Resumo Gardenia jasminoides Ellis é um fitoterápico chinês com valor medicinal e econômico, mas seu mecanismo de resposta ao estresse hídrico permanece obscuro. Neste estudo, o “método de potes duplos” foi usado para simular o estresse hídrico de G. jasminoides Ellis e explorar seu mecanismo de resposta fisiológica e transcriptômica. Não encontramos danos significativos aos lipídios da membrana de G. jasminoides Ellis durante o estresse. POD desempenhou um papel antioxidante vital, o enriquecimento KEGG mostrou que metabólitos secundários, como flavonoides, também podem desempenhar um papel antioxidante e PRO desempenhou um ajuste osmótico significativo. Os hormônios endógenos regulam o crescimento e o desenvolvimento de G. jasminoides Ellis e desempenham um papel na transdução de sinal. Entre eles, o estresse hídrico leve é retardado. Ao mesmo tempo, havia 19631, 23693 e 15045 genes diferencialmente expressos de G. jasminoides Ellis nos dias 5, 10 e 15 sob estresse hídrico. Esses genes foram intimamente associados ao proteassoma, endopeptidase, ribossomo, transdução de sinal MAPK e transdução de sinal de hormônio endógeno, interação planta-patógeno e biossíntese de fenilpropanoides e outras vias fisiológicas e metabólicas, que regulam o turnover e transporte de proteínas, o reforço e adesão das paredes celulares, a indução do fechamento estomático, reações alérgicas, reações de defesa, movimentos foliares, entre outros. Também podem absorver os raios ultravioleta para reduzir a geração de radicais livres de oxigênio, alterar a forma de utilização da energia e ajustar a pressão osmótica das células vegetais.

Abstract Gardenia jasminoides Ellis is a Chinese herbal medicine with medicinal and economic value, but its mechanism of response to waterlogging stress remains unclear. In this study, the “double pots method” was used to simulate the waterlogging stress of Gardenia jasminoides Ellis to explore its physiological and transcriptomic response mechanism. We found no significant damage to Gardenia jasminoides Ellis membrane lipid during stress. POD played a vital antioxidant role, KEGG enrichment showed that secondary metabolites such as flavonoids might also play an antioxidant role, and PRO played a significant osmotic adjustment. Endogenous hormones regulate the Gardenia jasminoides Ellis's growth and development and play a role in signal transduction. Among them, light waterlogging stress is delayed. At the same time, there were 19631, 23693, and 15045 differentially expressed genes on the 5th, 10d, and 15d of Gardenia jasminoides Ellis under waterlogging stress. These genes were closely associated with the proteasome, endopeptidase, ribosome, MAPK signal transduction, and endogenous hormone signal transduction, plant-pathogen interaction and phenylpropanoid biosynthesis and other physiological and metabolic pathways, which regulate the turnover and transportation of protein, the reinforcement and adhesion of cell walls, the induction of stomatal closure, allergic reactions, defense reactions, leaf movements and others. It also can absorb ultraviolet rays to reduce the generation of oxygen free radicals, change the way of energy utilization and adjust the osmotic pressure of plant cells.

ABSTRACT Purpose: This study aimed to report an experiment designed to determine anatomical changes in porcine corneas following placement of a novel polymer implant into the cornea. Methods: An ex vivo porcine eye model was used. A novel type I collagen-based vitrigel implant (6 mm in diameter) was shaped with an excimer laser on the posterior surface to create three planoconcave shapes. Implants were inserted into a manually dissected stromal pocket at a depth of approximately 200 μm. Three treatment groups were defined: group A (n=3), maximal ablation depth 70 μm; Group B (n=3), maximal ablation depth 64 μm; and group C (n=3), maximal ablation depth 104 μm, with a central hole. A control group (D, n=3) was included, in which a stromal pocket was created but biomaterial was not inserted. Eyes were evaluated by optical coherence tomography (OCT) and corneal tomography. Results: Corneal tomography showed a trend for a decreased mean keratometry in all four groups. Optical coherence tomography showed corneas with implants placed within the anterior stroma and visible flattening, whereas the corneas in the control group did not qualitatively change shape. Conclusions: The novel planoconcave biomaterial implant described herein could reshape the cornea in an ex vivo model, resulting in the flattening of the cornea. Further studies are needed using in vivo animal models to confirm such findings.

