Abstract Objective To explore the molecular mechanism of neuropathologic damage induced by radiofrequency ablation at different temperatures. Methods This is basic research, and 36 SD rats were used to construct the neuropathological injury model. The rats were subjected to radiofrequency stimulation at different temperatures and were divided into 6 groups according to the temperature injury: 42°, 47°, 52°, 57°, 62°, and 67°C groups. Conduction time, conduction distance, and nerve conduction velocity were recorded after temperature injury. HE-staining was used to observe the histopathological morphology of the sciatic nerve. The expression of SCN9A, SCN3B, and NFASC protein in sciatic nerve tissue were detected by western blot. Results With the increase in temperature, nerve conduction velocity gradually decreased, and neurons were damaged when the temperature was 67°C. HE-staining showed that the degrees of degeneration of neurons in rats at 47°, 52°, 57°, 62°, and 67°C were gradually increased. The expression of SCN9A, SCN3B protein in 57°, 62°, 67°C groups were much higher than that of NC, 42°, 47°, 52°C groups. However, the expression of NFASC protein in 57°, 62°, 67°C groups was much lower than that of the NC, 42°, 47°, 52°C groups. Conclusion There was a positive correlation between temperature caused by the radiofrequency stimulation to neuropathological damage. The mechanism is closely related to the expression of SCN9A, SCN3B, and NFASC protein in nerve tissue caused by heat transfer injury.

Glycemic variability (GV) may be linked to the development of diabetic complications by inducing inflammation, oxidative stress, and endothelial dysfunction. Flash glucose monitoring (FGM) provides a novel method of continuously monitoring interstitial glucose levels for up to 14 days. This study randomly assigned poorly controlled type 2 diabetes mellitus patients treated with metformin and multiple daily injections of insulin (n=60) to either continuous subcutaneous insulin infusion (CSII) treatment or CSII in combination with liraglutide (CSII+Lira) treatment for 14 days during hospitalization. GV was assessed using a FGM system; weight and cardiometabolic biomarkers were also evaluated. The coefficient of variation was significantly reduced in the CSII+Lira group (P<0.001), while no significant change was observed in the CSII group. The changes differed significantly between the two groups in mean amplitude of glycemic excursions (P=0.004), standard deviation (P=0.006), and the percentage of time in the target range (4–10 mmol/L, P=0.005 and >10 mmol/L, P=0.028). The changes in mean of daily differences, interquartile range, and percentage of time in hypoglycemia (<3.3 mmol/L) and hyperglycemia (>13.9 mmol/L) identified by FGM showed no difference. Treatment with liraglutide increased serum adiponectin [33.5 (3.5, 47.7) pg/mL, P=0.003] and heme oxygenase-1 levels [0.4 (–0.0, 1.8) ng/mL, P=0.001] and reduced serum leptin levels [–2.8 (3.9) pg/mL, P<0.001]. Adding the glucagon-like peptide-1 analog liraglutide improved GV, weight, and some cardiometabolic risk markers. The FGM system is, therefore, shown to be a novel and useful method for glucose monitoring.

Resumo: O objetivo deste estudo é a elaboração de uma ferramenta quantitativa, baseada em técnicas de Geomarketing e Aprendizado de Máquina, capaz de identificar oportunidades de negócio e contribuir para o processo estratégico de escolha locacional de uma rede de franquias, selecionando regiões que possuam uma alta previsão de demanda e uma carência na oferta do produto. Além disso, realizou-se uma análise qualitativa dos pontos comerciais selecionados com base em critérios definidos no decorrer do trabalho. Essa previsão se dá pela construção de um padrão de consumo, definido por um classificador, baseado nas características dos indivíduos que costumam comprar o produto. Inicialmente, para um melhor entendimento a respeito do assunto, foi feito um embasamento teórico abarcando os principais conceitos sobre Geomarketing e Aprendizado de Máquina e suas aplicações. Em seguida, para a demonstração dos resultados, optou-se pela aplicação do método para o mercado de chocolates finos (Cacau-Show) no Distrito Federal. As principais bases de dados utilizadas neste trabalho foram provenientes da Pesquisa de Orçamentos Familiares e do Censo Demográfico, ambos desenvolvidos pelo Instituto Brasileiro de Estatística e Geografia (IBGE). Como resultado, obteve-se o Gasto Padronizado, que indica o nível de demanda para cada Setor Censitário, as informações georreferenciadas da concorrência, contendo 44 lojas que possuem como principal produto o chocolate fino, e as malhas digitais do Distrito Federal. O cruzamento dessas informações permitiu a elaboração de um mapa que facilita identificação das oportunidades de negócio para o mercado de chocolates finos no Distrito Federal.

