Resumo Spondias mombin é uma espécie arbórea cujos frutos possuem sabor e aroma únicos e contêm compostos nutritivos e bioativos. Considerando que o conhecimento sobre a variabilidade genética em S. mombin é importante para o estabelecimento deum programa de melhoramento genético, objetivou-se caracterizar acessos de S. mombincoletados nos Estados do Maranhão e Piauí, Nordeste do Brasil, utilizando análisemolecular com repetições simples de sequência: marcadores (ISSR). Foram avaliados 26 acessos mantidos no banco de germoplasma da Embrapa Meio-Norte (Teresina-PI, Brasil) e dois acessos anões coletados em propriedade privada, em Teresina-PI. Os 12 marcadores ISSR empregadosapresentaram forte poder discriminatório demonstrado pelo alto percentual de polimorfismo(88,75%) e conteúdo médio de informação polimórfica (0,641). A altadiversidade genética entre os acessos foi demonstrada pelos índices de diversidade de Nei (H=0,37)e Shannon(I=0,55), enquanto o fluxo gênico entre populações foi considerado moderado(Nm=0,6268 migrantes por geração). A diversidade genética pode ser explicada,principalmente, pela variabilidade dentro das populações (88,71%) e não entre populações (11,29%), embora ocoeficiente de diferenciação genética entre populações tenha sido moderado (GST = 0,11) e estatisticamente significativo (P <0,01). O método de grupos de pares nãoponderados com agrupamento de média aritmética (UPGMA) e a análise de estrutura populacional mostraram grupos geneticamente distintos. Abstract: Spondias mombin is an arboreal species, the fruits of which possess a unique flavor and aroma and contain both nutritive and bioactive compounds. Considering that knowledge regarding the genetic variability in S. mombin is important for the establishment of a genetic improvement program, the aim was to characterize S. mombin accessions collected in the states of Maranhão and Piauí, northeastern Brazil, using molecular analysis with simple sequence repeats markers (ISSR). Twenty-six accessions maintained in the Embrapa Meio-Norte germplasm bank (Teresina, PI, Brazil) and two dwarf accessions collected from a private property in Teresina-PI were evaluated. The 12 ISSR markers employed presented strong discriminatory power as demonstrated by the high percentage of polymorphism (88.75%) and mean polymorphic information content (0.641). High genetic diversity among the accessions was demonstrated by the diversity indices of Nei (H=0.37) and Shannon (I=0.55), while gene flow between populations was considered moderate (Nm=0.6268 migrants per generation). The genetic diversity could be explained mainly by variability within-populations (88.71%) rather than between-populations (11.29%), although the genetic differentiation coefficient between-populations was moderate (GST=0.11) and statistically significant (P < 0.01). Unweighted pair group method with arithmetic mean (UPGMA) clustering and Population Structure analysis showed genetically distinct groups. The dwarf accessions were the most divergent and should be incorporated into the germplasm bank. Our study verified the existence of genetic differentiation between the tested S. mombin accessions and confirmed that such variability could be exploited through genetic improvement programs. ABSTRACT Objective Currently programmed cell death protein 1 (PD-1) inhibitors in combination with other therapies are being evaluated to determine their efficacy in cancer treatment. However, the effect of PD-ligand (L) 1 expression on disease outcomes in stage III (EC III) non-small cell lung cancer is not completely understood. Therefore, this study aimed to assess the influence of PD-L1 expression on the outcomes of EC III non-small cell lung cancer. Methods This study was conducted on patients diagnosed with EC III non-small cell lung cancer who underwent treatment at a tertiary care hospital. PD-L1 expression was determined using immunohistochemical staining, all patients expressed PD-L1. Survival was estimated using the Kaplan-Meier method. Relationships between variables were assessed using Cox proportional regression models. Results A total of 49 patients (median age=69 years) with EC III non-small