Screening for myeloid mutations in patients with myelodysplastic syndromes and AML with myelodysplasia-related changes

ABSTRACT Introduction: One of the most critical complications in myelodysplastic syndromes (MDS) is the progression to acute myeloid leukemia (AML). The dynamics of clonal evolution in MDS and how acquired mutations can be used as biomarkers to track disease progression remains under investigation. Objective and method: Herein, we investigated the frequency of common myeloid clonal mutations (FLT3, NPM1, JAK2, IDH1 and IDH2) in 88 patients with MDS and 35 AML patients with myelodysplasia-related changes, followed at a single reference center in northeastern Brazil. Results: Overall, 9/88 (10%) ofthe MDSpatients and 9/35 (26%) of the secondary AML patients had at least one mutation. While the JAK2 V617F mutation was the most frequent in the MDS patients, the FLT3, NPM1, IDH1 and IDH2 mutations were more frequently found in the secondary AML group. Furthermore, there was a higher frequency of FLT3, NPM1, IDH1 and IDH2 mutations in MDS patients classified as high-risk subtypes than in those of lower risk. Conclusion: Despite the limited sample size, our data suggest that mutations in FLT3, NPM1, IDH1 and IDH2 genes could be potential biomarkers to detect early disease progression in MDS.

INFLUENCE OF ALBUMIN ON GUAVA PULP POWDER OBTAINED BY FOAM-MAT DRYING

ABSTRACT Guava is a fruit that stands out for its aroma, intense flavor, vitamins, and minerals. Its availability as a powdered food enhances its commercial applications. This study aimed to evaluate mathematical models for adsorption isotherms and determine the properties and flowability of guava pulp powder obtained by foam-mat drying. The models of GAB, BET, Henderson and Oswin were fitted to determine the isotherms of powders with 4 and 8% albumin, at temperatures of 35°C and 45°C. The powders were evaluated by wall friction angle, flow index, apparent density, and particle microstructure analysis. The best fit to the isotherms was obtained by the GAB model. Wall friction angle ranged from 15.7 to 21.9° and from 13.6 to 20.4° for samples containing 4 and 8% albumin, respectively. Higher densities were observed in the powder containing 8% albumin. The powders were classified as easy flowing when the flow index was between 8.00 and 8.68. The presence of albumin in powders resulted in particles with less rough surfaces, less hygroscopicity, and improved flowability.

ABSTRACT The quality of sesame (Sesamum indicum L.) seeds is directly influenced by their physical characteristics, which are seldom studied notwithstanding their extreme importance in the entire production chain. Therefore, the objective of this study was to quantify the physical properties of two types of sesame seeds, collected at three points in the plant with different maturation levels, measured with digital caliper, and to evaluate the feasibility of using a digital image to determine their physical characteristics. The seeds were initially evaluated for water content and mass of 1000 seeds, and later with the data of the characteristic dimensions A (length), B (width), and C (thickness), obtained by a caliper and digital images, we determined their sphericity, circularity, projected area, and geometric diameter. We observed that water content did not affect the physical properties of the cream (CNPA G4) and black sesame cultivars. In addition, seeds of the black cultivar presented higher results than the seeds of the cream cultivar for the mass of 1000 seeds (2.7 ± 0.08 g), sphericity (58.8 ± 0.47%), projected area (4.03 ± 0.12 mm2), and geometric diameter (1.65 ± 0.03 mm). Seed batches harvested after 70% maturation had the highest mass of 1000 seeds (2.7 ± 0.12 g). The results of the physical analyses of sesame seeds performed with caliper and digital image are equivalent, which shows that digital images can be used as tools to obtain the physical properties of seeds.

The use of targeted selective treatments on controlling gastrointestinal nematodes in different sheep categories under grazing system

