Abstract The hybrid of Cattleya forbesii x Cattleya loddigesii has a short stature and early flowering, a light lilac tending to white color and a labellum with a velvety purple lobe and corrugated edges. The flower lifetime lasts approximately 20 days.

Resumo Este estudo teve como objetivo determinar a proporção de NO3-/NH4+, fornecidos por fertirrigação que permita o melhor crescimento de plantas de rosa do deserto cultivadas em vaso. As plantas foram cultivadas em casa de vegetação, em vasos de polipropileno, tendo como substrato uma mistura de areia e pó de pinus compostado (1:1, v v-1). Utilizou-se o delineamento experimental inteiramente casualizado, com seis tratamentos (controle, 0/100 25/75, 50/50, 75/25 e 100/0, de NO3– e NH4+ respectivamente), e dez repetições. As fertirrigações foram realizadas semanalmente na quantidade de 100 mL por vaso durante um período de 180 dias. As variáveis avaliadas foram: altura da parte aérea, diâmetro basal do cáudice, número dos ramos, massa seca de raízes, folhas e cáudice e determinação dos teores e acúmulo de macronutrientes de folha, cáudice e raiz. As proporções de NO3– e NH4+ promoveram maior alteração no teor de nutrientes nas folhas comparado ao cáudice e raízes. Recomenda-se para o cultivo de rosa do deserto a proporção 25/75 de NO3-/NH4+, por proporcionar melhor acúmulo de massa seca pelas plantas, bem como aumento na altura e diâmetro do cáudice das plantas.

Abstract This study aimed to determine NO3-/NH4+ proportion delivered by fertigation that allows greater desert rose plants growth in pots. Plants were cultivated in greenhouse in polypropylene pots, using a mixture of sand and composted Pinus powder (1:1, v v-1) as substrate. Experimental design was completely randomized, with six treatments (control, 0/100 25/75, 50/50, 75/25 and 100/0 of NO3– and NH4+ respectively), and ten replications. Fertigation was performed weekly in the amount of 100 mL per pot for 180 days. Variables evaluated were: shoot height, basal diameter of the caudice, number of branches, roots, leaves and caudice dry mass and macronutrient contents determination and accumulation in leaves, caudice and root. NO3– and NH4+ proportion promoted a greater change in nutrient content in leaves compared to caudice and roots. The proportion 25/75 NO3–/NH4+ is recommended for desert rose cultivation, as it provides the best results for most of the characteristics studied.

RESUMO Uma alternativa para a conservação de espécies de orquídeas ameaçadas de extinção é a preservação por longos períodos de tempo das sementes por criopreservação. O objetivo deste trabalho foi avaliar a influência de soluções criogênicas na criopreservação de sementes das orquídeas Epidendrum radicans e Arundina bambusifolia em nitrogênio liquído. Foram realizados sete tratamentos: sacarose 0,4 M; glicerol 2 M; PVS1; PVS2; PVS3; PVS2 com 1% de floroglucinol e o controle. Após a retirada do nitrogênio líquido, metade das sementes foi submetida ao teste de tetrazólio, e outra metade foi cultivado in vitro. Após 30 dias foi avaliada a germinação das sementes. No teste de tetrazólio e na avaliação da germinação das sementes de A. bambusifolia, não houve diferença significativa entre os tratamentos de E. radicans. Por outro lado, no teste de tetrazólio e no teste de germinação das sementes de A. bambusifolia, observou-se que as soluções de PVS1, PVS2 e PVS2 + 1% de floroglucinol apresentaram os melhores resultados. A criopreservação de sementes de E. radicans com 4% de teor de água não requer o uso de crioprotetores. Por outro lado, a utilização dos crioprotetores apresentou efeito positivo na manutenção e preservação da viabilidade de sementes de A. bambusifolia, sendo que a utilização das soluções de PVS1, PVS2 e PVS2 acrescentado de 1% floroglucinol, apresentam melhores taxas de sobrevivência das sementes após criopreservação em nitrogênio líquido.

ABSTRACT An alternative for the conservation of orchid species threatened with extinction is the seeds preservation for long periods by cryopreservation. This work aimed to evaluate the influence of cryogenic solutions on the orchids Epidendrum radicans and Arundina bambusifolia seeds cryopreservation in liquid nitrogen. Seven treatments were performed: 0.4 M sucrose; 2 M glycerol; PVS1; PVS2; PVS3; PVS2 + 1% phloroglucinol and the control. After liquid nitrogen removal, half of the seeds were subjected to tetrazolium test, and another half was in vitro cultured. After 30 days seed germination was evaluated. There was no significant difference between A. bambusifolia and E. radicans in tetrazolium test and seeds germination. On the other hand, it was observed that in A. bambusifolia tetrazolium and seed germination tests the solutions of PVS1, PVS2, PVS2 + 1% phloroglucinol showed the best results. E. radicans seeds with 4% water content cryopreservation does not require the use of cryoprotectant. On the other hand, cryoprotectant's use resulted in positive effect in maintenance and viability preservation of A. bambusifolia seeds. PVS1, PVS2 and PVS2 + 1% phloroglucinol showed best seeds survival rates after cryopreservation in liquid nitrogen.