cell lung cancer and PD-L1 expression were evaluated. More than half of the patients were men, and most were regular smokers. The patients were treated with neoadjuvant chemotherapy, surgery, or sequential or combined chemotherapy and radiotherapy. The median progression-free survival of the entire cohort was 14.2 months, and the median overall survival was 20 months. There was no significant association between PD-L1 expression and disease progression, clinical characteristics, or overall survival. Conclusions PD-L1 expression was not correlated with EC III non-small cell lung cancer outcomes. Whether these findings differ from the association with immune checkpoint inhibitors remains to be addressed in future studies. ABSTRACT The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. Abstract In this work, we analyzed cytogenetically eight Chactidae and Buthidae, including the localization of repetitive DNA sequences. The chactids possess monocentric chromosomes and the highest diploid numbers (2n=50 in Brotheas amazonicus, 2n=36 in Chactopsis amazonica, 2n=30 in Neochactas sp.) when compared with buthids (2n=10 in Tityus bahiensis, 2n=14 in Tityus apiacas and Tityus metuendus, 2n=18 in Tityus aba, 2n=26 in Ischnotelson peruassu). The localization of rDNA genes and (TTAGG)n sequences exhibited a conserved pattern of two terminal/subterminal ribosomal cistrons and terminal telomere signals. However, the comparison between the data of C-banding, DAPI after FISH and Cot-DNA fraction indicated a variable quantity and distribution of these regions, as follow: (i) positive heterochromatin and Cot-DNA signals (B. amazonicus and I. peruassu), (ii) small blocks of heterochromatin with large Cot-DNA signals (T. metuendus), (iii) positive heterochromatic regions and absence of Cot-DNA signals (T. aba and T. apiacas), and (iv) negative heterochromatin and Cot-DNA signals (T. bahiensis). BACKGROUND Cerebral malaria (CM) is a severe immunovasculopathy caused for Plasmodium falciparum infection, which is characterised by the sequestration of parasitised red blood cells (pRBCs) in brain microvessels. Previous studies have shown that some terpenes, such as perillyl alcohol (POH), exhibit a marked efficacy in preventing cerebrovascular inflammation, breakdown of the brain-blood barrier (BBB) and brain leucocyte accumulation in experimental CM models. OBJECTIVE To analyse the effects of POH on the endothelium using human brain endothelial cell (HBEC) monolayers co-cultured with pRBCs. METHODOLOGY The loss of tight junction proteins (TJPs) and features of endothelial activation, such as ICAM-1 and VCAM-1 expression were evaluated by quantitative immunofluorescence. Microvesicle (MV) release by HBEC upon stimulation by P. falciparum was evaluated by flow cytometry. Finally, the capacity of POH to revert P. falciparum-induced HBEC monolayer permeability was examined by monitoring trans-endothelial electrical resistance (TEER). FINDINGS POH significantly prevented pRBCs-induced endothelial adhesion molecule (ICAM-1, VCAM-1) upregulation and MV release by HBEC, improved their trans-endothelial resistance, and restored their distribution of TJPs such as VE-cadherin, Occludin, and JAM-A. CONCLUSIONS POH is a potent monoterpene that is efficient in preventing P. falciparum-pRBCs-induced changes in HBEC, namely their activation, increased permeability and alterations of integrity, all parameters of relevance to CM pathogenesis. ABSTRACT Objective: Evaluate the influence of isolated and associated prepregnancy obesity and gestational diabetes mellitus (GDM) on adverse perinatal outcomes. Materials and methods: Cross-sectional observational study with women who delivered at a Brazilian Maternity Hospital, between August and December 2020. Data were collected by interview with application form, and medical records. Sample was stratified by body mass index (BMI) and GDM screening in four groups: no obesity (BMI < 30 kg/m2) no GDM – reference; isolated GDM; isolated obesity (BMI ≥ 30 kg/m2); and obesity with GDM. Preeclampsia (PE), cesarean section (CS), large-for-gestational-age (LGA) newborn and admission to neonatal intensive care unit (NICU) were