Canonical correlation between clogging agents and performance of drippers operating with aquaculture effluents

RESUMO A correlação canônica é usada para correlacionar grupos de variáveis distintas. O objetivo deste estudo foi utilizar análise de correlação canônica para avaliar a interação entre atributos de qualidade da água causadores de entupimento e o desempenho hidráulico de gotejadores não autocompensantes operando com diluições de efluente de aquicultura em água de poço. O experimento foi conduzido em delineamento inteiramente casualizado, em esquema de parcelas subsubdivididas, com três repetições, com cinco diluições nas parcelas (Diluição 1: 100% de efluente de aquicultura; Diluição 2: 75% de efluente de aquicultura + 25% de água de poço; Diluição 3 : 50% efluente de aquicultura + 50% água de poço; Diluição 4: 25% efluente de aquicultura + 75% água de poço; e Diluição 5: 100% água de poço), três emissores nas subparcelas (ST (1,6 L h-1) , SL (1,6 L h-1) e NJ (1,7 L h-1)), e três tempos de operação nas subsubparcelas (início do experimento, 80 e 160 horas, em que 0 horas de operação significa o início das avaliações). As análises estatísticas realizadas foram matriz de correlação e análise canônica. A análise de correlação canônica indicou que quando as concentrações dos atributos aumentam, o desempenho hidráulico dos gotejadores diminui. A interação desse grupo de atributos indica que o entupimento dos gotejadores que operam com as cinco diluições de efluente de aquicultura em água de poço ocorre pela ação combinada de agentes físicos, químicos e biológicos. ABSTRACT The canonical correlation is used to correlate groups of distinct variables. The objective of this study was to use canonical correlation analysis to evaluate the interaction between water quality attributes clogging-causing and the hydraulic performance of non-pressure compensating drippers operating with dilutions of aquaculture effluent in well water. The experiment was in a completely randomized design in a split-split-plot scheme with three replicates, with five dilutions in the plots (Dilution 1: 100% aquaculture effluent; Dilution 2: 75% aquaculture effluent + 25% well water; Dilution 3: 50% aquaculture effluent + 50% well water; Dilution 4: 25% aquaculture effluent + 75% well water; and Dilution 5: 100% well water), three emitters in the sub-plots (ST (1.6 L h-1), SL (1.6 L h-1) and NJ (1.7 L h-1)), and three operating times in the sub-subplots (0, 80 and 160 hours, in which 0 hours operating time means the start of evaluations). ABSTRACT Waste pickers constitute a marginalized demographic engaged in the collection of refuse, facing considerable occupational hazards that heighten their susceptibility to contract infectious diseases. Moreover, waste pickers contend with societal stigmatization and encounter barriers to accessing healthcare services. To explore the viral profile of waste pickers potentially linked to their occupational environment, we conducted a metagenomic analysis on 120 plasma specimens sampled from individuals employed at the Cidade Estrutural dumpsite in Brasilia city, Brazil. In total, 60 blood donors served as a comparative control group. Specimens were pooled and subjected to Illumina NextSeq 2000 sequencing. Viral abundance among waste pickers revealed the presence of significant pathogens, including HIV, HCV, and Chikungunya, which were not detected in the control group. Additionally, elevated levels of anelloviruses and Human pegivirus-1 were noted, with a comparable incidence in the control group. These findings underscore the utility of metagenomics in identifying clinically relevant viral agents within underserved populations. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. RESUMO: A esporotricose é uma micose subcutânea causada por fungos do complexo Sporothrix, e no Brasil a principal espécie relatada é Sporothrix brasiliensis da qual o transmissor é o gato doente. Embora sua ocorrência tenha aumentado no estado da Paraíba, Brasil, desde 2016, os dados sobre a doença neste estado são limitados. Diante disso, esta pesquisa teve por objetivo identificar, molecularmente, isolados de Sporothrix spp. procedentes de felinos domésticos de cidades da Paraíba, e dessa maneira expandir a compreensão da enfermidade no estado. Foram analisadas 39 amostras, obtidas de lesões cutâneas de felinos domésticos, oriundos das seguintes cidades paraibanas: