Resumo: O objetivo deste trabalho foi selecionar genótipos de mandioca (Manihot esculenta) do banco de germoplasma brasileiro com funcionalidade semelhante à do amido ceroso. Um total de 881 genótipos foi pré-selecionado por análise de componentes principais e agrupamento hierárquico, e seu potencial industrial foi comparado ao dos clones 7745-5WX de mandioca cerosa e WX-Maize de milho ceroso, ambos usados como referência. Dois genótipos se destacaram: BGM0036 e BGM0083. Amostras desses quatro genótipos foram caracterizadas por meio de teor de amilose, microscopia eletrônica de varredura, difração de raios-X, calorimetria diferencial de varredura, viscosidade de pasta e distribuição da cadeia de amilopectina. As amostras apresentaram cristalinidade tipo A e nenhuma diferença estatística (p> 0,05) quanto ao grau de cristalinidade (25,3 a 30,0), o que mostra proporções similares das frações de amilose/amilopectina. Não foram observadas diferenças na microestrutura dos amidos de mandioca. A temperatura inicial de gelatinização e as cadeias curtas de amilopectina apresentaram correlação negativa muito forte, o que indica que uma menor proporção de cadeias curtas de amilopectina resulta em uma maior temperatura inicial de gelatinização. BGM0036 e BGM0083 apresentam baixa viscosidade final, próxima às dos amidos de milho e mandioca cerosos, sendo uma alternativa para a utilização em diferentes alimentos que necessitam de estabilidade durante o congelamento.

Abstract: The objective of this work was to select cassava (Manihot esculenta) genotypes from the Brazilian germplasm bank with a functionality similar to that of waxy starch. A total of 881 genotypes were pre-selected using principal component analysis and hierarchical clustering, and their industrial potential was compared with that of the 7745-5WX waxy cassava and the WX-Maize waxy maize clones, both used as references. Two genotypes stood out: BGM0036 and BGM0083. Samples from these four genotypes were characterized by means of amylose content, scanning electron microscopy, X-ray diffraction, differential scanning calorimetry, paste viscosity, and chain-length distribution of amylopectin. The samples presented A-type crystallinity and no statistical differences (p > 0.05) regarding crystallinity degree (25.3 to 30.0), which shows similar proportions of the amylose/amylopectin fractions. No differences were observed in the microstructure of the cassava starches. Initial gelatinization temperature and amylopectin short chains presented a very strong negative correlation, indicating that a lower proportion of short chains of amylopectin results in a higher initial temperature of gelatinization. BGM0036 and BGM0083 show a low final viscosity, close to that of the waxy maize and cassava starches, being an alternative for use in different foods that require stability during freezing.

Abstract Native cassava starch was gelatinized under different pretreatment conditions (preheating temperature, starch concentration and heating time) and spray-dried. A rotational compound central design was used to test the three independent variables: the preheating temperature, starch concentration, and heating time, and the behavior of the system was assessed by response surface methodology. The results showed a significant effect of both the preheating temperature and heating time on the cold viscosity. The combination of the preheating temperature and starch concentration affected the crystallinity, enthalpy change, solubility, and final viscosity. Modification by spray-drying resulted in a decrease in the crystalline regions, and enthalpy change, as well as an increase in the cold viscosity. The optimal conditions for obtaining partially gelatinized cassava starch with high cold viscosity and lower breakdown, desirable characteristics for commercial pregelatinized starch, are as follows: starch concentration of 25%, preheating temperature of 52 °C, and processing heating time of 10 min.

ABSTRACT This study aimed to evaluate the effect of the high and low molecular weight glutenin subunits on the grain traits of sixteen Brazilian wheat genotypes. Grain hardness index, milling traits, physicochemical and rheological properties of the flour, and specific volume and firmness of the bread were evaluated. Physicochemical properties of the flour were not influenced by glutenin subunits. Genotypes with subunits at the Glu-B1 (17+18 or 7+8), Glu-D1 (5+10), and Glu-A3 (b) were associated with strong flours and bread with high specific volume and low firmness. The subunits at the Glu-A1 and Glu-B3 had no effect on the rheological properties of the dough and bread quality, while the subunit 2+12 at Glu-D1 negatively affected the resistance to extension, and specific volume and firmness of the bread. Specific volume and firmness of the bread were influenced by the rheological properties of the dough, while the flour protein content was not important to define wheat quality. The identification of glutenin subunits at different loci along with the rheological tests of the flour are fundamental in estimating the potential use of different materials developed in wheat breeding.

