As an important component of solid tumors, mast cells show specific phenotypes in various tumor microenvironments. However, the precise mechanism of mast cell accumulation and the phenotypic features of thyroid cancer (TC) remain largely unknown. Here, we found that mast cells were obviously recruited to tumor tissue by TC-derived stem cell factor (SCF). With tumor progression, mast cell levels increased gradually. In addition, intratumoral mast cells expressed higher levels of the immunosuppressive molecule galectin-9, which effectively suppresses CD8+ T-cell antitumor immunity in vitro. Blocking galectin-9 on tumor-infiltrating mast cells reversed the immunosuppression of CD8+ T cells. In conclusion, our data elucidated novel protumorigenic and immunosuppressive roles of mast cells in TC. In addition, our results indicated that blocking mast cells may impede tumor progression and ameliorate the prognosis of TC patients.

ABSTRACT A closed-form analytical solution to rheological phenomena is obtained based on Hoek-Brown (HB) yield criterion and the generalized Bingham model. Once the circular tunnel is excavated, an initial plastic region is formed. When the total stresses fulfil the yield condition, the initial plastic region steps into the viscoplastic region, in which the displacement develops with time. The one-dimensional constitutive equation of the generalized Bingham model is transformed into a three-dimensional expression in polar coordinates by combining the plane strain condition. With the help of equilibrium differential equation, geometric equation and boundary condition, the stress and displacement in the initial plastic region can be expressed. At the moment of t, the stresses and displacements are described similar to the solutions of the elasto-plastic state. The stresses, strains and displacements after transformation can be expressed in the viscoplastic region. The proposed solution was verified by the closed-form solution of circular tunnels in elastic-brittle-plastic rock mass.

The aim of the current study was to explore the correlation between physical properties of wet masses and pellet quality by using powdered herbal slices as model drugs. Wet masses with 100 formulations were prepared by taking 20 kinds of powdered herbal slices as model drugs, microcrystalline cellulose as pelletization aid and five levels of added water as liquid binder. Physical properties of the wet masses such as hardness, adhesiveness, springiness, cohesiveness, chewiness, and resilience were measured by a texture analyzer. Meanwhile, the moisture retention capacities (MRC) of powdered herbal slices and wet masses were determined. Particles were classified after they were produced during spheronization. Principal component analysis, factor analysis and classification analysis were performed on the data. Wet masses could be classified into three groups by taking Ha as the first classification index and Ha/Sp as the second classification index. The correct rate of the classification was 91.00%. If Ha value of wet masses was greater than 15610 g, pellets of type ① would form, otherwise, pellets of type ② or type ③ would form based on Ha/Sp value. Then a classification plot of wet masses was developed to predict pellet formation of powdered herbal slices. Meanwhile, the probable mechanism of pellets formation during spheronisation was concluded in this study, which provided useful information to improve pellet quality.

RESUMO O uso de engenharia assistida por computador (CAE) na fase de projeto do conceito do automóvel tornou-se um ponto de acesso na pesquisa de fatores humanos. Com base no software computacional biomecânico musculoesquelético humano, foi construído um modelo musculoesquelético sentado para descrever a postura sentada natural de um condutor. A interação entre um motorista e um carro em várias combinações de ângulos de inclinação do assento-pan/encosto foi analisada usando uma abordagem dinâmica do verso. A fim de descobrir a postura de condução “mais confortável” do assento-pan/encosto, o efeito dos ângulos de inclinação do assento-pan/dorso sobre o grau de atividade muscular e a força de compressão intradiscal L4-L5 foi investigado. Os resultados mostraram que um ângulo de inclinação para trás muito maior, aproximadamente 15°, e um ligeiro assento-pan para trás, cerca de 7°, pode aliviar a fadiga muscular e levar a dirigir em uma posição confortável. Posteriormente, de acordo com as conclusões acima expostas, foi construída uma função preliminar de conforto ao dirigir.