João Pessoa, Pilões, Patos, Areia, Bananeiras e Guarabira. Realizou-se análise citológica, para triagem das amostras, a seguir cultura fúngica, e posteriormente a caracterização molecular dos isolados, utilizando a Reação em Cadeia da Polimerase (PCR), espécie-específica ou sequenciamento parcial do gene calmodulina. Todos os isolados foram identificados molecularmente como S. brasiliensis. O sequenciamento demonstrou 100% de similaridade com a cepa S. brasiliensis CBS 120339. Contudo, conclui-se que nas áreas do estudo a espécie envolvida em casos de esporotricose felina é S. brasiliensis, sua presença na Paraíba demonstra a disseminação do agente em regiões distantes dos epicentros no Brasil, alertando para a possível ocorrência de surtos zoonóticos, semelhantes aos encontrados nas regiões Sul e Sudeste do país. ABSTRACT: Sporotrichosis is a subcutaneous mycosis caused by fungus of the Sporothrix complex, and in Brazil the main species reported is Sporothrix brasiliensis, of which the diseased cat is the transmitter. Although, its occurrence has increased in the state of Paraíba, Brazil, since 2016, data on the disease in this state are limited. Therefore, this research aimed to identify molecularly isolates of Sporothrix spp. from domestic cats from cities in Paraíba, and in this way to expand the understanding of the disease in the state. Thirty-nine samples were analyzed, obtained from skin lesions of domestic felines, from the following cities in Paraíba: João Pessoa, Pilões, Patos, Areia, Bananeiras and Guarabira. Cytological analysis was performed to screen the samples, followed by fungal culture, and the molecular characterization of the isolates was performed, using the species-specific Polymerase Chain Reaction (PCR) or partial sequencing of the calmodulin gene. All isolates were identified as S. brasiliensis. The sequencing showed 100% similarity to the S. brasiliensis CBS 120339 strain. In view of this, it is concluded that in the study areas the species involved in cases of feline sporotrichosis is S. brasiliensis, its presence in Paraíba demonstrated the spread of the agent in regions distant from the epicenters in Brazil, alerting to the possible occurrence of zoonotic outbreaks similar to those found in the South and Southeast regions of the country. In addition, it highlights the emerging role of felines in the transmission of sporotrichosis in new endemic areas of Brazil. Abstract Healthcare workers present an increased risk of contagion for the SARS-CoV-2 virus due to their labor exposure. Here, we describe the clinical, laboratory, and immunological characteristics of healthcare workers, before vaccine application, exposed to SARS-CoV-2-infected patients. We collected sociodemographic, clinical, and laboratory information from 50 professionals who worked during the COVID-19 pandemic at the Clinical Hospital of the Northwest in Brazil. The results showed that most workers are women, over 50 years old, and worked as nursing technicians. Approximately 56% of workers were positive for a previous infection by RT-PCR and/or anti-SARS-CoV-2-immunoglobulin tests. Increased levels of hematocrit, neutrophils, NK lymphocytes, and fibrinogen, were found in positive healthcare workers, suggesting a light inflammatory status. The immunological findings showed an increase in IL-17 production and a Th2/Th17/Th22 profile followed by high serology for anti-SARS-CoV-2 IgM and IgG. Those data reveal the importance of studies with healthcare workers to investigate if the continuous exposition to the virus may result in chronic activation of the immune system and/or pulmonary inflammation in this target group. Abstract Different degrees in the biological activities of Canavalia rosea had been previously reported . In this study, our group assessed the cardioprotective effects of the ethyl acetate fraction (EAcF) of the Canavalia rosea leaves. Firstly, it was confirmed, by in vitro approach, that the EAcF has high antioxidant properties due to the presence of important secondary metabolites, as flavonoids. In order to explore their potential protector against cardiovascular disorders, hearts were previously perfused with EAcF (300 μg.mL-1) and submitted to the global ischemia followed by reperfusion in Langendorff system. The present findings have demonstrated that EAcF restored the left ventricular developed pressure and decreased the arrhythmias