Total rupture of Achilles tendon induces inflammatory response and glial activation on the spinal cord of mice

Rupture of Achilles tendon is a common accident affecting professional and recreational athletes. Acute and chronic pain are symptoms commonly observed in patients with rupture. However, few studies have investigated whether Achilles tendon rupture is able to promote disorders in the central nervous system (CNS). Therefore, the current study aimed to evaluate nociceptive alterations and inflammatory response in the L5 lumbar segment of Balb/c mice spinal cord after Achilles tendon rupture. We found increased algesia in the paw of the ruptured group on the 7th and 14th days post-tenotomy compared with the control group. This phenomenon was accompanied by overexpression of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase-2 (NOS-2) as well as hyperactivation of astrocytes and microglia in nociceptive areas of L5 spinal cord as evidenced by intense GFAP and IBA-1 immunostaining, respectively. Biochemical studies also demonstrated increased levels of nitrite in the L5 spinal cord of tenotomized animals compared with the control group. Thus, we have demonstrated for the first time that total rupture of the Achilles tendon induced inflammatory response and nitrergic and glial activation in the CNS in the L5 spinal cord region.

Abstract: Currently, the research of new natural compounds with biological potential demonstrates great ethnopharmacological importance. In this study, we evaluated the biological properties promoted by saline extract from Malpighia emarginata DC leaves, whose objective is to evaluate the antioxidant, antimicrobial and cytotoxicity potential. Phytochemical characterization was performed by UPLC-MS chromatography to identify the chemical compounds. For the antioxidant potential, DPPH, ATT and FRAP methods were used. The antibacterial and antifungal tests were performed evaluating the MIC50, MIC90, CMB and CMF parameters. Moreover, antibiofilm action was evaluated. Cytotoxicity and proliferation were performed using splenocytes from Balb/c mice and were evaluated by cytometry. We found a list of phenolic compounds among other bioactive compounds in the M. emarginata saline extract. In addition, higher antioxidant profile and antifungal activity against different strains of Candida spp. was promoted by the saline extract. Splenocytes showed greater cell viability (more than 90%) and showed higher proliferate index in 24 and 48 hours of incubation with the extract. Saline extract from Malpighia emarginata DC has potential action like antioxidant and antifungal agent without promote animal cell damage.

Resumo Apresenta-se uma síntese atualizada de informações sobre algas no Brasil objetivando refinar os dados reunidos até o presente, bem como avaliar os avanços sobre o conhecimento da diversidade de algas no país desde a publicação do Catálogo de Plantas e Fungos do Brasil. Os resultados de 2015 mostraram um acréscimo de 1.250 espécies (35.7%) a um total de 4.747 em relação a 2010. As classes mais diversas em número de espécies foram Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae e Euglenophyceae. Bacillariophyceae e Cyanophyceae tiveram o maior acréscimo de espécies no intervalo de cinco anos. A região Sudeste e Sul foram as mais diversas, porém, as regiões Nordeste com os estados do Piauí e Sergipe e Centro-Oeste com os estados de Mato Grosso, Goiás e Distrito Federal destacaram-se no cenário da biodiversidade nacional. Apesar da escassez de taxonomistas e da infraestrutura limitada, os resultados obtidos evidenciaram um avanço significativo no conhecimento da diversidade de algas no país nesse período de cinco anos, iniciando uma mudança quanto as diferenças geográficas regionais.
Abstract An updated synthesis of cyanobacteria and algae information is presented for Brazil aiming to refine the data gathered to date and evaluate the progress of the biodiversity knowledge about these organisms in the country since the publication of the Catálogo de Plantas e Fungos do Brasil. The results of 2015 showed an increase of 1,250 species (35.7%) when compared to 2010, reaching a total of 4,747 species. The most diverse classes in species number were the Bacillariophyceae, Conjugatophyceae, Florideophyceae, Cyanophyceae, Dinophyceae and Euglenophyceae. Bacillariophyceae and Cyanophyceae had the highest increase in species number in the five-year interval. The Southeast and South regions were the most diverse, however, the Northeast, with the states of Piauí and Sergipe, and the Central-west region, with Mato Grosso, Goiás and Distrito Federal, also stood out in the national algal biodiversity scenario. Despite the shortage of taxonomists and limited infrastructure, the results showed a significant improvement in the knowledge regarding the diversity of cyanobacteria and algae in the country during the study period, starting to even out regional geographical differences caused by subsampling.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

