The presence of Staphylococcus spp. has increasingly been reported in food products and poses a public health threat. The aim of this study was to determine the diversity of Staphylococcus spp. and the antibiotic resistance profiles of isolates obtained from freshly harvested and packed ready-to-eat mushrooms (n=432) and handlers' hands (n=150). A total of 56 Staphylococcus isolates [46.4% (n=26) from hands and 53.6% (n=30) from mushrooms] were recovered belonging to 10 species. Staphylococcus succinus isolates (n=21) were the most prevalent, of which 52.4% came from mushrooms and 47.6% from hands. This was followed by S. equorum isolates [n=12; 91.7% (n=11) from mushrooms and 8.3% (n=1) from hands] and S. saprophyticus [n=9; 66.7% (n=6) from mushrooms and 33.3% (n=3) from hands]. Six isolates that were characterised as multidrug resistant were isolated from hands of handlers. Most (83.9%; n=47) of the 56 isolates were resistant to penicillin [53.2% (n=25) from mushrooms and 46.8% (n=22) from hands] and 14.3% (n=8) were resistant to cephalosporin classes [25% (/=2) from mushrooms and 75% (n=6) from hands], both of which are used to treat staphylococcal infections. Antibiotic resistance genes blaZ [25.0% (n=14) of all isolates of which 71.4% (n=10) were from hands and 28.57% (n=4) from mushrooms], tetL and tetK [both 1.8% (n=1) from hands], mecA [5.4% (n=3) from hands] and ermA [1.8% (n=1) from mushrooms] were detected from the 56 isolates. Only two (25.0%) of the eight methicillin-resistant staphylococci harboured the mecA gene, while only 11 (23%) of the 47 penicillin-resistant isolates harboured the blaZ gene [36.4% (n=4) from mushrooms and 63.6% (n=7) from hands]. Our results demonstrate that food handlers and harvested and packed ready-to-eat mushrooms could be a source of diverse Staphylococcus spp. that exhibit antimicrobial resistance. Clinically relevant S. aureus was only detected on one handler's hand; however, the isolate was not multidrug resistant. The presence of diverse Staphylococcus spp. on mushrooms and the hands of handlers is a potential public health concern due to their potential to cause opportunistic infections.SIGNIFICANCE: • This study is the first to describe the antibiotic resistance profiles and antibiotic gene presence of Staphylococcus spp. isolated from fresh mushrooms and hands of pickers and packers. Mushrooms and handlers in this study were demonstrated to be possible routes of transmission of Staphylococcus spp. that are antibiotic resistant and which harbour antibiotic resistance genes, presenting a possible public health hazard.

This study considers expanding beyond the current collective understanding of research on the impact of global food and agriculture laws on Africa's food security. This paper aims to answer two basic questions: are the current global food and agriculture laws capable of facilitating and supporting the goal of ending hunger in Africa and increasing food security; will the existing global food law promote fair and equitable food production and supply practices to benefit all who need it? This paper will answer the questions by using a qualitative approach to Africa's experience in dealing with the existing global food and agriculture laws. This will provide insight into understanding the law, the behaviour of society and the outcome of the application of the law in real life. This will enable us to identify the gap in the global food law addressing food security. The qualitative data in the study will help the in-depth explanation, exploration and understanding of the root cause of food insecurity.

Limited information of fungicide efficacy on cultivated mushrooms and resistance development potential is available. Minor crop industries in general have a smaller arsenal of protectants to rely on and the likelihood of resistance build-up is of greater concern. This study focused on Cladobotryum mycophilum's sensitivity to carbendazim and prochloraz manganese following recent reports on decreased efficacy of both fungicides. The median effective dose (ED50) values for carbendazim ranged between 0.02 mg/L and 4.31 mg/L with 60% of the South African isolates being moderately resistant. The highest resistance factor for carbendazim was 215. Prochloraz manganese ED50 values varied from 0.00001 mg/L to 0.55 mg/L. A significant difference in mean ED50 values for both fungicides tested was observed. Using cluster analysis, no discrimination of isolates previously exposed and unexposed to prochloraz manganese was observed. A wide range of differences in ED50 values indicated moderate resistance to carbendazim and high sensitivity to prochloraz manganese among isolates under investigation. Discriminant analysis indicated significant differences between clusters contributed by one or a few variables. This study provided evidence that prochloraz manganese remains highly fungitoxic to C. mycophilum. However, prochloraz manganese is to be used in a disease management strategy in combination with strict farm hygiene management strategies to retain product efficacy and ensure crop protection.