analyzed by odds ratio (OR) adjusted for confounding factors, adopting 95% confidence interval (CI) and P < 0.05 statistically significant. Results: From 1,618 participants, isolated obesity group (233/14.40%) had high chance of PE (OR = 2.16; CI: 1.364-3.426; P = 0.001), isolated GDM group (190/11.74%) had high chance of CS (OR = 1.736; CI: 1.136-2.652; P = 0.011) and NICU admission (OR = 2.32; CI: 1.265-4.261; P = 0.007), and obesity with GDM group (121/7.48%) had high chance of PE (OR = 1.93; CI: 1.074-3.484; P = 0.028), CS (OR = 1.925; CI: 1.124-3.298; P = 0.017) and LGA newborn (OR = 1.81; CI: 1.027-3.204; P = 0.040), compared with reference (1,074/66.38%). Conclusion: Obesity and GDM enhances the chance of different negative outcomes, worsening this prognosis when associated. Resumo Tendo em vista a semelhança morfológica entre tumores estromais gastrointestinais (GIST) e outros sarcomas do intestino de cães, objetivou-se realizar o diagnóstico histomorfológico e imunoistoquímico desses tumores, associando raça, sexo e idade, localização e invasão tumoral. Foram avaliados 217 casos por histopatologia e incluídos 36 diagnosticados por imuno-histoquímica (24 GIST e 12 outros sarcomas intestinais). Cães sem raça definida foram os mais diagnosticados com GIST, principalmente fêmeas idosas (9,5±2,2 anos); nos demais sarcomas intestinais, mestiços e Dachshunds, machos e fêmeas, foram igualmente acometidos. O ceco foi o mais acometido pelos GISTs, com invasão tumoral das camadas intestinais em todos os casos. O intestino delgado foi o mais acometido pelos demais sarcomas intestinais, com invasão das camadas na maioria desses tumores. Abstract In view of the morphological similarity between gastrointestinal stromal tumors (GIST) and other sarcomas of the intestine of dogs, the aim was to carry out the histomorphological and immunohistochemical diagnosis of these tumors, associating breed, sex and age, location and tumor invasion. 217 cases were evaluated by histopathology and 36 diagnosed by immunohistochemistry were included (24 GIST and 12 other intestinal sarcomas). Mixed breed dogs were the most diagnosed with GIST, mainly elderly females (9.5±2.2 years); in the other intestinal sarcomas, crossbreeds and Dachshunds, males and females, were equally affected. The cecum was the most affected by GISTs, with tumor invasion of the intestinal layers in all cases. The small intestine was the most affected by the other intestinal sarcomas, with invasion of the layers in most of these tumors. GISTs expressed markers such as CD117 and DOG-1, unlike other intestinal sarcomas. Resumo Discute o modelo de eugenia empregado pelo médico-sanitarista Belisário Penna durante a campanha do saneamento do Brasil nas décadas de 1920 e 1930. São abordados dois conceitos fundamentais para o seu pensamento: “raça brasileira” e eugenia “preventiva”. A maneira como Penna avaliava a questão racial brasileira era fundamental para aderir à concepção eugênica aliada à medicina social e ao seu projeto de “consciência sanitária”. O texto oferece uma perspectiva de como foi bordada a concepção eugênica de Penna e o diálogo estabelecido com o movimento eugênico brasileiro, em especial com o eugenista Renato Kehl. Assim, a defesa de uma eugenia classificada como “preventiva” estabelecia coerência para um projeto reformista social via o saneamento defendido por Penna.
Abstract The purpose of this article is to discuss the eugenics model used by the medical-sanitarist Belisário Penna during the sanitation campaign in Brazil in the 1920s and 1930s. The article addresses two fundamental concepts for his thinking: “Brazilian race” and “preventive” eugenics. The way in which Penna saw the Brazilian racial issue was fundamental to adhere to the eugenic conception combined with social medicine and its project of “health awareness.” The text offers a perspective of how Penna’s eugenics was conceived and the dialogue established with the Brazilian eugenic movement, especially with the eugenicist Renato Kehl. Thus, the defense of an eugenics classified as “preventive” established a coherence for a social reform project through the sanitation defended by Penna.

