Abstract The peeling process of fresh ginger during factory production causes not only waste but also pollution to the environment. Now, by fermenting whole pieces of fresh ginger into black ginger, people are able to further enhance the nutritional value of ginger while lowering the burden on the environment. In this experiment, fresh ginger was fermented into black ginger, and superoxide dismutase (SOD) crudely extracted from both fresh ginger and black ginger were gavaged separately in type 2 diabetic rats to compare the SOD activity and effect in rat liver during the treatment of type 2 diabetes. It was found that the SOD activity of extracts from fermented black ginger was much higher than that of extracts from fresh ginger. In addition, the activity of SOD in the liver of rats gavaged with black ginger extract is higher than that in the liver of rats gavaged with fresh ginger extract, which shows that black ginger extract has certain protective effects on the liver.

OBJECTIVES: To investigate the role of miR-139-5p and the TLR4/MyD88/NF-κB signaling pathway in acute lung injury in septic mice. METHOD: A total of 140 healthy male SPF C57BL/6 mice were divided into seven groups, i.e., Normal, Control, NC, miR-139-5p mimic, miR-139-5p inhibitor, TAK-242, and miR-139-5p inhibitor+TAK-242 groups. The levels of miR-139-5p, proteins related to the TLR4/MyD88/NF-κB signaling pathway (TLR4, MyD88, and p-NF-κB p50), and MPO, SOD, GSH, and MDA in lung tissue were measured. The lung tissue wet-to-dry mass ratio (W/D), arterial oxygen partial pressure (PaO2), and carbon dioxide partial pressure (PaCO2) were measured. RESULTS: A web-based bioinformatic tool predicted that MyD88 was a target of miR-139-5p, which was verified by a dual luciferase reporter assay. Compared with those in the Normal group, the levels of miR-139-5p, PaO2, SOD, and GSH were significantly lower, while those of TLR4, MyD88, p-NF-κB p50, W/D, PaCO2, IL-1β, TNF-α, IL-6, MPO, and MDA were higher in all other groups. Moreover, compared with their levels in the Control group, these indicators exhibited contrasting results in the miR-139-5p mimic and TAK-242 groups, but were similar in the miR-139-5p inhibitor group. In the miR-139-5p inhibitor+TAK-242 group, acute lung injury, aggravated by miR-139-5p inhibitor, was partially rescued by TAK-242. CONCLUSION: miR-139-5p inhibits the TLR4/MyD88/NF-κB signaling pathway to alleviate acute lung injury in septic mice.

Abstract Purpose: To investigate the mechanism of Periplaneta americana extract promoting intestinal mucosal repair of OXZ-induced colitis in rat. Methods: All experiments used an equal number of male and female SD rats (n=48). We injected OXZ into the colon to induce UC rat model. To determine the optimal concentration of P. Americana's extract (PA-40), it was classified into low (L), medium (M), and high (H) doses. After OXZ treatment, each drug was administered by enema for 7 consecutive days. Rats were divided into the following 6 groups: (1) Saline treatment group (NC), (2) OXZ treatment UC model group (MC), (3) OXZ + budesonide group (BUN), (4) OXZ + PA-40 L group, (5) OXZ + PA-40 M group, (6) OXZ + PA-40 H group. Disease activity index (DAI) scores, colon length, histopathological score, serum cytokine level (IL-4, IL-10, iNOS, tNOS), and amount of MPO, EGF, IL-13 in colonic mucosa were measured. Results: PA treatment had a significant healing effect on the OXZ-colitis model and significantly reduced the lesioned area, especially in the PA-40H groups. PA treatment did not alter the expression of IL-10 and MPO level, but increased EGF (epidermal growth factor) and decrease IL-13 in the colonic tissue. PA inhibited the rise of NOSs (nitric oxide synthase) and decreased the serum IL-4 level. Conclusions: The data suggest that Periplaneta americana extract may be a potential compound for the treatment of colonic lesions. The mechanism may be related to inhibiting the secretion of IL-13 and promoting the formation of EGF.

