Resumen Antecedentes: La enfermedad de injerto contra huésped (EICH) es una complicación importante después del trasplante alogénico de células madre hematopoyéticas. Objetivos: Para dilucidar el papel de TLR4, el principal receptor de LPS bacteriano, en el desarrollo de GVHD, construimos un modelo de GVHD en ratones knockout para TLR4 (TLR4-/-) y monitoreamos el quimerismo celular. Métodos: En este estudio, usamos PCR para identificar si se establecieron ratones knockout para TLR4 (TLR4-/-). Antes del trasplante, pretratamos a los ratones con irradiación para obtener la dosis de irradiación adecuada. Se aplicó citometría de flujo para medir el estado de quimerismo, las distribuciones de APC y células T en ratones receptores TLR4+/+ y TLR4-/-. Resultados: El estado general de los receptores de TLR4-/- fue mejor que el de los receptores de TLR4+/+, y los ratones receptores de TLR4-/- mostraron manifestaciones de GVHD menos graves que los ratones receptores de TLR4+/+. La mayoría de las APC y las células T en el bazo del ratón huésped se derivaron de las células del donante, y las células T CD4+, incluidas las células T de memoria, se encontraban en su mayoría en los ratones huéspedes. Conclusión: En general, nuestros datos muestran que la eliminación de TLR4 atenuó el desarrollo de GVHD, lo que sugiere que TLR4 podría usarse como un nuevo objetivo y paradigma terapéutico en las terapias de GVHD.

Abstract Background: Graft-versus-host disease (GVHD) is a major complication after allogeneic hematopoietic stem cell transplantation. Objective: To elucidate the role of Toll-like receptor 4 (TLR4), the major receptor for bacterial lipopolysaccharide, in the development of GVHD, we constructed a GVHD model in TLR4 knockout (TLR4-/-) mice and monitored the cell chimerism. Methods: In this study, we used polymerase chain reaction to identify whether TLR4 knockout (TLR4-/-) mice were established. Before transplantation, we pretreated mice with irradiation so as to obtain an appropriate irradiation dose. Flow cytometry was applied to measure the chimerism status, the distributions of antigen-presenting cells (APCs), and T-cells in TLR4+/+ and TLR4-/- recipient mice. Results: The general condition of TLR4-/- recipients was better than that of TLR4+/+ recipients, and the TLR4-/- recipient mice showed less severe GVHD manifestations than the TLR4+/+ recipient mice. Most of the APCs and T-cells in the host mouse spleen were derived from donor cells, and CD4+ T-cells, including memory T-cells, were in the majority in host mice. Conclusion: In general, our data show that TLR4 deletion attenuated GVHD development, which suggests that TLR4 could be used as a novel target and therapeutic paradigm in GVHD therapies.

Abstract The higher acidity in fruits compared with grapes was one of the main obstacles to producing high-quality fruit wines. In the present research, the effect of the split batch fermentation pattern on the metabolites of greengage wines was investigated. This fermentation pattern included whole fruit immersion in the sucrose and coculturing Saccharomyces bayanus Y4 with Torulaspora delbrueckii Y7. These results showed that the contents of total acidity and dominant organic acids were significantly reduced among the batches. Aliphatic compounds were decreased with the batches, while aromatic compounds that contributed to flavor improvement were increased oppositely. The acetate esters, ketones, and terpenes contents were significantly increased in the first batch of coculture. In addition, acidic stress largely affected the kinetics characteristics of greengage wine on a pilot scale. Interestingly, the increment of total acidity was closely associated with the rate of total sugar consumption, which was affected by microbial activities. The acidity content was significantly declined and distinct flavour profiles were formed in split batch and coculture fermentation of greengage wines. Abstract Coronary heart disease (CHD) is one of the main diseases causing morbidity and mortality globally, with oxidative damage and lipid peroxidation associated with oxidative stress and atherosclerosis. Cinnamomum migao is a Miao ethnomedicine that has been shown to provide relief from CHD symptoms. A gas chromatograph-mass spectrometer (GC-MS) was used to determine the chemical components of ten batches of migao fatty oil (MFO), yielding a total of 35 chemical constituents. The antioxidant activity of MFO in vitro was assessed using the DPPH· and ABTS·+ assay methods, and both indicated good antioxidant activity. The network pharmacology predicted the active constituents of MFO and their therapeutic influence on the mechanism of oxidative damage-induced coronary heart disease (OD-CHD). Six compounds were discovered in MFO, including tau-cadinol acetate and dodecanoic acid, which may play a role in OD-CHD by acting on lipids and atherosclerosis, the PI3K-Akt signalling pathway, and other pathways. In conclusion, our study lays the foundation for investigating the use of MFO in the treatment of OD-CHD.