RESUMO Objetivo: Relatar um experimento projetado para determinar alterações anatômicas em córneas porcinas após a colocação de um novo implante de polímero na córnea. Métodos: Foi utilizado olho de porco ex vivo. Um novo agente modelador biocompatível, de colágeno tipo 1, com 6mm de diâmetro foi moldado com excimer laser em sua face posterior, para criar três formatos planocôncavos. Os implantes foram inseridos dentro de um bolsão, dissecado manualmente, a 200 micrômetros (μm). Foram definidos três grupos de tratamento: grupo A (n=3), teve a profundidade máxima de ablação de 70 μm; o grupo B (n=3), profundidade máxima de ablação de 64 μm; e o grupo C (n=3), profundidade máxima de ablação de 104 μm, com buraco central. O grupo controle, D (n=3), foi incluído, com a criação do bolsão estromal, porém sem inserir o material. A avaliação desses olhos foi realizada por tomografia de coerência óptica (OCT) e por tomografia corneana. Resultados: A tomografia corneana mostrou uma tendência para diminuição da ceratometria média em todos os 4 grupos. A tomografia de coerência óptica mostrou córneas com implantes localizados no estroma anterior e aplanamento visível, enquanto as córneas não mudaram qualitativamente o formato no grupo controle. Conclusões: O novo implante de biomaterial planocôncavo descrito aqui foi capaz de remodelar a córnea em modelo de animal ex vivo, resultando no aplanamento corneano. Novos estudos são necessários usando modelos animais in vivo para confirmar tais achados.

Resumo Os medicamentos combinados de β-lactâmicos / inibidores da lactamase (BLBLIs) são considerados uma alternativa eficaz aos carbapenêmicos. No entanto, existe uma preocupação crescente de que o aumento do uso de BLBLIs pode levar ao aumento da resistência. Este estudo determinou a associação temporal entre o consumo de BLBLI e a resistência antimicrobiana em bactérias gram-negativas. Neste estudo retrospectivo, os dados eletrônicos sobre as bactérias gram-negativas isoladas, incluindo A. baumannii, P. aeruginosa, E. coli e K. pneumoniae de pacientes internados e os resultados dos testes de suscetibilidade foram recuperados dos registros médicos do laboratório clínico. Uma regressão linear e análise de correlação cruzada foram realizadas nos dados adquiridos. Foram observadas tendências crescentes (p < 0,05) no consumo de BIBLI e carbapenem com uma mediana de uso de 27,68 e 34,46 DDD/1000 PD por trimestre, respectivamente. Foi observada uma tendência de diminuição (p = 0,023) no consumo de fluoroquinolonas com uma mediana de uso de 29,13 DDD/1000 PD por trimestre. A taxa de resistência de K. pneumoniae foi sincronizada com os consumos de BIBLI e carbapenem com coeficiente de correlação de 0,893 (p = 0,012) e 0,951 (p = 0,016), respectivamente. A análise de correlação cruzada contra o consumo de BIBLI e K. pneumoniae resistente ao meropenem atingiu o pico no intervalo de 0 quarto (r = 951, p = 0,016). Houve uma tendência de aumento no consumo de BLBLI e carbapenêmicos. A tendência crescente nas taxas de resistência a piperacilina/tazobactam, em linha com o consumo crescente de BLBLI, sugere que BLBLI deve ser usado com cautela e não pode ser considerado diretamente como alternativa de longo prazo aos carbapenêmicos.

Abstract The β-lactam/lactamase inhibitors (BLBLIs) combination drugs are considered an effective alternative to carbapenems. However, there is a growing concern that the increased use of BLBLIs may lead to increased resistance. This study determined the temporal association between the consumption of BLBLI and the antimicrobial resistance in Gram-negative bacteria. In this retrospective study, electronic data on the Gram-negative bacterial isolates, including A. baumannii, P. aeruginosa, E. coli, and K. pneumoniae from in-patients and susceptibility testing results were retrieved from the medical records of the clinical laboratory. A linear regression and cross-correlation analysis were performed on the acquired data. Increasing trends (p<0.05) in the consumption of BIBLI and carbapenem with a median use of 27.68 and 34.46 DDD/1000 PD per quarter were observed, respectively. A decreased trend (p=0.023) in the consumption of fluoroquinolones with a median use of 29.13 DDD/1000 PD per quarter was observed. The resistance rate of K. pneumoniae was synchronized with the BIBLI and carbapenem consumptions with a correlation coefficient of 0.893 (p=0.012) and 0.951 (p=0.016), respectively. The cross-correlation analysis against the consumption of BIBLI and meropenem resistant K. pneumoniae was peaked at 0-quarter lag (r=951, p=0.016). There was an increasing trend in the consumption of BLBLI and carbapenems. The increasing trend in the rates of resistance to piperacillin/tazobactam, in line with the increasing consumption of BLBLI, suggests that BLBLI has to be used with caution and cannot be directly considered as a long-term alternative to carbapenems.

