Abstract Introduction: Propofol is a widely used anesthetic and its dose is closely related to aging. Telomere length (TL) is a unique heritable trait, and emerging as a biomarker of aging, health and disease. Telomerase RNA component (TERC) plays an important role in maintaining TL. We proposed a hypothesis that propofol dose in general anesthesia can be predicted by measuring TL before operation, which greatly reduced the risk of anesthesia, especially the elderly. Methods: The association between the propofol dose in anesthesia induction and: TL in the DNA of peripheral blood leukocytes; body weight; sex; difference of the Bispectral Index (BIS) before and after anesthesia induction in patients was evaluated by multivariable linear regression analyses. The mutation at the 5’end or 3’end of TERC was detected. We recruited 100 patients of elective surgery. Results: We found that propofol dose in anesthesia induction was clearly correlated significantly with TL (r = 0.78, p < 0.001), body weight (r = 0.84, p = 0.004), sex (r = 0.83, p= 0.84, p = 0.004), sex (r = 0.83, p = 0.004), and difference of BIS before and after anesthesia induction (r = 0.85, p = 0.029). By comparing the absolute values of standardized regression coefficients (0.58, 0.21, 0.19, and 0.12) of the four variables, it can be seen that TL contributes the most to the propofol dose in anesthesia induction. However, the mutation at the 5’ end or 3’ end of TERC was not found. Conclusions: These findings provide preliminary evidence that the propofol dose in anesthesia induction was clearly correlated with genetically determined TL. TL may be a promising predictor of the propofol dose, which is beneficial to improve the safety of anesthesia and reduce perioperative complications.

Abstract Backgroud Homocitrulline (Hcit), is involved in the pathological processes of some diseases. However, the role and function of Hcit (CBL) in human skin remains largely obscure. Objective To investigate the correlation of the level of Hcit in seborrheic keratosis, skin aging, and its clinical significance. Methods Immunohistochemistry was used to analyze the level of Hcit in skin lesions of seborrheic keratosis (SK), unaffected skin (distant 0.5 centimeters from SK lesion), and normal skin of healthy subjects in the control group. ELISA test was used to detect the serum level of CBL in SK patients and healthy subjects of different ages. Results Hcit was mainly localized in the nucleus of epidermal cells. In healthy control skin, the expression of Hcit increased with age and showed a positive correlation with age (the correlation coefficient was 0.806, p = 0.0002). The expressional level of Hcit in SK lesions was higher than that in healthy control skin (Z = −3.703, p = 0.0002). The serum level of CBL in healthy subjects and in SK patients increased with age (the correlation coefficient were 0.5763, p = 0.0032; 0.682, p = 0.004. respectively). The serum level of CBL in SK patients was higher than that in healthy subjects (Z = −2.19, p = 0.030). Study limitations The small serum sample size in the study. Conclusion The high expressional level of Hcit is correlated with seborrheic keratosis and skin aging. HCit may be one of the potential biomarkers of skin aging.