Rainwater harvesting (RWH) is a relatively inexpensive technology that has the potential to provide safe water in communities where conventional technologies are difficult to implement. In this study, the microbiological quality of rainwater harvested from rooftops and ground-surface runoff was evaluated based on the concentrations of Escherichia coli, total coliforms and enterococci. Samples were collected from 15 roof-harvested rainwater (RHRW) tanks, 4 ground-surface runoff rainwater harvesting (GRWH) tanks, 3 rivers and 1 spring water source in the Eastern Cape Province of South Africa, and 14 RHRW tanks in Gauteng Province. In the Eastern Cape Province E. coli and enterococci were detected in 7 and 4 of the 15 RHRW tanks, respectively. Enterococci were detected only from one river whereas E. coli was detected in all three rivers; in spring water neither enterococci nor E. coli were detected. Samples from GRWH tanks were positive for E. coli and enterococci in 2 and 3 of the 4 tanks, respectively. In Gauteng Province, E. coli, coliforms and enterococci were detected from 6, 6 and 9 of the 14 rainwater tanks, respectively. On average, E. coli and enterococci were detected in 44.8% of the RHRW tanks, although enterococci concentrations were several times higher than those for E. coli. We further evaluated the significance of urban pigeons as the likely sources of contamination by isolating 156 enterococci from 30 pigeon faecal samples and 208 enterococci from RHRW samples collected from Gauteng Province. Matrix-assisted laser desorption ionisation identification of the various enterococci revealed 4 species - E. faecalis (20.5%), E. mundtii (20.51%), E. faecium (23.1%) and E. casseliflavus (17.3%) - to be dominant in faecal samples, whereas E. casseliflavus (34.6%) and E. mundtii (33.2%) were dominant in RHRW.

Strict quarantine measures for the export of South African citrus fruit to European and US markets require the development of sensitive and accurate detection methods for the pathogen Phyllosticta citricarpa - a fungus causing citrus black spot disease. Because of the presence of other, non-pathogenic Phyllosticta species, rapid and accurate verification of the Phyllosticta species present on exported citrus fruit is important to producers, exporters and regulatory authorities to prevent unnecessary losses. We have analysed over 800 samples collected over 7 years and have compared sample preparation and detection protocols applied in different environments: nurseries, production systems including phytosanitary inspections in orchards, pack houses and export terminals in order to compile protocols for the detection of P. citricarpa. Standard procedures of sample preparation and DNA extraction were adapted to suit diverse inoculum sources. Low pathogen numbers in symptomless green leaves, for example, obliged the use of a wet-dry enrichment technique constituting the stimulation of fungal growth for easier detection. Physical maceration was adapted for sturdy material using liquid nitrogen or bead beating. The use of a two-step polymerase chain reaction (PCR) with nested primers significantly increased both the sensitivity and the specificity of the PCR performed on soil samples, overcoming problems with relatively impure DNA extracts and low pathogen numbers. The assays have proven to be highly consistent, thereby providing a reliable, reproducible and highly sensitive detection and diagnostic service to the southern African citrus industries in order to sustain market access.

Three yeast antagonists (two strains of Cryptococcus laurentii and one of Candida sake) from orange trees reduced incidence of green mold by 80 to 95% when tested in wounded orange fruits inoculated with Penicillium digitatum and incubated at 7ºC for 30 days. The yeasts inhibited conidial germination of the pathogen, but did not kill the spores. Effectiveness of the three yeasts as antagonists was associated in part with their ability to rapidly colonize wound sites, despite low nutrient availability. Observations suggested that production of extracellular matrix by the yeasts may have facilitated rapid wound colonization. Germination of P. digitatum conidia was significantly inhibited when the pathogen and antagonists were in direct physical contact in a culture suspension. The results supported the view that competition for nutrients is also a mode of action of yeasts against P. digitatum.

Commercial producers of white button mushrooms utilise a casing material to cover the spawn run compost, which stimulates the mushrooms' reproductive stage. Certain bacteria in this casing are responsible for this stimulation, which is known as pinning. Bacterial species richness and diversity within peat and peat-based casing mixtures made from industrial waste materials (i.e. those containing coir, wattle bark, bagasse and filter cake) were examined using denaturing gradient gel electrophoresis (DGGE) at three phases of mushroom growth: (1) casing, (2) pinning and (3) harvesting. Results from the DGGE established that higher bacterial species richness occurred at pinning and harvesting than at casing. Increases in bacterial population density at pinning were greater in the peat-based mixtures, which contained industrial waste materials, than in peat alone. Peat mixtures containing these alternative materials are therefore favourable substrates for bacterial growth. The DGGE profiles for pasteurised casing materials reflected their ability to rapidly re-establish the original bacterial community. The bacteria found to be dominant in casing materials during pinning were closely related to Pseudomonas, Flavobacterium, alpha-Proteobacterium, beta-Proteobacterium, gamma-Proteobacterium, delta-Proteobacterium and uncultured species.