Resumo: Sementes de muitas espécies de peônia contêm grande conteúdo de lipídios. No entanto, a exploração de seu potencial para a produção de óleo é dificultada pela falta de informações básicas sobre a biologia do desenvolvimento das sementes. Objetivou-se determinar o acúmulo de vários compostos de reserva durante o desenvolvimento de sementes da espécie Paeonia lactiflora ‘Hangshao’. As sementes foram coletadas em cinco estádios de desenvolvimento: 30 dias após a floração (DAF), 45 DAF, 60 DAF, 75 DAF e 90 DAF. Foram realizadas análises anatômicas e histológicas e o peso de sementes, conteúdo de água, proteínas, amido e ácidos graxos foram determinados. O período de tempo para o completo desenvolvimento das sementes foi de 90 DAF. As sementes estavam fisiologicamente maduras aos 75 DAF. Durante o desenvolvimento das sementes, o conteúdo de amido e de lipídio aumentaram para, em seguida, diminuir, atingindo o conteúdo máximo aos 60 DAF e aos 75 DAF, respectivamente. O teor de proteínas apresentou uma ligeira diminuição e, em seguida, aumentou. Lipídio foi o principal composto de reserva presente nas sementes maduras. Um total de 26 tipos de ácidos graxos foram detectados entre os quais, sete tipos atingiram mais de 100 mg.Kg-1 em sementes de todos os estádios de desenvolvimento. Esses sete ácidos graxos foram ácido palmítico, ácido esteárico, ácido oleico, ácido trans-oleico, ácido linoleico, ácido α-linolenic e ácido erécico. Além disso, o teor de ácido α-linolenic representou mais de 40% do teor total de ácidos graxos em cada estádio.

Abstract: Seeds of many peony species contain a large amount of oil. However, the exploiting of its potential for oil production is hampered by a lack of basic information regarding the developmental biology of the seeds. Our aim was to obtain a detailed relationship between seed development and accumulation of various storage compounds of Paeonia lactiflora ‘Hangshao’ seed. Seeds were collected at five developmental stages including 30 days after flowering (DAF), 45 DAF, 60 DAF, 75 DAF and 90 DAF. Anatomical and histological analysis, seed weight and water content, proteins, starch, and fatty acids contents were determinated. The time span of seed development for P. lactiflora ‘Hangshao’ was 90 DAF. Seeds were physiologically mature by 75 DAF, with maximum dry matter content. During seed development, the starch and lipid content showed an increased and then decrease pattern, while they reached their maximum content differed with starch for 60 DAF and lipid for 75 DAF. Protein content showed a slight decreased and then increased pattern. Lipid was the main storage reserve of mature seeds. A total of 26 kinds of fatty acid were detected and among which, seven kinds was all more than 100 mg.Kg-1 in all developmental seeds. Those seven fatty acids were palmitic acid, stearic acid, oleic acid, trans-oleic acid, linoleic acid, α-linolenic acid, and erucic acid. Besides, the content of α-linolenic acid accounted for more than 40% of the total fatty acid content in each stage.

