Os mais importantes reservatórios do vírus influenza são os pássaros. A manutenção do vírus influenza em hospedeiros naturais, inclusive o homem, permite que esse vírus realize rearranjos entre as suas cepas. O recente relato de uma cepa influenza aviária A(H5N1), em humanos, se deu em uma criança com doença respiratória fatal, na China em 1977. O presente estudo foi conduzido para elucidar o transporte da influenza por pássaros que migram, anualmente, através de ambos hemisférios o do Norte e do Sul, com especial atenção voltada à espécies Vireo olivaceo [Juruviara(BR) e Red-eyed vireo(USA)] que viaja do USA para o Brasil, e vice-versa, e a espécie Elaenia mesoleuca [Tuque(BR) e (USA)] que voa por todo o Hemisfério Sul. Essas espécies de pássaros, que residem e migram em São Paulo, e que demonstram transportar o vírus influenza, foram selecionadas. As partículas virais isoladas foram observadas por microscópio eletrônico. O vírus influenza foi detectado pelos testes: House Duplex/PCR e Gloria. Os resultados revelam que os pássaros das espécies: Elaenia mesoleuca e Vireo olivaceus são transportes do vírus influenza enquanto cruzam ambos Hemisférios. Para o conhecimento da função que os pássaros migratórios podem desempenhar na epidemia de influenza, no Brasil, caracterização dos subtipos deste vírus estão sendo realizados.

Birds are the most important reservoirs of the influenza virus. Its maintenance in its natural hosts, including man, allows the influenza virus to reassorts its strains. The recent report of an avian influenza A (H5N1) virus in humans, was in a child with fatal respiratory illness in China, 1997. The current study was conducted to elucidate the transportation of the influenza by birds that migrate, annually, through the both Northern and Southern hemispheres, with special attention paid to the Vireo olivaceus [Juruviara(BR) or Red-eyed vireo(USA)] species, which travels from the USA to Brazil, and vice versa, and the Elaenia mesoleuca [Tuque(BR) or (USA)] species that flies over the entire Southern Hemisphere. There are two species of birds, which breed and migrate in São Paulo State, Brazil, and which were demonstrated to carry Influenza virus, were selected. The viral particles isolated were observed by electron microscopy. The influenza virus was detected by the House Duplex/PCR and Gloria molecular biology tests. The results demonstrated that the Elaenia mesoleuca and Vireo olivaceus bird species are carrying the Influenza virus whilst crossing both the Northern and Southern hemispheres. To understand the role that these migrating birds may play in epidemic influenza, in Brazil, characterization of avian influenza subtypes will be done.

A vigilância do vírus da influenza, no periodo de janeiro a outubro de 2002, detectou a circulação da estirpe B/Hong Kong/330/2001 nas regiões Sudeste e Centro Oeste do país. Esta estirpe é uma variante do vírus da influenza B/Victoria/02/88 a qual teve sua circulação circunscrita no continente asiático por volta de 1991. Variantes da estirpe B/Hong Kong/330/2001 (B/Hong Kong/1351/2002) e da estirpe B/Sichuan/379/99 (B/Shizuoka/5/2001) também foram detectadas. A porcentagem de vírus caracterizados durante a epidemia foi de 34,88%, 15,12%, 15,12%, 4,65%, 15,12%, 13,95%, nas faixas etárias de 0-4, 5-10, 11-15, 16-20, 21-30, 31-50, respectivamente. Observou-se maior incidência da doença entre os indivíduos menores de 4 anos. Atribuiu-se a ausência de notificação nos indivíduos maiores de 60 anos ao contato prévio, desta faixa etária, com o vírus na década de 80. No mesmo período, detectou-se a circulação do vírus da influenza A/Panamá/2007/99-like (H3N2), por outro lado evidenciou-se a ausência da circulação da estirpe A(H1N1) até a presente data.