severity index. Furthermore, EAcF significantly increased the glutathiones peroxidase activity with decreased malondialdehyde and creatine kinase levels. EAcF was effective upon neither the superoxide dismutase, glutationes reductase nor the catalase activities. In addition, the Western blot analysis revealed that ischemia-reperfusion injury significantly upregulates caspase 3 protein expression, while EAcF abolishes this effect. These results provide evidence that the EAcF reestablishes the cardiac contractility and prevents arrhythmias; it is suggested that EAcF could be used to reduce injury caused by cardiac reperfusion. Abstract The Santos Basin (SB) is the main petroliferous basin in the Brazilian continental margin and one of the most studied marine areas in Brazil. However, historical data suggest that new efforts should be carried out to acquire quantitative biological data, especially in the deep sea, to establish the baseline of essential ocean variables in different ecosystems for future monitoring programs. The Brazilian energy company Petrobras planned and executed 24 oceanographic cruises over a period of 2 years to assess the benthic (SANSED cruise) and pelagic (SANAGU cruise) systems of the SB (356 days at sea in 2019 and 2021/2022). These efforts were part of the Santos Project, which comprised a comprehensive environmental study aimed at investigating benthic and pelagic variables to characterize ecology, biogeochemistry, thermohaline properties of water masses, and ocean circulation patterns, geomorphology, and sedimentology, as well as organic and inorganic chemistry. Here we present the detailed sampling designs and the field methods employed on board, during the SB scientific cruises. All sampling protocols were based on standardized approaches. For the benthos analyses, triplicate sediment samples were performed using a GOMEX-type box corer (0.25 m²) or a large modified Van Veen grab (0.75 m²) at 100 stations ranging from 25 to 2400 m depth. At each station, 25 geochemical and physico-chemical parameters were analyzed in addition to micro-, meio-, and macrofauna and living foraminifera samples. For the pelagic system, 60 stations were selected to investigate the plankton community, ranging in size from pico- to macroplankton, through vertical, horizontal, and oblique net hauls (20, 200, and 500 μm mesh size), as well as 25 biogeochemical parameters collected with an aid of a CTD-rosette sampler. Part of this scientific information also serves the Regional Environmental Characterization Project (PCR-BS) in support of Petrobras’ Santos Basin drilling licensing process led by the Brazilian Environmental Agency – IBAMA. RESUMO: Pombos urbanos são conhecidos pela sua capacidade de carrear e disseminar diversos agentes zoonóticos. Estudos tem sugerido que pombos são também relevantes na disseminação de estirpes resistentes a múltiplas drogas. No presente estudo, pombos no ambiente de um hospital veterinário foram amostrados em três diferentes períodos e testados para a presença de Escherichia coli patogênica, Salmonella spp., Staphylococcus spp. e Clostridioides (Clostridium) difficile. Isolados de E. coli de 19 pombos (40.4%) foram positivos para o gene codificador da toxina EAST1. O gene codificador de intimina (eae) do patotipo E. coli enteropatogênica foi encontrada em um isolado (2.1%). Salmonella spp. foi encontrada em nove pombos (19.1%), sendo todos isolados do primeiro período de captura (P < 000.1). S. Typhimurium foi isolado de seis animais e S. Heidelberg de três. A tipagem molecular de isolados de Salmonella spp. por ERIC-PCR demonstrou que oito estirpes possuíam alta similaridade genética entre si, sugerindo a ocorrência de um surto de salmonelose nos animais carreadores. Vinte Staphylococcus (42.5%) foram isolados de 18 animais (38.3%). Oito diferentes espécies foram detectadas, sendo S. xylosus a mais frequente. Duas estirpes de C. difficile não-toxigênica (4.3%) foram isoladas. Uma estirpe de S. Typhimurium, uma de S. aureus e um isolado de C. difficile foram classificados como resistentes a múltiplas drogas antimicrobianas. O presente estudo sugere que pombos capturados no ambiente do hospital veterinário podem atuar como reservatórios e disseminadores de bactérias patogênicas e envolvidas em infecção hospitalar, incluindo E. coli diarreiogênica e Staphylococcus sp., C. difficile e Salmonella spp multirresistente.