A identificação das propriedades funcionais de farinhas de trigo através de testes específicos possibilita selecionar genótipos de trigo com características adequadas a cada uso industrial. O principal objetivo dos programas de melhoramento genético é melhorar a qualidade do banco de germoplasma para que seja possível desenvolver trigos com força de glúten e extensibilidade adequadas para produção de produtos panificáveis. O objetivo deste estudo foi avaliar 16 genótipos de trigo correlacionando as subunidades de gluteninas de alto e baixo peso molecular e as subunidades de gliadinas com as características físico-químicas do grão. Foram determinados o teor de proteína, volume de sedimentação, índice de sedimentação e número de queda. O peso hectolitro e massa de 1000 sementes também foram determinados. As subunidades de gluteninas e gliadinas foram separadas usando gel de poliacrilamida na presença do dodecil sulfato de sódio. Os dados foram submetidos à análise de variância, correlação de Pearson, análise de componentes principais e análise de cluster. Os genótipos IPR 85, IPR Catuara TM, T 091015 e T 091069 se destacaram dos demais, evidenciando possível superioridade na qualidade do grão, apresentando maior volume de sedimentação, índice de sedimentação e peso de mil sementes, e apresentaram em sua constituição as subunidades 1 (Glu-A1), 5 + 10 (Glu-D1), c (Glu-A3) e b (Glu-B3), com exceção do genótipo T 091069, que apresentou o alelo g em vez do b no Glu-B3.

Identification of functional properties of wheat flour by specific tests allows genotypes with appropriate characteristics to be selected for specific industrial uses. The objective of wheat breeding programs is to improve the quality of germplasm bank in order to be able to develop wheat with suitable gluten strength and extensibility for bread making. The aim of this study was to evaluate 16 wheat genotypes by correlating both glutenin subunits of high and low molecular weight and gliadin subunits with the physicochemical characteristics of the grain. Protein content, sedimentation volume, sedimentation index, and falling number values were analyzed after the grains were milled. Hectoliter weight and mass of 1000 seeds were also determined. The glutenin and gliadin subunits were separated using polyacrylamide gel in the presence of sodium dodecyl sulfate. The data were evaluated using variance analysis, Pearson's correlation, principal component analysis, and cluster analysis. The IPR 85, IPR Catuara TM, T 091015, and T 091069 genotypes stood out from the others, which indicate their possibly superior grain quality with higher sedimentation volume, higher sedimentation index, and higher mass of 1000 seeds; these genotypes possessed the subunits 1 (Glu-A1), 5 + 10 (Glu-D1), c (Glu-A3), and b (Glu-B3), with exception of T 091069 genotype that possessed the g allele instead of b in the Glu-B3.

Amidos e gomas são hidrocoloides frequentemente usados em sistemas alimentícios com a finalidade de fornecer textura, umidade e mobilidade de água. A interação amido-goma em sistemas alimentícios pode alterar o inchamento do grânulo de amido e as suas propriedades de gelatinização e reológicas. Neste trabalho, o efeito da adição de goma xantana (GX), carboximetilcelulose sódica (CMC) e carragena (CAR) nas concentrações de 0,15, 0,25, 0,35 e 0,45% (p/v) sobre as propriedades de pasta, térmicas e reológicas do amido de mandioca foi estudado. O Poder de inchamento (PI) e a Microscopia Eletrônica de Varredura (MEV) dos géis de amido também foram avaliados. Os resultados obtidos mostraram que a GX apresentou forte interação com o amido, penetrando entre os grânulos e provocando aumento das viscosidades de pasta, PI, G' e G", e redução da retrogradação do amido; CMCS aumentou as viscosidades de pasta, PI, G' e G" das misturas, principalmente em função da sua maior capacidade de reter água, e não por causa da interação com o amido; CAR não modificou qualquer das propriedades do amido, porque não houve nenhuma interação entre essa goma e o amido de mandioca nas concentrações usadas.