SUMMARY Using computer-aided engineering (CAE) in the concept design stage of automobiles has become a hotspot in human factor engineering research. Based on human musculoskeletal biomechanical computational software, a seated human-body musculoskeletal model was built to describe the natural sitting posture of a driver. The interaction between the driver and car in various combinations of seat-pan/back-rest inclination angles was analyzed using an inverse-dynamics approach. In order to find out the “most comfortable” driving posture of the seat-pan/back-rest, the effect of seat-pan/back-rest inclination angles on the muscle activity degree, and the intradiscal L4-L5 compression force were investigated. The results showed that a much larger back-rest inclination angle, approximately 15°, and a slight backward seat-pan, about 7°, may relieve muscle fatigue and provide more comfort while driving. Subsequently, according to the findings above, a preliminary driving-comfort function was constructed.

OBJECTIVES: Previous studies have not shown any correlation between bile acid metabolism and bone mineral density (BMD) in women with postmenopausal osteoporosis. Thus, the current study evaluated the association between bile acid levels as well as BMD and bone turnover marker levels in this group of women. METHODS: This single-center cross-sectional study included 150 postmenopausal Chinese women. According to BMD, the participants were divided into three groups: osteoporosis group, osteopenia group, and healthy control group. Serum bile acid, fibroblast growth factor 19 (FGF19), and bone turnover biomarker levels were assessed. Moreover, the concentrations of parathyroid hormone, 25-hydroxy vitamin D [25(OH)D], procollagen type I N-peptide (P1NP), and beta-CrossLaps of type I collagen containing cross-linked C-terminal telopeptide (β-CTX) were evaluated. The BMD of the lumbar spine and proximal femur were examined via dual-energy X-ray absorptiometry. RESULTS: The serum total bile acid levels in the osteoporosis and osteopenia groups (5.28±1.56 and 5.31±1.56 umol/L, respectively) were significantly lower than that in the healthy control group (6.33±2.04 umol/L; p=0.002 and 0.018, respectively). Serum bile acid level was positively associated with the BMD of the lumbar spine, femoral neck, and total hip. However, it negatively correlated with β-CTX concentration. Moreover, no correlation was observed between bile acid and P1NP levels, and the levels of the other biomarkers that were measured did not differ between the groups. CONCLUSION: Serum bile acid was positively correlated with BMD and negatively correlated with bone turnover biomarkers reflecting bone absorption in postmenopausal women. Thus, bile acid may play an important role in bone metabolism.

Resumo Fundamentos: A antraciclina gera uma disfunção ventricular esquerda progressiva associada a um prognóstico ruim. Objetivos: O propósito deste estudo foi avaliar se a análise layer específico de strain poderia avaliar disfunção ventricular esquerda subclínica após exposição a antraciclina. Métodos: Foram inscritos quarenta e dois sobreviventes tratados com antraciclina por linfoma não Hodgkin de células B grandes, de 55,83 ± 17,92 anos (grupo de quimioterapia) e 27 voluntários saudáveis, de 51,39 ± 13,40 anos (grupo controle). A dose cumulativa de epirrubicina no grupo de quimioterapia foi de 319,67 ± 71,71 mg/m2. O tempo desde a última dose de epirrubicina até o exame ecocardiográfico foi de 52,92 ± 22,32 meses. Analisaram-se o strain longitudinal global (GLS), o circunferencial (GCS) e o strain radial (GRS), os valores das camadas subendocárdica, média e subepicárdica so strain longitudinal (LS-ENDO, LS-MID, LS-EPI) e do strain circunferencial (CS-ENDO, CS-MID, CS-EPI). O gradiente de strain transmural foi calculado como a diferença no strain sistólico pico entre as camadas subendocárdicas e subepicárdicas. Um valor de p < 0,05 foi considerado significativo. Resultados: Os parâmetros convencionais da função sistólica e diastólica não mostraram diferenças significativas entre dois grupos. Comparados aos controles, os pacientes apresentaram GCS e GLS significativamente menores. A análise de speckle tracking multi-layer mostrou uma redução significativa no strain circunferencial da camada subendocárdica, o gradiente transmural CS e o strain longitudinal das três camadas. Em contraste, os dois grupos não diferiram no gradiente de strain longitudinal transmural e de strain radiais. Conclusões: Provou-se a deterioração preferencial do strain subendocárdico em sobreviventes de longa duração após exposição à antraciclina. O ecocardiograma de speckle tracking multi-layer pode facilitar o acompanhamento longitudinal dessa coorte de pacientes em risco. (Arq Bras Cardiol. 2018; 110(3):219-228)