RESUMO Introdução: O treinamento em artes marciais concentra-se na ciência, na metodologia e na prática. As artes marciais são um símbolo de aptidão física. A análise acadêmica da influência do treinamento de artes marciais na saúde física das crianças é de grande importância para a promoção da cultura tradicional chinesa e pode enriquecer os programas de intervenção esportiva para melhorar a saúde física infantil. Objetivo: Analisar o efeito do treinamento de qualidade sensitiva no treinamento de equilíbrio das artes marciais em crianças. Métodos: Selecionou-se crianças de 8 a 12 anos como objetos de pesquisa. Após repetidas deliberações, foi elaborado e implementado um conjunto de artes marciais infantis. O projeto piloto analisa a importância da qualidade sensitiva na etapa essencial do treinamento das artes marciais infantis. Resultados: Após o experimento, houve uma diferença significativa na qualidade do equilíbrio no grupo experimental (p<0,01). Após o experimento, a diferença no sentido de equilíbrio entre os grupos experimental e de controle foi significativa (p<0,01). Conclusão: Na fase de treinamento básico das artes marciais infantis, os treinadores devem exigir que os atletas dominem as habilidades básicas das artes marciais de forma mais abrangente. Todas as combinações de movimentos nas artes marciais exigem aptidão física e sensibilidade de equilíbrio abrangentes, incluindo velocidade, flexibilidade e coordenação. ABSTRACT Introduction: Martial arts training focuses on science, methodology, and practice. Martial arts are a symbol of physical fitness. The academic analysis of the influence of martial arts training on children’s physical health is of great importance for promoting traditional Chinese culture. It can enrich sports intervention programs to improve children’s physical health. Objective: Analyze the effect of sensory quality training on martial arts balance training in children. Methods: Children aged 8 to 12 years were selected as research subjects. After repeated deliberations, a children’s martial arts set was designed and implemented. The pilot project analyzes the importance of sensory quality in the essential stage of children’s martial arts training. Results: After the experiment, there was a significant difference in balance quality in the experimental group (p<0.01). After the experiment, the difference in balance sense between the experimental and control groups was significant (p<0.01). Conclusion: In the basic training phase of children’s martial arts, coaches should require athletes to master basic martial arts skills more comprehensively. All movement combinations in martial arts require comprehensive physical fitness and balance sensitivity, including speed, flexibility, and coordination. El análisis académico de la influencia del entrenamiento de artes marciales en la salud física de los niños es de gran importancia para la promoción de la cultura tradicional china y puede enriquecer los programas de intervención deportiva para mejorar la salud física de los niños. Objetivo: Analizar el efecto del entrenamiento de la calidad sensorial en el entrenamiento del equilibrio en artes marciales en niños. Métodos: Se seleccionaron niños de 8 a 12 años como sujetos de la investigación. Tras repetidas deliberaciones, se diseñó y puso en marcha un conjunto de artes marciales para niños. El proyecto piloto analiza la importancia de la calidad sensorial en la etapa esencial del entrenamiento de artes marciales de los niños. Resultados: Después del experimento, hubo una diferencia significativa en la calidad del equilibrio en el grupo experimental (p<0,01). Tras el experimento, la diferencia en el sentido del equilibrio entre los grupos experimental y de control fue significativa (p<0,01). Conclusión: En la fase de formación básica de las artes marciales infantiles, los entrenadores deberían exigir a los deportistas un dominio más completo de las habilidades marciales básicas. Todas las combinaciones de movimientos en las artes marciales requieren una amplia aptitud física y sensibilidad al equilibrio, incluidas la velocidad, la flexibilidad y la coordinación. Abstract In this research, a total of 47 samples from five types of fermented soybeans were selected. The physicochemical parameters, biogenic amines (BAs) and bioactive compounds such as isoflavones, total phenolic (TPC) and γ-aminobutyric acid (GABA) were investigated. The results showed that the contents of isoflavones ranged from 598.06 mg/kg to 2725.03 mg/kg in all samples and it varied from 598.96 mg/kg to 2241.67 mg/kg in Semen Sojae Preparatum (SSP) which was higher than other types of fermented soybeans. The maximum variation of GABA content was found in Aspergillus-type douchi (AD) samples. The maximum value of TPC in SSP was 4.53 g/kg. For BAs, the indicator of spoilage, 4.26% of fermented soybeans samples may lead to histamine poisoning and 42.55% of samples with total BAs contents exceeding 100 mg/kg may be harmful to human body. It indicated that different types of fermented soybeans had various bioactive compounds and BAs, so different fermented soybeans could be selected according to bioactive substances. This finding could provide comprehensive information of health beneficial compounds and BAs risks of fermented soybeans. It could guide consumers to select healthy, nutritious and safety fermented soybeans.

ABSTRACT As a common problem in rice (Oryza sativa L.) production, lodging will not only increase harvest costs and reduce rice production, but also affect rice taste seriously and reduce its commercial value. There is still a lack of systematic research on the effect of lodging resistance of rice after returning wheat (Triticum aestivum L.) straw to the field. The study evaluated the effects of continuous straw returning and different fertilizer input levels on the lodging resistance of rice stalks based on years of experiments, using a split zone design with two factors including full return of straw to the field and straw removal, and three fertilization levels (conventional fertilization, 50% of conventional fertilization and no fertilization) under each factor, for a total of six treatments. Results suggested that under the same level of chemical fertilizer application, treatments of straw returning significantly increased stalk flexural resistance up to 33.74% as well as decreased the lodging index by a maximum of 33.86% compared with straw removal. Decrease of chemical fertilizer application increased significantly stalk flexural resistance, while decreased significantly lodging index. Rice plant height, center of gravity height, internode length, stem wall thickness and internode dry weight were main factors that influence rice lodging resistance. Compared with straw removal, straw returning combined with chemical fertilizers applying could significantly increase rice yield up to 19.62%. The increase in rice yield increased the risk of rice lodging. The continuous return of straw to the field combined with the application of chemical fertilizers reduced the risk of rice lodging to a certain extent, and meanwhile it achieved successive annual increase in rice production, which has a good promotion prospect.

