Abstract The objective was to evaluate the chemical composition and in vitro fermentation of spineless cactus of the genus Nopalea, F-21 (Nopalea cochenillifera Dyck), IPA-Sertânia (Nopalea cochenillifera Dyck) and Miúda (Nopalea cochenillifera Salm Dyck), in different phenophases. There was no effect (P < 0.05) of the phenological phases of spineless cactus on DM, ash, OM, EE, and CP. Varieties F-21 and Miúda presented higher values of DM and OM, whereas the CP was higher for IPA-Sertânia. The contents of NDF, ADF, and ADL, as well as the fractions of carbohydrates B2 and C were higher in the mature stage, irrespective of the variety. The Miúda variety showed higher levels of NFC and fractions A + B1 and the lower levels of pectin compared to the F-21 and IPA-Sertânia varieties, but not differ of TC to F-21. The volume of gas produced via the degradation of NFC was higher for young phenological phases. The young and intermediate stages showed a higher in vitro digestibility of DM. Based on the results, varieties IPA-Sertânia and Miúda have a high potential for use in animal feed because of their high nutritional quality. Mature cladodes showed a higher fibrous fraction and lower digestibility in all varieties. Abstract Among the potential feed additives, β-glucans are known to positively affect the growth performance, blood parameters, and intestinal microbiota of fish, even the ornamental species. Therefore, the present study evaluated the effects of the dietary supplementation of different Saccharomyces cerevisiae β-glucans concentrations (0, 0.05, 0.1, and 0.2%) in juvenile angelfish (Pterophyllum scalare) over a 42-day period. Regarding growth performance, no effects were observed on most parameters. However, 0.2% β-glucans supplementation produced higher condition factor values, indicating a better nutritional status. Furthermore, β-glucans supplementation did not affect blood parameters. Regarding intestinal microbiota, β-glucans supplementation increased the abundance of the potentially beneficial bacterial genus Phascolarctobacterium. The high abundance of bacteria from the phylum Bacteroidetes, which can degrade β-glucans, may be attributed to the increased abundance of Phascolarctobacterium spp. In addition, 0.2% β-glucans supplementation produced more operational taxonomic units and higher Sobs (observed species richness), indicating effects on the overall bacterial community structure. These results demonstrate the potential application of β-glucans as a dietary supplement to improve the performance and modulate the intestinal microbiota of angelfish. additives βglucans β glucans parameters fish Therefore 0, 0 (0 005 05 0.05 01 1 0.1 02 2 Pterophyllum scalare 42day day 42 period However 0.2 values status Furthermore Bacteroidetes βglucans, glucans, spp addition richness, richness , richness) structure ( 00 0.0 0. 4