RESUMO Adenium obesum, também conhecida vulgarmente como rosa-do-deserto, tem se popularizado no Brasil nos últimos anos devido à sua exuberância e forma escultural. Porém, a produção da espécie em escala comercial tem enfrentado desafios em relação à propagação de seus híbridos por técnicas de cultivo in vitro, escolha de substratos, manejo da irrigação e fertilização, controle de doenças e pragas, dentre outros. Diferente de outras espécies comercializadas como planta de vaso, ainda não existem protocolos de produção comercial totalmente estabelecidos para Adenium, devido, principalmente, à escassez de informação técnica sobre a espécie. Assim, esse artigo técnico teve o intuito de trazer à luz da literatura algumas informações relacionadas ao cultivo da espécie, tendo em vista a sua produção em escala comercial. Além disso, abordou-se aspectos da morfologia floral, polinização e produção de sementes da espécie, bem como estratégias de propagação vegetativa.

ABSTRACT Adenium obesum, also known as desert rose, has recently become popular in Brazil, due to its exuberance and sculptural shape. However, the production of the species in commercial scale has been facing challenges regarding propagation of hybrids by in vitro cultivation techniques, potting medium composition, irrigation and fertilization management, disease and pest control. Unlike other species sold as potted plants, there are no commercial production protocols established for Adenium, mainly due to the scarcity of technical information about this species. Thus, this technical article aimed to provide information related to the Adenium growth, in view of its commercial scale production. In addition, aspects related to floral morphology, pollination and seed production of the species, as well as, vegetative propagation strategies were discussed.

RESUMO O híbrido, obtido do retrocruzamento entre (Cattleya labiata x Cattleya forbesii) x Cattleya labiata é uma planta vigorosa, bi ou unifoliada, apresenta pseudobulbos delgados e cilíndricos e folhas coriáceas com tonalidade verde escuro, com formato oblanceolado, de pontas obtusas, apresentando o primeiro florescimento após quatros anos da semeadura. O florescimento em Londrina ocorreu duas vezes no ano, entre os meses de abril e maio e outubro e novembro com 2-4 flores por pseudobulbo e durabilidade entre 15 a 20 dias. As flores do novo híbrido possuem coloração lilás e labelo com um centro amarelo e listras roxas.

ABSTRACT The hybrid, obtained by back crossing between (Cattleya labiata x Cattleya forbesii) x Cattleya labiata is a vigorous plant, bi-or unifoliate, features slender and cylindrical pseudobulbs and leathery dark-greenish leaves, with oblanceolate shape format of blunt tips with the first flowering four years after sowing. In Londrina, flowering occurred twice a year, between the months of April and May and October and November with 2-4 flowers per pseudobulb and durability ranging from 15 to 20 days. The flowers of the new hybrid were purple with a labellum with a yellow center and purple stripes.

RESUMO Dentre as técnicas utilizadas pela biotecnologia para multiplicação de mudas de orquídea, o cultivo in vitro destaca-se pelo elevado potencial de produção de novas plântulas. A presença de algumas substâncias químicas utilizadas no meio de cultura, dependendo da concentração, pode favorecer ou não o crescimento e o desenvolvimento da planta. Diante do exposto, objetivou-se, com este trabalho, avaliar o efeito do glifosato no cultivo in vitro da orquídea Cattleya nobilior Rchb. F. O delineamento experimental utilizado foi inteiramente casualizado, com seis tratamentos e dez repetições. Os tratamentos foram constituídos de seis concentrações de glifosato (0,0; 8,5; 17,0; 25,5; 34,0 e 42,5 mg L-1 e.a.), acrescidas ao meio de cultura MS modificado, com metade das concentrações de macronutrientes. As plântulas submetidas às diferentes concentrações de glifosato foram obtidas de sementes germinadas in vitro. As avaliações de comprimento e de biomassas secas de parte aérea e de raízes, de número de folhas, de comprimento da folha, de largura da folha, do número de brotos dos teores de clorofilas a, b e de carotenoides foram realizadas 200 dias após o transplantio das plântulas. Dentre os componentes fitométricos analisados, apenas o comprimento, a biomassa seca de raízes e o teor de carotenoides diminuíram com o incremento das doses do herbicida glifosato. Baixas concentrações de glifosato adicionadas ao meio de cultura resultam em efeito hormese, com aumento do crescimento da parte aérea de Cattleya nobilior Rchb. F.