ABSTRACT: Pigeons are known for their capacity to harbor and spread several zoonotic agents. Studies have suggested that pigeons are also relevant disseminators of multidrug-resistant strains. In this study, pigeons surrounding a veterinary hospital were sampled and tested for the presence of pathogenic Escherichia coli, Salmonella spp., Staphylococcus spp., and Clostridioides (Clostridium) difficile. E. coli isolates from 19 (40.4%) pigeons tested positive for the E. coli heat-stable enterotoxin 1 (EAST1)-encoding gene. The intimin-encoding gene (eae) of enteropathogenicE. coli (EPEC) was found in one isolate (2.1%). Salmonella spp. were found in nine (19.1%) pigeons, all from the first capture event (P < 000.1). S. Typhimurium and S. Heidelberg were isolated from six and three pigeons, respectively. Enterobacterial repetitive intergenic consensus (ERIC-PCR) of the Salmonella spp. isolates suggested that eight of the nine strains had a high genetic similarity, supporting the hypothesis of an outbreak of salmonellosis in these pigeons. Twenty (42.5%) staphylococcal isolates were recovered from 18 (38.3%) pigeons. Eight different species were detected, with S. xylosus being the most frequent. Two (4.3%) C. difficile strains were isolated. Three isolates, one each of S. Typhimurium, S. aureus, and C. difficile, were classified as multidrug-resistant strains. The present research suggested that pigeons residing in urban areas can act as reservoirs and disseminators of pathogenic bacteria, including nosocomial pathogens, such as diarrheagenicE. coli and multidrug-resistant Staphylococcus spp., C. difficile, and Salmonella spp.

Isolation and antimicrobial resistance of coagulase-negative staphylococci recovered from healthy tortoises in Minas Gerais, Brazil

RESUMO: Nas últimas décadas, houve um aumento global na adoção de répteis como animais de companhia, principalmente tartarugas e jabutis. Considerando a popularidade dos répteis como animais de estimação no Brasil e a notável falta de dados sobre estafilococos potencialmente patogênicos nesses animais, o objetivo deste estudo foi isolar e avaliar a susceptibilidade antimicrobiana de espécies estafilocócicas de jabutis (Chelonoidis carbonaria) saudáveis no Brasil. Durante um período de 12 meses (fevereiro de 2019 a fevereiro de 2020), suabes cloacais de 66 jabutis saudáveis foram coletados no Centro de Triagem de Animais Silvestres em Belo Horizonte, Minas Gerais, Brasil. Os suabes foram plaqueados em ágar manitol salgado para isolamento de estafilococos e a identificação das espécies foi realizada usando MALDI-TOF MS. A susceptibilidade aos antimicrobianos foi investigada pelo método de difusão em disco, e a presença do gene mecA foi investigada por PCR para detectar resistência à meticilina. Dos animais testados, 72,7% foram positivos para o isolamento estafilocócico. Todos os isolados eram estafilococos coagulase-negativos (CoNS), sendo Staphylococcus sciuri (81,3%) e S. xylosus (12,5%) as espécies mais frequentemente isoladas. A maioria dos isolados (56%) foi resistente a pelo menos um antimicrobiano. Alta frequência de resistência foi observada para penicilina (35,5%) e tetraciclina (29,1%). Todas as estirpes foram sensíveis à cefoxitina, cloranfenicol, ciprofloxacina, eritromicina e gentamicina. Todos os isolados foram negativos para o gene mecA. O presente trabalho sugere que jabutis saudáveis são colonizados principalmente por CoNS, especialmente S. sciuri. Metade dos isolados foram resistentes a pelo menos um antimicrobiano, levantando questões sobre o possível papel desses animais como reservatórios de genes de resistência aos antimicrobianos.
