Inadequate invasion and excessive apoptosis of trophoblast cells are associated with the development of preeclampsia. Vitamin D deficiency in pregnant women may lead to an increased risk of preeclampsia. However, the underlying mechanisms by which vitamin D is effective in preventing preeclampsia are not fully understood. The objectives of this study were to investigate the role of lysosome-associated membrane glycoprotein 3 (LAMP3) in the pathogenesis of preeclampsia and to evaluate whether vitamin D supplementation would protect against the development of preeclampsia by regulating LAMP3 expression. Firstly, the mRNA and protein levels of LAMP3 were significantly upregulated in the placentas of preeclampsia patients compared to normal placentas, especially in trophoblast cells (a key component of the human placenta). In the hypoxia/reoxygenation (H/R)-exposed HTR-8/Svneo trophoblast cells, LAMP3 expression was also upregulated. H/R exposure repressed cell viability and invasion and increased apoptosis of trophoblast cells. siRNA-mediated knockdown of LAMP3 increased cell viability and invasion and suppressed apoptosis of H/R-exposed trophoblast cells. We further found that 1,25(OH)2D3 (the hormonally active form of vitamin D) treatment reduced LAMP3 expression in H/R exposed trophoblast cells. In addition, 1,25(OH)2D3 treatment promoted cell viability and invasion and inhibited apoptosis of H/R-exposed trophoblast cells. Notably, overexpression of LAMP3 abrogated the protective effect of 1,25(OH)2D3 on H/R-exposed trophoblast cells. Collectively, we demonstrated trophoblast cytoprotection by vitamin D, a process mediated via LAMP3.

ABSTRACT Soil deterioration, yield decline and soil microbial activity reduction caused by banana (Musa × paradisiaca L.) monoculture is threatening the sustainability of banana production in China. Therefore, it is necessary to study the benefits of rotation on soil quality. This study aimed to assess the effects of rotation and tillage on soil properties in a banana plantation for 25 yr. Treatments consisted of three rotation methods (banana-pineapple, BA; banana-cowpea, BP; banana-rice, BR) and banana monoculture (CK) combined with two tillage intensities (no-tillage, NT; conventional tillage, CT). Soil samples were taken at depth of 0-40 cm in 2019-2020. In comparison with CK, BA and BR increased soil moisture, pH, total organic C and available P, but decreased soil bulk density. Microbial biomass C and N at booting stage were 46.1% and 39.2% higher in BA and BR than those in CK. Urease, dehydrogenase and β-glucosidase obtained a mean of 34.1% increase in BP and 23.8% increase in BR compared with BA. Higher total N, NO3-N, available K and macro aggregate were showed in NT compared with CT, whereas porosity was 24.8% lower in NT than in CT. CO2, N2O and CH4 emissions were in average around between one third and two fifth lower in no-tillage compared with conventional tillage. In general, rotations combined with no-tillage led to a positive effect on soil quality, as evidenced by increase of soil moisture, total N, microbial biomass C and urease and accompanying increase in banana yield. In order to sustain higher productivity, application of rotation and no-tillage is of considerable importance.

OBJECTIVES: Cerebral ischemia seriously threatens human health and is characterized by high rates of incidence, disability and death. Developing an ideal animal model of cerebral ischemia that reflects the human clinical features is critical for pathological studies and clinical research. The goal of this study is to establish a local cerebral ischemia model in rhesus macaque, thereby providing an optimal animal model to study cerebral ischemia. METHODS: Eight healthy rhesus monkeys were selected for this study. CT scans were performed before the operation to exclude cerebral vascular and intracranial lesions. Under guidance and monitoring with digital subtraction angiography (DSA), a microcatheter was inserted into the M1 segment of the middle cerebral artery (MCA) via the femoral artery. Then, autologous white thrombi were introduced to block blood flow. Immediately following embolization, multisequence MRI was used to monitor cerebrovascular and brain parenchymal conditions. Twenty-four hours after embolization, 2 monkeys were sacrificed and subjected to perfusion, fixation and pathological examination. RESULTS: The cerebral ischemia model was established in 7 rhesus monkeys; one animal died during intubation. DSA and magnetic resonance angiography (MRA) indicated the presence of an arterial occlusion. MRI showed acute local cerebral ischemia. HE staining revealed infarct lesions formed in the brain tissues, and thrombi were present in the cerebral artery. CONCLUSION: We established a rhesus macaque model of local cerebral ischemia by autologous thrombus placement. This model has important implications for basic and clinical research on cerebral ischemia. MRI and DSA can evaluate the models to ensure accuracy and effectiveness.

Weaning results in intestinal dysfunction, mucosal atrophy, transient anorexia, and intestinal barrier defects. In this study, the effect of prodigiosin (PG) on the intestinal inflammation of weaned rats was investigated by using 1H-NMR spectroscopy and biochemistry indexes to regulate the intestinal metabolism. After administration for 14 days, the body mass of the PG group was increased by 1.29- and 1.26-fold compared with those of the control and alcohol groups, respectively, using a dose of 200 µg PG·kg-1 body weight per day. PG increased organic acid content and decreased moisture, pH values, and free ammonia in feces. In addition, PG alleviated the intestinal inflammation of weaned rats. The analysis of 1H-NMR signal peak attribution and the model validation of metabolic data of feces contents showed that PG significantly affected the metabolism of small molecular compounds in the intestinal tract of weaned rats. This study presents the promising alternative of using PG to alleviate intestinal inflammation effectively in the intestinal tract of weaned rats.