ABSTRACT Among the techniques used by biotechnology for multiplication of orchid seedlings, the in vitro cultivation stands out due to the high production potential of new seedlings. The presence of some chemical substances used in the culture medium, depending on the concentration, may or may not favor the growth and development of the plant. Therefore, the objective of this work was to evaluate the effect of glyphosate on the in vitro cultivation of the orchid Cattleya nobilior Rchb. F. The experimental design was completely randomized, with six treatments and ten replications. The treatments consisted of six concentrations of glyphosate (0.0, 8.5, 17.0, 25.5, 34.0, and 42.5 mg L-1 a.i.), added to the modified MS culture medium with half the concentrations of macronutrients. Seedlings subjected to different glyphosate concentrations were obtained from seeds germinated in vitro. The length and dry biomass measurements of the shoot and root, number of leaves, leaf length, leaf width, number of shoots, chlorophyll a, b, and carotenoid contents were performed 200 days after transplanting of the seedlings. Among the phytometric components analyzed, only the length and dry biomass of root and the carotenoid content decreased with increasing doses of the glyphosate herbicide. Low concentrations of glyphosate added to the culture medium result in hormese effect, with increased growth of Cattleya nobilior Rchb. F. shoot.

RESUMO A rosa do deserto (Adenium obesum) tem se destacado no mercado interno, no entanto não existem muitos estudos relacionados às suas exigências quanto à nutrição e substratos. O objetivo deste estudo foi avaliar o efeito de diferentes fertilizações e suas interações com diferentes substratos no crescimento inicial de rosa do deserto. As mudas de rosa do deserto com 60 dias foram transplantadas para vasos com capacidade de 0,415 L, preenchidos com dois tipos de substrato: areia + fibra de coco e areia + pó de pinus compostado. Foram realizados quatro diferentes manejos de fertilização: sem fertilização, solução nutritiva de Hoagland e Arnon; aplicação de NH3NO4 na concentração de 590 mg L-1 de nitrogênio (N); e aplicação da solução nutritiva de Hoagland & Arnon modificada pela adição de NH3NO4, elevando a concentração de N para 800 mg L-1. As soluções foram aplicadas quinzenalmente adicionando 50 mL por vaso. O delineamento experimental foi inteiramente casualizado em esquema fatorial 2x4, com 10 repetições. Após 150 dias do início das fertilizações foram avaliados as seguintes variáveis fitométricas: altura da parte aérea; diâmetro basal do cáudice; número dos ramos; massa seca de raízes, cáudice e folhas; e determinação dos teores e acúmulo de macronutrientes da parte aérea, dos substratos foram avaliados o pH e condutividade elétrica. Os dados foram submetidos à análise de variância com posterior comparação de médias pelo teste de Tukey a 5% de probabilidade de erro. Para todas as variáveis fitométricas avaliadas os tratamentos contendo a mistura de areia + pó de pinus compostado, apresentaram maior desenvolvimento de mudas comparado a utilização de areia + fibra de coco. As mudas apresentaram melhor desempenho com a solução de Hoagland modificada para a altura, número de brotos, diâmetro de cáudice, e massa seca de folha em amos os substratos. O uso do substrato areia + pó de pinus e a fertilização com solução de Hoagland e Arnon combinada com solução de NH3NO4 proporcionou melhor crescimento inicial em rosa do deserto.

ABSTRACT The desert rose (Adenium obesum) has been highlighted in the internal market, however there are not many studies related to its demands regarding its nutrition and substrates. The aim of this study was to evaluate the effect of different fertilizations and its interactions with different substrates in the initial growth of the desert rose. The seedlings of desert rose with 60 days have been potted transplanted with a capacity of 0.415 L, filled with two kinds of substrates: sand + coconut fiber and sand + composted pine powder. Four different fertilization managements have been done: without fertilization, application of the Hoagland & Arnon nutrient solution; application of NH3NO4 at 590 g L-1 of nitrogen (N); and the application of the Hoagland & Arnon nutrient solution modified through the addition of NH3NO4, increasing the concentration of N to 800 g L-1. The solutions have been applied biweekly adding 50 mL per pot. The experiment has been entirely randomized in a 2x4 factorial design, with 10 repetitions. After 150 days from the beginning of the fertilizations the phytometrical parameters evaluated were: height of the shoot; diameter of the basal caudex; number of shoots; dry mass of the roots, caudex and leaves; and determination of the levels and accumulation of macronutrients in the shoot, and the pH and electrical conductivity of the substrates. The evaluated data have been submitted to an analysis of variance and the Tukey test compared through the test at 5% of error probability. For all the evaluated parameters, when the composted pine mixture was used, the seedlings showed a higher development compared to coconut fiber mixture. The seedlings had a better performance with the modified Hoagland solution for height, number of shoots, caudex diameter and leaf dry mass in both substrates. The use of the sand + pine powder (1:1 v v-1) and the fertilization with Hoagland & Arnon solution combined with the NH3NO4 solution provided a better initial growth on the desert rose.

