OBJECTIVES: Retinoblastoma (RB) is a highly malignant eye tumor with a low survival rate and a high metastatic rate. The current work was designed to investigate the potential roles of microRNA-144 (miR-144) in the diagnosis and prognosis of RB. METHODS: miR-144 expression levels in RB tissues and adjacent normal tissues, as well as serum samples from RB patients and healthy controls were measured. The association between miR-144 expression levels and clinical features were analyzed. Moreover, diagnostic and prognostic values of miR-144 in RB were verified by receiver operating characteristic analysis and Kaplan-Meier survival assays. RESULTS: The expression level of miR-144 was markedly decreased in tumor tissues of RB patients, and the expression level of miR-144 was positively associated with tumor size and metastasis in RB patients. Moreover, miR-144 can distinguish tumor tissues from normal tissues with high specificity and sensitivity, and RB patients with lower miR-144 expression have shorter overall and disease-free survival rates than those with higher miR-144 expression. Alternatively, miR-144 also decreased in the serum of RB patients in comparison with healthy subjects, and miR-144 expression levels in the tissue samples and serum were positively correlated. Furthermore, miR-144 levels in the serum of RB patients sensitively distinguished RB patients from healthy controls. CONCLUSIONS: miR-144 expression was downregulated in serum and tissue samples of RB patients and may function as a diagnostic and prognostic marker for RB.

RESUMEN Las fracturas estructurales de yacimientos tienes excelentes características fractales y autosimilares. Basados en la teoría fractal, se estudiaron las características fractales superficiales de las fallas y las características fractales de las fracturas en el núcleo del Cinturón Falla-Pliegue de la Formación Xujiahe, área central de Yuanba (China). Luego se estableció una relación cuantitativa para estas relaciones. Según las características fractales de las fallas, se predijo la zona favorable de fractura, lo que ofrece una nueva idea para el conocimiento de esta y a la vez brinda referencias teóricas para la exploración de la zona de desarrollo de la fractura durante la exploración de petróleo y gas. Los resultados muestran lo siguiente: el valor sísmico de la dimensión fractual de la falla en la Formación Xujiahe es de 1.5284; el coeficiente de correlación R2 es mayor a 0.99901; la dimensión del coeficiente de correlación de regresión lineal en el núcleo de la fractura de la Formación Xujiahe es mayor a 0.98; la dimensión fractal D puede reflejar la falla y el grado de desarrollo de la fractura, al igual que la complejidad del sistema de fractura; también puede calcular cuantitativamente la densidad de la fractura del yacimiento en el área; las áreas con capacidad mayor a 1.45 son las zonas de desarrollo de la fractura en el Cinturón Falla-Pliegue del área central de Yuanba; el grado de enriquecimiento de petróleo y gas es alta; las áreas con valor de dimensión fractal entre 0.95 y 1.45 son las zonas de fractura de desarrollo relativo; la dimensión fractal con valores menos a 0.95 son las áreas que carecen de fracturas.

ABSTRACT The reservoir structural fractures have excellent fractal characteristics, as well as self-similarities. Based on the fractal theory, the surface fractal characteristics of faults and the fractal characteristic of fractures in the core of the Xujiahe Formation in the Fault-Fold Belt of the central Yuanba area were studied, and a quantitative relationship was set up between them. Based on the fractal characteristics of faults, predictions were made of the favorable fracture zone, which provides a new idea for the research of fracture, as well as offers theoretical references for exploring the fracture development zone during oil-gas exploration. The results show the following: the seismic value of reflection fault fractal dimension of the Xujiahe Formation is 1.5284; the correlation coefficient R2 is bigger than 0.9901; the capacity dimension linear regression correlation coefficient of the fracture in core of the Xujiahe Formation is bigger than 0.98; the fractal dimension D can well reflect the fault and fracture development degree, as well as the complexity of the fracture system; it can quantitatively calculate the density of the fracture of the reservoir in the area; the areas of capacity dimension bigger than 1.45 are the fracture development zones in the Fault-Fold Belt of the central Yuanba area; the oil and gas enrichment degree is high; the areas with the fractal dimension value between 0.95 and 1.45 are the fracture relatively-developed zones; the fractal dimension with values smaller than 0.95 are the lack of fracture areas.