ABSTRACT Purpose To study the Periplaneta americana L. extract Ento-B on the treatment of chronic ulcerative colitis induced by 2,4-dinitrochlorobenzene and acetic acid in rats and to explore its primary mechanism of action. Methods Using 2,4-dinitrochlorobenzene combined with acetic acid to induce chronic ulcerative colitis (chronic UC) in rats. The sulfasalazine (400 mg/kg) and Ento-B (200 mg/kg, 100 mg/kg,50 mg/kg) were given by intragastric administration and the effect was evaluated according to the disease activity index (DAI) score, colon mucosal injury index (CMDI) score, histopathological score (HS) and the serum levels of Interleukin-4(IL-4), Interleukin-10(IL-10), Tumor necrosis factor-α(TNF-α), Malondialdehyde(MDA), Superoxide dismutase(SOD) and Inducible nitric oxide synthase(iNOS.) Results Compared with the model group, all doses of Ento-B could reduce the score of CMDI (p < 0.05), HS(p < 0.05 or p < 0.01), significantly increased the expression of IL-4, IL-10, SOD (p < 0.01) and decreased the levels of TNF-α, MDA, iNOS in serum of UC rats, significantly improving the degree of colon lesionsin UC rats. Conclusions Ento-B may play an important role in the treatment of ulcerative colitis induced byUC rats. The mechanism may be related to the increased expression of IL-4, IL-10, SOD and reduced expression of TNF-α, MDA, iNOS.

Total Panax notoginseng saponin (TPNS) is the main bioactivity compound derived from the roots and rhizomes of Panax notoginseng (Burk.) F.H. Chen. The aim of this study was to investigate the effectiveness of TPNS in treating vascular neointimal hyperplasia in rats and its mechanisms. Male Sprague-Dawley rats were randomly divided into five groups, sham (control), injury, and low, medium, and high dose TPNS (5, 10, and 20 mg/kg). An in vivo 2F Fogarty balloon-induced carotid artery injury model was established in rats. TPNS significantly and dose-dependently reduced balloon injury-induced neointimal area (NIA) (P<0.001, for all doses) and NIA/media area (MA) (P<0.030, for all doses) in the carotid artery of rats, and PCNA expression (P<0.001, all). The mRNA expression of smooth muscle (SM) α-actin was significantly increased in all TPNS groups (P<0.005, for all doses) and the protein expression was significantly increased in the medium (P=0.006) and high dose TPNS (P=0.002) groups compared to the injury group. All the TPNS doses significantly decreased the mRNA expression of c-fos (P<0.001). The medium and high dose TPNS groups significantly suppressed the upregulation of pERK1/2 protein in the NIA (P<0.025) and MA (P<0.004). TPNS dose-dependently inhibited balloon injury-induced activation of pERK/p38MAPK signaling in the carotid artery. TPNS could be a promising agent in inhibiting cell proliferation following vascular injuries.

Abstract Purpose: To investigate the mechanism of Periplaneta americana extract promoting intestinal mucosal repair of OXZ-induced colitis in rat. Methods: All experiments used an equal number of male and female SD rats (n=48). We injected OXZ into the colon to induce UC rat model. To determine the optimal concentration of P. Americana's extract (PA-40), it was classified into low (L), medium (M), and high (H) doses. After OXZ treatment, each drug was administered by enema for 7 consecutive days. Rats were divided into the following 6 groups: (1) Saline treatment group (NC), (2) OXZ treatment UC model group (MC), (3) OXZ + budesonide group (BUN), (4) OXZ + PA-40 L group, (5) OXZ + PA-40 M group, (6) OXZ + PA-40 H group. Disease activity index (DAI) scores, colon length, histopathological score, serum cytokine level (IL-4, IL-10, iNOS, tNOS), and amount of MPO, EGF, IL-13 in colonic mucosa were measured. Results: PA treatment had a significant healing effect on the OXZ-colitis model and significantly reduced the lesioned area, especially in the PA-40H groups. PA treatment did not alter the expression of IL-10 and MPO level, but increased EGF (epidermal growth factor) and decrease IL-13 in the colonic tissue. PA inhibited the rise of NOSs (nitric oxide synthase) and decreased the serum IL-4 level. Conclusions: The data suggest that Periplaneta americana extract may be a potential compound for the treatment of colonic lesions. The mechanism may be related to inhibiting the secretion of IL-13 and promoting the formation of EGF.

