Abstract INTRODUCTION COVID-19 sequelae, or the short-, medium-, and long-term manifestations of the disease are under continuous study. There are currently few reports on the evolution of hematological variables following a demonstrated absence of SARS-CoV-2 after infection. OBJECTIVE Identify hematological alterations in Cuban adults recovered from SARS-CoV-2 infection, and their relation with disease severity. METHODS We selected 348 persons recovered from COVID-19 residing in Havana, Cuba with an RT-PCR study negative for SARS-CoV-2 performed two weeks after hospital discharge; a structured survey was administered to obtain clinical–epidemiological data. Three groups were established according to COVID-19 clinical criteria: asymptomatic, mild/moderately symptomatic, and severely symptomatic, which, in turn, were divided according to hospital discharge date and blood sample collection date. We performed hemograms with differential leukocyte counts and compared results among groups. We then measured the associations between hematological variables, personal medical history, and relevant lifestyle habits (smoking). RESULTS All hematological variables were within normal reference limits, although men from the group of severely ill patients had increased total leukocytes, neutrophils and lymphocytes, and decreased hemoglobin and eosinophils, which was also evident in those with a recovery time of 31–90 days. CONCLUSIONS The relation between hematological variables and degree of clinical severity offers evidence as to persistence of systemic alterations (possibly inflammatory) associated with viral infection. Their identification and characterization can facilitate personalized patient followup and rehabilitation.

ABSTRACT Introduction: Alterations in the cellular redox state have been described as causal factors in various diseases. Reduced glutathione depletion has been fundamentally associated with neurodegenerative, pulmonary, liver, cardiovascular and immunological diseases. Objective: To determine the concentrations of reduced glutathione and the cellular redox status in pediatric patients with immunodeficiencies. Methods: We studied 21 patients with immunodeficiencies from the immunogenetic service, aged between 1 and 8 years and as a control group, 8 children in the same age range as the patients, with normal humoral and cellular immunity studies. Patients diagnosed with immunodeficiency were divided into two groups according to the affected component of the immune response: humoral and cellular. The intraerythrocyte concentrations of oxidized and reduced glutathione were determined by means of an HPLC-UV method. To evaluate the cellular redox state, the relationship between the reduced and oxidized forms of glutathione (GSH/GSSG) was calculated. Results: Reduced glutathione concentrations and cellular redox status were found to be decreased in both groups of patients in relation to children without immunodeficiency (p=0,031 and p=0,03; respectively). Oxidized glutathione showed no difference between the groups. Conclusions: In patients with immunodeficiency, the cellular redox state is affected as a consequence of the decrease in reduced glutathione. This first approach offers the potential for the use of these biomarkers in the comprehensive evaluation of patients with immunodeficiency.

RESUMEN Introducción: Las alteraciones en el estado redox celular se han descrito como factores causales en diversas enfermedades. La depleción del glutatión reducido se ha asociado fundamentalmente a enfermedades neurodegenerativas, pulmonares, hepáticas, cardiovasculares e inmunológicas. Objetivo: Determinar las concentraciones de glutatión reducido y el estado redox celular en pacientes pediátricos con inmunodeficiencias. Métodos: Se estudiaron 21 pacientes con inmunodeficiencias procedentes de la consulta de Inmunogenética, en edades comprendidas entre 1 y 8 años, de ambos sexos, y 8 niños en el mismo rango de edad de los pacientes, como grupo control, con estudios de inmunidad humoral y celular normales. Los pacientes con diagnóstico de inmunodeficiencia se dividieron para su estudio en 2 grupos según el componente afectado de la respuesta inmune: humoral y celular. Fueron determinadas las concentraciones intraeritrocitarias de glutatión reducido y oxidado, mediante un método de HPLC-UV. Para evaluar el estado redox celular se calculó la relación entre las formas reducidas y oxidadas del glutatión (GSH/GSSG). Resultados: Las concentraciones de glutatión reducido y el estado redox celular se encontraron disminuidos en ambos grupos de pacientes en relación con los niños sin inmunodeficiencia (p=0,031 y p=0,03; respectivamente). El glutatión oxidado no mostró diferencias entre los grupos. Conclusiones: En los pacientes con inmunodeficiencia se evidenció la afectación del estado redox celular como consecuencia de la disminución del glutatión reducido. Este primer acercamiento ofreció las potencialidades del empleo de estos biomarcadores en la evaluación integral de pacientes con inmunodeficiencia.