Abstract For deep-water long span bridges under earthquakes, the interaction between water and structure will inevitably induce the hydrodynamic force on the structures. Based on the Morison potential fluid theory, a simplified calculation method of hydrodynamic force was proposed. Taken the 3rd Nanjing Yangtze River Bridge in China as the prototype, the shaking table test for the elevated pile caps was performed. And the results from the experiment and the proposed simplified calculation method were analyzed and compared. The main conclusions include: the natural vibration frequency of the bridge in deep water is decreased, and the predominant frequency is reduced by 8.24% due to the water-structure interaction. In earthquake, the effect of hydrodynamic force on the dynamic response of the deep water bridge is significant that the bending moment of the main girder is increased 7.73% and the bottom bending moment of the most adverse pile is increased 14.22%. In the seismic design of deep-water bridges, the effect of water-structure interaction should not be ignored.

OBJECTIVE: To evaluate the safety and efficacy of combining artemether (AM) and praziquantel (PZQ) in different regimens for treating acute schistosomiasis japonica. METHODS: We undertook a randomized, double-blind, placebo-controlled trial within four specialized schistosomiasis hospitals in the Dongting Lake region, Hunan province, China, between May 2003 and December 2005. Study participants were randomized into one of four treatment regimes: group A received 60 mg/kg PZQ + 6 mg/kg AM; group B received 60 mg/kg PZQ + AM placebo; group C received 120 mg/kg PZQ + 6 mg/kg AM; and group D received 120 mg/kg PZQ + AM placebo. All participants were followed up over a 45-day period. The primary endpoint of the trial was human infection status (determined by positive stool examination). Secondary endpoints involved clinical observations and blood biochemistry, including monitoring haemoglobin and alanine aminotransferase levels over time. FINDINGS: Treatment efficacies of the four different treatment regimens were 98.0%, 96.4%, 97.7% and 95.7% for group A, B, C, and D respectively (P > 0.05). The group B had a greater treatment efficacy (96.4%) than the group D (95.7%) (P > 0.05). Group A treatment was better for clearance of fever (P < 0.05) and resulted in a shorter hospitalization time (P < 0.05). CONCLUSION: This is the first report of a randomized, double-blind, placebo-controlled trial for evaluating combined chemotherapy with AM and two different dosages (60 mg/kg and 120 mg/kg) of PZQ in the treatment of acute schistosomiasis japonica in China. The combination of AM and PZQ chemotherapy did not improve treatment efficacy compared with PZQ alone. PZQ given as a dosage of 60 mg/kg (1 day, 3 × 20 mg/kg doses at 4-5 hour intervals) may be as effective as a dosage of 120 mg/kg (6 days, 20 mg/kg for each day split into 3 doses at 4-5 hour intervals).

OBJETIVO: Evaluar la seguridad y eficacia de la combinación de artemetero (AM) y prazicuantel (PZQ) en diferentes posologías para tratar la esquistosomiasis japonesa aguda. MÉTODOS: Entre mayo de 2003 y diciembre de 2005 se realizó un ensayo aleatorizado a doble ciego controlado con placebo en cuatro hospitales especializados en la esquistosomiasis de la región del lago Dongting, en la provincia china de Hunan. Los participantes en el estudio se repartieron aleatoriamente entre cuatro grupos sometidos a distintas pautas de tratamiento: el grupo A recibió 60 mg/kg PZQ + 6 mg/kg AM; el grupo B, 60 mg/kg PZQ + placebo de AM; el grupo C, 120 mg/kg PZQ + 6 mg/kg AM; y el grupo D, 120 mg/kg PZQ + placebo de AM. Todos los participantes fueron sometidos a seguimiento durante un periodo de 45 días. El criterio principal de valoración empleado en el ensayo fue la presencia de la infección (examen coproparasitoscópico positivo). Como criterios secundarios de valoración se emplearon los resultados de la exploración clínica y de los análisis bioquímicos sanguíneos, en particular la evolución de los niveles de hemoglobina y de alanina-aminotransferasa. RESULTADOS: La eficacia de las cuatro pautas de tratamiento fue del 98,0%, 96,4%, 97,7% y 95,7% para los grupos A, B, C y D, respectivamente (p > 0,05). La eficacia fue mayor en el grupo B (98%) que en el D (95,7%) (p > 0,05). El tratamiento recibido por el grupo A eliminó más eficazmente la fiebre (p < 0,05) y acortó el tiempo de hospitalización (p < 0,05). CONCLUSIÓN: Este es el primer trabajo publicado sobre un ensayo aleatorizado a doble ciego controlado con placebo destinado a evaluar la antibioticoterapia combinada con AM y dos posologías diferentes (60 mg/kg y 120 mg/kg) de PZQ como tratamiento de la esquistosomiasis japonesa aguda en China. La combinación de AM y PZQ no mejoró la eficacia del tratamiento en comparación con el uso aislado de PZQ. El PZQ administrado en dosis de 60 mg/kg (1 día, 3 tomas de 20 mg/kg a intervalos de 4-5 horas) puede ser tan eficaz como en dosis de 120 mg/kg (6 días, 20 mg/kg cada día repartidos en 3 tomas a intervalos de 4-5 horas)

