Gelsolin (GSN) can sever actin filaments associated with autophagy. This study investigated how GSN-regulated actin filaments control autophagy in pancreatic ductal epithelial cells (PDECs) in acute pancreatitis (AP). AP was produced in a rat model and PDECs using caerulein (CAE). Rat pancreatic duct tissue and HPDE6-C7 cells were extracted at 6, 12, 24, and 48 h after CAE treatment. HPDE6-C7 cells in the presence of CAE were treated with cytochalasin B (CB) or silenced for GSN for 24 h. Pancreatic histopathology and serum amylase levels were analyzed. Cellular ultrastructure and autophagy in PDECs were observed by transmission electron microscopy after 24 h of CAE treatment. The expression of GSN and autophagy markers LC3, P62, and LAMP2 was evaluated in PDECs by immunohistochemistry and western blotting. Actin filaments were observed microscopically. Amylase levels were highest at 6 h of AP, and pancreatic tissue damage increased over time. Mitochondrial vacuolization and autophagy were observed in PDECs. CAE increased GSN expression in these cells over time, increased the LC3-II/LC3-I ratio and LAMP2 expression at 24 and 6 h of treatment, respectively, and decreased P62 expression at all time points. CB treatment for 24 h decreased the LC3-II/LC3-I ratio and LAMP2 expression, increased P62 levels, but had no impact on GSN expression in CAE-treated PDECs. CAE induced actin depolymerization, and CB potentiated this effect. GSN silencing increased the LC3-II/LC3-I ratio and LAMP2 expression and reduced actin depolymerization in CAE-treated PDECs. GSN may inhibit autophagosome biogenesis and autophagosome-lysosome fusion by increasing actin depolymerization in PDECs in AP.

SUMMARY OBJECTIVE: The aims of this study were to observe the regularity of blood glucose changes in hemodialysis patients with diabetes, time of onset of hypoglycemia and blood glucose level during dialysis, and to explore the sensitive early warning indicators of hypoglycemia in dialysis patients. BACKGROUND: Diabetes patients have a high incidence of hypoglycemia during hemodialysis. METHODS: A total of 124 maintenance hemodialysis patients with diabetes were selected for this study. Before dialysis, one, two, and three h after dialysis, and when hypoglycemia symptoms occurred, the blood glucose changes were monitored, the blood glucose drop range was observed when hypoglycemia symptoms occurred, and the correlation between the two was analyzed. RESULTS: After the start of the dialysis, the patient’s blood glucose showed a downward trend. The symptoms of hypoglycemia were most obvious within one-two hours, with an incidence rate of 57.9%. When the blood glucose drop percentage reached 37.7%, the specificity and sensitivity of early warning hypoglycemia symptoms were 84.6 and 73%, respectively. CONCLUSIONS: For hemodialysis patients with diabetes, attention should be paid to the symptoms of hypoglycemia during dialysis, and blood glucose should be monitored before dialysis and after 1-2 h of dialysis. If the blood glucose drop percentage is greater than 37.7%, the timely measures should be taken.

ABSTRACT Background: The spread of forest fire is a complex natural phenomenon. The cellular automata(CA) is a common model of forest fire spread, which fails to combine the unique combustion properties of forest fire spreading, resulting in inaccurate simulation results. In order to improve the accuracy of forest fire spread simulation, the Rothermel forest fire rate formula is simplified and combined with CA to form a multi-dimensional cellular automata(MD-CA) model of forest fire spread with different combustion properties in each cell. The formulas of the spread rate of forest fire in eight directions are obtained through the training datasets, and the testing datasets are used to compare the simulation results of CA model, MD-CA model and the actual fire spread. Results: It is concluded that the simulated areas and perimeters of the MD-CA are closer to the actual forest fire spread, the area error rate is 9.42% - 15.63%, and the perimeter error is 4.21% - 8.99%, The errors are less than CA model. Conclusion: The MD-CA model based on Rothermel has the strong ability to simulate the spread of fire in the laboratory, however, how to eliminate the errors over time is the task of the next stage.

RESUMO: Fungos micorrízicos arbusculares (FMA) tem atraído atenções de cientistas devido ao seu papel fundamental no crescimento e na restauração da vegetação de ecossistemas frágeis. No entanto, o conhecimento da composição e da diversidade da FMA em habitats cársticos e que retornem da agricultura para a silvicultura é limitado. Para entender: (1) a composição da Comunidade FMA e diversidade em fase diferente de retornar cultivo para a floresta; e (2) os efeitos da mudança do ambiente do solo sobre a diversidade das comunidades FMA, mostras de solo e de raiz de amoreira (Morus sp.) foram coletadas em Bijie e Libo, China, após um e dez anos de retorno ao cultivo dessa espécie, respectivamente, em sequência ao cultivo agrícola. Utilizando análise molecular e sequenciamento genético, constatou-se oito gêneros conhecidos e a distincão de oitenta e três espécies de FMA. Os gêneros Glomus, Paraglomus, Archaeospora e Diversispora foram dominantes, em solo e raízes. Em comparação as amostras de Libo, os gêneros Glomus, Paraglomus, Acaulospora e Claroideoglomus foram relativamente abundante nas raízes coletadas em Bijie, indicando que o retorno de uma área agrícola para o cultivo de amoreira é benéfico para a diversidade dos FMA, o que é atribuído as propriedades físico-químicas do solo. Essa conclusão é de grande importância para avaliar os efeitos do tipo de uso agrícola na microbiologia do solo.

ABSTRACT: Arbuscular mycorrhizal fungi (AMF) have been attracted more scientific attentions due to its critical role in enhancement of drought tolerance of plants for growth and vegetation restoration in karst fragile ecosystem. However, scientists know little about the AMF composition and diversity occurring in root systems of mulberry (Morus sp.), and in karst habitats which return land use from mulberry forestry, as well as the effects of soil environment change on the diversity of the AMF communities. To understand: (1) the AMF community composition and diversity at different stage of returning cropland to forest; and (2) the effects of soil environment change on the diversity of the AMF communities, soil and mulberry root samples were collected from Bijie and Libo sites, China, which experienced one and ten years, respectively, after returning croplands to forest. With the high throughput 454-sequencing technology, 8 known genera including 83 virtual species were distinguished and the genera Glomus, Paraglomus, Archaeospora and Diversispora were found to be dominant in soil and root sample. Compared to the samples in Libo, the genera Glomus, Paraglomus, Acaulospora and Claroideoglomus in root samples at Bijie site had a relatively abundance of species indicating that the returning cropland to forest is benefit to the AMF diversity and abundance, which was attribute to the variation of soil physiochemical properties. This conclusion is of great significance for guiding the return of farmland to forests.