RESUMO: Infecções por nematóides gastrointestinais podem prejudicar tanto a saúde como o desempenho animal. Diante disso, objetivou avaliar o uso de tratamentos seletivos como o sistema FAMACHA© e a contagem de ovos por grama de fezes no controle de nematóides gastrointestinais em diferentes categorias de ovinos sob sistema de pastejo. Trinta ovelhas Santa Inês, seis em cada categoria (seis cordeiros lactentes, seis cordeiros desmamados, seis ovelhas não-prenhes, seis ovelhas prenhes e seis ovelhas lactantes) foram utilizadas. Os ovinos foram pesados, amostras de sangue e fezes foram coletadas nos dias 0, 15, 30, 45, 60, 75 para determinação de ovos por grama de fezes (OPG), volume globular (VG), proteínas plasmáticas totais (PPT) e avaliação da mucosa ocular pelo sistema FAMACHA© pela manhã (1) e pela tarde (2). A contagem de ovos por grama de fezes foi maior (p=0,0039) nos cordeiros lactentes e desmamados comparado às ovelhas não prenhes, prenhes e lactantes. Também o OPG diferiu entre ovelhas com maior contagem em ovelhas lactantes. A observação da mucosa ocular mostrou que a frequência do escore FAMACHA© variou de 2,45 a 2,91 pela manhã, diferindo (p=0,0494) entre categorias, aonde cordeiros desmamados mostraram melhor escore. A interação categoria animal e período teve um efeito significante no escore FAMACHA© 1 com melhor escore em cordeiros desmamados comparado às ovelhas prenhes nos dias 0, 15 e 75. Além disso, cordeiros desmamados apresentaram melhor escore FAMACHA© comparado com os cordeiros lactentes e ovelhas lactantes aos 45 dias e com cordeiros lactentes nos dias 60 e 75. O volume globular pela manhã e pela tarde foi influenciado pelos dias de coleta com menor porcentagem aos 30 dias de experimento. O Sistema FAMACHA© mostrou ser um método eficaz para detectar infecções parasitárias. Contudo, o uso de todos os métodos disponíveis aumenta a acurácia do diagnóstico e promove melhor controle de nematóides gastrointestinais em sistemas de pastejo.
ABSTRACT: Infections with gastrointestinal nematodes can impair both the animal health and performance. Thereby, the aim of this study was to evaluate the use of targeted selective treatments as the FAMACHA© system and faecal egg count on controlling gastrointestinal nematodes in different sheep categories under grazing system. Thirty Santa Inês sheep, six in each category (six weaning, six weaned lambs, six non-pregnant ewes, six pregnant ewes and six lactating ewes were used. Sheep were weighed, and blood and faecal samples were collected on day 0, 15, 30, 45, 60 and 75 for determination of faecal egg count (FEC), packed cell volume (PCV), total plasma protein (PPT) and the mucosa colour by FAMACHA© system in the morning (1) and afternoon (2). The faecal egg count was higher (p=0.0039) in weaning and weaned lambs compared to the non-pregnant, pregnant and lactating ewes. Also, FEC differed between ewes with higher counts in lactating ewes. The observation of eye mucosae showed a frequency of 100% of FAMACHA© scores ranged of 2.45 to 2.91 in the morning, differing (p=0.0494) among the categories, where weaned lambs showed better score. The animal categories and period interaction had a significant (p=0.0148) effect on FAMACHA© score 1 with better score in weaned lambs compared to the pregnant ewes at day 0, 15 and 75. In addition, weaned lambs were presented better FAMACHA© score compared with weaning lambs and lactating ewes at day 45 than weaning lambs at day 60 and 75. Packed cell volume in the morning and in the afternoon was influenced by days of collection with lower percentage at 30 days of experiment. The FAMACHA© system seems to be a suitable method to detect parasite infections. However, use of all available methods increases the accuracy of the diagnosis and promotes better control of gastrointestinal nematodes in grazing sheep.