The efficiency of a recirculation system for the cultivation of C. auratus, fed with natural and artificial diets was evaluated. The experiment consisted of four treatments with four replicates. The first two treatments used recirculation system and the last two static system. In the first and fourth treatment, animals were fed with brine shrimp biomass (50% CP), while in the second and third treatments a commercial flake ration (42% CP) was used. At the end of cultivation (60 days), water samples were taken from all experimental units and subjected to standard plate count for determining the density of mesophilic bacteria. Final weight (g), final length (cm), specific growth rate (% day -1) and weight gain (%) were significantly different (p<0.05) among tested diets, but the culture systems did not affect fish performance. There was significant difference (p<0.05) between systems for total ammonia and nitrite. Animals fed with brine shrimp biomass acquired more intense and brighter colors than those fed only with commercial flake food. The values of standard plate count of mesophilic bacteria ranged from 7.0 × 10³ to 1.1 × 10(4) CFU.ml-1. In this study we observed that C. auratus is a species tolerant to adverse water conditions, perhaps with increased stock densities, the recirculation system could play some positive role on the zootechnical performance of cultured animals.
A eficiência de um sistema de recirculação para o cultivo de C. auratus, alimentados com dieta natural e artificial foi avaliada. O experimento consistiu de quatro tratamentos com quatro repetições. Os dois primeiros tratamentos utilizaram sistemas de recirculação e os dois últimos sistemas estáticos. No primeiro e quarto tratamento, os animais foram alimentados com biomassa de artêmia (PB 50%), enquanto nos tratamentos dois e três, com ração comercial em flocos (42% PB). Ao final do cultivo (60 dias), amostras de água foram coletadas em todas as unidades experimentais e submetidos a contagem padrão em placas para determinação da densidade de bactérias mesófilas. O peso final (g), comprimento final (cm), taxa de crescimento específico (% dia-1) e ganho de peso (%) foram significativamente diferentes (p<0,05) entre as dietas testadas, mas os sistemas de cultivo não afetaram o desempenho dos peixes. Houve diferença significativa (p<0,05) entre os sistemas para os níveis de amônia total e nitrito. Os animais alimentados com biomassa artêmia adquiriram cor e brilho mais intensos do que aqueles alimentados apenas com ração comercial em flocos. Os valores de contagem padrão em placas de bactérias mesófilas variaram de 7,0 × 10³ para 1,1 × 10(4) CFU.ml-1. Neste estudo, observou-se que C. auratus é uma espécie tolerante a condições adversas de água, talvez com maior densidade de estocagem, o sistema de recirculação poderia desempenhar papel positivo sobre o desempenho zootécnico dos animais cultivados.