ABSTRACT Background: the relationship between the reduced and oxidized form of glutathione, GSH/ GSSG, is frequently used as an indicator of the cellular redox state. Under conditions where high levels of oxidizing species are generated, reduced glutathione requirements can be increased, and therefore, the cellular redox state can be affected. Objective: to determine the relationship between the cellular redox state and systemic markers of inflammation. Methods: a descriptive study was carried out in a series of 56 cases referred from the immunogenetics clinic, aged between 1 and 76 years old, of both sexes. Erythrocyte sedimentation rate and serum levels of C-reactive protein were determined as systemic markers of inflammation. The GSH/GSSG ratio was calculated from the intraerythrocyte concentrations of reduced glutathione and its oxidized form, which were determined by an HPLC-UV method. Results: the average GSH/GSSG ratio was 7.9 (95 % CI: 6.4-9.4) and age did not influence this proportion. In the cases that had altered values of the inflammation markers, they showed a decrease in the GSH/GSSG ratio. Cellular redox status was negatively correlated with erythrocyte sedimentation values (r=0.41; p=0.017). However, this association does not appear when C-reactive protein levels are analyzed. Conclusions: the patients who present alterations in the inflammation markers show changes in the cellular redox state. The combined study of these biomarkers allows a more comprehensive analysis of the health status of patients with inflammatory diseases.

RESUMEN Fundamento: la relación entre la forma reducida y oxidada del glutatión, GSH/GSSG, es frecuentemente utilizada como indicador del estado redox celular. En las condiciones donde se generan altos niveles de especies oxidantes, se pueden aumentar los requerimientos de glutatión reducido, y por tanto, afectarse el estado redox celular. Objetivo: determinar la relación entre el estado redox celular y marcadores sistémicos de inflamación. Métodos: se realizó un estudio descriptivo en una serie de 56 casos remitidos de la consulta de inmunogenética, en edades comprendidas entre 1 y 76 años, de ambos sexos. Se determinaron la velocidad de eritrosedimentación y los niveles séricos de proteína C-reactiva como marcadores sistémicos de inflamación. La relación GSH/GSSG se calculó a partir de las concentraciones intraeritrocitarias de glutatión reducido y su forma oxidada, las que fueron determinadas mediante un método de HPLC-UV. Resultados: la relación GSH/GSSG promedio fue de 7,9 (IC 95 %:6,4-9,4) y la edad no influyó en esta proporción. En los casos que tenían valores alterados de los marcadores de inflamación, mostraban una disminución en la relación GSH/GSSG. El estado redox celular se correlacionó negativamente con los valores de la eritrosedimentación (r=-0,41; p=0,017). Sin embargo, esta asociación no aparece cuando se analizan los niveles de proteína C-reactiva. Conclusiones: los pacientes que presentan alteraciones en los marcadores de inflamación muestran cambios en el estado redox celular. El estudio combinado de estos biomarcadores permite realizar un análisis integral del estado de salud del paciente con enfermedades inflamatorias.

ABSTRACT Introduction: Havana city was the most complex territory at the national level facingthe first outbreak of COVID-19. Its condition of capital, the population density, the limited geographical delimitation between its municipalities, the high inter-municipal and inter-provincial flow of people, added to the fact that it is the province that receives the largest number of travelers from abroad, conferred particularities on this territory. Objective: To determine the main clinical-epidemiological characteristics of patients affected by COVID-19 living in Havana. Methods: An observational, analytical, cross-sectional study was carried out. The sample contained 431 patients, older than one year, with epidemiological discharge from the SARS-CoV-2 infection. To collect information, a questionnaire was applied through an interview. Results: The average age of the sample was 45 years. More than half of the cases evolved in a symptomatic way (not serious or serious). Fever was the most frequent clinical manifestation. The individuals included in the sample, with blood group O or with a background of asthma, showed less risk of presenting symptomatic forms of the disease. Conclusions: The results suggest that age is the main risk factor for developing symptomatic forms (not severe or severe) of COVID-19. On the contrary, blood group O and a history of asthma are protective factors for these forms of clinical evolution.