Starches and gums are hydrocolloids frequently used in food systems to provide proper texture, moisture, and water mobility. Starch-gum interaction in food systems can change the starch granule swelling and its gelatinization and rheological properties. In this study, the effect of the addition of xanthan gum (XG), sodium carboxymethyl cellulose (SCMC), and carrageenan (CAR) at the concentrations of the 0.15, 0.25, 0.35 and 0.45% (w/v) on the pasting, thermal, and rheological properties of cassava starch was studied. The swelling power (SP) and the scanning electron microscopy (SEM) of the starch gels were also evaluated. The results obtained showed that xanthan gum (XG) had a strong interaction with the cassava starch penetrating between starch granules causing increase in pasting viscosities, SP, storage and loss (G', and G", respectively) modulus and reduction in the setback of the starch; sodium carboxymethyl cellulose (SCMC) greatly increased the pasting viscosities, the SP, and the storage and loss (G', and G", respectively) modulus of the starch-mixtures, mainly due to its greater capacity to hold water and not due to the interaction with cassava starch. Carrageenan (CAR) did not change any of the starch properties since there was no interaction between this gum and cassava starch at the concentrations used.

A banana, uma das principais herbáceas cultivadas em grande quantidade no mundo e importante fonte de energia para as pessoas, tem obtido interesse como matéria-prima para a produção de farinha e fécula para uso alimentício e outros fins industriais, devido ao elevado conteúdo de amido nos frutos verdes. O trabalho teve como objetivo caracterizar os frutos verdes de sete genótipos de bananeira quanto à composição química e avaliar o conteúdo de amilose, propriedades de pasta e térmicas dos amidos destes genótipos. Os resultados obtidos mostraram diferenças significativas na composição química dos frutos, com maior teor de amido na cultivar Figo Cinza. As análises dos amidos mostraram valores de amilose de 26,68 a 33,95% e propriedades de pasta com elevado pico de viscosidade, baixa resistência à temperatura e agitação (quebra de viscosidade) e tendência à retrogradação para todos os amidos. Nas propriedades térmicas, os amidos mostraram um endoderma, temperatura de gelatinização na faixa de 60 a 73ºC com DH variando de 11,6 a 16,99 J g-1 na gelatinização e 4,11 a 7,97 J g-1 na retrogradação.

The banana, the largest herbaceous plant in the world, grown abundantly in many countries is considered to be one of the most important sources of energy for people. Due to high starch concentration, banana processing into flour and starch is of interest in view of a possibly important resource for food and other industrial purposes. The objective of this study was to characterize the chemical composition of green fruits of seven banana genotypes, and evaluate the content of amylose, pasting and thermal properties of starches. Results showed significant differences to chemical composition with higher level of starch in cultivar Figo cinza. The amylose content ranged from 26.68 to 33.95%. Starches analysis showed pasting properties with high cold peak and breakdown, with retrogradation tendency to all genotypes. The thermal properties showed one endoderm. The gelatinization temperature was in range of 60 to 73ºC with DH ranged from 11.6 to 16.99 J g-1 in gelatinization and 4.11 to 7.97 J g-1 in retrogradation.

Amido de mandioca modificado com ácido lático e radiação ultravioleta antes da secagem artificial têm mostrado boa capacidade de expansão, a exemplo da encontrada para o polvilho azedo. Neste trabalho, os efeitos do ácido lático e radiação UV sobre as características físico-químicas e estruturais de amidos de mandioca e milho foram investigados. Apenas o amido de mandioca modificado apresentou capacidade de expansão durante o forneamento. Do RVA, redução dos valores de viscosidade, boa estabilidade interna e nenhuma tendência a retrogradação para este amido modificado foi observado. O amido de milho não apresentou qualquer pico de viscosidade após modificação. Não foram observadas diferenças significativas nas propriedades térmicas, determinadas em DSC, entre os amidos nativos e modificados de ambas as fontes. As moléculas de amilopectina e amilose de ambos os amidos modificados mostraram alguma degradação. O peso molecular da amilopectina do amido de mandioca foi mais preservado, enquanto a amilopectina do amido de milho foi mais atacada em todo o grânulo. As cadeias ramificadas longas da amilopectina de mandioca, com DP~37, foram degradadas, enquanto as de milho mantiveram-se inalteradas.