This is a case report of lichen planus (LP) with multiple system involvement. A 35-year-old female patient was admitted in November 2014 with a 5-year history of painful/difficult sexual intercourse and loss of oral mucosa, and an 8-year history of focal hair loss. Earlier, the patient had been unable to adhere to corticosteroid therapy because of severe adverse side effects. In September 2014, labia minora mucosa defects and stricture of the urethral orifice (with dysuria), vaginal orifice, and vagina were identified. Biopsy was performed and a diagnosis of erosive LP was made. The patient was treated with an oral immunosuppressant (cyclosporine A) and urethral/vaginal dilatation. Urine flow rate and sex life were improved after 6 months and she discontinued medication. Four years later, the patient reported a good overall treatment efficacy. LP can involve multiple systems and should be considered in patients with dyspareunia. Immunosuppressive agents can achieve a satisfactory effect in patients with contraindication to corticosteroid.

OBJECTIVES: This study was conducted to investigate the risk factors for pulmonary abscess-related empyema by investigating the clinical characteristics and chest computed tomography imaging features of patients with pulmonary abscesses. METHODS: We retrospectively analyzed the chest computed tomography findings and clinical features of 101 cases of pulmonary abscess, including 25 cases with empyema (the experimental group) and 76 cases with no empyema (the control group). The potential risk factors for pulmonary abscess-related empyema were compared between the groups by using univariate and multivariate logistic regression analyses. RESULTS: The incidence of pulmonary abscess-related empyema was 24.8% (25/101). Univariate analysis showed that male gender, diabetes, pleuritic symptoms, white blood cells >10×109/L, albumin level <25 g/L, and positive sputum cultures were potential clinical-related risk factors and that an abscess >5 cm in diameter and transpulmonary fissure abscesses were potential computed tomography imaging-related risk factors for pulmonary abscess-related empyema. Multivariate logistic regression analysis showed that transpulmonary fissure abscesses (odds ratio=9.102, p=0.003), diabetes (odds ratio=9.066, p=0.003), an abscess >5 cm in diameter (odds ratio=8.998, p=0.002), and pleuritic symptoms (odds ratio=5.395, p=0.015) were independent risk factors for pulmonary abscess-related empyema. CONCLUSIONS: Transpulmonary fissure abscesses, diabetes, giant pulmonary abscesses, and pleuritic symptoms increased the risk of empyema among patients with pulmonary abscesses.

Abstract Background: Prunella vulgaris L. has been an important medicinal plant for the treatment of thyroid gland malfunction and mastitis in China for over 2000 years. There is an urgent need to select effective wavelengths for greenhouse cultivation of P. vulgaris as light is a very important factor in P. vulgaris growth. Here, we described the effects of natural light (control) and UV solar exclusion on the morphological and physiological traits, secondary metabolites contents and antioxidant activities of P. vulgaris. Results: The results showed that UV solar exclusion resulted in remarkable alterations to morphological and biomass traits; significantly reduced the chlorophyll a, chlorophyll b and total chlorophyll contents; significantly enhanced the ratio of chlorophyll a to b; and significantly increased the carotenoid and anthocyanin contents in P. vulgaris. UV solar exclusion significantly increased the catalase (CAT) and peroxidase (POD) activities, increased superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities and slightly decreased the glutathione (GSH) content. UV solar exclusion significantly increased the soluble sugar and H2O2 contents and increased the soluble protein content but significantly decreased the proline content and slightly decreased the MDA content. The secondary metabolite contents (total phenolics, rosmarinic acid, caffeic acid, hyperoside, ursolic acid and oleanolic acid) and in vitro antioxidative properties (DPPH· and ABTS·+scavenging activities) were significantly increased in P. vulgaris spicas under UV solar exclusion. Additionally, the total polysaccharide and total flavonoids contents were slightly increased by UV solar exclusion. The salviaflaside content was significantly reduced by UV solar exclusion. Conclusion: Our study demonstrated that P. vulgaris activates several antioxidant defence systems against oxidative damage caused by UV solar exclusion.