Abstract: The objective of this study is to develop a quantitative tool, based on Machine Learning and Geomarketing to identify business opportunities and contribute to the strategic process of local choice of franchises’ network selecting regions that have a high demand forecast and a lack of product supply. In addition, we conducted a qualitative analysis of the selected business places based on defined criteria. This prediction is given by constructing a consumption pattern, defined by a classifier, based on the characteristics of the reserved rights. Initially, for a better understanding on this subject, a theoretical background was made covering the main concepts about Geomarketing and Machine Learning and its applications. After that for a demonstration of the results, we opted for the application of the method for the market of fine chocolates (Cacau-Show) in the Distrito Federal. The main databases used in this paper were Pesquisa de Orçamentos Familiares and from Instituto Brasileiro de Estatística e Geografia (IBGE). As a result, the Standardized Spend was obtained, which indicates the requirement for each Censitar Sector, as georeferenced information of the competition, containing 44 stores that have as their main product of fine chocolate, and as digital meshes of the Federal District. The crossing is available for the elaboration of a map that facilitates the identification of the business opportunities for the market of fine chocolates in the Distrito Federal, Brazil.

ABSTRACT Traditional villages in the northern Jiangsu province have a local character and history unique artistic style, traditional culture and art Jiangsu indispensable part. How to make achieving sustainable and harmonious for development in rural areas, it is a major problem currently in these places. In this paper, field research and theoretical analysis a multidisciplinary approach, the traditional villages of northern Jiangsu space environment features, including location and spatial pattern are elaborated, analyzed the current conditions of rural development. Then taking Pengzhuang Village in the northern Jiangsu as example, discuss the current situation as well as its architectural characteristics of the space environment group and architectural types. Discussing the necessity of architectural features traditional villages in Northern implementation of conservation is proposed as the main ecological environmental protection and rural development policy space. Beautiful traditional village development should be coordinated development of economic and social environment, especially in the process of our socialist new rural construction, village spatial patterns of Jiangsu for basic research has positive practical value and far-reaching historical significance.

Intravesical chemotherapy is an important part of the treatment for superficial bladder cancer. However, the response to it is limited and its side effects are extensive. Functional single-walled carbon nanotubes (SWNT) have shown promise for tumor-targeted accumulation and low toxicity. In the present study, we performed in vivo and in vitro investigations to determine whether SWNT-based drug delivery could induce high tumor depression in rat bladder cancer and could decrease the side effects of pirarubicin (tetrahydropyranyl-adriamycin, THP). We modified SWNT with phospholipid-branched polyethylene glycol and constructed an SWNT-THP conjugate via a cleavable ester bond. The cytotoxicity of SWNT-THP against the human bladder cancer cell line BIU-87 was evaluated in vitro. Rat bladder cancer in situ models constructed by N-methyl-N-nitrosourea intravesical installation (1 g/L, 2 mg/rat once every 2 weeks for 8 weeks) were used for in vivo evaluation of the cytotoxicity of SWNT and SWNT-THP. Specific side effects in the THP group including urinary frequency (N = 12), macroscopic hematuria (N = 1), and vomiting (N = 7) were identified; however, no side effects were observed with SWNT-THP treatment. Flow cytometry was used to assess the cytotoxicity in vitro and in vivo. Results showed that SWNT alone did not yield significant tumor depression compared to saline (1.74 ± 0.56 and 1.23 ± 0.42%) in vitro. SWNT-THP exhibited higher tumor depression than THP-saline in vitro (74.35 ± 2.56 and 51.24 ± 1.45%) and in vivo (52.46 ± 2.41 and 96.85 ± 0.85%). The present findings indicate that SWNT delivery of THP for the treatment of bladder cancer leads to minimal side effects without loss of therapeutic efficacy. Therefore, this nanotechnology may play a crucial role in the improvement of intravesical treatment of bladder cancer.

Sichuan basin, situated in the west of China, is the fourth biggest basin in China. In order to describe a systematic study of the cry2-type genes resources from Bacillus thuringiensis strains of Sichuan basin, a total of 791 Bacillus thuringiensis strains have been screened from 2650 soil samples in different ecological regions. The method of PCR-restriction fragment length polymorphism (PCR-RFLP) was used to identify the type of cry2 genes. The results showed that 322 Bacillus thuringiensis strains harbored cry2-type genes and four different RFLP patterns were found. The combination of cry2Aa/cry2Ab genes was the most frequent (90.4%), followed by cry2Aa (6.8%) and cry2Ab alone (2.5%), and only one novel type of cry2 gene was cloned from one isolate (JF19-2). The full-length of this novel gene was obtained by the method of thermal asymmetric interlaced PCR (Tail-PCR), which was designated as cry2Ag1 (GenBank No. ACH91610) by the Bt Pesticide Crystal Protein Nomenclature Committee. In addition, the result of scanning electron microscopic (SEM) observation showed that these strains had erose, spherical, bipyramidal, and square crystal. And the results of sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) demonstrated that these strains harbored about one to three major proteins. These strains exhibited a wide range of insecticidal spectrum toxic to Aedes aegypti (Diptera) and Pieris rapae Linnaeus, 1758 (Lepidoptera). Particularly, JF19-2 contained cry2Ag gene had the highest insecticidal activity. All these researches mentioned above revealed the diversity and particularity of cry2-type gene resources from Bacillus thuringiensis strains in Sichuan basin.