Resumo A produção de ceramida ocorre em todo o corpo e desempenha um papel importante na manutenção da fisiologia normal. No entanto, os níveis de ceramidas são alterados em estados de doença, principalmente durante o desenvolvimento de diabetes e dislipidemia. A produção de ceramidas também está associada à instabilidade das placas ateroscleróticas. Estudos recentes revelam que pacientes com doença arterial coronariana instável apresentam níveis plasmáticos aumentados de ceramidas (principalmente C16, C18 e C24:1). Atualmente, são consideradas biomarcadores emergentes nas doenças cardiovasculares, sendo utilizadas na predição de instabilidade da placa aterosclerótica e eventos cardiovasculares adversos de forma independente aos fatores de risco tradicionais. Com o objetivo de descrever e discutir o papel das ceramidas na estratificação das doenças cardiovasculares, o desenvolvimento desta revisão narrativa contextualiza a importância desse biomarcador no cenário atual da cardiologia.
Abstract Ceramide production takes place throughout the body and plays a key role in the maintenance of normal physiology. However, ceramide levels are altered during disease states, particularly considering the development of diabetes and dyslipidemia. Ceramide production is also associated with atherosclerotic plaque instability. Recent studies revealed that patients with unstable coronary artery disease (CAD) presented increased plasma ceramide levels (especially C16, C18, and C24:1). These molecules are currently considered emerging biomarkers of cardiovascular diseases (CVD), being used for predicting atherosclerotic plaque instability and adverse cardiovascular events independently from traditional risk factors. With the aim of describing and discussing the role of ceramides in the stratification of cardiovascular diseases, this narrative review contextualizes the importance of this biomarker in the present cardiology scenario.

RESUMO O desenvolvimento de programas de melhoramento genético requer o conhecimento dos níveis de diversidade presente nas populações, desse modo o objetivo desse estudo foi caracterizar com base em descritores morfológicos a diversidade genética de variedades crioulas de feijão-fava (Phaseolus lunatus L.). A diversidade genética foi estimada a partir dos métodos multivariados que selecionaram variedades crioulas atendendo tanto os agricultores como o mercado consumidor. De acordo com as características morfológicas propostas pelo Biodiversity International, os genótipos UFPI-667 e UFPI-682 são os mais divergentes e apresentam alto potencial para serem utilizados em cruzamentos por serem geneticamente distantes e complementares. A análise de componentes principais detectou associações fenotípicas positivas e significativas entre as variáveis: comprimento da vagem, largura da vagem e produção de grãos. Ao usar métodos multivariados, as variedades crioulas UFPI-666, UFPI-650, UFPI-651, UFPI-687, UFPI-658, UFPI- 673, UFPI-667 e UFPI-674 são candidatos à seleção por apresentarem ciclos mais curtos e maiores produções de grãos, características importantes para o melhoramento do feijão-fava. Este estudo mostrou que as análises multivariadas podem ser usadas como uma ferramenta eficaz para encontrar características potenciais em feijão-fava e pode auxiliar os melhoristas de feijão-fava na seleção de variedades locais superiores. Os acessos UFPI- 667 e UFPI-682 podem ser indicados como genitores em programas de melhoramento genético, pois são distantes geneticamente e complementares em suas características.
