo principal componente é o eugenol tem sido utilizado em odontologia como anti-séptico e analgésico. O objetivo deste estudo foi avaliar as atividades antiinflamatória e antinociceptiva do eugenol de uso odontológico, administrado oralmente, em modelos experimentais in vivo. A atividade antiinflamatória do eugenol foi avaliada através do volume de exsudato e migração leucocitária no teste de pleurisia e do edema de pata de rato induzido pela carragenina. A atividade antinociceptiva foi avaliada através dos testes de contorções induzidas pelo ácido acético e da placa quente. O eugenol (200 e 400 mg/kg) reduziu o volume de exsudato pleural sem interferir na contagem de leucócitos totais presentes na pleura. Na dose de 200 mg/kg, o eugenol inibiu significativamente o edema de pata, 2-4 h após a injeção do agente flogístico. No teste da placa quente, a administração do eugenol (100 mg/kg) mostrou atividade significativa à reação de desconforto-tempo dependente, avaliada como a latência da resposta, inibida pela meperidina. Eugenol na doses de 50, 75 e 100 mg/kg apresentou efeito antinociceptivo significativo no teste de contorções abdominais induzidas pelo ácido acético em comparação com o grupo controle. Os dados obtidos indicam que o eugenol apresenta atividade antiinflamatória e antinociceptiva periférica.

Eugenia caryophyllata, popular name "clove", is grown naturally in Indonesia and cultivated in many parts of the world, including Brazil. Clove is used in cooking, food processing, pharmacy; perfumery, cosmetics and the clove oil (eugenol) have been used in folk medicine for manifold conditions include use in dental care, as an antiseptic and analgesic. The objective of this study was evaluated the anti-inflammatory and antinociceptive activity of eugenol used for dentistry purposes following oral administration in animal models in vivo. The anti-inflammatory activity of eugenol was evaluated by inflammatory exudates volume and leukocytes migration in carrageenan-induced pleurisy and carrageenan-induced paw edema tests in rats. The antinociceptive activity was evaluated using the acetic acid-induced writhing and hot-plate tests in mice. Eugenol (200 and 400 mg/kg) reduced the volume of pleural exudates without changing the total blood leukocyte counts. At dose of 200 mg/kg, eugenol significantly inhibited carrageenan-induced edema, 2-4 h after injection of the flogistic agent. In the hot-plate test, eugenol administration (100 mg/kg) showed unremarkable activity against the time-to-discomfort reaction, recorded as response latency, which is blocked by meperidine. Eugenol at doses of 50, 75 and 100 mg/kg had a significant antinociceptive effect in the test of acetic-acid-induced abdominal writhing, compared to the control animals. The data suggest that eugenol possesses anti-inflammatory and peripheral antinociceptive activities.