Abstract Comparsion of the effects of the chilling (4 ºC), superchilling (-2 ºC) and frozen (-18 ºC) storage on microbial spoilage and bacterial community structure of fresh beef was evaluated by high-throughput sequencing technology in this study. The results indicated that storage temperature can significantly affect the degree of microbial spoilage and bacterial community structure of fresh beef during storage. At the species level, the primary dominant bacterial species in beef samples was Pseudomonas fragi with relative abundance of 37.78% followed by Myroides phaeus and Brochothrix thermosphacta with 10.95% and 4.64%, respectively, at the beginning of storage. Both superchilling and frozen storage can effectively inhibit the growth of P. fragi, while the chilling storage failed to demonstrate strong inhibition for growth of P. fragi. Similar result was found for Myroides phaeus. The chilling, superchilling and frozen storage can not effectively inhibit the growth of Brochothrix thermosphacta and Acinetobacter johnsonii. The difference of microbial spoilage in beef stored in three kinds of storage may be related to the difference of the bacterial community structure and the formation of dominant spoilage bacteria. These results revealed that superchilling and frozen storage can effectively inhibit the formation of dominant spoilage bacteria, resulting in a good freshness preservation.

In South Africa, the family Heliozelidae in the order Lepidoptera is restricted to four known species. The grapevine leaf miner, Holocacista capensis, feeds between the epidermal layers of a grapevine leaf, predominantly along the leaf margin. A final instar larva will descend from the blotch mine/gallery to attach its cocoon casing (constructed from the epidermal layers of the mined gallery) to any object below the infested leaf. Five monophyletic clades and a polyphyletic group have been identified within the Heliozelidae, using a mitochondrially encoded gene cytochrome c oxidase I (COI) and a nuclear gene, histone 3 (H3). An exploratory study of the genetic diversity within H. capensis populations was conducted using these genes. The phylogenetic analyses of COI indicate that H. capensis that are currently being collected from South Africa fall within three clades/haplotypes, of which one is well supported and contains only one species from Gauteng, and one has three specimens from two different areas in the Western Cape province, while 80% belong to haplotype 1 (H1). The current study can be used as a starting point for future DNA-based studies aimed at gaining insight into possible patterns of diversity in H. capensis to confirm switching from native to commercial grapevine hosts. However, more samples need to be collected from different areas in South Africa.

Abstract Powders were prepared from fresh Phyllostachys pubescens bamboo shoots (P. pubescens), Dendrocalamopsis oldhami bamboo shoots (D.oldhami) and Dendrocalamus latiflorus bamboo shoots (D.latiflorus), and their physical, chemical, adsorption and powder properties were determined. All three of these powders were high in dietary fiber and protein, with P. pubescens powder containing the highest concentration of dietary fiber (50.34 ± 0.65 g∙100 g-1). The water holding, oil holding, and swelling capacities of this powder were all higher than those of the other two kinds of powders. The P. pubescens powder also showed the highest capacity to adsorb glucose, cholesterol and nitrite. In addition, the measured repose and slip angles of this powder were lower than those of the other two powders, indicative of it having good fluidity. And the bulk density and tap density of this powder were significantly higher than those of the others as well, indicative of its superior filling property.