RESUMEN Introducción: La Habana fue el territorio más complejo a nivel nacional en el enfrentamiento al primer brote de la COVID-19. Su condición de capital, la densidad poblacional, la escasa delimitación geográfica entre sus municipios, el elevado flujo intermunicipal e interprovincial de personas, sumado a que es la provincia que mayor número de viajeros recibe procedentes del extranjero, confirieron a este territorio particularidades en el enfrentamiento a la pandemia. Objetivo: Determinar las principales características clínico-epidemiológicas de pacientes cubanos residentes en La Habana afectados por la COVID-19. Métodos: Se realizó un estudio observacional, analítico, de corte transversal. La muestra quedó formada por 431 pacientes, mayores de un año, de alta epidemiológica de la infección por SARS-CoV-2. Para la recolección de información se aplicó un cuestionario a través de una entrevista. Resultados: La edad promedio de la muestra fue de 45 años. Más de la mitad de los casos evolucionaron de modo sintomático (no grave o grave). La fiebre fue la manifestación clínica más frecuente. Los individuos incluidos en la muestra, con grupo sanguíneo O, o con antecedentes de asma, mostraron menos riesgo de presentar formas sintomáticas de la enfermedad. Conclusiones: Los resultados sugieren que la edad constituye el principal factor de riesgo para desarrollar formas sintomáticas (no graves o graves) de la COVID-19. De modo contrario, el grupo sanguíneo O y el antecedente de asma son factores de protección para estas formas de evolución clínica.

ABSTRACT Non-infectious anterior uveitis is an inflammatory disease of the eye that affects the uveal tract and can cause total blindness and other visual disabilities. Autoimmune and inflammatory diseases are associated with qualitative and quantitative alterations in the immune response; therefore, the levels of tetanus and diphtheria antitoxin related to the HLA-B27 allele were evaluated in Cuban patients with non-infectious anterior uveitis. Tetanus and diphtheria antitoxin levels were determined by ELISA in 190 patients with non-infectious anterior uveitis and healthy control individuals. 97.37% of patients with uveitis showed protective tetanus antitoxin levels greater than and equal to 0.1 IU/mL as well as healthy controls (98.95%) (p=0.4385). The proportions of patients with non-infectious anterior uveitis and presumably healthy controls in the different levels of protective tetanus antitoxin were similar (p>0.05) at all levels of protection, as were the geometric mean titers for this antitoxin (p=0.2907). Patients with uveitis aged 65 years or older had a higher proportion of individuals with long-term reliable protective titers (>1.0 IU/mL) of diphtheria antitoxin (p=0.0065). In uveitis patients, no association was observed between the presence of the HLA-B27 allele and the antibody response against tetanus toxoid (p=0.6196) and diphtheria (p=0.1917). Similarly, 37.9% of patients with uveitis and 42% of their controls had non-protective titers (<0.1 IU/mL) of diphtheria antitoxin (0.1148). Most patients with anterior uveitis and control subjects were protected against tetanus (p>0.05), while in patients with uveitis and supposedly healthy controls, aged 18 years or older, the administration of booster doses with diphtheria toxoid should be reevaluated to achieve higher levels of protection against diphtheria.