ABSTRACT: In the last few decades, there has been a global increase in the adoption of reptiles as companion animals, mainly turtles and tortoises. Considering the popularity of reptiles as pets in Brazil, and a notable lack of data about potentially pathogenic staphylococci in these animals, this study isolated and evaluate the antimicrobial susceptibility of staphylococcal species from healthy tortoises (Chelonoidis carbonaria) in Brazil. During a 12-month period (February 2019 to February 2020), cloacal swabs from 66 healthy tortoises were collected at the Wild Animals Screening Center in Belo Horizonte, Minas Gerais, Brazil. The swabs were plated onto mannitol salt agar for staphylococci isolation, and species identification was performed using MALDI-TOF MS. Antimicrobial susceptibility was investigated using the disk diffusion method, and the presence of the mecA gene was investigated by PCR to detect methicillin resistance. Of the tested animals, 72.7% were positive for staphylococcal isolation. All isolates were coagulase-negative staphylococci (CoNS), and Staphylococcus sciuri (81.3%), and S. xylosus (12.5%) were the most frequently isolated species. The majority of the isolates (56%) were resistant to at least one antimicrobial agent. A high frequency of resistance was observed for penicillin (35.5%) and tetracycline (29.1 %). All strains were susceptible to cefoxitin, chloramphenicol, ciprofloxacin, erythromycin, and gentamicin. All isolates were negative for the mecA gene. The present work suggests that healthy tortoises are mainly colonized by CoNS, especially S. sciuri. Half of the isolates were resistant to at least one antimicrobial, raising questions regarding the possible role of these animals as reservoirs of antimicrobial resistance genes.

ABSTRACT The Big Data phenomenon has imposed maturity on companies regarding the exploration of their data, as a prerogative to obtain valuable insights into their clients and the power of analysis to guide decision-making processes. Therefore, a general approach that describes how to extract knowledge for the execution of the business strategy needs to be established. The purpose of this research paper is to introduce and evaluate the implementation of a process for the experimental development of Data Mining (DM), AI and Data Science applications aligned with the strategic planning. A case study with the proposed process was conducted in a federal educational institution. The results generated evidence showing that it is possible to integrate a strategic alignment approach, an experimental method, and a methodology for the development of DM applications. Data Mining (DM) and Data Science (DS) applications also present the risks of other Information Systems, and the adoption of strategy-driven and scientific method processes are critical success factors. Moreover, it was possible to conclude that the application of the scientific method was facilitated, besides being an important tool to ensure the quality, reproducibility and transparency of intelligent applications. In conclusion, the process needs to be mapped to foment and guide the strategic alignment.

Abstract Introduction: We investigated the clinical course and outcomes of patients submitted to cardiovascular surgery in Brazil and who had developed symptoms/signs of coronavirus disease 2019 (COVID-19) in the perioperative period. Methods: A retrospective multicenter study including 104 patients who were allocated in three groups according to time of positive real time reverse transcriptase-polymerase chain reaction (RT-PCR) for the SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2): group 1, patients who underwent cardiac surgery > 10 days after positive RT-PCR; group 2, patients with a positive RT-PCR within 10 days before or after surgery; group 3, patients who presented positive RT-PCR > 10 days after surgery. The primary outcome was mortality and secondary outcomes were postoperative complications, intensive care unit (ICU) length of stay, and postoperative days of hospitalization. Results: The three groups were similar with respect to age, the European System of Cardiac Operative Risk Evaluation score, and comorbidities, except hypertension. Postoperative complications and death were significantly higher in groups 2 and 3 than in group 1, and no significant difference between groups 2 and 3 was seen. Group 2 showed a high prevalence of surgery performed as an urgent procedure. Although no significant differences were observed in ICU length of stay, total postoperative hospitalization time was significantly higher in group 3 than in groups 1 and 2. Conclusion: COVID-19 affecting the postoperative period of patients who underwent cardiovascular surgery is associated with a higher rate of morbidity and mortality. Delaying procedures in RT-PCR-positive patients may help reduce risks of perioperative complications and death.