Background: Finding molecular markers linked to quantitative trait loci is the first step in marker-assisted selection (MAS). Microsatellites are excellent molecular markers because of their large numbers, even distribution in the genome, and high polymorphism. In this study, the polymerisation effect of four microsatellites (OarAE101, BM1329, BM143, and LSCV043) on litter size was analysed using microsatellite markers and pedigrees. Results: The results indicate that the polymerisation effect of four microsatellite loci significantly affected the litter size. E5E10F2F6G1G5H6H11 and E3E8F5F7G1G5H3H9 had the highest and lowest litter sizes in the F2 generation, respectively. The polymerisation effect value (v) of the E5E10 genotype was 3.18% higher than that of the E2E7 genotype. The v of genotype F2F6 was 14.47% higher than that of the F5F7 genotype. The v of genotype G1G5 was 58.99% higher than that of the G2G7 genotype. The v of the H6H11 genotype was 5.60% to 49.74% higher than those of the H4H10 and H1H7 genotypes. The v of the H3H9 genotype was 17.22% higher than that of the H1H7 genotype. Conclusions: The results of the present study are vital to improving the reproductive performance in goat breeds MAS.

OBJECTIVES: This pilot study was designed to utilize stroke volume variation and cardiac index to ensure fluid optimization during one-lung ventilation in patients undergoing thoracoscopic lobectomies. METHODS: Eighty patients undergoing thoracoscopic lobectomy were randomized into either a goal-directed therapy group or a control group. In the goal-directed therapy group, the stroke volume variation was controlled at 10%±1%, and the cardiac index was controlled at a minimum of 2.5 L.min-1.m-2. In the control group, the MAP was maintained at between 65 mm Hg and 90 mm Hg, heart rate was maintained at between 60 BPM and 100 BPM, and urinary output was greater than 0.5 mL/kg-1/h-1. The hemodynamic variables, arterial blood gas analyses, total administered fluid volume and side effects were recorded. RESULTS: The PaO2/FiO2-ratio before the end of one-lung ventilation in the goal-directed therapy group was significantly higher than that of the control group, but there were no differences between the goal-directed therapy group and the control group for the PaO2/FiO2-ratio or other arterial blood gas analysis indices prior to anesthesia. The extubation time was significantly earlier in the goal-directed therapy group, but there was no difference in the length of hospital stay. Patients in the control group had greater urine volumes, and they were given greater colloid and overall fluid volumes. Nausea and vomiting were significantly reduced in the goal-directed therapy group. CONCLUSION: The results of this study demonstrated that an optimization protocol, based on stroke volume variation and cardiac index obtained with a FloTrac/Vigileo device, increased the PaO2/FiO2-ratio and reduced the overall fluid volume, intubation time and postoperative complications (nausea and vomiting) in thoracic surgery patients requiring one-lung ventilation.

Nitric oxide (NO), synthesized as needed by NO synthase (NOS), is involved in spinogenesis and synaptogenesis. Immature spine morphology is characteristic of fragile X syndrome (FXS). The objective of this research was to investigate and compare changes of postnatal neuronal NOS (nNOS) expression in the hippocampus of male fragile X mental retardation 1 gene knockout mice (FMR1 KO mice, the animal model of FXS) and male wild-type mice (WT) at postnatal day 7 (P7), P14, P21, and P28. nNOS mRNA levels were analyzed by real-time quantitative PCR (N = 4-7) and nNOS protein was estimated by Western blot (N = 3) and immunohistochemistry (N = 1). In the PCR assessment, primers 5’-GTGGCCATCGTGTCCTACCATAC-3’ and 5’-GTTTCGAGGCAGGTGGAAGCTA-3’ were used for the detection of nNOS and primers 5’-CCGTTTCTCCTGGCTCAGTTTA-3’ and 5’-CCCCAATACCACATCATCCAT-3’ were used for the detection of β-actin. Compared to the WT group, nNOS mRNA expression was significantly decreased in FMR1 KO mice at P21 (KO: 0.2857 ± 0.0150, WT: 0.5646 ± 0.0657; P < 0.05). Consistently, nNOS immunoreactivity also revealed reduced staining intensity at P21 in the FMR1 KO group. Western blot analysis validated the immunostaining results by demonstrating a significant reduction in nNOS protein levels in the FMR1 KO group compared to the WT group at P21 (KO: 0.3015 ± 0.0897, WT: 1.7542 ± 0.5455; P < 0.05). These results suggest that nNOS was involved in the postnatal development of the hippocampus in FXS and impaired NO production may retard spine maturation in FXS.