Non-alcoholic fatty liver disease (NAFLD) is a common disease associated with metabolic syndrome and can lead to life-threatening complications like hepatic carcinoma and cirrhosis. Exenatide, a glucagon-like peptide-1 (GLP-1) receptor agonist antidiabetic drug, has the capacity to overcome insulin resistance and attenuate hepatic steatosis but the specific underlying mechanism is unclear. This study was designed to investigate the underlying molecular mechanisms of exenatide therapy on NAFLD. We used in vivo and in vitro techniques to investigate the protective effects of exenatide on fatty liver via fat mass and obesity associated gene (FTO) in a high-fat (HF) diet-induced NAFLD animal model and related cell culture model. Exenatide significantly decreased body weight, serum glucose, insulin, insulin resistance, serum free fatty acid, triglyceride, total cholesterol, low-density lipoprotein, aspartate aminotransferase, and alanine aminotransferase levels in HF-induced obese rabbits. Histological analysis showed that exenatide significantly reversed HF-induced lipid accumulation and inflammatory changes accompanied by decreased FTO mRNA and protein expression, which were abrogated by PI3K inhibitor LY294002. This study indicated that pharmacological interventions with GLP-1 may represent a promising therapeutic strategy for NAFLD.

A series of unsaturated esters were prepared via condensation of sodium carboxylates and alkenyl halide under the condition of macroporous polystyrene grafted quarternary ammonium salt as recyclable phase transfer catalyst, NaI as co-catalyst, Cu powder as inhibitor and H2O as solvent. Under optimal conditions, products yields are 78.2~ 96.0%. The catalyst can be convenient recycled and reutilized for about five times without losing its activity obviously.

OBJECTIVE: To estimate influenza-associated mortality in urban China. METHODS: Influenza-associated excess mortality for the period 2003-2008 was estimated in three cities in temperate northern China and five cities in the subtropical south of the country. The estimates were derived from models based on negative binomial regressions, vital statistics and the results of weekly influenza virus surveillance. FINDINGS: Annual influenza-associated excess mortality, for all causes, was 18.0 (range: 10.9-32.7) deaths per 100000 population in the northern cities and 11.3 (range: 7.3-17.8) deaths per 100000 in the southern cities. Excess mortality for respiratory and circulatory disease was 12.4 (range: 7.4-22.2) and 8.8 (range: 5.5-13.6) deaths per 100000 people in the northern and southern cities, respectively. Most (86%) deaths occurred among people aged >65 years. Influenza-associated excess mortality was higher in B-virus-dominant seasons than in seasons when A(H3N2) or A(H1N1) predominated, and more than half of all influenza-associated mortality was associated with influenza B virus. CONCLUSION: Between 2003 and 2008, seasonal influenza, particularly that caused by the influenza B virus, was associated with substantial mortality in three cities in the temperate north of China and five cities in the subtropical south of the country.

MÉTODOS: Se calculó el exceso de mortalidad asociado a la gripe durante el periodo comprendido entre 2003 y 2008 en tres ciudades del norte de China con clima templado y en cinco ciudades del sur del país con clima subtropical. Los cálculos se obtuvieron de modelos basados en regresiones binomiales negativas, estadísticas vitales y de los resultados de la vigilancia semanal del virus de la gripe. RESULTADOS: El exceso de mortalidad anual asociado a la gripe, por todas las causas, fue de 18,0 (rango: 10,9-32,7) muertes por cada 100000 habitantes en las ciudades del norte y de 11,3 (rango: 7,3-17,8) muertes por cada 100000 habitantes en las ciudades del sur. La mayor parte de este exceso de mortalidad - 12,4 (rango: 7,4-22,2) y 8,8 (rango: 5,5-13,6) muertes por cada 100000 habitantes en las ciudades del norte y del sur, respectivamente - se atribuyeron a una enfermedad respiratoria y/o circulatoria. La mayoría de las muertes (el 86%) ocurrió en personas con una edad >65 años. El exceso de mortalidad asociado a la gripe fue superior en épocas con un virus B dominante que en épocas en las que predominaron los virus A(H3N2) o A(H1N1). Más de la mitad de la mortalidad total asociada a la gripe se asoció al virus B de la gripe. CONCLUSIÓN: Entre 2003 y 2008, la gripe estacional, particularmente la causada por el virus B de la gripe, estuvo asociada a la mortalidad sustancial en tres ciudades de China con clima templado y en cinco ciudades del sur del país con clima subtropical.

RÉSULTATS: La mortalité annuelle excessive associée à la grippe, dans tous les cas, a été de 18 (plage: 10,9-32,7) décès pour une population de 100 000 personnes dans les villes du nord et de 11,3 (plage: 7,3-17,8) décès pour une population de 100 000 personnes dans les villes du sud. La plus grande partie de cette mortalité excessive - respectivement 12,4 (plage: 7,4-22,2) et 8,8 (plage: 5,5-13,6) décès pour une population de 100 000 personnes dans les villes du nord et du sud - a été attribuée à des maladies respiratoires et/ou circulatoires. La plupart des décès (86%) sont survenus chez des personnes de >65 ans. La mortalité excessive associée à la grippe a été plus élevée lors des saisons où prédominait le virus B plutôt que lors de celles où prédominaient les virus A(H3N2) ou A(H1N1) et plus de la moitié de l'ensemble de la mortalité associée à la grippe a été associée au virus B de la grippe. CONCLUSION: De 2003 à 2008, la grippe saisonnière, surtout celle provoquée par le virus B, a été associée à une mortalité substantielle dans trois villes du nord tempéré de Chine et dans cinq villes du sud subtropical du pays.