RESUMEN La uveítis anterior no infecciosa es una enfermedad inflamatoria del ojo que afecta al tracto uveal y que puede causar ceguera total y otras discapacidades visuales. Esta enfermedad se ubica en el espectro de enfermedades autoinmunes y autoinflamatorias. Se han descrito respuestas no adecuadas a la vacunación en enfermedades mediadas por el sistema inmune, por lo que se evaluaron los niveles de antitoxina tetánica y diftérica en pacientes cubanos con uveítis anterior no infecciosa, relacionada con el alelo HLA-B27. Se determinaron los niveles de antitoxina tetánica y diftérica mediante ELISA en 190 pacientes con uveítis anterior no infecciosa y controles supuestamente sanos. El 97,37% de los pacientes con uveítis mostraron niveles de protección de antitoxina tetánica mayor o igual a 0,1 UI/mL, similar a lo observado en los controles sanos (98,95%) (p=0,4385). Las proporciones de pacientes con uveítis anterior no infecciosa y sus controles en los diferentes niveles de protección de antitoxina tetánica fueron similares (p>0,05), al igual que los títulos medios geométricos (p=0,2907). En los pacientes con uveítis, de 65 años o más, se detectó una mayor proporción de individuos con títulos protectores de larga duración (>1,0 UI/mL) de antitoxina diftérica (p=0,0065). En los pacientes con uveítis no se observó asociación entre la presencia del alelo HLA-B27 y la respuesta de anticuerpos frente al toxoide tetánico (p=0,6196) y diftérico (p=0,1917). El 37,9% de los pacientes con uveítis y el 42% de los controles, presentaron títulos no protectores (<0,1 UI/mL) de antitoxina diftérica (0,1148). La mayoría de los pacientes con uveítis anterior no infecciosa y los controles supuestamente sanos presentaron protección frente al toxoide tetánico; mientras que, en los pacientes con uveítis, así como en los controles supuestamente sanos, con edad igual o más de 18 años, se debe reevaluar incluir refuerzos con toxoide diftérico para alcanzar mayores niveles de protección frente a la difteria.

ABSTRACT Introduction: Until June 11, 2020, 10.4% of the total positive cases of SARS-CoV-2 in Cuba corresponded to patients under 18 years of age. Clinical manifestations in children affected by SARS-CoV-2 are usually mild and few deaths occur compared to other age groups. Objective: Characterize from the clinical, epidemiological and immunological point of view the Cuban pediatric patients infected by SARS-CoV-2 until June 11, 2020. Methods: Observational, cross-sectional study in 137 pediatric patients infected with COVID-19, who were epidemiologically discharged. Results: 56.9 % of the patients presented asymptomatic form of the disease. Havana was the most affected province and the average age was 11.3 years. The most frequent symptoms were: fever, cough and diarrhea and occurred more frequently in patients with black skin (Odd Ratio = 3.40). Antibody concentrations for tetanus and Haemophilus influenzae type b provided protection in 92.7% to 100% of children, while for diphtheria 24.8% had no reliable protection. Of the symptomatic patients, 62.5% developed specific total antibodies against SARS-CoV-2. Conclusions: Patients in pediatric ages can contribute significantly to the transmission dynamics. Reliable protection for specific vaccines, the development of total antibodies to SAR-Cov-2, and general favorable prognosis without complications require continued research to identify biological and immunological differences between children and other age groups.

RESUMEN Introducción: Las manifestaciones clínicas en niños afectados por el SARS-CoV-2 suelen ser más leves, un número reducido evoluciona hacia la gravedad y se producen pocas muertes en comparación con otros grupos de edad. Objetivo: Caracterizar desde el punto de vista clínico, epidemiológico e inmunológico a los pacientes pediátricos en Cuba infectados por SARS-CoV-2 a partir del 11 de marzo de 2020 y que se encontraban de alta epidemiológica el 11 de junio del mismo año. Métodos: Estudio observacional, de corte transversal en 137 pacientes en edad pediátrica infectados por la COVID-19, que se encontraban de alta epidemiológica. Resultados: Presentaron forma asintomática de la enfermedad 56,9 %. La Habana fue la provincia más afectada y la edad promedio de 11,3 años. Los síntomas más frecuentes fueron: fiebre, tos y diarreas y se presentaron con mayor frecuencia en pacientes de color de piel negra (Odd Ratio= 3,40). Las concentraciones de anticuerpos para tétano y Haemophilus influenzae tipo b proporcionaron protección entre 92,7 y 100 % de los niños, mientras que para la difteria 24,8 % no tuvo protección confiable. De los sintomáticos, 62,5 % desarrollaron anticuerpos totales específicos frente al SARS-CoV-2. Conclusiones: Los pacientes en edades pediátricas pueden contribuir de manera importante a la dinámica de la transmisión. La protección confiable para vacunas específicas, el desarrollo de anticuerpos totales frente a SAR-Cov-2 y el pronóstico general favorable sin complicaciones, obliga a continuar las investigaciones para identificar las diferencias biológicas e inmunológicas entre niños y otros grupos de edad.