RESUMO O cultivo in vitro é um método de exímia importância na conservação de germoplasma e na multiplicação de plantas ameaçadas de extinção, porém necessita de estudos sobre a adequação dos nutrientes utilizados para um bom desenvolvimento de cada espécie. O objetivo foi avaliar o desenvolvimento in vitro de Laelia marginata Lindl. em meios de cultura contendo diferentes relações nitrato/amônio. As plântulas foram obtidas a partir de sementes germinadas in vitro. Os tratamentos consistiram em diferentes relações nitrato/amônio, sendo cinco proporções NO3- e NH4+: T1-0/100; T2-25/75; T3-50/50; T4-75/25 e T5-100/0. Aos 200 dias após o transplante das plântulas, foram avaliados altura da parte aérea, comprimento de raiz, número de folhas, número de brotos, comprimento da folha, largura da folha, massa seca da parte aérea e de raiz, teor de clorofila a, b e carotenoides. Utilizou-se delineamento inteiramente casualizado, sendo cinco tratamentos e dez repetições. A combinação de 50/50 nitrato/amônio resultou nos maiores valores de comprimento de parte aérea e raiz, massa seca de parte aérea e raiz, bem como comprimento e largura da folha. A proporção de 50/50 de nitrato/amônio resultou no melhor desenvolvimento inicial das plântulas de L. marginata.

ABSTRACT In vitro cultivation is a very important method for the conservation of germplasm and the multiplication of endangered plants; however, studies on the adequacy of the nutrients used for a good development of each species are needed. The objective of this study was to evaluate the in vitro development of Laelia marginata Lindl. in culture media containing different nitrate/ ammonium ratios. Seedlings were obtained from seeds germinated in vitro. The treatments consisted of different nitrate/ ammonium ratios, with five proportions of NO3- and NH4+: T1-0/100; T2-25/75; T3-50/50; T4-75/25 and T5-100/0. At 200 days after seedling transplanting, height of the aerial part, root length, number of leaves, number of shoots, leaf length, leaf width, aerial and root dry mass and contents of chlorophyll a, b and carotenoids were evaluated. A completely randomized design was used, with five treatments and ten replicates. The combination of 50/50 nitrate/ammonium resulted in the highest values of aerial part and root length, dry mass of the aerial part and root, as well as leaf length and width. The proportion of 50/50 nitrate/ammonium resulted in the best initial development of L. marginata seedlings.

Abstract The hybrid between Cattleya forbesii x Cattleya bowringiana shows flowers of lilac coloration, labellum with a light yellow center and striated with purple stripes of the same tone of other floral whorls. The plant flourished four years after seeding and produced, on average, six flowers.

Resumo Objetivou-se nesse estudo realizar a descrição de frutos e sementes e do processo de germinação de rosa do deserto, a partir de dois anos de observações. Os frutos e as sementes foram caracterizados com base no comprimento, diâmetro e número de sementes por fruto; também, descreveu-se a estrutura interna das sementes e o processo de germinação. Realizaram-se testes de germinação às temperaturas de 25 e 30 ºC, determinando-se a porcentagem e índice de velocidade de germinação. As sementes colhidas em 2013 foram armazenadas por 12 meses e submetidas a novo teste de germinação. Paralelo a esses testes determinou-se a curva de embebição das sementes em substrato sobre e entre papel às temperaturas de 15, 20, 25 e 30 ºC. Os frutos e as sementes apresentaram comprimentos e diâmetros semelhantes nos dois anos de observações. As sementes podem ser armazenadas por até 12 meses sem perdas na viabilidade e as temperaturas de 25 e 30 ºC são adequadas para realizar o teste de germinação dessa espécie. A curva de absorção de água das sementes de rosa do deserto segue um padrão trifásico de embebição.

This study has aimed to carry out the description of fruits and seeds and germination process of desert rose, from two years of observations. The fruits and seeds were characterized based on length, diameter and number of seeds per fruit. The seeds internal structure and germinating process were also described. Germination test was performed at 25 and 30 °C temperatures, determining the germination percentage and germination speed index. Seeds harvested in 2013 were stored for 12 months and submitted to a new germination test. Parallel to these tests, the seeds imbibition curve was determined in substrates over and between sheets of paper at 15, 20, 25 and 30 °C temperatures. The fruits and seeds had similar lengths and diameters in both years of observations. The seeds can be stored for up to 12 months without loss in viability and temperatures of 25 and 30 °C are suitable for performing the germination test of this species. The water absorption curve of desert rose seeds follows a triphasic pattern of soaking.