Abstract Background: Anthracycline generates progressive left ventricular dysfunction associated with a poor prognosis. Objectives: The purpose of this study was to evaluate whether layer-specific strain analysis could assess the subclinical left ventricular dysfunction after exposure to anthracycline. Methods: Forty-two anthracycline-treated survivors of large B-cell non-Hodgkin lymphoma, aged 55.83 ± 17.92 years (chemotherapy group) and 27 healthy volunteers, aged 51.39 ± 13.40 years (control group) were enrolled. The cumulative dose of epirubicin in chemotherapy group was 319.67 ± 71.71mg/m2. The time from last dose of epirubicin to the echocardiographic examination was 52.92 ± 22.32 months. Global longitudinal (GLS), circumferential (GCS) and radial strain (GRS), subendocardial, mid and subepicardial layer of longitudinal (LS-ENDO, LS-MID, LS-EPI) and circumferential strain (CS-ENDO, CS-MID, CS-EPI) values were analyzed. Transmural strain gradient was calculated as differences in peak systolic strain between the subendocardial and subepicardial layers. A value of p < 0.05 was considered significant. Results: Conventional parameters of systolic and diastolic function showed no significant difference between two groups. Compared with controls, patients had significantly lower GCS and GLS. Multi-layer speckle tracking analysis showed significant reduction of circumferential strain of subendocardial layer, transmural CS gradient and longitudinal strain of all three layers. In contrast, the two groups did not differ in transmural longitudinal strain gradient and radial strains. Conclusions: It proved the preferential impairment of subendocardial deformation in long-term survivors after exposure to anthracycline. Multi-layer speckle tracking echocardiography might facilitate the longitudinal follow-up of this at-risk patient cohort.

ABSTRACT We compared the morphology of the erythrocytes of five anurans, two toad species - Bufo gargarizans (Cantor, 1842) and Duttaphrynus melanostictus (Schneider, 1799) and three frog species - Fejervarya limnocharis (Gravenhorst, 1829), Microhyla ornata (Duméril & Bibron, 1841), and Rana zhenhaiensis (Ye, Fei & Matsui, 1995). We then reconstructed the ancestral state of erythrocyte size (ES) and nuclear size (NS) in amphibians based on a molecular tree. Nine morphological traits of erythrocytes were all significantly different among the five species. The results of principal component analysis showed that the first component (49.1% of variance explained) had a high positive loading for erythrocyte length, nuclear length, NS and ratio of erythrocyte length/erythrocyte width; the second axis (28.5% of variance explained) mainly represented erythrocyte width and ES. Phylogenetic generalized least squares analysis showed that the relationship between NS and ES was not affected by phylogenetic relationships although there was a significant linear relationship between these two variables. These results suggested that (1) the nine morphological traits of erythrocytes in the five anuran species were species-specific; (2) in amphibians, larger erythrocytes generally had larger nuclei.