ABSTRACT The knowledge of genetic diversity in plant population is essential to the success of plant breeding programs. Thus, previous characterization of landraces is one of the first steps before the indication of genotypes to these plant breeding programs. This study aimed to characterize, through morphological traits, and estimate the genetic diversity in landraces of lima bean (Phaseolus lunatus L.). Genetic diversity was estimated by using multivariate methods that selected individual landraces to be indicated for farmers and consumers. According to morphological traits proposed by Biodiversity International, the genotypes UFPI-667 and UFPI- 682 showed higher dissimilarity and high potential to be used in crosses since they are genetically distant and complementary. The principal component analysis identified positive and significant phenotypic associations between variables: pod length, pod width, and grain production. According to the multivariate methods used, the landraces UFPI-666, UFPI-650, UFPI-651, UFPI-687, UFPI-658, UFPI-673, UFPI-667, and UFPI-674 are potential candidates for selection since they have relevant features for lima bean breeding, such as shorter cycles and higher grain production. This study showed that multivariate analysis can be used as an effective tool to find potential traits in lima bean and can assist the lima bean breeders in the selection of potential landraces. Accessions UFPI-667 and UFPI-682 can be indicated as genitors for breeding programs, as they are genetically distant and complementary in their characteristics.

RESUMO O feijão-fava (Phaseolus lunatus L.) é a segunda espécie de maior importância socioeconômica do gênero, constituindo alternativa alimentar na forma de grãos verdes ou maduros. Sendo uma opção de renda para agricultores familiares, a falta de variedades superiores dificulta a recomendação considerando a peculiar variabilidade do feijão-fava. Objetivou-se selecionar variedades crioulas de feijão-fava com base em caracteres agronômicos, visando a utilização em programas de melhoramento e posterior recomendação para agricultores. Foram conduzidos ensaios de avaliação com 14 variedades crioulas de feijão-fava, nos municípios de São Domingos do Maranhão - MA, Teresina - PI, Bom Jesus - PI e Tianguá - CE. Foram avaliados: número de dias até o início da floração, número de dias até a maturação das vagens, comprimento da vagem, largura da vagem, espessura da vagem, número de sementes por vagem, peso de 100 sementes e produtividade de grãos. Os dados foram submetidos à análise de variância univariada, para determinação da variabilidade genética em diferentes ambientes, e, em seguida, às análises multivariada e de agrupamento. As variedades crioulas apresentam divergência genética, mas não se agrupam segundo origem geográfica, demonstrando a existência de similaridade entre o germoplasma de comunidades rurais estados vizinhos. As variedades Boca de Moça, Raio de Sol e Fava Branca CE são as mais precoces; Boca de Moça, Rajada e Raio de Sol as de vagens mais longas e sementes maiores; e Boca de Moça, Rajada e Mulatinha são as mais produtivas, o que as credencia para recomendação para agricultores familiares e/ou incorporação em programas de melhoramento do feijão- fava.
ABSTRACT Lima bean (Phaseolus lunatus L.) is the second most important socioeconomic species of the genus, consisting of a food alternative as green or mature beans. It is an income option for family farmers and the lack of superior varieties makes its recommendation difficult, considering the peculiar lima bean variability. Thus, aimed to select landraces of lima beans based on desirable agronomic traits, enabling their use in breeding programs and later recommendations to family farmers. Evaluation trials were carried out with 14 landraces of lima beans in the municipalities of São Domingos do Maranhão - MA, Teresina - PI, Bom Jesus - PI, and Tianguá - CE. The agronomic traits were evaluated: number of days until flowering, number of days until pod maturation, pod length, pod width, pod thickness, number of seeds per pod, 100-seed weight, and grain yield. The data were initially subjected to univariate analysis of variance to determine the genetic variability in different environments and, subsequently, to multivariate and cluster analyses. The evaluated landraces showed genetic divergence, not being grouped according to geographic origin, demonstrating the existence of similarity between germplasms of rural communities in neighboring states. The varieties Boca de Moça, Raio de Sol, and Fava Branca CE are the earliest; Boca de Moça, Rajada, and Raio de Sol presented the longest pods and largest seeds; and Boca de Moça, Rajada, and Mulatinha are the most productive. Therefore, it qualifies them for recommendation to family farmers and/or incorporation in lima bean breeding programs.