Through the influenza virus surveillance from January to October 2002, influenza B/Hong Kong-like strains circulating in the Southeast and Centre East regions of Brazil have been demonstrated. This strain is a variant from B/Victoria/02/88 whose since 1991 and until recently have been isolated relatively infrequently and have been limited to South-Eastern Asia. A total of 510 respiratory secretions were collected from patients 0 to 60 years of age, with acute respiratory illness, living in the Southeast and Centre East regions of Brazil, of which 86 (17.13%) were positive for influenza virus. Among them 12 (13.95%) were characterized as B/Hong Kong/330/2001; 3 (3.49%) as B/Hong Kong/1351/2002 a variant from B/Hong Kong/330/2001; 1 (1.16%) as B/Sichuan/379/99; 1 (1.16%) as B/Shizuoka/5/2001, until now. The percentages of cases notified during the surveillance period were 34.88%, 15.12%, 15.12%, 4.65%, 15.12%, 13.95%, in the age groups of 0-4, 5-10, 11-15, 16-20, 21-30, 31-50, respectively. The highest proportion of isolates was observed among children younger than 4 years but serious morbidity and mortality has not been observed among people older than 65 years, although B influenza virus component for vaccination campaign 2002 was B/Sichuan/379/99 strain. This was probably due to the elderly protection acquired against B/Victoria/02/88. In addition, in influenza A/Panama/2007/99-like (H3N2) strains 22 (25.58%) were also detected, but influenza A(H1N1) has not been detected yet.

Durante os meses de junho e julho de 1999, foram notificados 324 casos de doença respiratória aguda no Município de Iporanga-SP. O isolamento do vírus da influenza do tipo A/Bayern/07/95 (H1N1) e a conversão sorológica para a estirpe viral (H1N1) foram de 57,1% e 100%, respectivamente. A porcentagem de casos com diagnóstico clínico notificados durante a epidemia foi de 28,4%, 29,0%, 20,7%, 6,2% e 15,7%, nas faixas etárias de 0-4, 5-9, 10-14, 15-19 anos e indivíduos acima de 20 anos de idade, respectivamente. Observou-se maior incidência da doença entre os indivíduos menores de 14 anos. Atribui-se a ausência de notificação de casos em indivíduos maiores de 65 anos à campanha de vacinação, na população idosa de Iporanga, que em 1999 atingiu 72,4%. O virus isolado é genética e antigenicamente semelhante à estirpe A/Beijing/262/95 (H1N1), o componente H1 da vacina de 1999. Vacinas contendo a estirpe A/Beijing/262/95 (H1N1) estimularam, após imunização, anticorpos inibidores da hemaglutinação, os quais foram equivalentes em freqüência e título para ambas as estirpes: A/Bayern/07/95 e A/Beijing/262/95.

From June to July 1999 an outbreak of acute respiratory illness occurred in the town of Iporanga. Out of a total of 4,837 inhabitants, 324 cases were notified to the Regional Surveillance Service. Influenza virus was isolated from 57.1% of the collected samples and 100% seroconversion to influenza A (H1N1) was obtained in 20 paired sera tested. The isolates were related to the A/Bayern/07/95 strain (H1N1). The percentages of cases notified during the outbreak were 28.4%, 29.0%, 20.7%, 6.2% and 15.7% in the age groups of 0-4, 5-9, 10-14, 15-19 and older than 20 years, respectively. The highest proportion of positives was observed among children younger than 14 years and no cases were notified in people older than 65 years, none of whom had been recently vaccinated against influenza. These findings suggest a significant vaccine protection against A/Bayern/7/95, the H1 component included in the 1997-98 influenza vaccine for elderly people. This viral strain is antigenically and genetically related to A/Beijing/262/95, the H1 component of the 1999 vaccine. Vaccines containing A/Beijing/262/95 (H1N1) stimulated post-immunization hemagglutination inhibition antibodies equivalent in frequency and titre to both A/Beijing/262/95-like and A/Bayern/7/95-like viruses. Thus, this investigation demonstrates the effectiveness of vaccination against influenza virus in the elderly.