Abstract This study compares the effects of virus-cell interactions among SARS-CoV-2 variants of concern (VOCs) isolated in Brazil in 2021, hypothesizing a correlation between cellular alterations and mortality and between viral load and transmissibility. For this purpose, reference isolates of Alpha, Gamma, Zeta, and Delta variants were inoculated into monolayers of Vero-E6 cells. Viral RNA was quantified in cell supernatants by RT‒PCR, and infected cells were analyzed by Transmission Electron Microscopy (TEM) for qualitative and quantitative evaluation of cellular changes 24, 48, and 72 hours postinfection (hpi). Ultrastructural analyses showed that all variants of SARS-CoV-2 altered the structure and function of mitochondria, nucleus, and rough endoplasmic reticulum of cells. Monolayers infected with the Delta variant showed the highest number of modified cells and the greatest statistically significant differences compared to those of other variants. Viral particles were observed in the cytosol and the cell membrane in 100 % of the cells at 48 hpi. Alpha showed the highest mean particle diameter (79 nm), and Gamma and Delta were the smallest (75 nm). Alpha and Gamma had the highest particle frequency per field at 48 hpi, while the same was observed for Zeta and Delta at 72 hpi and 24 hpi, respectively. The cycle threshold of viral RNA varied among the target protein, VOC, and time of infection. The findings presented here demonstrate that all four VOCs evaluated caused ultrastructural changes in Vero-E6 cells, which were more prominent when infection occured with the Delta variant. viruscell virus SARSCoV2 SARSCoV SARS CoV 2 SARS-CoV- (VOCs 2021 transmissibility purpose VeroE6 VeroE Vero E6 E Vero-E RTPCR RT PCR RT‒PCR TEM (TEM 7 . ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. The importance of the CTFB is evidenced by the elevated number of citations in the scientific literature in diverse areas of biology, law, anthropology, education, forensic science, and veterinary science, among others. publications problem uptodate up date classifications context exception (CTFB http//fauna.jbrj.gov.br/, httpfaunajbrjgovbr http //fauna.jbrj.gov.br/ , jbrj gov br (http://fauna.jbrj.gov.br/) 2015 Brazil 80 specialists 1 2024 133691 133 691 133,69 125138 125 138 125,13 82.3%, 823 82 3 (82.3% 102000 102 000 102,00 7.69%, 769 7 69 (7.69% 11000 11 11,00 . 3,567 3567 567 (3,56 2,292 2292 2 292 (2,29 1,833 1833 833 (1,83 1,447 1447 447 (1,44 1000 1,00 831 (83 628 (62 606 (60 520 (52 50 users science health biology law anthropology education others http//fauna.jbrj.gov.br/ faunajbrjgovbr //fauna.jbrj.gov.br (http://fauna.jbrj.gov.br/ 201 8 202 13369 13 133,6 12513 12 125,1 82.3% (82.3 10200 10 00 102,0 7.69% 76 6 (7.69 1100 11,0 3,56 356 56 (3,5 2,29 229 29 (2,2 1,83 183 83 (1,8 1,44 144 44 (1,4 100 1,0 (8 62 (6 60 52 (5 5 http//fauna.jbrj.gov.br (http://fauna.jbrj.gov.br 20 1336 133, 1251 125, 82.3 (82. 1020 0 102, 7.69 (7.6 110 11, 3,5 35 (3, 2,2 22 (2, 1,8 18 (1, 1,4 14 4 ( 82. ABSTRACT Brazilian semi-arid tropical is responsible for the production and export of fine table grapes, but the demand for phosphate fertilizers and other agricultural inputs increases the cost of production and threatens local water bodies by leaching of phosphorus (P). The objectives of this study were to quantify the organic and inorganic fractions of P and to determine chemical and microbiological attributes related to its mineralization in soils cultivated with vines in São Francisco Valley. It was hypothesized that P fractions and microbial activity would increase in areas fertilized with phosphate. Soil samples from vines were collected in cultivated areas and in fallow systems. P fractionation, macro-elements analysis, microbial biomass phosphorus (Pmic), and phosphatase enzyme activity were determined and correlated with each other and with the total proportion of synthesized glomalin. In both treatments, the inorganic P fractions exceeded the organic ones, and the total phosphorus available in those soils