Abstract Atrial fibrillation (AF) represents the most common type of sustained cardiac arrhythmia in humans and confers a significantly increased risk for thromboembolic stroke, congestive heart failure and premature death. Aggregating evidence emphasizes the predominant genetic defects underpinning AF and an increasing number of deleterious variations in more than 50 genes have been involved in the pathogenesis of AF. Nevertheless, the genetic basis underlying AF remains incompletely understood. In the current research, by whole-exome sequencing and Sanger sequencing analysis in a family with autosomal-dominant AF and congenital patent ductus arteriosus (PDA), a novel heterozygous variation in the PRRX1 gene encoding a homeobox transcription factor critical for cardiovascular development, NM_022716.4:c.373G>T;p.(Glu125*), was identified to be in co-segregation with AF and PDA in the whole family. The truncating variation was not detected in 306 unrelated healthy individuals employed as controls. Quantitative biological measurements with a reporter gene analysis system revealed that the Glu125*-mutant PRRX1 protein failed to transactivate its downstream target genes SHOX2 and ISL1, two genes that have been causally linked to AF. Conclusively, the present study firstly links PRRX1 loss-of-function variation to AF and PDA, suggesting that AF and PDA share a common abnormal developmental basis in a proportion of cases.

RESUMO: Com o rápido desenvolvimento da industrialização e da urbanização, o empreendedorismo voltando para cidade natal tem recebido cada vez mais atenção dos círculos sociais e acadêmicos. Nesse texto, adotando um método de análise estatística descritiva, analisamos a situação atual e o processo pelo qual os empreendedores do ecossistema de empreendedorismo voltando para cidade natal participam do desenvolvimento de oportunidades empreendedoras nas duas dimensões: aquisição de recursos e apoio institucional. E adotando o método de análise de rede social, exploramos o mecanismo de desenvolvimento de oportunidades de cada sujeito do ecossistema de empreendedorismo de retorno de cidade natal. Os resultados mostram que durante o desenvolvimento das oportunidades de empreendedorismo de retorno de cidade natal, todos os sujeitos do ecossistema de empreendedorismo de retorno de cidade natal participaram com seus próprios recursos e capacidades e promoveram conjuntamente o desenvolvimento de oportunidades. Durante o desenvolvimento dessas oportunidades, cada sujeito do ecossistema do empreendedorismo de retorno de cidade natal participou de diferentes formas e com diferentes ênfases. Este texto ajuda a enriquecer a pesquisa de integração da teoria do empreendedorismo de retorno de cidade natal e da teoria do ecossistema de inovação, e além disso fornece uma referência para realizar a revitalização rural e promover o desenvolvimento econômico regional.

ABSTRACT: With the rapid development of industrialization and urbanization, returning-home entrepreneurship has received increasing attention from social and academic circles. This paper adopted the method of descriptive statistics to analyze the current situation, and the process by which entrepreneurs in the returning-home entrepreneurship ecosystem participate in the development of entrepreneurial opportunities from two dimensions: resource acquisition and institutional support. This research adopted the method of social network analysis to explore the mechanism of opportunity development of each subject of the returning-home entrepreneurship ecosystem. The results showed that during the opportunity development of returning-home entrepreneurship, all the subjects in the returning-home entrepreneurship ecosystem participated with their own resources and capabilities and jointly promoted the development of opportunities. During the development of these opportunities, each subject in the returning-home entrepreneurship ecosystem participated in different ways and with different emphases. This paper enriched the integration research of returning-home entrepreneurship theory and innovation ecosystem theory, and provided a reference for realizing rural revitalization and promoting regional economic development.