Resumo Apresenta-se uma síntese atualizada de informações sobre algas no Brasil objetivando refinar os dados reunidos até o presente, bem como avaliar os avanços sobre o conhecimento da diversidade de algas no país desde a publicação do Catálogo de Plantas e Fungos do Brasil. Os resultados de 2015 mostraram um acréscimo de 1.250 espécies (35.7%) a um total de 4.747 em relação a 2010. As classes mais diversas em número de espécies foram Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae e Euglenophyceae. Bacillariophyceae e Cyanophyceae tiveram o maior acréscimo de espécies no intervalo de cinco anos. A região Sudeste e Sul foram as mais diversas, porém, as regiões Nordeste com os estados do Piauí e Sergipe e Centro-Oeste com os estados de Mato Grosso, Goiás e Distrito Federal destacaram-se no cenário da biodiversidade nacional. Apesar da escassez de taxonomistas e da infraestrutura limitada, os resultados obtidos evidenciaram um avanço significativo no conhecimento da diversidade de algas no país nesse período de cinco anos, iniciando uma mudança quanto as diferenças geográficas regionais.
Abstract An updated synthesis of cyanobacteria and algae information is presented for Brazil aiming to refine the data gathered to date and evaluate the progress of the biodiversity knowledge about these organisms in the country since the publication of the Catálogo de Plantas e Fungos do Brasil. The results of 2015 showed an increase of 1,250 species (35.7%) when compared to 2010, reaching a total of 4,747 species. The most diverse classes in species number were the Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae and Euglenophyceae. Bacillariophyceae and Cyanophyceae had the highest increase in species number in the five-year interval. The Southeast and South regions were the most diverse, however, the Northeast, with the states of Piauí and Sergipe, and the Central-west region, with Mato Grosso, Goiás and Distrito Federal, also stood out in the national algal biodiversity scenario. Despite the shortage of taxonomists and limited infrastructure, the results showed a significant improvement in the knowledge regarding the diversity of cyanobacteria and algae in the country during the study period, starting to even out regional geographical differences caused by subsampling.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

Total phytosterol content in drug materials and extracts from roots of Acanthospermum hispidum by UV-VIS spectrophotometry

Acanthospermum hispidum DC., Asteraceae, is widely used in folk medicine in Brazil to treat respiratory diseases; this biological property has been attributed to its phytosterol content. This study evaluated the spectrophotometric assay method to quantify the total phytosterol content in raw materials and extracts from roots of A. hispidum. The procedure was based on the quantification at 625 nm after the Liebermann-Burchard reaction. The method was evaluated for linearity, repeatability, intermediate precision, accuracy and robustness. The date indicated that the procedure is a valid analytical tool for materials and herbal derivatives from A. hispidum.

Anemia Fanconi (AF) é uma síndrome autossômica recessiva, caracterizada por pancitopenia progressiva com hipoplasia de MO, em associação com várias anormalidades constitucionais, tendo como único recurso terapêutico com possibilidade potencial de cura o transplante de medula óssea, e sendo tais pacientes propensos ao desenvolvimento de malignidades hematológicas e carcinoma de células escamosas (CEC) em diversos locais: reto, vagina, cérvice, esôfago, cavidade bucal, faringe ou pele, mas especialmente em cabeça e pescoço. Relatamos aqui três casos de pacientes portadores de AF, que após TMO desenvolveram CEC em língua. Além disso, mencionamos fatores de risco relatados para tal evento, como diagnóstico de AF, condicionamento pré-transplante (quimioterápicos e irradiação), terapia com drogas imunossupressoras para tratamento de doença enxerto contra hospedeiro (DECH) aguda ou crônica, sexo e idade avançada. Além do que, discorremos sobre a existência de três mecanismos postulados que predispõem indivíduos com AF ao desenvolvimento de neoplasia: (1) defeito na reparação do DNA; (2) defeito na detoxificação de radicais de oxigênio; e (3) imunodeficiência.
Fanconi's Anemia, first described in 1927, is a rare autonomic recessive disease characterized by progressive pancytopenia, congenital malformations, spontaneous or chemically induced chromosome breakage and increased incidence of leukemia and other cancers. The onset of bone marrow hypoplasia and its hematological manifestations is usually in the 3 - 7 year age range. The disease has traditionally been managed clinically through administration of blood products, treatment of infections and prolonged administration of androgens, growth factors and more recently with gene therapy. The value of bone marrow transplantation in correcting the hematological manifestations of Fanconi's anemia has been established. Alkilanting agents and radiation have been utilized as a conditioning regimen in bone marrow transplantation for Fanconi's anemia, but these patients are particularly hypersensitive to these agents and its toxicity is the main cause of mortality and morbidity. Bone marrow transplantation is at the moment the only therapeutic modality able to bring hematological cure. Fanconi's anemia patients are particularly susceptible to the development of hematological malignancies and squamous cell carcinoma in the epithelium of the rectum, vagina, cervix, esophagus, oral cavity, pharynx or skin in general but more frequently on the head and neck; probably not only because the basic cellular defect but also because of the conditioning regimen. We report on three cases of Fanconi's anemia patients developing squamous cell carcinoma of the tongue after bone marrow transplantation. Furthermore, there is mention of the risk factors linked in an event, such as the diagnosis of Fanconi's anemia, pre-transplant conditioning regimen that usually includes alkylating agents and irradiation; immunosuppressive prophylactic therapy for acute and chronic graft-versus-host-disease, gender and age. Additionally, we discussed the existence of three postulated mechanisms that make individuals with Fanconi's anemia susceptible to the development of neoplasias: (1) deficiency in the DNA repair system, (2) deficiency in oxygen radical detoxification; and (3) immunodeficiency.