was associated with the most recalcitrant fractions, reducing risks of leaching of this element and contamination of water bodies. Pmic was higher in the winery areas due to the presence of phosphate fertilizers and organic matter, hence the activity of alkaline phosphatase enzymes and the production of total glomalin increased respectively in three and four evaluated areas, indicating that P-mineralized bacteria and arbuscular mycorrhizal fungi have been positively affected by the current agricultural system. semiarid semi arid grapes P. . Abstract Although Três Lagoas municipality is located in a rare and highly fragmented Atlantic Forest-Cerrado ecotone, there is scarcity of biodiversity data to guide conservation decisions. Therefore, this study aimed to compile the first bird checklist in 31 localities in this municipality to propose conservation strategies based on avian richness, conservation and migratory status. A total of 419 bird species were recorded, of which 412 were exclusive to the Atlantic Forest-Cerrado ecotone and seven to the Cerrado domain (C. minor, L. xenopterus, P. unicinctus, T. curucui, S. pileata, S. melanopis and S. luteola). According to the results, 12 species are included in the Near Threatened category, four are vulnerable (C. fasciolata, L. xenopterus, A. tricolor and S. hypoxantha) and one is endangered (U. coronata). Regarding migratory birds, 112 were recorded (95 intracontinental, 17 intercontinental). Since the majority of these species are recorded in grasslands, aquatic habitats and Seasonal Deciduous Alluvial Forest, they necessarily have highest priority for conservation and restoration. Thus, we recommend that: (1) areas 2, 11 and 24 be considered as both Important Bird Area and “Area de Proteção Ambiental”; (2) riparian forest restoration along the Paraná river act as a local ecological corridor among these areas, allowing the recolonization, genetic exchange and restoration of locally reduced populations of migratory and near/threatened bird species. ABSTRACT Alternative management measures have been used to reduce nematode population levels in affected areas. In this perspective, the objective was to evaluate the efficiency of nematicides based on fungus and bacterium, associated with poultry litter applied via ground in the management of Pratylenchus brachyurus and Heterodera glycines in soybean and corn crops in succession. The experiment was conducted in the 2018/19 growing season in the municipality of Ipameri– GO (-17°34’55.58”S−48°12’02.35”O) in naturally infested field, arranged in a randomized block design, with 10 treatments and 4 replicates, being: T1- Witness/ control, T2- Poultry litter, T3- P. lilacinum + T. harzianum, T4- B. subtilis + B. licheniformis, T5- P. chlamydosporia, T6- Abamectina, T7- P. lilacinum + T. harzianum + Poultry litter, T8- B. subtilis + B. licheniformis + Poultry litter, T9- P. chlamydosporia + Poultry litter, T10- Abamectina + Poultry litter. The poultry litter was added in the soil 10 days before planting. The plots were composed of 6 lines of 6 m, with a spacing of 0.5 m. In the soybean crop at 45 and 90 days after sowing (DAS), the fresh weight of the roots (MFR), nematological variables and productivity were evaluated. In corn, only nematological variables were evaluated at 60 DAS. The treatments using P. lilacinum + T. harzianum and B. subtillis + B. licheniformes increased the MFR and reduced the number of P. brachyurus and H. glycines at the root. P. chlamydosporia associated with poultry litter increased in productivity in soybean plants. areas perspective bacterium succession 201819 2018 19 2018/1 Ipameri 17°3455.58S−48°1202.35O 17345558S48120235O SO 17°34 55.58 S−48°12 02.35 O 17 34 55 58 S 48 12 02 35 (-17°34’55.58”S−48°12’02.35”O field design 1 replicates being T1 T Witness control T2 T3 P T4 B T5 T6 T7 T8 T9 T10 planting m 05 0 5 0. 9 DAS, DAS , (DAS) MFR, (MFR) H root plants 20181 201 2018/ 3455 58S 1202 35O 1734 17°3 5558 55.5 S4812 S−48°1 0235 02.3 3 (DAS (MFR 20 345 120 173 17° 555 55.