ABSTRACT Introduction: Flow cytometry allows quantification of lymphocyte subpopulations with high sensitivity, specificity and objectivity. These advantages are only achieved through the hardworking process of individualized and controlled design for each experiment. Objective: To design a flow cytometry protocol of a single polychromatic tube for peripheral lymphocyte immunophenotype. Methods: An experimental in vitro study was carried out, in March 2019, with peripheral blood samples obtained from three healthy volunteers, at the National Center for Medical Genetics. The tube was made up of six lineage markers for identifying natural B and T lymphocytes, natural killers and natural killer T cells. A protocol was developed for red blood cell lysis without washing. Fluorochrome-conjugated monoclonal antibodies were used. The optimal point of concentration corresponded to the highest staining index and preservation of the positivity percentages of each population. Progressive tube construction was performed and a logical window sequence strategy was proposed for data analysis. Results: The chosen markers allowed to carry out correct peripheral lymphocyte immunophenotype. Good discriminations between positive and negative signals were observed at the five titration points, except for anti-CD56, which presented a decreasing trend in the staining index. The total volume of conjugates required for determination of the six antigens was 3.75 μL per tube. Conclusions: A polychromatic tube was obtained that allows to carry out peripheral immunophenotype quickly and precisely by six lymphocyte antigens simultaneously, with the use of small volumes of conjugate and blood.

RESUMEN Introducción: La citometría de flujo permite la cuantificación de las subpoblaciones de linfocitos con una elevada sensibilidad, especificidad y objetividad. Estas ventajas solo se logran con un proceso laborioso de diseño individualizado y controlado para cada experimento. Objetivo: Diseñar un protocolo de un solo tubo policromático de citometría flujo para inmunofenotipo linfocitario periférico. Métodos: Se realizó un estudio experimental in vitro con muestras de sangre periférica obtenidas de tres voluntarios sanos, en el Centro Nacional de Genética Médica, en marzo de 2019. El tubo se compuso de seis marcadores de linaje para identificar linfocitos B, T, natural killer y natural killer T. Se desarrolló un protocolo de lisis de hematíes sin lavado. Se emplearon anticuerpos monoclonales conjugados con fluorocromos. El punto óptimo de concentración correspondió al mayor índice de tinción y conservación de los porcentajes de positividad de cada población. Se realizó la construcción progresiva del tubo y se propuso una estrategia lógica de secuencia de ventanas para el análisis de datos. Resultados: Los marcadores seleccionados permitieron realizar correctamente el inmunofenotipo linfocitario periférico. En los cinco puntos de titulación se observaron buenas discriminaciones entre las señales positivas y negativas, excepto para el anti-CD56 que presentó una tendencia decreciente del índice de tinción. El volumen total de conjugados requeridos para la determinación de los 6 antígenos fue de 3,75 μL por tubo. Conclusiones: Se obtuvo un tubo policromático que permite el inmunofenotipo periférico de forma rápida y precisa por seis antígenos linfocitarios simultáneamente, con el empleo de pequeños volúmenes de conjugado y sangre.

ABSTRACT In Cuba, the UMELISA HCG, produced by the Immunoassay Center, detects the presence of human chorionic gonadotropin. With the aim to purify and characterize human chorionic gonadotropin (hCG) for diagnostic use, was developed a procedure for its purification from the urine of pregnant women, with a capture stage and a phase of immune affinity chromatography, using a monoclonal antibody anti-chain β hCG. The highest mass of hCG (74,27 %) in the precipite was obtained at 50 % saturation of (NH4)2SO4, 22 (C and 3 h of rest,. The specific activity was 12 417,65 IU/mg. PAGE-SDS showed the bands of the α and β chains of the hormone, with similar mobility to the control. The purity by densitometry was 97,3 %. By gel filtration a molar mass of 40 kDa was obtained. Using this procedure human chorionic gonadotropin with the required quality to get biological reagents for the UMELISA HCG was obtain.