Abstract To understand differential expression of genes (DEGs) based on transcriptome and transcription features of fatty acid desaturase genes (fads) in development of oil flax capsules. Three varieties of flax with high, medium and low linolenic acid (C18:3) content was investigated. The differential expressed genes (DEGs) of seed developing periods (15 d and 30 d after flowering, DAF) were compared with that of the flowering day based on RNA sequencing technology. DEGs related to fatty acid accumulation were distinguished and compared with Gene Ontolygy (GO) database and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway. In the synthesis of unsaturated fatty acids and α-linolenic acid metabolism, DEGs acted in multiple processes and signaling pathways. Lus_GLEAN_10046787, Lus_GLEAN_10024616, Lus_GLEAN_10024710, Lus_GLEAN_10046516, scaffold280_unigene_193 and scaffold272_unigene_19 participated in 23, 11, 12, 12, 12, 13 processes in GO description, respectively. And they acted in fatty acid degradation (ath00071), metabolism (ath01212), biosynthesis (ath00061) and elongation (ath00062) in the KEGG signaling pathway. The scaffold96_unigene_47 was differentially expressed between FR46 and FR48 with different linolenic acid contents. The DEGs can be utilized describe the structure and transcription expression features, which contribute to the selection and utilization of flax breeding lines.

Resumo: Governança significa a política e a estratégia adotadas no processo de gestão de um país. Na longa história de mais de dois mil anos, o confucionismo tornou-se o núcleo cultural e a doutrina política da autocracia imperial, e a teoria da “governança de todo o país” contida nele tornou-se uma categoria importante na cultura tradicional chinesa. Deve-se aprofundar na rica conotação do pensamento de governança confucionista e construir um sistema de governança nacional que se adapte à sociedade, à situação geral e ao futuro, por meio de desenvolvimento inovador e transformação criativa, de modo a permitir que a excelente cultura tradicional chinesa retorne à base, para abrir um novo caminho e fazer o passado servir ao presente, fornecendo, assim, importante orientação teórica e referência histórica para o grande rejuvenescimento da nação chinesa.

Abstract: Governance means the policy and strategy adopted in the process of running a country. In the long history of more than two thousand years, Confucianism had become the cultural core and political doctrine of the imperial autocracy, and the theory of “governance of the whole country” contained in it had become an important category in traditional Chinese culture. We should dig deep into the rich connotation of Confucian governance thought, and build a national governance system that suits society, the overall situation and future through innovative development and creative transformation, so as to let excellent traditional Chinese culture return to open a new road and make the past serve the present, thus providing important theoretical guidance and historical reference for the great rejuvenation of the Chinese nation.

Although metastasis is the major cause of death in cervical cancer, the mechanism of metastasis is still unclear. The mRNA expression and protein level of latent transforming growth factor beta binding protein 1 (LTBP1) were detected in tumor tissues and paracancerous tissues from in-house samples. Cell proliferation, cell cycle, migration, and in vivo metastasis were determined after LTBP1 was knocked down. Then, 13 drugs were screened, and the changes in cell apoptosis and proliferation and tumor metastasis were detected after drug treatment in shRNA cells. In our in-house samples, LTBP1 was lowly expressed in cervical cancer tissues. After LTBP1 knockdown, cell proliferation was increased, and the ability of in vitro migration and in vivo metastasis was enhanced. At the same time, the proportion of myeloid derived suppressor cells (MDSC) in situ increased, the proportion of T cells decreased, and transforming growth factor beta-1 (TGFβ1) signaling was activated. After carboplatin treatment, LTBP1 shRNA cell line apoptosis increased, metastasis in vivo was limited, and the proportion of MDSC in situ decreased. LTBP1 was lowly expressed in cervical cancer, and the inhibition of LTBP1 can improve the malignant degree of the tumor, and this process can be blocked by carboplatin.

Abstract Serum uric acid (UA) is a traditional biomarker in the clinical diagnosis of gout and hyperuricemia. However, serum treatment and storage are cumbersome, and wounds are susceptible to infection. Therefore, in this study, a simple and noninvasive method was developed to detect the UA in human saliva to monitor the gout. An Inertsil ODS-3 column was used for the analysis under the condition of isocratic elution with the mixed solution phosphate buffer (74 mM, pH=2.2): Methanol=98:2 (v:v) and the UV detection at 284 nm. Using salivary UA data from healthy volunteers (HVs) (n=68) and gout patients (GPs) (n=14), we examined the salivary UA difference in their content. The intra-and inter-day accuracy and precision (RSD %) were less than 2.56%, the limit of detection (LOD) of UA was 5.0 ng/mL, the mean recoveries of the corresponding compounds were 102.48%. Saliva levels of UA in HVs and GPs were 35.26±14.06 µg/mL and 91.96±23.90 µg/mL, respectively. The concentrations of salivary UA in GPs were significantly higher than those in HVs ( p < 0.001). This method was also expected to monitor the hyperuricemia and other metabolic disorders in the future.