OBJECTIF: Evaluer l'innocuité et l'efficacité de l'association artéméther (AM)/praziquantel (PZQ) dans le cadre de différents schémas thérapeutiques contre la schistosomiase asiatique aiguë. MÉTHODES: Nous avons entrepris un essai contrôlé randomisé en double aveugle dans quatre hôpitaux spécialisés dans la schistosomiase de la région du lac de Dongting, dans la province du Hunan, en Chine, entre mai 2003 et décembre 2005. Aux participants à l'étude, on a affecté au hasard l'un des quatre schémas thérapeutiques suivants : le groupe A a reçu 60 mg/kg de PZQ + 6 mg/kg d'AM ; le groupe B a reçu 60 mg/kg de PZQ + placebo de l'AM ; le groupe C a reçu 120 mg/kg de PZQ + 6 mg/kg d'AM ; et le groupe D a reçu 120 mg/kg de PZQ + placebo de l'AM. Tous les participants ont été suivis sur une période de 45 jours. La principale mesure de résultat pour l'essai était le statut infectieux des sujets (déterminé par le résultat de l'examen de selles). Parmi les mesures de résultat secondaires, figuraient des observations cliniques et des analyses biochimiques du sang, notamment la surveillance des taux d'hémoglobine et d'alanine aminotransférase au cours du temps. RÉSULTATS: L'efficacité du traitement par les quatre schémas thérapeutiques était respectivement de 98,0 %, 96,4 %, 97,7 % et 95,7 % pour les groupes A, B, C et D (p > 0,05). Le traitement du groupe B s'est révélé plus efficace (98 %) que celui du groupe D (95,7 %) (p > 0,05). Le traitement du groupe A a permis une meilleure élimination de la fièvre (p < 0,05) et une durée d'hospitalisation plus courte (p < 0,05). CONCLUSION: C'est la première fois qu'on rapporte un essai contrôlé randomisé en double aveugle visant à évaluer l'association médicamenteuse AM/PZQ et deux doses différentes (60 et 120 mg/kg) de PZQ dans le traitement de la schistosomiase asiatique aiguë en Chine. Le recours à l'association médicamenteuse AM/PZQ n'a pas amélioré l'efficacité du traitement par rapport à l'administration de PZQ seul. Le PZQ pourrait être aussi efficace à la dose de 60 mg/kg (sur une journée, 3 doses de 20 mg/kg à 4-5 heures d'intervalle) qu'à celle de 120 mg/kg (sur 6 jours, 20 mg/kg chaque jour, répartis en 3 doses à 4-5 heures d'intervalle).

OBJECTIVE: To determine the prevalence and intensity of Schistosoma japonicum infection and associated morbidity, and to estimate the infected human and buffalo populations in the Dongting Lake region, Hunan province, China. METHODS: We used data from the third national schistosomiasis periodic epidemiological survey (PES) of 2004. These included 47 144 human serological and 7205 stool examinations, 3893 clinical examinations and questionnaire surveys, and 874 buffalo stool examinations, carried out in 47 villages in Hunan province. Serological examinations were performed using the enzyme linked immunosorbent assay technique and human stool samples were examined by the Kato-Katz method. Stools from buffaloes and other domestic animals were examined for schistosome infection by the miracidial hatching test. FINDINGS: Sero-prevalence was 11.9% (range: 1.3-34.9% at the village level), and the rate of egg-positive stools was estimated at 1.9% (0-10.9%) for the same population. The prevalence of infection among buffaloes was 9.5% (0-66.7%). Extrapolating to the entire population of the Dongting Lake region, an estimated 73 225 people and 13 973 buffaloes were infected. Most frequently reported symptoms were abdominal pain (6.2%) and bloody stools (2.7%). More than half of the clinically examined people reported having had at least one prior antischistosomal treatment. CONCLUSION: There was a significant reduction in the number of humans infected with S. japonicum since the previous national PES carried out in 1995, partially explained by large-scale chemotherapy campaigns. However, a near-stable number of buffalo infections suggest continuing human re-infection, which may lead to future increases in human prevalence.