In this work, the effect of lactic acid and UV irradiation on the physicochemical and structural characteristics of cassava and corn starches was evaluated. Only the modified cassava starch presented baking expansion capacity. From RVA, reduction of viscosity values, greater internal stability and none set back for modified cassava starch were observed. Modified corn starch did not show any peak viscosity. There were no significant differences in DSC thermal properties of treated and native starches. Amylopectin and amylose molecules from both the modified starches displayed some degradation. Molecular weight of cassava amylopectin was mostly preserved, whereas corn amylopectin was evenly attacked through the granule. Nevertheless, the B long branched chains of cassava amylopectin, with DP~37, were degraded whereas they were unchanged for corn amylopectin.

No presente trabalho duas variedades comerciais de mandioquinha-salsa ('Amarela de Carandaí' e 'Senador Amaral') obtidas dos municípios de Castro e de Piraí do Sul (Paraná) foram processadas para a obtenção de farinha, farelo e amido e essas três frações foram caracterizadas quimicamente. O amido foi extraído, modificado com H2O2 e caracterizado por meio de análises físico-químicas. Na coloração com azul de metileno os grânulos dos amidos oxidados apresentaram coloração azul mais intensa. O teor de carboxilas, o poder redutor e a quantidade de água liberada das pastas foram maiores para as amostras de amidos modificados. As pastas dos amidos modificados foram mais claras que as dos nativos. Na análise viscoamilográfica (RVA) os amidos modificados apresentaram as menores viscosidades e houve diferenças entre as variedades, sendo que o amido modificado da variedade Senador Amaral de Piraí do Sul apresentou os menores valores de pico de viscosidade. Na análise térmica, as temperaturas de pirólise foram mais altas para os amidos nativos das variedades da mandioquinha-salsa (310,37, 299,08, 311,18 ºC) do que para os amidos modificados (294,16, 296,65 e 293,29 ºC) e esta diferença pode estar relacionada a maior superfície dos grânulos de amido parcialmente degradados pela modificação oxidativa. Os resultados revelaram diferenças entre os amidos das duas variedades, mas não entre os locais de cultivo.

Two commercial varieties of Peruvian carrot ('Amarela de Carandaí' and 'Senador Amaral') were processed into flour, starch and bagasse and chemically evaluated. The starch was extracted, modified with H2O2 and characterized by the physicochemical methods. By using the methylene blue dyeing, the granules of the modified starches showed intense blue color. The carboxyl content, the reducing power and the amount of the water liberated from the pastes after the freeze-thawing were higher for the oxidized starches and their pastes were clearer than those of the native starches of the two varieties from the two production areas. The RVA viscoamylography showed that the modified starches had lower viscosities with differences between the varieties. In the thermal analysis, the temperatures of the pyrolysis were higher for the native (310.37, 299.08, 311.18 ºC) than for the modified starches (294.16, 296.65 e 293.29 ºC) for both the varieties. This difference could be related with the larger surface of the granules due to the partial degradation promoted by the chemical modification. In almost all results, the differences were evident between the varieties but not for the cultivation places.

O amido apresenta propriedades físico-químicas importantes que são dependentes de sua fonte botânica e de suas características estruturais. O amido de mandioquinha-salsa possui características especiais que o tornam elegível para utilização em vários alimentos processados. Neste trabalho, amidos de duas variedades de mandioquinha-salsa, Amarela de Carandaí (AC) e Amarela de Senador Amaral (ASA), foram isolados e suas características físico-químicas e estruturais determinadas. O amido obtido da variedade ASA apresentou grânulos com diâmetro médio maior que aqueles da variedade AC e mais que o dobro da quantidade de grânulos com tamanho >20 µm, quando comparado à outra variedade. Os amidos de ambas as variedades, observados em microscópio eletrônico de varredura, mostraram superfícies granulares lisas com formatos circulares e poliédricos para grânulos grandes e pequenos, respectivamente. A distribuição de tamanho molecular, viscosidade intrínseca e índice de cristalinidade dos amidos de ambas as variedades foram semelhantes, porém o teor de amilose foi maior para o amido da variedade AC. Maiores valores de viscosidade, poder de inchamento, e temperaturas de gelatinização foram observados para o amido da variedade ASA e podem estar relacionados ao menor teor de amilose e à maior proporção de grânulos grandes apresentados por este amido.