Abstract Studies evaluating the structure and diversity of bacterial communities in arid environments including the rhizosphere of local and adapted plant species are important. Therefore, we used a sequencing of the 16S ribosomal RNA gene for describing the structure and diversity of soil bacterial community in three zones: Agreste, Transition and Sertão. The bacterial community was clustered in 9,838 OTUs in Agreste, 8,388 OTUs in the transition, and 14,849 OTUs for Sertão. Among the most abundant phyla, Proteobacteria and Acidobacteria were abundant in Agreste and Sertão, respectively, while Actinobacteria were abundant in Transition and Sertão. Specific taxa of Proteobacteria, in Agreste, and Actinobacteria, in Sertão, exhibited differences according to biotic and abiotic conditions. Thus, the structure and diversity of bacterial community were different in these areas and were influenced by environmental and soil conditions.

ABSTRACT Many infants are nurtured with milk supplied by human banks, whose bacteriological and physical-chemical profiles are a major issue. We investigated the bacteriological and physical-chemical characteristics, as well as genotypic and phenotypic and profiles of Staphylococcus species isolated from 240 samples of breast milk from a bank in a teaching hospital. Dornic acidity of milk revealed that 95.4% (229/240) had acceptable limits (< 8.0 oD). Caloric intake showed a wide variation in cream content (4%), fat (4%) and energy values (559.81 Kcal/L). Staphylococcus (105/186 or 56.5%) and Enterobacter (25/186 or 13.4%) were the most prevalent genera, although other microorganisms were identified, including Klebsiella pneumoniae and Pseudomonas aeruginosa. Amoxicillin/clavulanic acid (125/157 or 79.6%), vancomycin (115/157 or 73.2%), and cephalexin (112/157 or 71.3%) were the most effective antimicrobials. High resistance rates of isolates were found to penicillin G (141/157 or 89.8%), ampicillin (135/157 or 86%), and oxacillin (118/157 or 75.2%). Multidrug resistance to ≥ 3 antimicrobials occurred in 66.2% (123/186) of the isolates. Residues of microbial multiplication inhibitory substances were found in 85% (204/240) of samples. Among the coagulase-positive-CPS and negative-CoNS staphylococci, the mecA gene was detected in 53.3% (8/15) and 75% (30/40), respectively. Genes sea, seb and sec were detected in 20% (3/15) of CPS, while tsst-1 was detected in 13.34% (2/15). In addition, 13.3% (2/15) of S. aureus were toxin-producers. Genes sea, seb and sec were detected in 90% (36/40), 5% (2/40) and 15% (6/40) CoNS, respectively. Enterotoxin production was identified in 5% (2/40) of CoNS. The identification of multidrug-resistant bacteria, staphylococci species toxin-producers harboring methicillin-resistance genes, and residues of microbial multiplication inhibitory substances reinforce the need for a continuous vigilance of milk quality offered to infant consumption by human banks.

A chemical investigation of Anaxagorea dolichocarpa Sprague & Sandwith, a member of Annonaceae family, was carried out. The ethanolic extract from the roots of this plant led, by chromatography tecniches, to isolation of the new azaphenanthrene alkaloids dolichocarpine (1) and 9-methoxyeupolauramine (2), besides the known alkaloids eupolauramine (3), 3-methoxyeupolauridine (4), eupolauramine (5) and 4-methylsampangine (6). The structures of isolated compounds were established by 1D and 2D NMR, HRESIMS, tandem MSn and IR data. The cytotoxicity of compounds 1 - 5 was evaluated against HCT-116 (human colorectal carcinoma) and L929 (murine fibroblast) cell lines.