OBJETIVO: Comparar a sensibilidade do método de difusão em ágar e do método de extração utilizando as linhagens celulares RC-IAL (células fibroblásticas de rim de coelho) e HeLa (células epiteliais de carcinoma do colo do útero humano), na avaliação da citotoxicidade "in vitro" de materiais de uso médico-hospitalar. MATERIAL E MÉTODO: Foram testadas 50 amostras escolhidas por sorteio, entre as já conhecidamente positivas e negativas e identificadas como: algodão, espuma, borracha, látex, celulose e acrílico. Além, das amostras citadas foram testadas experimentalmente várias concentrações de SDS (duodecil sulfato de sódio) nas culturas celulares RC-IAL e HeLa. RESULTADOS: Das 50 amostras testadas , 44 (88%) foram positivas para os dois métodos. Mas quando comparado o SDS nos dois métodos foram observados resultados positivos nas concentrações de 0,5 a 0,05 µg/ml no método de difusão em ágar e no método de extração somente foi observado efeito citotóxico até a concentração de 0,25 µg/ml. CONCLUSÃO: Os resultados encontrados são similares aos observados por outros autores que testaram materiais como, por exemplo, ligas metálicas. Quando foi usado o SDS observou-se, nas duas linhagens celulares, diferenças favoráveis ao método de difusão em ágar em duas concentrações, isto é, a sensibilidade deste método foi significantemente maior, por inspecção, em relação ao método de extração, além de se constituir em método mais simples de ser realizado.

OBJECTIVE: A comparison of the sensitivity of the agar diffusion method with that of extraction using cell-lines RC-IAL (fibroblastic of rabbit kidney) and HeLa (epithelial carcionoma cells from the cervix uteri of the humam uterus), in the in vitro evaluation of materials of medical and hospital. MATERIAL AND METHODS: Fifteen samples chosen at random, from among the already known positives and negatives in our stock, were tested and identified as cotton, form, latex, cellulose and acrylic. Besides the samples mentioned, many SDS (GIbco) concentrations were tested experimentally in RC-IAL and HeLa cell cultures. RESULTS: Of the 50 samples tested, 44 (88%) were positive by both methods. However, when the SDS were compared by using the two methods, positive results were noted in the concentrations of from 0.5 to 0.05µg/ml in the agar diffusion ans extraction methods. A cytotoxic effect was only noted in the concentrations of up to 0.25 µg/ml. CONCLUSION: When the SDS was used, differences favorable to the agar diffusion method were observed in the two cell lines, in two concentrations; that is, the sensitivity of this method was quantitatively greater on inspection than that of the extraction method, as well as being the simpler method to use.

Uma linhagem celular de rim de coelho (denominada RC-IAL), que foi isolada em 1976, e que atualmente está na 150ª passagem, teve suas características analisadas. As células apresentaram morfologia semelhante aos fibroblastos desde o início de seu cultivo. A proporção de crescimento celular não se alterou desde seu isolamento, com uma eficiência de clonagem ao redor de 9%. A linhagem mostrou crescimento dependente de ancoragem, e a análise cromossômica apresentou o número modal da espécie com pequenas variações para mais ou menos um cromossomo, resultando uma somatória de 50%. Sua espécie de origem foi comprovada através da reação de imunofluorescência indireta e a susceptibilidade da linhagem a alguns vírus, com demonstração do efeito citopático, foi verificada com os vírus da vacínia, cowpox, herpes simples tipo 1 e 2 e da rubéola. Esse substrato celular está livre de contaminantes, satisfazendo assim, as condições para seu uso em trabalhos científicos, principalmente os relacionadas à saúde pública.

A rabbit kidney cell line RC-IAL, isolated in 1976 and at apresent at 150ª passage, has had its characteristics analysed. The cells presented morphology similar to fibroblasto throughout their culture. The cellular growth proportion remained unaltered from its isolation, with a cloning efficiency of around 9%. The line showed growth dependent on anchorage and chromosomic analysis presented the modal number of the species with small variations to about one chromosome, to a total of about 50%. The line's species of origin was confirmed through indirect imunofluorescence reaction and susceptibility to some viruses with cytopatic effect was verified with vaccinia, cowpox, herpes simplex types 1 and 2 and rubella viruses. This cellular substract is free from contaminating agents, thus satisfying the conditions for its use in scientific work, especially that relating to public health.