RESUMEN En Cuba, el UMELISA HCG, producido por el Centro de Inmunoensayo, detecta la presencia de gonadotropina coriónica humana. Con el objetivo de purificar y caracterizar gonadotropina coriónica humana (hCG) para uso diagnóstico, se desarrolló un procedimiento para su obtención a partir de orina de gestantes, con una etapa de captura y una de purificación por cromatografía de inmunoafinidad, empleando un anticuerpo monoclonal anticadena β hCG. La mayor masa de hCG en el precipitado (74,27 %) se obtuvo al 50 % de saturación del (NH4)2SO4, 22 (C y 3 h de reposo. La actividad específica fue 12 417,65 UI/mg. El PAGE-SDS mostró las bandas de las cadenas α y β, con movilidad similar al control. La pureza por densitometría fue 97,3 %. Por filtración en gel se obtuvo una masa molar de 40 kDa. Con la metodología empleada se obtuvo gonadotropina coriónica humana con la calidad requerida para la obtención de los reactivos biológicos del UMELISAHCG.

ABSTRACT Introduction: Diphtheria still persists in many countries. In Cuba, studies conducted in different age groups have demonstrated that there are non-protective levels of diphtheria antitoxin in the population, so it is necessary to have methods that allow the serologic survey of population immunity. The quantification of antibodies against vaccine antigens such as diphtheria toxin is also a useful, rapid and economic method to evaluate the immune response. Objective: To validate an ELISA-type immune-enzymatic test to quantify the levels of diphtheria antitoxin in human serum. Material and Method: An experimental study of technological development was carried out in the Immunology Laboratory of the National Medical Genetics Center, Havana, Cuba. The optimal values of the variables that influence on the result of the indirect heterogeneous immune-enzymatic test for the quantification of diphtheria antitoxin were determined. The calibration curve obtained was evaluated against the WHO standard (Diphtheria Antitoxin Human Serum 00/496). The analytical validation of the standardized method was performed. Results: The intra-assay and inter-assay coefficients of variation were less than 10% and 20%, respectively. Recovery values between 90 and 110% were found in accuracy and selectivity. The parallelism between the standard curve and the samples studied showed a coefficient of variation lower or equal to 10%. The limit of quantification was 0,015 IU/mL and the one of detection was 0,0039 IU/mL. Conclusions: The result obtained in the precision, accuracy and selectivity of the ELISA-type immune-enzymatic test developed and validated in the National Medical Genetics Center demonstrated that it can be used in the clinical practice to quantify the values of diphtheria antitoxin in human serum.

RESUMEN Introducción: La difteria aún persiste en numerosos países. En Cuba, estudios realizados en diferentes grupos etarios han demostrado que existen niveles no protectores de antitoxina diftérica en la población, por lo que es necesario contar con métodos que permitan la estimación serológica de la inmunidad poblacional. La cuantificación de anticuerpos contra antígenos vacunales como la toxina diftérica es además un método útil, rápido y económico para evaluar la respuesta inmune. Objetivo: Validar un ensayo inmunoenzimático tipo ELISA para cuantificar los niveles de antitoxina diftérica en suero humano. Material y Método: Se realizó un estudio experimental de desarrollo tecnológico, en el cual se determinaron los valores óptimos de las variables que influyen en el resultado de un ensayo inmunoenzimático heterogéneo indirecto para la cuantificación de antitoxina diftérica, desarrollado en el laboratorio de Inmunología del Centro Nacional de Genética Médica de Cuba. La curva de calibración se evaluó contra el estándar de la OMS (Diphtheria Antitoxin Human Serum 00/496). Se realizó la validación analítica del método estandarizado. Resultados: Los coeficientes de variación intraensayo e interensayo fueron inferiores a 10% y 20%, respectivamente. En la exactitud y selectividad se encontraron valores de recobrado entre 90 y 110%. El paralelismo entre la curva estándar y las muestras estudiadas presentó un coeficiente de variación menor o igual a 10%. El límite de cuantificación fue 0,015 UI/mL y el de detección 0,0039 UI/mL. Conclusiones: El resultado obtenido en la precisión, exactitud y selectividad del ensayo inmunoenzimático tipo ELISA desarrollado demostró que puede ser utilizado en la práctica clínica para cuantificar los valores de antitoxina diftérica en suero humano.

