Resumo: O envelhecimento da semente durante o armazenamento ou transporte pode afetar o rendimento e o custo da produção agrícola. O milho doce é um mutante natural com fraca vitalidade da semente durante o armazenamento. Neste estudo, mostramos que a dinâmica do nível de hormônio endógeno de sementes de milho doce recém-colhidas e envelhecidas foi diferente durante o processo de geminação precoce. A análise transcriptômica revelou vários novos transcritos durante o estágio inicial da germinação do milho doce nas vias de transdução de sinal e processamento de RNA. O perfil transcriptômico comparativo revelou que a expressão de muitos genes foi ativada mais cedo nas sementes envelhecidas do que nas recém-colhidas. As análises funcionais mostraram que, como duas redes principais, a sinalização de hormônios vegetais e as vias de defesa eram diferentes entre sementes de milho doce recém-colhidas e envelhecidas. As vias de sinalização de auxina, citocinina, giberelina, ácido salicílico e ácido jasmônico foram ativadas mais cedo nas sementes envelhecidas do que nas recém-colhidas no primeiro dia após a embebição. Abstract: Seed ageing during storage or shipping can affect the yield and cost of agricultural production. Sweet corn is a naturally mutated corn cultivar with weak seed vitality during storage. In this study, we have shown that endogenous hormone level dynamics of fresh and aged sweet corn seeds were different during early geminating process. Transcriptomic assembling uncovered several novel transcripts during early stage of sweet corn germination in signal transduction and RNA processing pathways. Comparative transcriptomic profiling revealed that expression of many genes was activated earlier in aged seeds than fresh ones. Functional analyses showed that, as two major networks, plant hormone signaling and defense pathways were different between fresh and aged sweet corn seeds. Auxin, cytokinin, gibberellin, salicylic acid and jasmonic acid signaling pathways were activated earlier in aged seeds than fresh ones on the first day after imbibition. These results provided knowledge for understanding the ageing process of sweet corn seeds and several candidate genes for genetic study of seed ageing mechanism. To solve the technical, economic and environmental problems in the remanufacturing process of decommissioned machine tools, this study proposes a module partition method for remanufacturing retired machine tools. Through an analysis of the economic, accuracy, environment-friendly, and functional and physical interactivity principles, this study applies the fuzzy cluster method (FCM) to analyse the modularity of the components to be assembled. It establishes the correlation matrix between the parts, from which a module division scheme is obtained. Finally, a case study is presented to verify the feasibility of the module division method.

Abstract Swertia L. is a large genus in Swertiinae (Gentianaceae). In China, many Swertia species are used as traditional Tibetan medicines, known as “Zangyinchen” or “Dida”. However, the phylogenetic relationships among Swertia medicinal plants and their wild relatives have remained unclear. In this study, we sequenced and assembled 16 complete chloroplast (cp) genomes of 10 Swertia species, mainly distributed in Qinghai Province, China. The results showed that these species have typical structures and characteristics of plant cp genomes. The sizes of Swertia cp genomes are ranging from 149,488 bp to 154,097 bp. Most Swertia cp genomes presented 134 genes, including 85 protein coding genes, eight rRNA genes, 37 tRNA genes, and four pseudogenes. Furthermore, the GC contents and boundaries of cp genomes are similar among Swertia species. The phylogenetic analyses indicated that Swertia is a complex polyphyletic group. In addition, positive selection was found in psaI and petL genes, indicating the possible adaptation of Qinghai Swertia species to the light environment of the Qinghai-Tibet plateau. These new cp genome data could be further investigated to develop DNA barcodes for Swertia medicinal plants and for additional systematic studies of Swertia and Swertiinae species.

Abstract To study the purification conditions of polysaccharide from Solanum nigrum L. by S-8 macroporous adsorption resin. The effect of multi-factors on the purification of polysaccharide from Solanum nigrum were studied by static - dynamic adsorption and desorption methods with indexes of adsorption and desorption rates. The optimized purification process condition was as follows: the concentration of sample was 5 mg/mL, pH level was 7.0, temperature was 30 °C, the adsorption velocity was 0.5 mL/min, the maximun loading amount was 10 BV, 0.5 mol/L NaCl solution 5BV was used as eluent, elution flow rate was 4 mL/min. With this condition, the purity of Solanum nigrum polysaccharide was 91.00%, which was 1.989 times higher than before. S-8 macroporous resin can be used to separate and purify the solanum nigrum polysaccharide and improve the purity and quality of polysaccharides.