Starch has very important physicochemical properties that depend on its botanical source and structural characteristics. Peruvian carrot starch displays special characteristics, which make it appropriate for industrial application in many processed foods. In this work, starch from two Peruvian carrot varieties, Amarela de Carandaí (AC) and Amarela de Senador Amaral (ASA), were isolated and their physicochemical and structural properties were determined. Starch from the ASA variety exhibited granules with a greater average diameter than those from the AC variety and more than double the number of granules with size >20 µm, when compared to the other variety. Starches from both varieties, observed in a Scanning Electron Microscope, showed smooth granule surface with circular and polyhedral shapes for large and small granules, respectively. Molecular size distribution, intrinsic viscosity and degree of cristallinity of starches from both varieties were similar; however, amylose content was higher for the starch from the AC variety. Higher values of viscosity, swelling power and gelatinization temperatures were observed for the starch from the ASA variety, which could be related to the lower amylose content and the higher proportion of large granules exhibited by this starch.

Amidos oxidados em elevadas concentrações produzem pastas fluidas, são estáveis à retrogradação e aplicados em indústrias de papel, têxtil e de alimentos. Esta propriedade é atribuída à presença de carboxilas, com cargas negativas e mais volumosas que as hidroxilas. Neste trabalho, amidos de batata, batata doce, mandioca, mandioquinha salsa, milho e milho ceroso foram modificados com KMnO4/ácido lático e NaOCl/ácido lático e caracterizados quanto ao teor de carboxilas, poder redutor, coloração diferencial, propriedade de expansão e propriedades viscoamilográficas, em água e tampões acetato (pH 4,0) e fosfato (pH 7,0). Amidos de milho ceroso e de mandioca modificados com KMnO4/ácido lático apresentaram elevadas expansões (25,8 e 24,1 mL.g-1, respectivamente). As pastas de amido de milho ceroso resistiram a ciclos de congelamento/descongelamento. A coloração diferencial e o teor de carboxilas não permitiram diferenciar as amostras; apenas o amido de batata reagiu com azul de metileno devido aos grupos fosfato. Quanto ao pH e acidez, as amostras apresentaram valores próximos da neutralidade, exceto o amido de milho e milho ceroso modificados com NaOCl. Amidos oxidados com KMnO4/ácido lático apresentaram picos de viscosidade inferiores aos tratados com NaOCl. Os picos de viscosidade em tampão fosfato foram inferiores aos em água e em tampão acetato.

Oxidized starches produce low-viscosity pastes even in high concentrations, are stable to retrogradation and are used in the paper, textile and food industries. This property is attributed to the presence of carboxyl groups, which have negative charges and are bulkier than hydroxyls. In this work, potato, sweet potato, Peruvian carrot, cassava, corn and waxy corn starches were modified with oxidative reagents and characterized with respect to their carboxyl content, reducing power, differential dyeing, expansion power and viscographic properties, in water, acetate buffer (pH 4.0) and phosphate buffer (pH 7.0). The results showed that waxy corn and cassava starches presented high expansions, when modified with KMNO4/lactic acid (25.8 and 24.1 mL.g-1, respectively). The starch pastes of waxy corn showed freeze-thaw stability in all treatments. Differential dyeing and carboxyl content did not vary between the native and modified samples; only potato starch reacted with methylene blue, due to the presence of phosphate groups. With respect to pH and acidity, almost all samples presented values next to neutrality, except corn and waxy corn starch modified with NaOCl/lactic acid. The starches oxidized with KMnO4/lactic acid presented lower viscosity peaks than those oxidized with NaOCl. The viscosity peaks of starches analyzed in phosphate buffer were lower than in water and acetate buffer.