ABSTRACT The mixture of mesosulfuron-methyl + iodosulfuron-methyl-sodium (ME+IMS) as post-emergence dual-purpose herbicide exhibits phytotoxicity in foxtail millet (Setaria italica (L.) P. Beauv.) Reducing ME+IMS stress in foxtail millet is therefore of practical significance. This study assessed the effects of seed pre-soaking with different concentrations of brassinolide (0.01, 0.1, 1.0 and 2.0 mg L-1) on foxtail millet grown under ME+IMS (2.81+0.56 g ai ha-1) stress. The agronomic characteristics, photosynthetic pigment, gas exchange parameters, chlorophyll fluorescence and P700 parameters of two foxtail millet cultivars (‘Jingu 21’ and ‘Zhangza 5’) were analyzed 7 d after ME+IMS treatment at the 5th leaf stage. Compared with ME+IMS treatment alone, brassinolide treatment within a concentration range of 0.1-1.0 mg L-1 had a positive effect on all plant parameters to varying degrees. Pre-soaking of seeds with 0.1 mg L-1 brassinolide increased the leaf area (41.8%), fresh mass (179.2%), net photosynthetic rate (Pn, 38.4%), transpiration rate (Tr, 61.5%), stomatal conductance (gs, 103.1%), photosystem II maximum quantum yield (Fv/Fm, 8.6%), effective quantum yield (Y(II), 16.0%), electron transport rate (ETR(II), 17.8%), photosystem I photochemical quantum yield (Y(I), 16.7%) and electron transport rate (ETR(I), 38.7%) in ‘Zhangza 5’ leaves under ME+IMS stress. An increase in plant height (53.0%), total chlorophyll (38.3%), carotenoid contents (40.0%), and maximal P700 change (Pm, 34.3%) were also observed following 1.0 mg L-1 brassinolide treatment compared with ME+IMS in ‘Jingu 21’. Comprehensive analysis showed that pre-sowing seed treatment with 0.1-1.0 mg L-1 brassinolide could enhance the growth and photosynthetic resistance of foxtail millet seedlings to ME+IMS stress.

RESUMO Porcos tibetanos são caracterizados por diferenças fenotípicas significativas em relação aos porcos de planície. Nosso estudo anterior demonstrou que os genes CRYAB e CTGF eram expressos diferentemente nos tecidos do coração entre os porcos tibetanos (raça das terras altas) e Yorkshire (raça das terras baixas), indicando que eles poderiam participar da adaptação à hipoxia. CRYAB (ɑB-crystallin) e CTGF (fator de crescimento do tecido conjuntivo) também foram relatados como estando associados ao desenvolvimento pulmonar. Entretanto, os padrões de expressão do CRYAB e CTGF nos tecidos pulmonares em diferentes altitudes e sua caracterização genética não são bem compreendidos. Neste estudo, o qRT-PCR e a mancha ocidental de tecido pulmonar revelou níveis de expressão de CRYAB mais elevados em porcos tibetanos e Yorkshire de altitude e média altitude do que em seus pares de planície. Com um aumento na altitude, o nível de expressão do CTGF aumentou nos porcos tibetanos, enquanto diminuiu nos porcos Yorkshire. Além disso, foram identificados dois novos polimorfismos de um único nucleotídeo na região flanqueadora de CRYAB (g.39644482C>T e g.39644132T>C) e CTGF (g.31671748A>G e g.31671773T>G). O polimorfismo pode contribuir parcialmente com as diferenças nos níveis de expressão entre grupos a uma mesma altitude. Estas descobertas proporcionam novos conhecimentos sobre as adaptações de hipoxia a alta altitude dos porcos tibetanos.

Our previous study demonstrated that the genes CRYAB and CTGF were differentially expressed in heart tissues between Tibetan (highland breed) and Yorkshire (lowland breed) pigs, indicating that they might participate in hypoxia adaptation. CRYAB (ɑB-crystallin) and CTGF (connective tissue growth factor) have also been reported to be associated with lung development. However, the expression patterns of CRYAB and CTGF in lung tissues at different altitudes and their genetic characterization are not well understood. In this study, qRT-PCR and western blot of lung tissue revealed higher CRYAB expression levels in highland and middle-highland Tibetan and Yorkshire pigs than in their lowland counterparts. With an increase in altitude, the expression level of CTGF increased in Tibetan pigs, whereas it decreased in Yorkshire pigs. Furthermore, two novel single-nucleotide polymorphism were identified in the 5′ flanking region of CRYAB (g.39644482C>T and g.39644132T>C) and CTGF (g.31671748A>G and g.31671773T>G). The polymorphism may partially contribute to the differences in expression levels between groups at the same altitude. These findings provide novel insights into the high-altitude hypoxia adaptations of Tibetan pigs.

Abstract Peucedanum praeruptorum Dunn is a natural plant with expectorant, antitussive, anti-asthmatic, anti-inflammatory, antispasmodic and sedative effects. It is used in traditional Chinese medicine. The key active substances extracted after separation are also used to process functional food. Recent advances in the development of Peucedanum praeruptorum Dunn industrially have increased the market demand. In particular, the study of the physiological activity of P. praeruptorum Dunn has always been a research hotspot; however, limited data exist currently. This study reviews the research developments associated with the physiological activities of P. praeruptorum Dunn in terms of cardiovascular, expectorant and antitussive, antitumor, and anti-inflammatory effects. The results serve as a standard of reference for further investigation into the pharmacologic role of Peucedanum praeruptorum Dunn.