Abstract In this study, the effects of the starter culture Lactobacillus sakei on pH, the total lactic acid concentration, nitrite depletion and histamine reduction of chili sauce were investigated. The nitrite concentration in the CPN sample was significantly lower (p < 0.05) than that of the CPS sample (as control), and decreased by 30.87%.The putrescine and histamine concentration in starter culture inoculated chili sauce were significantly lower (p < 0.05) than that of the control sample, and were decreased by 38.08% and 49.88%, respectively. These results revealed that L. sakei could be used as a potential starter culture for nitrite depletion and biogenic amines reduction of chili sauce with good sensory attributes.

Abstract In this study, the effects of the starter culture Lactobacillus sakei on microbial quality and the bacterial community diversity of chili sauce were investigated. In starter culture inoculated chili sauce (CPN sample), Lactobacillus spp. was predominant bacteria with 86.08% of the relative abundance on the 30th day of fermentation. Meanwhile, the spoilage organisms and pathogenic bacteria, including Staphylococcus spp., Enterobacter spp., Pseudomonas spp. and Neorhizobium spp. were nearly undetectable in starter culture inoculated chili sauce. In contrast, Lactobacillus spp. and Weissella spp. were predominant bacteria, and trace Pseudomonas spp. and Neorhizobium spp. were still detectable at the end of fermentation in the spontaneous fermentation (CPS sample). These results revealed that L. sakei could be used as a potential starter culture for the microbial quality improvement.

Resumo As plantas ajustam o crescimento da parte aérea para se adaptarem a fatores ambientais variáveis, como o aumento da radiação ultravioleta B (UVB). No entanto, as pessoas ignoram que as raízes das plantas também podem responder à luz UVB. Neste estudo, verificamos a morfologia das raízes enroladas de trigo sob radiação UVB, o que chamamos de “raízes dobradas”. A região encaracolada é a zona de transição da raiz no nível celular. Depois de exposição à radiação UVB aprimorada por 2 dias (10,08 KJ/m2/d), o tamanho das células diminuiu, e os filamentos de actina se reuniram. Também constatamos que a produção de H2O2 aumentou e que o conteúdo do ácido indol-3-acético (IAA) aumentou notavelmente. O experimento farmacológico revelou que os filamentos de actina se reuniram e polimerizaram em feixes nas células da raiz de trigo após irrigação com H2O2 e IAA. Esses resultados indicam que os filamentos de actina alteraram sua distribuição e formaram a “raiz dobrada”, relacionada à produção de H2O2 e ao aumento do IAA. No geral, os filamentos de actina nas células da raiz de trigo podem ser um alvo subcelular da radiação UVB, e sua interrupção determina a morfologia da raiz.

Abstract Plants adjust their shoot growth to acclimate to changing environmental factors, such as to enhanced Ultraviolet-B (UV-B) radiation. However, people have ignored that plant roots can also respond to UV-B light. Here, we find the morphology curled wheat roots under UV-B radiation, that we call, “bending roots.” The curly region is the transition zone of the root after observed at the cellular level. After exposed to enhanced UV-B radiation for 2 d (10.08 KJ/m2/d), cell size decreased and actin filaments gathered in wheat roots. We also find that H2O2 production increased and that content of the indole-3-acetic acid (IAA) increased remarkably. The pharmacological experiment revealed that actin filaments gathered and polymerized into bundles in the wheat root cells after irrigated H2O2 and IAA. These results indicated that actin filaments changed their distribution and formed the “bending root,” which was related to H2O2 production and increase in IAA. Overall, actin filaments in wheat root cells could be a subcellular target of UV-B radiation, and its disruption determines root morphology.

Abstract Cytoplasmic male sterility (CMS) is a maternally inherited trait that derives from the inability to produce functional pollen in higher plants. CMS results from recombination of the mitochondrial genome. However, understanding of the molecular mechanism of CMS in pepper is limited. In this study, comparative transcriptomic analyses were performed using a near-isogenic CMS line 14A (CMS-14A) and a maintainer line 14B (ML-14B) as experimental materials. A total of 17,349 differentially expressed genes were detected between CMS-14A and ML-14B at the PMC meiosis stage. Among them, six unigenes associated with CMS and 108 unigenes involved in energy metabolism were identified. The gene orf165 was found in CMS-14A. When orf165 was introduced into ML-14B, almost 30% of transgenic plants were CMS. In addition, orf165 expression in transgenic CMS plants resulted in abnormal function of some genes involved in energy metabolism. When orf165 in transgenic CMS plant was silenced, the resulted orf165-silenced plant was male fertile and the expression patterns of some genes associated with energy metabolism were similar to ML-14B. Thus, we confirmed that orf165 influenced CMS in pepper.