A utilização de amidos modificados em alimentos tem sido uma alternativa para melhorar as características que os amidos nativos não podem conferir. Os objetivos deste trabalho foram: avaliar características físico-químicas de amidos modificados com peróxido de hidrogênio, utilizar os amidos obtidos para a elaboração de doce de leite e bala de goma americana e verificar a aceitação sensorial destes produtos. Para obtenção das amostras modificadas, os amidos foram suspensos em solução de Fe2SO4.7H2O 0,01% e, em seguida, adicionou-se peróxido de hidrogênio 1,25%, ajustou-se o pH para 3,0, deixou-se reagir a 45 °C/15 minutos, lavou-se e secou-se a amostra em estufa de ventilação forçada a 45 °C/24 horas. Os amidos modificados apresentaram maior expansão, conteúdo de carboxilas, poder redutor e susceptibilidade à sinérese. Em relação à viscosidade aparente, verificou-se que com a modificação química houve diminuição no pico de viscosidade, especialmente quando as amostras foram analisadas em pH 7,0. A análise sensorial do doce de leite revelou que houve preferência pelas amostras elaboradas com amido de mandioquinha-salsa modificado e com o amido de milho regular ácido-modificado. A bala de goma produzida com amido de mandioquinha-salsa modificado ocupou o segundo lugar entre a preferência dos julgadores.

The use of modified starches in foods is made to improve characteristics that the native cannot confer. The objective of this work was to evaluate physicochemical characteristics of starches from several botanical sources after chemical modification with H2O2 considering technological properties of interest to the food industry. The modified starches were used in dulce de leche and starch gum and the sensorial acceptance of these products was verified. The chemical treatment included the suspension of the native starches in a 0.01% Fe2SO4, followed by addition of 1.25% H2O2 at pH 3.0 and reaction time of 15 min at 45 °C. After this period the starch was recovered by filtration, washed and oven-dried at 45 °C for 24 hours. The modified starches had higher expansion, carboxyl content, reducing power and susceptibility to syneresis. There was a decrease in the viscosity peak of the modified starches especially when the samples were analyzed in pH 7.0. The sensorial evaluation indicated that dulce de leche produced with addition of Peruvian carrot modified starch and regular corn acid-modified starch were the preferred samples. The starch gum made with modified starch of Peruvian carrot reached the second position on global preference between the panelists.

Amidos nativos e modificados têm grande importância na indústria de alimentos, sendo empregados principalmente como espessantes e/ou estabilizantes. As limitações das pastas e géis obtidos a partir de amidos nativos tornaram necessário o desenvolvimento de muitos tipos de amidos modificados para aplicações alimentícias. Neste trabalho, algumas amostras de amidos modificados (n=20) disponíveis no Brasil foram recebidas de empresas produtoras e analisadas em relação a algumas características físico-químicas e propriedades tecnológicas. Um levantamento do uso de amidos modificados em alimentos também é apresentado, revelando crescente interesse pela indústria nesses ingredientes. Constatou-se que, enquanto alguns alimentos industrializados, como maioneses contêm em suas formulações amidos modificados, outros como condimento preparado de mostarda contêm apenas amido nativo. As análises físico-químicas permitiram concluir que alguns amidos modificados apresentavam teores elevados de acidez, relacionados à presença de reagentes utilizados em sua obtenção, não havendo presença de carboxilas nas suas macromoléculas. De maneira geral, os resultados de algumas propriedades tecnológicas avaliadas, tais como viscosidade aparente das pastas, resistência a congelamento/descongelamento e propriedade de expansão, estavam de acordo com a descrição dos produtos. As fontes mais observadas nas modificações foram, em ordem decrescente de importância, a mandioca, o milho ceroso e o milho regular.

Native and modified starches are very valued for the food industry, being employed mainly as thickeners or stabilizers. Several technological limitations of native starch pastes or gels made it necessary to develop modified starches for the food industry. In the present work modified starch samples (n=20) available in Brazil were received from some producers and evaluated in relation to selected physicochemical characteristics and technological properties. Also a very simple evaluation of food starch market was done and allowed to conclude that Brazilian food industry is deeply interested on modified starches. It was possible to note that some foods like mayonnaise have on its composition modified starch whereas others like mustard sauce are produced only with native starch. The physicochemical analyses showed that some of the studied samples had high acidity levels, related with the presence of excess reagents employed on the starch modification processes; the results of carboxyl groups content allowed this conclusion. Almost all results were in agreement with the producers' specifications, for example for the freeze-thaw stability, apparent viscosity and the expansion power. The most frequent botanical sources among the studied samples were cassava, waxy corn and regular corn.