OBJETIVO: Determinar la prevalencia e intensidad de la infección por Schistosoma japonicum y la morbilidad a ella asociada, y estimar las poblaciones humanas y de búfalos infectadas en la región del lago Dongting, en la provincia china de Hunan. MÉTODOS: Usamos datos del tercer estudio epidemiológico nacional periódico sobre la esquistosomiasis de 2004. Como parte del mismo se hicieron 47 144 pruebas serológicas y 7205 análisis de heces en personas, 3893 exploraciones clínicas y encuestas a base de cuestionarios, y 874 análisis de heces en búfalos, en un total de 47 aldeas de la provincia de Hunan. Los análisis serológicos se realizaron mediante una técnica de inmunosorción enzimática, y las muestras de heces humanas fueron examinadas mediante el método de Kato-Katz. La presencia de infección por esquistosoma en las heces de búfalos y otros animales domésticos se analizó mediante la prueba de incubación de miracidia. RESULTADOS: La seroprevalencia fue del 11,9% (intervalo: 1,3%-34,9% a nivel de aldea), y la tasa de resultados positivos para la presencia de huevos en las heces fue del 1,9% (0%-10,9%) para la misma población. La prevalencia de infección entre los búfalos fue del 9,5% (0%-66,7%). Extrapolando a la totalidad de la población de la región del lago Dongting, se estimó que estaban infectadas unas 73 225 personas y 13 973 búfalos. Los síntomas notificados con más frecuencia fueron el dolor abdominal (6,2%) y las heces sanguinolentas (2,7%). Más de la mitad de los participantes explorados clínicamente declararon haber sido sometidos antes a tratamiento antiesquistosómico por lo menos una vez. CONCLUSIÓN: Se observó una reducción importante del número de personas infectadas por S. japonicum desde el estudio nacional realizado en 1995, lo que puede atribuirse en parte a las campañas de antibioticoterapia emprendidas a gran escala. Sin embargo, el número prácticamente inalterado de infecciones detectadas en la población de búfalos lleva a pensar que persisten las reinfecciones humanas, y ello podría traducirse en un futuro aumento de la prevalencia en el hombre.

OBJECTIF: Déterminer la prévalence et l’intensité de l’infection à Schistosoma Japonicum et la morbidité associée. Estimer l’importance des populations d’êtres humains et de buffles contaminés dans la région du lac de Dongting, dans la province du Hunan, en Chine. MÉTHODES: Nous avons utilisé des données provenant de la troisième enquête épidémiologique périodique sur la schistosomiase (PES) de 2004. Ces données étaient les résultats de 47 144 examens sérologiques, de 7205 examens de selles et de 3893 examens cliniques et enquêtes par questionnaire pratiqués chez des êtres humains, ainsi que de 874 examens d’excréments de buffles, effectués dans 47 villages du Hunan. Les examens sérologiques ont été réalisés par la méthode immuno-enzymatique en phase solide et les examens de selles humaines par la technique de Kato-Katz. On a recherché la présence d’une infection à schistosomes sur des selles de buffles et d’autres animaux domestiques par le test d’éclosion des miracidia. RÉSULTATS: La séroprévalence était de 11,9 % (plage : 1,3 - 34,9 % au niveau du village) et la proportion de selles positives pour le test d’éclosion a été estimée à 1,9 % (0 - 10,9 %) dans la même population. La prévalence de l’infection parmi les buffles était de 9,5 % (0 - 66,7 %). En extrapolant ces résultat à l’ensemble de la population de la région du Lac Dongting, on a estimé que 73 225 personnes et 13 973 buffles étaient contaminés. Les symptômes les plus fréquemment rapportés étaient des douleurs abdominales (6,2 %) et des selles sanglantes (2,7 %). Plus de la moitié des participants ayant subi un examen clinique ont signalé au moins un traitement antischistosomique antérieur. CONCLUSION: L’étude a permis de constater une réduction notable du nombre d’êtres humains contaminés par S. japonicum depuis la précédente enquête PES de 1995, réduction partiellement explicable par les campagnes de chimiothérapie à grande échelle. Cependant, le nombre relativement stable de buffles contaminés laisse prévoir une recontamination permanente des humains, susceptible de conduire à une recrudescence de la prévalence de la schistosomiase chez l’homme.