In recent studies, the isonicotinoylhydrazone and phthalazinylhydrazone groups were considered innovative in the development of antileishmania drugs. Molecular hybridization is a strategy for the rational development of new prototypes, where the main compound is produced through the appropriate binding of pharmacophoric subunits. Aims: To synthesize four hybrids of α- and β-lapachones, together with the isonicotinoylhydrazone and phthalazinylhydrazone groups and to determine the antileishmania activity against the promastigotic forms of L. amazonensis, L. infantum and L. major. Results: β-lapachone derivatives were more active against all tested leishmania species. βACIL (IC50 0.044μM) and βHDZ (IC50 0.023μM) showed 15-fold higher activity than amphotericin B. The high selectivity index exhibited by the compounds indicates greater safety for vertebrate host cells. Conclusion: The results of this work show that the hybrids βACIL and (3HDZ are promising molecules for the development of new antileishmania drugs.

ABSTRACT Malaria is a disease of global tropical distribution, being endemic in more than 90 countries and responsible for about 212 million cases worldwide in 2016. To date, the strategies used to eradicate this disease have been ineffective, without specific preventive measures such as vaccines. Currently, the existing therapeutic arsenal is limited and has become ineffective against the expansion of artemisinin-resistant Plasmodium, demonstrating the need for studies that would allow the development of new compounds against this disease. In this context, we studied the volatile oil obtained from rhizomes of Cyperus articulatus (VOCA), a plant species commonly found in the Amazon region and popularly used as a therapeutic alternative for the treatment of malaria, in order to confirm its potential as an antimalarial agent by in vitro and in vivo assays. We cultured Plasmodium falciparum W2 (chloroquine-resistant) and 3D7 (chloroquine-sensitive) strains in erythrocytes and exposed them to VOCA at different concentrations in 96-well microplates. In vivo antimalarial activity was tested in BALB/c mice inoculated with approximately 106 erythrocytes infected with Plasmodium berghei. VOCA showed a high antimalarial potential against the two P. falciparum strains, with IC50 = 1.21 μg mL-1 for W2 and 2.30 μg mL-1 for 3D7. VOCA also significantly reduced the parasitemia and anemia induced by P. berghei in mice. Our results confirmed the antimalarial potential of the volatile oil of Cyperus articulatus.

ABSTRACT Asian soybean rust (ASR), caused by the fungus Phakopsora pachyrhizi, is one of the most severe diseases that affect the crop. Among management strategies is the use of genotypes with horizontal or partial resistance, which are still restricted in the market but promising due to their resistance durability. The objective of this study was to evaluate the resistance of different genotypes to Asian soybean rust for future use in breeding programs. In the 2015/2016 crop season, 11 soybean genotypes from LAGER-UFU and cv. Desafio8473 (susceptibility pattern) were evaluated for artificial and natural infection by P. pachyrhizi in the field. ASR severity data were used to calculate the area under the disease progress curve (AUDPC). The percentage of affected leaf area and the AUDPC values of the evaluated genotypes ranged from 1.56 to 80.75% and from 380.62 to 975.62, respectively. The genotypes L-210, L-166, L-268, L-203, L-279, L-224, L-216 and L-218 have lower AUDPC values, indicating partial resistance, while the genotypes L-144, L-104, L-266 and cv. Desafio 8473 showed higher AUDPC values and therefore lower partial resistance or susceptibility. Even so, the genotypes L-144, L-104 and L-266 showed coefficients of disease severity progress 1.8 to 2.0-fold lower than those of cv. Desafio 8473. In conclusion, the genotypes from UFU Breeding Program (LAGER-UFU) showed potential to be used in the development of soybean cultivars resistant to P. pachyrhizi.

Abstract The aim of the present study was to compare periodontal conditions between intensive care unit (ICU) in-patients and non-ICU patients through clinical and microbiological periodontal parameters. This case-control study included 88 individuals hospitalized in ICU and 176 non-hospitalized controls. All individuals underwent a complete periodontal examination and microbiological sampling. The total bacterial load and counts of Porphyromonas gingivalis, Treponema denticola and Aggregatibacter actinomycetemcomitans were evaluated using qPCR. Data were analyzed through the Chi-square, Fisher exact, and t-Student tests, and the Spearman correlation, as appropriate. The prevalence of periodontitis was 39.7% among controls and 59.0% among ICU in-patients (OR=2.18; p=0.002). ICU in-patients had a significantly higher occurrence of cardiovascular disease (p=0.002; OR=2.20) and history of periodontal disease (p=0.031; OR=1.92) than controls. Bacterial counts of A. actinomycetemcomitans, T. denticola and P. gingivalis were significantly higher in ICU in-patients with periodontitis than in controls. The correlation between periodontal parameters and microbiological findings among cases and controls showed a significant and positive correlation between: total bacteria load and % of sites with probing depth (PD) ≥ 4 mm (cases: r=0.22 and controls: r=0.13) and P. gingivalis and % sites with bleeding on probing (BOP) (cases: r=0.22 and controls: r=0.23). Thus, ICU in-patients presented a higher prevalence of periodontitis and worse periodontal condition (higher mean plaque index, BOP, clinical attachment level ≥3 mm, and sites with PD 4 to 6 mm).