ABSTRACT Background: The biomarker for left ventricular hypertrophy (LVH) in patients with essential hypertension (EH) remains an unmet clinical need. The microRNA-30 (miR-30) family has been associated with LVH in cellular and animal studies, but not in a clinical setting. Objective: The objective of the study was to investigate the clinical significance of circulating levels of miR-30 family as a biomarker for LVH in EH patients. Methods: A total of 239 EH patients and 239 healthy controls were enrolled in this study. Circulating levels of miR-30 family members, namely, miR-30a, miR-30b, miR-30c-1, miR-30c-2, miR-30d, and miR-30e, were evaluated using real-time polymerase chain reaction assays. Results: The circulating miR-30a, miR-30b, and miR-30e were significantly reduced in EH patients in contrast to controls. EH patients with LVH (EH-LVH) had substantially lower circulating miR-30b and miR-30e levels compared to EH patients without LVH (EH-nLVH). Moreover, the expression levels of miR-30b and miR-30e were positively related to LVMI, respectively. Receiver operating curve analysis showed that circulating miR-30e levels distinguished EH patients from controls, and EH-LVH from EH-nLVH patients. Logistic regression analysis identified the circulating miR-30e as a risk factor for LVH in EH patients. Conclusion: Circulating miR-30e level can be used as a biomarker in distinguishing EH-LVH from EH-nLVH. A further prospective study is warranted to validate this finding.

Severe blockage in myeloid differentiation is the hallmark of acute myeloid leukemia (AML). Trdmt1 plays an important role in hematopoiesis. However, little is known about the function of Trdmt1 in AML cell differentiation. In the present study, Trdmt1 was up-regulated and miR-181a was down-regulated significantly during human leukemia HL-60 cell differentiation after TAT-CT3 fusion protein treatment. Accordingly, miR-181a overexpression in HL-60 cells inhibited granulocytic maturation. In addition, our “rescue” assay demonstrated that Trdmt1 3′-untranslated region promoted myeloid differentiation of HL-60 cells by sequestering miR-181a and up-regulating C/EBPα (a critical factor for normal myelopoiesis) via its competing endogenous RNA (ceRNA) activity on miR-181a. These findings revealed an unrecognized role of Trdmt1 as a potential ceRNA for therapeutic targets in AML.

Abstract Rapeseed is one of important oil crops in China. Better understanding of the regulation network of main agronomic traits of rapeseed could improve the yielding of rapeseed. In this study, we obtained an influrescence mutant that showed a fusion phenotype, similar with the Arabidopsis clavata-like phenotype, so we named the mutant as Bnclavata-like (Bnclv-like). Phenotype analysis illustrated that abnormal development of the inflorescence meristem (IM) led to the fused-inflorescence phenotype. At the stage of protein abundance, major regulators in metabolic processes, ROS metabolism, and cytoskeleton formation were seen to be altered in this mutant. These results not only revealed the relationship between biological processes and inflorescence meristem development, but also suggest bioengineering strategies for the improved breeding and production of Brassica napus.

The melamine and cyanuric acid (CA) complex has been suggested to cause the toxic effects observed in melamine-contaminated food or milk. However, the cytotoxic and genotoxic effects of co-exposure to melamine and CA are not fully clear. Therefore, the cytotoxic effects of melamine and CA were first examined by co‐exposure in human kidney 293 cells using the MTT assay. During a 24-h period for the three concentrations tested (0.5, 1, and 5 mg/mL), neither melamine nor CA alone showed significant toxic effects on 293 cells at 0.5 mg/mL, while higher concentrations led to decreased in cell viability. However, co-exposure to several combinations of melamine and CA [100:1, 10:1, 1:10, and 1:100 (v:v), at a final concentration of 0.5 mg/mL] did cause cytotoxicity with higher levels of CA leading to higher cytotoxicity. By contrast, while neither melamine nor CA alone induced phosphorylated-H2AX (γH2AX) foci formation, melamine and CA at a 100:1 ratio induced γH2AX foci 24 h post-treatment. The alkaline comet assay also revealed the presence of DNA damage following melamine and CA co-exposure. In vivo assay also revealed the presence of melamine-CA complex in the kidney. These data indicated that the cytotoxic and genotoxic effects of melamine and CA co-exposure differ from those of melamine or CA alone.