Additives have been used to improve quality diet and animal development. The objective was to assess the effects of probiotic or phytase supplementation on apparent digestibility of nutrients in 24 Mangalarga Marchador foals (16 males, 8 females), aged 241.6 ± 23.2 days (mean ± SD/SEM), and weighing 216.7 ± 27.8 kg. The study lasted 28 days, the first 23 for diet and handling adaptation, and the last five days for fecal collection. The basal diet consisted of Cynodon pasture and pelleted concentrate (1.2 kg/100 kg body weight), as well as water and mineral salt ad libitum. There were four treatments: basal diet (T1); basal diet + 5 g/day of 10(9) CFU/g of s. cerevisiae (T2); basal diet + 2 g/day of 10(8)CFU/g of bacteria B. bifidum, E. faecium, L. acidophilus, L. plantarum and 10(5) CFU/g of s. cerevisiae (T3); and basal diet + 1250 FTU/day of phytase (T4). The DM intake was estimated with an external marker (LIPE©) associated with cecal in situ digestibility of DM. All foals were randomized by replicate using a 4 x 2 factorial, and results were compared using student's t-test (p<0.05). Group T3 showed better digestibility of crude protein, total minerals, calcium and phosphorus; whereas T2 had better digestibility of hemicellulose and calcium. Group T4 had better digestibility of neutral detergent fiber and intermediary values of minerals digestibility. Probiotics can be used as a nutritional strategy to improve the digestibility of a foal's diet, particularly when live bacteria and yeast culture are used. A phytase dose of 205 FTU/kg DM in the basal diet improved fiber digestibility, indicating its potential in equine nutrition.
El uso de aditivos se ha desarrollado mucho actualmente, con el fin de mejorar el aprovechamiento de la dieta. El presente trabajo se realizó con el objetivo de evaluar los efectos de la suplementación con probióticos o fitasa sobre la digestibilidad aparente (DA) de los nutrientes en 24 potros (16 machos y 8 hembras) Mangalarga Marchador con 241,6 ± 23,2 días de edad y 216,7 ± 27,8 kg de peso vivo (PV). El experimento tuvo duración de 28 días (23 para la adaptación y cinco para la recogida de muestras de heces) y la dieta basal se compuso de pasto de Cynodon y pienso comercial (1,2 kg /100 kg PV), además de agua y sal mineralizada ad libitum. Los tratamientos fueron: dieta basal (T1); dieta basal + 5 g/día de 10(9)UFC/g de levadura S. cerevisiae (T2); dieta basal + 2 g/día de 10(8) UFC/g de bacterias B. bifidum, E. faecium, L. acidophilus, L. plantarum y 10(5) UFC/ g de levadura S. cerevisiae (T3); y dieta basal + 1250 FTU (unidades de fitasa)/dia (T4). La estimación del consumo se realizó utilizando el indicador externo LIPE® y la técnica de digestibilidad in situ. El diseño estadístico fue enteramente al azar, en esquema factorial 4 x 2, y las medias fueron comparadas por el test t de student (p<0,05). El grupo T3 tuvo mejor DA de la proteína bruta, cenizas, calcio y fósforo. El grupo T2 tuvo mejor DA de la hemicelulosa y calcio. El grupo T4 tuvo mejor DA de la fibra neutrodetergente y fue intermedio en DA de los minerales. Los resultados indicaron que los probióticos pueden ser una estrategia para la mejora del aprovechamiento de la dieta de potros, sobre todo cuando están compuestos por bacterias y levaduras vivas. La fitasa mejoró el aprovechamiento de la fibra cuando se administró en dosis de 205 FTU/kg de la dieta total, demostrando su potencial para la utilización en la nutrición equina.

The vasorelaxant effects of SR 47063 (4-(2-cyanimino-1,2-dihydropyrid-1-yl)-2,2-dimethyl-6-nitrochromene), a new K+-channel opener structurally related to levcromakalim, were examined in isolated human saphenous vein (HSV) and rat aorta (RA). HSV or RA rings were precontracted with either KCl or noradrenaline and cumulative relaxant concentration-response curves were obtained for SR 47063 (0.1 nM to 1 µM) in the presence or absence of 3 µM glibenclamide. SR 47063 potently relaxed HSV and RA precontracted with 20 mM (but not 60 mM) KCl or 10 µM noradrenaline in a concentration-dependent manner, showing slightly greater activity in the aorta. The potency of the effect of SR 47063 on HSV and RA was 12- and 58-fold greater, respectively, than that reported for the structurally related K+-channel opener levcromakalim. The vasorelaxant action of SR 47063 in both blood vessels was strongly inhibited by 3 µM glibenclamide, consistent with a mechanism of action involving ATP-dependent K+-channels.