Abstract Introduction During the acute phase of infection with Hepatitis B Virus (HBV) and the initial period of chronic infection, DNA is episomally (free or extrachromosomal) and replicates in hepatocytes producing among other virions, DNA polymerase and virus surface antigen (HBsAg). Objective Validate an ELISA to quantify HBsAg levels in patients with chronic hepatitis B. Method An experimental study of technological developmentwas conducted. The normalization and validation of heterogeneous double antibody enzyme linked inmunoassay for quantitation of HBsAg in human sera was carried out. 115 serum samples from patients with chronic hepatitis B with viral load results were correlated with its HBsAg concentrations. Results and discussion The developed method presented variation coefficients intra and interassay of 9.8 and 13.2% respectively. The working range was estimated between 0.15 and 60 ng/mL. The recovery rate was between 90 and 110% and the linear fit of the standard curve presented a determination coefficient greater than 0, 99. The correlation between the DNA levels and HBsAg concentration was 62.5%. Conclusions The assessment of the ELISA for quantitation of HBsAg developed in the laboratory showed that it achieved validation parameters for its clinical use.

Resumen Introducción Durante la etapa aguda de la infección por el Virus de la Hepatitis B (VHB) y el período inicial de una infección crónica, el DNA está en forma episomal (libre o extracromosomal) y se replica en el hepatocito produciendo, entre otros, viriones infectivos, DNA polimerasa y antígeno de superficie del virus (HBsAg). Objetivo Validar un inmunoensayo tipo ELISA para cuantificar los niveles de HBsAg en pacientes con hepatitis B crónica. Método Se realizó un estudio experimental de desarrollo tecnológico. Se llevó a cabo la normalización y validación de un inmunoensayo enzimático heterogéneo de doble anticuerpo para la cuantificación de HBsAg en sueros de seres humanos. 115 muestras de pacientes con hepatitis B crónica con resultados de carga viral se correlacionaron con las concentraciones de HBsAg. Resultados y discusión El método presentó coeficientes de variación intra e interensayo de 9,8 y 13,2% respectivamente. El rango de trabajo se estimó entre 0.15 y 60 ng/mL. El porcentaje de recuperación estuvo entre el 90 y 110% y el ajuste lineal de la curva estándar presentó un coeficiente de determinación superior a 0,99. La correlación alcanzada entre los niveles de DNA y la concentración de HBsAg fue de 62.5%. Conclusiones La evaluación del ELISA para la cuantificación de HBsAg desarrollado en el laboratorio mostró que cumple los parámetros de validación para su uso clínico.

Introduction: the Titanic antitoxin quantification is useful to assess the immune response against vaccinal antigens. Objective: to develop a standardization and validation technique of the indirect ELISA to quantify titanic antitoxin in human serum. Materials and Methods: the standard preparing to the assay curve was calibrated front the preparing Finlay Institute. The optimal indicators were normalized to each technique step developed. Results: a good intra-assay precision was reach during validation in all cases. The coefficient of variation to parallelism was less than 10% and 110%. Detection limit was 0.002 UI/ ml. Analytic sensitivity reached was 0.01 UI/ ml. Conclusions: the assay may be used to quantify Titanic antitoxin in human serum.

Introducción: la cuantificación de antitoxina tetánica es útil para la evaluación de la respuesta inmune humoral frente a antígenos vacunales. Objetivo: desarrollar la estandarización y validación técnica de un ensayo de inmunoabsorción ligado a enzima (ELISA) de tipo indirecto para cuantificar antitoxina tetánica en suero humano. Material y Método: el estándar preparado para la curva del ensayo se calibró frente al preparado en el Instituto Finlay. Se normalizaron los indicadores óptimos para el desarrollo de cada paso de la técnica. Resultados y Discusión: durante la validación se alcanzó una buena precisión intraensayo e interensayo en todos los casos. Al evaluar el paralelismo el coeficiente de variación fue menor de 10%. El ensayo mostró una excelente exactitud, con valores de recuperación entre 90% y 110%. El límite de detección fue 0,002 UI/mL. La sensibilidad analítica alcanzada fue de 0,01 UI/mL. Conclusiones: el ensayo puede ser utilizado para la cuantificación de antitoxina tetánica en suero humano.