ABSTRACT Objective To verify the presence of variants in HNF1B in a sample of the Brazilian population selected according to the presence of renal cysts associated with hyperglycemia. Subjects and methods We evaluated 28 unrelated patients with clinical suspicion of HNF1B mutation because of the concomitant presence of diabetes mellitus (DM) or prediabetes and renal cysts. Genotyping was accomplished using Sanger sequencing or multiplex ligation-dependent probe amplification (MLPA). In positive cases, available relatives were recruited. Results We found two patients with HNF1B mutations. The first presented the variant p.Pro328Leufs*48(c.983delC) and had DM, renal cysts, and hypomagnesemia. The second presented a heterozygous whole gene deletion in HNF1B, DM, renal cysts, body and tail pancreatic agenesis, and hypomagnesemia; this alteration was also found in his two siblings and his father. Conclusion The recruitment of suspected cases of HNF1B gene mutations in Brazilians due to hyperglycemia and renal cysts presents two positive cases. Our cases contribute to the annotation of clinical and biochemical phenotypes of this rare form of maturity-onset diabetes of the young (MODY).

Natural naphthoquinones such as lapachol (Lp) have demonstrated promising biological activities. However, these quinones present low systemic biodisponibility due to its poor aqueous solubility. To overcome this problem, nanoemulsions (NE) formed by oily core are able to incorporate hydrophobic molecules thus enabling them to be dispersed into an aqueous phase. The present study reports the development, characterization, and physical stability of NE containing lapachol. NE formulations (F1, F2, F3, F4, and F2-Lp) were prepared using emulsion phase inversion (EPI) method and characterized in relation to droplet size, size distribution (PDI), zeta potential, and physical stability. The formulation chosen to incorporate lapachol (F2-Lp) showed droplet size in nanometric scale with homogeneous size distribution (PDI < 0.2) and negative zeta potential (about −30 mV). In addition, good physical stability of F2 and F2-Lp was demonstrated using analytical centrifugation with photometric detection where the light transmission profiles did not change throughout the dispersions. Nanoemulsion containing lapachol presented a strong reddish aspect; however, the incorporation of this naphthoquinone did not alter the main physicochemical parameters of NE formulation. The in vitro release study demonstrated a sustained release profile from NE with about 60% of lapachol released within 54 h.

This work describes the tissue culture of Plectranthus ornatus Codd. (Lamiaceae) on Murashige and Skoog (MS) medium with naphthalene acetic acid (NAA) (1.0, 2.0 and 4.0 mg L-1) and 6-benzylaminopurine (BAP) (1.0, 2.0 and 4.0 mg L-1) as growth regulators to obtain the overproduction of rosmarinic acid. The content of rosmarinic acid and its biosynthetic precursors were determined by high performance liquid chromatography (HPLC). The antioxidant activities (AA) of the extracts were also evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. The best growing condition was observed with the addition of 1.0 mg L-1 of both NAA and BAP in the culture medium yielded an increase of 94 times of rosmarinic acid comparing with the wild plant. All extracts diplayed antioxidant activity, as evidenced by the DPPH free radical scavenging assay. However, those with 2.0 mg L-1 NAA and, 1.0 mg L-1 NAA and BAP in culture medium showed the lowest EC50 (26.0 and 29.8 µg mL-1, respectively). At concentration higher than 10 µg mL-1 of rosmarinic acid it was not observed the correlation with AA, suggesting some other anti-oxidant mechanism acting.

The objective of present study was to evaluate different storage conditions on the physiological classification and quality of P. pacifica seeds. To this end, there was the seed storage of this specie during periods of 0, 30, 60, 90 and 120 days in three conditions: in laboratory environment (25 ± 4 °C; 40% of U.H.) in permeable paper and semipermeable plastic, and in a refrigerator (5 ± 4 °C; 28% of U.H.) in semipermeable plastic. The following variables were evaluated in each storage period: moisture content (%), emergency of seedlings (%) in plastic trays with sand in laboratory environment (25 ± 4 °C; 40% of U.H.), total length (aerial part + root) (cm) and total dry weight (mg) of seedlings. The seeds stored in laboratory environment showed a progressive increase on percentage of seedling emergence to 37 and 45 days for plastics (67%) and paper (74%) respectively, rather than those packed in plastic bag in refrigerator in which the seedling emergence was zero (0%) after 50 days. Seeds of P. pacifica may be physiologically classified as intermediate because are intolerant to storage at low temperatures but maintains the physiological quality when stored in permeable or semipermeable package, in laboratory environment condition.