Boron isotopes in teeth has been a new proxy for dietary reconstructions and its resistance to diagenetic alteration. In this study a method using inductively coupled plasma source mass spectrometry (ICP-MS) for the measurement of boron isotope ratio in human dental enamel has been developed. Human dental enamel were digested with HNO3-H2O2 in a microwave system. Boron in solution was separated from the matrix components using Amberlite IRA-743 resin. The factors that may affect precision and accuracy in isotope ratio determination by ICP-MS, including memory effects, mass bias drift, and concentration effects, were investigated to obtain optimum conditions. Then, the 10B/11B ratios in teeth were measured. The results showed that 2% of HNO3 + 2% of NH3•H2O, selected as the diluent/rinse solution could be effective in the elimination of boron memory effect. There was no concentration effect on boron isotope ratios when the ratio of samples B concentration to standard B concentration (refers to Csample/Cstd) varied from 0.5 to 2. The result of 10B/11B ratios in tooth enamel by sex and age fluctuated over a broad range, ranged from 0.2007 to 0.2574. This method is expected to be used for boron isotope ratio analyses in archeometry, forensic identification, paleoecology, and other disciplines in the future.

Particular climate conditions in a low-latitude and high-altitude region endow grape berries with distinctive quality characteristics. So far, few reports have been concerned with the formation of berry flavour in such a region. This study aimed to investigate the accumulation pattern of flavonoids in Vitis vinifera L. cv. Cabernet Sauvignon grape berries growing at different altitudes of the highland in southwest China in two consecutive vintages. In addition to the 3-0-monoglucosides and 3-0-acyl monoglucosides of the five main anthocyanidins (delphinidin, cyanidin, peonidin, petunidin and malvidin), some uncommon anthocyanins, such as three diglucosides of anthocyanidins and pelargonidin-3-O-glucoside, were detected in the grape berries. Higher altitude cultivation greatly promoted the production of anthocyanins and flavonols, particularly cyanidin-type anthocyanins and quercetin-type flavonols from the F3'H branch of the flavonoid biosynthetic pathway. Flavan-3-ols from both branches were comparatively less influenced by vineyard altitude. Vintage in this high-altitude region also had a dramatic influence on the accumulation of flavonoids. Most of the anthocyanin and flavonol components were affected more by vineyard altitude than by vintage, whereas the accumulation of flavan-3-ols differed mainly between vintages. The present data will not only improve the understanding of flavonoid accumulation in grapes from a high-altitude region with different climates, but also provide practical guidance for the production of high-quality grapes and wine.

Um método empregando "bulk liquid membrane" (BLM) e o complexo ácido di(2-etilhexil) fosfórico (D2EHPA) e ácido O,O'-dibenzoil-(2R, 3R)-tartárico (L-(-)-DBTA) foi desenvolvido para a separação por extração do ácido mandélico racêmico (rac MA). Os efeitos de tempo de resolução, concentração de D2EHPA na fase orgânica, concentração de rac MA na fase doadora, velocidade de agitação e temperatura sobre a separação por extração foram estudados. Os resultados experimentais evidenciaram que o maior fator de separação (α) 2,74 e o valor do excesso enantiomérico (ee, %) 46,47% foram obtidos quando o tempo de resolução foi de aproximadamente 4 h, a razão de concentração molar de rac MA, D2EHPA e L-(-)-DBTA foi 0,05:0,2:0,3, a velocidade de agitação, 400 r min-1 e a temperatura, 35 ºC. O método desenvolvido é útil para a otimização do transporte de sistemas BLM e para a produção em grande escala de enantiômero puro.

A method of bulk liquid membrane (BLM) using complex of di(2-ethylhexyl) phosphoric acid (D2EHPA) and O,O'-dibenzoyl-(2R, 3R)-tartaric acid (L-(-)-DBTA) was developed for the extractive resolution of racemic mandelic acid (rac MA). The effects of resolution time, concentration of D2EHPA in the organic phase, concentration of rac MA in the feed phase, stirring speed and temperature on extractive resolution were studied. The experimental results have shown that the greatest separation factor (α) 2.74 and the enantiomeric excess value (ee, %) 46.47% were obtained when the resolution time was about 4 h, the molar concentration ratio of rac MA, D2EHPA and L-(-)-DBTA 0.05:0.2:0.3, the stirring speed 400 r min-1 and the temperature 35 ºC. The developed method is helpful for optimizing the transport of BLM systems and for large-scale production of pure enantiomer.