Preeclampsia is a major reason of morbidity and mortality in pregnant women and perinatal fetus. Hence, it is of prime importance that diagnostic markers are defined to predict chances of preeclampsia in pregnant women. It has been previously shown that microRNA (miRNA)-376c expression is decreased in the placenta of preeclampsia patients at term. Even though this decrease was not mimicked in the placenta at the pre-term stage, miR-376c expression was decreased in the plasma of these patients as early as the second trimester. Plasma and placenta specimens were obtained from pregnant women having unifetal gestation undergoing perinatal care between January 2014 and December 2016 (n=49). Early trimester placentas were collected from patients undergoing terminated pregnancies through dilation and curettage procedure. Our results showed that in addition to miR-376c, miR-441 levels were decreased in the placenta of preeclampsia patients, and this decrease occurred both at pre-term and at term. This decrease is also mimicked in the plasma levels at both early and late weeks of pregnancy, highlighting that miR-441 levels can serve as a diagnostic marker of risk of preeclampsia in pregnant women. Overexpression of the miR-441, as well as miR-376c, promoted cell viability, migration, and invasion in the human immortalized cytotrophoblast cell line HTR8/SVneo, indicating that their decrease in pregnant women would result in anomalous apoptosis and functional imbalance resulting in premature abortion and other complications. MiR-441 level can thus potentially serve as diagnostic marker of preeclampsia in pregnant women.

Abstract This paper deals with a deep circular tunnel excavated in infinite homogeneous and isotropic elasto-viscoplastic rock mass subjected to a hydrostatic initial stresses. The tunnel is divided into the initial plastic, viscoplastic, and elastic zones. By combining the generalized Bingham model with Mohr-Coulomb yield criteria, analytical solutions of the circular tunnel are derived with the consideration of non-associated flow rule and elasto-viscoplasticity. The initial plastic zone is defined as the instantaneous change of rock mass excavation. Based on the initial plastic zone, the stresses in viscoplastic zone are transferred to the deep surrounding rock with time due to the initial earth stresses. The results are compared with the elastic-brittle solution at static conditions, and the solutions of this paper are validated. Moreover, the presented results shows that the stress and displacement of the surrounding rock varies with time, and the solutions can be obtained at different instants of time.

In this work, thermal properties and kinetics of Al-nanoparticles/α-MnO2 nanorods thermite were reported. The α-MnO2 nanorods were synthesized using a hydrothermal method and were characterized by X-ray powder diffraction (XRD) and X-ray photoelectron spectra (XPS), then combined with Al nanoparticles based on the ultrasonic mixing method to prepare the nanostructure thermite. Besides, both pure components and mixture were characterized by field emission scanning electron microscopy (FE-SEM) to observe their morphologies and structures. Subsequently, the thermal properties of Al/α-MnO2 nanostructure thermite were studied on the basis of thermogravimetric-differential scanning calorimetry (TG-DSC). According to the TG-DSC tests, the calculation results of activation energy for kinetics of Al/α-MnO2 thermite were obtained using different isoconversional methods. It was found that Al/α-MnO2 nanostructure thermite has high heat release and low onset temperature, and the heat release of the nanostructure thermite was approximately 1146.6 J g -1.

Pre-compression 3% and pre-stretch 3% subsequent annealing at 200ºC for 2h are conducted on AZ31 magnesium alloys, then inverse tensile and compressive deformation are carried out at room temperature, respectively. During inverse tension 3% deformation on 1st pre-compression samples, detwinning behavior happens; after 2nd pre-compression 3%, the volume fraction of {10-12} extension twins decreases comparing with 1st pre-compression. Due to the interaction of dislocation and induced twinning lamellas, strain hardening rate (θ) increases on 1st and 2nd pre-compression samples. {10-12} tensile twinning is restrained during inverse compressive deformation by pre-stretch process. Owing to the decreasing amount of twins, the texture strengthening in compressive deformation weakens. So the slop of stage III in strain hardening rate sustaining reduces after 1st pre-stretch 3% and 2nd pre-stretch 3% deformation during inverse compression deformation.