Intravenous whole blood transfusion results in faster recovery of vascular integrity and increased survival in experimental cerebral malaria

BACKGROUND Cerebral malaria is a lethal complication of Plasmodium falciparum infections in need of better therapies. Previous work in murine experimental cerebral malaria (ECM) indicated that the combination of artemether plus intraperitoneal whole blood improved vascular integrity and increased survival compared to artemether alone. However, the effects of blood or plasma transfusion administered via the intravenous route have not previously been evaluated in ECM. OBJECTIVES To evaluate the effects of intravenous whole blood compared to intravenous plasma on hematological parameters, vascular integrity, and survival in artemether-treated ECM. METHODS Mice with late-stage ECM received artemether alone or in combination with whole blood or plasma administered via the jugular vein. The outcome measures were hematocrit and platelets; plasma angiopoietin 1, angiopoietin 2, and haptoglobin; blood-brain barrier permeability; and survival. FINDINGS Survival increased from 54% with artemether alone to 90% with the combination of artemether and intravenous whole blood. Intravenous plasma lowered survival to 18%. Intravenous transfusion provided fast and pronounced recoveries of hematocrit, platelets, angiopoietins levels and blood brain barrier integrity. MAIN CONCLUSIONS The outcome of artemether-treated ECM was improved by intravenous whole blood but worsened by intravenous plasma. RESUMO Objetivo: Analisar os efeitos da estimulação elétrica neuromuscular do quadríceps femoral associado ou não à suplementação com whey protein na atividade eletromiográfica e distribuição de massa corporal em voluntários submetidos à reconstrução do ligamento cruzado anterior. Métodos: 24 voluntários foram divididos em três grupos: controle basal, whey protein associado com estimulação elétrica neuromuscular e estimulação elétrica neuromuscular isolada. Resultados: Na avaliação pós-operatória, durante o miniagachamento, o grupo controle basal demonstrou diminuição da atividade eletromiográfica do vasto medial (p = 0,005, olhos abertos; p = 0,003, olhos fechados), vasto lateral (p = 0,005, olhos abertos; p = 0,020, olhos fechados) e reto femoral (p = 0,075, olhos abertos; p = 0,074, olhos fechados) e da distribuição de massa corporal no membro operado (p < 0,001, olhos abertos; p < 0,001, olhos fechados) e membro lesionado (p < 0,001, olhos abertos; p < 0,001, olhos fechados). Conclusão: O uso precoce de estimulação elétrica neuromuscular do quadríceps femoral, independentemente do uso de whey protein, foi eficaz para manter a atividade eletromiográfica dos músculos vasto medial e vasto lateral, e prevenir assimetrias na distribuição de massa corporal 15 dias após a reconstrução do ligamento cruzado anterior. ABSTRACT Objective: To analyze the effects of neuromuscular electrical stimulation of the femoral quadriceps associated or not with whey protein supplementation on the electromyographic activity and body mass distribution in volunteers undergoing anterior cruciate ligament reconstruction. Methods: 24 volunteers were randomly divided into three groups: basal control, whey protein in association with neuromuscular electrical stimulation, and neuromuscular electrical stimulation alone. Results: In the postoperative evaluation, during the mini squat, the basal group showed a decrease in the electromyographic activity of the vastus medialis (p = 0.005, eyes open; p = 0.003, eyes closed), vastus lateralis (p = 0.005, eyes open; p = 0.020; eyes closed) and rectus femoris (p = 0.075, eyes open; p = 0.074, eyes closed) and of body mass distribution in the injured limb (p < 0.001, eyes open; p < 0.001, eyes closed), and in the healthy limb (p < 0.001, eyes open; p < 0.001, eyes closed). Conclusion: The early use of neuromuscular electrical stimulation of the quadriceps femoris maintained the electromyographic activity of the vastus medialis and vastus lateralis muscles and prevented asymmetries in body mass distribution 15 days after anterior cruciate ligament reconstruction.