ABSTRACT Sugarcane (Saccharum L. spp. hybrids) is a globally important crop. While roots contribute to its net primary production, knowledge of sugarcane root growth is incomplete and limited in scope. The objective of this study was to determine cumulative root density (LA), root distribution, effective rooting depth (ERD), and root growth rate for four sugarcane genotypes across three crop cycles (plant cane [PC], first ratoon [FR], and second ratoon [SR]). The experiment was conducted in a rain shelter on four sugarcane genotypes (ROC22, YZ04-241, YZ05-194, and YZ05-51) using minirhizotron tubes to a soil profile depth of 1.08 m. Most roots in were found at 0–0.36 m-depth, and about 85.0% were within a 0–0.72 m depth in PC. In FR and SR, however, > 81.8% of roots were found at 0–0.54 m-depth regardless of genotype. The LA of ROC22 was often significantly higher than that of the other genotypes at depths < 0.54 m regardless of crop cycle. The genotype ROC22 had greater root length and faster root growth rate than YZ05-194 at 0.18–0.36 m in PC and FR, while SR YZ05-194 had slower root growth than the other genotypes at 0–0.18m. Ratooning ability of sugarcane genotypes may be related to these root growth attributes.

Abstract The measurement of gene expression can provide important information about gene function and the molecular basis for developmental processes. We analyzed the transcriptomes at three different developmental stages of pepper flower [sporogenous cell division, stage (B1); pollen mother cell meiosis, stage (B2); and open flower (B3)]. In the cDNA libraries for B1, B2, and B3: 82718, 77061, and 91491 unigenes were assembled, respectively. A total of 34,445 unigene sequences and 128 pathways were annotated by KEGG pathway analysis. Several genes associated with nectar biosynthesis and nectary development were identified, and 8,955, 12,182, and 23,667 DEGs were identified in the B2 vs B1, B3 vs B1, and B3 vs. B2 comparisons. DEGs were involved in various metabolic processes, including flower development, nectar biosynthesis, and nectary development. According to the RNA-seq data, all 13 selected DEGs showed similar expression patterns after q-PCR analysis. Sucrose-phosphatase, galactinol-sucrose galactosyltransferase, and sucrose synthase played very important roles in nectar biosynthesis, and CRABS CLAW could potentially be involved in mediating nectary development. A significant number of simple sequence repeat and single nucleotide polymorphism markers were predicted in the Capsicum annuum sequences. The new results provide valuable genetic information about flower development in pepper.

Abstract Background: ADP-glucose pyrophosphorylase (AGPase), the key enzyme in plant starch biosynthesis, is a heterotetramer composed of two identical large subunits and two identical small subunits. AGPase has plastidial and cytosolic isoforms in higher plants, whereas it is mainly detected in the cytosol of grain endosperms in cereal crops. Our previous results have shown that the expression of the TaAGPL1 gene, encoding the cytosolic large subunit of wheat AGPase, temporally coincides with the rate of starch accumulation and that its overexpression dramatically increases wheat AGPase activity and the rate of starch accumulation, suggesting an important role. Methods: In this study, we performed yeast one-hybrid screening using the promoter of the TaAGPL1 gene as bait and a wheat grain cDNA library as prey to screen out the upstream regulators of TaAGPL1 gene. And the barley stripe mosaic virus-induced gene-silencing (BSMV-VIGS) method was used to verify the functional characterization of the identified regulators in starch biosynthesis. Results: Disulfide isomerase 1-2 protein (TaPDIL1-2) was screened out, and its binding to the TaAGPL1-1D promoter was further verified using another yeast one-hybrid screen. Transiently silenced wheat plants of the TaPDIL1-2 gene were obtained by using BSMV-VIGS method under field conditions. In grains of BSMV-VIGS-TaPDIL1-2-silenced wheat plants, the TaAGPL1 gene transcription levels, grain starch contents, and 1000-kernel weight also significantly increased. Conclusions: As important chaperones involved in oxidative protein folding, PDIL proteins have been reported to form hetero-dimers with some transcription factors, and thus, our results suggested that TaPDIL1-2 protein could indirectly and negatively regulate the expression of the TaAGPL1 gene and function in starch biosynthesis.