Resumo Fundamento A associação entre o status de saúde cardiovascular ideal ( ideal cardiovascular health ( ICVH) e diagnóstico de fibrilação ou flutter atrial (FFA) foi menos estudado em comparação a outras doenças cardiovasculares. Objetivos Analisar a associação entre o diagnóstico de FFA e métricas e escores de ICVH no Estudo Longitudinal de Saúde do Adulto (ELSA-Brasil). Métodos Este estudo analisou dados de 13141 participantes com dados completos. Os traçados eletrocardiográficos foram codificados de acordo com o Sistema de Minnesota, em um centro de leitura centralizado. As métricas do ICVH (dieta, atividade física, índice de massa corporal, tabagismo, glicemia de jeju, e colesterol total) e escores do ICVH foram calculados conforme proposto pela American Heart Association . Modelos de regressão logística bruta e ajustada foram construídos para analisar associações de métricas e escores do ICVH com diagnóstico de FFA. O nível de significância foi estabelecido em 0,05. Resultados A idade mediana da amostra foi de 55 anos, e 54,4% eram mulheres. Nos modelos ajustados, os escores de ICVH não apresentaram associação significativa com diagnóstico de FFA prevalente [odds ratio (OR):0,96; intervalo de confiança de 95% (IC95%):0,80-1,16; p=0,70). Perfis de pressão arterial ideal (OR:0,33; IC95%:0,1-0,74; p=0,007) e colesterol total ideal (OR:1,88; IC95%:1,19-2,98; p=0,007) foram significativamente associados com o diagnóstico de FFA. Conclusões Não foram identificadas associações significativas entre escores de ICVH global e diagnóstico de FFA após ajuste multivariado em nossas análises, devido, ao menos em parte, às associações antagônicas da FFA com métricas de pressão arterial e de colesterol total do ICVH. Nossos resultados sugerem que estimar a prevenção da FFA por meio de escore de ICVH global pode não ser adequado, e as métricas do ICVH devem ser consideradas separadamente.

Abstract Background The association between ideal cardiovascular health (ICVH) status and atrial fibrillation or flutter (AFF) diagnosis has been less studied compared to other cardiovascular diseases. Objective To analyze the association between AFF diagnosis and ICVH metrics and scores in the Brazilian Longitudinal Study of Adult Health (ELSA-Brasil). Methods This study analyzed data from 13,141 participants with complete data. Electrocardiographic tracings were coded according to the Minnesota Coding System, in a centralized reading center. ICVH metrics (diet, physical activity, body mass index, smoking, blood pressure, fasting plasma glucose, and total cholesterol) and scores were calculated as proposed by the American Heart Association. Crude and adjusted binary logistic regression models were built to analyze the association of ICVH metrics and scores with AFF diagnosis. Significance level was set at 0.05. Results The sample had a median age of 55 years and 54.4% were women. In adjusted models, ICVH scores were not significantly associated with prevalent AFF diagnosis (odds ratio [OR]:0.96; 95% confidence interval [95% CI]:0.80-1.16; p=0.70). Ideal blood pressure (OR:0.33; 95% CI:0.15–0.74; p=0.007) and total cholesterol (OR:1.88; 95% CI:1.19–2.98; p=0.007) profiles were significantly associated with AFF diagnosis. Conclusions No significant associations were identified between global ICVH scores and AFF diagnosis after multivariable adjustment in our analyses, at least partially due to the antagonistic associations of AFF with blood pressure and total cholesterol ICVH metrics. Our results suggest that estimating the prevention of AFF burden using global ICVH scores may not be adequate, and ICVH metrics should be considered in separate.

Abstract Gut bacterial β-glucuronidase (GUS) can reactivate xenobiotics that exert enterohepatic circulation- triggered gastrointestinal tract toxicity. GUS inhibitors can alleviate drug-induced enteropathy and improve treatment outcomes. We evaluated the inhibitory effect of Polygonum cuspidatum Siebold & Zucc. and its major constituents against Escherichia coli GUS (EcGUS), and characterized the inhibitory mechanism of each of the components. Trans-resveratrol 4’-O-β-D-glucopyranoside (HZ-1) and (-)-epicatechin gallate (HZ-2) isolated from P. cuspidatum were identified as the key components and potent inhibitors. These two components displayed strong to moderate inhibitory effects on EcGUS, with Ki values of 9.95 and 1.95 μM, respectively. Results from molecular docking indicated that HZ-1 and HZ-2 could interact with the key residues Asp163, Ser360, Ile 363, Glu413, Glu504, and Lys 568 of EcGUS via hydrogen bonding. Our findings demonstrate the inhibitory effect of P. cuspidatum and its two components on EcGUS, which supported the further evaluation and development of P. cuspidatum and its two active components as novel candidates for alleviating drug-induced damage in the mammalian gut.