Produção e consumo de carbono orgânico na comunidade planctônica da represa da Pampulha, Minas Gerais, Brasil

O presente estudo tem por objetivo investigar a importância relativa do fitoplâncton como recurso alimentar para o zooplâncton em um reservatório hipereutrófico tropical. Para atingir esta meta, dois enfoques foram adotados. O primeiro deles consistiu num monitoramento regular (mensal) dos teores de biomassa do zooplâncton e das suas fontes de alimento expressas em termos de biomassa de carbono de partículas sestônicas. O segundo enfoque consistiu numa abordagem experimental com o objetivo de determinar o balanço entre a produção primária e o consumo de carbono orgânico na comunidade planctônica do reservatório. Tal avaliação deu-se através da mensuração simultânea das taxas de produção primária líquida potencial do fitoplâncton e das taxas de assimilação da comunidade mesozooplanctônica, derivadas a partir das taxas de respiração. A produção primária foi estimada em duas frações distintas: a) 50-160 µm e b) < 50 µm. Tanto a produção primária quanto a respiração do zooplâncton foram estimadas simultaneamente no laboratório sob condições controladas de iluminação e temperatura. O programa de monitoramento indicou que, excetuando breves períodos nos quais ocorreu o florescimento de Cyanobacteria coloniais (Microcystis spp.), a biomassa da comunidade fitoplanctônica foi quase sempre reduzida na zona limnética do reservatório (< 30 µg.L-1). A faixa de oscilação da clorofila-a restringiu-se a 0-126 µg.L-1. Adicionalmente, a maior parte do carbono particulado foi de origem não pigmentada. Os estudos experimentais indicaram que a demanda energética do zooplâncton, em termos de carbono, não pode ser suprida pela produção devida ao fitoplâncton no Reservatório da Pampulha. A produção primária potencial oscilou entre 2,0 e 37,3 mgC.m-3.h-1, valores estes que ficaram muito abaixo das taxas de assimilação devidas ao zooplâncton em diferentes épocas do ano. Considerando as evidências obtidas no campo e aquelas provenientes dos estudos experimentais, o presente estudo conclui que a demanda de carbono do mesozooplâncton durante o período de estudos foi em grande parte coberta através da cadeia de detritos.
The basic aim of the present investigation was related to the question: How important is the phytoplankton as a food resource for zooplankton in a tropical hypereutrophic reservoir? For answering this question, two different approaches were adopted. The first one included a regular monitoring program aimed to follow the composition and biomass of zooplankton as well as the major food resources, expressed in terms of C-biomass of sestonic particles. A second approach included the experimental determination of the balance between production and consumption of organic carbon in the planktonic community of Pampulha Reservoir. This was done through simultaneous determination of maximum potential primary production and assimilation rates of mesozooplankton estimated from respiration rates. The algal production was evaluated considering two different size fractions: a) nanno- (< 50 µm) and b) net phytoplankton (50-160 µm). The respiration of zooplankton community was restricted to larger organisms (mesozooplankton). These rates were determined at the laboratory under optimal conditions of illumination and temperature. Field data from the monitoring program showed that the limnetic zone of the reservoir supported a relatively low algal biomass for most parts of the seasonal cycle (< 30 µg.L-1). Except the short term algal (Microcystis spp.) blooming periods, chlorophyll-a concentrations in the water column remained at relatively low levels. The annual range of chlorophyll-a was 0-126 µg.L-1. Furthermore, field data also indicated that the bulk of the sestonic carbon was concentrated on non-pigmented particles. Evidences from laboratory suggested that the carbon demand of zooplankton could not be supplied by primary producers from the limnetic zone alone. The potential primary production ranged from low to moderate values (2.0-37.3 mgC. m-3h-1) and, as a consequence, assimilation rates of zooplankton remained higher than algal production (< 50 µm) for most parts of the year. Considering these evidences from field and laboratory, this study concludes that the energetic demand of (meso)zooplankton was, to a large extent, supplied by the detrital food chain.