Resumo Kiwis são uma fruta popular em todo o mundo; No entanto, o crescimento das plantas é ameaçado por estresses abióticos como a seca e as altas temperaturas. O tratamento com niacina em plantas mostrou aumentar os níveis de NADPH, aumentando assim a tolerância a stress abiótico. Aqui, avaliamos o efeito do tratamento com spray de solução de niacina sobre os níveis de NADPH nos cultivares de kiwis Hayward e Xuxiang. Descobrimos que o tratamento por spray com solução de niacina promoveu níveis de NADPH e NADP + e diminuiu a produção de O2·- e os teores de H2O2 nas folhas durante um curto período. Nos frutos, os teores de NADPH aumentaram durante o desenvolvimento precoce, mas diminuíram mais tarde. No entanto, não se observou qualquer efeito nos níveis de NADP + ao longo do desenvolvimento do fruto. Em resumo, este relatório sugere que a niacina pode ser utilizada para aumentar NADPH oxidases, aumentando assim a tolerância ao estresse em kiwis durante o encontro de condições estressantes de curto prazo.

Abstract Kiwifruit are a popular fruit worldwide; however, plant growth is threatened by abiotic stresses such as drought and high temperatures. Niacin treatment in plants has been shown to increase NADPH levels, thus enhancing abiotic stresses tolerance. Here, we evaluate the effect of niacin solution spray treatment on NADPH levels in the kiwifruit cultivars Hayward and Xuxiang. We found that spray treatment with niacin solution promoted NADPH and NADP+ levels and decreased both O2·- production and H2O2 contents in leaves during a short period. In fruit, NADPH contents increased during early development, but decreased later. However, no effect on NADP+ levels has been observed throughout fruit development. In summary, this report suggests that niacin may be used to increase NADPH oxidases, thus increasing stress-tolerance in kiwifruit during encounter of short-term stressful conditions.

ABSTRACT Bemisia tabaci (Gennadius), the vector of Tomato chlorosis virus (ToCV), is one of the major pests of tomato (Solanum lycopersicum L. var. lycopersicum), potentially causing up to 100% yield loss. The purpose of this research was to effectively reduce intrusion by B. tabaci and control damage from ToCV in protected cultivations. The treatments included the use of a regular solar greenhouse as a control (CK); greenhouses I and II with 60- and 80-mesh insect-proof nets, respectively, that were installed in both houses at the front and upper ventilations; and greenhouse III with the addition of not only the 80-mesh insect-proof net as in greenhouse II but also a ventilating pipeline to the back wall. The effects of mesh size and back wall ventilation on the greenhouse temperature and humidity, number of B. tabaci, and level of ToCV infection were studied. Under all conditions tested (from 2014-2016), the temperature of greenhouse III with installed netting was reduced by drilling holes through the back wall and nonsignificant difference existed in the average relative humidity between greenhouses II and III. Importantly, the number of B. tabaci and ToCV infection rate were effectively controlled in greenhouse III, ranging from 0.03 to 0.33 adults per sampled plant and from 0% to 6.67% of virus incidence. Thus, installing 80-mesh insect-proof netting at the front and upper ventilation areas as well as adding a ventilating pipeline to the back wall could effectively reduce the number of B. tabaci and prevent ToCV damage.

Two new Mg(II)-based and Zn(II)-based coordination polymers, {[Mg3(BTB)(DMA)4](DMA)2}n (1, H3BTB=1,3,5-benzenetrisbenzoic acid, DMA=N,N-dimethylacetamide) and {(H2NMe2)2[Zn3(BTB)2(OH)(Im)](DMF)9(MeOH)7}n (2, Im=imidazole, DMF=N,N-dimethylformamide), have been successfully synthesized and structurally characterized under solvothermal conditions. 1 contains a linear [Mg3(COO)6] cluster that connected by the fully deprotonated BTB3- ligands to give a kgd-type 2D bilayer structure; 2 represents a microporous 3D pillar-layered system based on the binuclear Zn units and pillared Im ligands, which shows a (3,5)-connected hms topological net. In addition, in vitro anticancer activities of compounds 1 and 2 on 4 human liver cancer cells (HB611, HHCC, BEL-7405 and SMMC-7721) were determined.

Glutathione synthetase deficiency (GSSD) is a rare inborn error of glutathione metabolism with autosomal recessive inheritance. The severe form of the disease is characterized by acute metabolic acidosis, usually present in the neonatal period with hemolytic anemia and progressive encephalopathy. A case of a male newborn infant who had severe metabolic acidosis with high anion gap, hemolytic anemia, and hyperbilirubinemia is reported. A high level of 5-oxoproline was detected in his urine and a diagnosis of generalized GSSD was made. DNA sequence analysis revealed the infant to be compound heterozygous with two mutations, c.738dupG in exon 8 of GSS gene resulting in p.S247fs and a repetitive sequence in exon 3 of GSS gene. Treatment after diagnosis of GSSD included supplementation with antioxidants and oral sodium hydrogen bicarbonate. However, he maintained a variable degree of metabolic acidosis and succumbed shortly after his parents requested discontinuation of therapy because of dismal prognosis and medical futility when he was 18 days old.