OBJECTIVES: The chemokine ligand (CCL) 21 regulates the maturation, migration, and function of dendritic cells, and has been implicated in the pathogenesis of asthma. This study aimed to investigate the association between serum CCL21 levels and asthma control. METHODS: The serum levels of CCL21 and other inflammatory cytokines were analyzed in patients with asthma (n=44) and healthy controls (n=35) by enzyme-linked immunosorbent assay. IgE levels and eosinophil counts were determined by turbidimetric inhibition immunoassay and fully automatic blood analysis, respectively. The Asthma Control Test (ACT) questionnaire was used, and spirometry and fractional exhaled nitric oxide (FENO) measurements were performed. A multiple unpaired Student’s t-test was performed to analyze the differences in CCL21 and interleukin levels between patients with asthma and healthy controls. The correlation of CCL21 levels with disease severity was evaluated using the Pearson’s rank correlation test. RESULTS: Serum CCL21 levels were lower in patients with asthma (254.78±95.66 pg/mL) than in healthy controls (382.95±87.77 pg/mL) (p<0.001). Patients with asthma had significantly higher levels of IL-1β (19.74±16.77 vs. 2.63±5.22 pg/mL), IL-6 (7.55±8.65 vs. 2.37±2.47 pg/mL), and tumor necrosis factor-α (12.70±12.03 vs. 4.82±3.97 pg/mL) compared with the controls. CCL21 levels were positively correlated with the ACT score (rs=0.1653, p=0.0062), forced expiratory volume in 1s (FEV1)/forced vital capacity (rs=0.3607, p<0.0001), and FEV1 (rs=0.2753, p=0.0003), and negatively correlated with FENO (rs=0.1060, p=0.0310). CCL21 levels were negatively correlated with serum IgE levels (rs=0.1114, p=0.0268) and eosinophil counts (rs=0.3476, p<0.0001). CONCLUSIONS: Serum CCL21 levels may be a new biomarker for assessing asthma control.

RESUMO Pholiota adiposa é um cogumelo com excelentes propriedades nutricionais e medicinais. No entanto, o rendimento do corpo de frutificação deste é baixo e o potencial de cultivo comercial deste fungo é limitado. 279 linhagens cruzadas foram obtidas pelo cruzamento mono-mono de linhagens monocarióticas derivadas de P. adiposa HS5 e HS4. Enzimas ligninolíticas e taxas de crescimento micelial foram utilizadas como marcadores para a triagem das linhagens cruzadas, e 18 linhagens foram seleccionadas para análise posterior. A linhagem cruzada A10B4 apresentou o maior rendimento (ie 165,91 ± 12,56 g por sacola), o qual 31,34 g e, 74,48 g a mais do que as linhagens HS5 e HS4, respetivamente. O tempo de colonização micelial de A10B4 foi 25,18 ± 1,33 d, sendo 5,64 d menor que HS5. A10B4 foi caracterizado, por marcadores moleculares de repetição de sequência inter-simples. Foram observadas diferenças nos produtos de PCR de linhagens parentais e cruzadas. Portanto, a linhagem híbrida recém-desenvolvida A10B4 (Renominada P. adiposa HS54), exibindo alto rendimento, pode ser adequada para cultivo comercial.

ABSTRACT Pholiota adiposa is a mushroom with excellent nutritional and medicinal properties. However, fruiting body yields are low, and the commercial cultivation potential of this fungus is limited. Here, 279 crossbred strains were obtained by the mono-mono crossing of monokaryotic strains derived from P. adiposa HS5 and HS4. Laccase enzymes and mycelial growth rates were used as markers to screen the crossbred strains, and 18 strains were selected for further analysis. Crossbred strain A10B4 displayed the highest yield (i.e. 165.91 ± 12.56 g per bag), which was 31.34 g and 74.48 g more than that of strains HS5 and HS4, respectively. The mycelial colonization time of A10B4 was 25.18 ± 1.33 d, which was 5.64 d shorter than that of HS5. A10B4 was characterized by inter-simple sequence repeat molecular markers. Differences in PCR products from parental and crossbred strains were observed. Therefore, the newly developed hybrid strain A10B4, named P. adiposa HS54 exhibiting high yield, might be suitable for commercial cultivation.
